Abstract:Chrysanthemum variety ‘Hanxiaobai’ plants infecting Chrysanthemum virus B(CVB) were made CVB-free plants by using meristem-tips culture.Different sizes(<0.3 mm,0.3-0.7 mm,0.7-1.0 mm) of shoot tips were cultured on MSmedium with 2.0 mg·L-16-BA and 0.5 mg·L-1NAA,0.7-1.0 mm of shoot tips were first cultured on MSmedium,and then 0.3-0.7 mm shoot tips cut from the plants were cultured on MSmedium with 2.0 mg·L-16-BA and 0.5 mg·L-1NAA(secondary culture).In the secondary culture,tips of 0.7-1.0 mm were first cultured on MSmedium,and then tips with 0.3-0.7 mm cut from the plants were cultured on MSmedium with 2.0 mg·L-16-BA and 0.5 mg·L-1NAA.The plants were indexed by RT-PCR.The result showed that in the first tissue culture,the size of shoot tip was proportional to the livability rate and in inverse proportion to the elimination of CVB,meristem-tips with length less than 0.3 mm,produced the lowest re-growth plants(6%),but obtained the most virus-free plants(66.67%);meristemtips 0.7-1.0 mm in length,produced the most re-growth plants(90%),but obtained the least virus-free plants(6.67%).Secondary tissue culturing was the most effective method to the elimination of CVB,since 60% plants survived and 43.33% virus-free plants can be obtained by this way.

Abstract:Virus-free materials of Lilium spp.were obtained from meristem culture in vitro.A series of optimization experiments for cultural medium composition and concentration of phytohormones were conducted with a view to aceelerate the propagation of virus-free materlals.The best condition of chromosome determination was established.The obtained results indicated that the most suitable rapid propagation media of Lililum spp.was MS+ 6-BA 1.0mg/L+ IAA0.2mg/L+ KT1.0mg/L+ PP3334.0mg/L.The optimized medium for rooting was 1/2MS+IBA0.5mg/L.These results could meet the technical requirements of rapid-propagation in large scale production.Virus-free materials were observed repeatedly under electron microscope,10 virus-free samples were screened out which could be used to the fast propagation for production.Some physiological data were tested among the virus-free lines and control lines.There was difference between the lines.

Abstract:Shoot tips as explants of field Colocasia esculenta (cv.Fenghuayunaitou) were cultured on MS basal salts medium supplemented with 2.0mg/L 6-BA and 0.2mg/L NAA.75%,New buds can be obtained,and 4-times new buds were proliferated within one month.If 10% organism(such as taro or coconut milk) is added in media,the new buds were proliferated faster and plantlets with roots were obtained.In addition,(0.5-1mg/L) triacontanol increased the number and length of roots,also enhanced shoot growth and frest weight.

Abstract:Almost all plants in their natural environment are commonly infected by viruses. These viral infections can cause devastating diseases and result in severe yield and economic losses, making viral diseases an important limiting factor for agricultural production and sustainable development. However, these losses can be effectively reduced through the productions and applications of virus-free plantlets. In vitro culture techniques are the most successful approaches for efficient eradication of various viruses from almost all the most economically important crops. Techniques for producing virus-free plantlets include meristem tip culture, somatic embryogenesis, chemotherapy, thermotherapy, electrotherapy, shoot tip cryotherapy, and micrografting. Among them, meristem tip culture is currently the most widely used. Here, we describe a detailed protocol for producing virus-free plantlets of Chrysanthemum morifolium Ramat using tissue culture techniques.

Abstract:ABSTRACT The pathogen, Rhizoctonia solani Kuhn is a polyphagous fungus and has a vast potential of attacking a wide range of host plants causing host decay, damping –off, stem rot, crown rot ,stemcanker, root rot, fruit decay and foliage diseases. The stem rot is an emerging devastating disease incarnation rating next in importance to Fusarium wilt with a seedling mortality of 37% under optimum climatic conditions. Because of high incidence and crop importance, the present studywas carried out to manage the disease with application of different plant extracts (botanicals), biofumigant releasing crop residues and biocontrol agents both under in vitro and field conditions. Out of 11 botanicals including two commercial formulations of neem (neemgold, neemazil) testedat 5,10,20,30,40% and 1,2,3,4,5% concentrations respectively revealed that seed extracts of Meliaazedarach and leaf extract of Adhatoda vasica showed maximum inhibition in mycelial growthwithin the range of 44.38 to 44.25% followed by Murraya koenigii (37.77%) and Tagetes erecta(37.62%).The commercial formulations of neem compared to other treatments were foundstatistically superior to inhibits mycelial growth. In general, higher concentration resulted in moreinhibition of the R. solani, above 50 % inhibition was recorded at concentration of 40 per cent, maximum being in neemgold (60.53%) compared to 5% or 10%. Among different crop residues, cauliflower and cabbage were found most effective in reducing the incidence of carnation stem rot to 22.28 and 24.25% from 48.34% registered in control. The plant and flower characters such as plant height, stem length, no. of flowers/plant, flower size/plant and no. of days to first floweringget reduced to 142 compared to 157 days. The average colony forming unit of the pathogen reducedto 22.83x103 from 58.12x103 in one cropping season. Antagonists, Trichoderma viride, T. harzianum and Bacillus subtilis resulted into mycelial inhibition of 65.08, 63.70 and 55.05%. However in field the highest disease control of stem rot was achieved in integrated treatments whenthe rooted cuttings of carnation were pretreated with neemgold or neemazil for 20 minutes as diptreatment and planted in soil amended with T. viride and T. harzianum by giving disease control between 63-73%.

Abstract:OBJECTIVE To investigate the significance and the influence of transforming growth factor beta 1 (TGF beta 1) on fibroblast proliferation and collagen synthesis. METHODS The fibroblasts derived from healthy human dermis and pathologic scar were harvested and cultured in vitro with TGF beta 1. The effects of TGF beta 1 on the proliferation and collagen synthesis of the fibroblasts were investigated during the early phase (24 h) of cell confluence by means of nonradioactive cellular proliferation detection, 3H thymidine and 3H proline incorporation and DNA quantification analysis. RESULTS After the co-culture with TGF beta 1, the incorporation of 3H thymidine and 3H proline and the proliferation were enhanced significantly in the fibroblasts from pathologic scar (P < 0.01) but remained at normal level in those from healthy human dermis(P > 0.05). CONCLUSION The cell proliferation and collagen synthesis of scar-derived fibroblasts could be positively modulated and controlled by TGF beta 1, which might be very important in the scar formation.

Abstract:Using Hot pepper-Yunnan xiao mi jiao as experimental material, aseptic seedings with 2～3 leaves are cultured on 1/2 Ms at 35℃ for 4 weeks. Shoot tips 1～1.5 mm in length ,are cultvated on the basic medium of Ms supplemented with 6-BA, ZT, KT, GA 3,IAA,NAA etc. Ms+6-BA7.0mg/L+IAA0.05mg/L selected form experiments is favourable for shoot tip culture,Ms+ZT8.0 mg/L +GA 32.0 mg/L for shoot tip elongation, and 1/2Ms+IAA0.2 mg/L+NAA0.1 mg/L for root growth. The virus free rate of hot pepper produced by shoot tips 1～1.5mm in length reaches up to 96.3% in the study.

Abstract:This paper studied on the difference of a shallow liquid culture system and the solid culture system in the system of rapid propagation technology of virus-free ginger. The multiplication coefficient?the length of root and plant height in shallow liquid culture were prior to those of solid culture. As the regeneration frequency of root and the average root number of each explant were analyzed, there was no significant difference between the two systems. The survival rate of transplantation of seedlings from the shallow liquid system was much higher than that from solid culture system. Furthermore, the cost of chemicals of shallow liquid system was only less than 1/3 compared to that of solid system. Meanwhile, the load of manual work and time were reduced much more. So it was more economical and convenient. For all of the above, the shallow liquid culture system was proposed during rapid propagation process of virus-free ginger.

Abstract:The virus-free yacon was got by culturing the meristem-tip from axillary buds of its tuber.The cucumber mosaic virus(CMV) was detected by DAS-ELISA.The results showed that 0.3 mm meristem-tip culture was an effective means for virus elimination and the ratio of virus elimination amount to 60%.A reliable and reproducible method for plant regeneration of yacon was developed which utilizes the stem(with nodes) and leaves from the virus-free plantlet as explants.The optimal proliferating shoot cultures medium from stems was MS supplemented with BAP 1 mg·L~(-1) and NAA 0.1 mg·L~(-1).Highly regenerative callus was obtained from the leaves following cultivation on MS medium supplemented with BA 0.2 mg·L~(-1),TDZ 0.01 mg·L~(-1) and NAA 0.05 mg·L~(-1).Regenerated shoots could produce roots in 1/2MS medium containing NAA 0.01 mg·L~(-1) and the plantlets were successfully transferred to soil,the survival rate being up to 80%.

Abstract:We use the technique of anther culture to induce haploids and homozygous diploids from TCF_(2) of OguCMS×Ad-6(F4). Buds containing uninucleate microspores were harvested and either pretreated (5 day at 4℃) or dissected immediately after harvest, various culture media and incubation temperature were tested. The result showed first that bud pretreatment can improve the induction rate of callus and secondly that an incubation teatment for 24h at 33℃ followed by 6 days at 30℃ increased the induction rate of callus. High numbers of callus were induced by culturing the anthers on B5 medium containing 10% or 8% sucrose addition with 0.5mg/L 2, 4-D、0.2mg/L BA and 0.2mg/L NAA. Shoots regenerated from callus on the MS medium addition with 5mg/L BA.

Abstract:Abstract Bcakground The dry root and rhizome of Salvia miltiorrhiza are used to treat cardiovascular diseases, chronic pain, and thoracic obstruction over 2000 years in Asian countries. For high quality, Sichuan Zhongjiang is regarded as the genuine producing area of S. miltiorrhiza. Given its abnormal pollen development, S. miltiorrhiza from Sichuan ( S.m. -SC) relies on root reproduction and zymad accumulation; part of diseased plants present typical viral disease symptoms and seed quality degeneration. This study aim to detected unknown viruses from mosaic-diseased plants and establish a highly efficient virus-free regeneration system to recover germplasm properties. Results Tobacco mosaic virus (TMV) and cucumber mosaic virus (CMV) were detected from mosaic-diseased plants. Primary apical meristem with two phyllo podium in 0.15–0.5 mm peeled from diseased plants were achieved 73.33% virus-free rate. The results showed that the medium containing MS, 0.5 mg/L 6-BA, 0.1 mg/L NAA, 0.1 mg/L GA 3 , 30 g/L sucrose and 7.5 g/L agar can achieve embryonic-tissue (apical meristem, petiole and leaf callus) high efficient organogenesis. For callus induction, the optimal condition was detected on the medium containing MS, 2 mg/L TDZ, 0.1 mg/L NAA by using secondary petiole of virus-free plants under 24 h dark/d condition for 21 d. The optimal system for root induction was the nutrient solution with 1/2 MS supplemented with 1 mg/L NAA. After transplant, the detection of agronomic metric and salvianolic acid B content confirmed the great germplasm properties of S.m.-SC virus-free plants. Conclusions A highly efficient virus-free regeneration system of S.m.- SC was established based on the detected viruses to recover superior seed quality. The proposed system laid support to control disease spread, recover good germplasm properties in S.m.- SC.

Abstract:Virus is the most menacing factor for plant, which causes enormous economic losses in agriculture worldwide. Tobacco mosaic virus is most hazardous virus among the plants that can spread through biological and non-biological sources. TMV is ancient virus that causes huge economic losses to pepper cucumber ornamental crops and tobacco. It can be controlled by reducing the population of vector through pesticide application. However, the rapid usage of synthetic chemicals causes environmental pollution and destroys our ecosystem. Consequently, different approaches just like natural derivatives should be adopted for the environmental friendly management for TMV. This in vitro study demonstrated the potential role of natural metabolites such as poultry manure and plant extracts such as salicylic acid and citric acid for the control of TMV. Two different concentrations of poultry manure 60G and 30G were used. Poultry manure was mixed with the soil at the time of sowing. Disease severity was minimum at maximum concentration as compared to the control. Meanwhile, two different concentrations of salicylic acid and citric acid 60% and 90% were applied by foliar sprayer after three-leaf stages. Disease severity was observed after 5, 10,15,20,25, and 30 days after disease inoculation. Here also maximum concentration showed the minimum disease severity and higher concentration of both animal and plants extracts were used for following experiment. Quantitative real-time PCR (RT-qPCR) results demonstrated that different plant defense-related genes such as PR1a, PAL, PR5, NPR1, PRIb, and PDF1.2 were up-regulated. Furthermore, applications of each treatment-induced systemic resistance against a wide range of pathogen including TMV and fungal pathogen Botrytis cinerea.

Abstract:Plant virus elimination is commonly used to rejuvenate the plant, and improve yield and quality, but how it affects belowground growth and the soil environment remains unknown. Here, the root morphological and metabolic traits, soil physicochemical properties, and bacterial diversity and composition of the rhizosphere and surrounding bulk soil of virus-free (VF) and common virus- carrying (VC) Chrysanthemum morifolium Ramat were identifed using LC-MS-MS and high-throughput MiSeq sequencing analysis. These plants were cultivated in the same field and experienced similar management practices. The results showed that the VF chrysanthemum cultivation improved soil properties, including soil carbon and aggregate stability, and exhibited significantly higher plant growth parameters than the VC plants. Plant virus elimination significantly enhanced root biomass production and proliferation, and promoted the synthesis of root metabolites, in addition to the quantity of rootreleased organic carbon. Further analysis indicated that root-associated microbial diversity increased after plant virus elimination. The shift from soil oligotrophy to copiotrophy bacteria were mainly driven by changes in root morphological and metabolic traits, indicating a strong bottom-up regulatory effect in VF chrysanthemum roots. In conclusion, plant virus elimination has great potential for improving soil quality, bacterial diversity, aboveand belowground productivity, which may provide a new approach for future environmental breeding programs.
Combined thermotherapy and cryotherapy for efficient virus eradication: relation of virus distribution, subcellular changes, cell survival and viral RNA degradation in shoot tips

Abstract:Accumulation of viruses in vegetatively propagated plants causes heavy yield losses. Therefore, supply of virus-free planting materials is pivotal to sustainable crop production. In previous studies, Raspberry bushy dwarf virus (RBDV) was difficult to eradicate from raspberry (Rubus idaeus) using the conventional means of meristem tip culture. As shown in the present study, it was probably because this pollen-transmitted virus efficiently invades leaf primordia and all meristematic tissues except the least differentiated cells of the apical dome. Subjecting plants to thermotherapy prior to meristem tip culture heavily reduced viral RNA2, RNA3 and the coat protein in the shoot tips, but no virus-free plants were obtained. Therefore, a novel method including thermotherapy followed by cryotherapy was developed for efficient virus eradication. Heat treatment caused subcellular alterations such as enlargement of vacuoles in the more developed, virus-infected cells, which were largely eliminated following subsequent cryotherapy. Using this protocol, 20-36% of the treated shoot tips survived, 30-40% regenerated and up to 35% of the regenerated plants were virus-free, as tested by ELISA and reverse transcription loop-mediated isothermal amplification. Novel cellular and molecular insights into RBDV-host interactions and the factors influencing virus eradication were obtained, including invasion of shoot tips and meristematic tissues by RBDV, enhanced viral RNA degradation and increased sensitivity to freezing caused by thermotherapy, and subcellular changes and subsequent death of cells caused by cryotherapy. This novel procedure should be helpful with many virus-host combinations in which virus eradication by conventional means has proven difficult.

Elimination of PPV and PNRSV through thermotherapy and meristem-tip culture in nectarine

Abstract:The plum pox virus (PPV) and prunus necrotic ringspot virus (PNRSV) cause serious disease problems in stone-fruit trees. In this work, the possibility of obtaining plant material free from these viruses through thermotherapy and meristem-tip culture from infected nectarine shoots (Prunus persica var. nectarina Max, cv. 'Arm King') was studied. In addition, the detection of these viruses in in vitro cultures and young acclimatized plantlets with double antibody sandwich-enzyme-linked immunosorbent assay (DAS-ELISA) and multiplex reverse transcriptase-polymerase chain reaction (RT-PCR) was studied. Meristem-tip explants (0.8-1.3 mm) derived from sprouted buds of winter wood and spring shoots from field grown plants had a 2-5% regeneration response. However, application of thermotherapy to potted nectarine trees (3 weeks at a maximum temperature of 35 degrees C) facilitated excision of longer meristem tips (1.3-2.0 mm) that resulted in a significantly higher regeneration response (38%) in woody plant medium (WPM) without plant growth regulators. Such explants formed multiple shoots with the addition of 8 microM benzylaminopurine and 0.8 microM indoleacetic acid. When they were tested for the presence of PPV and PNRSV, 86% and 81% were found to be virus-free as detected by DAS-ELISA and multiplex RT-PCR, respectively. Individual shoots excised from virus-free cultures readily rooted in vitro (half-strength WPM plus 2 microM indolebutyric acid) and grew to plantlets. The combination of an efficient protocol for virus elimination and the establishment of highly sensitive diagnostics resulted in the production of nectarine plants free from PPV and PNRSV.
Inactivation of T cell receptor peptide-specific CD4 regulatory T cells induces chronic experimental autoimmune encephalomyelitis (EAE)

Abstract:T cell receptor (TCR)-recognizing regulatory cells, induced after vaccination with self-reactive T cells or TCR peptides, have been shown to prevent autoimmunity. We have asked whether this regulation is involved in the maintenance of peripheral tolerance to myelin basic protein (MBP) in an autoimmune disease model, experimental autoimmune encephalomyelitis (EAE). Antigen-induced EAE in (SJL x B10.PL)F1 mice is transient in that most animals recover permanently from the disease. Most of the initial encephalitogenic T cells recognize MBP Ac1-9 and predominantly use the TCR V beta 8.2 gene segment. In mice recovering from MBP-induced EAE, regulatory CD4+ T cells (Treg) specific for a single immunodominant TCR peptide B5 (76-101) from framework region 3 of the V beta 8.2 chain, become primed. We have earlier shown that cloned B5-reactive Treg can specifically downregulate responses to Ac1- 9 and also protect mice from EAE. These CD4 Treg clones predominantly use the TCR V beta 14 or V beta 3 gene segments. Here we have directly tested whether deletion/blocking of the Treg from the peripheral repertoire affects the spontaneous recovery from EAE. Treatment of F1 mice with appropriate V beta-specific monoclonal antibodies resulted in an increase in the severity and duration of the disease; even relapses were seen in one-third to one-half of the Treg-deleted mice. Interestingly, chronic disease in treated mice appears to be due to the presence of Ac1-9-specific T cells. Thus, once self-tolerance to MBP is broken by immunization with the antigen in strong adjuvant, TCR peptide- specific CD4 Treg cells participate in reestablishing peripheral tolerance. Thus, a failure to generate Treg may be implicated in chronic autoimmune conditions.

Abstract:Using three Citrullus lanatus cultivars of F1 generation 'Chunguang','Nabite','Xidu' which including lots of merits as materials,the technology of anther culture were studied.The results showed that more callus were formed on MS+KT 1.5 mg/L+BA 2.0 mg/L+NAA 1.5 mg/L;the best concentration of sucrose for callus induction was 6%;the results indicated that the treating of 4℃,72 hours was the proper condition that make the frequencies of callus induction;the studying of culture conditions and culture means indicated that callus inducement was better when it was cultured at 30℃ for 72 h and then cultured at 25℃.The quality of watermelon callus induced in dark was much better than in light.
