Abstract:BACKGROUND Androgenic alopecia (AGA) is the most common type of hair loss in men. However, the pathogenesis is not yet fully understood and therapeutic approaches are limited. This retrospective study investigated the association between levels of androgen-associated hormones and curative effect in androgenic alopecia in young male AGA patients. MATERIAL AND METHODS By using chemiluminescence immunoassay, serum levels of androgens and upstream regulated hormones were measured in 178 young male patients with AGA and in 61 normal controls before therapy, 1 and 2 weeks after administration of finasteride. RESULTS Before oral finasteride therapy, we found significantly higher levels of serum free testosterone (FT) and dihydrotestosterone (DHT) in AGA patients than in normal controls. The levels of serum sex hormone-binding globulin (SHBG), luteinizing hormone (LH), and follicle-stimulating hormone (FSH) were similar in the 2 groups. There were no significant differences in serum androgen levels, including DHT and FT, among AGA patients with different grades of hair loss severity (p>0.05). After finasteride therapy, the levels of DHT decreased significantly (p<0.05). Increased serum levels of LSH or LH were also observed in 55 patients after therapy (p<0.05). The levels of SHGB did not change significantly after therapy (p>0.05). Patients with lower levels of serum FT and DHT than before who accepted finasteride therapy had a higher ratio of curative effect manifested by improved severity grade (p<0.05). Patients with higher levels of LSH or LH had a lower curative rate compared to those without change after therapy (p<0.05). CONCLUSIONS We confirmed the role of the androgens hypothalamus-hypophysis-sexual gland axis in the pathogenesis of AGA and the treatment effect of oral anti-androgen therapy in young male Chinese patients.

Abstract:Objective To investigate AR(androgen receptor)expression in male pattern baldness.Methods Obtain follicles from different scalp areas(occipital area of non-hair loss,hair loss transition and hair loss zone area) of 10 male AGA(androgenetic alopecia) patients and 3 normal scalp.Use immunohistochemistry method to study AR expressions in hair follicles in different regions.Results In the hair follicle bulge and infundibulum,AR expressions in hair loss area and the transitional area are obviously stronger than those in the occipital and the normal control group(P0.05).In epidermis,AR expressions show no significant difference(P0.05) in occipital area,hair loss area and the transitional area,but there are significant differences compared to normal control group(P0.05).In the sebaceous gland,AR expressions in each group is of no significant difference(P0.05).Conclusion The AR expressions are different between different regions.In the hair follicle bulge and infundibulum,AR expressions in hair loss area and the transitional area are obviously stronger than those in the occipital.This may be one of the key factors leading to male pattern hair loss.

Abstract:Objective To investigate the proliferation and apoptosis of hair follicle stem cells in male patients with AGA.Methods The follicle from different scalp areas(occipital of non-hair loss,hair loss transition and hair loss zone) of 15 AGA patients and 3 normal scalp.Using double immunofluorescence method to study the proliferation and apoptosis of hair follicle stem cell in scalp different regions.Results In hair follicle bulge area of normal group and patients' occipital area,there is no significant difference between the percentage of PCNA-positive cells in CK15 positive cells(P>0.05),they both are higher than that of the hair loss area and the hair loss transition area(P<0.05).There are no TUNEL-positive cells in the CK15-positive cells in each group,TUNEL-positive cells are mainly distributed in the inner hair follicle root sheath,The rate of TUNEL-positive cells is no significant difference among the groups(P>0.05).Conclusion Decrease of the proliferation activity of the hair follicle stem cells may be involved in the pathogenesis of male patients with AGA.

Abstract:BackgroundAndrogenic alopecia (AGA) is a major type of human scalp hair loss, which is caused by two androgens: testosterone (T) and 5α-dihydrotestosterone (5α-DHT). Both androgens bind to the androgen receptor (AR) and induce androgen-sensitive genes within the human hair dermal papilla cells (HHDPCs), but 5α-DHT exhibits much higher binding affinity and potency than T does in inducing the involved androgen-sensitive genes. Changes in the induction of androgen-sensitive genes during AGA are caused by the over-production of 5α-DHT by the 5α-reductase (5α-R) enzyme; therefore, one possible method to treat AGA is to inhibit this enzymatic reaction.MethodsRT-PCR was used to identify the presence of the 5α-R and AR within HHDPCs. A newly developed AGA-relevant HHDPC-based assay combined with non-radioactive thin layer chromatography (TLC) detection was used for screening crude plant extracts for the identification of new 5α-R inhibitors.ResultsHHDPCs expressed both 5α-R type 1 isoform of the enzyme (5α-R1) and AR in all of the passages used in this study. Among the thirty tested extracts, Avicennia marina (AM) displayed the highest inhibitory activity at the final concentration of 10 μg/ml, as the production of 5α-DHT decreased by 52 % (IC50 = 9.21 ± 0.38 μg/ml).ConclusionsAvicennia marina (AM) was identified as a potential candidate for the treatment of AGA based on its 5α-R1-inhibitory activity.
Expression of BMP type IA receptor in the scalp of AGA patients

Abstract:Objective: To study the expression of bone morphogenetic protein receptor I A (BMPR- I A) in the scalp of androgenetic alopecia (AGA) patients and its significance. Methods: Specimens were obtained from the frontoparietal alopecic scalp and occipital non-alopecic scalp of 8 AGA patients and healthy scalp of 11 healthy volunteers with informed consent and subjected to Western blot, immunohistochemistry and RT-PCR for the detection of BMPR- I A. Hair follicles were cultured with DHT(dihydrotestosterone)10 -9mol/L to observe the growth velocity, morphology of the hair follicles, and BMPR- I A was detected at the tenth day compared with the control group. Results: BMPR- I A expression was observed in the hair follicles of all groups. The average absorbance of BMPR- I A was significantly lower in alopecic area of AGA than in normal area of healthy volunteers(0.223±0.062 vs 0.097±0.038, t = 4.39, P<0.05), and the average absorbance of BMPR- I A in occipital scalp of AGA was 0.216±0.051(P>0.05). Decreased protein expression of BMPR- I A was also noted in the alopecia group compared with the normal group. Moreover, the gray ratio between BMPR- I A protein and GAPDH in the frontoparietal scalp of AGA patient reduced from 1.733±0.058 to 0.396±0.036(P<0.05), and the gray ratio of occipital scalp of AGA was 1.574±0.039 with no statistic difference to the control group. The gray ratio of DHT group was 0.367±0.026, which was much lower than 3.441±0.172 of control group(P<0.05). RT-PCR results revealed that the mRNA expression level of BMPR- I A of the alopecia group decreased by 47 percent compared with the control group and decreased by 87 percent in the DHT group compared with the control group. Conclusions: The decrease of BMPR- I A is partly induced by DHT, whether the slowdown of the growth and degeneration of hair follicles induced by DHT is due to the the down regulation of BMPR- I A is still to be made clear; and the reduction of BMPR- I A may be associated with obstacle of the formation of hair shaft and inner root sheath.

Abstract:This thesis examines two main hypotheses: 1. Obese knee osteoarthritis (KOA) patients can achieve symptomatic improvements following diet intervention regardless of their level of structural damage and overall joint malfunctioning: 2. Rapid weight-loss in obese patients with KOA will lead to improvements in KOA related pathology that can be assessed and evaluated by MRI. Data for the studies were obtained from obese KOA patients who were recruited for a 16 week diet intervention trial, the CAROT-trial (ClinicalTrials.gov identification no.: NCT00655941). Inclusion criteria were age ≥ 50 years, BMI ≥ 30 kg/square metro plus symptomatic and verified KOA. Patients underwent a 16 weeks dietary programme with formula products and counselling. Baseline and week 16 assessments included clinical examinations, MRI and CR of the most symptomatic knee, muscle strength tests, gait analyses, blood samples and collection of patient-reported outcomes with a variety of generic and specific health status questionnaires. MRI scans were graded by the BLOKS and CR was analysed by measuring the mJSW and grading the knee as described by KL. 388 possible subjects were pre-screened, 192 were enrolled. Following the 16 weeks diet intervention 175 patients remained in the study. 187 (97%) MRI scans were completed at baseline, 172 (98 %) MRI scans obtained at week 16 and this left the study with 169 (97%) patients with complete MRI datasets at week 16. No statistical significant differences were detected between baseline characteristics of all the initially included patients (n = 192) and the 169 patients included in the per protocol analyses performed in study III (p < 0.05). In order to apply BLOKS, an extensive MRI scoring system, in study II and III we examined the inter- and intra-observer reliability of the various BLOKS items in study I. Results showed that our assessment team performed as described in the original study defining BLOKS and that the patients in the CAROT-trial were graded as expected. In study II we investigated the impact of diet intervention on KOA symptoms whatever the patient's individual level of joint damage and malfunctioning, and the explanatory variables included high-field MRI, radiographs, and muscle strength in m. quadriceps as well as measurements of the knee-joint alignment axis. Results showed that diet intervention resulted in a symptomatic relief in obese KOA patients, irrespective of their level of structural damage, measures of joint malfunctioning and general pre-study patient characteristics. The final study examined whether or not weight-loss had an immediate impact on MRI assessed BMLs. The results showed that changes seen in the total TF sum of BML scores and maximal BML scores did not differ between patients achieving a major weight loss (> 10%) and those who did not. Furthermore, changes in clinical symptoms and BML scores were not associated. The limitations of this thesis were that the MRI analyses were based on single determinations of MRI variables and that the studies did not assess between scan reliability. The MRI protocol for this study did not include all the recommended sequences for BLOKS. Analysing BMLs with the use of only coronal STIR and T1w sequences is considered adequate for a reasonable assessment of the tibial and femoral bones. However, we recognize the limitations this strategy withholds in terms of correctly assessing BMLs located at the margins of our slices when only having a single plane view included in our MRI protocol. Due to an inadequate coverage we did not analyse BMLs in patella, and this confined the thesis to only study changes in the tibial and femoral bones. BLOKS contains separate scores for effusion and synovitis and we have assessed all MRI scans according to this discrimination well knowing that this procedure is biased and that a recent paper has proposed the combination of the two scores. MRI technology allows for an excellent discrimination and delineation of synovitis and synovial effusion by performing MRI with I.V. gadolinium and post-contrast T1 FS images, but due to extensive requirements and longer scans times for such examinations we proceeded with our, in this matter, suboptimal MRI protocol. The optimal assessment of KOA would be achieved by performing three radiographic views, posteroanterior, lateral and skyline, but for this study we chose a radiographic protocol only including the first two mentioned as this procedure was somewhat similar to the routine examination for KOA applied on a daily basis at our Department of Radiology. The results of this thesis support existing guidelines suggesting that diet intervention in obese KOA patients is beneficial for symptomatic improvements. The new information from the thesis is that improvement in clinical symptoms is possible for the majority of patients, independent of their pre-study level of structural damage and measures of joint malfunctioning. The present results also demonstrated that a rapid weight-loss had no association to changes in BML scores and established that changes observed in symptoms and BML scores, following a 16 weeks diet intervention, were not related.

Abstract:A micro-ELISA technique has been developed for the diagnosis of onchocerciasis. With the use of a low molecular weight (LMW) fraction of a soluble, adult Onchocerca volvulus extract as an antigen, the problem of nonspecificity observed with the crude worm extract is negligible. Seventy-three sera from proven cases of other filarial infections were weakly reactive or nonreactive. From cross-sectional and longitudinal surveys in a West African village hyperendemic for onchocerciasis, we concluded that this assay detects antibodies elicited when female worms start to produce microfilariae. Thirty-two of 33 children aged less than or equal to 15 years with positive skin snips were seropositive. High antibody levels could also be found in children who only became parasitologically positive 1-4 years after seroconversion. This leads to the conclusion that serology is more sensitive than the 2-snip method. In individuals with chronic infection (exposed for greater than 15 years), antibody titers were consistently lower (independent of the density of skin microfilariae) than in the early phase of a patent infection. A comparative serological study in a mesoendemic village revealed that the assay can serve to assess onchocerciasis prevalence by testing individuals aged 5-15 years.

Abstract:Androgenic alopecia (AGA) is the most prevalent type of progressive hair loss and has psychological repercussions. Nevertheless, the effectiveness of current pharmacological treatments remains limited, in part because the molecular basis of the disease has not been fully elucidated. Our group previously highlighted the important roles of aromatase and 5α-reductase (5α-R) in alopecia in young women with female pattern hair loss. Additionally, an association has been proposed between AGA and prostate cancer (PCa), suggesting that genes implicated in PCa would also be involved in AGA. A low-invasive, sensitive, and precise method was used to determine mRNA levels of aromatase, 5α-R isozymes, and 84 PCa-related genes in samples of plucked hair from young men with AGA and controls. Samples were obtained with a trichogram from the vertex scalp, and mRNA levels were quantified using real-time RT-PCR. The men with AGA had significantly higher 5α-R2 mRNA levels in comparison to controls; interestingly, some of them also showed markedly elevated mRNA levels of 5α-R1 or 5α-R3 or of both, which may explain the varied response to 5α-R inhibitor treatments. The men with AGA also showed significant changes versus controls in 6 out of the 84 genes implicated in PCa. This study contributes greater knowledge of the molecular bases of AGA, facilitating early selection of the most appropriate pharmacological therapy and opening the way to novel treatments.

Abstract:Pathogenetic mechanisms in androgenetic alopecia are not yet fully understood; however, it is commonly accepted that androgens like testosterone (T) and 5alpha-dihydrotestosterone (5alpha-DHT) inhibit hair follicle activity with early induction of the catagen. Thus, we investigated the influence of T and 5alpha-DHT on proliferation, cell death and bcl-2/bax expression in cultured dermal papilla cells (DPC) from nonbalding scalp regions of healthy volunteers. T and 5alpha-DHT induced apoptosis in DPC in a dose-dependent and time-related manner; in addition a necrotic effect due to T at 10(-5) M was found. Interestingly, bcl-2 protein expression was decreased in T- and 5alpha-DHT-treated cells, leading to an increase in the bax/bcl-2 ratio. In addition, T and 5alpha-DHT induced proteolytic cleavage of caspase 8 and inhibited proliferation of DPC at 10(-5) M. High concentrations of T and 5alpha-DHT were needed to induce apoptotic effects in DPC. These data suggest that DPC from nonbalding scalp regions do have the capacity to undergo apoptosis, but need a high androgen stimulus. The present study provides an interesting new pathogenetic approach in androgenetic alopecia.

Abstract:Androgenetic alopecia (AGA) is the most common type of hair loss, and is mainly caused by the biological effects of testosterone on dermal papilla cells (DPCs). In vitro culturing of DPCs might be a useful tool for the screening of target molecule of AGA. However, primary DPCs cannot continuously proliferate owing to cellular senescence and cell culture stress. In this study, we introduced mutant cyclin-dependent kinase 4 (CDK4), Cyclin D1, and telomerase reverse transcriptase (TERT) into DPCs. We confirmed protein expression of CDK4 and Cyclin D1, and enzymatic activity of TERT. Furthermore, we found the established cell line was free from cellular senescence. We also introduced the androgen receptor gene using a recombinant retrovirus, to compensate the transcriptional suppressed endogenous androgen receptor in the process of cell proliferation. Furthermore, we detected the efficient nuclear translocation of androgen receptor into the nucleus after the treatment of dihydrotestosterone, indicating the functionality of our introduced receptor. Our established cell line is a useful tool to identify the downstream signaling pathway, which activated by the testosterone.

Abstract:Androgenetic alopecia (AGA) is a prevalent hair loss condition in males that develops due to the influence of androgens and genetic predisposition. With the aim of elucidating genes involved in AGA pathogenesis, we modelled AGA with three-dimensional culture of keratinocyte-surrounded dermal papilla (DP) cells. We co-cultured immortalised balding and non-balding human DP cells (DPCs) derived from male AGA patients with epidermal keratinocyte (NHEK) using multi-interfacial polyelectrolyte complexation technique. We observed up-regulated mitochondria-related gene expression in balding compared with non-balding DP aggregates which indicated altered mitochondria metabolism. Further observation of significantly reduced electron transport chain complex activity (complexes I, IV and V), ATP levels and ability to uptake metabolites for ATP generation demonstrated compromised mitochondria function in balding DPC. Balding DP was also found to be under significantly higher oxidative stress than non-balding DP. Our experiments suggest that application of antioxidants lowers oxidative stress levels and improves metabolite uptake in balding DPC. We postulate that the observed up-regulation of mitochondria-related genes in balding DP aggregates resulted from an over-compensatory effort to rescue decreased mitochondrial function in balding DP through the attempted production of new functional mitochondria. In all, our three-dimensional co-culturing revealed mitochondrial dysfunction in balding DPC, suggesting a metabolic component in the aetiology of AGA.

Abstract:Androgenetic alopecia (AGA) is a common heritable and androgen-dependent hair loss condition in men. Twelve genetic risk loci are known to date, but it is unclear which genes at these loci are relevant for AGA. Dermal papilla cells (DPCs) located in the hair bulb are the main site of androgen activity in the hair follicle. Widely used monolayer-cultured primary DPCs in hair-related studies often lack dermal papilla characteristics. In contrast, immortalized DPCs have high resemblance to intact dermal papilla. We derived immortalized human DPC lines from balding (BAB) and non-balding (BAN) scalp. Both BAB and BAN retained high proportions of dermal papilla signature gene and versican protein expression. We performed expression analysis of BAB and BAN and annotated AGA risk loci with differentially expressed genes. We found evidence for AR but not EDA2R as the candidate gene at the AGA risk locus on chromosome X. Further, our data suggest TWIST1 (twist family basic helix-loop-helix transcription factor 1) and SSPN (sarcospan) to be the functionally relevant AGA genes at the 7p21.1 and 12p12.1 risk loci, respectively. Down-regulated genes in BAB compared to BAN were highly enriched for vasculature-related genes, suggesting that deficiency of DPC from balding scalps in fostering vascularization around the hair follicle may contribute to the development of AGA.
Weekly treatment with SAMiRNA targeting the androgen receptor ameliorates androgenetic alopecia

Abstract:Androgenetic alopecia (AGA) is the most common type of hair loss in men and women. Dihydrotestosterone (DHT) and androgen receptor (AR) levels are increased in patients with AGA, and DHT-AR signaling correlates strongly with AGA pathogenesis. In this study, treatment with self-assembled micelle inhibitory RNA (SAMiRNA) nanoparticle-type siRNA selectively suppressed AR expression in vitro. Clinical studies with application of SAMiRNA to the scalp and massaging to deliver it to the hair follicle confirmed its efficacy in AGA. For identification of a potent SAMiRNA for AR silencing, 547 SAMiRNA candidates were synthesized and screened. SAMiRNA-AR68 (AR68) was the most potent and could be efficiently delivered to human follicle dermal papilla cells (HFDPCs) and hair follicles, and this treatment decreased the AR mRNA and protein levels. We confirmed that 10 µM AR68 elicits no innate immune response in human PBMCs and no cytotoxicity up to 20 µM with HFDP and HaCaT cells. Clinical studies were performed in a randomized and double-blind manner with two different doses and frequencies. In the low-dose (0.5 mg/ml) clinical study, AR68 was applied three times per week for 24 weeks, and through quantitative analysis using a phototrichogram, we confirmed increases in total hair counts. In the high-dose (5 mg/ml) clinical study, AR68 was given once per week for 24 weeks and showed 83% efficacy in increasing hair counts compared with finasteride. No side effects were observed. Therefore, SAMiRNA targeting AR mRNA is a potential novel topical treatment for AGA.

Abstract:The identification of crucial targets for hair regrowth in androgenetic alopecia (AGA) involves determining important characteristics and different stages during the process of hair follicle regeneration. Traditional methods for assessing key features and different stages of hair follicle primarily involve taking skin tissue samples and determining them through various staining or other methods. However, non-invasive assessment methods have been long sought. Therefore, in this study, endogenous fluorescence signals from skin keratin and second harmonic signals from skin collagen fibers were utilized as probes, two-photon excited fluorescence (TPEF) and second harmonic generation (SHG) imaging techniques were employed to non-invasively assess hair shafts and collagen fibers in AGA mice in vivo . The TPEF imaging technique revealed that the alternation of new and old hair shafts and the different stages of the growth period in AGA mice were delayed. In addition, SHG imaging found testosterone reduced hair follicle area and miniaturized hair follicles. The non-invasive TPEF and SHG imaging techniques provided important methodologies for determining significant characteristics and different stages of the growth cycle in AGA mice, which will facilitate future non-invasive assessments on human scalps in vivo and reduce the use of animal testing.

Abstract:Androgenetic alopecia (AGA) is the most prevalent form of progressive hair loss disorder in both men and women, significantly impacting their appearance and overall quality of life. Overactivation of the AR signaling pathway in dermal papilla cells (DPCs) plays a crucial role in the development and progression of AGA. Considering the severe systemic side effects associated with oral AR antagonists, the idea of developing of topical AR antagonists with rapid metabolic deactivation properties emerged as a promising approach. Herein, through systematic structural optimization, we successfully identified compound 30a as a potent and selective AR antagonist with favorable pharmacokinetic properties, resulting in high skin exposure and low plasma exposure following topical administration. Importantly, in both hair-growth and AGA mouse models, compound 30a showed potent hair-growth-promoting effects without any noticeable toxicity. These findings suggest that compound 30a holds significant potential as a topical AR antagonist for treating AGA patients.

Abstract:Female pattern hair loss (FPHL) is an important hair disorder, especially when young women are affected. However, pharmacological treatments are not successful in all women. Androgens, especially dihydrotestosterone (DHT), may play a role in FPHL, but many women with this disorder have normal serum androgen levels. It therefore appears that hair follicle levels of DHT depend on in situ testosterone (T) metabolism. Because T can be converted to DHT or estradiol (E2) by 5α-reductase (5α-R) and aromatase, respectively, these enzymes would determine DHT and E2 concentrations and their ratio. We propose and apply a low-invasive, sensitive and precise method for the absolute quantification of mRNA levels of aromatase and 5α-R isozymes (type 1, type 2 and type 3) in plucked hair from young women with FPHL. Normoandrogenic women with FPHL and controls were studied. Plucked hair samples were obtained by trichogram from vertex scalp and mRNA levels quantified by real-time RT-PCR. We revealed for the first time the presence of 5α-R3 mRNA in human hair. Interestingly, one, two, or even three 5α-R isozymes were increased in some women with FPHL but not in others, which may explain the lack of response to 5α-R inhibitors in some FPHL cases. Aromatase mRNA levels were significantly lower in women with FPHL than in controls. It may therefore produce a reduction in oestrogen levels and an increase in the androgen/oestrogen ratio in hair. The proposed low-invasive technique offers a molecular aetiologic diagnosis of FPHL for the selection of more appropriate pharmacological treatments with early predicted effectiveness.

Abstract:OBJECTIVE To compare monocular visual field extent at 6 years of age in eyes with high-risk prethreshold retinopathy of prematurity (ROP) randomized to early treatment (ET) with eyes that underwent conventional management (CM) and were treated at threshold or regressed without treatment. METHODS Subjects were 370 surviving study participants who developed high-risk prethreshold ROP and were enrolled in the Early Treatment for Retinopathy of Prematurity Study between October 1, 2000, and September 30, 2002. When the participants were 6 years of age, vision testers unaware of ROP status used white-sphere kinetic perimetry to measure visual field extent along the superotemporal, inferotemporal, inferonasal, and superonasal meridians. RESULTS The extent of the visual field was 0.1° to 3.7° larger in ET eyes when blind eyes were assigned a score of 0°. When data were examined from eyes of participants with 1 sighted ET eye and 1 sighted CM eye, ET eyes showed a small (1.3°-3.1°) reduction, which was statistically significant only along the superonasal meridian (P = .005). In bilaterally sighted children, visual field extent was not significantly reduced for high-risk type 1 ET eyes (-0.9° to 1.8°). However, in ET eyes with high-risk type 2 disease, visual field extent was significantly smaller compared with that of CM eyes (3.6°-8.7° superonasal field [P = .003]; inferonasal field [P < .001]). CONCLUSION Early treatment preserves peripheral vision, with only a small reduction of visual field extent. APPLICATION TO CLINICAL PRACTICE Early treatment for high-risk prethreshold ROP does not adversely affect visual field extent clinically.

Abstract:Context/objective: Androgen induces androgenetic alopecia (AGA), which has a regressive effect on hair growth from the frontal region of the scalp. Conversely, Wnt proteins are known to positively affect mammalian hair growth. We hypothesized that androgen reduces hair growth via an interaction with the Wnt signaling system. The objective of this study was to investigate the effect of androgen on Wnt signaling in dermal papilla (DP) cells. Design: The effect of androgen and Wnt3a on keratinocyte proliferation was measured by use of a coculture system consisting of DP cells and keratinocytes. The molecular mechanisms of androgen and Wnt pathway interactions in DP cells were examined by analyzing the expression, intracellular localization, and activity of the androgen receptor (AR) and also downstream Wnt signaling molecules. Results: Wnt3a-dependent keratinocyte growth was suppressed by the addition of dihydrotestosterone in coculture with DP cells that were derived from AGA patients, but growth was not suppressed in coculture with DP cells from non-AGA males. Whereas DP cells from both scalp regions expressed AR protein, the expression levels of AR and cotranslocation with beta-catenin, a downstream Wnt signaling molecule, were higher in DP cells of AGA patients than in DP cells from non-AGA males. In addition, significant suppression of Wnt signal-mediated transcription in response to dihydrotestosterone treatment was observed only in DP cells from AGA patients. Conclusion: These results suggest that Wnt signaling in DP cells is regulated by androgen and this regulation plays a pivotal role in androgen's action on hair growth.
