Abstract:Manidipine (MP) is a dihydropyridine drug, which is treated for the reduction of high blood pressure. The aim of this study is to clarify the photochemical behavior of MP in the case of ultraviolet light (UV) irradiation for MP tablets (Calslot ® tablets). The tablets and its altered forms (powders and suspensions) were UV-irradiated using a black light, and residual amounts of active pharmaceutical ingredients (APIs) were monitored by high-performance liquid chromatography (HPLC). Due to the photoproducts of MP were detected in HPLC chromatograms, the elucidation of their chemical structures was carried out utilizing electrospray ionization tandem mass spectrometry (LC–ESI–MS/MS). As a result, APIs in Calslot ® tablets were almost completely photodegraded in the case that Calslot ® tablets were suspended in an aqueous media along with the generation of some MP photoproducts. LC–ESI–MS/MS analysis clarified the chemical structures of three MP photoproducts, indicating that they were a pyridine analogue, benzophenone and a hydrolysate. Benzophenone was a main MP photoproduct. It was possible that MP might be firstly oxidized to form its pyridine analogue, followed by the oxidation of a dimethyl methylene moiety. This moiety seemed to be eliminated as a benzophenone, and the cleavage of an ester bond of the residual moiety resulted in the generation of a hydrolysate. Finally, toxicological potencies of MP and its photoproducts were predicted in silico toxicity evaluation, suggesting some of biological effects of the photoproducts might be altered compared with MP.

A new similarity measure for interval type-2 fuzzy sets: application in fuzzy risk analysis

Abstract:Here, we present a fuzzy risk analysis method based on a new similarity measure of interval type-2 fuzzy sets (IT2-FSs). First, we present a new similarity measure for IT2-FSs. This measure considers features of IT2-FSs such as cardinality, height and distance. We establish some properties of the proposed similarity measure. We make an experiment to compare the experimental results obtained by the proposed method with the ones obtained by other existing similarity measures. The proposed method shows not to have the drawbacks of the existing methods. Applying the proposed method, we develop a fuzzy risk analysis approach extending the utility of fuzzy risk analysis methods in dealing with sophisticated problems. The factors used in the fuzzy risk analysis are the probability of failure, severity of loss, probability of failure detection and economic disbenefits of failure occurrence, features not being possible to use in existing interval type-2 fuzzy risk analysis methods.

Photostability of extended-release matrix formulations

Abstract:The photostability of drugs has been widely studied while less attention is devoted to the possible modifications that UV light may induce on the excipients of a dosage form, in particular, on the functional polymers used to modulate drug delivery. In this work we have evaluated the effect of UV light on the release characteristics of extended-release matrix tablets containing hydroxypropylmethylcellulose (HPMC) or polyethylene oxide (PEO) as retarding polymers. Two different model drugs have been used: nifedipine (insoluble and photolabile drug) and diltiazem (soluble and photostable drug). Photodecomposition of nifedipine was evaluated and the formation of the photoproducts was followed during the dissolution process. Regarding the dissolution stability, the matrix tablets containing HPMC exposed to UV light have not shown significant differences in drug release profiles compared to the same non-irradiated formulation, while the matrix tablets containing PEO and exposed to the same conditions of UV light have shown a remarkable increase of drug release rate within the first minutes of the dissolution test (burst effect) which is particularly critical because it can cause the loss of the desired therapeutic control.

Abstract:1,4-dihydropyridines (1,4-DHPs) are widely recognized as highly effective L-type calcium channel blockers with significant therapeutic benefits in the treatment of cardiovascular disorders. 1,4-DHPs can also target T-type calcium channels, making them promising drug candidates for neurological conditions. When exposed to light, all 1,4-DHPs tend to easily degrade, leading to an oxidation product derived from the aromatization of the dihydropyridine ring. Herein, the elaboration of a quantitative structure–property relationships (QSPR) model was carried out by correlating the light sensitivity of structurally different 1,4-DHPs with theoretical molecular descriptors. Photodegradation experiments were performed by exposing the drugs to a Xenon lamp following the ICH rules. The degradation was monitored by spectrophotometry, and experimental data were elaborated by Multivariate Curve Resolution (MCR) methodologies to assess the kinetic rates. The results were confirmed by the HPLC-DAD method. PaDEL-Descriptor software was used to calculate molecular descriptors and fingerprints related to the chemical structures. Seventeen of the 1875 molecular descriptors were selected and correlated to the photodegradation rate by means of the Ordinary Least Squares (OLS) algorithm. The chemometric model is useful to predict the photosensitivity of other 1,4-DHP derivatives with a very low relative error percentage of 5.03% and represents an effective tool to design new analogs characterized by higher photostability.

Forced degradation studies and structural characterization of related substances of bisoprolol fumarate in finished drug product using LC-UV-MS/MS

Abstract:Methods for determination of bisoprolol and related substances mostly use UV detection and a phosphate buffer and are not suitable for MS detection. In this study, LC?UV?MS/MS for separation and characterization of bisoprolol related substances was developed, validated and applied for studying the degradation products of bisoprolol when exposed to hydrolytic stress, heat and light. The method uses a C18 column, formic acid in water and acetonitrile as mobile phases, gradient elution and UV and MS detection. Forced degradation revealed that acid hydrolysis produces the most intensive transformation of bisoprolol to its impurity A, along with impurities L and D. Alkaline hydrolysis produced impurities A, L, Q, G and K; oxidative and thermal degradation produced impurities A, L and K, while photodegradation produced impurities A, L, G and K, all characterized by their mass spectral data. The developed method using two detection systems was demonstrated as efficient since mass spectra allowed identification of the related substances of bisoprolol and quantification was possible using absorbance measurements at 270 nm. The obtained results will fill in the lack of data on the fragmentation patterns of bisoprolol and related substances that could be used by researchers and practitioners in research and quality control laboratories.

Effect of atmosphere and relative humidity on photodegradation of clopidogrel under artificial solar and indoor light irradiation

Abstract:Knowledge of the chemical stability of active pharmaceutical ingredients (APIs) is an important issue in the drug development process. This work describes a methodical approach and a comprehensive protocol for forced photodegradation studies of solid clopidogrel hydrogen sulfate (Clp) under artificial sunlight and indoor irradiation at different relative humidities (RHs) and atmospheres. The results showed that, at low RHs (up to 21%), this API was relatively resistant to simulated sunlight as well as indoor light. However, at higher RHs (between 52% and 100%), more degradation products were formed, and the degradation rate increased with rising RH. The influence of oxygen on the degradation was relatively low, and most degradation reactions proceeded even in humid argon atmosphere. The photodegradation products (DP) were analyzed with two different HPLC systems (LC-UV, LC-UV-MS) and selected impurities were separated by a semi-preparative HPLC and identified by high resolution mass spectrometry (ESI-TOF-MS) and 1H NMR techniques. Based on the obtained results, a light induced degradation pathway could be proposed for Clp in solid state.
Subleading jet functions in inclusive B decays

Abstract:The contribution of subleading jet functions to inclusive decay distributions of B mesons are derived from a systematic two-step matching of QCD current correlators onto soft collinear and heavy quark effective theory. Focusing on the tree level matching of QCD onto soft collinear effective theory, the subleading jet functions are defined to all orders in αs(μi) (with μi2 ~ mbΛQCD) and are calculated explicitly at first order in αs(μi). We present explicit expressions for the decay rates of → Xu l and the Q7γ−Q7γ contribution to → Xs γ, where the subleading jet functions are multiplied by a tree level hard function and appear in a convolution with the leading order shape function. Together with the recent two loop calculation of the leading order hard function for → Xu l , this paper will allow for a more precise description of inclusive B decays in the end point region.

Abstract:Rationale Lomefloxacin hydrochloride ear drops are highly unstable to light and prone to produce photodegradation impurities. These impurities might be related to the phototoxicity of lomefloxacin, which could seriously threaten the health of patients. In this article, the photodegradation impurity profile in lomefloxacin hydrochloride ear drops was studied for further improvement of quality control of the drug. Methods By studying the chromatographic behavior of photodegradation impurities, the photodegradation impurities in lomefloxacin hydrochloride ear drops were separated and detected effectively. Liquid chromatography combined with ion trap/time‐of‐flight mass spectrometry was applied to characterize the structures of the photodegradation impurities in lomefloxacin hydrochloride ear drops. Results The structures of 17 impurities in lomefloxacin hydrochloride ear drops were elucidated based on high‐resolution MS n data in positive ion mode, 12 of them being unknown impurities. Conclusions The structural characteristics and fragmentation patterns of the photodegradation impurities were also studied. The study of the photodegradation impurity profile in lomefloxacin hydrochloride ear drops provides a scientific basis for quality control of these ear drops and ensures the safety of drug use by the public.

Micelle-enhanced spectrofluorimetric method for determination of lacidipine in tablet form; application to content uniformity testing

Abstract:A highly sensitive, simple and rapid spectrofluorimetric method was developed for the determination of lacidipine (LCP) in tablets. The proposed method is based on the investigation of the fluorescence spectral behavior of LCP in both sodium dodecyl sulphate (SDS) and the tween-80 micellar system. In aqueous solutions of acetate buffer (pH 4.5), the fluorescence intensities of LCP were greatly enhanced (ca. 2.4 and 4.3 folds) in the presence of either SDS or tween-80, respectively. The fluorescence intensity was measured at 444 nm after excitation at 277 nm using either SDS or tween-80 as a surfactant. The fluorescence-concentration plots were rectilinear over the ranges of 50.0-500.0 ng/ml and 5.0-200.0 ng/ml with lower detection limits of 5.11 and 2.06 ng/ml and lower quantification limits of 17 and 6.87 ng/ml using SDS and tween-80, respectively. The method was successfully applied to the analysis of LCP in commercial tablets and the results were in good agreement with those obtained with the comparison method. Furthermore, content uniformity testing of pharmaceutical tablets was also conducted.

Abstract:Ethiprole (ETH) is a phenylpyrazole insecticide that is used worldwide as an alternative to fipronil (FIP). Research on the photodegradation of ETH in aquatic environments has been limited compared with that on FIP. In this study, to clarify the photodegradation of ETH in aquatic systems, the photodegradation pathway and products were investigated using liquid chromatography and liquid chromatography-tandem mass spectrometry. We also determined the photochemical half-lives ( t 1/2 ) of ETH and its main degradation products. The primary photodegradation pathway was cyclization/dechlorination and hydroxylation/dechlorination of ETH to form the didechlorinated products (benzimidazole of des-chloro-hydroxy-ETH). Some newly identified photodegradation products and analogs of FIP photodegradation products were also detected as minor products. We compared the photodegradation of ETH with that of FIP under the same conditions. Didechlorinated products of ETH and FIP had the highest photostability. However, although the photochemical t 1/2 of EHT was 2.7 times that of FIP, the photochemical t 1/2 of the didechlorinated product of ETH was approximately one-third that of the didechlorinated product of FIP. This comparison of the photochemical processes of ETH and FIP provides new insight into the persistence and characteristics of both insecticides in the environment.

Solvent-shift strategy to identify suitable polymers to inhibit humidity-induced solid-state crystallization of lacidipine amorphous solid dispersions

Abstract:The solvent-shift strategy was used to identify appropriate polymers that inhibit humidity-induced solid-state crystallization of amorphous solid dispersions (ASDs). Lacidipine with the polymers, PVP-K30, HPMC-E5 or Soluplus, were combined to form amorphous solid dispersions prepared by solvent evaporation. The formulations were characterized by differential scanning calorimetry (DSC), powder X-ray diffraction (PXRD), and Fourier-transform infrared spectroscopy (FT-IR) and were subjected to in vitro dissolution testing. The moisture had a significant impact on the amount dissolved for the solid dispersions. Molecular docking studies established that hydrogen bonding was critical for the stabilization of the solid dispersions. The rank order of the binding energy of the drug-polymer association was Soluplus (-6.21 kcal/mol)>HPMC-E5 (-3.21 kcal/mol)>PVP-K30 (-2.31 kcal/mol). PVP-K30 had the highest water uptake among the polymers, as did ASD system of lacidipine-PVP-K30 ASDs. In the Soluplus ASDs, with its strong drug-polymer interactions and low water uptake, moisture-induced solid-state crystallization was not observed.
Degradation of Selected Pharmaceuticals in Aqueous Solution with UV and UV/H(2)O(2).

Abstract:The degradation of four pharmaceutical compounds (PhACs), ibuprofen (IBU), diphenhydramine (DP), phenazone (PZ), and phenytoin (PHT) was investigated via ultraviolet (UV) photolysis and UV/H(2)O(2) process with a low-pressure (LP) UV lamp. For each PhAC tested, direct photolysis quantum yields at 254 nm were found to be ranging from 6.32 x 10(-2) to 2.79 x 10(-1)molE(-1) at pH 7. The second-order rate constants of the reaction between the PhACs and OH were determined to be from 4.86 x 10(9) to 6.67 x 10(9)M(-1)s(-1) by using a competition kinetic model which utilized para-chlorobenzoic acid (pCBA) as a reference compound. The overall effect of OH radical scavenging from humic acid (HA) and anions HCO(3)(-), NO(3)(-) was measured utilizing R(OH,UV) method through examining the aqueous photodegradation of pCBA as a probe compound. Moreover, these fundamental direct and indirect photolysis parameters were applied in the model prediction for oxidation rate constants of the PhACs in UV/H(2)O(2) process. It was found that the predicted oxidation rate constants approximated the observed ones. The results indicated that the new R(OH,UV) probe compound method was applicable for measuring background OH radical scavenging effects in water treatment process of UV/H(2)O(2). Furthermore, by GC-MS analysis, most of the intermediates created during the photodegradation of the selected PhACs in UV/H(2)O(2) process were identified. For the photodegradation of PZ, a competition mechanism existed between the direct UV photolysis and the oxidation of OH. An appropriate dosage of H(2)O(2) could hinder the occurrence of the direct photolysis.
Improvement of The Photostability of Nimodipine by Using Liquisolid Compacts Technique

Abstract:Liquisolid compacts are one of the modern drug formulation techniques used for drug formulation for several objectives including protection against photosensitivity. This study evaluates the photoprotective effect of the liquisolid compact technique on a calcium channel blocker antihypertensive drug, nimodipine, which is considered a photosensitive drug model. Liquisolid compact tablets of nimodipine were prepared from; Avicel PH-102 as a carrier material, (colloidal silicon dioxide, Cab-O-sil, and titanium dioxide as coating materials), croscarmellose sodium (CSS), sodium starch glycolate (SSG), crospovidone (CP) as super disintegrants, and propylene glycol as the liquid vehicle. However, the ratio of carrier to coating materials was kept constant in all formulations at 35:1. This ratio was chosen after testing the ratios 15:1, 20:1, 25:1, 30:1 and 35:1. The feasibility of the prepared liquisolid systems was also evaluated for first; the pre-compression and post-compression characteristics. Second, the possible drug-excipients interaction using FT-IR. Third, the photostability of Nimodipine in pure form, conventional tablets, and in the prepared liquisolid tablets through irradiation with a light dose of 0.55 W/m2/h visible light, 50 W/m2/h UVA and 0.27 W/m2/h UVB for 12 h in order to estimate the effect of coating material type on Nimodipine stability especially in the prepared liquisolid formulations. The results showed that all of the prepared liquisolid tablets showed satisfactory properties regarding pre and post-compression properties. Interestingly, the FTIR spectra study ruled out any interaction between nimodipine and excipients in the prepared liquisolid tablets. Besides, all of the prepared liquisolid tablets had shown a superior drug dissolution profile compared to the conventional and direct compressed tablets. Finally, 92.43% of the drug remaining was found in the liquisolid formulation (F7) after 12 h photo-irradiation as compared to 65.48% drug remaining for the pure drug substance and 75.12% for directly compressed tablets indicating that liquisolid technique had significantly inhibited the photo-degradative effect of light in the prepared liquisolid formulations. It can be concluded that the liquisolid compacts technique is a promising approach for the improvement of photostability of this photosensitive Nimodipine with an acceptable stability as well as enhanced dissolution rate.
Lacidipine Amorphous Solid Dispersion Based on Hot Melt Extrusion: Good Miscibility, Enhanced Dissolution, and Favorable Stability

Abstract:The present study aimed to increase the in vitro dissolution rate of lacidipine, a poorly water-soluble drug, by formulating amorphous solid dispersions (ASDs) using hot-melt extrusion (HME). Differential scanning calorimetry, powder X-ray diffraction, polarized light microscopy, and Fourier transform infrared were used to characterize the optimal formulations and evaluate the physical stability for the stress test. Film-casting method and hot-stage microscopy were applied to study the miscibility of lacidipine and the drug carriers. In vitro dissolution tests were conducted as the final evaluation index. The optimal formulations were successfully obtained with Soluplus and PVP VA64 at a drug/carrier ratio of 1:10 (w/w), Fourier transform infrared studies revealed the hydrogen bonding between drug and polymers, and in vitro dissolution rates of the optimal formulations were extremely enhanced compared to bulk lacidipine and physical mixtures, similar with that of the commercial tablet. The ASD formulated with Soluplus showed better physical stability than that with PVP VA64. A strong hydrogen bonding and good drug-polymer miscibility were essential to hinder the recrystallization of lacidipine ASDs. In conclusion, the lacidipine ASD formulated with Soluplus showed a significant increase in in vitro dissolution rate and favorable physical stability in the stress test.
Degradation of Anticonvulsant Drug Primidone in Aqueous Solution by Uv Photooxidation Processes

Abstract:This study reports on the photooxidation of antiepileptic drug primidone by direct UV-C irradiation, UV-C/H2O2, and UV-C/Fe(II). The influence of different parameters, such as pH, hydrogen peroxide concentration, Fe(II) concentration, Cl- concentration, HCO3- concentration, and primidone initial concentration, has been studied. Experiments were carried out by utilizing a UV high-pressure mercury lamp with 254 nm wavelength to irradiate aqueous solutions of primidone. Kinetic experiments showed that primidone followed pseudo-first-order kinetics. H2O2 concentration and Fe(II) concentration had significant influence on photodegradation speed of primidone, and their optimal concentrations were 1 and 0.1 mM, respectively. According to results of photooxidation experiments, effectiveness of specific photooxidation processes could be arranged in the following order: UV-C/Fe(II)>UV-C/H2O2>UV-C. Optimum conditions for primidone UV-C and UV-C/H2O2 photodegradation were observed at a pH value of 6.3. Increase of concentration of Cl- in UV-C/H2O2 system slightly increased primidone degradation rate. However, presence of HCO3- inhibited photodegradation of primidone. Saturation of primidone aqueous solution with O-2 proved to be beneficial to degradation of primidone in UV-C/Fe(II) system. Phenobarbital, 4-hydroxyphenobarbital, and 1-(2-nitrobenzyl)-5-oxoproline were identified by liquid chromatography-mass spectrometry-mass spectrometry analysis as main by-products of primidone degradation. Finally, primidone and its by-products were subjects to complete mineralization to ammonia, carbon dioxide, and water. This study obtained the fundamental data on the UV degradation efficiency and the by-products of primidone, and data obtained in our study can be used to evaluate the application of advanced oxidation processes.
Photochemical degradation of acifluorfen in aqueous solution

Abstract:To elucidate the photochemical behavior of diphenyl ether herbicides in superficial waters, the photodegradation of acifluorfen, 5-[2-chloro-4-(trifluoromethyl)phenoxy]-2-nitrobenzoïc acid (CAS Registry No. 50594-66-6), was studied in water and acetonitrile. All experiments were carried out under laboratory conditions using a solar simulator (xenon arc) or jacket Pyrex reaction cell equipped with a 125 W high-pressure mercury lamp. The calculated polychromatic quantum efficiencies (Phi(solvent)) of acifluorfen in different solvents are as follows (units are degraded molecules photon(-1)): Phi(water) = 10(-4), Phi(acetonitrile) = 10(-4), Phi(methanol) = 10(-4), and Phi(hexane) = 10(-2). The results obtained in this work are in good agreement with the literature value of monochromatic quantum yield. HPLC-MS analysis (APCI and ESI in positive and negative modes) was used to identify acifluorfen photoproducts. These results suggest that the photodegradation of acifluorfen proceeds via a number of reaction pathways: (1) decarboxylation, (2) dehalogenation, (3) substitution of chlorine group by hydroxyl or hydrogen groups, and (4) cleavage of ether linkage, giving phenols. Photorearrangement products were studied by other investigators. No such products were observed. In addition, it was found that the trifluoro functional group on acifluorfen was not affected by any transformation, and no products of a nitro group reduction were found.
Haemodynamic Parameters in Hypertensive Patients: Changes Induced by Lacidipine and Nifedipine.

Abstract:We conducted a randomly allocated, double-blind study in 16 essential hypertensive patients, eight of whom were treated with nifedipine and eight with lacidipine. The antihypertensive efficacy was evaluated and any modifications to peripheral haemodynamic parameters were observed in the brachial artery by a mechanographic method and B-mode scanner with a 10-MHz probe. Statistically significant reductions in blood pressure from basal values were observed after 1 and 6 months' treatment. Enhanced compliance (P less than 0.005), reduced characteristic impedance (P less than 0.001) and lower peripheral resistances (P less than 0.01) were also noted. Variations in pulse wave velocity and mean blood pressure showed a statistically significant correlation as early as the first month of treatment (P less than 0.01). Our results suggest that therapy with nifedipine and lacidipine allows an improvement in peripheral haemodynamics in hypertensive patients. This response is maintained in chronic treatment, even just before the next dose administration at the end of the longest dose interval.
Chemiluminescence method for evaluating photooxidative degradation of dispensed drugs: a potential new drug information tool

Abstract:Background: Dispensed drugs stored by patients are often in single-dose packages (SDPs) or are crushed and mixed after being removed from a press-through package (PTP) sheet. Information on their stability is extremely limited. To address this, we explored using chemiluminescence (CL) measurements to detect oxidative degradation. Methods: Eight amlodipine, 14 telmisartan, and two warfarin preparations were used as specimens. These preparations were stored at room temperature under various conditions, after which CL was measured. Cellopoly packaging paper was used for SDP. Three light conditions were used (Condition A: darkness, Condition B: indoor diffused light (approximately 400 lx), and Condition C: exposure to 4,000 lx). CL cumulative light output was measured every minute under nitrogen gas conduction and with a sample chamber temperature of 150 °C, for a maximum of 10 min. Luminescence images were obtained simultaneously with the CL measurements. Results: CL was observed on light-exposed tablet surfaces. For each preparation, an increase in the CL value was observed with the duration of light exposure. In the same preparation with the same exposure time, CL tended to be higher in the order of Condition A < B < C. Moreover, CL increased even when no changes in color were observed by the naked eye. A comparison between preparations with the same main ingredients showed differences in the rate of increase in CL with exposure, and each was found to show a different reactivity to light. Conclusions: To the best of our knowledge, this is the first study to visually capture the surface oxidation of tablets exposed to light using the CL method. The CL values, thought to be derived from photooxidation, increased with exposure of tablets and powders to light after SDP. This method can sensitively assess drug degradation due to photooxidation. Further research is needed to establish a CL method for assessing the stability of preparations in clinical settings.
Lacidipine: a calcium antagonist with potent and long-lasting antihypertensive effects in animal studies

Abstract:Lacidipine, currently being evaluated as a once-daily antihypertensive agent, acted as a calcium entry blocker on rabbit ear artery (pA2 = 9.4) with a markedly slower onset of action than that of nitrendipine; this effect was not reversed after 9 h of drug washout. Calcium entry blocker activity was also evaluated on nonvascular smooth muscles: Lacidipine showed a more pronounced vascular selectivity than nitrendipine; for both drugs, concentrations required to induce negative inotropic effects in guinea pig ventricular strip were approximately 100 times higher than concentrations needed to antagonize calcium contraction in vascular smooth muscle. In spontaneously hypertensive rats (SHR), by the tail-cuff method, lacidipine (ED25 = 0.35 mg/kg orally, p.o.) proved approximately 30 times more potent, slower in onset, and longer-acting than nitrendipine in reducing blood pressure. These features were confirmed in chronically implanted SHR after oral and intravenous (i.v.) administration (ED25 = 0.19 mg/kg p.o. and 0.006 mg/kg i.v.). A short-lasting tachycardia was detected with both drugs. No evidence of acquired tolerance emerged after repeated oral administrations over a 3-week period. Lacidipine induced a natriuretic effect in saline-loaded SHR at antihypertensive doses. In renal hypertensive dogs, lacidipine proved more potent (three to seven times), slower in onset, and longer-lasting than nitrendipine after p.o. (ED25 = 0.22 mg/kg) and i.v. (ED25 = 0.004 mg/kg) administrations.
