Abstract:Ferroptosis is highly associated with reactive oxygen species (ROS), especially the hydroxyl radical (•OH) that induces lipid peroxide accumulation, which may have significant effect on polarity changes, but with no clear evidence of the relationship. Therefore, it is vital and challenging to monitor polarity and •OH simultaneously for further exploration of the mechanisms of ferroptosis. Therefore, taking advantage of a structural modulation strategy to optimize the probe molecule stepwise, dual-site fluorescent sensor ( ZY-5 ) was developed to meet the need of analyzing polarity and •OH changes independently with little crosstalk. With excellent sensitivity and selectivity, ZY-5 could accomplish the quantitative analysis for polarity and •OH in their own channels rapidly. And the variations of exogenous and endogenous polarity and •OH in cells can be monitored through different fluorescence channels by ZY-5 . More significantly, with the appealing imaging potential of ZY-5 , the relationship between polarity and •OH during different ferroptosis-induced pathways were revealed preliminarily and it uncovered the differences in redox and the homeostatic regulations of the microenvironment.
