Abstract:The pathology of the fibrotic proliferative vitreoretinopathy (PVR) membrane represents an excessive wound healing response characterised by cells' proliferation, migration and secretion of extracellular matrix molecules (ECMs). Retinal pigment epithelial (RPE) cells are a major cellular component of the fibrotic membrane. Endothelin-1 (ET-1) has been reported to be involved in the development of PVR in vivo research. However, little is known about the role of ET-1 in RPE cells in vitro. In the present study, we investigated the role of ET-1 in the proliferation, migration and secretion of ECMs (such as type I collagen and fibronectin) in RPE cells in vitro. Our results illustrated that ET-1 promoted the proliferation, migration and secretion of ECMs through the protein kinase B (Akt) and extracellular signal-regulated kinase (Erk) signaling pathways in RPE cells in vitro. These findings strongly suggested that ET-1 may play a vital role in the development of PVR.

Abstract:The BioCD is an optical biosensor that uses highspeed spinning-disk interferometry for direct detection of antigen binding in multiplexed immunoassay protein arrays. The high-frequency sampling suppresses the noise floor and enables a metrology-limited height resolution of 1 picometer for measuring surface-bound protein. In assay protocols there is a direct trade-off between multiplex degree and limit of detection. In this paper, we demonstrate quantitative assays and the scaling properties of direct detection on the BioCD in assays of clinical relevance for human diagnostics, including haptoglobin assays and prostate-specific antigen assays in human patient sera.
Long-Term Effects on Retinal Structure and Function in a Mouse Endothelin-1 Model of Retinal Ganglion Cell Degeneration

Abstract:Purpose: To study the long-term effects of endothelin-1 (ET-1)-induced retinal pathologies in mouse, using clinically relevant tools. Methods: Adult C57BL/6 mice (7-9 weeks old) were intravitreally injected with PBS (n = 10) or 0.25 (n = 8), 0.5 (n = 8), or 1 nmol ET-1 (n = 9) and examined using electroretinogram, optical coherence tomography (OCT), and Doppler OCT at baseline and postinjection days 10, 28, and 56. Retinal ganglion cell (RGC) survival in retinal whole mount was quantified at days 28 and 56. Results: ET-1 induced immediate retinal arterial constriction. The significantly reduced total blood flow and positive scotopic threshold response in the 0.5- and 1-nmol ET-1 groups at day 10 were recovered at day 28. A-wave magnitude was also significantly reduced at days 10 and 28. While a comparable and significant reduction in retinal nerve fiber layer thickness was detected in all ET-1 groups at day 56, the 1-nmol group was the earliest to develop such change at day 28. All ET-1 groups showed a transient inner retinal layer thinning at days 10 and 28 and a plateaued outer layer thickness at days 10 to 56. The 1-nmol group showed a significant RGC loss over all retinal locations examined at day 28 as compared with PBS control. As for the lower-dosage groups, significant RGC density loss at central and midperipheral retina was detected at day 56 when compared with day 28. Conclusions: ET-1 injection in mice resulted in a transient vascular constriction and reduction in retinal functions, as well as a gradual loss of retinal nerve fiber layer and RGC in a dose-dependent manner.

Abstract:AIM:In order to study the effects of retinal glial cells on glaucomatous retinal ganglion cells damage,expression of TNF-α and TNF-R1 in retinal glial cells was observed in rat model with chronic elevated intraocular pressure glaucoma.METHODS:(1)Rats were rendered elevated intraocular pressure by ligating 2 episcleral veins with subconjunctival injection of 5-Fu.(2)Four weeks after operation,immuno-histological assays were carried out on cryostat sections.The co-expressions of TNF-α-GFAP,TNF-α-OX42,TNFR-1-GFAP,and TNFR-1-NeuN were observed via a confocal laser scanning microscope,respectively.RESULTS:(1)Elevated intraocular pressure was consistently maintained for up to 4 weeks in model group.(2)The co-expressions of TNF-α and GFAP,TNF-α and OX42 were detected in retina in treatment group,respectively.(3)The co-expressions of TNFR-1 and GFAP were also detected in retina,but the co-expressions of TNFR-1 and NeuN were not detected in retina in treatment group.CONCLUSION:TNF-α that activated retinal glial cells may play an important role in glaucomatous retinal ganglion cell damage.
Effect of Dual Endothelin Receptor Antagonist on a Retinal Degeneration Animal Model by Regulating Choroidal Microvascular Morphology

Abstract:Objective. Clinical studies have found that increasing levels of plasma endothelin-1 (ET-1) might inhibit choroidal blood flow (BF) and promote choroidal vasoconstriction. This study was designed to investigate ET-1 levels and its effect on choroidal microvascular morphology in a retinitis pigmentosa (RP) animal model. Methods. Mice with retinal degeneration (rd10) were intragastrically administered bosentan, a dual endothelin receptor antagonist. We detected plasma ET-1 levels using an enzyme-linked immunosorbent assay (ELISA) kit at P14, P21, and P28 and evaluated ET-1 expression in RPE/choroid/sclera complexes using western blot and whole mount immunofluorescence staining at P28. Retinal thickness was measured using hematoxylin and eosin (H&E) staining at P28. At the same time, we also estimated choroidal microvascular densities using vascular luminal casting with a scanning electron microscope (SEM). Results. Plasma ET-1 levels were increased significantly in rd10 mice at P21 (65.48 +/- 24.83 pg/ml) and P28 (85.89 +/- 20.23 pg/ml) compared with C57BL/6J mice at P21 (33.52 +/- 16.33 pg/ml) and P28 (42.38 +/- 17.53 pg/ml); the expression of ET-1 was also upregulated in RPE/choroid/sclera complexes at P28. Bosentan inhibited ET-1 expression in plasma (P<0.05) and RPE/choroid/sclera complexes at P28 in rd10 mice. Choroidal microvascular densities were decreased in rd10 mice, and bosentan could weaken these changes. Conclusion. Plasma and local ET-1 was elevated in an animal model of RP, suggesting that it likely participates in the pathological progression of retinal degeneration and may thus provide a new intervention target. ET-1 blockade might exert its protective effect by elevating choroidal microvascular density via inhibition of ET-1.
Chemokine expression in retinal pigment epithelial ARPE-19 cells in response to coculture with activated T cells

Abstract:Purpose: To investigate the effects of T-cell-derived cytokines on gene and protein expression of chemokines in a human RPE cell line (ARPE-19). Methods: We used an in vitro coculture system in which the RPE and CD3/CD28-activated T-cells were separated by a membrane. RPE cell expression of chemokine genes was quantified using three different types of microarrays. Protein expression was determined by single and multiplex ELISA and immunoblotting. Results: Coculture with activated T-cells increased RPE mRNA and protein expression of chemokines CCL2 (MCP-1); CCL5 (RANTES); CCL7 (MCP-3); CCL8 (MCP-2); CXCL1 (GRO-α); IL8 (CXCL8); CXCL9 (MIG); CXCL10 (IP10); CXCL11 (ITAC); and CX3CL1 (fractalkine). CCL7, CXCL9, CXCL10, and CXCL11 were secreted significantly more in the apical direction. Using recombinant human cytokines and neutralizing antibodies, we identified IFNγ and TNFα as the two major T-cell-derived cytokines responsible for the RPE response. For CCL5, CXCL9, CXCL10, CXCL11, CXCL16, and CX3CL1, we observed a synergistic effect of IFNγ and TNFα in combination. CCL20, CXCL1, CXCL6, and IL8 were negatively regulated by IFNγ. Conclusions: RPE cells responded to exposure to T-cell-derived cytokines by upregulating expression of multiple chemokines related to microglial, T-cell, and monocyte chemotaxis and activation. This inflammatory stress response may have implications for immune homeostasis in the retina, and for the further understanding of inflammatory ocular diseases such as uveitis and AMD.

Abstract:Purpose: Endothelin-1 (ET-1) is a potent vasoactive factor implicated in development of diabetic retinopathy, which is commonly associated with retinal edema and hyperglycemia. Although the vasomotor activity of venules contributes to the regulation of tissue fluid homeostasis, responses of human retinal venules to ET-1 under euglycemia and hyperglycemia remain unknown and the ET-1 receptor subtype corresponding to vasomotor function has not been determined. Herein, we addressed these issues by examining the reactivity of isolated human retinal venules to ET-1, and results from porcine retinal venules were compared.Methods: Retinal tissues were obtained from patients undergoing enucleation. Human and porcine retinal venules were isolated and pressurized to assess diameter changes in response to ET-1 after exposure to 5 mM control glucose or 25 mM high glucose for 2 hours.Results: Both human and porcine retinal venules exposed to control glucose developed similar basal tone and constricted comparably to ET-1 in a concentration-dependent manner. ET-1-induced constrictions of human and porcine retinal venules were abolished by ET<sub>A</sub> receptor antagonist BQ123. During high glucose exposure, basal tone of human and porcine retinal venules was unaltered but ET-1-induced vasoconstrictions were enhanced.Conclusions: ET-1 elicits comparable constriction of human and porcine retinal venules by activation of ET<sub>A</sub> receptors. In vitro hyperglycemia augments human and porcine retinal venular responses to ET-1.Translational Relevance: Similarities in vasoconstriction to ET-1 between human and porcine retinal venules support the latter as an effective model of the human retinal microcirculation to help identify vascular targets for the treatment of retinal complications in patients with diabetes.

Abstract:Purpose To explore the relationship between plasmatic ET-1 and glaucoma especially NTG.Methods The plasmatic ET-1 in NTG、POAG and normal persons was measured with radioimmunoassay.Results The concentrations of plasmatic ET-1 were 34.5±5.70 pg/ml,37.3±6.19 pg/ml,39.2±6.15 pg/ml in NTG、POAG and normal persons respectively.The difference between NTG and normal persons was statistically significant(P<0.05),whereas the concentrations between NTG and POAG.POAG and normal persons were not statistically different(P=0.087,0.273> 0.05).Conclusion It is suggested that ET-1 may play an active vascular role locally to modulate the blood supply of optic nerve which is related with glancomatous optic neuropathy.(Chin J Ophthalmol and Otorhinolaryngology,2005,5:353～354)

Abstract:Abstract Background Neuroinflammation plays an important role in the pathogenesis of glaucoma. Tumor necrosis factor-alpha (TNF-α) is a major pro-inflammatory cytokine released from activated retinal glial cells in glaucoma. Here, we investigated how TNF-α induces retinal ganglion cell (RGC) hyperexcitability and injury. Methods Whole-cell patch-clamp techniques were performed to explore changes in spontaneous firing and evoked action potentials, and Na + currents in RGCs. Both intravitreal injection of TNF-α and chronic ocular hypertension (COH) models were used. Western blotting, immunofluorescence, quantitative real-time polymerase chain reaction (q-PCR), and terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) techniques were employed to investigate the molecular mechanisms of TNF-α effects on RGCs. Results Intravitreal injection of soluble TNF-α significantly increased the spontaneous firing frequencies of RGCs in retinal slices. When the synaptic transmissions were blocked, more than 90% of RGCs still showed spontaneous firing; both the percentage of cells and firing frequency were higher than the controls. Furthermore, the frequency of evoked action potentials was also higher than the controls. Co-injection of the TNF-α receptor 1 (TNFR1) inhibitor R7050 eliminated the TNF-α-induced effects, suggesting that TNF-α may directly act on RGCs to induce cell hyperexcitability through activating TNFR1. In RGCs acutely isolated from TNF-α-injected retinas, Na + current densities were upregulated. Perfusing TNF-α in RGCs of normal rats mimicked this effect, and the activation curve of Na + currents shifted toward hyperpolarization direction, which was mediated through p38 MAPK and STAT3 signaling pathways. Further analysis revealed that TNF-α selectively upregulated Nav1.6 subtype of Na + currents in RGCs. Similar to observations in retinas of rats with COH, intravitreal injection of TNF-α upregulated the expression of Nav1.6 proteins in both total cell and membrane components, which was reversed by the NF-κB inhibitor BAY 11-7082. Inhibition of TNFR1 blocked TNF-α-induced RGC apoptosis. Conclusions TNF-α/TNFR1 signaling induces RGC hyperexcitability by selectively upregulating Nav1.6 Na + channels, thus contributing to RGC apoptosis in glaucoma.

[The Effect of Vascular Endothelial Growth Factor and Hypoxia on the Secretion of Opticin in Retinal Pigment Epithelium Cells].

Abstract:OBJECTIVE:To evaluate the role of vascular endothelial growth factor or hypoxia on the secretion of opticin in retinal pigment epithelium cells. METHODS:Human RPE cells were cultured, the third to sixth passage of retinal pigment epithelium (RPE) cells were placed in 6-well culture plates at a density of 4 × 10(4)/well. For hypoxia experiment, the cells were cultured under hypoxic condition for different times. For vascular endothelial growth factor (VEGF) experiment, the media was changed to DMEM containing different concentration VEGF (1, 10, 50, 100 µg/L) for 24 h respectively. VEGF mRNA levels were determined by RT-RCR method. The protein content of opticin in RPE cells or culture media was detected by Western blot. Matrix metalloproteinase activity in culture media was analysis by zymography. One way ANOVA was used to test the comparisons between experimental groups and control group. RESULTS:Western blot experiment showed the opticin expression was not changed in RPE cells after hypoxia treatment, however was significantly decreased in culture media. Compared with control group (0.21 ± 0.03). The relative density of VEGF mRNA levels (0.81 ± 0.04, 0.67 ± 0.07) in RPE cells were increased after 12 h or 24 h hypoxia treatment (F = 483.60, P < 0.05). Opticin expression in RPE cells was also remain unchanged after vary concentration VEGF addition treatment (F = 2.16, P > 0.05), the relative density of opticin expression in VEGF conditioned culture medium were 0.65 ± 0.02, 0.52 ± 0.04, 0.23 ± 0.03, 0.30 ± 0.03 respectively, and the difference in culture media was significant compared to control group (0.73 ± 0.04) (F = 141.38, P < 0.05). Zymography indicate a matrix metalloproteinases type 2 digest band, the activities were enhanced with VEGF increasing. The decrease of opticin in culture media after VEGF treatment could be inhibited by low condition of EDTA. CONCLUSION:VEGF and hypoxia have an effect on the on the secretion of opiticin in RPE cells, it may be contributed to the increasing levels of matrix metalloproteinases type 2.

Abstract:Objectives Diabetic retinopathy (DR) is a retinal disease that arises from impaired glucose tolerance and leads to retinal microvascular leakages. Recent studies have indicated that DR pathogenesis is linked to dysfunctional retinal pigment epithelial (RPE) cells. Methods Investigating the potential interplay between endothelial cells (ECs) and RPE cells by treating ECs with high glucose (HG) and evaluating the function of cytokines released from ECs on the growth of RPE cells. Results The results revealed that high glucose-stimulated Human Umbilical Vein Endothelial Cells (HUVECs) activated the NF-κB signaling pathway, increased intracellular levels of reactive oxygen species (ROS) and expression of caspase 3 while also elevating HUVECs delivery of cytokines such as VEGF, TNF-α, IL-6, and IL-1β. Conclusions As a result of our study, cytokines released from HG-treated HUVECs impede the growth of ARPE-19 in vitro, highlighting the importance of functional ECs for exploring the underlying mechanisms of vascular-associated retinal dysfunction. Endothelial Notch Signaling Regulates the Function of the Retinal Pigment Epithelial Barrier via EC Angiocrine Signaling

Abstract:The outer blood-retina barrier (oBRB), comprises tightly connected retinal pigment epithelium (RPE) cells, Bruch's membrane, and choroid blood vessels, and is essential for retinal health and normal visual function. Disruption of the RPE barrier and its dysfunction can lead to retinal disorders such as age-related macular degeneration (AMD). In the present study, we investigated the essential role of choroid endothelial cells (ECs) in the RPE barrier formation process and its dysfunction. We discovered that ECs promoted RPE barrier formation through angiocrine signaling. Through blocking or activating endothelial Notch signaling and conducting experiments in vitro and in vivo, we confirmed that endothelial Notch signaling regulated the expression of heparin-binding epidermal growth factor (HBEGF) and consequently impacted the expression and activity of matrix metalloproteinases (MMP)-9 in RPE cells. This modulation influenced the RPE extracellular matrix deposition, tight junctions and RPE barrier function. In in vivo experiments, the intravitreal administration of recombinant HBEGF (r-HBEGF) alleviated the RPE barrier disruption induced by subretinal injection (SI) or laser treatment and also rescued RPE barrier disruption in endothelial Notch-deficient mice. Our results showed that the endothelial Notch signaling drove HBEGF expression through angiocrine signaling and effectively improved RPE barrier function by regulating the MMP-9 expression in RPE cells. It suggests that the modulation of Notch signaling in the choroidal endothelium may offer a novel therapeutic strategy for retinal degenerative diseases.

Abstract:Introduction: Endothelin-1 (ET-1) is a vasoconstrictor produced by the vascular endothelium [1]. It binds to endothelial B receptor(ETB) on the endothelial cells(EC) [2]. Increased ET-1 has been observed in the EC of patients with moderate to severe hypertension [3]. Studies suggest that the EC cytoskeletal disruptions could mediate its production[4,5,6]. ETB is a plasma membrane bound receptor present in the EC surface with EC glycocalyx (GCX) [7,8]. GCX is flow sensitive and consists of several transmembrane solid core proteins [10,11,12]. Linked to the core protein chains include heperan sulfate (HS) [12]. GCX HS and ETB are located in the surface of the EC and are both flow sensitive. We hypothesize that the shear stress induced upregulation of the GCX HS could lead to expression of ETB which will trigger the release of ET1. Methods: Human Lung Microvascular Endothelial cells (HLMVEC) were cultured until 70-80% confluence. To mimic disease progression the GCX were degraded with Heparinase III which targets the HS component of the GCX. Immunostaining was used to determine the expression of GCX HS, ETB and ET-1. Results: Data revealed that the degradation of GCX HS resulted in the significant decrease in the expression of ETB. Conversely, the removal of GCX HS caused a significant increase in the expression of ET-1. Abstract:Purpose: Retinal microglia can be activated by retinal neuronal monocyte chemoattractant protein-1 (MCP-1) and play a pivotal role in early retinal degeneration. The current study investigates the pathways via which retinal neuronal MCP-1 stimulates tumor necrosis factor-alpha (TNF-alpha) expression in rat microglia. Methods: Primary rat retinal neurons and microglia were separated and cocultured in a Transwell apparatus. The levels of TNF-alpha mRNA and soluble TNF-a produced by the microglia in response to advanced glycation end product (AGE)-induced retinal neuronal MCP-1 were measured with real-time PCR and enzyme-linked immunosorbent assay (ELISA). The ability of neuronal MCP-1 to stimulate microglia activation was examined by preexposing the retinal neurons to AGEs and an MCP-1 antibody or by pretreating microglia with AGEs and siRNA specific for CC-chemokine receptor 2 (CCR2) knockdowns. Additionally, we investigated the effects of microglial activation on neuronal MCP-1-induced nuclear factor-kappa B (NF-kappa B) activation and phosphorylation of mitogen-activated protein kinases (MAPKs). Results: Stimulation with AGEs significantly increased the expression of TNF-alpha mRNA and soluble TNF-alpha in the microglial cells. Retinal neurons that had been pretreated with AGEs and an MCP-1 antibody or microglia that were CCR2 knockdowns displayed greatly reduced TNF-a secretion. Using signaling pathway-specific inhibitors, we showed that blocking the p38, extracellular signal-regulated kinase (ERK), and NF-kappa B signaling pathways significantly reduced the expression of TNF-alpha by retinal neuronal MCP-1-stimulated microglia. Conclusions: This study indicates that TNF-alpha was released from the activated microglia induced by retinal neuronal MCP-1 via the p38, ERK, and NF-kappa B pathways, but not c-Jun N-terminal kinase (JNK), which may be an important finding in diabetic retinopathy pathogenesis.

Abstract:The formation of retinal neovascularization (RNV) is the primary pathological process underlying retinopathy of prematurity (ROP). Previous studies have shown that inflammatory factors are related to the formation of RNV. Tumor necrosis factor-α (TNF-α), as an important factor in the inflammatory response, is involved in the regulation of RNV formation. However, the mechanism through which TNF-α inhibition reduces RNV formation is not fully clarified. Therefore, the purpose of this study was to explore the effect of etanercept, an inhibitor of TNF-α, on RNV, and its possible mechanism.

Abstract:Endothelin-1 (ET-1) stimulates vascular smooth muscle and mesangial cells to release prostaglandin E2 (PGE2), which can attenuate the vasoconstrictor and mitogenic effects of this peptide. Phospholipase A2 (PLA2)-mediated release of arachidonic acid from the sn-2 position of membrane phospholipids is thought to be one of the rate-limiting steps in prostaglandin (PG) synthesis. We evaluated the role of ET-1 to regulate gene expression, protein synthesis and enzymatic activity of cytosolic PLA2 (cPLA2), an intracellular form of the PLA2 enzyme family, in cultured rat mesangial cells using both acute and chronic incubation protocols. Acute ET-1-induced stimulation of cPLA2 activity was maximal after 10 minutes (181.1 +/- 6.84% of control), persisted for 40 minutes and did not require new protein synthesis. Heparin, a potent inhibitor of intracellular Ca2+ increase as well as mitogen-activated protein (MAP) kinase activation and cell proliferation, did not affect the rapid cPLA2 stimulation by ET-1. Chronic incubation of glomerular mesangial cells with ET-1 (1 to 24 hr) led to time- and dose-dependent increases in cPLA2 mRNA expression which was maximal after six hours, persisted up to 24 hours and which was accompanied by both cPLA2 protein formation, as assessed by Western analysis, as well as by stimulation of enzymatic activity. Inhibition of protein synthesis by cycloheximide increased basal cPLA2 mRNA accumulation in quiescent mesangial cells, and the combination of ET-1 and cycloheximide resulted in a greater induction of cPLA2 gene expression when compared to ET-1 alone. Actinomycin D treatment blocked the effect of ET-1 on cPLA2 mRNA accumulation.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract:Ocular diseases such as diabetic retinopathy (DR) and uveitis are associated with injury to the blood–retinal barrier (BRB). Whereas high glucose (HG) and advanced glycation end products (AGE) contribute to DR, bacterial infections causing uveitis are triggered by endotoxins such as lipopolysaccharide (LPS). It is unclear how HG, AGE, and LPS affect human retinal endothelial cell (HREC) junctions. Moreover, tumor necrosis factor-α (TNFα) is elevated in both DR and ocular infections. In the current study, we determined the direct effects of HG, AGE, TNFα, and LPS on the expression and intracellular distribution of claudin-5, VE-cadherin, and β-catenin in HRECs and how these mediators affect Akt and P38 MAP kinase that have been implicated in ocular pathologies. In our results, whereas HG, AGE, and TNFα activated both Akt and P38 MAPK, LPS treatment suppressed Akt but increased P38 MAPK phosphorylation. Furthermore, while treatment with AGE and HG increased cell-junction protein expression in HRECs, LPS elicited a paradoxical effect. By contrast, when HG treatment increased HREC-barrier resistance, AGE and LPS stimulation compromised it, and TNFα had no effect. Together, our results demonstrated the differential effects of the mediators of diabetes and infection on HREC-barrier modulation leading to BRB injury.

Abstract:In their article, Krueger and Eaton (pp. 97-118, this issue) advocate for a "more complete and extensive integration of personality traits in future DSMs [Diagnostic and Statistical Manuals of Mental Disorders], via the explicit inclusion of an empirically based, dimensional personality trait model" (p. 97) and review the "diverse ways in which the inclusion of an empirically based personality trait model could constitute a critical innovation in the transition from DSM-IV to DSM-5" (p. 113). Krueger and Eaton's proposal that (DSM-5) adopt a dimensional personality trait approach exemplifies the difficulties of devising a classification scheme that satisfies the needs of the both the clinical and research communities. In this instance, the challenge is adapting a complex dimensional trait scheme developed and utilized almost exclusively by the research community, for clinical use.
