Abstract:A remote control system has been developed to deliver stimuli into the rat brain through a wireless micro-stimulator for animal behavior training. The system consists of the following main components: an integrated PC control program, a transmitter and a receiver based on Bluetooth (BT) modules, a stimulator controlled by C8051 microprocessor, as well as an operant chamber and an eight-arm radial maze. The micro-stimulator is featured with its changeable amplitude of pulse output for both constant-voltage and constant-current mode, which provides an easy way to set the proper suitable stimulation intensity for different training. The system has been used in behavior experiments for monitoring and recording bar-pressing in the operant chamber, contriolling rat roaming in the eight-arm maze, as well as navigating rats through a 3D obstacle route. The results indicated that the system worked stably and that the stimulation was effective for different types of rat behavior controls. In addition, the results showed that stimulation in the whisker barrel region of rat primary somatosensory cortex (SI) acted like a cue. The animals can be trained to take different desired turns upon the association between the SI cue stimulation and the reward stimulation in the medial forebrain bundle (MFB).

Abstract:To study decoding information in the neural system, invasive brain-machine interfaces (BMIs) in rat were investigated. In this paper we developed a remote control training system for rat navigation. The system consists of an integrated PC control program with functions of signal communication, parameter setting and data file management, a transmitter and a receiver based on Bluetooth modules, and a stimulator controlled by a microprocessor. Stimuli delivered into the rat brain through a wireless micro-stimulator brought the rat to bear virtual cues or rewards and follow the commands to move right, left and forward in 3D obstacle environments. We designed a portable bi-directional telemetry communication system, which can deliver four-channel stimuli and receive two-channel neuronal signals in real time. It helps us in studying brain stimulation and neuronal activity recording in freely behaving rats. The firing patterns of neural units in motor cortex (M1) were detected by adjusted matched filter and classified by principal component analysis (PCA) and 3-D approximate entropy (ApEn). After synchronous analysis of the forelimb movement behavior images and neuronal signals, different statuses of neural activities during rats performing forelimb extend-flex tasks were clearly discriminated. The accuracy is 96.3%. The invasive brain-machine interfaces in rat are helpful to further explore the relationship between stimulation and responses in the neural network in free-moving status. The experiment results provide some new tools to brain-machine interfaces technologies. The system is useful for neuron behavior encoding mechanism research.
A rat-robot control system based on optogenetics

Abstract:We developed a rat-robot system based on optogenetic techniques for the precise freezing behavior. Rat-robots were built up by optogenetic modulation at the dlPAG of rat brains. We conducted track navigation for the rat-robots and found they were able to exhibit precise freezing at given positions with high spatiotemporal accuracy. Different types of optical stimulation were compared and their influence on the rat-robots was investigated. Furthermore we recorded the neural electrical activity in real time during the optical stimulation. The system could be used to explore the mechanism of freezing behaviors and to build up a more integrated rat-robot navigation system based on optical modulations.
A Telecontrol Navigation System for Bio-robot

Abstract:A telecontrol navigation system for bio-robot is developed,which is applicable to the navigation control of rats by stimulating some particular brain areas.The system basically consists of two parts,i.e.,wireless parameters transmitter device(WPTD) and implantable neural stimulation system(INSS).The WPTD contains a single-chip microcomputer (C8051F330),a wireless frequency modulation(FM) transmitter chip(QN8027) and power amplifier(PA) circuits.The INSS includes a C8051F330,a wireless FM receiver chip(QN8025),analog switches,and implantable electrodes.The INSS is fabricated of surface mount devices(SMD) with the dimension of 20 mm×15 mm×3 mm,weight of 6 g(including a rechargeable lithium battery,3 g).Lateral medial forebrain bundle(MFB) and ventral posterlateral thalamic nucleus(VPL) are selected as the brain sites to be stimulated,and each brain site is implanted into a pair of electrodes.Each pair of electrodes as one channel is connected with INSS,so that WPTD can regulate the signal parameters of the neural stimulation in each channel in real time.Experiments indicate that the current stimulation of MFB with proper parameters can drives the rat to move forward as virtual reward,while the current stimulation of VPL with proper parameters can cause the rat to turn left or right.

Abstract:In this paper, we present a novel intelligent control system for rat navigation based on video tracking and state machine techniques. And we demonstrate the automatic navigation of a rat to follow a preset course. The system consists of these components: (1) a wireless backpack stimulator comprised of stimulating circuit, control processor and bluetooth transceivers, (2) a video camera device capturing the rat's movement, (3) a state machine based control program generating navigation commands automatically. The main contribution of this work is the intelligent control system for navigating a rat accurately and automatically in a comparatively narrow space. Experiment results show that the system is stable and practical.
A Remote Constant Current Stimulator Designed For Rat-Robot Navigation

Abstract:In this paper, a remote stimulator is developed for rat-robot navigation based on the technique of Brain-Computer-Interface (BCI). The stimulator can output constant current from 0 to 1000 mu A, which overcome several shortages of our previous constant voltage stimulator. The constant current stimulator consists of four major components, including power supply, micro control unit (MCU), constant current source and bluetooth transceiver for downloading stimulation commands. The stimulator has a weight of about 20g and size of 32mm*25mm*6mm It has five channels of stimulation, which are connected with implanted microelectrodes in rat brain. The electrical parameters were characterized on three rats with different recovery time after brain surgery. Increasing current stimulations were applied on the dorsolateral periaqueductal gray (d1PAG) area to prove the effect of current stimulation on rat behavior.

Abstract:As a cutting-edge technology, animal robots based on living organisms are being extensively studied, with potential for diverse applications in the fields of neuroscience, national security, and civil rescue. However, it remains a significant challenge to reliably control the animal robots with the objective of protecting their long-term survival, and this has seriously hindered their practical implementation. To address this issue, this work explored the use of a bio-friendly neurostimulation system that includes integrated stimulation electrodes together with a remote wireless stimulation circuit to control the moving behavior of rat robots. The integrated electrodes were implanted simultaneously in four stimulation sites, including the medial forebrain bundle (MFB) and primary somatosensory cortex, barrel field (S1BF). The control system was able to provide flexibility in adjusting the following four stimulation parameters: waveform, amplitude, frequency, and duration time. The optimized parameters facilitated the successful control of the rat's locomotion, including forward movement and left and right turns. After training for a few cycles, the rat robots could be guided along a designated route to complete the given mission in a maze. Moreover, it was found that the rat robots could survive for more than 20 days with the control system implanted. These findings will ensure the sustained and reliable operation of the rat robots, laying a robust foundation for advances in animal robot regulation technology.

Abstract:Studies on behavior control are important for bio-robots designation. For auto or manual navigation of the bio-robots, the accuracy of the command execution is especially critical. In this paper, we reported a precise "STOP" command for the rat-robots by optical stimulation of the central nervous system (CNS). We labeled dorsolateral periaqueductal gray (dlPAG) neurons with light sensitive channelrhodopsin-2 (ChR2) and directly probed the optical fiber to reactivate these neurons. The rats showed freezing behavior only upon the optical stimulation with an appropriate range of laser intensity and stimulation frequency. Neuron spikes and local field potential (LFP) were simultaneously recorded with optical stimulation by optrodes on free moving rat-robots. Together, our findings demonstrated the utility of deep brain optical stimulation for the stopping behavior of rat-robot control and indicated a potential application of optogenetics for precise control of bio-robots in further work.

Abstract:Traditional automatic navigation methods for bio-robots are constrained to configured environments and thus can't be applied to tasks in unknown environments. With no consideration of bio-robot's own innate living ability and treating bio-robots in the same way as mechanical robots, those methods neglect the intelligence behavior of animals. This paper proposes a novel ratbot automatic navigation method in unknown environments using only reward stimulation and distance measurement. By utilizing rat's habit of thigmotaxis and its reward-seeking behavior, this method is able to incorporate rat's intrinsic intelligence of obstacle avoidance and path searching into navigation. Experiment results show that this method works robustly and can successfully navigate the ratbot to a target in the unknown environment. This work might put a solid base for application of ratbots and also has significant implication of automatic navigation for other bio-robots as well.
Live Demonstration: A BLE Wireless Neural-Recording Headstage on Freely-Moving Rat

Abstract:Wirelessly neural recording offers an effective way in continuously monitoring the neural behaviors of animals. In this work, we demonstrate a BLE wireless headstage system based on a low-power neural-recording chip, which consists of an 8-channel analog front end (AFE) and a 10-bit successive-approximation-register (SAR) analog-to-digital converter (ADC). A kickback-reduction technique is proposed to reduce the power of neural-recording chip, minimizing the battery size of the headstage system. The chip is implemented in 65nm CMOS process, and measured results show that: 1) The AFE achieves a gain of 43.3dB, an input referred noise (IRN) of 9.68µVrms, a bandwidth of 0.9Hz−7.2kHz, and a common-mode rejection ratio (CMRR) of 82.75dB. 2) The SAR ADC achieves an effective number of 9.66 bits with only 315nW power consumption. 3) The neural-recording chip consumes totally 249µW. In addition, invivo demonstration on a Sprague-Dawley rat is presented, where the 2.6g headstage induces little impact to the freely-moving activities. In addition, the in-vivo neural signals are transferred wirelessly to a commodity cellphone through BLE, which are displayed in 8 channels by a self-developed software.

Abstract:The technology of optogenetics provides a new method to modulate neural activity with spatial specificity and millisecond-temporal scale. This nonelectrical modulation method also gives chance for simultaneous electrophysiological recording during stimulations. Here, we describe our locomotor activity modulation on free-behaving rats using optogenetic techniques. The target sites of the rat brain were dorsal periaqueductal gray (dPAG) and ventral tegmental area (VTA) for the modulation of defensive and reward behaviors, respectively.

Abstract:Electric brain stimulation is frequently used in bio-robot control. However, one possible limitation of electric stimulation is the resultant wide range of influences that may lead to unexpected side-effects. Although there has been prior research done towards optogenetics based brain activation, there has not been much development regarding the comparisons between electric and optical methods of brain activation. In this study, we first encode “Stop” and “Escape” commands by optical stimulation in the dorsal periaqueductal grey (dPAG). The rats behavioral comparisons are then noted down under these two methods. The dPAG neural activity recorded during optical stimulation suggests rate and temporal coding mechanisms in behavioral control. The behavioral comparisons show that rats exhibit anxiety under the “Stop” command conveyed through both optical and electric methods. However, rats are able to recover more quickly from freezing only under optical “Stop” command. Under “Escape” commands, also conveyed through optical means, the rat would move with lessened urgency but the results are more stable. Moreover, c-Fos study shows the optical stimulation activates restricted range in midbrain: the optical stimulation affected only dPAG and its downstreams but electric stimulation activates both the upstream and downstream circuits, in which the glutamatergic neurons are largely occupied and play important role in “Stop” and “Escape” behavior controls. We conclude that optical stimulation is more suited for encoding “Stop” and “Escape” commands for rat–robot control.

Abstract:A rat robot is a type of animal robots, where an animal is connected to a machine system via a brain-computer interface. Electrical stimuli can be generated by the machine system and delivered to the animal's brain to control its behavior. The sensory capacity and flexible motion ability of rat robots highlight their potential advantages over mechanical robots. However, most existing rat robots require that a human observes the environmental layout to guide navigation, which limits the applications of rat robots. This work incorporates object detection algorithms to a rat robot system to enable it to find 'human-interesting' objects, and then use these cues to guide its behaviors to perform automatic navigation. A miniature camera is mounted on the rat's back to capture the scene in front of the rat. The video is transferred via a wireless module to a computer and we develop some object detection/identification algorithms to allow objects of interest to be found. Next, we make the rat robot perform a specific motion automatically in response to a detected object, such as turning left. A single stimulus does not allow the rat to perform a motion successfully. Inspired by the fact that humans usually give a series of stimuli to a rat robot, we develop a closed-loop model that issues a stimulus[GRAPHICS]sequence automatically according to the state of the rat and the objects in front of it until the rat completes the motion successfully. Thus, the rat robot, which we refer to as a rat cyborg, is able to move according to the detected objects without the need for manual operations. The object detection methods and the closed-loop stimulation model are evaluated in experiments, which demonstrate that our rat cyborg can accomplish human-specified navigation automatically.

Abstract:Rat cyborg is a rat which can receive cerebral commands and act as human wants. The navigation task is a widely used scenario in which the rat cyborg is required to walk along a specified path according to commands. Mostly the commands are given by human operators, but there's a need for automatic control which is quite challenging since several modules need to be elaborately combined together to work as a whole, among which the monitoring system and the commands model are two major components. Previously, few attempts were made for behavior monitoring of rat cyborg. Existing works often implement an over-simple rat information extractor which is capable of giving only a few parameters of the rat cyborg, which greatly limited the performance of their automatic control accuracy. In response to this requirement, we develop a monitoring system that is capable of giving detailed motion parameters and accurate body postures of the rat cyborg. We explore the possibility of recognizing rat postures using shape information, which is described by the powerful Zernike moments. We propose several simple shape descriptors which are fast to compute and achieve acceptable performance.
A novel rat robot controlled by electrical stimulation of the nigrostriatal pathway

Abstract:Objective: Artificial manipulation of animal movement could offer interesting advantages and potential applications using the animal's inherited superior sensation and mobility. Although several behavior control models have been introduced, they generally epitomize virtual reward-based training models. In this model, rats are trained multiple times so they can recall the relationship between cues and rewards. It is well known that activation of one side of the nigrostriatal pathway (NSP) in the rat induces immediate turning toward the contralateral side. However, this NSP stimulation-induced directional movement has not been used for the purpose of animal-robot navigation. In this study, the authors aimed to electrically stimulate the NSP of conscious rats to build a command-prompt rat robot. Methods: Repetitive NSP stimulation at 1-second intervals was applied via implanted electrodes to induce immediate contraversive turning movements in 7 rats in open field tests in the absence of any sensory cues or rewards. The rats were manipulated to navigate from the start arm to a target zone in either the left or right arm of a T-maze. A leftward trial was followed by a rightward trial, and each rat completed a total of 10 trials. In the control group, 7 rats were tested in the same way without NSP stimulation. The time taken to navigate the maze was compared between experimental and control groups. Results: All rats in the experimental group successfully reached the target area for all 70 trials in a short period of time with a short interstimulus interval (< 0.7 seconds), but only 41% of rats in the control group reached the target area and required a longer period of time to do so. The experimental group made correct directional turning movements at the intersection zone of the T-maze, taking significantly less time than the control group. No significant difference in navigation duration for the forward movements on the start and goal arms was observed between the two groups. However, the experimental group showed quick and accurate movement at the intersection zone, which made the difference in the success rate and elapsed time of tasks. Conclusions: The results of this study clearly indicate that a rat-robot model based on NSP stimulation can be a practical alternative to previously reported models controlled by virtual sensory cues and rewards.
A Radio-Telemetry System for Navigation and Recording Neuronal Activity in Free-Roaming Rats

Abstract:A radio-telemetry recording system is presented which is applied to stimulate specific brain areas and record neuronal activity in a free-roaming rat. The system consists of two major parts: stationary section and mobile section. The stationary section contains a laptop, a Micro Control Unit (MCU), an FM transmitter and a receiver. The mobile section is composed of the headstage and the backpack (which includes the mainboard, FM transmitter, and receiver), which can generate biphasic microcurrent pulses and simultaneously acquire neuronal activity. Prior to performing experiments, electrodes are implanted in the Ventral Posterolateral (VPL) thalamic nucleus, primary motor area (M1) and Medial Forebrain Bundle (MFB) of the rat. The stationary section modulates commands from the laptop for stimulation and demodulates signals for neuronal activity recording. The backpack is strapped on the back of the rat and executes commands from the stationary section, acquires neuronal activity, and transmits the neuronal activity singles of the waking rat to the stationary section. All components in the proposed system are commercially available and are fabricated from Surface Mount Devices (SMD) in order to reduce the size (25 mm × 15 mm × 2 mm) and weight (10 g with battery). During actual experiments, the backpack, which is powered by a rechargeable Lithium battery (4 g), can generate biphasic microcurrent pulse stimuli and can also record neuronal activity via the FM link with a maximum transmission rate of 1 kbps for more than one hour within a 200 m range in an open field or in a neighboring chamber. The test results show that the system is able to remotely navigate and control the rat without any prior training, and acquire neuronal activity with desirable features such as small size, low power consumption and high precision when compared with a commercial 4-channel bio-signal acquisition and processing system.

Abstract:Bio-robots based on brain computer interface (BCI) suffer from the lack of considering the characteristic of the animals in navigation. This paper proposed a new method for bio-robots' automatic navigation combining the reward generating algorithm base on Reinforcement Learning (RL) with the learning intelligence of animals together. Given the graded electrical reward, the animal e.g. the rat, intends to seek the maximum reward while exploring an unknown environment. Since the rat has excellent spatial recognition, the rat-robot and the RL algorithm can convergent to an optimal route by co-learning. This work has significant inspiration for the practical development of bio-robots' navigation with hybrid intelligence.

Abstract:Background: Bran-computer interface (BCI) is an important technique used in brain science. However, the large size of equipment and wires severely limit its practical applications. New Methods: This study presents a wearable system with bidirectional brain-computer interface based on Wi-Fi technology, which can be used for olfactory electrophysiological recording and animal motion control. Results: On the "brain-to-computer" side, the results show that the wireless system can record high-quality olfactory electrophysiological signals for over a month. By analyzing the recorded data, we find that the same mitral/tufted (M/T) cells can be activated by many odorants and different M/T cells can be activated by a single odorant. Further, we find neurons in dorsal lateral OB are highly sensitive to isoamyl acetate. On the "computerto-brain" side, the results show that we can efficiently control rats' motions by applying electrical stimulations to electrodes implanted in specific brain regions. Comparison with existing methods: Most existing wireless BCI systems are designed for either recording or stimulating while our system is a bidirectional BCI featured with both functions. Taking advantage of our years of experience in olfactory decoding, we developed the first wireless system for olfactory electrophysiological recording and animal motion control. It provides high-quality recording and efficient motion control for a long time. Conclusions: The system provides possibility of practical SCI applications, such as in vivo bioelectronic nose and "rat-robot".
Bio-Robots Automatic Navigation With Electrical Reward Stimulation

Abstract:Bio-robots that controlled by outer stimulation through brain computer interface (BCI) suffer from the dependence on realtime guidance of human operators. Current automatic navigation methods for bio-robots focus on the controlling rules to force animals to obey man-made commands, with animals' intelligence ignored. This paper proposes a new method to realize the automatic navigation for bio-robots with electrical micro-stimulation as real-time rewards. Due to the reward-seeking instinct and trial-and-error capability, bio-robot can be steered to keep walking along the right route with rewards and correct its direction spontaneously when rewards are deprived. In navigation experiments, rat-robots learn the controlling methods in short time. The results show that our method simplifies the controlling logic and realizes the automatic navigation for rat-robots successfully. Our work might have significant implication for the further development of bio-robots with hybrid intelligence.

Abstract:A multi-channel implantable electrical stimulator is described based on wireless microcontroller CC2430. PWM conversion circuits to DAC and the voltage-controlled current source are designed, the latter of which is needed for electrical stimulator circuits. Through the multi-channel analogy switches, positive and negative pulses on the rat brain areas can be achieved. The positive and negative waves are shown to verify the validity of the circuits.
