Abstract:Abstract Rhizodegradation is a promising cleanup technology where microorganisms degrade soil contaminants in the rhizosphere. A symbiotic relationship is expected to occur between plant roots and soil microorganisms in contaminated soils that enhance natural microbial degradation in soils. However, little is known about how this initial microbiota influences the rhizodegradation outcome in a context of different soil microbiotas. Recent studies have hinted that soil initial diversity has a determining effect on the outcome of contaminant degradation. To test this hypothesis, we planted (P) or not (NP) balsam poplars ( Populus balsamifera ) in two soils of contrasting diversity (agricultural and forest) that were contaminated or not with 50 mg kg -1 of phenanthrene (PHE). The DNA from the rhizosphere of the P and the bulk soil of the NP pots was extracted and the bacterial genes encoding for the 16S rRNA, the PAH ring-hydroxylating dioxygenase alpha subunits (PAH-RHDα) of gram-positive (GP) and gram-negative (GN) bacteria, and the fungal ITS region were sequenced to characterize the microbial communities. and the abundance of the PAH-RHDα genes were also quantified by real-time quantitative PCR. Plant presence had a significant effect on PHE degradation only in the forest soil, whereas both NP and P agricultural soils degraded the same amount of PHE. Bacterial communities were principally affected by the soil type, and upon contamination the dominant PAH degrading community was similarly constrained by soil type. Our results highlight the crucial importance of soil microbial and physicochemical characteristics in the outcome of rhizoremediation.
Substrate Interactions in Btex and Mtbe Mixtures by an Mtbe-Degrading Isolate

Abstract:Groundwater contaminant plumes from recent accidental gasoline releases often contain the fuel oxygenate MTBE (methyl tert-butyl ether) together with BTEX (benzene, toluene, ethylbenzene, o-xylene, m-xylene and p-xylene) compounds. This study evaluates substrate interactions during the aerobic biotransformation of MTBE and BTEX mixtures by a pure culture, PM1, capable of utilizing MTBE for growth. PM1 was unable to degrade ethylbenzene and two of the xylene isomers at concentrations of 20 mg/L following culture growth on MTBE. In addition, the presence of 20 mg/L of ethylbenzene or the xylenes in mixtures with MTBE completely inhibited MTBE degradation. When MTBE-grown cells of PM1 were exposed to MTBE/ benzene and MTBE/toluene mixtures, MTBE degradation proceeded, while the degradation of benzene and toluene was delayed for several hours. Following this initial lag, benzene and toluene were degraded rapidly, while the rate of MTBE degradation slowed significantly. MTBE degradation did not increase to previous rates until benzene and toluene were almost entirely degraded. The lag in benzene and toluene degradation was presumably due to the induction of the enzymes necessary for BTEX degradation. Once these enzymes were induced, sequential additions of benzene or toluene were degraded rapidly, and growth on benzene and toluene was observed. The results of this study suggest that BTEX and MTBE degradation occurs primarily via two independent and inducible pathways. If subsurface microbial communities behave similarly to the culture used in this study, the observed severe inhibition of MTBE degradation by ethylbenzene and the xylenes and the partial inhibition by benzene and toluene suggest that the biodegradation of MTBE in subsurface environments would most likely be delayed until MTBE has migrated beyond the BTEX plume.

Abstract:In this study, degradation of methyl tert-butyl ether (MTBE) by resting cells of Methylibium petroleiphilum PM1 was performed in poor nutrition solution, major component of which was MTBE. It was found that the biomass was hard to increase in poor nutrition, and the MTBE degradation activity was enhanced by 4.65-fold when 1 mM of Ba2+ was added into deionized water. It was also found that the MTBE degradation could be significantly improved by the dissolved oxygen level. All of 50 mg L−1 MTBE could be degraded under aerobic condition, while only 5% was degraded under anaerobic condition by resting cells in poor nutrition solution after 12 h. In the above solution, the degradation of tert-butyl alcohol (TBA) was found to be faster than that of MTBE, which suggested that TBA degradation might not be the limiting step in MTBE metabolism. While in the poor nutrition solution with the mixture of MTBE and TBA, the addition of TBA did not affect MTBE degradation while MTBE inhibited TBA degradation weakly, which suggested that different and independent enzymes were responsible for degrading such compounds. The success of MTBE degradation by PM1 cells in real contaminated groundwater demonstrated its feasibility to biodegrade MTBE under poor environment, and it also indicated the great potential of MTBE bioremediation by entrapped cells in future application.

Abstract:Methyl tert-butyl ether(MTBE)is widely used as reformulated gasoline oxygenate.It is so much used that the amount of MTBE discharged into the environment is increasing continuously.MTBE is of great water-solubility,so it diffuses rapidly in the water and composes potential pollution to the surface water and the groundwater.Furthermore,MTBE is hard to be biodegraded,so it may endanger the groundwater persistently.The studies on the toxicology of MTBE indicate that it has definite toxicity to the animals' livers,kidneys and the immunity functions,but really weaker toxicity to the procreation.MTBE could metabolize to methyl alcohol and tert-butyl alcohol(TBA),which are more toxic than MTBE itself in the bodies of animals and human beings.Therefore,some countries begin to restrict or forbid the use of MTBE.However,at present,lead-free gasoline with MTBE as reformulated gasoline oxygenate is still widely used in China.So we should pay more attention to its pollution as early as possible.The source and distributing of MTBE in the environment,its environmental behavior,the ecologic effects and toxicology of MTBE,including toxicity of oral exposure and inhalation exposure,toxicity of body exposure and toxicity of MTBE to plants and marine organisms,and metabolism study were reviewed.
A Field Test Comparison of Hiking Stick Use on Heartrate and Rating of Perceived Exertion

Abstract:The purpose of this study was to compare heartrate carrying a load and rating of perceived exertion with and without hiking sticks while ascending and descending a slope. 11 novice, moderately fit volunteers, ages 18 to 21 years (M = 19 3 yr.) completed two alternate 50-meter, uphill and downhill hikes on a 40° slope during randomly ordered trials with and without fitted hiking sticks and backpacks (15 kg). Paired t test comparisons for 4 trials indicated that mean heartrate was significantly lower only following the first ascent by those using hiking sticks than those without sucks. Rating of perceived exertion also was significantly lower (p<.05) for those using hiking sticks. Perhaps heartrate may be lower at the onset of climbing using hiking sticks, but as the duration the hike is extended, heartrates become comparable, presumably due to the transfer of energy utilization from the legs to the upper body.

Abstract:Widespread contamination of groundwater by methyl tertiary butyl ether (MTBE) has triggered the exploration of different technologies for in situ removal of the pollutant, including biostimulation of naturally occurring microbial communities or bioaugmentation with specific microbial strains known to biodegrade the oxygenate. After laboratory studies revealed that bacterial strain PM1 rapidly and completely biodegraded MTBE in groundwater sediments, the organism was tested in an in situ field study at Port Hueneme Naval Construction Battalion Center in Oxnard, California. Two pilot test plots (A and B) in groundwater located down-gradient from an MTBE source were intermittently sparged with pure oxygen. Plot B was also inoculated with strain PM1. MTBE concentrations up-gradient from plots A and B initially varied temporally from 1.5 to 6 mg MTBE/L. Six months after treatment began, MTBE concentrations in monitoring wells down-gradient from the injection bed decreased substantially in the shallow zone of the groundwater in plots A and B, thus even in the absence of the inoculated strain PM1. In the deeper zone, downstream MTBE concentrations also decreased in plot A and to a lesser extent in plot B. Difficulties in delivery of oxygen to the deeper zone of plot B, evidenced by low dissolved oxygen concentrations, were likely responsible for low rates of MTBE removal at that location. We measured the survival and movement of strain PM1 in groundwater samples using two methods for detection of DNA sequences specific to strain PM1: TaqMan quantitative polymerase chain reaction, and internal transcribed spacer region analysis. A naturally occurring bacterial strain with > 99% 16S rDNA sequence similarity to strain PM1 was detected in groundwater collected at various locations at Port Hueneme, including outside the plots where the organism was inoculated. Addition of oxygen to naturally occurring microbial populations was sufficient to stimulate MTBE removal at this site. In some cases, however, inoculation with an MTBE-degrading culture may be warranted.

Abstract:Methyl tert-butyl ether (MTBE) has been used in many countries as an octane- enhancing replacement for lead, primarily in mid- a nd high-grade gasoline. The prevalent use of MTBE has led to its introduction into environmen t. MTBE is poorly adsorbed, biologically and chemically stable, making it very persistent in the environment. MTBE has been classified as a possible human carcinogen. Many studies have been carried out to find a better method for the degradation and removal of MTBE in the environment. Activated carbon, clay mineral, resin and zeolite have been used as adsorbents for removal of MTBE. Persulfate, potassium permanganate, Fenton oxidation and photocatalytic oxidation have been studied for chemical degradation of MTBE. In addition, different bacteri a have been isolated and identified for biodegradation of MTBE. This review summarizes current knowledge on the adsorption, chemical degradation and biodegradation of MTBE.

Abstract:Abstract Understanding anaerobic biodegradation of ether oxygenates beyond MTBE in groundwater is important, given that it is replaced by ETBE as a gasoline additive in several regions. The lack of studies demonstrating anaerobic biodegradation of ETBE, and its product TBA, reflects the relative resistance of ethers and alcohols with a tertiary carbon atom to enzymatic attack under anoxic conditions. Anaerobic ETBE- or TBA-degrading microorganisms have not been characterized. Only one field study suggested anaerobic ETBE biodegradation. Anaerobic (co)metabolism of ETBE or TBA was reported in anoxic microcosms, indicating their biodegradation potential in anoxic groundwater systems. Non-isotopic methods, such as the detection of contaminant loss, metabolites, or ETBE- and TBA-degrading bacteria are not sufficiently sensitive to track anaerobic biodegradation in situ . Compound- and position-specific stable isotope analysis provides a means to study MTBE biodegradation, but isotopic fractionation of ETBE has only been studied with a few aerobic bacteria (εC −0.7 to −1.7‰, εH −11 to −73‰) and at one anoxic field site (δ 2 H-ETBE +14‰). Similarly, stable carbon isotope enrichment (δ 13 C-TBA +6.5‰) indicated TBA biodegradation at an anoxic field site. CSIA and PSIA are promising methods to detect anaerobic ETBE and TBA biodegradation but need to be investigated further to assess their full potential at field scale. Graphical abstract

Abstract:This study employed innovative technologies to evaluate multiple lines of evidence for natural attenuation (NA) of methyl tertiary-butyl ether (MTBE) in groundwater at the 22 Area of Marine Corps Base (MCB) Camp Pendleton after decommissioning of a biobarrier system. For comparison, data from the 13 Area Gas Station where active treatment of MTBE is occurring was used to evaluate the effectiveness of omic techniques in assessing biodegradation. Overall, the 22 Area Gas Station appeared to be anoxic. MTBE was detected in large portion of the plume. In comparison, concentrations of MTBE at the 13 Area Gas Station were much higher (42,000 μg/L to 2800 μg/L); however, none of the oxygenates were detected. Metagenomic analysis of the indigenous groundwater microbial community revealed the presence of bacterial strains known to aerobically and anaerobically degrade MTBE at both sites. While proteomic analysis at the 22 Area Gas Station showed the presence of proteins of MTBE degrading microorganisms, the MTBE degradative proteins were only found at the 13 Area Gas Station. Taken together, these results provide evidence for previous NA of MTBE in the groundwater at 22 Area Gas Station and demonstrate the effectiveness of innovative-omic technologies to assist monitored NA assessments.
Reconstructed metagenomes reveal changes of microbial functional profiling during PAHs degradation along a rice (Oryza sativa) rhizosphere gradient.

Abstract:AIMS:The response of microbial metagenome to polycyclic aromatic hydrocarbons (PAHs) degradation in the rice rhizosphere remains poorly understood. We investigated the spatial and temporal variations of microbial communities and reconstructed metagenomes along the rice rhizosphere gradient during PAHs degradation. METHODS AND RESULTS:The experiment was performed in rhizoboxes, in which the rhizosphere region was divided into five 1-mm thick layers. Based on denaturant gradient gel electrophoresis profiling and sequencing of bacterial and archaeal 16S rRNA genes, predicted metagenomes were reconstructed. The microbial communities in the rice rhizosphere were influenced by the PAHs concentration and distance from the root surface during PAHs degradation. Correlation network analysis showed that archaea played an important role in PAHs degradation. Predicted metagenomes can be clustered into two groups with high and low PAHs degrading potential, respectively. The relative abundance of genes for defense mechanisms, replication, recombination and reparation was significantly higher in samples with high PAHs degrading potentials. The relative abundance of the dioxygenase gene was greater near the root surface of the rice. However, the abundance of aldolase and dehydrogenase was constant in rhizosphere soils at different distances from the root surface. CONCLUSIONS:Distance from root surface and PAH concentrations affected the microbial communities and metagenomes in rice rhizosphere. The abundance of dioxygenase genes relating to PAH degradation in metagenomes mirrored the PAH degradation potential in rice rhizosphere. SIGNIFICANCE AND IMPACT OF THE STUDY:Our findings suggested that the predicted metagenomes reconstructed from 16S rRNA marker gene sequences provide further insights into the spatial variation and dynamics of microbial functioning that occur during bioremediation.
Phytoremediation: past promises and future practises

Abstract:Plant-based bioremediation technologies have received recent attention as strategies to clean-up contaminated soils and water. These strategies have collectively been termed phytoremediation and refer to the use of green plants and their associated microbiota for the in situ treatment of soil, sediment, and ground water. Biologically based remediation strategies, including phytoremediation, have been estimated to be four to 1000 times cheaper, on a per volume basis, than current non-biological technologies. Compounds targeted for phytoremediation strategies include heavy metals, chlorinated solvents, polycyclic aromatic hydrocarbons, polychlorinated biphenyls, pesticides, munitions and radionuclides. While some of these contaminants are more readily degraded or detoxified than others, plants or their attendant rhizosphere microbes have been shown in several instances to transform these compounds to some degree. The main types of phytoremediation strategies used include the stimulation of non-specific and specific authochthonous and zymogenous rhizosphere microorganisms (both bacteria and fungi) for the accelerated biodegradation of herbicide and solvent contaminants, the use of "hyperaccumulating" plants for remediation of soils contaminated with metals, and the use of plants to transform soluble contaminants to less soluble or less toxic forms. The ultimate goal of all phytoremediation technologies is to either remove the contaminant from the affected area, a process termed phytodecontamination, or to stabilize the contaminant to prevent movement or toxicological affects. Below ground phytodecontamination processes are thought to chiefly rely on rhizosphere degradation activity (either plant enzyme-or microbiologically-driven) to transform hazardous waste materials. Future biotechnological strategies for enhancing phytoremediation include enlarging root mass to increase adsorption area, using Agrobacterium rhizogenes, the direct genetic engineering of plants for altered biodegradation potential, and the genetic engineering of rhizosphere microorganisms. However, while phytoremediation processes hold great promise as means to remediate contaminated soils and water, there are advantages and disadvantages associated with these strategies that must be carefully considered. Whereas attractions of phytoremediation processes include cost effectiveness and non-invasiveness, they require relatively long periods of time, often require the disposal of toxic vegetation, are ineffective at remediating sites containing pollutants located deep into the soil profile, do not work on all contaminants, are sensitive to contaminant types and concentrations, may end up producing secondary metabolites which are more toxic than parent compounds, and in many instances don’t remove environmentally significant quantities of pollutants. Abstract:Trichloroethylene (TCE) poses a significant environmental threat in groundwater and soil, necessitating effective remediation strategies. Phytoremediation offers a cost-effective and environmentally friendly approach to remediation. However, the mechanisms governing plant uptake, volatilisation, and degradation of TCE remain poorly understood. This review explores the mechanisms of TCE phytoremediation, metabolic pathways, and influencing factors, emphasizing future research directions to improve the understanding of TCE phytoremediation. The results showed that although the proportion of TCE phytovolatilisation is limited, it is important at sites chronically contaminated with TCE. The rhizosphere is a key microzone for pollutant redox reactions that significantly enhance its effectiveness when its characteristics are fully utilised and manipulated through reinforcement. Future research should focus on manipulating microbial communities through methods such as the application of endophytic bacteria and genetic modification. However, practical applications are in their infancy and further investigation is needed. Furthermore, many findings are based on non-uniform parameters or unstandardised methods, making them difficult to compare. Therefore, future studies should provide more standardised experimental parameters and employ accurate and standardised methods to develop suitable prediction models, enhancing data comparability and deepening our understanding of plant detoxification mechanisms.
Unveiling the novel role of ryegrass rhizospheric metabolites in benzo[a]pyrene biodegradation

Abstract:Rhizoremediation is a promising remediation technology for the removal of soil persistent organic pollutants (POPs), especially benzo[a]pyrene (BaP). However, our understanding of the associations among rhizospheric soil metabolites, functional microorganisms, and POPs degradation in different plant growth stages is limited. We combined stable-isotope probing (SIP), high-throughput sequencing, and metabolomics to analyze changes in rhizospheric soil metabolites, functional microbes, and BaP biodegradation in the early growth stages (tillering, jointing) and later stage (booting) of ryegrass. Microbial community structures differed significantly among growth stages. Metabolisms such as benzenoids and carboxylic acids tended to be enriched in the early growth stage, while lipids and organic heterocyclic compounds dominated in the later stage. From SIP, eight BaP-degrading microbes were identified, and most of which such as Ilumatobacter and Singulisphaera were first linked with BaP biodegradation. Notably, the relationship between the differential metabolites and BaP degradation efficiency further suggested that BaP-degrading microbes might metabolize BaP directly to produce benzenoid metabolites (3-hydroxybenzo[a]pyrene), or utilize benzenoids (phyllodulcin) to stimulate the co-metabolism of BaP in early growth stage; some lipids and organic acids, e.g. 1-aminocyclopropane-1-carboxylic acid, might provide nutrients for the degraders to promote BaP metabolism in later stage. Accordingly, we determined that certain rhizospheric metabolites might regulate the rhizospheric microbial communities at different growth stages, and shift the composition and diversity of BaP-degrading bacteria, thereby enhancing in situ BaP degradation. Our study sheds light on POPs rhizoremediation mechanisms in petroleum-contaminated soils.
Dissipation of Pentachlorophenol in the Aerobic-Anaerobic Interfaces Established by the Rhizosphere of Rice (oryza Sativa L.) Root

Abstract:Phytoremediation is an emerging technology for the detoxification and remediation of organic pollutants such as pentachlorophenol (PCP). To investigate the dissipation behavior of PCP in the aerobic–anaerobic interfaces established by the rhizosphere of rice (Oryza sativa L.) root, a glasshouse experiment was conducted using a specially designed rhizobox. The possible biogeochemical mechanisms were also studied through illustration of the dynamic behavior of important electron acceptors and donors that are potentially involved in the reductive dechlorination and aerobic catabolism processes of PCP. The soil was spiked with 20 ± 0.25 and 45 ± 0.25 mg of PCP kg−1 soil. Soil in the rhizobox was divided into five different compartments at various distances from the root surface. Maximum dissipation of PCP in planted soil was observed at 3‐mm distance from the root zone as well as rapid changes in concentrations of sulfate, chloride, nitrate, and ammonium at the same distance from the root. In contrast, in the unplanted soil, no difference was observed in the PCP concentration with increasing distance. After 45 d, a significantly higher concentration of PCP was degraded in planted soil compared with unplanted soil. In the unplanted microcosms, about 45% of the initial PCP was lost at both low and high added rates, respectively. This was, proportionately, a significantly smaller percentage compared with the planted rhizosphere (an average of 66 and 64.5%, respectively). Moreover, the correlations of PCP dissipation with SO42−, NO3−, and Fe3+ were significantly negative, while the correlations of PCP dissipation with NH4+, Fe2+, and Cl− were significantly positive. This suggested the oxidization of soil constituents can inhibit aerobic catabolism of PCP by consuming O2, and the reduction of soil constituents can inhibit anaerobic reductive dechlorination of PCP. Therefore, the significance of the rhizosphere in phytoremediation of chlorinated compounds such as PCP differs significantly between wetland and rainfed systems.

Abstract:A methyl tert-butyl ether degradation strain A1 was isolated from the soil under an old Gingko tree. It was identified preliminarily as Comamonas testosterone by 16S rDNA sequence analysis. The main factors including inoculation amount of microbes, pH, temperature and MTBE concentration that may affect the degradation efficiency of MTBE were further studied. The results indicated that the optimum conditions were as following: pH 7.0, temperature 25 degrees C, inoculation amount of microbes was 2 mL (D600 = 2.523 A), initial MTBE concentration 50 mg/L. Under that condition, MTBE can be reduced by 98.89% with seven days (compared with the blank, the volatilization of MTBE was 46.55%). In addition, the biodegradation process of MTBE can be well described by enzymatic reaction of high concentration inhibition, with the maximum substrate utilization rate 0.872 d(-1), Michaelis-Menten constant 7.832 mg x L(-1), inhibitory constant 130.75 mg x L(-1) respectively.

Abstract:Biodegradation of methyl tert-butyl ether (MTBE) by Methylibium petroleiphilum PM1 was studied in this paper. The results showed that MTBE could not be degraded until it underwent a long induction period. It was observed that DO decreased and pH was slightly reduced during the process of 50 mg·L-1 MTBE degradation. Tert-butyl alcohol (TBA), a frequently detected intermediate of MTBE degradation, was also detected by GC/MS in this paper. Tert-butyl formate (TBF), TBA and acetone could be degraded quickly, while isopropanol showed absolute resistance to biodegradation by PM1. Moreover, MTBE was inhibited completely by the addition of isopropanol.

Abstract:Spillage from oil refineries, pipelines, and service stations consistently leads to soil, food and groundwater contamination. Bacterial-assisted phytoremediation is a non-invasive and sustainable solution to eliminate or decrease the concentration of xenobiotic contaminants in the environment. In the present study, a protected area interested by a fuel discharge was considered to assess a bioremediation intervention. From the spill point, a plume of contamination flowed South-West into the aquifer, eventually reaching a wetland area. Soils, groundwaters and plants belonging to the species Scirpus sylvaticus (L.) were sampled. In the majority of the soil samples, concentrations of total petroleum hydrocarbons, both C ≤ 12 and C > 12, exceeded legal limits set forth in Directive 2000/60/EC. The analysis of diatom populations, used as ecological indicators, evidenced morphology alterations and the presence of Ulnaria ulna and Ulnaria biceps species, previously detected in hydrocarbon-polluted waters. Tests for phytotoxicity and phytodegradation, carried out in soil mesocosms, planted with Zea mays and Helianthus annuus , demonstrated that both species significantly contributed to the removal of total petroleum hydrocarbons. Removal of C ≤ 12 and C > 12 petroleum hydrocarbons was in the range of 80%–82% for Z. mays and 71%–72% for H. annuus . Microbial communities inhabiting high organic carbon and vegetated soils were more active in hydrocarbon degradation than those inhabiting subsoils, as evidenced by soil slurry experiments. The abundance of functional genes encoding toluene-benzene monooxygenase ( tbmD ) and alkane hydroxylase ( alkB ), quantified in environmental samples, confirmed that the plant rhizosphere recruited a microbial community with higher biodegradation capacity. Bacterial strains isolated from the sampling site were able to grow on model hydrocarbons (hexane, hexadecane and o-, m-, p -xylene) as sole carbon and energy sources, indicating that a natural bio-attenuation process was on-going at the site. The bacterial strains isolated from rhizosphere soil, rhizoplane and endosphere showed plant growth promoting traits according to in vitro and in vivo tests on Z. mays and Oryza sativa , allowing to forecast a possible application of bacterial assisted rhizoremediation to recover the protected area.

Abstract:Despite the effectiveness of microbial-phytoremediation for remediating total petroleum hydrocarbons (TPH)-contaminated soil, the underlying mechanisms remain elusive. This study investigated the whole-genome and biological activity of Rhodococcus erythropolis KB1, revealing its plant growth promotion (PGP), TPH degradation, and stress resistance capabilities. Phytoremediation (using alfalfa) and plant-microbial remediation (using alfalfa and KB1) were employed to degrade TPH. The highest TPH degradation rate, reaching 95%, was observed with plant-microbial remediation. This is attributed to KB1's ability to promote alfalfa growth, induce the release of signaling molecules to activate plant antioxidant enzymes, actively recruit TPH-degrading bacteria (e.g., Sphingomonas, Pseudomonas, C1-B045), and increase soil nitrogen and phosphorus levels, thereby accelerating TPH degradation by both plants and microorganisms. This study demonstrates that R. erythropolis KB1 holds great potential for enhancing the remediation of TPH-contaminated soil through its multifaceted mechanisms, particularly in plant-microbial remediation strategies, providing valuable theoretical support for the application of this technology.
