Abstract:Synthetic musks, as an alternative product of natural musks, are widely used in almost all fragrances of consumer products, such as perfumes, cosmetics and detergents. During the past few decades, the production of synthetic musks has been increasing year by year, subsequently followed by large concern about their adverse effects on ecosystems and human beings. Until now, several studies have reviewed the latest development of analytical methods of synthetic musks in biological samples and cosmetics products, while there is still lack of a systematic analysis of their global distribution in different environmental media. Thus, this review summarizes the occurrence of synthetic musks in the environment including biota around the world and explores their global distribution patterns. The results show that galaxolide (HHCB), tonalide (AHTN), musk xylene (MX) and musk ketone (MK) are generally the most frequently detected synthetic musks in different samples with HHCB and AHTN being predominant. Higher concentrations of HHCB and AHTN are normally found in western countries compared to Asian countries, indicating more consumptions of these musks in western countries. The persistence, bioaccumulation and toxicity (PBT) of synthetic musks (mainly for polycyclic musks and nitro musks) are also discussed. The risk quotients (RQs) of HHCB, AHTN, MX and MK in most waters and sediments are below 0.1, reflecting a low risk to aqueous and sediment-dwelling species. In some sites, e.g., close to STPs, high risks (RQs>1) are characterized. Currently, limited data are available for macrocyclic musks and alicyclic musks in terms of either occurrence or PBT properties. More studies with an expanded scope of chemical type, geographical distribution and (synergic) toxicological effects especially from a long-term point of view are needed.

Abstract:Synthetic musks (SMs) have gained widespread utilization in daily consumer products, leading to their widespread dissemination in aquatic environments through various pathways. Over the past few decades, the production of SMs has consistently risen, prompting significant concern over their potential adverse impacts on ecosystems and human health. Although several studies have focused on the development of analytical techniques for detecting SMs in biological samples and cosmetic products, a comprehensive evaluation of their global distribution in diverse aquatic media and biological matrices remains lacking. This review aims to provide an up-to-date overview of the occurrence of SMs in both aquatic and various biological matrices, investigating their worldwide distribution trends, assessing their ecological toxicity, and comparing different methodologies for processing and analysis of SMs. The findings underscore the prevalence of polycyclic musks as predominant SMs, with consumption of various products in different countries leading to contrasting distribution of contaminants. Furthermore, the migration of SMs from sediments to the water phase is investigated, indicating the role of solid-phase reservoirs. Incomplete degradation of SMs in the environment could contribute to their accumulation in aquatic systems, impacting the growth and oxidative stress of aquatic organisms, and having a possibility of genotoxicity to them. Human exposure data highlight substantial risks for vulnerable populations such as pregnant women and infants. Moreover, contemporary methods for SMs analysis are presented in this review, particularly focusing on advancements made in the last five years. Finally, research enhancement and critical questions regarding the analysis of SMs are provided, offering suggestions for future research endeavors.

Abstract:The discharge of municipal wastewater and sewage sludge is the main pathway of synthetic musk entering soil environment. Parameters for the discharge are necessary for ecological risk assessment models,but few have been reported regarding the ecological risk assessment of synthetic musk in soil partly due to the shortage of convenient,quick and efficient techniques for the detection of synthetic musk in the environment. To promote the analytic methodology of synthetic musk,we compared and optimized the techniques for extracting two typical kinds of synthetic musk,galaxolide (HHCB) and tonalid (AHTN),in municipal wastewater by solid-phase extraction and liquid-liquid extraction,and in dehydrated sludge by soxhlet extraction and accelerated solvent extraction (ASE). The results showed that liquid-liquid extraction and ASE followed by silica gel column cleaning process had higher recovery in detecting synthetic musk in water and sludge samples. The two optimized methods were adopted to detect the HHCB and AHTN concentrations in the inflow and effluent water and the dehydrated sludge from a Beijing sewage treatment plant,and their concentrations were 897-1270 ng·L-1 and 331-488 ng·L-1 in inflow water,201-226 ng·L-1 and 65-130 ng·L-1 in effluent water,and 137-348 ng·g-1 and 64-138 ng·g-1 in dehydrated sludge,respectively.

Abstract:A method has been developed for the simultaneous determination of 11 synthetic musks (cashmeran, celestolide, phantolide, musk ambrette, traseolide, galaxolide, musk xylene, tonalide, musk moskene, musk tibetene, and musk ketone) in imported seafood. The method combines solid phase extraction and gas chromatography-mass spectrometry. Samples were extracted by n-hexane and purified using a Florisil column. Internal standards were used to correct for matrix effects. The calibration curves showed good linearity with correlation coefficients greater than 0.99. The limits of detection (S/N>3) ranged from 0.35 to 2.08 μg/kg, and the limits of quantification (S/N>10) were between 1.18 and 5.00 μg/kg. The average recoveries measured at three spiked levels (10, 20, and 30 μg/kg) were in the range 83.1%-117%, with relative standard deviations ranging from 5.1% to 8.5%. Further, the concentrations of 11 synthetic musks in 30 imported seafood in Shanghai were investigated. Galaxolide was detected in 93.3% of samples analysed, in concentration as high as 3.82 μg/kg. Musk ambrette and musk moskene were found in concentrations as high as 15.4 μg/kg and 10.5 μg/kg, respectively. The established method demonstrates high sensitivity and selectivity for the determination and confirmation of 11 synthetic musks in imported seafood.

Abstract:Synthetic musks (SMs), as an indispensable odor additive, are widely used in various personal care products. However, due to their physico-chemical properties, SMs were detected in various environmental media, even in samples from arctic regions, leading to severe threats to human health (e.g., abortion risk). Environmentally friendly and functionally improved SMs have been theoretically designed in previous studies. However, the synthesizability of these derivatives has barely been proven. Thus, this study developed a method to verify the synthesizability of previously designed SM derivatives using machine learning, 2D-QSAR, 3D-QSAR, and high-throughput density functional theory in order to screen for synthesizable, high-performance (odor sensitivity), and environmentally friendly SM derivatives. In this study, three SM derivatives (i.e., D52, D37, and D25) were screened and recommended due to their good performances (i.e., high synthesizability and odor sensitivity; low abortion risk; and bioaccumulation ability in skin keratin). In addition, the synthesizability mechanism of SM derivatives was also analyzed. Results revealed that high intramolecular hydrogen bond strength, electrostatic interaction, qH+ value, energy gap, and low EHOMO would lead to a higher synthesizability of SMs and their derivatives. This study broke the synthesizability bottleneck of theoretically designed environment-friendly SM derivatives and advanced the mechanism of screening functional derivatives.

Distribution and Elimination of Polycyclic Musks in Three Sewage Treatment Plants of Beijing, China.

Abstract:The two representative polycyclic musks, 1-(5,6,7,8-tetrahydro-3,5,5,6,8,8-hexamethyl-2-naphthenyl)-ethanone (AHTN) and 1,3,4,6,7,8-hexahydro-4,6,6,7,8,8-hexamethylcyclopenta[g]-2-benzopyrane (HHCB), were measured in aqueous samples and sludge samples of three sewage treatment plants (STPs) in Beijing, China using a gas chromatograph-mass spectrometry. The HHCB and AHTN concentration ranges in the influents were 1251.4-3003.8 ng/L and 111.9-286.3 ng/L, respectively. Meanwhile, the ranges of 492.8-1285.3 ng/L for HHCB and 47.3-89.3 ng/L for AHTN were present in the effluents. The musks in the sludges were three to four orders of magnitude higher than those in aqueous sewages. The removal efficiencies of the two musks varied in the ranges of 41.7%-70.1% for HHCB, and 25.5%-68.8% for AHTN. Adsorption onto suspended particulate matter played an important role in removing musks from the sewages. The HHCB/AHTN ratio along the treatment processes showed that the two musks had high similarity of removal from sewage by each reactor in STP. The musks in the effluent sewages may pose a low potential risk to aquatic environment in terms of the predicted-non-effect concentration. Nevertheless, considering the possible additive and synergistic effects with other compounds emitted via STPs, their bioconcentration and bioaccumulation in aquatic organisms and so on, it is essential to monitor these compounds in various compartments and to study their environmental fate.
Comparison of Sample Preparation and Detection Methods for the Quantification of Synthetic Musk Compounds (SMCs) in Carp Fish Samples

Abstract:This study deals with the separation and detection methods for 12 synthetic musk compounds (SMCs), which are some of the emerging contaminants in fish samples, are widely present in environmental media, and can be considered serious risks due to their harmful effects. For the separation of co-extracted substances and the target SMCs in fish samples after ultrasonic extraction, four solid-phase extraction (SPE) sorbents were investigated. The recoveries of SMCs from 10 mL of eluent, as optimized by the elution profile, were within the acceptable range of 80–120% in all SPE types, and it was found that nitro musk and polycyclic musk compounds were separated more clearly in Florisil SPE than others (Aminopropyl, Alumina-N, PSA). Furthermore, the results of measuring the matrix effects by each SPE through the spiking experiments showed that Florisil SPE was superior. The comparison of a gas chromatograph-single quadrupole mass spectrometer (GC-SQ/MS) with selected ion monitoring (SIM) mode and GC-triple quadrupole mass spectrometer (GC-QqQ-MS/MS) with multiple reaction monitoring (MRM) modes regarding the detection method of SMCs showed that the method detection limits (MDLs) of SMCs were on average ten times lower when GC-QqQ-MS/MS with MRM mode was used. The differences between the two methods can provide essential information for selecting an analytical method in related research fields that require appropriate detection levels, such as risk assessment or pollution control.

Identification of proteomic signatures of exposure to marine pollutants in mussels (Mytilus edulis)

Abstract:Bivalves and especially mussels are very good indicators of marine and estuarine pollution, and so they have been widely used in biomonitoring programs all around the world. However, traditional single parameter biomarkers face the problem of high sensitivity to biotic and abiotic factors. In our study, digestive gland peroxisome-enriched fractions of Mytilus edulis (L., 1758) were analyzed by DIGE and MS. We identified several proteomic signatures associated with the exposure to several marine pollutants (diallyl phthalate, PBDE-47, and bisphenol-A). Animals collected from North Atlantic Sea were exposed to the contaminants independently under controlled laboratory conditions. One hundred and eleven spots showed a significant increase or decrease in protein abundance in the two-dimensional electrophoresis maps from the groups exposed to pollutants. We obtained a unique protein expression signature of exposure to each of those chemical compounds. Moreover a set of proteins composed a proteomic signature in common to the three independent exposures. It is remarkable that the principal component analysis of these spots showed a discernible separation between groups, and so did the hierarchical clustering into four classes. The 14 proteins identified by MS participate in alpha- and beta-oxidation pathways, xenobiotic and amino acid metabolism, cell signaling, oxyradical metabolism, peroxisomal assembly, respiration, and the cytoskeleton. Our results suggest that proteomic signatures could become a valuable tool to monitor the presence of pollutants in field experiments where a mixture of pollutants is often present. Further studies on the identified proteins could provide crucial information to understand possible mechanisms of toxicity of single xenobiotics or mixtures of them in marine ecosystems.
Derivation of predicted no effect concentration and ecological risk assessment of polycyclic musks tonalide and galaxolide in sediment

Abstract:Polycyclic musks (PMs) have drawn increased attention in recent years because of their persistence, bioaccumulation and toxicity. As two typical PMs contaminants, tonalide (AHTN) and galaxolide (HHCB) are widely detected in sediment worldwide. Acute and chronic toxicity data of AHTN and HHCB to freshwater and seawater organisms in water and sediments are collected and screened. The predicted no effect concentrations (PNECsediment) for AHTN and HHCB is derived according to the equilibrium partitioning method recommended by the EU technical guidance document (TGD) and the species sensitivity distribution (SSD) method based on the measured sediment toxicity data. The concentration levels of AHTN and HHCB are investigated and evaluated in freshwater and seawater sediments. Results show the difference between native and non-native freshwater species is not statistically significant. AHTN is more toxic to freshwater and seawater organisms than HHCB, and seawater organisms are more sensitive to 2 musks than freshwater organisms. The chronic PNECsediment values of AHTN and HHCB are 194.48 and 416.47 ng/g in freshwater sediment, 88.93 and 128.34 ng/g in seawater sediment respectively. The AHTN and HHCB linear correlation analysis exhibited a strong positive linear correlation in both domestic (R2=0.9054) and foreign (R2 = 0.9645) sediment. Preliminary risk assessment shows that the risks posed by AHTN and HHCB in sediment based on individual or combined concentrations of two musks are at medium to high levels in some regions. Further risk assessment results indicate that, for HHCB, 1.72% of foreign freshwater sediment may pose an ecological risk to 5% species; for AHTN, 8.06% of foreign freshwater sediment and 1.02% of domestic freshwater sediment may pose an ecological risk to 5% species, and 5.86% of seawater sediment may pose an ecological risk to 5% species. The above results indicate that there are some negligible risks in domestic and foreign sediments posed by these two musks, we should continue to pay attention to the toxic effects and pollution level of both musks in environment.

Abstract:Chinese sturgeon (Acipenser sinensis) was listed as a Grade I protected animal in China in 1989, and the observed intersexual phenomenon and sex ratio deviation have suggested that chemicals have posed a risk as environment pollutants. This study analyzed seven musk fragrances in liver, muscle, heart, gonad, stomach, intestines, adipose, gill, pancreas, kidney, gallbladder, and roe from 13 female Chinese sturgeons, and the toxicokinetic behavior of musks were studied and compared with some organochlorines. Of the seven musks, HHCB, AHTN, and musk xylene were detected, and the highest concentrations were observed in adipose tissue: from 33.7 to 62.1 ng/g wet weight (ww), from 1.0 to 5.4 ng/g ww, and from 1.1 to 13 ng/g ww, respectively. Similar to the tissue distribution of DDTs and HCB, musks were detected frequently in high lipid content tissues such as roe, adipose, and liver, suggesting that tissue distribution of musks is controlled by the affinity to lipids. The concentration ratios based on lipid weight between roe and adipose were estimated to be 0.47 for HHCB, 0.58 for AHTN, and 0.51 for musk xylene, and those for the total DDTs and HCB were 0.27 and 0.61, which were relatively low compared with mammals. Relatively high concentrations of p,p'-DDE (68.4-449 ng/g ww) were detected in 10 of total 11 samples, which would cause the feminization of Chinese sturgeons during embryonic development. It was found that lipid-corrected concentrations of HHCB, AHTN, p,p'-DDE, and p,p'-DDD increased with age in female sturgeon, of which the trends were similar to those in fishes and different from those in mammals.
Spatial distribution characteristics of polycyclic musks as a chemical marker in river water and sediment compared with other typical pollutants.

Abstract:Polycyclic musks (PMs), widely used in the cosmetic and perfume industries, are gaining attention as a new type of persistent organic pollutant (POP). The related contamination in developing countries, such as China, where the use of these compounds is increasing, needs to be closely monitored. This work systematically studied the distribution profile (water-suspended particulate-sediment-porewater) of PMs in Haihe River, China. The average concentrations of PMs were comparable to those of the traditional POPs. The typical PMs (galaxolide and tonalide) exhibited different environmental behaviors in various media. Water played a more significant role than suspended particulates in the transport of pollutants. Importantly, this work explored the relationships among PMs, surfactant, phosphorus, carbon, salinity, heavy metals and pesticides. Unlike previous reports focusing on the relationships among PMs, wastewater discharge and population distribution, this work innovatively studied the distribution characteristics of pollutants by principal components analysis. The results suggest that PMs can be used as a chemical marker indicating domestic contamination.

Abstract:Musk is a secretion of the forest musk deer ( Moschus berezovskii ). Normal musk is a brown solid secretion with a light fragrance. In this study, abnormal types of musk, namely, white and black musks, were discovered during the musk collection process. Researchers have long been concerned with the components of musk. Herein, GC-MS, headspace solid-phase microextraction (HS-SPME), and nonmetric multidimensional scaling (NMDS) were used to analyze the nonpolar organic components, volatile organic components, and sample similarities among different musks, respectively. Abundant steroid hormones and proteins were also found in the musk. The steroid hormone concentrations were detected using a radioimmunoassay (RIA). Proteins in the samples were hydrolyzed and the amino acids concentrations were detected. The steroid hormone and amino acid concentrations in white musk were significantly lower than in normal and black musks (p<0.05). The components were subjected to NMDS analysis to understand the differences in components among different types of musk, with the results suggesting that white musk was different from normal and black musks.

Abstract:Abstract Background Moschus is a rare and precious natural medicine. Due to the properties of resources scarcity and expensive price of natural musk, artificial musk has been developed as substitute materials in some prescriptions. Rapid and accurate identification of natural or artificial musk in complex traditional Chinese medicine (TCM) preparations is also a challenge. Method A strategy from non-targeted to targeted gas chromatography-mass spectrometry (GC–MS) metabolomics was developed for discrimination of natural and artificial musk. Firstly, GC–MS-based non-targeted analysis combined with chemometrics was used to find the potential chemical markers to distinguish natural musk and artificial musk. Subsequently, targeted metabolomics was used to analyze musk in preparations with multiple reaction monitoring (MRM) mode by use gas chromatography coupled with triple quadrupole mass spectrometry (GC-QQQ MS). Results Two chemical markers named prasterone and androsterone have been selected and could be detected in all Compound Pien Tze Huang preparations (CPZHs) containing artificial musk, while the CPZHs containing natural musk did not detect two markers with S/N (signal to noise ratio) less than 3. Conclusion Our work provides an applicable approach to select the practical chemical markers for the assessment of musk in preparations to realize the traceability of musk in TCM and improve the quality control of musk-containing preparations.

Abstract:Emerging contaminants such as synthetic musks and UV-filters as ingredients personal care products were widely used in human daily life in Thailand. The occurrence and fate of four synthetic musks and nine UV-filters were investigated in eight full-scale sewage treatment plants (STPs) and their receiving aquatic environments in Bangkok and Pattaya, Thailand. All target compounds were detected in every single sample from STPs and surface water with magnitude from ng/L to μg/L. HHCB-lactone and HMS were found as the predominant musk and UV filter in influent and effluent of STPs, respectively. HHCB-lactone was also found with the highest concentration up to 79501 ng/g (dw) in the sludge. Low removal efficiency range from −37% (HHCB-lactone) to 58% (AHTN) were found for four musks in the STPs. The total emission of Σ4musks and Σ9UV-filters were estimated to be up to 16.7 mg/person/day and 0.28 mg/person/day by the STPs. Three synthetic musks and seven UV-filters were detected in fish from the receiving river. Concentration and emission of musks and UV filters found in this study from Thailand were much higher than those reported in many other countries worldwide. The preliminary ecological risk assessment showed that Musk xylene, 4-MBC and OC may pose high risk to aquatic organisms in the riverine and estuarine environment in Thailand.

Analytical and Bioanalytical Chemistry

Abstract:Concern about the environmental fate and potential effects of synthetic organic chemicals used in soaps, lotions, toothpaste, and other personal care products continues to increase. This review describes procedures used for the analysis of five classes of these compounds–synthetic musk fragrances, antimicrobials, ultraviolet filters, insect repellents, and parabens–in water, sediment, sewage sludge, air, and aquatic biota. The primary focus is on sample extraction and preparation methods for these compounds. Instrumental methods commonly used for these compounds are also discussed.

Abstract:Abstract Musk, the dried secretion from the musk sac gland which is located between the navel and genitals of mature male musk deer, is utilized as oriental medicine in east Asia. It has been utilized to treat conditions such as stroke, coma, neurasthenia, convulsions, and heart diseases in China since ancient times. This paper aims to provide a comprehensive overview of musk in zoology, chemical composition, pharmacology, clinical applications, and quality control according to the up-to-date literature. Studies found that musk mainly contains macrocyclic ketones, pyridine, steroids, fatty acids, amino acids, peptides, and proteins, whilst the main active ingredient is muscone. Modern pharmacological studies have proven that musk possesses potent anti-inflammatory effects, neuroprotective effects, anti-cancer effects, antioxidant effects, etc. Moreover, muscone, the main active ingredient, possesses anti-inflammatory, neuroprotective, antioxidant, and other pharmacological effects. In the quality control of musk, muscone is usually the main detection indicator, and the common analytical method is GC, and researchers have established novel and convenient methods such as HPLC-RI, RP-UPLC-ELSD, and Single-Sweep Polarography. In addition, quality evaluation methods based on steroids and the bioactivity of musk have been established. As for the identification of musk, due to various objective factors such as the availability of synthetic Muscone, it is not sufficient to rely on muscone alone as an identification index. To date, some novel technologies have also been introduced into the identification of musk, such as the electronic nose and DNA barcoding technology. In future research, more in vivo experiments and clinical studies are encouraged to fully explain the pharmacological effects and toxicity of musk, and more comprehensive methods are needed to evaluate and control the quality of musk.

Distribution of Typical Polycyclic Musks in Municipal Sewage

Abstract:The distribution and migration of two typical polycyclic musks,namely galaxolide(HHCB) and tonalide(AHTN) were investigated in the processes of three sewage treatment plants(STPs) in Beijing,China.The results show that HHCB and AHTN are 1 251.4 to 3 003.8 ng/L and 111.9 to 435.4 ng/L in the influent,and 457.2 to 729.9 ng/L and 35.5 to 190.9 ng/L in the effluent respectively.The average removal rates of HHCB and AHTN are 62.2% and 53.1% respectively.HHCB and AHTN in the sludge are 2.5 to 17.0 μg/g and 0.7 to 13.9 μg/g respectively.The polycyclic musks are removed mainly in biological treatment stage.The mechanism for their removal is basically adsorption to sludge.The potential environmental risk caused by the musk discharge from STPs as well as the reduction of the compounds in the excess sludge should be paid attention to.

Abstract:Twenty-nine surface sediments from Chaohu Lake in China and from its six main tributaries were sampled to investigate the concentrations of two important polycyclic musks (PCMs), 1,3,4,6,7,8-hexahydro-4,6,6,7,8,8-hexamethylcyclopenta(g)-2-benzopyran (galaxolide, HHCB) and 7-acetyl-1,1,3,4,4,6-hexamethyl-1,2,3,4-tetrahydronaphthalene (tonalide, AHTN), as well as the concentration of 4,6,7,8-hexahydro-4,6,6,7,8,8-hexamethylcyclopenta(g)-2-benzopyran-1-one (galaxolidon, HHCB-lactone), which is the main degradation product of HHCB. Except for the high concentrations of AHTN and HHCB measured in the Nanfei River (879 ng/g dw and 5,513 ng/g dw, respectively), the levels of AHTN and HHCB in the river sediments were 7.08–44.9 ng/g dw and 20.6–268 ng/g dw, respectively, which are slightly lower than those documented in various areas worldwide. The concentrations of AHTN and HHCB in the sediments of Chaohu Lake were one or two orders of magnitude lower than those in the tributary rivers and showed a clear regional distribution. The concentrations of HHCB-lactone were comparable to those of HHCB and presented a significant positive correlation with the concentrations of HHCB, suggesting that the HHCB-lactone originated directly from the degradation of HHCB in wastewater treatment plants (WWTPs) or in the natural environment. The diagnostic ratios of HHCB/AHTN and HHCB-lactone/HHCB and the enantiomeric fractions (EFs) of these PCMs showed that the direct origins of the target PCMs in the study area were municipal and industrial wastewaters discharged from adjacent cities or point sources and that the HHCB-lactone in sediment originated from the natural degradation of HHCB in the rivers and the lake. The results of the risk assessment showed that the PCMs in the watershed sediments were unlikely to pose a threat to aquatic species. However, the effluents of industrial and municipal wastewaters that are discharged into the Nanfei River should be investigated in future research.

Abstract:Musk xylene, ketone, ambrette, tibetene and moskene are synthetic nitroaromatic compounds. Because of their scent, they are used as substitutes for natural musk. In particular musk xylene and musk ketone are added to cosmetics, washing agents and other products. Acute toxicity of nitro musks has been estimated to be low. Knowledge on the toxicokinetics, chronic toxicity, as well as mutagenic and carcinogenic potential is based so far on only few studies. First evidence for the persistence of these lipophilic compounds in the environment and their potential to accumulate in organisms came from their identification in the aquatic ecosystem. Nitro musks have now been identified in mother's milk. The analysis of 391 samples from Southern Bavaria, Germany, in 1991 and 1992 revealed concentrations of musk xylene ranging from 0.01-1.22 mg/kg fat with a mean content of 0.10 mg/kg fat. This was 2-3 times higher than the mean contents of musk ketone or musk ambrette. Average levels of the nitro musks were in the range of those usually found for various chlorinated compounds like hexachlorocyclohexanes and cis-heptachloroepoxide. Because of their presence in cosmetic products and washing agents dermal absorption could be an important route of uptake of nitro musks in man. Toxicologic relevance of the present findings for humans is still unclear. In order to assess consequences for health, further investigation of effects caused by a continual uptake of small amounts of these substances is necessary.
