Abstract:Areas V1 and V2 of Macaque monkey visual cortex are characterized by unique cytochrome-oxidase (CO)-staining patterns. Initial electrophysiological studies associated CO blobs in V1 with processing of surface properties such as color and brightness and the interblobs with contour information processing. However, many subsequent studies showed controversial results, some supporting this proposal and others failing to find significant functional differences between blobs and interblobs. In this study, we have used optical imaging to map color-selective responses in V1 and V2. In V1, we find striking "blob-like" patterns of color response. Fine alignment of optical maps and CO-stained tissue revealed that color domains in V1 strongly associate with CO blobs. We also find color domains in V1 align along centers of ocular dominance columns. Furthermore, color blobs in V1 have low orientation selectivity and do not overlap with centers of orientation domains. In V2, color domains coincide with thin stripes; orientation-selective domains coincide with thick and pale stripes. We conclude that color and orientation-selective responses are preferentially located in distinct CO compartments in V1 and V2. We propose that the term "blob" encompasses both the concept of "CO blob" and "color domain" in V1.

Abstract:Because at least some squirrel monkeys lack ocular dominance columns (ODCs) in the striate cortex (V1) that are detectable by cytochrome oxidase (CO) histochemistry, the functional importance of ODCs on stereoscopic 3-D vision has been questioned. However, conventional CO histochemistry or trans-synaptic tracer study has limited capacity to reveal cortical functional architecture, whereas the expression of immediate-early genes (IEGs), c-FOS and ZIF268, is more directly responsive to neuronal activity of cortical neurons to demonstrate ocular dominance (OD)-related domains in V1 following monocular inactivation. Thus, we wondered whether IEG expression would reveal ODCs in the squirrel monkey V1. In this study, we first examined CO histochemistry in V1 of five squirrel monkeys that were subjected to monocular enucleation or tetrodotoxin (TTX) treatment to address whether there is substantial cross-individual variation as reported previously. Then, we examined the IEG expression of the same V1 tissue to address whether OD-related domains are revealed. As a result, staining patterns of CO histochemistry were relatively homogeneous throughout layer 4 of V1. IEG expression was also moderate and homogeneous throughout layer 4 of V1 in all cases. On the other hand, the IEG expression was patchy in accordance with CO blobs outside layer 4, particularly in infragranular layers, although they may not directly represent OD clusters. Squirrel monkeys remain an exceptional species among anthropoid primates with regard to OD organization, and thus are potentially good subjects to study the development and function of ODCs.
Identification of Ocular Dominance Domains in New World Owl Monkeys by Immediate-Early Gene Expression

Abstract:Ocular dominance columns (ODCs) have been well studied in the striate cortex (V1) of macaques, as well defined arrays of columnar structure that receive inputs from one eye or the other, whereas ODC expression seems more obscure in some New World primate species. ODCs have been identified by means of eye injections of transneuronal transporters and examination of cytochrome oxidase (CO) activity patterns after monocular enucleation. More recently, live-imaging techniques have been used to reveal ODCs. Here, we used the expression of immediate-early genes (IEGs), protooncogene, c-Fos, and zinc finger protein, Zif268, after monocular inactivation (MI) to identify ODCs in V1 of New World owl monkeys. Because IEG expression is more sensitive to activity changes than CO expression, it is capable of revealing activity maps in all layers throughout V1 and demonstrating brief activity changes within a couple of hours. Using IEGs, we not only revealed apparent ODCs in owl monkeys but also discovered a number of unique features of their ODCs. Distinct from those in macaques, these ODCs sometimes bridged to other columns in layer 4 (Brodmann layer 4C). CO blobs straddled ODC borders in the central visual field, whereas they centered ODC patches in the peripheral visual field. In one case, the ODC pattern continued into V2. Finally, an elevation of IEG expression in layer 4 (4C) was observed along ODC borders after only brief MI. Our data provide insights into the structure and variability of ODCs in primates and revive debate over the functions and development of ODCs.
Multiple Visuotopically Organized Subdivisions of the Lateral Pulvinar/Central Lateral Inferior Pulvinar Project into Thin and Thick Stripe Compartments of V2 in Macaques.

Abstract:The lateral and central lateral inferior pulvinar (PL/PIcl) of primates has been implicated in playing an important role in visual processing, but its physiological and anatomical characteristics remain to be elucidated. It has been suggested that there are two complete visuotopic maps in the PL/PIcl, each of which sends afferents into V2 and V4 in primates. Given that functionally distinct thin and thick stripes of V2 both receive inputs from the PL/PIcl, this raises the possibility of a presence of parallel segregated pathways within the PL/PIcl. To address this question, we selectively injected three types of retrograde tracers (CTB-488, CTB-555, and BDA) into thin or thick stripes in V2 and examined labeling in the PL/PIcl in macaques. As a result, we found that every cluster of retrograde labeling in the PL/PIcl included all three types of signals next to each other, suggesting that thin stripe- and thick stripe-projecting compartments are not segregated into domains. Unexpectedly, we found at least five topographically organized retrograde labeling clusters in the PL/PIcl, indicating the presence of more than two V2-projecting maps. Our results suggest that the PL/PIcl exhibits greater compartmentalization than previously thought. They may be functionally similar but participate in multiple cortico-pulvinar-cortical loops.
Specificity of Color Connectivity Between Primate V1 and V2.

Abstract:To examine the functional interactions between the color and form pathways in the primate visual cortex, we have examined the functional connectivity between pairs of color oriented and nonoriented V1 and V2 neurons in Macaque monkeys. Optical imaging maps for color selectivity, orientation preference, and ocular dominance were used to identify specific functional compartments within V1 and V2 (blobs and thin stripes). These sites then were targeted with multiple electrodes, single neurons isolated, and their receptive fields characterized for orientation selectivity and color selectivity. Functional interactions between pairs of V1 and V2 neurons were inferred by cross-correlation analysis of spike firing. Three types of color interactions were studied: nonoriented V1/nonoriented V2 cell pairs, nonoriented V1/oriented V2 cell pairs, and oriented V1/nonoriented V2 cell pairs. In general, interactions between V1 and V2 neurons are highly dependent on color matching. Different cell pairs exhibited differing dependencies on spatial overlap. Interactions between nonoriented color cells in V1 and V2 are dependent on color matching but not on receptive field overlap, suggesting a role for these interactions in coding of color surfaces. In contrast, interactions between nonoriented V1 and oriented V2 color cells exhibit a strong dependency on receptive field overlap, suggesting a separate pathway for processing of color contour information. Yet another pattern of connectivity was observed between oriented V1 and nonoriented V2 cells; these cells exhibited interactions only when receptive fields were far apart and failed to interact when spatially overlapped. Such interactions may underlie the induction of color and brightness percepts from border contrasts. Our findings thus suggest the presence of separate color pathways between V1 and V2, each with differing patterns of convergence and divergence and distinct roles in color and form vision.
Perceptual Color Map In Macaque Visual Area V4

Abstract:Colors distinguishable with trichromatic vision can be defined by a 3D color space, such as red-green-blue or hue-saturation-lightness (HSL) space, but it remains unclear how the cortex represents colors along these dimensions. Using intrinsic optical imaging and electrophysiology, and systematically choosing color stimuli from HSL coordinates, we examined how perceptual colors are mapped in visual area V4 in behaving macaques. We show that any color activates 1-4 separate cortical patches within "globs," millimeter-sized color-preferring modules. Most patches belong to different hue or lightness clusters, in which sequential representations follow the color order in HSL space. Some patches overlap greatly with those of related colors, forming stacks, possibly representing invariable features, whereas few seem positioned irregularly. However, for any color, saturation increases the activity of all its patches. These results reveal how the color map in V4 is organized along the framework of the perceptual HSL space, whereupon different multipatch activity patterns represent different colors. Wepropose that such distributed and combinatorial representations may expand the encodable color space of small cortical maps and facilitate binding color information to other image features.

Abstract:Visual area V4 in the macaque monkey is a cortical area that is strongly involved in color and shape perception. However, fundamental questions about V4 are still debated. V4 was initially characterized as a color-processing area, but subsequent studies revealed that it contains a diverse complement of cells, including those with preference for color, orientation, disparity and higher-order feature preferences. This has led to disputes and uncertainty about the role of V4 in vision. Using intrinsic signal optical imaging methods in awake, behaving monkeys, we found that different feature preferences are functionally organized in V4. Optical images revealed that regions with preferential response to color were largely separate from orientation-selective regions. Our results help to resolve long-standing controversies regarding functional diversity and retinotopy in V4 and indicate the presence of spatially biased distribution of featural representation in V4 in the ventral visual pathway.

Abstract:By combining optical imaging, single unit electrophysiology and cytochrome oxidase (CO) histology, we sought to reveal in greater detail the functional organization within the CO stripes of visual area V2 of primates. To visualize the disparity selective regions of V2, the imaging of binocular interaction was employed. These imaging maps guided single unit penetrations that then revealed a columnar organization for disparity. Our studies also showed a pattern of intermixing between the color and disparity pathways of V2, including the existence of single cells tuned for both color and disparity. While previous studies have suggested that the CO stripes of V2 constitute the fundamental organizational unit within V2, our results show a further level of organization consisting of functionally distinct subcompartments, 0.7–1.5 mm in diameter, within individual stripes. These subcompartments, which are not clearly revealed by CO histochemistry, lie within each of the thin, pale, and thick CO dense stripes in V2 and are specific for aspects of color, orientation and retinal disparity, respectively. The present results favor an architectural view of V2, not unlike that of V1, as a collection of functionally distinct subcompartments or modules situated within each of the V2 stripes. These modules also support the notion that for each cortical area (e.g. V1, V2, V4), there exists a stereotyped cortical module with a geometry that is characteristic for each area. These modules exist as a middle tier in a hierarchy of functional organization within V2.
Organization of hue selectivity in macaque V2 thin stripes

Abstract:V2 has long been recognized to contain functionally distinguishable compartments that are correlated with the stripelike pattern of cytochrome oxidase activity. Early electrophysiological studies suggested that color, direction/disparity, and orientation selectivity were largely segregated in the thin, thick, and interstripes, respectively. Subsequent studies revealed a greater degree of homogeneity in the distribution of response properties across stripes, yet color-selective cells were still found to be most prevalent in the thin stripes. Optical recording studies have demonstrated that thin stripes contain both color-preferring and luminance-preferring modules. These thin stripe color-preferring modules contain spatially organized hue maps, whereas the luminance-preferring modules contain spatially organized luminance-change maps. In this study, the neuronal basis of these hue maps was determined by characterizing the selectivity of neurons for isoluminant hues in multiple penetrations within previously characterized V2 thin stripe hue maps. The results indicate that neurons within the superficial layers of V2 thin stripe hue maps are organized into columns whose aggregated hue selectivity is closely related to the hue selectivity of the optically defined hue maps. These data suggest that thin stripes contain hue maps not simply because of their moderate percentage of hue-selective neurons, but because of the columnar and tangential organization of hue selectivity.
Optical imaging of contrast response in functional domains in V1 and V2 of macaque visual cortex

Abstract:Our recent optical imaging and single unit studies in visual cortex suggested a preferential involvement of V1 blobs and V2 thin stripes in real and illusory brightness response (Hung et al. Soc Neurosci 2002; cf. Hung et al Vision Res 2001). To further explore this possibility and to better relate full field brightness response to standard contrast sensitivity responses, we have measured contrast response of different functional domains in V1 and V2 with optical imaging and electrophysiological recording. Visual stimuli consisted of drifting horizontal and vertical luminance sine wave gratings with different spatial frequencies, ranging from 0.84 — 7.6 cyc/deg. Luminance contrast of the gratings was varied over four different levels (10%, 20%, 40%, 80%). Intrinsic signals were imaged from area V1 and V2 of anesthetized macaque monkeys. Average reflectance values from functional domains (orientation domains in V1 and V2, blobs in V1, and thin and thick/pale stripes in V2) were obtained and were also compared to single cell responses sampled from superficial layers of imaged domains. Results showed that, as expected, cortical reflectance values changed systematically with the contrast level of the stimulus: the greater the contrast, the greater the response. As measured by reflectance values, we find that, at low spatial frequencies and low contrast levels, V1 blobs have greater response than V1 interblobs. In comparison to V1 domains, V2 domains exhibit higher contrast gain and saturate at a low-mid contrast levels. V2 thin stripes also showed higher contrast gain than thick/pale stripes. Functional Subdivisions in Macaque V4 Revealed by Optical Imaging in the Behaving Macaque Monkey

Abstract:In macaque V4, it has been proposed that there are several subdivisions based on patterns of connectivity and visual topography (Zeki, 1971; Maguire and Baizer, 1984). Zeki (1983) reported that color selective cells were more frequently found in the anterior bank of lunate sulcus (V4 proper), as compared with the cortex on the prelunate gyrus (V4A). However, this finding was based on qualitative observation and there has been no study to confirm this finding. Here, we studied the functional organization of V4 using intrinsic signal optical imaging in two alert macaque monkeys and examined whether there is any functional subdivision in V4 in relation to color, luminance, and orientation preferences. Intrinsic signals were obtained through a chronic chamber over the portion of V4 representing foveal and parafoveal visual space ([[lt]]1.5° and 3–7° eccentricity, respectively). To reveal feature representation, a patch (0.25–8°) or full-screen stimulus filled with isoluminant color-varying or achromatic luminance-varying gratings was presented while the monkey was performing a fixation task. Visual activations in V4 showed a stimulus-size dependence: small grating patches activated the cortex more strongly than the full-screen gratings, consistent with the presence of strong surround suppression. In the foveal V4, color/luminance and orientation mapped in the different cortical locations. Color and luminance-sensitive regions were located posteriorly, close to the lunate sulcus (within 2 mm), alternating with a band-like appearance (color/luminance domains). The width of these domains was approximately 0.5 mm. In a region anterior to these domains, orientation-sensitive regions were observed (orientation domains). These two types of domains were largely separated but with some overlap. Within the color/luminance domains, different colors activated separate sub-domains. This functional compartmentalization was also observed in the parafoveal V4. These results indicate that there are distinct functional subdivisions in V4, which appear to correspond to the subdivisions proposed previously (V4 proper/V4A).

Abstract:Our studies on brightness information processing in Macaque monkey visual cortex suggest that the thin stripes in the secondary visual area (V2) are preferentially activated by brightness stimuli (such as full field luminance modulation and illusory edge-induced brightness modulation). To further examine this possibility, we used intrinsic signal optical imaging to examine contrast response of different functional domains in primary and secondary visual areas (V1 and V2). Color and orientation stimuli were used to map functional domains in V1 (color domains, orientation domains) and V2 (thin stripes, thick/pale stripes). To examine contrast response, sinusoidal gratings at different contrasts and spatial frequencies were presented. We find that, consistent with previous studies, the optical signal increased systematically with contrast level. Unlike single-unit responses, optical signals for both color domains and orientation domains in V1 exhibit linear contrast response functions, thereby providing a large dynamic range for V1 contrast response. In contrast to domains in V1, domains in V2 exhibit nonlinear responses, characterized by high gain at low contrasts, saturating at a mid-high contrast levels. At high contrasts, thin stripes exhibit increasing response, whereas thick/pale stripes saturate, consistent with a strong parvocellular input to thin stripes. These findings suggest that, with respect to contrast encoding, thin stripes have a larger dynamic range than thick/pale stripes and further support a role for thin stripes in processing of brightness information.
The Organization of Orientation-Selective, Luminance-Change and Binocular- Preference Domains in the Second (V2) and Third (V3) Visual Areas of New World Owl Monkeys As Revealed by Intrinsic Signal Optical Imaging

Abstract:Optical imaging was used to map patterns of visually evoked activation in the second (V2) and third (V3) visual areas of owl monkeys. Modular patterns of activation were produced in response to stimulation with oriented gratings, binocular versus monocular stimulation, and stimuli containing wide-field luminance changes. In V2, luminance-change domains tended to lie between domains selective for orientation. Regions preferentially activated by binocular stimulation co-registered with orientation-selective domains. Co-alignment of images with cytochrome oxidase (CO)-processed sections revealed functional correlates of 2 types of CO-dense regions in V2. Orientation-responsive domains and binocular domains were correlated with the locations of CO-thick stripes, and luminance-change domains were correlated with the locations of CO-thin stripes. In V3, orientation preference, luminance-change, and binocular preference domains were observed, but were more irregularly arranged than those in V2. Our data suggest that in owl monkey V2, consistent with that in macaque monkeys, modules for processing contours and binocularity exist in one type of compartment and that modules related to processing-surface features exist within a separate type of compartment.
The Representation of Colored Objects in Macaque Color Patches

Abstract:An important question about color vision is how does the brain represent the color of an object? The recent discovery of "color patches" in macaque inferotemporal (IT) cortex, the part of the brain responsible for object recognition, makes this problem experimentally tractable. Here we recorded neurons in three color patches, middle color patch CLC (central lateral color patch), and two anterior color patches ALC (anterior lateral color patch) and AMC (anterior medial color patch), while presenting images of objects systematically varied in hue. We found that all three patches contain high concentrations of hue-selective cells, and that the three patches use distinct computational strategies to represent colored objects: while all three patches multiplex hue and shape information, shape-invariant hue information is much stronger in anterior color patches ALC/AMC than CLC. Furthermore, hue and object shape specifically for primate faces/bodies are over-represented in AMC, but not in the other two patches.
Hierarchical Representation for Chromatic Processing Across Macaque V1, V2, and V4

Abstract:The perception of color is an internal label for the inferred spectral reflectance of visible surfaces. To study how spectral representation is transformed through modular subsystems of successive cortical areas, we undertook simultaneous optical imaging of intrinsic signals in macaque V1, V2, and V4, supplemented by higher-resolution electrophysiology and two-photon imaging in awake macaques. We find a progressive evolution in the scale and precision of chromotopic maps, expressed by a uniform blob-like architecture of hue responses within each area. Two-photon imaging reveals enhanced hue-specific cell clustering in V2 compared with V1. A phenomenon of endspectral (red and blue) responses that is clear in V1, recedes in V2, and is virtually absent in V4. The increase in mid- and extra-spectral hue representations through V2 and V4 reflects the nature of hierarchical processing as higher areas read out locations in chromatic space from progressive integration of signals relayed by V1.
Representation of Cone-Opponent Color Space in Macaque Early Visual Cortices

Abstract:In primate vision, the encoding of color perception arises from three types of retinal cone cells (L, M, and S cones). The inputs from these cones are linearly integrated into two cone-opponent channels (cardinal axes) before the lateral geniculate nucleus. In subsequent visual cortical stages, color-preferring neurons cluster into functional domains within “blobs” in V1, “thin/color stripes” in V2, and “color bands” in V4. Here, we hypothesize that, with increasing cortical hierarchy, the functional organization of hue representation becomes more balanced and less dependent on cone opponency. To address this question, we used intrinsic signal optical imaging in macaque V1, V2, and V4 cortices to examine the domain-based representation of specific hues (here referred to as “hue domains”) in cone-opponent color space (4 cardinal and 4 intermediate hues). Interestingly, we found that in V1, the relative size of S-cone hue preference domain was significantly smaller than that for other hues. This notable difference was less prominent in V2, and, in V4 was virtually absent, resulting in a more balanced representation of hues. In V2, hue clusters contained sequences of shifting preference, while in V4 the organization of hue clusters was more complex. Pattern classification analysis of these hue maps showed that accuracy of hue classification improved from V1 to V2 to V4. These results suggest that hue representation by domains in the early cortical hierarchy reflects a transformation away from cone-opponency and toward a full-coverage representation of hue.
Functional Architecture of the Foveal Confluence in Macaque Visual Cortex

Abstract:Visual behavior in primates is dominated by moving the eyes (more specifically the fovea) to objects of interest. Representing the most central part of visual space, the fovea is crucial for providing high spatial acuity needed for tasks such as reading, form identification, face recognition, and is key to natural visual attentional behavior. In early visual cortex, cortical representation disproportionately represents the fovea. However, beyond this, surprisingly little is known about its cortical representation (e.g. neurophysiological data comes primarily from extra-foveal cortex). We hypothesize that, given the known specialized functions performed by the fovea, there should be differences in foveal vs extra-foveal cortical organization. This thinking is based on studies which suggest that the unique functional organizations present in V1, V2, and V4 enable different integrative capabilities within those areas, thereby resulting in area-specific functionalities. To examine this hypothesis, we have conducted optical imaging in the region of the foveal confluence in awake and anesthetized macaque monkeys. We predict differences in the organization (either qualitative or quantitative) of ocular dominance, orientation, color, disparity, and motion in foveal vs extra-foveal cortex. We consider the possibility that at the foveal confluence there are no clear borders (i.e. retintopic reversals) between cortical areas and that the foveal confluence is structured as a single specialized cortical region shared by V1, V2, and V4. Also, given the lack of blue cone representation in the foveal retina, we examine whether there is blue color representation in foveal V1, V2, and V4. It may also be possible to address the question of whether V3 extends to the foveal confluence. Thus far, our preliminary data suggest that ocular dominance columns may be less regularly organized and wider in foveal regions of V1 and that there may be a lack of strong representation of blue in the foveal confluence. Abstract:Previous studies have revealed modular projections from area V2 to area V4 in macaques. Specifically, V2 neurons in cytochrome oxidase (CO)-rich thin and CO-sparse pale stripes project to distinct regions in V4. However, how these modular projections relate to the functional subcompartments of V4 remains unclear. In this study, we injected retrograde fluorescent tracers into V4 regions with different functional properties (color, orientation, and direction) that were identified with intrinsic signal optical imaging (ISOI). We examined the labeled neurons in area V2 and their locations with respect to the CO patterns. Covariation was observed between the functional properties of the V4 injection sites and the numbers of labeled neurons in particular CO stripes. This covariation indicates that the color domains in V4 mainly received inputs from thin stripes in V2, whereas V4 orientation domains received inputs from pale stripes. Although motion-sensitive domains are present in both V2 and V4, our results did not reveal a functional specific feedforward projection between them. These results confirmed previous findings of modular projections from V2 to V4 and provided functional specificity for these anatomical projections. Together, these findings indicate that color and form remain separate in ventral mid-level visual processing.

Abstract:V1 neurons in primates respond to binocular stimulation, but with various degrees of eye preferences. We used two-photon calcium imaging (GCaMP5) to study a) ocularity-contingent monocular and binocular responses of macaque V1 superficial neurons and their contributions to stable perception when the world is viewed with one or both eyes open; b) functional organization of ocularity and its relationships to orientation and SF maps. The stimulus was a high-contrast (0.9) drafting (2 cycles/sec) Gabor of 12 orientations (in 15° steps) and 6 SFs (0.25-8 cpd), and was presented binocularly or monocularly to each eye. Data analysis identified a total of 7335 neurons that were orientation selective with at least one eye. Most V1 superficial-layer neurons responded to stimulations from both eyes, and only a few were strictly monocular. When monocularly stimulated, the best responses of more monocular neurons were markedly stronger than those of more binocular neurons, approximately in the form of a quadratic function. However, stronger responses of monocular neurons were suppressed, and weaker responses of binocular neurons were enhanced by binocular stimulation. As a result, the net population responses remain little changed, which may be related to the homeostasis of perception under monocular and binocular viewing. The monocular and binocular response changes can be described with a binocular integration and gain control modal, which also predicts binocular summation at low stimulus contrasts. V1 neurons clustered with similar ocularity index within the same recording plane. The median clustering index was 1.87, significantly higher than permuted data. Neurons also formed ocular dominance columns across depth planes. Neuron-wise, ocularity, SF, and orientation functional maps intersected each other in a wide range of angles, with the median around 40°. The vector sums of individual angles were near 0°, but with large circular variances (>0.8), suggesting no reliable relationships among these functional maps.
