Abstract:Poly(ethylene glycol) (PEG) segments were introduced into a polyanhydride main chain by copolymerization of terminal-carboxylated poly(ethylene glycol) with diacidic monomers (sebacic acid and trimellitylimidoglycine). IR and 1H NMR spectroscopy confirmed the copolymer structures. DSC analysis showed that these polyanhydrides have low Tg and low crystallinity. In vitro degradation tests indicated that introducing PEG segments accelerated the degradation rate of these polymers and the degradation duration could be manipulated from 3 days to 3 weeks. The pH of the environment caused by the polymer degradation was lower than 5.0; therefore, the polyanhydrides could be used as components of a newly designed pulsed-release device for peptide and protein delivery. © 1999 Society of Chemical Industry

Abstract:Poly(glycerol sebacate) (PGS), a biodegradable elastomer, has been extensively explored in biomedical applications for its favorable mechanical properties and biocompatibility. Efforts have been made to fabricate multifunctional PGS copolymer in recent years, in particular PGS-co-PEG (poly(glycerol sebacate)-co-polyethylene glycol) polymers. However, rare research has been systematically conducted on the effect of reactant ratios on physicochemical properties and biocompatibility of PGS copolymer till now. In this study, a serial of PEGylated PGS (PEGS) with PEG content from 20% to 40% and carboxyl to hydroxyl from 0.67 to 2 were synthesized by thermal curing process. The effects of various PEGS on the mechanical strength and biological activity were further compared and optimized. The results showed that the PEGS elastomers around 20PEGS-1.0C/H and 40PEGS-1.5C/H exhibited the desirable hydrophilicity, degradation behaviors, mechanical properties and cell viability. Subsequently, the potential applications of the 20PEGS-1.0C/H and 40PEGS-1.5C/H in bone repair scaffold and vascular reconstruction were investigated and the results showed that 20PEGS-1.0C/H and 40PEGS-1.5C/H could significantly improve the mechanical strength for the calcium phosphate scaffolds and exhibited preferable molding capability for fabrication of the vascular substitute. These results confirmed that the optimized PEGS elastomers should be promising multifunctional substrates in biomedical applications.

Abstract:A biodegradable poly(sebacic-co-propanetriol) ester elastomer(PGS) was synthesized with propanetriol and sebacic acid by melting cocondensation.The structure of the PGS was characterized.The mechanical properties,hydrophilicity and degradation of the PGS were discussed.The results showed that the ester bond,carboxyl and hydroxyl groups in the PGS endowed with certain hydrophilicity and biodegradation of PGS. The mechanical properties,hydrophilicity and degradation of the PGS could be adjusted by altering mole ratio of propanetriol and sebacic acid.

Abstract:We have developed a family of synthetic biodegradable polymers that are composed of structural units endogenous to the human metabolism, designated poly(polyol sebacate) (PPS) polymers. Material properties of PPS polymers can be tuned by altering the polyol monomer and reacting stiochiometric ratio of sebacic acid. These thermoset networks exhibited tensile Young's moduli ranging from 0.37+/-0.08 to 378+/-33 MPa with maximum elongations at break from 10.90+/-1.37% to 205.16+/-55.76%, and glass transition temperatures ranging from approximately 7-46 degrees C. In vitro degradation under physiological conditions was slower than in vivo degradation rates observed for some PPS polymers. PPS polymers demonstrated similar in vitro and in vivo biocompatibility compared to poly(L-lactic-co-glycolic acid) (PLGA).

Abstract:Development of biodegradable shape memory elastomers (SMEs) is driven by the growing need for materials to address soft tissue pathology using a minimally invasive surgical approach. Composition, chain length and crosslinking of biocompatible polymers like PCL and PLA have been investigated to control mechanical properties, shape recovery and degradation rates. Depending on the primary mechanism of degradation, many of these polymers become considerably stiffer or softer resulting in mechanical properties that are inappropriate to support the regeneration of surrounding soft tissues. Additionally, concerns regarding degradation byproducts or residual organic solvents during synthesis accelerated interest in development of materials from bioavailable monomers. We previously developed a biodegradable SME, poly(glycerol dodecanoate) (PGD), using biologically relevant metabolites and controlled synthesis conditions to tune mechanical properties for soft tissue repair. In this study, we investigate the influence of crosslinking density on the mechanical and thermal properties of PGD during in vitro and in vivo degradation. Results suggest polymer degradation in vivo is predominantly driven by surface erosion, with no significant effects of initial crosslinking density on degradation time under the conditions investigated. Importantly, mechanical integrity is maintained during degradation. Additionally, shifts in melt transitions on thermograms indicate a potential shift in shape memory transition temperatures as the polymers degrade. These findings support the use of PGD for soft tissue repair and warrant further investigation towards tuning the molecular and macromolecular properties of the polymer to tailor degradation rates for specific clinical applications.

Abstract:Poly(glycerol sebacate) (PGS), a crosslinked network synthesized by polycondensation of glycerol and sebacic acid, has found successful utilization in various tissue engineering applications because of its biocompatibility, ease of synthesis, surface-erosion biodegradability, rubber-like elasticity, and adjustable mechanical properties. However, its fast in vivo degradation and lack of intrinsic antibacterial activity may be a concern for specific applications. The functional groups of glycerol and sebacic acid can react with those of other monomers, offering unlimited possibilities for generating new crosslinked networks tailored for particular purposes. Herein, we synthesized a series of poly(glycerol sebacate dithiodiglycolate) (PGSDTG) by incorporation of dithiodiglycolic acid (DTG) in the synthesis of PGS. PGSDTG exhibited the reduced stiffness, sensitive to glutathione (GSH), resistance to lipase degradation, and inhibition of the adhesion and growth of bacteria. The PGSDTG containing 45.6 wt% of DTG exhibited the lowest degradation rate with more than 80 % of PGSDTG weight remained at day 28 and the lowest bacterial inactivation rates of 99.2 ± 0.5 % and 82.5 ± 11.0 % against S. aureus and E. coli . This study suggested that PGSDTG may serve as a suitable antibacterial scaffold material for repairing or regenerating tissues characterized by a slow healing rate.

Abstract:Poly(glycerol sebacate) (PGS) is a biodegradable elastomer that has attracted increasing attention as a potential material for applications in biological tissue engineering. The conventional method of synthesis, first described in 2002, is based on the polycondensation of glycerol and sebacic acid, but it is a time-consuming and energy-intensive process. In recent years, new approaches for producing PGS, PGS blends, and PGS copolymers have been reported to not only reduce the time and energy required to obtain the final material but also to adjust the properties and processability of the PGS-based materials based on the desired applications. This review compiles more than 20 years of PGS synthesis reports, reported inconsistencies, and proposed alternatives to more rapidly produce PGS polymer structures or PGS derivatives with tailor-made properties. Synthesis conditions such as temperature, reaction time, reagent ratio, atmosphere, catalysts, microwave-assisted synthesis, and PGS modifications (urethane and acrylate groups, blends, and copolymers) were revisited to present and discuss the diverse alternatives to produce and adapt PGS.

Abstract:Poly(glycerol sebacate) (PGS), a soft, tough elastomer with excellent biocompatibility, has been exploited successfully in many tissue engineering applications. Although tunable to some extent, the rapid in vivo degradation kinetics of PGS is not compatible with the healing rate of some tissues. The incorporation of L-glutamic acid into a PGS network with an aim to retard the degradation rate of PGS through the formation of peptide bonds was conducted in this study. A series of poly(glycerol sebacate glutamate) (PGSE) containing various molar ratios of sebacic acid/L-glutamic acid were synthesized. Two kinds of amino-protected glutamic acids, Boc-L-glutamic acid and Z-L-glutamic acid were used to prepare controls that consist of no peptide bonds, denoted as PGSE-B and PGSE-Z, respectively. The prepolymers were characterized using 1H-NMR spectroscopy. Cured elastomers were characterized using FT-IR, DSC, TGA, mechanical testing, and contact angle measurement. In vitro enzymatic degradation of PGSE over a period of 28 days was investigated. FT-IR spectroscopy confirmed the formation of peptide bonds. The glass transition temperature for the elastomer was found to increase as the ratio of sebacic acid/glutamic acid was increased to four. The decomposition temperature of the elastomer decreased as the amount of glutamic acid was increased. PGSE exhibited less stiffness and larger elongation at break as the ratio of sebacic acid/glutamic acid was decreased. Notably, PGSE-Z was stiffer and had smaller elongation at break than PGSE and PGSE-B at the same molar ratio of monomers. The results of in vitro enzymatic degradation demonstrated that PGSE has a lower degradation rate than does PGS, whereas PGSE-B and PGSE-Z degrade at a greater rate than does PGS. SEM images suggest that the degradation of these crosslinked elastomers is due to surface erosion. The cytocompatibility of PGSE was considered acceptable although slightly lower than that of PGS. The altered mechanical properties and retarded degradation kinetics for PGSE reflect the influence of peptide bonds formed by the introduction of L-glutamic acid. PGSE displaying a lower degradation rate compared to that for PGS can be used as a scaffold material for the repair or regeneration of tissues that are featured by a low healing rate.

Abstract:Poly(glycerol sebacate) (PGS) belongs to the hyperbranched polyesters family (HBP), which possesses an extensive variety of applications due to its tunable chemical and mechanical properties, together with its biocompatibility and biodegradability. However, the understanding of PGS synthesis becomes a challenge due to the lack of consistency when determining its synthesis parameters. Understanding these parameters is fundamental to control PGS synthesis and obtain a scalable and reproducible final product for biomedical applications. To unveil their effect on diverging PGS properties, diols are used as glycerol analogs and the reaction is chemically and thermally monitored, suggesting a heterogeneous reactivity of the exposed hydroxyl groups. Also, confinement of the prepolymerization is proven to be fundamental In order to maintain the equimolar ratio of initial monomers during synthesis. Early stages of the polycondensation (first 4 h) tend to linear and less branched oligomers by consuming primary hydroxyls rather than secondary hydroxyls. However, physicochemical characterization determines that a high degree of conversion (40%) is reached during these early stages. Afterward, the oligomers tend to condense through the secondary hydroxyls into a more crosslinked elastomer. This study demonstrates how hydroxyl affinity, water presence, and glycerol loss are crucial for the scalability and reproducibility of its final product.

Abstract:A kind of biodegradable poly(sebacic-co-propanetriol) ester elastomer(PGS) was synthesized by condensing sebacic acid and propanetriol with different mole ratio and its composition and crystallization property were characterized by swelling test, X-ray diffraction and differential scanning calorimetry.The results showed that PGS was a polyester with low crosslinking density, and composed of sol and gel with sol content more than 75%.The glass transition temperature of PGS was below -25 ℃.PGS was a crystallizable elastomer and had microphase separation structure with crystal regions and amorphous regions with increasing the sebacic acid.

Abstract:The present study synthesized a poly (glycerol–sebacate–lactic acid) (PGSL) with 1:1:0.5 mole ratio of glycerol, sebacate and lactic acid and investigated the degradation characteristics of the polymer in phosphate buffered saline (PBS) at 37°C in vitro by means of mass loss tests, geometry, differential scanning calorimeter (DSC) measurements, tensile analysis and scanning electron microscopy (SEM). The maintained geometry, linear mass loss, and minor crack formation on the surface during degradation characterized both the bulk degradation and surface erosion of the polymer. By day 30 of degradation, the mass lost reached 16%. The elastic modulus, tensile strength and elongation at breakage of PGSL were correlative to the period of degradation.
Biodegradable xylitol-based elastomers: in vivo behavior and biocompatibility

Abstract:Biodegradable elastomers based on polycondensation reactions of xylitol with sebacic acid, referred to as poly(xylitol sebacate) (PXS) elastomers have recently been developed. We describe the in vivo behavior of PXS elastomers. Four PXS elastomers were synthesized, characterized, and compared with poly(L-lactic-co-glycolic acid) (PLGA). PXS elastomers displayed a high level of structural integrity and form stability during degradation. The in vivo half-life ranged from approximately 3 to 52 weeks. PXS elastomers exhibited increased biocompatibility compared with PLGA implants.

Abstract:Poly (glycerol sebacate) (PGS) elastomer scaffolds with different porosity for skin tissue engineering were fabricated via particulate leaching. The introduction of pores lowers the hydrophilicity but improves the water uptake capability of PGS. The gel content of PGS increases with the increase of salt mass ratio, but the degree of swelling goes the opposite way due to the existence of the porous structure. The degradation rate of PGS can be tailored and controlled by the porous structure, which is of great value for its applications in tissue engineering. The feasibility of these porous PGS scaffolds for skin tissue engineering was evaluated by seeding mouse dermal fibroblasts (MDFs) onto the scaffold. In vitro cell culture results indicate good attachment, proliferation and deep penetration of MDFs into porous PGS scaffolds, which confirms the excellent biocompatibility of these scaffolds. A Poly(glycerol Sebacate) Based Photo/thermo Dual Curable Biodegradable and Biocompatible Polymer for Biomedical Applications

Abstract:Due to its biomimetic mechanical properties to soft tissues, excellent biocompatibility and biodegradability, poly (glycerol sebacate) (PGS) has emerged as a representative bioelastomer and been widely used in biomedical engineering. However, the typical curing of PGS needs high temperature (>120 degrees C), high vacuum (>1Torr), and long duration (>12h), which limit its further applications. Accordingly, we designed, synthesized and characterized a photo/thermo dual curable polymer based on PGS. Treatment of PGS with 2-isocyanatoethyl methacrylate without additional reagents readily produced a methacrylated PGS (PGS-IM). Photo-curing of PGS-IM for 10min at room temperature using salt leaching method efficiently produced porous scaffolds with a thickness up to 1mm. PGS-IM was adapt to thermo-curing as well. The combination of photo and thermo curing provided a further way to modulate the properties of resultant porous scaffolds. Interestingly, photo-cured scaffolds exhibited hierarchical porous structures carrying extensive micropores with a diameter from several to hundreds micrometers. All the scaffolds showed good elasticity and biodegradability. In addition, PGS-IM exhibited good compatibility with L929 fibroblast cells. We expect this new PGS based biomaterial will have a wide range of biomedical applications.

Abstract:Poly(glycerol sebacate) (PGS), a synthetic biorubber, is characterised by its biocompatibility, high elasticity and tunable mechanical properties; however, its inherent hydrophobicity and insolubility in water make it unsuitable for use in advanced biomaterials like hydrogels fabrication. Here, we developed new hydrophilic PGS-based copolymers that enable hydrogel formation through use of two different types of polyethylene glycol (PEG), polyethylene glycol (PEG2) or glycerol ethoxylated (PEG3), combined at different ratios. A two-step polycondensation reaction was used to produce poly(glycerol sebacate)-co-polyethylene glycol (PGS-co-PEG) copolymers that were then crosslinked thermally without the use of initiators or crosslinkers, resulting in PGS-co-PEG2 and PGS-co-PEG3 amphiphilic polymers. It has been illustrated that the properties of PGS-co-PEG copolymers can be controlled by altering the type and amount of PEG. PGS-co-PEG copolymers containing PEG≥40% showed high swelling, flexibility, stretching, bioadhesion and biocompatibility, and good biodegradation and mechanical properties. Also, the addition of PEG created hydrogels that demonstrated pH-responsive behaviours, which can be used for bioapplications requiring responding to physicochemical dynamics. Interestingly, PGS-co-40PEG2 and PGS-co-60PEG3 had the highest shear strengths, 340.4 ± 49.7 kPa and 336.0 ± 35.1 kPa, and these are within the range of commercially available sealants or bioglues. Due to the versatile multifunctionalities of these new copolymer hydrogels, they can have great potential in soft tissue engineering and biomedicine.

Abstract:Biodegradable elastomers represent a useful class of biomaterials. In this paper, a novel biodegradable elastomer, poly(PEG-co-CA) (PEC), was synthesized by condensation of poly(ethylene glycol) (PEG) and citric acid (CA) under atmospheric pressure without any catalyst. We first synthesized a pre-polymer by carrying out a controlled condensation reaction between PEG and citric acid, and then post-polymerised and simultaneously cross-linked the pre-polymer in the mould at 120°C. The pre-polymer was characterized by FT-IR, 1H NMR, 13C NMR, GPC and DSC. A series of polymers were prepared at different post-polymerisation time and different monomer ratios. Measurements on the mechanical properties of PEC testified that the new polymers are elastomers with low hardness and big elongation, and hydrolytic degradation of the polymer films in a buffer of pH 7.4 at 37°C showed that PEC had excellent degradability (all the films show the weight losses more than 60% after 96h incubation). The different post-polymerisation time and monomer ratio had strong influence on the degradation rates and mechanical performances. The material is expected to be useful for controlled drug delivery and other biomedical applications.
In Vitro and in Vivo Degradation Study on Novel Blends Composed of Polyphosphazene and Polyester or Polyanhydride

Abstract:Poly[bis(glycine ethyl ester)phosphazene] (PGP) was blended with poly(D,L-lactide-co-glycolide) (80:20 by mole) (PLGA) and poly[sebacic anhydride-co-trimellitylimidoglycine)-block-poly(ethylene glycol)] (30:50:20 by mole) (PSTP) in various ratios using a solvent-mixing technique. The in vitro degradation of polymer blends was examined by treatment in distilled water with or without lipase at 37degreesC; the degradation rate of the blends could be regulated by adjusting the composition. PGP/PLGA (70:30 by wt) slabs took 120 days to disappear completely, while PGP/PSTP (70:30 by wt) needed only 20 days. Furthermore, Rhizopus delemer lipase showed the strongest acceleration activity on PGP/PLGA blends. The degree of enzymatic degradation was proportional to the percentage of PLGA in the blend. For comparison, the in vivo degradation of polymer blends was investigated by implanting them subcutaneously in the back of mice, some difference in degradation being observed. Abstract:We studied the condensing reaction of sebacic acid and glycerol and prepared biodegradable elastomers. Swelling experiments proved that the elastomers were crosslinked polyesters consisting of both insoluble parts (gel) and soluble parts (sol), but the content of sol was higher than get. X-ray diffraction analysis showed that some ordered and crystallized structures existed in most of the elastomers. Differential scanning calorimetry measurement showed that there were both crystal regions and amorphous regions with low glass-transition temperatures in the products, which indicated the elastomers had a microphase separation structure. The elastomers exhibited thermal processing abilities, such as mold-shaping performance, and a certain elasticity, and hydroxyl, carboxyl, and ester groups in the molecular chains endowed the elastomers with good biodegradation abilities. Furthermore, by altering the molar ratio of the reactants, we were able to adjust the mechanical properties, biodegradable performance, and so on of the elastomers. Glycerol and polymers containing sebacic acid have been approved for biological medical uses by the U.S. Food and Drug Administration, so the elastomers we prepared would have broad application in medical fields such as implants and drug-delivery systems. Effects of the species of crosslinking reagents on the structures and properties of biodegradable poly (butanediol sebacate ‐ butanediol terephthalate) copolyester

Abstract:In the present study, poly(butylene sebacate‐co‐terephthalate)s having different crosslinking reagents were synthesized with a random distribution by a two‐step esterification and one‐step polycondensation process. In detail, the using crosslinking agents were included trimethylolethane (TME), trimethylolpropane, tris(hydroxymethyl)aminoethane, glycerol (GL). Particularly, CS refers to a sample with no crosslinking agent added. The copolyester with TME was the least crystalline sample, and the melting peaks of all copolyesters corresponding to both, sebacate and terephthalate‐rich phases were still observable in second calorimetric heating runs. These copolyesters were associated with interesting thermal and mechanical properties. The melting points of all samples were higher than 118°C and the puncture resistance and tensile strength of GL, the tear strength of TME, and the Young's modulus of all samples have been improved. Enzymatic degradability was assessed and the effect of composition and crystallinity on the degradation rate was investigated. The copolyester adding GL appears as a highly promising biodegradable material since it showed a significant weight loss during exposure to all selected degradation media, but also exhibited good performance and properties that were comparable to those characteristic of polyethylene. In brief, the improvement of PBSeT's puncture resistance, tear strength, and degradation performance is extremely important for the development and promotion of degradable foam, film, and elastomer materials.
