Abstract:The factors which influence the saponification of Hippophae rhamnoides L.(HRL)seed oil including time,temperature and the alkali concentration were studied.The results showed that the optimum conditions for the saponification of HRL seed oil were that the saponification time,temperature and the alkali concentration were 60min,70℃,1mol/L,respectively.Under these conditions,a fatty acid yield of 91.08% was obtained,and the product contained 17.8%oleic acid,27.2%linoleic acid and 24.9%linolenic acid.
Simultaneous Chemical Fingerprinting and Quantitative Analysis of Crude and Processed Radix Scrophulariae from Different Locations in China by HPLC.

Abstract:A validated liquid chromatography method was first developed to evaluate the quality of crude and processed Radix Scrophulariae extracts through establishing chromatographic fingerprint and simultaneous determination of five bioactive compounds, namely 5-hydroxymethylfurfural (5-HMF), acteoside, angroside C, harpagoside and cinnamic acid. The chromatographic were separated on an Agilent Zorbax Extend C(18) column (250 mm × 4.6 mm, 5 μm) and detected by diode array detector (DAD). Mobile phase was composed of (A) aqueous phosphoric acid (0.03%, v/v) and (B) acetonitrile using a gradient elution. Analytes were performed at 30 °C with a flow rate of 1.0 mL/min and UV detection at 280 nm. All calibration curves showed good linear regression (r(2) ≥0.9996) within the tested ranges, and the recovery of the method was in the range of 98.12-103.38%, with RSD values ranging from 0.6 to 2.8%. In addition, the contents of those five bioactive compounds in crude and processed Radix Scrophulariae prepared by different locations of China were determined to establish the effectiveness of the method. The results demonstrate that the developed method is accurate and reproducible and could be readily utilized as a suitable quality control method for the quantification of Radix Scrophulariae.

Abstract:The extraction and refinement of rosa laevigata's seed oil investigates the physicochemical properties of seed oil.The ingredients and contents of fatty acids in the seed oil are determined with GC-MS.The result shows that the oil yield of rosa laevigata's seed oil is 5.24%;the acid value is 0.36 mgKOH / g;the peroxide value is 46.10 mmol / kg;the iodine value is 160.90 gI2/100 g;and the saponification value is 193.76 mgKOH / g.Five fatty acids are identified in the rasa laevigata's seed oil.The content of unsaturated fatty acid is 86.10% which includes 43.99% linoleic acid and 42.11% α-linolenic acid.But the saturated fatty acid is 13.90% which is composed of palmitic acid,stearic acid and arachidic acid.The seed oil has plentiful unsaturated fatty acids and it is a potential source for nutritional and healthy functions.

Abstract:Hippophae rhamnoides L. (sea buckthorn), consumed as a food and health supplement worldwide, has rich nutritional and medicinal properties. Different parts of H. rhamnoides L. were used in traditional Chinese medicines for relieving cough, aiding digestion, invigorating blood circulation, and alleviating pain since ancient times. Phytochemical studies revealed a wide variety of phytonutrients, including nutritional components (proteins, minerals, vitamins, etc.) and functional components like flavonoids (1-99), lignans (100-143), volatile oils (144-207), tannins (208-230), terpenoids (231-260), steroids (261-270), organic acids (271-297), and alkaloids (298-305). The pharmacological studies revealed that some crude extracts or compounds of H. rhamnoides L. demonstrated various health benefits, such as anti-inflammatory, antioxidant, hepatoprotective, anticardiovascular disease, anticancer, hypoglycemic, hypolipidemic, neuroprotective, antibacterial activities, and their effective doses and experimental models were summarized and analyzed in this paper. The quality markers (Q-markers) of H. rhamnoides L. were predicted and analyzed based on protobotanical phylogeny, traditional medicinal properties, expanded efficacy, pharmacokinetics and metabolism, and component testability. The applications of H. rhamnoides L. in juice, wine, oil, ferment, and yogurt were also summarized and future prospects were examined in this review. However, the mechanism and structure-activity relationship of some active compounds are not clear, and quality control and potential toxicity are worth further study in the future.

Abstract:Plant genus Hippophae contains a variety of bioactive substances and has high nutritional and medicinal value. To help the classification of Hippophae plants and ensure its quality, a high-performance liquid chromatography-diode array detector (HPLC-DAD) fingerprinting method was established based on the distribution and relative contents of flavonoids. Chemometrics analysis were used to analyze and compare the differences of Hippophae samples. Differences were observed among Hippophae varieties, and different plants within the same variety showed good similarity. Chemometrics analysis showed that 16 samples of Hippophae fruit and 40 samples of Hippophae leaf were divided into three groups. Four flavonoids were identified in fruit (isorhamnetin-3-O-glucoside-7-O-rhamnoside, rutin, isorhamnetin-3-O-rutinoside, and isorhamnetin) and five were identified in leaf (quercetin-7-O-glucoside, isorhamnetin-3-O-glucoside-7-O-rhamnoside, rutin, guaijaverin, and kaempferol-3-O-rutinoside). The highest flavonoid content in fruit was observed in Hippophae rhamnoides ssp. yunnanensis (average 3.651 mg/g). In leaf, Hippophae neurocarpa had the highest flavonoid content (average 13.303 mg/g) and a higher flavonoid content was observed in male plants (average 8.348 mg/g) than in female plants (average 7.273 mg/g). Correlation was observed between flavonoid content and climate factors. HPLC fingerprinting combined with chemometric analysis and quantitative analysis can provide a good foundation for subsequent production and processing.

[Explore antioxidant quality markers of Hippophae tibetana based on "dry-method + wet-method" technology]

Abstract:The cross combination of dry-method(network pharmacology analysis) and wet-method(high-resolution mass spectro-metry with antioxidation experiment) was used to predict antioxidant quality markers(Q-markers) of Hippophae tibetana. Ultra-high performance liquid chromatography coupled with hybrid quadrupole-orbitrap mass spectrometry(UPLC-Q-Exactive Orbitrap-MS) was developed to rapidly separate and identify the chemical constituents in H. tibetana. Then in DPPH free radicals and superoxide anion scavenging experiment, the antioxidant activity of the four different polar parts with extracts of petroleumether, ethyl acetate, n-butanol and water was evaluated. Network pharmacology method was used for functional enrichment and pathway analysis to screen antioxidant-related components and preliminarily explain the mechanism of action. On this basis, multi-source information was integrated to predict the antioxidant Q-markers. The results showed that 51 components in H. tibetana were identified, including 18 flavonoids, 14 terpenoids, 6 alkaloids, 4 coumarins and phenylpropanoids, 3 volatile components and 2 polyphenols. The antioxidant capacity of different fractions: ethyl acetate > n-butanol > water > petroleum ether. The medicine mainly acted on PI3 K-Akt and FoxO signaling pathways to perform antioxidant effects through flavonoids such as quercetin, luteolin and kaempferol. According to the results of dry-method and wet-method, quercetin, luteolin and kaempferol, the representatives of poly-hydroxy flavone, may be the antioxidant Q-markers of H. tibetana. In this study, with the antioxidant Q-markers of H. tibetana as an example, an investigation model of predicting Q-marker was discussed based on the ternary system of composition, function and informatics, providing a scientific basis for the establishment of quality evaluation standards for H. tibetana.

Abstract:The total flavonoids in different parts of Wuqi wild Hippophae rhamnoides L.was studied in the paper.The results showed that the total flavonoids was the highest in mashed of Hippophae rhamnoides L.fruit(814.03 mg/100 g),the next were the fruit peel,the pulp oil,the marc of fruit in order,and the total flavonoids was 90.39 mg/mL in rod of Hippophae rhamnoides L.fruit at least.This is the basic of theory for the further studies of Wuqi wild Hippophae rhamnoides L..

Abstract:Radix Ophiopogonis and Radix Liriopes are two widely used herbal medicines and functional foods with similar pharmacological activities and clinical efficacies. In order to figure out whether phytoequivalence of species leads to therapeutic equivalence and consistency, in this study, a novel sensitive and selective high performance liquid chromatography-tandem mass spectrometric method (HPLC-MS/MS) has been developed for the simultaneous analysis of 11 bioactive components (6 steroidal saponins and 5 flavonoids) and then was applied for comparing in Radix Ophiopogonis and Radix Liriopes from different habitats. Separation was performed using a C18 analytical column with gradient elution of acetonitrile and 0.3‰ acetic acid (v/v) at a flow rate of 0.8mL/min. Multiple-reaction monitoring (MRM) scanning was employed with switching electrospray ion source polarity between positive and negative modes in a single run. All calibration curves showed good linearity (r2>0.9915) within the test ranges. The intra-day and inter-day precision for 11 analytes were less than 3.7% and 4.2%, respectively. The recoveries were between 95.3% and 106.7%. Fifty-three batches of commercial samples from different sources were analyzed using the developed method. Hierarchical clustering analysis (HCA) and content analysis were performed to differentiate and classify the samples. The results demonstrated that steroidal saponins were the major components in both Liriopes species, and a low content of flavonoids could become an important symbol of distinction between Ophiopogonis and Liriopes. The developed method could successfully differentiate between Ophiopogonis and Liriopes.

Abstract:Objective To establish a method to control the quality of Xiao'er Changweikang Granules.Methods Herba Kummerowiae Striatae,Radix Paeoniae Rubra,Radix et Rhizoma Glycyrrhizae were indentified by TLC.Paeoniflorin and glycyrrhizic acid were determined by HPLC.The chromatographic separation was performed on the Kromasil C18 column(250 mm×4.6 mm,5 μm) by gradient elution with 0.1%phosphoric acid-acetonitrile at the flow rate of 1.0 mL/min,the detection wavelength was set at 230 nm and 250 nm.Results The developed TLC spots were fairly clear and the blank test showed no interference.The linear range of paeoniflorin was 2.9-174 μg/mL(r2=1.000 0),the average recovery rate was 99.25%,RSD=1.55%(n=6).The linear range of glycyrrhizic acid was 1.3-79 μg/mL(r2=0.999 9),the average recovery rate was 98.38%,RSD=1.27%(n=6).Conclusion The method is effective for the quality control of Xiao'er Changweikang Granules.
Systematical Quality Evaluation of Fructus Crataegi and Its Processed Drugs Planted in China

Abstract:OBJECTIVE To systematically evaluate the quality of Fructus Crataegi and its processed drugs planted in China.METHODS The contents of hyperoside and organic acid in 15 samples of Fructus Crataegi and its processed drugs were analyzed by HPLC and by titration method,respectively.Ursolic acid in samples was measured by TLC.Some physical and chemical tests (H_2O,total ash and EtOH extract) were done to investigate their quality.RESULTS Hyperoside was found in 15 samples and was a predominant flavone constituent in Fructus Crataegi.There were significant differences in contents of hyperoside and organic acid as well as some physical and chemical parameters among Fructus Crataegi and its processed drugs,which indicated that the quality of drugs was affected by different processing method.CONCLUSION The study provided theoretical and experimental data in order to systematically evaluate the quality of Fructus Crataegi and its processed drugs planted in China.

Abstract:Objective:To establish a sensitive and specific HPLC method and a standard fingerprint for quality control of Carthamus tinctorius.Method: About 46 samples from different sources were analyzed by HPLC.By the similarity evaluation system for chromatographic fingerprint of traditional chinese medicine(Version 2004A),mean chromatogram was generated as the representative standard fingerprint and the similarity of each chromatogram against the standard chromatogram was also calculated.Result: Twelve main marker peaks were selected in the standard fingerprint and 10 peaks of them were identified.The similarity of 76% samples was over 0.8.Conclusion: The quality control of crude drug can be evaluated by comparing with the standard fingerprint.The method can be applied for quality assessment of C.tinctorius.
Influence of different extraction methods on the chemical composition, antioxidant activity, and overall quality attributes of oils from peony seeds (Paeonia suffruticosa Andr.)

Abstract:Peony ( Paeonia suffruticosa Andr.) seed oil is full of nutrition and exerts positive effects on human's health. The study detected and compared the quality, lipid characteristics, and antioxidant activities of peony seed kernel and shell oil using three extraction techniques (cold pressing, supercritical fluid extraction, and ultrasonic-assisted solvent extraction). Results indicated that the peony seed oil was rich in α-linolenic acid (43.19–44.24%) and γ-tocopherol (403.31–535.15 mg/kg). The extraction methods and shells significantly affected the beneficial phytochemicals (polyphenols, phytosterols, and tocopherols) and antioxidant activities ( p < 0.05). Meanwhile, the highest lipid yield was obtained using ultrasonic-assisted solvent extraction (30.60%), while pressing showed the lowest value (17.12%). Supercritical fluid extraction had higher tocopherol (492.68–576.40 mg/kg) and phytosterol (3421.18–4006.23 mg/kg) than other methods. Additionally, ten individual polyphenols were quantified, with the number of catechin being higher. Although the pressing led to a lower lipid yield, its extracts presented a significant higher polyphenol content. In general, peony seed shell oils had a significant advantage in obtaining polyphenols, while seed kernel oil showed higher sterol and tocopherol content. The above information is valuable for manufacturers to select suitable approaches to produce peony seed oil with the required quality attributes and chemical compositions for targeted end-use.

Abstract:Objective: To develop a qNMR method for quantitative analysis of triacylglycerols in fruit oil of Hippophae rhamnoides (seabuckthorn, SBT) and analyze commercial samples of SBT oils using GC-MS and FTIR. Methods: SBT fruit oil (IPHRFH) was extracted with hexane and the triglyceride (TAG) was isolated by vacuum liquid chromatography. Six different branded SBT oils purchased from e-commerce suppliers (Amazon) and in-house prepared SBT oil was analyzed by qNMR and fatty acyl composition of TAGs determined by using NMR. In-house oil was also analysed by GC-MS and FTIR spectroscopy. Results: The qNMR results showed that the oil contained 80.3% of triacylglycerol (TAG). The SBT oil TAGs comprised of linolenate 6.6%, palmitoleate/oleate 65.4%, and total saturated fatty acyl chain including palmitate 28% as determined by qNMR. GC-MS analysis revealed that the major acyl functionalities present in the TAG were palmitoleic acid 36.5%, oleic acid 12.9%, palmitic acid 21.2%, and linoleic acid 18%. Of the six commercial samples analyzed, samples from only one supplier (SW) were fruit oil; All others were the seed oils or mix of fruit oil and seed oil. The labels for samples except for the SW did not indicate whether it was fruit oil or seed oil. Conclusion: The results suggest that SBT oil should be analyzed by combination of GC-MS, FTIR and qNMR for factual content of free fatty acid or TAGs, which are chemically different in nature and affect the quality of oil. GC-MS showed the content of omega free fatty acids after hydrolysis, while qNMR and FTIR showed the content of TAGs. The major acyl functionalities found in SBT fruit oil TAGs are palmitoleate/palmitate/oleate, while linoleate and linonelate make up a minor fraction. Furthermore, analysis of commercial samples showed discrepancies between label claims and actual content.

Abstract:The contents of constant metal elements,trace metal elements and seventeen kinds of amino acids in desert plant——Hippophae rhamnoides in Helingeer county of Huhhot were measured and analyzed.The results indicate that constant elements Ca、K、Na、Al、Mg show the same content orders in different members of the plant,namely,CaKNaAlMg.The trace elements to plant vital activities such as Cu,Fe,Mn,Zn exist differences,the content orders are FeMnCuZn in root,FeCuMnZn in trunk,FeMnCuZn in branch,respectively.The contents of seven kinds amino acids required in human body are relatively high,and the order in every member is branchroottrunk.

Abstract:Flavonoids in the water extract of Carthamus tinctorius L. exhibit potent biological activities such as anti-coagulant, vasodilation, anti-oxidant, neuroprotection and immunosuppressant. A high-performance liquid chromatographic method was established to evaluate the quality of Carthamus tinctorius through a simultaneous quantitation of eight flavonoids, hydroxysafflor yellow A (2), 6-hydroxykaempferol 3,6-di-O-β-glucoside-7-O-β-glucuronide (3), 6-hydroxykaempferol 3,6,7-tri-O-β-glucoside (4), 6-hydroxykaempferol 3-O-β-rutinoside-6-O-β-glucoside (6), 6-hydroxykaempferol 3,6-di-O-β-glucoside (7), 6-hydroxyapigenin 6-O-glucoside-7-O-glucuronide (8), anhydrosafflor yellow B (9), and kaempferol 3-O-β-rutinoside (10), together with two other compounds named guanosine (1) and syringin (5). Among them, compound 8 was identified as a new compound. The compounds were separated on an Alltech Alltima-C18 column with gradient elution of acetonitrile and 0.01% trifluoroacetic acid. The detection wavelength was 280nm. All the compounds showed good linearity (r2≥0.9989). The recoveries, measured at three concentration levels, varied from 94.9% to 105.2%. This method was also validated with respect to precision, repeatability and accuracy, and was successfully applied to quantify the 10 components in 46 batches of C. tinctorius samples from different areas. Significant variations were found in the contents of these compounds in these samples. Compared with the reported analytical methods of C. tinctorius, this simple and reliable method provided a new basis for overall assessment on quality of C. tinctorius and should be considered as a suitable quality control method.
Optimized extraction, enrichment, identification and hypoglycemic effects of triterpenoid acids from Hippophae rhamnoides L pomace

Abstract:The Hippophae rhamnoides L. pomace was generated in the production process for juice, wine of food industry. To expand the application of pomace, the extraction process optimization, enrichment and identification of triterpene acids were performed in this study. The extraction yield was 14.87% under optimal ultrasound-assisted extraction techniques performed via response surface methodology. The extract was subsequently purified to obtain the triterpenoid acid enrichment fraction (TPF) with the content of 75.23% ± 1.45%. 13 triterpenoid acids were identified via UPLC-Triple-TOF MS/MS and further semi-quantified through comparison with triterpenoid acid standards. TPF exhibited a strong inhibitory effect on α-glucosidase with IC50 value of 5.027 ± 0.375 μg/mL, as determined via enzyme inhibition experiment and molecular docking. Additionally, the TPF significantly reduced postprandial glucose levels, as revealed via carbohydrate tolerance tests, as well as ameliorate serum lipid profiles. Therefore, pomace may be a promising resource of functional food components with therapeutic and commercial values.
Analysis of components and study on antioxidant and antimicrobial activities of oil in apple seeds

Abstract:In order to improve the comprehensive utilization of major by-products in apple-juice processing, the components, antioxidant and antimicrobial activities of oil in two species apple seeds, Fuji and New Red Star, were investigated. The Soxhlet extracted oil content of apple seeds raged from 20.69 to 24.32 g/100 g. The protein, fiber and ash contents were found to be 38.85-49.55 g/100 g, 3.92-4.32 g/100 g and 4.31-5.20 g/100 g, respectively; the extracted oils exhibited an iodine value of 94.14-101.15 g I/100 g oil; refractive index (40 degrees C) was 1.465-1.466; density (25 degrees C) was 0.902-0.903 mg/ml; saponification value was 179.01-197.25 mg KOH/g oil; and the acid value was 4.036-4.323 mg KOH/g oil. The apple seed oils mainly consisted of linoleic acid (50.7-51.4 g/100 g) and oleic acid (37.49-38.55 g/100 g). Other prominent fatty acids were palmitic acid (6.51-6.60 g/100 g), stearic acid (1.75-1.96 g/100 g) and arachidic acid (1.49-1.54 g/100 g). Apple seed oil was proven to possess interesting properties, emerging from its chemical composition and from the evaluation of its in vitro biological activities. The apple seed oil was almost completely active against bacteria, mildews were less sensitive to apple seed oil than yeasts, and the minimum inhibitory concentration (MIC) of apple seed oil ranged from 0.3 to 0.6 mg/ml. The observed biological activities showed that the oil had a good potential for use in the food industry and pharmacy.
[New method of processing on Rhei Radix Et Rhizoma and study on quality control of its processing products]

Abstract:Objective: To establish a simple, rapid and usable new method of processing on Rhei Radix Et Rhizoma and the quality control standard on its processing products. Methods: The studies of processing on Rhei Radix Et Rhizoma were proceed using yellow rice wine as solvent, through spray, soften and dry at 60-70 degrees C. The contents of total and uncombined chrysophanol and emodin in multi-Rhei Radix Et Rhizoma and its processing products were determined by HPLC. Results: The new method of processing on Rhei Radix Et Rhizoma was simple, rapid and usable. The contents of uncombined chrysophanol and emodin in its processing products was 80%. Conclusion: This study provides a new method of processing on Rhei Radix Et Rhizoma and quality control standard on its processing products.
Comparative study of adsorption polysaccharide on bioactive components and in vitro antioxidant activity of sea buckthorn (Hippophaë rhamnoides L.) pulp oil

Abstract:Sea buckthorn (Hippophaë rhamnoides L.) pulp oil (SPO) is enriched in natural bioactive components with high pharmacological activities. However, the presence of polysaccharides affects oil quality. Herein, the effects of different polysaccharide-adsorption treatments on the bioactive components and the in vitro antioxidant activities of SPO were investigated. The results demonstrated that the microwave-assisted activated clay (MAE-ACL) adsorption was more effective than the ultrasound-assisted activated clay adsorption for removing polysaccharides. Although the bioactive component levels and antioxidant activities of the SPO were reduced during the adsorption, the squalene content showed no significant difference. By performing principal component and correlation analyses that further evaluated the connection between the bioactive components and the free radical scavenging activities of SPO, MAE-ACL was determined to be the more suitable polysaccharide-adsorption treatment.
Quality Status Analysis and Intrinsic Connection Research of Growing place, Morphological Characteristics, and Quality of Chinese Medicine: Cyperi Rhizoma (Xiangfu) as a Case Study

Abstract:Aim of the Study. We set out to investigate the quality status of CR in China objectively according to the Chinese Pharmacopeia (ChP). We also explored the correlation between its growth area, morphological characteristics, and quality by analyzing physical and chemical indexes from 47 batches of CR collected from 12 provinces belonging to six geographic divisions. Materials and Methods. The macroscopic characteristics of CR as well as its moisture, ethanol extract, essential oil, total ash, and acid-insoluble ash contents were examined and calculated. In addition, qualitative identification and quantitative determination of α-cyperone, cyperotundone, and nootkatone were simultaneously performed, and a high-performance liquid chromatography (HPLC) fingerprint chromatogram was established. Inductively coupled plasma mass spectrometry and gas chromatography methods recorded in ChP were used to measure the contents of residues of heavy metal and deleterious elements as well as residues of organochlorine pesticide, respectively. Hierarchical cluster analysis and typical canonical correlation analysis were performed using Origin 9.1 and SPSS 23.0 to explore the correlation between CR’s growth area, morphological characteristics, and quality. Results. Of the 47 batches of CR analyzed, only 4 collected from the province of Shandong had a flat appearance, which did not accord with the macroscopic characteristics of CR. Overall, only 4 batches met ChP standards for CR. In addition, 30 and 38 batches did not meet the requirements for moisture content and essential oil content, respectively. The similarity values of HPLC fingerprints ranged from 0.568 to 0.986. Results of hierarchical cluster analysis for ethanol extracts, essential oil, α-cyperone, cyperotundone, and nootkatone and the HPLC fingerprints (total peak time and peak area) suggested that the samples could be classified into four clusters, with no significant difference in growth geographic areas among them. Results of canonical correlation analysis indicated that the first canonical pair could represent the correlation between macroscopic characteristics (vector 1) and chemical quality (vector 2), with shorter diameter and length denoting lower ethanol extract content and higher nootkatone content in a single grain of CR. Conclusions. Crude medicinal materials were collected and examined in this study to reflect the overall quality status of CR in China. The methods chosen to detect, calculate, and analyze the quality of CR were suitable to the investigation, and the results are crucial not only for estimating the current quality status of CR, but also for conducting further research into its cultivation, quality assurance, and commodity specification. Besides, this mode of investigation could be used to evaluate other medicines.
