Abstract:In contrast to higher animals, plants maintain the capacity of generating new organs throughout their lifespan. Stem cells in shoot meristem provide the source of all above-ground organs. In Arabidopsis, shoot stem cell homeostasis is maintained by homeodomain (HD) transcription factors in two main regulatory pathways. The WUSCHEL (WUS) gene is transcribed in the organizing center (OC) below the stem cells. WUS protein moves into the stem cells where it directly activates CLAVATA3 (CLV3) expression and thus specifies the stem cell identity. SHOOT MERISTEMLESS (STM) is expressed throughout the shoot meristem and represses stem cell differentiation (Aichinger et al., 2012Aichinger E. Kornet N. Friedrich T. Laux T. Whether and how these two pathways interact to regulate shoot stem cell homeostasis were not clear until now. A recent study by Su et al., 2020Su Y. Zhou C. Li Y. Yu Y. Tang L. Zhang W. Yao W. U S A. 2020; 117: 22561-22571Crossref PubMed Scopus (10) Google Scholar revealed that the WUS and STM pathways interact, and their interactions play an essential role in inducing CLV3 expression and maintaining stem cell homeostasis. Direct interactions between STM and WUS were demonstrated by both in vitro and in vivo studies. Interestingly, the co-expression of WUS and STM displayed an additive effect on CLV3 induction, and downregulation of WUS expression in stm knockdown mutants enhanced the shoot meristem defect of these mutants. A set of elegant experiments were then conducted to test how the physical protein-protein interaction and genetic interaction between WUS and STM play roles in regulating CLV3 expression and shoot meristem development. The results suggest that STM serves as a direct activator of CLV3 by binding the CLV3 promoter at a different site from that of WUS. Both binding sites of STM and WUS are required for CLV3 expression and CLV3-mediated stem cell control. Moreover, the CLV3 promoter region containing the WUS binding site was enriched when the STM-VENUS fusion protein was immunoprecipitated, suggesting that a STM-WUS heterodimer may regulate CLV3 expression at the WUS binding site. Indeed, both in vitro and in vivo experiments showed that STM-WUS interaction could enhance WUS binding to the CLV3 promoter and activation of CLV3 transcription, strongly suggesting that CLV3 expression is simultaneously regulated by STM, WUS, and their complex (Figure 1B). Finally, the authors showed that WUS activates STM expression in shoot meristem, consistent with the previous finding that the function of STM in CLV3 induction relies on WUS activity (Lenhard et al., 2002Lenhard M. It is intriguing to know that the spatial expression and the interplay among the three important players regulate stem cell activity. Further investigations are needed to understand how the STM-WUS complex activates CLV3 expression at the STM binding site and why there is little CLV3 induction in the OC where both WUS and STM expression domains overlap.
