Abstract:Transition metal ions are served as disinfectant thousand years ago. However, the in vivo antibacterial application of metal ions is strongly restricted due to its high affinity with proteins and lack of appropriate bacterial targeting method. Herein, for the first time, Zn2+-gallic acid nanoflowers (ZGNFs) are synthesized by a facile one-pot method without additional stabilizing agents. ZGNFs are stable in aqueous solution while can be easily decomposed in acidic environments. Besides, ZGNFs can specifically adhere onto Gram-positive bacteria, which is mediated by the interaction of quinone from ZGNFs and amino groups from teichoic acid of Gram-positive bacteria. ZGNFs exhibit high bactericidal effect toward various Gram-positive bacteria in multiple environments, which can be ascribed to the in situ Zn2+ release on bacterial surface. Transcriptome studies reveal that ZGNFs can disorder basic metabolic processes of Methicillin-resistant Staphylococcus aureus (MRSA). Moreover, in a MRSA-induced keratitis model, ZGNFs exhibit long-term retention in the infected corneal site and prominent MRSA elimination efficacy due to the self-targeting ability. This research not only reports an innovative method to prepare metal-polyphenol nanoparticles, but also provides a novel nanoplatform for targeted delivery of Zn2+ in combating Gram-positive bacterial infections.

Abstract:Silver nanoparticles (AgNPs) are widely used as antibacterial agents because of their significant antimicrobial activities and little sign of antimicrobial resistance. However, the relatively high toxicity to healthy cells and low penetration efficiency into bacterial biofilms prevent their further use in biomedical applications. In order to decrease the cytotoxicity of the AgNPs to mammalian cells while increasing their antibacterial and antibiofilm efficiency, a novel nanocomposite composed of AgNPs decorated with carboxyl betaine groups (AgNPs-LA-OB) was prepared. The zwitterion modified AgNPs showed a pH responsive transition from a negative charge to a positive charge, which enabled the AgNPs to be compatible with mammalian cells and red blood cells (RBCs) in healthy tissues (pH ∼ 7.4), while strongly adhering quickly to negatively charged bacterial surfaces at infectious sites (pH ∼ 5.5) based on electrostatic attraction. The AgNPs penetrated deeply into bacterial biofilms and killed the bacteria living in an acidic environment. The results indicated that the designed zwitterion NPs for antibacterial applications and eradication of bacterial biofilms, which also had particles that did not harm the healthy cells showed promise for future use in humans. The satisfactory selectivity for bacteria compared to RBCs, together with their potent eradication of bacterial biofilms make AgNPs-LA-OB a promising antibacterial nanomedicine in biomedical fields.
Responsive Assembly of Silver Nanoclusters with a Biofilm Locally Amplified Bactericidal Effect to Enhance Treatments Against Multi-Drug-Resistant Bacterial Infections.

Abstract:Bacterial biofilms pose a major threat to public health because they are resistant to most current therapeutics. Conventional antibiotics exhibit limited penetration and weakened activity in the acidic microenvironment of a biofilm. Here, the development of biofilm-responsive nanoantibiotics (rAgNAs) composed of self-assembled silver nanoclusters and pH-sensitive charge reversal ligands, whose bactericidal activity can be selectively boosted in the biofilm microenvironment, is reported. Under neutral physiological conditions, the bactericidal activity of rAgNAs is self-quenched because the toxic silver ions' release is largely inhibited; however, upon entry into the acidic biofilm microenvironment, the rAgNAs not only exhibit charge reversal to facilitate local accumulation and retention but also disassemble into small silver nanoclusters, thus enabling deep penetration and accelerated silver ions release for dramatically amplified bactericidal activity. The superior antibiofilm activity of rAgNAs is demonstrated both in vitro and in vivo, and the mortality rate of mice with multi-drug-resistant biofilm-induced severe pyomyositis can be significantly reduced by rAgNAs treatment, indicating the immense potential of rAgNAs as highly efficient nanoscale antibacterial agents to combat resistant bacterial biofilm-associated infections.

Abstract:Integrating kinds of bactericides as one entirety shows the prospect for improving their antibacterial efficiency, thus a new type of nanoclusters based on bactericide-directed herein emerged as a combination for achieving the higher antibacterial effect. To be specific, an easily-operated approach for preparing the bacitracin-directed silver, gold and copper nanoclusters (AgNCs, AuNCs and CuNCs) has been proposed. Meanwhile, AgNCs@Bacitracin, AuNCs@Bacitracin and CuNCs@Bacitracin respectively emitted cyan, yellow and red color of fluorescence, and different sizes and nitrogen contents of these nanoclusters played the critical role for their fluorescence alterations. Significantly, AgNCs@Bacitracin exhibited a robust bacteria-killing efficiency rather than other nanoclutsers, owing to the dramatic damage on the bacterial membrane by the as-prepared AgNCs@Bacitracin. Additionally, propidium iodide (PI) staining indicated the bacterial membrane damage for 72.3% of the bacteria population followed by the treatment with AgNCs@Bacitracin, and the introduction of AgNCs@Bacitracin lead to the higher Reactive Oxygen Species (ROS) production, facilitating their attractive antibacterial activity. Moreover, this strategy of developing new nanoclusters broadened the avenues for designing the improved antibacterial materials.

Abstract:In this study, silver nanoparticles (Ag NPs) coated with catechol-conjugated chitosan (CSS) were prepared using green methods. Interestingly, we uncovered that CSS-coated Ag NPs (CSS-Ag NPs) exhibited a higher toxicity against gram-negative Escherichia coli (E. coli) bacteria than against grampositive Staphylococcus aureus (S. aureus) bacteria. The differences revealed that the CSS-Ag NPs killed gram bacteria with distinct, species-specific mechanisms. The aim of this study is to further investigate these underlying mechanisms through a series of analyses. The ultrastructure and morphology of the bacteria before and after treatment with CSS-Ag NPs were observed. The results demonstrated the CSS-Ag NPs killed gram-positive bacteria through a disorganization of the cell wall and leakage of cytoplasmic content. In contrast, the primary mechanism of action on gram-negative bacteria was a change in membrane permeability, induced by adsorption of CSS-Ag NPs. The species-specific mechanisms are caused by structural differences in the cell walls of gram bacteria. Gram-positive bacteria are protected from CSS-Ag NPs by a thicker cell wall, while gram-negatives are more easily killed due to an interaction between a special outer membrane and the nanoparticles. Our study offers an in-depth understanding of the antibacterial behaviors of CSS-Ag NPs and provides insights into ultimately optimizing the design of Ag NPs for treatment of bacterial infections.

Abstract:Novel polymer–silver hybrid core–shell nanoparticles (SNSs) which showed long term bacteriostatic, photohyperthermia induced bactericidal and triggered Ag+ release abilities were reported. Abstract:In this study, catechol-conjugated chitosan (CSS) was synthesized to prepare silver nanoparticles in aqueous solution as both a reducing and stabilizing agent. The entire reaction process complied with the principles of green chemistry. The stability and physicochemical properties of CSS-coated silver nanoparticles (CSS–Ag NPs) were well characterized by a series of techniques. A narrow diameter distribution and excellent stability were observed in the target CSS–Ag NPs. Determination of the minimum inhibitory concentration (MIC) and the disk diffusion test were applied to evaluate the antibacterial activities with respect to quantity and quality, respectively. The cytotoxic effects on HepG2 cells were also evaluated with a series of assays. The resulting CSS–Ag NPs combined the unique properties of chitosan and silver nanoparticles, showing effective antibacterial activities and low cytotoxicity. The effective toxic concentration of CSS–Ag NPs towards E. coli and S. aureus is too low to damage HepG2 cells. The results demonstrated that CSS–Ag NPs could be a potential candidate for use in biological and pharmaceutical areas to prevent infections caused by microorganisms.

Abstract:Nanotechnology is a highly promising field, with nanoparticles produced and utilized in a wide range of commercial products. Silver nanoparticles (AgNPs) has been widely used in clothing, electronics, bio-sensing, the food industry, paints, sunscreens, cosmetics and medical devices, all of which increase human exposure and thus the potential risk related to their short- and long-term toxicity. Many studies indicate that AgNPs are toxic to human health. Interestingly, the majority of these studies focus on the interaction of the nano-silver particle with single cells, indicating that AgNPs have the potential to induce the genes associated with cell cycle progression, DNA damage and mitochondrial associated apoptosis. AgNPs administered through any method were subsequently detected in blood and were found to cause deposition in several organs. There are very few studies in rats and mice involving the in vivo bio-distribution and toxicity, organ accumulation and degradation, and the possible adverse effects and toxicity in vivo are only slowly being recognized. In the present review, we summarize the current data associated with the increased medical usage of nano-silver and its related nano-materials, compare the mechanism of antibiosis and discuss the proper application of nano-silver particles.
Cationic antibacterial metal nanoclusters with traceable capability for fluorescent imaging the nano—bio interactions

Abstract:A thorough understanding of antimicrobial mechanism is of great importance for developing novel, efficient antibacterial agents. While cationic nanoparticles, such as metal nanoclusters (NCs), represent an attractive type of antibacterial nanoagents, their interactions with bacteria remains largely un-elucidated. Herein, we report the synthesis of cationic bovine serum albumin-protected AuAgNCs (cBSA-AuAgNCs), which exhibit both near-infrared (NIR) fluorescence properties and significant antimicrobial effects. With E. coli and S. aureus as the representative bacteria, we investigated the antimicrobial process of cBSA-AuAgNCs in real-time based on their intrinsic fluorescence properties via fluorescence imaging. Our results showed that these cBSA-AuAgNCs exert their antimicrobial effects primarily by attaching on the outer membrane of bacteria without obvious internalization, which is significantly different from the antibacterial process of negatively-charged metal NCs. Further mechanistic investigation showed that these cationic NCs will cause serious disruption to the bacterial membrane due to strong electrostatic interactions, which then leads to over accumulation of reactive oxygen species (ROS) that finally causes the bactericidal action. This study demonstrates the great potential of cationic luminescent metal NCs as novel, traceable antimicrobial agents, which also provides new tools for further understanding microbial interactions of nanomedicines.

Abstract:A single layer zwitterionic polymer brush grafted surface with pH-responsive charge reversal capacity was prepared in a simple and widely used strategy. The amide bond of the zwitterionic polymer will break under acidic condition and undergo a charge reversal, resulting the surface to convert from an antifouling zwitterionic surface to a bactericidal cationic surface. The surface we constructed can successfully resist the attachment of initial bacteria at physiological conditions and kill bacteria on demand to defend bacterial infection, showing an attractive application in implantable medical devices.
Hybrids of Upconversion Nanoparticles and Silver Nanoclusters Ensure Superior Bactericidal Capability via Combined Sterilization

Abstract:It is highly desired to develop new antibacterial agents with superior bactericidal efficiency for minimizing the damage to biological cells. We developed a combined antibacterial nanohybrid exhibiting a superb bactericidal effect and excellent biocompatibility by integrating upconversion nanoparticles (UCNPs) with silver nanoclusters (AgNCs). UCNPs and methylene blue (MB) molecules were encapsulated with silica microspheres via microemulsion, with MB as the photosensitizer. Silver ions (Ag+) were reduced by amino groups on the surface of silica spheres, wherein silver nanoclusters (AgNCs) were formed in situ to produce the nanohybrid, UCNPs@SiO2(MB)@AgNCs. UCNPs emit visible light at 655 nm under excitation by near-infrared radiation (NIR, 980 nm). MB absorbs the emission from UCNPs to generate toxic singlet oxygen (1O2), which leads to the apoptosis of bacteria cells. Meanwhile, silver ions released from AgNCs destroy the bacteria membrane structure. Upon NIR irradiation at 980 nm for 10 min, 8.33 μg mL-1 nanohybrid results in a 100% killing rate for both Gram-positive S. aureus (+) and Gram-negative E. coli (-).
Conjugating AIE-featured AuAg nanoclusters with highly luminescent carbon dots for improved visible-light-driven antibacterial activity

Abstract:Metal nanoclusters (NCs) have emerged as a novel antibacterial agent featuring broad-spectrum antibacterial activity without drug resistance for bacteria, but suffer from fast antibacterial invalidation due to the consumption by bacteria. Herein we report the design of a visible-light-driven photodynamic antibacterial agent based on conjugating aggregation-induced emission (AIE)-featured AuAg NCs with highly luminescent carbon dots (CDs). The conjugation of CDs with AuAg NCs could not only enhance the visible-light harvest, but also promote charge carrier generation/separation via charge/energy transfer, leading to the production of abundant reactive oxygen species (ROS) for bacterial killing under visible-light irradiation. Consequently, the as-obtained CDs@AuAg NCs display excellent photodynamic antibacterial activity against both gram-positive and gram-negative bacteria with 4–5 orders of magnitude reduction in colony forming units, which is different from the conventional metal NCs-based analogue relying on the self-consumption for bacterial killing. In addition, the CDs@AuAg NCs are found to be free of cytotoxicity; the ROS capture experiments indicate that the photoproduced H2O2 by CDs@AuAg NCs is the main active species for bacterial killing, accounting for nearly 48% of total antibacterial efficacy. This study provides a paradigm in the design of metal NCs-based photodynamic antibacterial agents for diversified bactericidal applications.

Abstract:The use of traditional Ag‐based antibacterial agents is usually accompanied by uncontrollable silver release, which makes it difficult to find a balance between antibacterial performance and biosafety. Herein, we prepared a core‐shell system of ZIF‐8‐derived amorphous carbon‐coated Ag nanoparticles (Ag@C) as an ideal research model to reveal the synergistic effect and structure‐activity relationship of the structural transformation of carbon shell and Ag core on the regulation of silver release behavior. It is found that Ag@C prepared at 600 °C (AC6) exhibits the best ion release kinetics due to the combination of relatively simple shell structure and lower crystallinity of Ag core, thereby exerting stronger antibacterial properties (> 99.999%) at trace doses (20 μg mL‐1) compared with most other Ag‐based materials. Meanwhile, the carbon shell prevents the metal Ag from being directly exposed to the organism and thus endows AC6 with excellent biocompatibility. In animal experiments, AC6 can effectively promote wound healing by inactivating drug‐resistant bacteria while regulating the expression of TNF‐α and CD31. This work provides theoretical support for the scientific design and clinical application of controllable ion‐releasing antibacterial agents.

Abstract:Superbugs are bacteria that have grown resistant to most antibiotics, seriously threating the health of people. Silver (Ag) nanoparticles are known to exert a wide-spectrum antimicrobial property, yet remains challenging against superbugs. Here, Ag clusters are assembled using porphyrin-based linkers and a novel framework structure (Ag9 -AgTPyP) is produced, in which nine-nuclearity Ag9 clusters are uniformly separated by Ag-centered porphyrin units (AgTPyP) in two dimensions, demonstrating open permeant porosity. Ag9 -AgTPyP eliminates over 99.99999% and 99.999% methicillin-resistant Staphylococcus aureus (MRSA) and Pseudomonas aeruginosa (P. aeruginosa) within 2 h upon visible-light irradiation, which are superior to a majority of bacteria inactivation photocatalysts. The novel-established long-term charge-transfer states from AgTPyP to adjacent Ag9 cluster that has preferential affinity to O2 greatly promote reactive oxygen species (ROS) production efficiency; and its unique framework accelerates the ROS transportation. Personal protective equipment (masks and protective suits) incorporating Ag9 -AgTPyP film also shows excellent performances against superbugs. This superbugs-killing efficiency is unprecedented among silver complexes and porphyrin derivatives. Utilizing efficient photogenerated electrons and holes between metal cluster and linkers can open up new interests of research in photocatalytic areas.

Abstract:Nanosilver has been regarded as a promising alternative to traditional antibiotics for fighting pathogen-associated infections due to its efficacy toward a broad spectrum of pathogens. However, bacterial resistance to nanosilver has emerged recently. In this contribution, a surface engineering strategy based on N-halamine chemistry to address bacterial resistance to nanosilver was proposed. Using 1,3-dichloro-5,5-dimethylhydantoin (DCDMH) as an N-halamine source, AgCl nanodots were deposited on the surface of Ag nanowires (Ag NWs) via in situ redox reaction to prepare AgCl-on-Ag NWs. After in vitro and in vivo tests, AgCl-on-Ag NWs effectively inactivated two antibiotic-resistant bacteria, ampicillin-resistant Escherichia coli (AREC) and methicillin-resistant Staphylococcus aureus (MRSA) with the minimum bactericidal concentration (MBC) as low as 10 μg·ml−1 and exhibited good biosafety against normal cells. The experimental and theoretical tests demonstrated that AgCl-on-Ag NWs worked on AREC and MASA by generating high level of reactive oxygen species under visible light irradiation, coupled with the sustained Ag+ ion release. Meanwhile, the antibacterial mechanism of AgCl-on-Ag NWs against MRSA was verified at the gene level by transcriptome analysis (RNA sequencing). Moreover, the full-thickness defect model verified that AgCl-on-Ag NWs reduced inflammatory cell infiltration and dramatically accelerated wound healing. This work provides a synergistic mechanism based on nanosilver surface engineering to eradicate the resistant bacteria that can alleviate drug resistance and develop an innovative approach for the treatment of bacterial infections.

Abstract:Abstract Nanomaterials, specifically metal nanoclusters (NCs), are gaining attention as a promising class of antibacterial agents. Metal NCs exhibit antibacterial properties due to their ultrasmall size, extensive surface area, and well-controlled surface ligands. The antibacterial mechanisms of metal NCs are influenced by two primary factors: size and surface charge. In this review, we summarize the impacts of size and surface charge of metal NCs on the antibacterial mechanisms, their interactions with bacteria, and the factors that influence their antibacterial effects against both gram-negative and gram-positive bacteria. Additionally, we highlight the mechanisms that occur when NCs are negatively or positively charged, and provide examples of their applications as antibacterial agents. A better understanding of relationships between antibacterial activity and the properties of metal NCs will aid in the design and synthesis of nanomaterials for the development of effective antibacterial agents against bacterial infections. Based on the remarkable achievements in the design of metal NCs, this review also presents conclusions on current challenges and future perspectives of metal NCs for both fundamental investigations and practical antibacterial applications. Silver‐Infused Porphyrinic Metal–Organic Framework: Surface‐Adaptive, On‐Demand Nanoplatform for Synergistic Bacteria Killing and Wound Disinfection

Abstract:The fabrication of functional nanoplatforms for combating multidrug‐resistant bacteria is of vital importance. Among them, silver nanoparticles (Ag NPs) have shown an antibacterial effect; however, the remainder cores of Ag NPs after use might have a toxic effect on humans. Thus, Ag ions based materials have been fabricated to substitute Ag NPs for antibacterial applications. Nevertheless, the always‐on release state leads to the low biocompatibility, which limits their biomedical applications. In addition, the single effect also restricts their antibacterial ability. Herein, a powerful surface‐adaptive, on‐demand antimicrobial nanoplatform is fabricated by coating hyaluronic acid (HA) on Ag ions loaded photosensitive metal‐organic frameworks to exhibit a strong synergistic effect. The nanoplatform shows good biocompatibility with nontargeted cells, as negatively charged HA can prevent the release of Ag ions. While in the presence of targeted bacteria, the secreted hyaluronidase can degrade HA on the nanoplatform and produce positively charged nanoparticles, which display increased affinity to bacteria and show a strong synergistic antibacterial effect owing to the released Ag ions and generated reactive oxygen species under visible light irradiation. Importantly, due to the outstanding on‐demand antimicrobial performance, the nanoplatform also shows great effects on treating multidrug‐resistant bacteria infected wounds in mice models.
Synthesis of highly stable luminescent silver nanoclusters in metal–organic framework for heightened antibacterial activity

Abstract:Size of the nanoparticles plays a very crucial role in their bactericidal activity as smaller size render better perforation via bacterial cell wall and are thus found to be more effective against resistant bacterial infections also. In this regard, fluorescent silver nanoclusters (Ag NCs) having a size less than 2 nm ought to be a potent bactericidal agent against both Gram positive and Gram negative bacterial strains. However, stabilization of such small sized NPs in aqueous medium that is amenable for biological applications is very challenging. Herein, a simple and fast method of synthesis of Ag NCs was proposed using metal–organic framework (MOFs) as scaffold, which was found to be very stable in the ambient conditions. As-synthesized Ag NCs-in-MOF nanocomposite was characterized by UV–Vis spectroscopy, transmission electron microscopy (TEM), and X-ray photoelectron spectroscopy (XPS). The nanocomposite was highly bactericidal against both Gram positive and Gram negative bacterial strains and caused cell wall perforation that was evident from flow cytometry analysis. Results also showed that DNA damage had occurred due to treatment of Ag NCs-in-MOF, ultimately leading to bacterial cell death with considerably lower dose of silver.

Abstract:A novel composite material named sliver nanoparticles@carbon dots (AgNPs/CDs) was fabricated with CDs as the capping agents through one simple method. The images of TEM showed that CDs were embedded in the gaps of AgNPs and the as-prepared AgNPs/CDs can be self assembled into nanochains through electrosatic interaction between silver nanoparticles and carbon dots. With higher redox properties than AgNPs, the as-prepared com-posites exhibited excellent synergistic antibacterial performance against Gram-positive bacterium and Gram-negative bacterium in the antibacterial assays, which indicates potential application as an effective antibacte-rial material. And the results of antibacterial assy showed that the minimum inhibitory concentrations (MIC) of AgNPs/CDs were 40 mu g/mL for E. coli and 20 mu g/mL for S. aureus in LB medium. In addition, the oxidative stress induced by AgNPs/CDs-mediated reactive oxygen species further confirmed good application prospects in antibacterial.
