Abstract:In this work, an amperometric L-lactate biosensor based on a graphene-modified screen-printed carbon electrode (SPCE) was constructed. First, the electrocatalytic performance of the SPCE modified with graphene by a one-step electrodeposition process (OerGO/SPCE) was investigated. The cyclic voltammogram of OerGO/SPCE, which showed a well-defined redox peak, had a smaller peak potential separation than that of SPCE, revealing the improvement in electron transfer speed brought about by modifying with graphene. Next, lactate oxidase and potassium ferricyanide were dropped on the OerGO/SPCE to construct a graphene-modified L-lactate biosensor (LOD/K3[Fe(CN)6]/OerGO/SPCE). The proposed biosensor, with a detection limit of 60 04, had a high sensitivity (42.42 mu A mM(-1) cm(-2)) when working at a low working potential (0.15V). The linear range was 0.5 mM-15mM, covering the detecting range of L-lactate in clinical applications. The L-lactate biosensor had a short response time (10s) and required only 10 mu l of the sample. This L-lactate sensor modified with electrodeposited graphene had a larger sensitivity than that based on the bare SPCE. Thus, our low-cost and disposable L-lactate biosensor enhanced by graphene can perform as an attractive electrochemical device that can be manufactured for point-of-care testing (POCT) devices and be employed in POCT applications.

Abstract:This paper deals with recent progress in the use of laser-induced graphene sensors for the electrochemical detection of glucose molecules. The exponential increase in the exploitation of the laser induction technique to generate porous graphene from polymeric and other naturally occurring materials has provided a podium for researchers to fabricate flexible sensors with high dynamicity. These sensors have been employed largely for electrochemical applications due to their distinct advantages like high customization in their structural dimensions, enhanced characteristics and easy roll-to-roll production. These laser-induced graphene (LIG)-based sensors have been employed for a wide range of sensorial applications, including detection of ions at varying concentrations. Among the many pivotal electrochemical uses in the biomedical sector, the use of these prototypes to monitor the concentration of glucose molecules is constantly increasing due to the essentiality of the presence of these molecules at specific concentrations in the human body. This paper shows a categorical classification of the various uses of these sensors based on the type of materials involved in the fabrication of sensors. The first category constitutes examples where the electrodes have been functionalized with various forms of copper and other types of metallic nanomaterials. The second category includes other miscellaneous forms where the use of both pure and composite forms of LIG-based sensors has been shown. Finally, the paper concludes with some of the possible measures that can be taken to enhance the use of this technique to generate optimized sensing prototypes for a wider range of applications.

Laser-Induced Graphene-Based Enzymatic Biosensor for Glucose Detection

Abstract:Laser-induced graphene (LIG) has recently been receiving increasing attention due to its simple fabrication and low cost. This study reports a flexible laser-induced graphene-based electrochemical biosensor fabricated on a polymer substrate by the laser direct engraving process. For this purpose, a 450 nm UV laser was employed to produce a laser-induced graphene electrode (LIGE) on a polyimide substrate. After the laser engraving of LIGE, the chitosan–glucose oxidase (GOx) composite was immobilized on the LIGE surface to develop the biosensor for glucose detection. It was observed that the developed LIGE biosensor exhibited good amperometric responses toward glucose detection over a wide linear range up to 8 mM. The GOx/chitosan-modified LIGE biosensor showed high sensitivity of 43.15 μA mM−1 cm−2 with a detection limit of 0.431 mM. The interference studies performed with some possible interfering compounds such as ascorbic acid, uric acid, and urea exhibited no interference as there was no difference observed in the amperometric glucose detection. It was suggested that the LIGE-based biosensor proposed herein was easy to prepare and could be used for low-cost, rapid, and sensitive/selective glucose detection.

Abstract:<p>Glucose is one of the most important metabolites and a primary nutrient in human body metabolism. Abnormal levels of glucose can cause diabetes mellitus and related complications. Here, we developed a disposable and portable non-enzymatic glucose sensor strip based on gold-nanoparticles (AuNPs)-modified laser-scribed graphene electrode (LSGE). Disposable LSGEs, with a highly porous structure, were fabricated by laser scribing on a commercially available polyimide plastic sheets, followed by electrodepositing of AuNPs, that are electrocatalytic for glucose oxidation. The laser scribing process was optimised in term of laser power, laser speed and pulse per inch (PPI). The effect of number of the electrochemical deposition cycles of AuNPs on LSGE was also investigated and optimised. The sensor provides a linear detection range for the glucose concentrations between 10 µM to 10 mM with a limit of detection of 6.3 µM and shows a good selectivity towards interfering agents, such as urea L-ascorbic acid, uric acid, dopamine, and Na<sup>+</sup>. The LSGE/AuNP strips have a potential to be used as disposable glucose sensor strips for point-of-care glucose measurements, including self-monitoring and nutrition management.</p>

Abstract:Laser-inducing provides a cost-effective, easily-manufacturable, and environment-friendly approach to directly transfer carbon-rich polymers into graphene materials, which attracts attention from various fields, such as sensors, electrocatalysts, micro-supercapacitors, etc. Laser-induced graphene (LIG) benefits from the intrinsic properties of graphene, for example, high conductivity, high electroactivity, and high specific area. In this work the potential of laser-induced graphene in constructing an enzymatic electrochemical biosensing interface is evaluated. Here, a laminar-structured laser-induced graphene material is fabricated by laser engraving with polyimide. After deposition of the electron mediator ferrocene, a conjugated enzyme complex of bovine serum albumin-glucose oxidase (BSA-GOx) is modified on the laser-induced graphene by cross-linking. The fabricated glucose oxidase/ferrocene/LIG (GOx/Fc/LIG) biosensor achieves high sensitivity of 11.3 & mu;A mM-1 cm-2), wide linear range of 0-11 mM, and low detection of limit of 0.04 & mu;M. The LIG electrodes exhibit high flexibility with bending angle as high as 60 & DEG; without observed conductivity change. The repeatability and robustness of the developed LIG biosensor in detection of real serum samples empower it with great potential in clinical implementation in the future. A high sensitivity and robust enzymatic electrochemical biosensing interface based on laser-induced graphene is developed by a combined low-cost fabrication strategy of ultraviolet laser pre-treatment and infrared laser defocused processing in together with conductive organic network-mediated conjugation of enzymes and electron mediators on the 3D porous graphene for enzyme immobilization.image

Abstract:Enzyme-free electrochemical glucose (Glu) sensors, adopting electrochemically active catalysts, are of vital significance to effectively monitoring and analyzing the blood Glu level in diabetes diagnosis. Meanwhile, developing novel electrode substrates have been the hot research pot to meet the demand for constructing flexible sensors. In this work, porous laser-induced graphene (LIG) was prepared through a facile one-step laserengraving technique on a polyimide thin film, and then used directly as the electrode. Furthermore, NiPt alloyed nanoparticles (NPs) were electrodeposited on the LIG electrode surface to gain the NiPt/LIG composite electrode, which was applied to construct the nonenzymatic electrochemical Glu sensor. The morphology, element and electrochemical performance of NiPt/LIG composite was characterized through various techniques. NiPt alloyed NPs were homogeneously distributed on the LIG scaffolds, which possess plenty of porous structure, high surface area and many active sites. The cooperative effect between the outstanding electrocatalytic capacity of NiPt alloyed NPs, the extraordinary electroconductibility of LIG and the numerous exposed active sites endow the NiPt/LIG sensor outstanding electrocatalytic performance for electrochemical oxidation of Glu. Under the most appropriate conditions, the obtained NiPt/LIG sensor was adopted to detect Glu, showing a broad detection concentration range of 0.5 mu M to 2.1 mM and 2.1 mM to 5.6 mM, good sensitivities of 1.824 and 0.467 mu A mu M-1 cm-2, a low detection limit of 0.03 mu M. The as-fabricated nonenzymatic Glu sensor also displays high selectivity, stability considering ageing effects under different environment conditions and prominent reproducibility. Additionally, this novel NiPt/LIG nonenzymatic Glu sensor was applied to quantify Glu successfully in blood serum and food samples and the results were in good agreement with that by UV-vis spectrophotometer, demonstrating high precision and accuracy as well as excellent recovery, showing good application prospects in Glu monitoring. This research confirmed the potential of exploiting NiPt/LIG as a highly sensitive Glu nonenzymatic electrochemical sensor and provided a practicable and suitable approach for construction of novel electrochemical sensors.

Abstract:Non-enzymatic glucose sensors outperform enzymatic ones in terms of cost, sensitivity, stability, and operating duration. Though highly sensitive, it is still desirable to further improve the sensitivity of non-enzymatic glucose sensors to detect a trace amount of glucose in sweat and other biofluids. Among the demonstrated effective approaches using bimetals or 3D porous structures, the porous laser-induced graphene (LIG) on flexible polymers showcases good conductivity and a simple fabrication process for the integration of sensing materials. The uniform electroless plating of the nickel and gold layer on LIG electrodes demonstrates significantly enhanced sensitivity and a large linear range for glucose sensing. The sensor with the porous LIG foam exhibits a high sensitivity of 1080 μA mM−1 cm−2, whereas a further increased sensitivity of 3500 μA mM−1 cm−2 is obtained with LIG fibers (LIGF). Impressively, a large linear range (0 – 30 mM) can be achieved by changing the bias voltage from 0.5 to 0.1 V due to the Au coating. Because the existing non-enzymatic glucose sensors are limited to use in basic solutions, their application in wearable electronics is elusive. In addition to the reduced requirement for the base solution, this work integrates a porous encapsulating reaction cavity containing alkali solutions with a soft, skin-interfaced microfluidic component to provide integrated microfluidic non-enzymatic glucose sensors for sweat sampling and glucose sensing. The accurate glucose measurements from the human sweat and cell culture media showcase the practical utility, which opens up opportunities for the non-enzymatic glucose sensors in wearable electronics.

Abstract:Dependable and sensitive glucose (Glu) testing in foodstuff and blood serum is highly desirable to prevent and treat diabetes. Electrochemical quantification of Glu has attracted great interests due to the advantages, including simple operation, higher sensitivity, easy miniaturization, ease of on-site and wearable detection as well as fast response. High costs and environmental dependence of enzymes pose a challenge to the electrochemical enzymatic biosensors. Nonenzymatic electrochemical Glu sensors are urgently needed to aid the Glu detection in human serum and food samples. To fabricate flexible Glu electrochemical sensors, designing suitable electrode substrate and efficient electrocatalyst is of paramount significance. Herein, the porous patterned laser-induced graphene (LIG) was fabricated on polyimide substrates through an efficient laser-inducing technology, and then used directly as the electrode substrate. Electrochemical deposition of NiCo layered double hydroxide (LDH) nanoflakes on the LIG surface was then conducted to achieve NiCo-LDH/LIG electrode as a Glu sensor. Under optimal conditions, this sensor displays a low detection limit of 0.05 μM. Two sets of broad detection linear ranges were found to be from 0.5 to 270 μM and from 0.27 to 3.6 mM, with high sensitivities of 9.750 μA μM -1 cm -2 and 3.760 μA μM -1 cm -2 , respectively. The enhanced performance was ascribed to the cooperative action of NiCo-LDH and LIG, in which porous LIG provides extraordinary electroconductibility and a high surface area, while NiCo-LDH offers numerous exposed active sites and outstanding electrocatalytic performance. Practical application was further verified during the Glu detection in human serum and food samples. This research confirms that the NiCo-LDH/LIG composite is a prospective electrode for high-performance Glu sensor and provides a way of developing nonenzymatic electrochemical sensors to analyze the Glu in human serum and food samples, opening new avenues in electrochemical sensing.

Abstract:Laser-induced graphene (LIG) has been applied in many different sensing devices, from mechanical sensors to biochemical sensors. In particular, LIG fabricated on paper (PaperLIG) shows great promise for preparing cheap, flexible, and disposable biosensors. Distinct from the fabrication of LIG on polyimide, a two-step process is used for the fabrication of PaperLIG. In this study, firstly, a highly conductive PaperLIG is fabricated. Further characterization of PaperLIG confirmed that it was suitable for developing biosensors. Subsequently, the PaperLIG was used to construct a biosensor by immobilizing glucose oxidase, aminoferrocene, and Nafion on the surface. The developed glucose biosensor could be operated at a low applied potential (−90 mV) for amperometric measurements. The as-prepared biosensor demonstrated a limit of detection of (50–75 μM) and a linear range from 100 μM to 3 mM. The influence of the concentration of the Nafion casting solution on the performance of the developed biosensor was also investigated. Potential interfering species in saliva did not have a noticeable effect on the detection of glucose. Based on the experimental results, the simple-to-prepare PaperLIG-based saliva glucose biosensor shows great promise for application in future diabetes management.

Abstract:Designing and fabricating a highly sensitive non-enzymatic glucose sensor is crucial for the early detection and management of diabetes. Meanwhile, the development of innovative electrode substrates has become a key focus for addressing the growing demand for constructing flexible sensors. Here, a simple one-step laser engraving method is applied for preparing laser-induced graphene (LIG) on polyimide (PI) film, which serves as the sensor substrate. NiCo-layered double hydroxides (NiCo-LDH) are synthesized on LIG as a precursor, utilizing the zeolitic imidazolate framework (ZIF-67), and then reacted with Ni(NO3)2 via solvent-thermal methods. The sensitivity of the non-enzymatic electrochemical glucose sensor is significantly improved by employing NiCo-LDH/LIG as the sensing material. The porous and interconnected structure of NiCo-LDH, derived from ZIF-67, enhances the accessibility of electrochemically active sites, while the incorporation of LIG ensures exceptional conductivity. The combination of NiCo-LDH with LIG enables efficient electron transport, leading to an increased electrochemically active surface area and enhanced catalytic efficiency. The fabricated electrode achieves a low glucose detection limit of 0.437 μM and demonstrates a high sensitivity of 1141.2 and 631.1 μA mM−2 cm−2 within the linear ranges of 0–770 μM and 770–1970 μM, respectively. Furthermore, the NiCo-LDH/LIG glucose sensor demonstrates superior reliability and little impact from other substances. A flexible integrated LIG-based non-enzymatic glucose sensor has been developed, demonstrating high sensitivity and suggesting a promising application for LIG-based chemical sensors.

Abstract:Accurate and convenient detection of human glucose level is an effective method for early diagnosis of diabetes and prevention of complications. The flexible electrochemical glucose electrode with low cost, fast responsiveness, good stability, reliability, and high sensitivity has attracted much attention in the monitoring of glucose concentration. The preparation of a conductive layer with catalytic activity on a flexible substrate is the key to make a flexible glucose electrode. Here, we have successfully prepared graphene composite materials sintered with copper nanoparticles on a polyimide film by direct laser writing, and fabricated a flexible non-enzymatic glucose electrode using laser-ablated graphene (LAG) as a conductive electrode. The Cu/LAG electrode exhibits high catalytic activity for glucose. The glucose electrode shows a linear relationship with the sensitivity of 0.708 mA mM-1 cm-2 in 0.05-1.5 mM. In addition, The Cu/LAG electrode also exhibits excellent response time(~2s). The electrode has good application potential and can be used to monitor glucose in human sweat.

Abstract:Laser-induced graphene (LIG) has emerged as a promising technique for fabricating porous graphene electrodes for many applications, including biosensors and medical devices. However, improving electrochemical properties of LIG hinges on not only understanding the kinetics and energetics governing the physicochemical process of lasing, but also the tunability of surface chemistry and morphology of the produced LIG. This talk reveals the spatiotemporal laser control for achieving different LIG morphology and atomic structures with demonstrations of seamless transitions between conductive networked LIG structures and insulating fibrous LIG morphologies. The effect of laser fluence and multi-pass lasing on LIG morphology and properties will be shown. In addition, molecular engineering of the polymer substrate is explored for controlling the chemical and physical properties of the produced LIG. Finally, the electrochemical detection of neurotransmitters in the nanomolar range will be demonstrated. Our results show that heteroatom doping of graphene is possible by including sulfur or fluorine atoms into the backbone of the polyimide synthesized by two-step polycondensation. Accordingly, both the surface hydrophobicity and sensing sensitivity are shown to depend on the heteroatom-doping content. Taken together, our work paves the way for scalable fabrication of functional graphene-based electrodes and coatings on polymers for flexible and wearable electronics, as well as implantable devices, such as neural probes.
Solid-state, reagent-free and one-step laser-induced synthesis of graphene-supported metal nanocomposites from metal leaves and application to glucose sensing

Abstract:The laser-induced method to prepare three-dimensional (3D) porous graphene has been widely used in many fields owing to its low-cost, easy operation, maskless patterning and ease of mass production. Metal nanoparticles are further introduced on the surface of 3D graphene to enhance its property. The existing methods, however, such as laser irradiation and electrodeposition of metal precursor solution, suffer from many shortcomings, including complicated procedure of metal precursor solution preparation, strict experimental control, and poor adhesion of metal nanoparticles. Herein, a solid-state, reagent-free, and one-step laser-induced strategy has been developed for the fabrication of metal nanoparticle modified-3D porous graphene nanocomposites. Commercial transfer metal leaves were covered on a polyimide film followed by direct laser irradiation to produce 3D graphene nanocomposites modified with metal nanoparticles. The proposed method is versatile and applicable to incorporate various metal nanoparticles including gold silver, platinum, palladium, and copper. Furthermore, the 3D graphene nanocomposites modified with AuAg alloy nanoparticles were successfully synthesized in both 21 Karat (K) and 18K gold leaves. Its electrochemical characterization demonstrated that the synthesized 3D graphene-AuAg alloy nanocomposites exhibited excellent electrocatalytic properties. Finally, we fabricated LIG-AuAg alloy nanocomposites as enzyme-free flexible sensors for glucose detection. The LIG-18K electrodes exhibited the superior glucose sensitivity of 1194 μA mM-1 cm-2 and low detection limits of 0.21 μM. The LIG-21K nanocomposite sensors showed two linear ranges from 1 μM to 1 mM and 2 mM-20 mM with good sensitivity. Furthermore, the flexible glucose sensor showed good stability, sensitivity, and ability to sense in blood plasma samples. The proposed one-step fabrication of reagent-free and metal alloy nanoparticles on LIG with excellent electrochemical performance opens up possibilities for diversifying potential applications of sensing, water treatment and electrocatalysis.
Label-free Glucose Biosensor Based on Enzymatic Graphene Oxide-Functionalized Tilted Fiber Grating

Abstract:A label-free biosensor based on graphene oxide (GO) and glucose oxidase (GOD) functionalized tilted fiber grating (TFG) with large tilted angle is proposed for low concentration glucose detection. Taking advantages of sufficient binding sites of the GO with oxygen-containing groups, the enzymes (GOD) are covalently immobilized onto GO-deposited TFG via 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide and N-hydroxyl succinimide cross-liner. Surface characterizations with optical microscopy, scanning electron microscopy, Raman and infrared spectroscopy provide detailed assessments and evidences about the homogeneity of GO deposition and the effectiveness of enzyme modification. Through the specific catalysis reaction of GOD on the glucose, a considerable refractive index change in local microenvironment around the TFG results in the resonant wavelength shifts of cladding modes. The detection results of the low-concentration glucose demonstrate that the resonant wavelength has a linear response to the glucose concentration in the range of 0-8 mM with a response coefficient of 0.24 nm/mM, showing an enhanced sensitivity and bio-selectivity compared with the pristine TFG. The miniaturized size and remote label-free sensing capacity of the proposed device permit a multitude of opportunities for single-point measurement in harsh conditions and hard-to-reach spaces, presenting a promising candidate for label-free glucose detection for disease diagnosis, pharmaceutical research and bioengineering applications. Fabricating process-electrochemical property correlation of laser-scribed graphene and smartphone-based electrochemical platform for portable and sensitive biosensing

Abstract:Laser-scribed graphene (LSG), a promising electrode material has attracted special research interest in recent years. Here, the fabricating process-electrochemical property correlation of laser-scribed graphene (LSG) devices was discussed emphatically and a pertinent optimization was performed to achieve better electroanalytical performance. Experiment results indicated that the laser scribing technique possessed great process latitude and reducing laser power and scribing speed facilitated fabricating high-quality graphene electrodes. Benefiting from its binder-free 3D porous network structure and high active/geometric area ratio, the optimized LSG electrode was superior to the screen-printed carbon electrode (SPCE) on electrochemical performance in the [Fe(CN)6]3-/4- redox system. Integrating the LSG electrode with a homemade hand-held detector, a portable electrochemical sensing platform with smartphone readout was developed. It realized a specific detection of H2O2 (linear range: 0.02-3.4 mM, sensitivity: 24.56 μA mM-1 cm-2), glucose (linear range: 0.04-4.0 mM, sensitivity: 16.35 μA mM-1 cm-2) by directly decorating biological enzymes without artificial redox mediator and featured a satisfactory comprehensive performance. The constructed immunosensor for tumor necrosis factor-α exhibited a wide linear range (2-500 pg mL-1) and a 4.3-fold enhancement in sensitivity compared with that of SPCE. With satisfactory selectivity, reproducibility, and sensitivity, the developed smartphone-based electrochemical sensing platform held great promise in accurate detection on the spot. This work also provided a significant reference for tailoring binder-free carbonaceous electrode materials toward the desired application.
Flexible Nonenzymatic Glucose Sensing with One‐Step Laser‐Fabricated Cu2O/Cu Porous Structure

Abstract:Integrated flexible nonenzymatic electrochemical glucose sensors have attracted substantial interest because they enable portable noninvasive glucose detection stably and accurately. Manufacturing of these sensors generally requires complex step‐by‐step processes, including synthesis of noble and functional materials, patterning the as‐synthesized noble materials into electrodes/contacts, and subsequent working electrode functionalizing. Herein, the one‐step fabrication of a working electrode and electrical contacts on a flexible substrate for an integrated sensor is realized through laser writing of a low‐cost Cu ionic precursor. The obtained Cu2O/Cu porous structure not only has a low sheet resistance of 1.3 Ω sq−1 to ensure signal transmission in the sensor, but also provides a remarkable sensing performance for glucose detection, especially at a low detection potential of 0.3 V and a low detection limit of 0.34 μm. Due to its anti‐interference and flexibility, the integrated sensor has the potential to directly detect trace amounts of glucose in body fluids as demonstrated in sweat sensing. This work highlights an efficient route to fabricate low‐cost integrated flexible electrochemical sensors.
Graphene Bridged Enzyme Electrodes for Glucose Biosensing Application

Abstract:The fabrication of glucose oxidase (GOx) enzyme electrodes with controlled alternate enzyme and graphene layers is described. GOx was first modified with pyrene functionalities in order to be self-assembled onto a graphene basal plane via non-covalent π-π stacking interaction. Fluorescence spectroscopy analysis revealed that about 5.4 pyrene functional groups were attached to each GOx and the pyrene functionalized GOx retained more than 76% of the biocatalytical activity compared with the native enzyme. Via alternate layer-by-layer self-assembly of graphene and pyrene functionalized GOx, mono- and multi-layered enzyme electrodes with controlled biocatalytical activity can be easily fabricated. The biocatalytical activity of the as-prepared enzyme electrodes increased with increasing graphene and GOx layers and increased insignificantly when the layers reached four. Such multi-layered enzyme electrodes with controlled nanostructure exhibited reliable application in human serum samples analysis with high detection sensitivity, good stability and repeatability. A broad linear detection limit of 0.2 to 40 mM was obtained.

Abstract:Glucose monitoring is of great significance for the health management of diabetic patients. Herein, non-enzymatic glucose sensors were developed by modifying Au@CuO or Au@CuO/V2CTx MXene nanocomposites (NCs) on the surface of disposable laser-induced graphene (LIG) electrode. The structures and morphologies were confirmed by scanning electron microscopy (SEM), transmission electron microscopy (TEM) and elemental analysis. The electrochemical performances of NCs were investigated by electrochemical impedance spectros-copy (EIS), cyclic voltammetry (CV) and chronoamperometry (CA). The results demonstrated that both Au@CuO/LIG and Au@CuO/V2CTx MXene/LIG have excellent electrocatalytic activity for glucose oxidation in an alkaline medium. However, it was found that Au@CuO/V2CTx MXene/LIG was easily oxidized. Hence, the combination of Au@CuO/LIG was preferred for glucose test. Under optimized conditions, the electrochemical sensor achieved a detection range of 0.005 to 5.0 mM with a sensitivity of 1.124 x 106 mu A mM-1 cm-2. The response time was 1.5 s and the limit of detection (LOD) was as low as 1.8 mu M. The sensor has good selectivity and stability (84.12 % activity retained after 6 weeks). Its affordable price, disposable usage and portable essence are suitable for clinical application. Our study presented a novel strategy to the field of non-enzymatic glucose sensing.
