NO Oxidation States in Nonheme Iron Nitrosyls: A DMRG-CASSCF Study of {FeNO}6 –10 Complexes

Abstract:Building upon an earlier study of heme-nitrosyl complexes (Inorg. Chem. 2023, 62, 20496–20505), we have examined a wide range of nonheme {FeNO}6–10 complexes, as well as two dinitrosyl iron complexes (DNICs), using DMRG-CASSCF theory and DFT. We found striking parallels between heme and nonheme complexes in terms of molecular spin density profiles and the local oxidation state of the NO moiety. For spin densities, classic pure functionals were found to give the closest agreement to DMRG-CASSCF theory. Analysis of DMRG-CASSCF wave functions identified NO0 resonance forms as the largest contributors to both {FeNO}6 and {FeNO}7 complexes. Our results thus suggest that FeIII-NO0 and FeII-NO0 are the dominant electronic configurations for {FeNO}6 and {FeNO}7 complexes, respectively, mirroring our previous findings on heme-nitrosyl complexes. However, multiple strong-field ligands such as cyanide or thiolate bring about a substantial contribution from an NO− resonance form to the overall {FeNO}7 wave functions. A trigonal-bipyramidal S = 1 {FeNO}8 complex with an equatorial triscarbene ligand set was best described as a resonance hybrid of FeI-NO0 and FeII-NO−, echoing a similar description for the S = 0 {FeNO}8 heme complex {Fe[P](NO)}– (P = porphyrinato). Reduction to the corresponding S = 1/2 {FeNO}9 state was found to involve the whole molecule, i.e., both the metal and the NO, leading to an essentially FeI-NO− complex. Further reduction to the {FeNO}10 state was found to be primarily metal-centered, leading to a predominantly Fe0-NO− configuration. A linear correlation between the NO bond distance and NO stretching frequency was also observed, which could be mapped onto a similar correlation between the NO bond distance and NO π* occupancy, allowing a simple readout of the NO oxidation state from the NO bond distance.

Formation of the complex of nitrite with the ferriheme b beta-barrel proteins nitrophorin 4 and nitrophorin 7

Abstract:The interaction of ferriheme proteins with nitrite has recently attracted interest as a source for NO or other nitrogen oxides in mammalian physiology. However, met-hemoglobin (metHb), which was suggested as a key player in this process, does not convert nitrite unless small amounts of NO are added in parallel. We have recently reported that, in contrast, nitrophorins (NPs) convert nitrite as the sole substrate to form NO even at pH 7.5, which is an unprecedented case among ferrihemes [He, C., and Knipp, M. (2009) J. Am. Chem. Soc. 131, 12042-12043]. NPs, which comprise a class of unique heme b proteins from the saliva of the blood-sucking insect Rhodnius prolixus, appear in a number of concomitant isoproteins. Herein, the first spectroscopic characterization of the initial complexes of the two isoproteins NP4 and NP7 with nitrite is presented and compared to the data reported for metHb and met-myoglobin (metMb). Because upon nitrite binding, NPs, in contrast to metHb and metMb, continue to react with nitrite, resonance Raman spectroscopy and continuous wave electron paramagnetic resonance spectroscopy were applied to frozen samples. As a result, the existence of two six-coordinate ferriheme low-spin complexes was established. Furthermore, X-ray crystallography of NP4 crystals soaked with nitrite revealed the formation of an eta(1)-N nitro complex, which is in contrast to the eta(1)-O-bound nitrite in metMb and metHb. Stopped-flow kinetic experiments show that although the ligand dissociation constants of NP4 and NP7 (15-190 M(-1)) are comparable to those of metHb and metMb, the rates of ligand binding and release are significantly slower. Moreover, not only the reaction kinetics but also electron paramagnetic resonance spectroscopy reveals notable differences between the two isoproteins.
Spectroscopic characterization of mononitrosyl complexes in heme--nonheme diiron centers within the myoglobin scaffold (Fe(B)Mbs): relevance to denitrifying NO reductase.

Abstract:Denitrifying NO reductases are evolutionarily related to the superfamily of heme-copper terminal oxidases. These transmembrane protein complexes utilize a heme-nonheme diiron center to reduce two NO molecules to N2O. To understand this reaction, the diiron site has been modeled using sperm whale myoglobin as a scaffold and mutating distal residues Leu-29 and Phe-43 to histidines and Val-68 to a glutamic acid to create a nonheme Fe-B site. The impact of incorporation of metal ions at this engineered site on the reaction of the ferrous heme with one NO was examined by UV-vis absorption, EPR, resonance Raman, and FTIR spectroscopies. UV-vis absorption and resonance Raman spectra demonstrate that the first NO molecule binds to the ferrous heme, but while the apoproteins and Cu-I- or Zn-II-loaded proteins show characteristic EPR signatures of S = 1/2 six-coordinate heme {FeNO}(7) species that can be observed at liquid nitrogen temperature, the Fe-II-loaded proteins are EPR silent at >= 30K Vibrational modes from the heme [Fe-N-O] unit are identified in the RR and FUR spectra using (NO)-N-15 and (NO)-N-15-O-18. The apo and Cu-I-bound proteins exhibit v(FeNO) and v(NO) that are only marginally distinct from those reported for native myoglobin. However, binding of Fe-II at the Fe-B site shifts the heme v(FeNO) by 17 cm(-1) and the v(NO) by 50 cm(-1) to 1549 cm(-1). This low v(NO) is without precedent for a six-coordinate heme {FeNO}(7) species and suggests that the NO group adopts a strong nitroxyl character stabilized by electrostatic interaction with the nearby nonheme Fe-II. Detection of a similarly low v(NO) in the Zn-II-loaded protein supports this interpretation.

Abstract:Flavodiiron NO reductases (FNORs) are important enzymes in microbial pathogenesis, as they equip microbes with resistance to the human immune defense agent nitric oxide (NO). Despite many efforts, intermediates that would provide insight into how the non-heme diiron active sites of FNORs reduce NO to N2O could not be identified. Computations predict that iron-hyponitrite complexes are the key species, leading from NO to N2O. However, the coordination chemistry of non-heme iron centers with hyponitrite is largely unknown. In this study, we report the reactivity of two non-heme iron complexes with preformed hyponitrite. In the case of [Fe(TPA)(CH3CN)2](OTf)2, cleavage of hyponitrite and formation of an Fe2(NO)2 diamond core is observed. With less Lewis-acidic [Fe2(BMPA-PhO)2(OTf)2] (2), reaction with Na2N2O2 in polar aprotic solvent leads to the formation of a red complex, 3. X-ray crystallography shows that 3 is a tetranuclear iron-hyponitrite complex, [{Fe2(BMPA-PhO)2}2(μ-N2O2)](OTf)2, with a unique hyponitrite binding mode. This species provided the unique opportunity to us to study the interaction of hyponitrite with non-heme iron centers and the reactivity of the bound hyponitrite ligand. Here, either protonation or oxidation of 3 is found to induce N2O formation, supporting the hypothesis that hyponitrite is a viable intermediate in NO reduction.
Heme protonation affects iron-NO binding in the NO transport protein nitrophorin

Abstract:The nitrophorins (NPs) comprise an unusual group of heme proteins with stable ferric heme iron nitric oxide (Fe-NO) complexes. They are found in the salivary glands of the blood-sucking kissing bug Rhodnius prolixus, which uses the NPs to transport the highly reactive signaling molecule NO. Nuclear resonance vibrational spectroscopy (NRVS) of both isoform NP2 and a mutant NP2(Leu132Val) show, after addition of NO, a strong structured vibrational band at around 600 cm-1, which is due to modes with significant Fe-NO bending and stretching contribution. Based on a hybrid calculation method, which uses density functional theory and molecular mechanics, it is demonstrated that protonation of the heme carboxyl groups does influence both the vibrational properties of the Fe-NO entity and its electronic ground state. Moreover, heme protonation causes a significant increase of the gap between the highest occupied and lowest unoccupied molecular orbital by almost one order of magnitude leading to a stabilization of the Fe-NO bond.
Spectroscopic and Computational Study of A Nonheme Iron Nitrosyl Center in A Biosynthetic Model of Nitric Oxide Reductase

Abstract:A major barrier to understanding the mechanism of nitric oxide reductases (NORs) is the lack of a selective probe of NO binding to the nonheme FeB center. By replacing the heme in a biosynthetic model of NORs, which structurally and functionally mimics NORs, with isostructural ZnPP, the electronic structure and functional properties of the FeB nitrosyl complex was probed. This approach allowed observation of the first S=3/2 nonheme {FeNO}(7) complex in a protein-based model system of NOR. Detailed spectroscopic and computational studies show that the electronic state of the {FeNO}(7) complex is best described as a high spin ferrous iron (S=2) antiferromagnetically coupled to an NO radical (S=1/2) [Fe(2+)-NO(.)]. The radical nature of the FeB -bound NO would facilitate N-N bond formation by radical coupling with the heme-bound NO. This finding, therefore, supports the proposed trans mechanism of NO reduction by NORs.
Complex Interplay of Heme-Copper Oxidases with Nitrite and Nitric Oxide

Abstract:Nitrite and nitric oxide (NO), two active and critical nitrogen oxides linking nitrate to dinitrogen gas in the broad nitrogen biogeochemical cycle, are capable of interacting with redox-sensitive proteins. The interactions of both with heme-copper oxidases (HCOs) serve as the foundation not only for the enzymatic interconversion of nitrogen oxides but also for the inhibitory activity. From extensive studies, we now know that NO interacts with HCOs in a rapid and reversible manner, either competing with oxygen or not. During interconversion, a partially reduced heme/copper center reduces the nitrite ion, producing NO with the heme serving as the reductant and the cupric ion providing a Lewis acid interaction with nitrite. The interaction may lead to the formation of either a relatively stable nitrosyl-derivative of the enzyme reduced or a more labile nitrite-derivative of the enzyme oxidized through two different pathways, resulting in enzyme inhibition. Although nitrite and NO show similar biochemical properties, a growing body of evidence suggests that they are largely treated as distinct molecules by bacterial cells. NO seemingly interacts with all hemoproteins indiscriminately, whereas nitrite shows high specificity to HCOs. Moreover, as biologically active molecules and signal molecules, nitrite and NO directly affect the activity of different enzymes and are perceived by completely different sensing systems, respectively, through which they are linked to different biological processes. Further attempts to reconcile this apparent contradiction could open up possible avenues for the application of these nitrogen oxides in a variety of fields, the pharmaceutical industry in particular.
Molecular alignment in a liquid induced by a nonresonant laser field: Molecular dynamics simulation.

Abstract:We carried out molecular dynamics (MD) simulations for a dilute aqueous solution of pyrimidine in order to investigate the mechanisms of field-induced molecular alignment in a liquid phase. An anisotopically polarizable molecule can be aligned in a liquid phase by the interaction with a nonresonant intense laser field. We derived the effective forces induced by a nonresonant field on the basis of the concept of the average of the total potential over one optical cycle. The results of MD simulations show that a pyrimidine molecule is aligned in an aqueous solution by a linearly polarized field of light intensity I approximately 10(13) W/cm2 and wavelength lambda = 800 nm. The temporal behavior of field-induced alignment is adequately reproduced by the solution of the Fokker-Planck equation for a model system in which environmental fluctuations are represented by Gaussian white noise. From this analysis, we have revealed that the time required for alignment in a liquid phase is in the order of the reciprocals of rotational diffusion coefficients of a solute molecule. The degree of alignment is determined by the anisotropy of the polarizability of a molecule, light intensity, and temperature. We also discuss differences between the mechanisms of optical alignment in a gas phase and a liquid phase.

Characterization of ferrous FixL-nitric oxide adducts by resonance Raman spectroscopy

Abstract:Resonance Raman spectra of the nitric oxide adducts of the ferrous forms of two soluble truncations of Rhizobium meliloti FixL, FixL* and FixLN, are reported. At room temperature, four isotope sensitive vibrations are observed for both ferrous FixL*-NO and ferrous FixLN-NO. For FixL*-NO, they are observed at 558, 525, 450, and 1675 cm(-1) and are assigned to v(Fe-NO) of a six-coordinate nitrosyl adduct, v(Fe-NO) of a five-coordinate nitrosyl adduct, delta(Fe-NO) of a six-coordinate nitrosyl adduct, and v(N-O) of a five-coordinate nitrosyl adduct, respectively. Similar frequencies are observed for the FixLN-NO isotope sensitive bands. On the basis of the frequencies and spectral separation of the v(Fe-NO) and delta(Fe-NO) modes, the Fe-N-O unit is concluded to have a bent geometry similar to those observed for the nitrosyl adducts of ferrous hemoglobin and myoglobin. Both proteins can be converted to predominantly five-coordinate nitrosyl adducts by lowering the temperature. In low-temperature resonance Raman spectra of FixL*-NO and FixLN-NO, the 558 cm(-1) bands are significantly decreased in intensity and v(Fe-NO)5-c (the Fe-NO stretching vibration for the five-coordinate nitrosyl adduct) is observed at 529 and 526 cm(-1), respectively. Analysis of the v3 and v8 vibrations for these nitrosyl adducts also supports the presence of both five- and six-coordinate nitrosyl adducts of FixL* and FixLN at room temperature and the conversion to predominantly five-coordinate nitrosyl adducts at low temperatures. This temperature-dependent interconversion is reversible. The possible physiological relevance of the thermally accessible five-coordinate state is discussed. The width of v(Fe-NO)6-c at half-height is 1.3 times broader in FixLN-NO than in FixL*-NO, suggesting that the Fe-N-O geometry is more homogeneous in FixL*-NO. In low-temperature spectra of FixLN-NO, a second v(N-O)5-c band is observed, indicating that more than one conformation is attainable in the five-coordinate FixLN-NO. This second v(N-O)5-c is not observed for five-coordinate FixL*-NO, further suggesting a more conformationally restricted nitrosyl heme in FixL*. These variations in the vibrations involving the Fe-NO unit indicate that the kinase domain influences the heme structure. The spectral differences are discussed in terms of the interdomain interactions that result in ligation-dependent mediation of the kinase activity.
Effect of Outer-Sphere Side Chain Substitutions on the Fate of the Trans Iron-Nitrosyl Dimer in Heme/Nonheme Engineered Myoglobins (fe(b)mbs): Insights into the Mechanism of Denitrifying No Reductases

Abstract:Denitrifying NO reductases are transmembrane protein complexes that utilize a heme/nonheme diiron center at their active sites to reduce two NO molecules to the innocuous gas N2O. Fe(B)Mb proteins, with their nonheme iron sites engineered into the heme distal pocket of sperm whale myoglobin, are attractive models for studying the molecular details of the NO reduction reaction. Spectroscopic and structural studies of Fe(B)Mb constructs have confirmed that they reproduce the metal coordination spheres observed at the active site of the cytochrome c-dependent NO reductase from Pseudomonas aeruginosa. Exposure of Fe(B)Mb to excess NO, as examined by analytical and spectroscopic techniques, results primarily in the formation of a five-coordinate heme-nitrosyl complex without N2O production. However, substitution of the outer-sphere residue Ile107 with a glutamic acid (i.e., I107E) decreases the formation rate of the five-coordinate heme-nitrosyl complex and allows for the substoichiometric production of N2O. Here, we aim to better characterize the formation of the five-coordinate heme-nitrosyl complex and to explain why the level of N2O production increases with the I107E substitution. We follow the formation of the five-coordinate heme-nitrosyl inhibitory complex through the sequential exposure of Fe(B)Mb to different NO isotopomers using rapid-freeze-quench resonance Raman spectroscopy. The data show that the complex is formed by the displacement of the proximal histidine by a new NO molecule after the weakening of the Fe(II)-His bond in the intermediate six-coordinate low-spin (6cLS) heme-nitrosyl complex. These results lead us to explore diatomic migration within the scaffold of myoglobin and whether substitutions at residue 107 can be sufficient to control access to the proximal heme cavities. Results on a new Fe(B)Mb construct with an I107F substitution (Fe(B)Mb3) show an increased rate for the formation of the five-coordinate low-spin heme-nitrosyl complex without N2O production. Taken together, our results suggest that production of N2O from the [6cLS heme {FeNO}(7)/{Fe(B)NO}(7)] trans iron-nitrosyl dimer intermediate requires a proton transfer event facilitated by an outer-sphere residue such as E107 in Fe(B)Mb2 and E280 in P. aeruginosa cNOR.
One‐Electron NO to N2O Pathways via Heme Models and Lewis Acid: Metal Effects and Differences from the Enzymatic Reaction

Abstract:Some pathogens use heme‐containing nitric oxide reductases (NORs) to reduce NO to N2O as their defense mechanism to detoxify NO and reduce nitrosative stress. This reduction is also significant in the global N cycle. Our previous experimental work showed that Fe and Co porphyrin NO complexes can couple with external NO to form N2O when activated by the Lewis acid BF3. A key difference from conventional two‐electron enzymatic reaction is that one electron is sufficient. However, a complete understanding of the entire reaction pathways and the more favorable reactivity for Fe remains unknown. Here, we present a quantum chemical study to provide such information. Our results confirmed Fe's higher experimental reactivity, showing advantages in all steps of the reaction pathway: easier metal oxidation for NO reduction and N‐O cleavage as well as a larger size to expedite the N/O coordination mode transition. The Co system, with a similar product energy as the enzyme, shows potential for further development in catalytic NO coupling. This work also offers the first evidence that this new one‐electron NO reduction is both kinetically competitive and thermodynamically more favorable than the native pathway, supporting future initiatives in optimizing NO reduction agents in biology, environment, and industry.

Photoinduced linkage isomers in a model ruthenium nitrosyl complex: Identification and assignment of vibrational modes

Abstract:Photoinduced NO-linkage isomers were investigated in the solid state of labelled trans-[Ru(14/15NO)(py4)F](ClO4)2 complex by combined IR-spectroscopy and DFT calculations. Based on the experimental data and the DFT calculations of this isotopically labelled 14/15NO nitrosyl compound, we present a complete assignment of the vibrational bands of three nitrosyl linkage isomers in a range from 4000 to 200 cm-1. The calculated IR-spectra match well with the experimental data allowing reliable assignment of the vibrational bands. The structural change from the Ru-NO (GS) to the Ru-ON (MS1) and Ru-η2-(NO) (MS2) linkage configuration leads to the downshift of the ν(NO) and ν(Ru-(NO)) bands, and a corresponding increase of the energy of the ν(Ru-F) band. The shift of the bands corresponds to the change of the Ru-(NO) and Ru-F bond lengths: increase of the Ru-(NO) bond length leads to the decrease of the energy of the ν(Ru-(NO)) band; decrease of the Ru-F bond length leads to the increase of the energy of the ν(Ru-F) band. These observations can be extrapolated to the family of related nitrosyl complexes and therefore be used for the qualitative prediction of the Ru-(NO) and Ru-Ltrans-to-NO bond lengths of different linkage isomers in the framework of one complex. While the formation of linkage isomers is a reversible process, long-time irradiation sometimes induces irreversible reactions such as the release of NO. Here, we show that the photolysis of trans-[Ru(14/15NO)(py4)F](ClO4)2 in KBr pellets may lead to the release of nitrous oxide N2O, conceivably through the formation of a {Ru-(κ2-ONNO)} intermediate.

Abstract:Abstract Uncommon metal oxidation states in porphyrinoid cofactors are responsible for the activity of many enzymes. The F 430 and P450nor co‐factors, with their reduced Ni I ‐ and Fe III ‐containing tetrapyrrolic cores, are prototypical examples of biological systems involved in methane formation and in the reduction of nitric oxide, respectively. Herein, using a comprehensive range of experimental and theoretical methods, we raise evidence that nickel tetraphenyl porphyrins deposited in vacuo on a copper surface are reactive towards nitric oxide disproportionation at room temperature. The interpretation of the measurements is far from being straightforward due to the high reactivity of the different nitrogen oxides species (eventually present in the residual gas background) and of the possible reaction intermediates. The picture is detailed in order to disentangle the challenging complexity of the system, where even a small fraction of contamination can change the scenario.
Phenoxide coordination to Fe(III) tetraphenylporphyrin: Exploring antibacterial and electrocatalytic HER activity

Abstract:Herein, we report synthesis, solid and solution state structure as well as properties of three five coordinate (unsubstituted, 4-NO 2 substituted and 2,4,6 trinitro substituted) phenoxide axially ligated to Fe(III)-tetraphenyl porphyrins. Spectroscopic characterization revealed high-spin (S = 5/2) nature of the phenoxide bound Fe(III) porphyrins ( Fe1 and Fe2 ). However, the 2,4,6 trinitro substituted phenoxide axially ligated Fe(III) porphyrin ( Fe3 ) demonstrate spin admixed nature (S = 5/2 , 3/2) with predominate high spin contribution. (Phenolato)(porphinato)iron(III) complexes merit attention owing to structural analogy with the catalase enzyme. In this study we investigated their antibacterial as well as electrocatalytic hydrogen evolution reaction (HER) activity. Fe1 demonstrate potent antibacterial activity against gram-negative E. coli (MTCC 443) and gram-positive Staphylococcus aureus (MTCC 3160) human pathogenic bacteria. It also produce catalytic current for electrocatalytic HER activities upon gradual addition of PTSA ( para -tolunesulphonic acid) as proton source. Compounds Fe2 and Fe3 also produce enhanced catalytic wave which originate possibly from reduction of proton coupled with reduction of –NO 2 group.

Vibrational properties of heme-nitrosoalkane complexes in comparison with those of their HNO analogs, and reactivity studies towards nitric oxide and Lewis acids

Abstract:C-Nitroso compounds (RNO, R = alkyl and aryl) are byproducts of drug metabolism and bind to heme proteins, and their heme-RNO adducts are isoelectronic to ferrous nitroxyl (NO-/HNO) complexes. Importantly, heme-HNO compounds are key intermediates in the reduction of NO to N2O and nitrite to ammonium in the nitrogen cycle. Ferrous heme-RNO complexes act as stable analogs of these species, potentially allowing for the investigation of the vibrational and electronic properties of unstable heme-HNO intermediates. In this paper, a series of six-coordinate ferrous heme-RNO complexes (where R = iPr and Ph) were prepared using the TPP2- and 3,5-Me-BAFP2- co-ligands, and tetrahydrofuran, pyridine, and 1-methylimidazole as the axial ligands (bound trans to RNO). These complexes were characterized using different spectroscopic methods and X-ray crystallography. The complex [Fe(TPP)(THF)(iPrNO)] was further utilized for nuclear resonance vibrational spectroscopy (NRVS), allowing for the detailed assignment of the Fe-N(R)O vibrations of a heme-RNO complex for the first time. The vibrational properties of these species were then correlated with those of their HNO analogs, using DFT calculations. Our studies support previous findings that RNO ligands in ferrous heme complexes do not elicit a significant trans effect. In addition, the complexes are air-stable, and do not show any reactivity of their RNO ligands towards NO. So although ferrous heme-RNO complexes are suitable structural and electronic models for their HNO analogs, they are unsuitable to model the reactivity of heme-HNO complexes. We further investigated the reaction of our heme-RNO complexes with different Lewis acids. Here, [Fe(TPP)(THF)(iPrNO)] was found to be unreactive towards Lewis acids. In contrast, [Fe(3,5-Me-BAFP)(iPrNO)2] is reactive towards all of the Lewis acids investigated here, but in most cases the iron center is simply oxidized, resulting in the loss of the iPrNO ligand. In the case of the Lewis acid B2(pin)2, the reduced product [Fe(3,5-Me-BAFP)(iPrNH2)(iPrNO)] was identified by X-ray crystallography.

Abstract:Metalloporphyrins and imidazole(ate) have long been employed to mimic metalloenzymes such as five-coordinate heme complexes. In this work, several five-coordinate manganese(II) porphyrin complexes utilizing sterically hindered 2-methylimidazole (2-MeHIm) or 2-methylimidazolate (2-MeIm ̅) as axial ligand are isolated and characterized by single-crystal X-ray, UV–vis, DFT and electronic paramagnetic resonance (EPR) spectroscopies to explore their geometric and electronic structures. Both UV–vis and crystal structural investigations revealed noteworthy differences between the two species (e.g. the unusual Soret band split of 2-MeIm ̅ derivatives) which are consistent with the stronger ligand nature of 2-methylimidazolate. Low temperature (4 K) EPR measurements have been carried out for all the products in both solid and solution states which gave characteristic signals of a high-spin (S = 5/2) d 5 system with g⊥ ≈ 5.9 and g// ≈ 2.0. Notably, the positive correlation between λ (E/D) and the numbers of hyperfine splittings are confirmed by double measurements e.g. [Mn(TPP)(2-MeHIm)] exhibits two more hyperfine splittings than [Mn(TMP)(2-MeIm ̅)] ̅ with a larger λ (0.018 vs. 0.002).

Abstract:The dimerization of nitrogen monoxide (NO) is highly relevant in homo- and heterogeneous biochemical and environmental redox processes, but a broader understanding is challenged by the endergonic nature of this equilibrium. The present work describes NO-dimerization leveraged by structurally constrained aluminum and metal-ligand cooperativity at the anionic calix[4]pyrrolato aluminate(III). Quantum chemical calculations reveal the driving force for N-N bond formation, while reactivity tests shed light on subsequent redox chemistry and NO decomposition at metal surfaces. Inhibiting the dimerization pathway by saturating NO's unpaired electron with a phenyl group (nitrosobenzene) allows trapping the 1,2‐adduct as a key intermediate. Elevated temperatures result in an unprecedented and high-yielding rearrangement of the calix[4]pyrrolato ligand scaffold. Kinetic and theoretical studies provide a comprehensive picture of the rearrangement mechanism and delineate systematics for ring modification of the prominent calix[4]pyrrole macrocycle.
