The long-term negative effects of early life stress on SGK1 modulation: the key role of epigenetic mechanisms

Abstract:BACKGROUND Serum- and Glucocorticoid-inducible Kinase 1 (SGK1) is a serine/threonine kinase implicated in cellular stress response and neuronal functions. It has been proposed as a vulnerability gene for depression, especially in association with early life stress (ELS). Here, we aim characterizing the molecular mechanisms underlying the ELS-induced upregulation of SGK1, focusing on epigenetics. METHODS We measured SGK1 methylation levels in the hippocampus of prenatal stress (PNS) rats and in peripheral blood of depressed subjects characterized for childhood trauma (CT). We analyzed by Real-Time PCR the modulation of several miRNAs targeting SGK1 (miR-19a; miR-19b; miR-20b; miR-21; miR-28; miR-29a; miR-29b; miR- 29c) in the same PNS animals and cohort of depressed patients and controls. The expression levels of these miRNAs were also measured in human hippocampal progenitor cells (HIP009, PhoenixSongs Biologicals) treated with cortisol or vehicle. RESULTS We observed an increased methylation status in SGK1 locus both in PNS animals and in depressed subjects in association with ELS. Among the selected miRNAs, we found decreased levels of miR-19a, miR-19b, miR-29c and miR-28 (p<0.01) in the hippocampus of PNS rats characterized for vulnerability or resilience and a trend of downregulation, although not significant, in subjects exposed to CT. Interestingly, cortisol modulated the levels of several miRNAs (p<0.05) in the in vitro model. CONCLUSION The long-lasting expression alterations of SGK1 are associated with changes in miRNAs rather than in DNA methylation. Treatments acting on these miRNAs may prevent the SGK1 ELS-related vulnerability.
