Abstract:Ganglioside glycans are ubiquitous and complex biomolecules that are involved in a wide range of biological functions and disease processes. Variations in sialylation and sulfation render the structural complexity and diversity of ganglioside glycans, and influence protein–carbohydrate interactions. Structural and functional insights into the biological roles of these glycans are impeded due to the limited accessibility of well-defined structures. Here we report an integrated chemoenzymatic strategy for expeditious and systematic synthesis of a comprehensive 65-membered ganglioside glycan library covering all possible patterns of sulfation and sialylation. This strategy relies on the streamlined modular assembly of three common sialylated precursors by highly stereoselective iterative sialylation, modular site-specific sulfation through flexible orthogonal protecting-group manipulations and enzymatic-catalysed diversification using three sialyltransferase modules and a galactosidase module. These diverse ganglioside glycans enable exploration into their structure–function relationships using high-throughput glycan microarray technology, which reveals that different patterns of sulfation and sialylation on these glycans mediate their unique binding specificities.

Abstract:There has also been a growing interest in the application of carbohydrates as pharmaceutical drugs in the field of invasive bacterial diseases, inflammatory diseases, and viral infections. Chemical oligosaccharide synthetic procedures often require tedious protection and deprotection steps. Enzymatic synthesis offers tremendous benefits because of its high regio- and stereoselectivities, stability, organic solvent compatibility, and low cost. Glycosyltransferase-catalyzed synthesis of carbohydrates has been recognized as one of the most practical approaches for its high efficiency and specificity under very mild conditions. The two main obstacles in the enzymatic synthesis of carbohydrates are the inadequate availability of glycosyltransferases and the high cost of sugar nucleotides, which serve as intermediates. Significant progress in the study of the enzymatic biosynthesis of complex carbohydrates has been made with developments in protein purification, molecular genetics, and new methods of enzymological analysis. Bioinformatics provide a large amount of putative candidates for carbohydrate active enzymes. The combined enzymatic and genetic approach has overcome formidable obstacles ubiquitous to the study of glycans, and has begun to yield new information that definitively addresses basic enzymological issues relevant to these complex carbohydrates. In fact, complex carbohydrates representing the glycan chain of gangliosides such as GT2, GM1, and GD1a were synthesized using glycosyltransferases. Although traditional cell-free enzymatic synthesis of oligosaccharides will continue to be used in laboratory scale synthesis, the future direction for large-scale preparation of complex oligosaccharides and polysaccharides will be whole-cell systems. Further development of these enzymatic methods will allow synthetic biochemists to create important molecular tools for biochemical, biophysical, and medical applications.

Abstract:Recent progress in the development of efficient one-pot three-enzyme chemoenzymatic synthesis and application of functionalized sialosides is described. By taking the advantage of relaxed substrate specificity of several bacterial sialoside biosynthetic enzymes, the method has been used for the prepactive scale synthesis of a2,3- and α2,6-linked sialoside libraries containing naturally and non-naturally occurring sialic acid modifications. Starting from the hexose precursors (ManNAc or mannose) of sialic acids, a library of pNP-tagged sialyl disaccharides with various naturally occurring sialic acid forms, different sialyl linkages, and different penultimate monosaccharides have also been prepared and used in the substrate specificity studies of bacterial sialidases in a 96-well plate-based colorimetric high-throughput screening platform. The combination of efficient chemoenzymatic synthesis and high-throughput screening is a powerful approach to studying proteins that recognizing sialic acid-containing carbohydrates.

Abstract:Gangliosides play vital biological regulatory roles and are associated with neurological system diseases, malignancies, and immune deficiencies. They have received extensive attention in developing targeted drugs and diagnostic markers. However, it is difficult to obtain enough structurally defined gangliosides and analogs especially at an industrial-relevant scale, which prevent exploring structure-activity relationships and identifying drug ingredients. Here, we report a highly modular chemoenzymatic cascade assembly (MOCECA) strategy for customized and large-scale synthesis of ganglioside analogs with various glycan and ceramide epitopes. We typically accessed five gangliosides with therapeutic promising and systematically prepared ten GM1 analogs with diverse ceramides. Through further process amplification, we achieved industrial production of ganglioside GM1 in the form of modular assembly at hectogram scale. Using MOCECA-synthesized GM1 analogs, we found unique ceramide modifications on GM1 could enhance the ability to promote neurite outgrowth. By comparing the structures with synthetic analogs, we further resolved the problem of contradicting descriptions for GM1 components in different pharmaceutical documents by reinterpreting the exact two-component structures of commercialized GM1 drugs. Because of its applicability and stability, the MOCECA strategy can be extended to prepare other glycosphingolipid structures, which may pave the way for developing new glycolipid drugs.

Abstract:The modular enzymatic synthesis of a library of 24 ganglioside glycans is successfully achieved by using bacterial glycosyltransferases in a reprogrammed synthetic route. Glycan microarray analysis reveals that these synthetic ganglioside glycans, featuring various spatial arrangements of sialic acids, are specifically bound to sialic acid‐binding immunoglobulin‐type lectins (Siglec‐7 and Siglec‐9). In the present work, bacterial glycosyltransferases are utilized to construct ganglioside glycans in a convergent approach via a sugar‒nucleotide regeneration system and one‐pot multienzyme reactions. Starting from β‐lactoside enables the diversification of both the glycan moieties and the linkages in the lower α‐arm and upper β‐arm. Overall, a comprehensive panel of 24 natural a‐series (GM3, GM2, GM1a, GD1a, GT1a, and fucosyl‐GM1), b‐series (GD3, GD2, GD1b, GT1b, and GQ1b), c‐series (GT3, GT2, GT1c, GQ1c, and GP1c), α‐series (GM1α, GD1aα, and GT1aα), and o‐series (GA2, GA1, GM1b, GalNAc‐GM1b, and GD1c) ganglioside glycans are prepared, which are suitable for biological studies and further applications. Moreover, a microarray is constructed with these synthesized ganglioside glycans to investigate their binding specificity with recombinant Fc‐fused Siglec‐7 and Siglec‐9, which are immune checkpoint‐like glycan recognition proteins on natural killer cells. The microarray binding results reveal that GD3 and GT1aα are specific ligands for Siglec‐7 and Siglec‐9, respectively, and this discovery can lead to the identification of appropriate ligands for investigating the roles of these Siglecs in immunomodulation.

Abstract:Sulfated N-glycans are present in many glycoproteins, which are implicated in playing important roles in biological recognition processes. Here, we report the systematic chemoenzymatic synthesis of a library of sulfated and sialylated biantennary N-glycans and assess their binding to Siglecs and glycan-specific antibodies that recognize them as glycan ligands. The combined use of three human sulfotransferases, GlcNAc-6-O-sulfotransferase (CHST2), Gal-3-O-sulfotransferase (Gal3ST1), and keratan sulfate Gal-6-O-sulfotransferase (CHST1), resulted in asymmetric and symmetric branch-selective sulfation of the GlcNAc and/or Gal moieties of N-glycans. The extension of the sugar chain using α-2,3- and α-2,6-sialyltransferases afforded the sulfated and sialylated N-glycans. These synthetic glycans with different patterns of sulfation and sialylation were evaluated for binding to selected Siglecs and sulfoglycan-specific antibodies using glycan microarrays. The results confirm previously documented glycan-recognizing properties and further reveal novel specificities for these glycan-binding proteins, demonstrating the utility of the library for assessing the specificity of glycan-binding proteins recognizing sulfated and sialylated glycans.

Abstract:A systematic, efficient means of producing diverse libraries of asymmetrically branched N-glycans is needed to investigate the specificities and biology of glycan-binding proteins. To that end, we describe a core pentasaccharide that at potential branching positions is modified by orthogonal protecting groups to allow selective attachment of specific saccharide moieties by chemical glycosylation. The appendages were selected so that the antenna of the resulting deprotected compounds could be selectively extended by glycosyltransferases to give libraries of asymmetrical multi-antennary glycans. The power of the methodology was demonstrated by the preparation of a series of complex oligosaccharides that were printed as microarrays and screened for binding to lectins and influenza-virus hemagglutinins, which showed that recognition is modulated by presentation of minimal epitopes in the context of complex N-glycans.

Abstract:Despite the versatility of enzyme-mediated oligosaccharide assembly, it has as a limitation that not all glycosyl transferases or glycan-modifying enzymes are readily available to install all natural occurring terminal epitopes. Here a chemoenzymatic strategy is described in which a core oligosaccharide is assembled enzymatically that is subjected to chemical manipulations to install complex terminal epitopes. It provided an unprecedented panel of human natural killer-1 (HSO 3 –3GlcAβ1–3Galβ1–4GlcNAc)-containing oligosaccharides and derivatives thereof. The compounds were printed as a microarray to examine binding specificities of serum antibodies of patients suffering from anti-myelin-associated glycoprotein neuropathy. All samples required glucuronic acid for antibody binding; however, variable dependence was observed for the length of the LacNAc chain and sulfation of glucuronic acid. Most serum samples required a lacto-neohexaose backbone indicating glycosphingolipids are being targeted. The clinical spectrum of immunoglobulin M monoclonal gammopathy varies, and the glycan microarray provides a more reliable platform for disease diagnosis and prognosis.

Abstract:O-GalNAc glycans (or mucin O-glycans) play pivotal roles in diverse biological and pathological processes, including tumor growth and progression. Structurally defined O-GalNAc glycans are essential for functional studies but synthetic challenges and their inherent structural diversity and complexity have limited access to these compounds. Herein, we report an efficient and robust chemoenzymatic modular assembly (CEMA) strategy to construct structurally diverse O-GalNAc glycans. The key to this strategy is the convergent assembly of O-GalNAc cores 1–4 and 6 from three chemical building blocks, followed by enzymatic diversification of the cores by 13 well-tailored enzyme modules. A total of 83 O-GalNAc glycans presenting various natural glycan epitopes are obtained and used to generate a unique synthetic mucin O-glycan microarray. Binding specificities of glycan-binding proteins (GBPs) including plant lectins and selected anti-glycan antibodies towards these O-GalNAc glycans are revealed by this microarray, promoting their applicability in functional O-glycomics. Serum samples from colorectal cancer patients and healthy controls are assayed using the array reveal higher bindings towards less common cores 3, 4, and 6 than abundant cores 1 and 2, providing insights into O-GalNAc glycan structure-activity relationships.
The Structural Role of Gangliosides: Insights from X-ray Scattering on Model Membranes

Abstract:Background: Gangliosides are an essential component of eukaryotic plasma membranes implicated in multiple physiological processes. Little is known about molecular mechanisms underlying the distribution and functions of membrane gangliosides. The overwhelmingly complex organization of glycocalyx impedes the structural analysis on cell surface and the interplay between the lipid components. Advanced X-ray analytical tools applicable to studying biological interfaces call for the simplistic models that mimic ganglioside-enriched cellular membranes. Objective: To summarize the mechanistic evidences of ganglioside interactions with lipid environment and biologically active ligands using high-resolution synchrotron X-ray scattering. Methods: A comprehensive review of studies published over the last decade was done to discuss recent accomplishments and future trends. Results: Langmuir monolayers represent an adequate model system to assess the effect of gangliosides on membrane structure. Grazing incidence X-ray diffraction reveals a condensation effect by gangliosides on zwitterionic phospholipids with the cooperative packing of sialo- and phosphate groups. In turn, the arrangement of negatively charged lipids in ganglioside mixture remains unchanged due to the stretched conformation of carbohydrate moieties. Upon interaction with biological ligands, such as cholera toxin and galectins, the ganglioside redistribution within the ordered regions of monolayer follows distinct mechanistic patterns. The cholera toxin pentamer attached to the oligosaccharide core induces local transition from oblique to the hexagonal lattice resulting in phase coexistence. The incorporation of the A subunit responsible for endocytosis is further promoted by the acidic environment characteristic for endosomal space. X-ray reflectivity shows in-plane orientation of galectin dimers with the spatial mismatch between the lectin binding sites and ganglioside carbohydrates to perturb ceramide alkyl chains. Recent data also demonstrate sialic acid groups to be potential targets for novel peptide mimicking anticancer therapeutics. Conclusion: Coupled with surface X-ray scattering, the membrane mimetic approach allows for better understanding the biological role of gangliosides and their potential applications.

Abstract:Abstract Cellular glycosylation is characterized by chemical complexity and heterogeneity, which is challenging to reproduce synthetically. Here we show chemoenzymatic synthesis on phage to produce a genetically-encoded liquid glycan array (LiGA) of complex type N -glycans. Implementing the approach involved by ligating an azide-containing sialylglycosyl-asparagine to phage functionalized with 50–1000 copies of dibenzocyclooctyne. The resulting intermediate can be trimmed by glycosidases and extended by glycosyltransferases yielding a phage library with different N -glycans. Post-reaction analysis by MALDI-TOF MS allows rigorous characterization of N -glycan structure and mean density, which are both encoded in the phage DNA. Use of this LiGA with fifteen glycan-binding proteins, including CD22 or DC-SIGN on cells, reveals optimal structure/density combinations for recognition. Injection of the LiGA into mice identifies glycoconjugates with structures and avidity necessary for enrichment in specific organs. This work provides a quantitative evaluation of the interaction of complex N -glycans with GBPs in vitro and in vivo.

Abstract:Heparan sulfate (HS) is a member of the glycosaminoglycans (GAG) family that plays essential roles in biological processes from animal sources. Heparin, a highly sulfated form of HS, is widely used as anticoagulant drug worldwide. The high diversity and complexity of HS and heparin represent a roadblock for structural characterization and biological activity studies. Access to structurally defined oligosaccharides is critical for the successful development of HS and heparin structure-activity relationships. In this study, a library of 66 HS and heparin oligosaccharides covering different sulfation patterns and sizes was prepared through an efficient method of chemoenzymatic synthesis. A systematic nuclear magnetic resonance spectroscopy study was firstly undertaken for every oligosaccharide in the library. In addition to the availability of different oligosaccharides, this work also provides spectroscopic data helpful for characterizing more complicated polysaccharide structures providing a safeguard to ensure the quality of the drug heparin. This HS/heparin library will be useful for activity screening and facilitate future structure-activity relationship studies.

Abstract:A novel. chemoenzymatic approach for the synthesis of oligosacchrides containing free amino groups has been developed in which thermophilic glycosidases and a fusion enzyme containing catalytic domains of uridine-5'-diphospho-galactose 4-epimerase and alpha(1-->3) galactosyltransferase were used. This methodology, in conjunction with a convenient purification procedure employing ion exchange chromatography, facilitates the lengthy and high-cost process of carbohydrate synthesis. The prepared oligosaccharides range from disaccharide lactosamine (1a), trisaccharide alpha-Gal-(1-->3)-beta-Gal-(1-->4)-GlcNH(2) (2), tetrasaccharide beta-Gal-(1-->4)-beta-GlcNH(2)-(1-->3)-beta-Gal-(1-->4)-beta-Glc-N-3 (7), to pentasaccharide alpha-Gal-(1-->3)-beta-Gal-(1-->4)-beta-GlcNH(2)-(1-->3)-beta-Gal-(1-->4)-beta-Glc-N-3 (15). Compounds 2 and 15 are derivatives of natural alpha-Gal epitopes. Both of them have shown comparable activities with their natural parent compounds toward human anti-Gal IgG. This method provides a practical approach for the preparation and purification of oligosaccharides containing free amino groups, which can be further derivatized for the enhancement of biological activities.

Abstract:Structural glycobiology has traditionally been a challenging field due to a limited set of tools available to investigate the diverse and complexity of Glycan molecules. However, we cannot ignore that glycans play critical roles in health as well as in disease, are present in more than 50% of all proteins and on over 80% of all surface proteins. Chemoenzymatic Glycoengineering (CEG) methods are a powerful set of tools to synthesize complex glycans but its full potential has not been explored in cell biology yet. Herein, we report the labeling of live CHO cells by employing three highly specific glycosyltransferases: a sialyltransferase, a galactosyltransferase, and an N-acetyl-glucosaminyl transferase. We verified our results by bio-orthogonal blots and further rationalized them by computational modeling. We expect CEG applications in cell biology to rise and their implementation will assist in structural-functional discoveries in glycobiology. This research will contribute to this effort.

Abstract:Over the past decade, considerable progress has been made with respect to the analytical methods for analysis of glycans from biological sources. Regardless of the specific methods that are used, glycan analysis includes isolation, identification, and quantitation. Derivatization is indispensable to increase their identification. Derivatization of glycans can be performed by permethylation or carbodiimide coupling/esterification. By introducing a fluorophore or chromophore at their reducing end, glycans can be separated by electrophoresis or chromatography. The fluorogenically labeled glycans can be quantitated using fluorescent detection. The recently developed approaches using solid-phase such as glycoprotein immobilization for glycan extraction and on-tissue glycan mass spectrometry imaging demonstrate advantages over methods performed in solution. Derivatization of sialic acids is favorably implemented on the solid support using carbodiimide coupling, and the released glycans can be further modified at the reducing end or permethylated for quantitative analysis. In this review, methods for glycan isolation, identification, and quantitation are discussed.

Abstract:Gangliosides, a diverse class of glycosphingolipids, are highly abundant in neural tissue and have been implicated in numerous aging-related diseases. Their characterization with methods such as liquid chromatography-tandem mass spectrometry is often precluded by their structural complexity, isomeric heterogeneity, and lack of commercially available authentic standards. In this work, we coupled high-resolution cyclic ion mobility spectrometry with multiple collision-induced dissociation-based tandem mass spectrometry strategies to sequence the sialic acid positions in various ganglioside isomers. Initially, as a proof-of-concept demonstration, we were able to characterize the sialic acid positions in several GD1 and GT1 species. From there, we extended our approach to identify the location of N-glycolylneuraminic acid (NeuGc) residues in previously uncharacterized GD1 and GQ1 isomers. Our results highlight the potential of this presented methodology for the de novo characterization of gangliosides within complex biological matrices without the need for authentic standards.
