Abstract:
Cancer-on-a-chip has recently emerged and attracted wide attention in the field of anticancer drug screening. Compared with traditional cancer models, it enables the faithful recapitulation of the native cancer microenvironment in a high-throughput and cost-effective manner. Moreover, the application of cancer-on-a-chip usually simplifies the process of drug loading and cellular characterization. Therefore, it is considered a promising tool for drug screening in both drug development and personal drug administration. Here, we review recent advances in cancer-on-a-chip assisted drug screening, with a special focus on the concrete process of constructing the microfluidic chip. It is thought that cancer-on-a-chip will strongly promote the development of drug screening by serving as a reliable and convenient cancer model.

What problem does this paper attempt to address?
The main problem that this paper attempts to solve is the limitations of traditional cancer models in drug screening. Specifically, traditional models such as 2D cell culture, cancer spheroids and animal models cannot fully simulate the microenvironment of human cancers, resulting in a deviation between the tumor's response to anticancer drugs and clinical results, and thus affecting the reliability and effectiveness of drug screening. Therefore, this paper explores how to improve the drug - screening process by constructing "cancer - on - a - chip", especially how to use this technology to predict drug effects more accurately, thereby promoting the development of new drugs and the selection of personalized treatment regimens. "Cancer - on - a - chip" combines in vitro biological cancer models with microfluidic chip technology and can faithfully reproduce the native cancer microenvironment in a high - throughput and cost - effective manner. In addition, it simplifies the processes of drug loading and cell feature analysis and is considered a promising tool in the field of drug screening. The paper focuses on the construction methods of "cancer - on - a - chip" and the progress of its application in drug screening, emphasizing its potential in improving the efficiency and accuracy of drug screening.
