Abstract:Wistar rats were fed a control diet or a diet containing either cholestyramine or high fat and cholesterol throughout gestation and the first 14 d of lactation. New-born litters were cross-fostered from rats fed the control diet to rats fed either cholestyramine or high fat and cholesterol, or from rats fed cholestyramine to rats fed the control diet. Hepatic hydroxymethylglutaryl-coenzyme A (HMG-CoA) reductase activity, plasma cholesterol and triglycerides were assayed on gestation d 20 and postnatal d 8, 14, 22 and 30. Cholestyramine had no effect on maternal or fetal plasma lipid levels but increased fetal hepatic HMG-CoA reductase activity by approximately 50%. The increased reductase activity persisted on postnatal d 8 and 14. Control pups suckled by dams fed cholestyramine also had significantly increased HMG-CoA reductase activities on postnatal d 8 and 14. The high fat and cholesterol diet significantly increased maternal plasma cholesterol but had no effect on HMG-CoA reductase activity in the fetus or suckling pups. Neither cholestyramine nor high fat and cholesterol altered the rat milk cholesterol levels. The studies demonstrate that HMG-CoA reductase activity in the developing rat can be altered by factors dependent on maternal diet. They do not support a hypothesis for regulation by maternal dietary or milk cholesterol supply.
Early Tumor Detection Afforded by in Vivo Imaging of Near-Infrared Ii Fluorescence

Abstract:Cell-intrinsic reporters such as luciferase (LUC) and red fluorescent protein (RFP) have been commonly utilized in preclinical studies to image tumor growth and to monitor therapeutic responses. While extrinsic reporters that emit near infrared I (NIR-I: 650-950 nm) or near-infrared II (NIR-II: 1000 -1700 nm) optical signals have enabled minimization of tissue autofluorescence and light scattering, it has remained unclear as to whether their use has afforded more accurate tumor imaging in small animals. Here, we developed a novel optical imaging construct comprised of rare earth lanthanide nanoparticles coated with biodegradable diblock copolymers and doped with organic fluorophores, generating NIR-I and NIR-II emissive bands upon optical excitation. Simultaneous injection of multiple spectrally-unique nanoparticles into mice bearing tumor implants established via intraperitoneal dissemination of LUC+/ RFP+ OVCAR-8 ovarian cancer cells enabled direct comparisons of imaging with extrinsic vs. intrinsic reporters, NIR-II vs. NIR-I signals, as well as targeted vs. untargeted exogenous contrast agents in the same animal and over time. We discovered that in vivo optical imaging at NIR-II wavelengths facilitates more accurate detection of smaller and earlier tumor deposits, offering enhanced sensitivity, improved spatial contrast, and increased depths of tissue penetration as compared to imaging with visible or NIR-I fluorescent agents. Our work further highlights the hitherto underappreciated enhancements in tumor accumulation that may be achieved with intraperitoneal as opposed to intravenous administration of nanoparticles. Lastly, we found discrepancies in the fidelity of tumor uptake that could be obtained by utilizing small molecules for in vivo as opposed to in vitro targeting of nanoparticles to disseminated tumors. Digital optical imaging of green fluorescent proteins for tracking vascular gene expression: feasibility study in rabbit and human cell models.

Abstract:PURPOSE: To investigate the feasibility of using a sensitive digital optical imaging technique to detect green fluorescent protein (GFP) expressed in rabbit vasculature and human arterial smooth muscle cells. MATERIALS AND METHODS: A GFP plasmid was transfected into human arterial smooth muscle cells to obtain a GFP-smooth muscle cell solution. This solution was imaged in cell phantoms by using a prototype digital optical imaging system. For in vivo validation, a GFP-lentivirus vector was transfected during surgery into the carotid arteries of two rabbits, and GFP-targeted vessels were harvested for digital optical imaging ex vivo. RESULTS: Optical imaging of cell phantoms resulted in a spatial resolution of 25 mum/pixel. Fluorescent signals were detected as diffusely distributed bright spots. At ex vivo optical imaging of arterial tissues, the average fluorescent signal was significantly higher (P < .05) in GFP-targeted tissues (mean <plus/minus> SD, 9,357.3 absolute units of density +/- 1,001.3) than in control tissues (5,633.7 absolute units of density +/- 985.2). Both fluorescence microscopic and immunohistochemical findings confirmed these differences between GFP-targeted and control vessels. CONCLUSION: The digital optical imaging system was sensitive to GFPs and may potentially provide an in vivo imaging tool to monitor and track vascular gene transfer and expression in experimental investigations.

Abstract:Objective:To establish a human lung carcinoma cell line with stable expression of green fluorescent protein, and discuss the application of in vivo biofluorescence imaging system in subcutaneously xenografted tumor model. Methods:Human lung carcinoma NCI-H460-GFP cell line was established by tranfection of NCI-H460 cells with green fluorescent protein (GFP) gene via lentiviral vector. The NCI-H460-GFP cells were inoculated into the nude mice to prepare the subcutaneously xenografted tumor model. The dynamic growth of xenografted tumor was observed using in vivo fluorescence imaging system for subsequent 5 weeks. Results:We successfully built up a human lung carcinoma NCI-H460-GFP cell line with transfection rate of nearly 100%, which can stably express high level of GFP protein in vitro and in vivo. In vivo fluorescence imaging found that the mean fluorescent intensity gradually increased with the increase in tumor volume in nude mice from one week to four weeks, however, the mean fluorescent intensity tended to decline when the tumor had apparent necrosis at the fifth week. Conclusion:NCI-H460-GFP cell line with stable expression of GFP and its animal model provide useful experimental materials for lung cancer study. The in vivo fluorescence imaging system could be used to quantitatively eva-luate the tumor growth in vivo.

Abstract:A distributed algorithm is described for finding a common fixed point of a family of <inline-formula> <tex-math notation="LaTeX">$m>1$</tex-math></inline-formula> nonlinear maps <inline-formula> <tex-math notation="LaTeX">$M_i:\mathbb {R}^n\rightarrow \mathbb {R}^n$</tex-math></inline-formula> assuming that each map is a paracontraction and that at least one such common fixed point exists. The common fixed point is simultaneously computed by <inline-formula><tex-math notation="LaTeX">$m$</tex-math></inline-formula> agents assuming each agent <inline-formula><tex-math notation="LaTeX">$i$</tex-math></inline-formula> knows only <inline-formula> <tex-math notation="LaTeX">$M_i$</tex-math></inline-formula>, the current estimates of the fixed point generated by its neighbors, and nothing more. Each agent recursively updates its estimate of a fixed point by utilizing the current estimates generated by each of its neighbors. Neighbor relations are characterized by a time-varying directed graph <inline-formula><tex-math notation="LaTeX">$\mathbb {N}(t)$</tex-math></inline-formula>. Establishment of Orthotopic Lung Cancer Model Expressing Enhanced Green Fluorescent Protein

Abstract:BACKGROUND AND OBJECTIVE:In vivo molecular imaging with mouse model could continuously and in real-time monitor the changes of the tumor. The aim of this study is to establish stable enhanced green fluorescent protein (EGFP) expressing NCI-H460 cell lines and relevant mouse model via orthotopic transplantation, and to study the characteristic of this model and the quantitative detection method of the primary tumor and metastatic lesions.METHODS:Human lung cancer NCI-H460 cells were transfected with retroviral vector containing the EGFP. Stable high-level expression of EGFP was maintained in the subcutaneously-growing tumors. Fragments of the subcutaneously growing tumor, which were comprised of EGFP-expressing cells, were implanted by surgical orthotopic implantation (SOI) in the lung of nude mice. The dynamic growth of orthotopic tumor was observed using in vivo fluorescence imaging. The correlation of fluorescence area and tumor volume was tested.RESULTS:After the model established, green fluorescent can be observed through the flap in day 7. Tumor formation rate was 100%. Mean survival time of tumor-bearing nude mice was 34.2 days. The metastasis sites were the contralateral lung, mediastinal and hilar lymph nodes, pleura and diaphragm; metastasis rates were 87.5%, 75%, 25% and 12.5%, respectively. Tumor volume and fluorescence area was correlated (r = 0.873, P = 0.001).CONCLUSIONS:The nude mouse model bearing orthotopic human lung cancer expressing EGFP has been successfully established. The model might be used for further molecular studies on tumor metastasis, angiogenesis and also be applied to anti-tumor drug screening in preclinical stage.
Optical imaging of green fluorescent protein markers for tracking vascular gene expression: a feasibility study in human tissue-like phantoms

Abstract:Vascular gene therapy is an exciting approach to the treatment of cardiovascular diseases. However, to date, there are no imaging modalities available for non-invasive detection of vascular gene expression. We have developed an optical imaging method to track vascular gene expression by detecting fluorescent signals emitted from arterial walls following gene transfer. To investigate the feasibility of this new technique, we performed experiments on a set of human tissue-like phantoms using a common biological marker in gene therapy, the green fluorescent protein (GFP). The phantoms were constructed to mimic the arterial geometry beneath a tissue layer. Human smooth muscle cells transfected with GFP were embedded in a capillary tube in the phantom. Monte Carlo modeling of the phantom experiment was performed to optimize the performance of the optical imaging system. We compared the fluence rates among three types of light beams, including ring beam, Gaussian beam, and flat beam. The results showed that our optical imaging system was able to detect fluorescent signals up to 5-mm depth in the phantom, and that flat beam geometry would produce the optimum fluorescence remittance. This study provides valuable insights for improvements to the optical imaging system and refinement of the new technique to non-invasively detect/track vascular gene expression.

Abstract:Non-invasive in vivo imaging is emerging as an important tool for basic and preclinical research. Near-infrared (NIR) fluorescence dyes and probes have been used for non-invasive optical imaging since in the NIR region absorption and auto fluorescence by body tissue is low, thus permitting for greater penetration depths and high signal to noise ratio. Currently, cell tracking systems rely on labeling cells prior to injection or administering probes targeting the cell population of choice right before imaging. These approaches do not enable imaging of tumor growth, as the cell label is diluted during cell division. In this study we have developed cell lines stably expressing the far-red fluorescence protein E2-Crimson, thus enabling continuous detection and quantification of tumor growth. In a xenograft rat model, we show that E2-Crimson expressing cells can be detected over a 5 week period using optical imaging. Fluorescence intensities correlated with tumor volume and weight and allowed for a reliable and robust quantification of the entire tumor compartment. Using a novel injection regime, the seeding of MDA-MB-231 breast cancer cells in the lungs in a rat model was established and verified.

Abstract:Objective: To establish an eGFP-gene-marked tumor cells model in murine for long-term in vivo research and to study its preliminary application.Methods:High titer eGFP retrovirus (RV) was prepared and was used to transfect mouse leukemia cells—P388 cells. P388-eGFP clones were obtained by limited dilution method. Wild type P388 (wtP388) was used as control; DBA/2 mice were inoculated with P388-eGFP abdominally or intravenously (n=10). eGFP~+ cells from abdominal cavities or bone marrow of the dyed mice were used to inoculate new mice. eGFP~+ P388 cells were studied in different tissues by Flow cytometry (FCM), semi-solid culture, fluorescence microscope, light microscope and PCR, etc.Results: It was found that 80.2% P388 cells were eGFP~+ 48 h after retrovirus transfection. P388-eGFP clone selection increased the eGFP~+ rate to 99.2%, and the rate remained at 95.2% after 3 months′ passaging. After inoculation in abdominal cavities for (56.3±1.25) d, the eGFP~+ rate of P88-eGFP cells increased to (93.3±0.50)% (n=3). eGFP gene marking had no influence on the bioactivity of P388 cells. Paraformaldehydum and liquid nitrogen both effectively protected the fluorescence signals in the tissues. Perfusion fixation method was suitable for various kinds of pathological observation approach. FCM could be used to observe eGFP~+ cells in the bone marrow, liver, spleen, thymus, peripheral blood, ascitic fluid,etc. The dynamic changes of eGFP~+ cells could reflect the response of tumor cells to treatment. eGFP~+ cells could be used for in vitro culture and in vivo transplantation. PCR is sensitive in detecting eGFP~+ tumor cells in tissues. Conclusion: The present model allows for long-term, dynamic and sensitive tracing of fluorescence-labeled cells in vivo, and can be widely used in tumor and cell engineering research.
Orthotopic metastatic (MetaMouse) models for discovery and development of novel chemotherapy

Abstract:Currently-used rodent tumor models, including transgenic tumor models, or subcutaneously growing human tumors in immunodeficient mice, do not sufficiently represent clinical-cancer, especially with regard to metastasis and drug sensitivity. In order to obtain clinically-accurate models, we have developed the technique of surgical orthotopic implantation (SOI) to transplant histologically intact fragments of human cancer, including tumors taken directly from the patient, to the corresponding organ of immunodeficient rodents. It has been demonstrated in approx 100 publications describing 10 tumor types that SOI allows the growth and metastatic potential of the transplanted tumors to be expressed and reflect clinical cancer. These clinically-accurate and relevant SOI models of human cancer have enabled discovery and evaluation of novel antitumor and antimetastatic agents including antiangiogenic drugs. The green fluorescent protein (GFP) and red fluorescent protein (RFP) genes, cloned from bioluminescent organisms, have now been introduced into a series of human and rodent cancer cell lines in vitro to stably express GFP and RFP in vivo in SOI mouse models. With these fluorescent tools, tumors and metastasis in host organs can be externally imaged down to the single-cell level. The combination of fluorescent protein-based imaging in SOI models enables real-time antitumor, antimetastatic, and antiangiogenic drug evaluation including high-throughput in vivo screening. These SOI models are uniquely useful for innovative drug discovery and mechanism studies and serve as a bridge linking preclinical and clinical drug development.
Assessment of a Novel VEGF Targeted Agent Using Patient-Derived Tumor Tissue Xenograft Models of Colon Carcinoma with Lymphatic and Hepatic Metastases

Abstract:The lack of appropriate tumor models of primary tumors and corresponding metastases that can reliably predict for response to anticancer agents remains a major deficiency in the clinical practice of cancer therapy. It was the aim of our study to establish patient-derived tumor tissue (PDTT) xenograft models of colon carcinoma with lymphatic and hepatic metastases useful for testing of novel molecularly targeted agents. PDTT of primary colon carcinoma, lymphatic and hepatic metastases were used to create xenograft models. Hematoxylin and eosin staining, immunohistochemical staining, genome-wide gene expression analysis, pyrosequencing, qRT-PCR, and western blotting were used to determine the biological stability of the xenografts during serial transplantation compared with the original tumor tissues. Early passages of the PDTT xenograft models of primary colon carcinoma, lymphatic and hepatic metastases revealed a high degree of similarity with the original clinical tumor samples with regard to histology, immunohistochemistry, genes expression, and mutation status as well as mRNA expression. After we have ascertained that these xenografts models retained similar histopathological features and molecular signatures as the original tumors, drug sensitivities of the xenografts to a novel VEGF targeted agent, FP3 was evaluated. In this study, PDTT xenograft models of colon carcinoma with lymphatic and hepatic metastasis have been successfully established. They provide appropriate models for testing of novel molecularly targeted agents.
Establishment of Green Fluorescent Protein-Expressing Hepatocellular Carcinoma Cell Lines with Different Metastatic Potential: Relevant Models for in Vivo Monitoring of Metastasis and Angiogenesis

Abstract:Purpose To establish stable green fluorescent protein (GFP)-expressing metastatic human hepatocellular carcinoma (HCC) cell lines with different metastatic potential for long-term in vivo studies of metastasis and angiogenesis. Methods The pIRES2-EGFP vector, which contains an enhanced GFP gene, was transfected into MHCC97-H and MHCC97-L, HCC cell lines with different metastatic potential. The stability of GFP expression, basic biological characteristics, invasion abilities in vitro, and spontaneous metastasis in vivo of the new cell lines (MHCC97-HG and MHCC97-LG) were studied. Microvessel density (MVD) of orthotopic implanted tumors was compared by anti-CD31 immunohistochemical staining, and real-time angiogenesis and metastasis of GFP-transfected tumors were detected by intravital fluorescent microscope. Results The GFP-transfected cell lines stably expressed green fluorescence in the absence of G418 over a 36-day period. Compared with the parental cell lines, they exhibited no distinct differences in biological characteristics. MHCC97-HG showed more aggressive invasion and spontaneous metastatic behavior than MHCC97-LG, and even its parental cell line, MHCC97-H ( P <0.01). MVD levels induced by MHCC97-HG orthotopic implanted tumors were significantly higher than MHCC97-LG ( P <0.01). Real-time angiogenesis and sequential steps of metastasis could be detected clearly under intravital fluorescent microscope. Conclusions These two stable GFP-expressing HCC cell lines with the same genetic background and different metastatic potential were established, which could be useful models for monitoring metastasis and angiogenesis of HCC.
Model Development of Visible Orthotopic Mouse Bladder Tumor

Abstract:Objective To label a human bladder cancer cell line and establish a novel human bladder cancer mouse model in which the fluorescent signal of the orthotopic tumor can be detected using the whole-body optical imaging system. Method T24, a human bladder transitional cell carcinoma cell line, were transfected with GFP plasmid to screen stable GFP expressing clones. The latter were implanted into the wall of bladder of the nude mice. The growth, invasion, and metastasis of the tumour were observed and evaluated by the whole-body optical imaging system. The findings were compared with those of HE staining on routine paraffin sections. Result GFP-labeled tumor cells displayed green fluorescence under fluorescent microscopy and showed stable GFP expression in vitro and in vivo. One week after in situ transplantation of 5×105 T24 cells, the new bladder cancer could be observed and evaluated by the whole body optical imaging system. Two weeks later, the new bladder tumor could be palpated with hands. Four weeks later, the metastasis to regional drained lymph node in pelvic and retroperitoneal lymph node occurred. The new bladder tumor growth and metastasis were easily observed and accurately evaluated by fluorescent microscope.Conclusion GFP-labeled tumor cells displayed green fluorescence under fluorescent microscopy and showed stable GFP expression. GFP labeled T24 and the novel human bladder cancer model described here provided a simple and reliable tool for studying the human bladder cancer in vivo.

Abstract:BACKGROUND/AIM: Although the 5-year survival rate for localized prostate cancer is nearly 100%, prognosis for patients with metastases, of which the bone is the most common site, is poor. In order to evaluate efficacy of treatments against metastatic prostate cancer, experimental tibia-bone-metastasis mouse models of prostate cancer have been previously established. In the present study, we used a novel procedure for establishment of an experimental tibiabone metastasis mouse model, with human PC-3 prostate cancer expressing green fluorescent protein (GFP), that more closely matches prostate cancer growing in the bone.MATERIALS AND METHODS: PC-3 human prostate cancer cells, labeled with GFP, were initially subcutaneously injected into the flank of five male nude mice to obtain tumor tissues. Once the tumor tissue grew larger than 10 mm in diameter, the tumor tissue was harvested and minced into fragments of 1 mm3 A 1-mm hole was made in the proximal left tibia of eight male nude mice, using the tip of a 5-mm blade, and a tumor fragment was implanted into the hole for an exact fit. Tumor size was measured once a week, by non-invasive imaging of GFP fluorescence. The mice were sacrificed four weeks after tumor implantation.RESULTS: Tumors grew in 8 out of 8 mice (100%). All tumors were non-invasively detectable with GFP fluorescence, through the skin. Increased tumor growth in the tibia was observed every week.CONCLUSION: The establishment in the tibia of the novel experimental bone-metastatic mouse model of human prostate cancer enables facile screening, in a clinically-relevant system, of improved therapeutics for this recalcitrant disease.

Luciferase/mCherry-labeled cholangiocarcinoma cells and 24 mice with subcutaneously implanted cholangiocarcinoma cells were treated by: (i) a combination of green fluorescent protein (GFP)/HSV-TK/plasmid gene transfection followed by RFH and ganciclovir exposure; (ii) gene therapy alone; (iii) RFH alone; and (iv) phosphate-buffered saline (PBS) to serve as a control (6 mice per group). Cell viabilities were quantified by MTS assay. Bioluminescent optical imaging and transcutaneous ultrasound imaging were used to follow up the tumor growth over 2 weeks. For in-vivo technical validation of the new technique, pigs in two groups received either intrabiliary delivery of GFP/HSV-TK/plasmid using a needle-integrated balloon catheter flowed by RFH enhancement (n = 6) or gene delivery only (n = 6). GFP signal in bile duct walls were quantified by optical imaging.The non- parametric Mann-Whitney U test was used for statistical comparison of variables. MTS assay demonstrated the lowest cell proliferation in combination therapy compared with three control groups (24.0 ± 5.3% vs 41.9 ± 6.4% vs 72.0 ± 9.0% vs 100%, p<0.001). GFP optical imaging confirmed that GFP/HSV-TK gene expression peaked at day 2 after gene transfection. Bioluminescent optical imaging demonstrated the decreased bioluminescence signals in tumors of combination therapy, compared with other control groups (0.71 ± 0.16 vs 1.46 ± 0.21 vs 2.10 ± 0.27 vs 2.36 ± 0.28, p<0.001). Ultrasound imaging showed the smallest tumor volumes of the combination therapy group, compared with other groups (0.90 ± 0.21 VS 1.61 ± 0.21 VS 2.22 ± 0.25 VS 2.50 ± 0.30, p<0.001). optical imaging confirmed higher GFP expression in the bile ducts with RFH enhancement (signal to noise ratio: 1.28 ± 0.61 VS 0.49 ± 0.31, P<0.05). It is feasible to use intratumoral RFH to enhance GFP/HSV-TK/plasmid gene expression and HSV-TK/GCV gene therapy of cholangiocarcinomas.This concept may pave a new avenue for managing pancreatobiliary malignancies.(supported by an NIH RO1EBO12467)
Value of Life Liabilities with Embedded Options : an Application to a Portfolio of Italian Insurance Policies

Abstract:In this paper we present a pricing application analyzing, in a contingent-claims framework, the two most common types of life policies sold in Italy during the last two decades. These policies, characterized by different premium payment styles (single and constant periodical), are endowments including both a bonus option and a surrender option. The bonus option’s benefit is annually adjusted according to the performance of a reference fund and a minimum return is guaranteed to the policyholder. The surrender option is an American-style put option that enables the policyholder to give up the contract receiving the surrender value. We propose to price this American-style put option by Montecarlo simulation according to the Longstaff and Schwartz Least-Squares approach [17] giving a comparative analysis with the results obtained by Grosen and Jørgensen [15] according to a Recursive Tree Binomial approach. We then proceed to present an application to a relevant portion of a major Italian life policies’ portfolio. We make use of a Black&Scholes-CIR economy to simulate the reference fund, composed by equities and bonds, according to De Felice and Moriconi [14] and Pacati [21], and we estimate the fair value of portfolio’s liabilities extending the framework in order to price also the embedded surrender options.

Abstract:Background/Aim: Transgenic nude mice expressing green fluorescent protein (GFP), red fluorescent protein (RFP), or cyan fluorescent protein (CFP) were previously developed by our laboratory, AntiCancer Inc. In the present study, we demonstrate imaging of the GFP, RFP, or CFP nude mice with single-nanometer-tuning laser fluorescence excitation with a single instrument. Materials and Methods: Female transgenic C57/B6 nude GFP, RFP, and CFP mice aged six weeks were used. The images were obtained using the UVP Biospectrum Advanced system (Analytik Jena US LLC) with excitation at 480 nm and peak emission at 513 nm for GFP; 520 nm and 605 nm, respectively, for RFP; and 405 nm and 480 nm, respectively, for CFP. Results: For each color transgenic fluorescent mouse, images without background could be obtained individually with the UVP Biospectrum Advanced system. Conclusion: Using a single instrument, brilliant and well-defined images of GFP, RFP, and CFP mice were obtained with single-nanometer-tuning laser fluorescence excitation. This imaging system will be used in future studies to analyze cancer cells in the colored mice that are spectrally distinct in order to determine how stromal cells and cancer interact in the tumor microenvironment.
Combined Bioluminescence and Fluorescence Imaging Visualizing Orthotopic Lung Adenocarcinoma Xenograft in Vivo.

Abstract:Lung adenocarcinoma is the most common type of lung cancer. A close monitor of in vivo tumor development may help to better understand the pathogenesis and pathological processes of this disease. A bimodal imaging strategy has been developed, which is a very important tool to investigate the growth and metastasis of lung adenocarcinoma. In the present study, we used a combined labeling strategy in p53RE-luc-A549 cells via transfecting the reporter gene EGFP. In order to unambiguously identify the growth and metastasis of transfected A549 tumor cells, we established and observed subcutaneous and orthotopic xenografts in nude mice by in vivo bioluminescence and fluorescence imaging, which was verified by our post-mortem histological analysis. In vivo bioluminescence signal was observed for the progression of both subcutaneous and orthotopic xenografts in EGFP-p53RE-luc-A549 cells; in vivo fluorescence was only observed for the growth of subcutaneous xenograft of EGFP-p53RE-luc-A549 cells. Moreover, EGFP-p53RE-luc-A549 cells allow for the improved identification of implanted cells within host tissue during histological analysis. In conclusion, we presented a combined labeling strategy for bimodal A549 cell imaging which leads to improved detection of cellular grafts.
A Numerical Investigation of the Local Ocean Response to Westerly Wind Burst Forcing in the Western Equatorial Pacific

Abstract:Numerical simulations of the local equatorial ocean response to idealized westerly wind burst (WWB) forcing are described. In particular, the authors examine the development and evolution of the subsurface westward jet (SSWJ) that has been observed to accompany these wind events. This westward current is interpreted as the signature of equatorial waves that accompany the downwelling and upwelling that occurs along the edges of the wind forcing region. Some important features of the SSWJ include maximum intensity toward the eastern edge of the forcing region, a time lag between the wind forcing and peak SSWJ development, and an eastward spreading of the SSWJ with time. The effect of wind burst zonal profile, magnitude, duration, and fetch on the SSWJ are explored. The response of an initially resting ocean to WWB forcing is compared with that for model oceans that are spun up with annual-mean surface fluxes and monthly varying fluxes. It is demonstrated that the gross features of the response for the spun up simulations can be well approximated by adding the background zonal current structure prior to the introduction of the wind burst to the initially resting ocean current response to the WWB. This result suggests that the zonal current structure that is present prior to the commencement of WWB forcing plays a key role in determining whether or not a SSWJ will develop.
Precise navigation surgery of tumours in the lung in mouse models enabled by in situ fluorescence labelling with a killer-reporter adenovirus

Abstract:BACKGROUND: Current methods of image-guided surgery of tumours of the lung mostly rely on CT. A sensitive procedure of selective tumour fluorescence labelling would allow simple and high-resolution visualisation of the tumour for precise surgical navigation.METHODS: Human lung cancer cell lines H460 and A549 were genetically transformed to express red fluorescent protein (RFP). Tumours were grown subcutaneously for each cell line and harvested and minced for surgical orthotopic implantation on the left lung of nude mice. Tumour growth was measured by fluorescence imaging. After the tumours reached 5 mm in diameter, they were injected under fluorescence guidance with the telomerase-dependent green fluorescent protein (GFP)-containing adenovirus, OBP-401. Viral labelling of the lung tumours with GFP precisely colocalised with tumour RFP expression. Three days after administration of OBP-401, fluorescence-guided surgery (FGS) was performed.RESULTS: FGS of tumours in the lung was enabled by labelling with a telomerase-dependent adenovirus containing the GFP gene. Tumours in the lung were selectively and brightly labelled. FGS enabled complete lung tumour resection with no residual fluorescent tumour.CONCLUSIONS: FGS of tumours in the lung is feasible and more effective than bright-light surgery.
