Abstract:Ace -AMP1 is a potent antifungal peptide found in onion ( Allium cepa ) seeds with sequence similarity to plant lipid transfer proteins. Transgenic plants over-expressing Ace-AMP1 gene have enhanced disease resistance to some fungal pathogens. However, mass production in heterologous systems and in vitro application of this peptide have not been reported. In this study, Ace -AMP1 was highly expressed in a prokaryotic Escherichia coli system as a fusion protein. The purified protein inhibited the growth of many plant fungal pathogens, especially Alternaria solani , Fusarium oxysporum f. sp. vasinfectum , and Verticillium dahliae . The inhibitory effect was accompanied by hyphal hyperbranching for V. dahliae but not for F. oxysporum f. sp. vasinfectum and A. solani , suggesting that the mode of action of Ace -AMP1 on different fungi might be different. Application of Ace -AMP1 on tomato leaves showed that the recombinant protein conferred strong resistance to the tomato pathogen A. solani and could be used as an effective fungicide.

Abstract:Canola (Brassica napus), an agriculturally important oilseed crop, can be significantly affected by diseases such as sclerotinia stem rot, blackleg, and alternaria black spot resulting in significant loss of crop productivity and quality. Cysteine-rich antimicrobial peptides isolated from plants have emerged as a potential resource for protection of plants against phytopathogens. Here we report the significance of an antimicrobial peptide, PmAMP1, isolated from western white pine (Pinus monticola), in providing canola with resistance against multiple phytopathogenic fungi. The cDNA encoding PmAMP1 was successfully incorporated into the genome of B. napus, and it’s in planta expression conferred greater protection against Alternaria brassicae, Leptosphaeria maculans and Sclerotinia sclerotiorum. In vitro experiments with proteins extracted from transgenic canola expressing Pm-AMP1 demonstrated its inhibitory activity by reducing growth of fungal hyphae. In addition, the in vitro synthesized peptide also inhibited the growth of the fungi. These results demonstrate that generating transgenic crops expressing PmAMP1 may be an effective and versatile method to protect susceptible crops against multiple phytopathogens.

Signaling pathways coordinating the alkaline pH response confer resistance to the hevein-type plant antimicrobial peptide Pn-AMP1 in Saccharomyces cerevisiae

Abstract:Genome-wide screening of Saccharomyces cerevisiae revealed that signaling pathways related to the alkaline pH stress contribute to resistance to plant antimicrobial peptide, Pn-AMP1. Plant antimicrobial peptides (AMPs) are considered to be promising candidates for controlling phytopathogens. Pn-AMP1 is a hevein-type plant AMP that shows potent and broad-spectrum antifungal activity. Genome-wide chemogenomic screening was performed using heterozygous and homozygous diploid deletion pools of Saccharomyces cerevisiae as a chemogenetic model system to identify genes whose deletion conferred enhanced sensitivity to Pn-AMP1. This assay identified 44 deletion strains with fitness defects in the presence of Pn-AMP1. Strong fitness defects were observed in strains with deletions of genes encoding components of several pathways and complex known to participate in the adaptive response to alkaline pH stress, including the cell wall integrity (CWI), calcineurin/Crz1, Rim101, SNF1 pathways and endosomal sorting complex required for transport (ESCRT complex). Gene ontology (GO) enrichment analysis of these genes revealed that the most highly overrepresented GO term was "cellular response to alkaline pH". We found that 32 of the 44 deletion strains tested (72 %) showed significant growth defects compared with their wild type at alkaline pH. Furthermore, 9 deletion strains (20 %) exhibited enhanced sensitivity to Pn-AMP1 at ambient pH compared to acidic pH. Although several hundred plant AMPs have been reported, their modes of action remain largely uncharacterized. This study demonstrates that the signaling pathways that coordinate the adaptive response to alkaline pH also confer resistance to a hevein-type plant AMP in S. cerevisiae. Our findings have broad implications for the design of novel and potent antifungal agents.
Enhanced tolerance to Phytophthora root and stem rot by over-expression of the plant antimicrobial peptide CaAMP1 gene in soybean

Abstract:Abstract Background Antimicrobial peptides play important roles in both plant and animal defense systems. Moreover, over-expression of CaAMP1 ( Capsicum annuum antimicrobial protein 1 ), an antimicrobial protein gene isolated from C. annuum leaves infected with Xanthomonas campestris pv. vesicatoria , confers broad-spectrum resistance to hemibiotrophic bacterial and necrotrophic fungal pathogens in Arabidopsis . Phytophthora root and stem rot (PRR), caused by the fungus Phytophthora sojae , is one of the most devastating diseases affecting soybean ( Glycine max ) production worldwide. Results In this study, CaAMP1 was transformed into soybean by Agrobacterium -mediated genetic transformation. Integration of the foreign gene in the genome of transgenic soybean plants and its expression at the translation level were verified by Southern and western blot analyses, respectively. CaAMP1 over-expression ( CaAMP1- OX) lines inoculated with P. sojae race 1 exhibited enhanced and stable PRR tolerance through T 2 –T 4 generations compared with the wild-type Williams 82 plants. Gene expression analyses in the transgenic plants revealed that the expression of salicylic acid-dependent, jasmonic acid-dependent, and plant disease resistance genes ( R -genes) were significantly up-regulated after P. sojae inoculation. Conclusions These results indicate that CaAMP1 over-expression can significantly enhance PRR tolerance in soybean by eliciting resistance responses mediated by multiple defense signaling pathways. This provides an alternative approach for developing soybean varieties with improved tolerance against soil-borne pathogenic PRR.

Abstract:Polyamines play an important role in plant response to abiotic stress. S-adenosyl-l-methionine decarboxylase (SAMDC) is one of the key regulatory enzymes in the biosynthesis of polyamines. In order to better understand the effect of regulation of polyamine biosynthesis on the tolerance of high-temperature stress in tomato, SAMDC cDNA isolated from Saccharomyces cerevisiae was introduced into tomato genome by means of Agrobacterium tumefaciens through leaf disc transformation. Transgene and expression was confirmed by Southern and Northern blot analyses, respectively. Transgenic plants expressing yeast SAMDC produced 1.7- to 2.4-fold higher levels of spermidine and spermine than wild-type plants under high temperature stress, and enhanced antioxidant enzyme activity and the protection of membrane lipid peroxidation was also observed. This subsequently improved the efficiency of CO2 assimilation and protected the plants from high temperature stress, which indicated that the transgenic tomato presented an enhanced tolerance to high temperature stress (38 degrees C) compared with wild-type plants. Our results demonstrated clearly that increasing polyamine biosynthesis in plants may be a means of creating high temperature-tolerant germplasm.
Dual Role of Endophytic Entomopathogenic Fungi: Induce Plant Growth and Control Tomato Leafminer Phthorimaea Absoluta

Abstract:BACKGROUNDEntomopathogenic fungi (EPF) are multifunctional microorganisms acting not only as biopesticides against insect pests but also as endophytes which regulate plant growth. The tomato leafminer, Phthorimaea absoluta (Tuta absoluta) is a devastating invasive pest of tomatoes worldwide. However, effective alternatives are needed for a sustainable management of this invasive pest. In this study, the functional effects of five EPF isolates Metarhizium flavoviride, M. anisopliae, M. rileyi, Cordyceps fumosorosea and Beauveria bassiana were evaluated on tomato growth promotion and pest protection against P. absoluta. RESULTSWhen directly sprayed with conidia, P. absoluta larvae showed high cumulative mortality of 100% to M. anisopliae under 1 x 10(8) conidia/mL, whereas M. flavoviride, B. bassiana, C. fumosorosea and M. rileyi caused cumulative mortality of 92.65%, 92.62%, 92.16% and 68.95%, respectively. Moreover, all five EPF isolates can successfully colonize tomato plants, whilst the colonization rate for each EPF depends on the inoculation method used. The most efficient inoculation method for M. flavoviride and M. rileyi was root dipping, for M. anisopliae and C. fumosorosea it was coating seed, and for B. bassiana it was foliage spraying. The highest plant colonization was obtained by M. flavoviride. Meanwhile, all these isolates promoted tomato plant growth upon inoculation. Furthermore, endophytic colonization of plants by the five EPF negatively affected the performance of P. absoluta, among them M. anisopliae and C. fumosorosea showed strong negative effects on the performance of P. absoluta. CONCLUSIONOur results highlight the potential of incorporating entomopathogenic fungi as endophytes in integrated pest management practices to protect tomatoes against P. absoluta. & COPY; 2023 Society of Chemical Industry.
Fungal effector protein AVR2 targets diversifying defense-related cys proteases of tomato

Abstract:The interaction between the fungal pathogen Cladosporium fulvum and its host tomato (Solanum lycopersicum) is an ideal model to study suppression of extracellular host defenses by pathogens. Secretion of protease inhibitor AVR2 by C. fulvum during infection suggests that tomato papain-like cysteine proteases (PLCPs) are part of the tomato defense response. We show that the tomato apoplast contains a remarkable diversity of PLCP activities with seven PLCPs that fall into four different subfamilies. Of these PLCPs, transcription of only PIP1 and RCR3 is induced by treatment with benzothiadiazole, which triggers the salicylic acid-regulated defense pathway. Sequencing of PLCP alleles of tomato relatives revealed that only PIP1 and RCR3 are under strong diversifying selection, resulting in variant residues around the substrate binding groove. The doubled number of variant residues in RCR3 suggests that RCR3 is under additional adaptive selection, probably to prevent autoimmune responses. AVR2 selectively inhibits only PIP1 and RCR3, and one of the naturally occurring variant residues in RCR3 affects AVR2 inhibition. The higher accumulation of PIP1 protein levels compared with RCR3 indicates that PIP1 might be the real virulence target of AVR2 and that RCR3 acts as a decoy for AVR2 perception in plants carrying the Cf-2 resistance gene.

Abstract:Plant diseases are important issues in agriculture, and the development of effective and environment-friendly means of disease control is crucial and highly desired. Antimicrobial peptides (AMPs) are known as potential alternatives to chemical pesticides because of their potent broad-spectrum antimicrobial activity and because they have no risk, or have only a low risk, of developing chemical-resistant pathogens. In this study, we designed a series of amphipathic helical peptides with different spatial distributions of positive charges and found that the peptides that had a special sequence pattern “BBHBBHHBBH” (“B” for basic residue and “H” for hydrophobic residue) displayed excellent bactericidal and fungicidal activities in a wide range of economically important plant pathogens. The peptides with higher helical propensity had lower antimicrobial activity. When we modified the peptides with a long acyl chain at their N-terminus, their plant protection effect improved. Our application of the fatty acyl-modified peptides on the leaves of tomato and Arabidopsis plants lessened the infection caused by Pectobacterium carotovorum subsp. carotovorum and Botrytis cinerea . Our study provides important insights on the development of more potent novel AMPs for plant protection.

Characterization of rhizosphere fungi that mediate resistance in tomato against bacterial wilt disease

Abstract:Plant immunization for resistance against a wide variety of phytopathogens is an effective strategy for plant disease management. Seventy-nine plant growth-promoting fungi (PGPFs) were isolated from rhizosphere soil of India. Among them, nine revealed saprophytic ability, root colonization, phosphate solubilization, IAA production, and plant growth promotion. Seed priming with four PGPFs exhibited early seedling emergence and enhanced vigour of a tomato cultivar susceptible to the bacterial wilt pathogen compared to untreated controls. Under greenhouse conditions, TriH_JSB27 and PenC_JSB41 treatments remarkably enhanced the vegetative and reproductive growth parameters. Maximum NPK uptake was noticed in TriH_JSB27-treated plants. A significant disease reduction of 57.3% against Ralstonia solanacearum was observed in tomato plants pretreated with TriH_JSB27. Furthermore, induction of defence-related enzymes and genes was observed in plants pretreated with PGPFs or inoculated with pathogen. The maximum phenylalanine ammonia lyase (PAL) activity (111U) was observed at 24h in seedlings treated with TriH_JSB27 and this activity was slightly reduced (99U) after pathogen inoculation. Activities of peroxidase (POX, 54U) and β-1,3-glucanase (GLU, 15U) were significantly higher in control plants inoculated with pathogen after 24h and remained constant at all time points. A similar trend in gene induction for PAL was evident in PGPFs-treated tomato seedlings with or without pathogen inoculation, whereas POX and GLU were upregulated in control plus pathogen-inoculated tomato seedlings. These results determine that the susceptible tomato cultivar is triggered after perception of potent PGPFs to synthesize PAL, POX, and GLU, which activate defence resistance against bacterial wilt disease, thereby contributing to plant health improvement.

Abstract:Pectin is one of the main components of the plant cell wall that functions as the primary barrier against pathogens. Among the extracellular pectinolytic enzymes, pectin methylesterase (PME) demethylesterifies pectin, which is secreted into the cell wall in a highly methylesterified form. Here, we isolated and functionally characterized the pepper (Capsicum annuum L.) gene CaPMEI1, which encodes a pectin methylesterase inhibitor protein (PMEI), in pepper leaves infected by Xanthomonascampestris pv. vesicatoria (Xcv). CaPMEI1 transcripts are localized in the xylem of vascular bundles in leaf tissues, and pathogens and abiotic stresses can induce differential expression of this gene. Purified recombinant CaPMEI1 protein not only inhibits PME, but also exhibits antifungal activity against some plant pathogenic fungi. Virus-induced gene silencing of CaPMEI1 in pepper confers enhanced susceptibility to Xcv, accompanied by suppressed expression of some defense-related genes. Transgenic ArabidopsisCaPMEI1-overexpression lines exhibit enhanced resistance to Pseudomonas syringae pv. tomato, mannitol and methyl viologen, but not to the biotrophic pathogen Hyaloperonospora parasitica. Together, these results suggest that CaPMEI1, an antifungal protein, may be involved in basal disease resistance, as well as in drought and oxidative stress tolerance in plants.

Abstract:MiAMP1 is a recently discovered 76 amino acid residue, highly basic protein from the nut kernel of Macadamia integrifolia which possesses no sequence homology to any known protein and inhibits the growth of several microbial plant pathogens in vitro while having no effect on mammalian or plant cells. It is considered to be a potentially useful tool for the genetic engineering of disease resistance in transgenic crop plants and for the design of new fungicides. The three-dimensional structure of MiAMP1 was determined through homonuclear and heteronuclear ((15)N) 2D NMR spectroscopy and subsequent simulated annealing calculations with the ultimate aim of understanding the structure-activity relationships of the protein. MiAMP1 is made up of eight beta-strands which are arranged in two Greek key motifs. These Greek key motifs associate to form a Greek key beta-barrel. This structure is unique amongst plant antimicrobial proteins and forms a new class which we term the beta-barrelins. Interestingly, the structure of MiAMP1 bears remarkable similarity to a yeast killer toxin from Williopsis mrakii. This toxin acts by inhibiting beta-glucan synthesis and thereby cell wall construction in sensitive strains of yeast. The structural similarity of MiAMP1 and WmKT, which originate from plant and fungal phyla respectively, may reflect a similar mode of action.

Abstract:The tomato Pto kinase confers resistance to the causative agent of bacterial speck disease, Pseudomonas syringae pv. tomato, by recognizing the pathogen effector proteins AvrPto or AvrPtoB. Pto-mediated resistance requires multiple signal transduction pathways and has been shown to activate many defense responses including an oxidative burst, rapid changes in the expression of over 400 genes, and localized cell death. We have tested the role in Pto-mediated resistance in tomato of a set of 21 genes from other species known to be involved in defense-related signaling. Expression of each gene was suppressed by virus-induced gene silencing (VIGS) and the effect on disease symptoms and bacterial growth during the tomato-Pseudomonas incompatible interaction was determined. We found that Pto-mediated resistance was compromised by silencing of genes encoding two mitogen-activated protein (MAP) kinase kinases, MEK1 and MEK2, two MAP kinases, NTF6 and wound-induced protein kinase (WIPK), a key regulator of systemic acquired resistance (SAR), NPR1, and two transcription factors, TGA1a and TGA2.2. A lesser impact on Pto-mediated resistance was observed in plants silenced for RAR1 and COI1. The identification of nine genes that play a role in resistance to bacterial speck disease both advances our knowledge of Pto signal transduction and demonstrates the conservation of many defense signaling components among diverse plant species.
Molecular cloning of a thaumatin-like protein gene from Pyrus pyrifolia and overexpression of this gene in tobacco increased resistance to pathogenic fungi

Abstract:Thaumatin-like proteins (TLPs) belong to the pathogenesis-related proteins 5 family, and members of TLP gene family accumulate in plants in response to pathogen infection or environmental stress. The biological functions of plant TLPs are believed to be diverse, and they are involved in host defense against both the biotic and abiotic stresses as well as the other physiological processes. Lots of TLPs were well studied as antifungal proteins in vivo or in vitro. To isolate TLP genes involved in self-defense during fruit development, a novel gene PpTLP was isolated from the fruit of Pyrus pyrifolia Nakai cv Huobali which is a kind of local sand pear with high resistance to biotic and abiotic stresses in Yunnan province of China. The nucleotide sequence of PpTLP was highly homologous with TLPs from other plant species, and the protein encoded by PpTLP has 16 conserved cysteine residues and 5 amino acid residues related with antifungal activity of plant TLPs, an arginine residue, a glutamic acid residue, and 3 aspartic acid residues. What is more, the putative protein PpTLP has the highly conserved main-chain conformation and the folding patterns as other plant TLPs with antifungal activity. PpTLP was abundantly expressed in pericarp and leaf of P. pyrifolia Nakai cv Huobali. Furthermore, in order to verify the function of PpTLP, the constitutive plant expression vector of PpTLP was constructed and transferred into tobacco (Nicotiana tabacum L. cv Xanthi). The Southern blot and quantitative reverse transcription-polymerase chain reaction (QRT–PCR) analyses demonstrated that the PpTLP gene was integrated into the genome of the tobacco transformants and highly expressed in the transgenic lines. The antifungal activity of PpTLP was detected in vitro plates, and the crude protein extract of transgenic tobacco plants inhibited the hyphal growth of Sclerotinia sclerotiorum, Phomopsis sp., Phytophthora parasitica var. nicotianae, and Alternaria sp. in different degrees.

Abstract:Pseudomonas syringae pv. actinidiae biovar 3 (Psa3) causes a devastating canker disease in yellow‐fleshed kiwifruit (Actinidia chinensis). The effector HopZ5, which is present in all isolates of Psa3 causing global outbreaks of pandemic kiwifruit canker disease, triggers immunity in Nicotiana benthamiana and is not recognised in susceptible A. chinensis cultivars. In a search for N. benthamiana nonhost resistance genes against HopZ5, we found that the nucleotide‐binding leucine‐rich repeat receptor NbPTR1 recognised HopZ5. RPM1‐interacting protein 4 orthologues from N. benthamiana and A. chinensis formed a complex with NbPTR1 and HopZ5 activity was able to disrupt this interaction. No functional orthologues of NbPTR1 were found in A. chinensis. NbPTR1 transformed into Psa3‐susceptible A. chinensis var. chinensis 'Hort16A' plants introduced HopZ5‐specific resistance against Psa3. Altogether, this study suggested that expressing NbPTR1 in Psa3‐susceptible kiwifruit is a viable approach to acquiring resistance to Psa3 and it provides valuable information for engineering resistance in otherwise susceptible kiwifruit genotypes.

Abstract:▪ Abstract The gene Mi, which confers resistance to several species of root-knot nematode, is present in many modern tomato cultivars. Recent cloning of this gene revealed that it encodes a member of the plant resistance protein family characterized by the presence of a putative nucleotide binding site and a leucine-rich repeat. Analysis of transgenic plants revealed the unexpected result that Mi also confers resistance to potato aphids. Although highly effective in many conditions, Mi fails to confer resistance at high soil temperature, and Mi-virulent nematode isolates have been identified in many areas of the world. These findings have stimulated efforts to identify new sources of root-knot nematode resistance. Resistance genes that differ from Mi in properties and genetic position have been identified in Lycopersicon peruvianum. These genes, as well as the cloned Mi gene, provide a resource for broadening the base of root-knot nematode resistance in tomato and other crops.

Abstract:Late blight (caused by Phytophthora infestans) poses a serious threat to tomato production but the number of late blight resistance genes isolated from tomato is limited, making resistance gene mining a high research priority. In this study, highly resistant CLN2037E and susceptible No. 5 tomato inbred lines were used to identify late blight resistance genes. Using transcriptome sequencing, we discovered 36 differentially expressed genes (DEGs), including 21 nucleotide binding site-leucine-rich repeat and 15 pathogenesis-related (PR) disease resistance genes. Cluster analysis and real-time quantitative PCR showed that these 36 genes possessed similar expression patterns in different inbred lines after inoculation with P. infestans. Moreover, two PR genes with unique responses were chosen to verify their functions when exposed to P. infestans: Solyc08g080660 and Solyc08g080670, both of which were thaumatin-like protein genes and were clustered in the tomato genome. Functions of these two genes were identified by gene overexpression and gene editing technology. Overexpression and knockout of single Solyc08g080660 and Solyc08g080670 corresponded to an increase and decrease in resistance to late blight, respectively, and Solyc08g080660 led to a greater change in disease resistance compared with Solyc08g080670. Cotransformation of dual genes resulted in a much greater effect than any single gene. This study provides novel candidate resistance genes for tomato breeding against late blight and insights into the interaction mechanisms between tomato and P. infestans.

Abstract:Species of the genus Phytophthora,the plant killer,cause disease and reduce yields in many crop plants.Although many Resistance to Phytophthora infestans(Rpi)genes effective against potato late blight have been cloned,few have been cloned-against other Phytophthora species.Most Rpi genes encode nucleotide-binding domain,leucine-rich repeat-containing(NLR)immune receptor proteins that recognize RXLR(Arg-X-Leu-Arg)effectors.However,whether NLR proteins can recognize RXLR effectors from mul-tiple Phytophthora species has rarely been investigated.Here,we identified a new RXLR-WY effector AV-Ramr3 from P.infestans that is recognized by Rpi-amr3 from a wild Solanaceae species Solanum ameri-canum.Rpi-amr3 associates with AVRamr3 in planta.AVRamr3 is broadly conserved in many different Phytophthora species,and the recognition of AVRamr3 homologs by Rpi-amr3 activates resistance against multiple Phytophthora pathogens,including the tobacco black shank disease and cacao black pod disease pathogens P.parasitica and P.palmivora.Rpi-amr3 is thus the first characterized resistance gene that acts against P.parasitica or P.palmivora.These findings suggest a novel path to redeploy known R genes against different important plant pathogens.
