Abstract:The understanding of many nanoscale processes occurring in liquids such as colloidal crystal formation, aggregation, nanowire growth, electrochemical deposition, and biological interactions would benefit greatly from real-time, in situ imaging with the nanoscale resolution of transmission electron microscopes (TEMs) and scanning transmission electron microscopes (STEMs). However, these imaging tools cannot readily be used to observe processes occurring in liquid media without addressing two experimental hurdles: sample thickness and sample evaporation in the high vacuum microscope chamber. To address these challenges, we have developed a nano-Hele-Shaw cell, dubbed the nanoaquarium. The device consists of a hermetically-sealed, 100 nm tall, liquid-filled chamber sandwiched between two freestanding, 50 nm thick, silicon nitride membranes. Embedded electrodes are integrated into the device. This fluid dynamics video features particle motion and aggregation during in situ STEM of nanoparticles suspended in liquids. The first solution contains 5 nm gold particles, 50 nm gold particles and 50 nm polystyrene particles in water. The second solution contains 5 nm gold particles in water. The imaging was carried out with a FEI Quanta 600 FEG Mark II with a STEM detector. In the footage of the multi-particle solution, note that the 50 nm gold particles prominently decorate the clusters and are clearly distinguished. In the footage of the 5 nm gold particles, diffusion-limited aggregation is observed. Individual particles and small clusters are seen diffusing throughout the field of view, bumping into each other and bonding irreversibly to form a fractal structure. The rate of aggregation and the fractal dimension of the aggregates are consistent with light scattering measurements, indicating that the electron beam does not greatly alter the observed phenomenon.

Abstract:We have directly observed motion of inorganic nanoparticles during fluid evaporation using a transmission electron microscope. Tracking real-time diffusion of both spherical (5-15 nm) and rod-shaped (5 x 10 nm) gold nanocrystals in a thin film of water-15% glycerol reveals complex movements, such as rolling motions coupled to large-step movements and macroscopic violations of the Stokes-Einstein relation for diffusion. As drying patches form during the final stages of evaporation, particle motion is dominated by the nearby retracting liquid front.

Imaging Translational and Rotational Diffusion of Single Anisotropic Nanoparticles with Planar Illumination Microscopy

Abstract:Here we demonstrated a simple yet powerful method, planar illumination microscopy, to directly track the rotational and translational diffusion dynamics of individual anisotropic nanoparticles in solution and living cells. By illuminating gold nanorods (GNRs) with two orthogonal sheets of light and resolving the polarized scattering signal with a birefringent crystal, we readily achieved three-dimensional angular resolving capability for single GNRs in noisy surroundings. The rotational dynamics of individual GNRs dispersed in glycerol/water mixtures with different chemical modification were tracked, and the measured rotational diffusion coefficient was well fitted to a previously reported theoretical model (Torre, J. G. d. l.; Martinez, M. C. L. Macromolecules 1987, 20, 661-666; Tirado, M. M.; Torre, J. G. d. l. J. Chem. Phys. 1980, 73, 1986-1993). In addition, the translational and rotational movements of individual GNRs transported by kinesin motor protein on microtubules inside living cells were directly imaged. Compared to its motion in free solution, a GNR attached to motor-protein did not rotate significantly while moving forward. Our method can be further generalized to allow determination of three-dimensional orientation of single dipoles using many different illumination modes.

Abstract:In real time and space, four-dimensional electron microscopy (4D EM) has enabled observation of transient structures and morphologies of inorganic and organic materials. We have extended 4D EM to include liquid cells without the time resolution being limited by the response of the detector. Our approach permits the imaging of the motion and morphological dynamics of a single, same particle on nanometer and ultrashort time scales. As a first application, we studied the rotational dynamics of gold nanoparticles in aqueous solution. A full transition from the conventional diffusive rotation to superdiffusive rotation and further to a ballistic rotation was observed with increasing asymmetry of the nanoparticle morphology. We explored the underlying physics both experimentally and theoretically according to the morphological asymmetry of the nanoparticles.

Abstract:In order to get the real-time observations on nanoparticle-liquid two-phase flow, an experimental apparatus with a fluorescence microscope, a CCD camera, a syringe pump and other assistant equipment was setup. Using the experimental apparatus, the two-phase flow in which diameter of the solid phase is about 55 nm was observed. Experimental results indicated that nanoparticles followed the flow in micro channels. So in the research of microfluid, nanoparticles can be used as the seeds to track the flow of liquid. With the increase of flow the velocities of particles became dispersed. The solid concentration had some effects on the speed distribution in the cross section of the channel.

Abstract:The dynamic behavior of octahedral gold nanoparticles (NPs) and nanoparticle clusters (NPCs) in aqueous solution is studied by in-situ liquid-cell transmission electron microscopy (TEM). The octahedral Au NPs/NPCs show preferential orientations in the liquid cell, due to the interaction with the SiNx window. The Au NPs show long-range reversible hopping and three-dimensional (3D) rotational motions in the liquid environment. At the same time, the Au NPCs and NPs perform slow stick-slip and stick-roll motions, respectively, with a centripetal trend. The centripetal motions were explained by a liquid evaporation-induced radial flow model, in which the NPCs/NPs trajectories are controlled by Stokes forces and surface friction by the silicon nitride window. The calculated radius-dependent force (Fc) on the NPCs/NPs shows a semi-linear correlation with the distance r between the NPCs/NPs and the center of mass, accompanied with stochastic fluctuations, in agreement with the model predictions. This work thus demonstrates the effectiveness of in situ liquid-cell TEM for the in-depth understanding of complicated liquid flow and force interactions in nanomaterials.

Abstract:Single nanoparticle (NP) electrochemical measurements are widely described, both theoretically and experimentally, as they enable visualization of the electrochemical signal of a single NP that is masked in ensemble measurements. However, investigating the behavior of individual NPs using electrochemical signals remains a significant challenge. Here we report experiments and simulations demonstrating that multiple distinct motion trajectories could be discerned from time-resolved current traces by dynamic Monte Carlo simulations. We show that continuous monitoring and quantification of electrochemical oxidation of individual AgNPs using a low-noise electrochemical measurement platform produce significantly distinguished current traces due to the size-dependent motions of AgNPs. Our findings offer a view of the electrochemical signals of individual NPs that are largely different from that in the literature, and underscore the significance of motion behaviors in single NP electrochemistry.

Abstract:Dynamics of active or propulsive Brownian particles in nonequilibrium status have recently attracted great interest in many fields including artificial micro/nanoscopic motors and biological entities. Understanding of their dynamics can provide insight into the statistical properties of physical and biological systems far from equilibrium. We report the translational dynamics of photon-activated gold nanoparticles (NPs) in water imaged by liquid-cell four-dimensional electron microscopy (4D-EM) with high spatiotemporal resolution. Under excitation of femtosecond laser pulses, we observed that those NPs exhibit superfast diffusive translation with a diffusion constant four to five orders of magnitude greater than that in the absence of laser excitation. The measured diffusion constant follows a power-law dependence on the laser fluence and a linear increase with the laser repetition rate, respectively. This superfast diffusion of the NPs is induced by a strong random driving force arising from the photoinduced steam nanobubbles (NBs) near the NP surface. In contrast, the NPs exhibit a superfast ballistic translation at a short time scale down to nanoseconds. Combining with a physical model simulation, this study reveals a photoinduced NB propulsion mechanism for propulsive motion, providing physical insights into better design of light-activated artificial micro/nanomotors. The liquid-cell 4D-EM also provides the potential of studying other numerical dynamical behaviors in their native environments.

Abstract:Convective transportation of materials in the solid state occurring in a prototype solid bilayer system of Al and Si with negligible mutual solubility has been directly imaged in real time at nanoscale using a valence energy-filtered transmission electron microscope. Such solid-state convection is driven by the stress gradient developing in the bilayer system due to the amorphous to crystalline phase transformation of the Si sublayer. The process is characterized by compression experienced in the Si phase crystallizing within the Al sublayer, as well as by the development of mushroom-shaped "plumes" of Al nanocrystals in the Si sublayer as a result of compressive stress relaxation and discrete, new nucleation of crystalline Al. The real-time, atomistic observation and the thus-obtained fundamental understanding of solid-state convection enable highly sophisticated applications of such a complex process in advanced fabrication and processing of nanomaterials and solid-state devices.

Abstract:In single-particle tracking (SPT), fluorescence video microscopy is used to record the motion images of single particle or single molecule. Here, by using a total-internal-reflection microscope equipped with an argon ion laser and a charge-coupled device (CCD) camera with high-speed and high-sensitivity, video images of single nanobeads in solutions were obtained. From the trajectories, the diffusion coefficient of individual nanobead was determined by the mean square displacements as a function of time. The sizes of nanobeads were calculated by Stokes-Einstein equation, and the results were compared with the actual values.

Abstract:Nanoparticle self-assembly has been well studied theoretically, but it remains challenging to directly observe and quantify individual nanoparticle interactions. With our custom image analysis method, we track the trajectories of nanoparticle movement with high precision from a stack of relatively noisy images obtained using liquid cell transmission electron microscopy. In a time frame of minutes, Pt-Fe nanoparticles self-assembled into a loosely packed hcp lattice. The energetics and stability of the dynamic assembly were studied quantitatively. From velocity and diffusion measurements, we experimentally determined the magnitude of forces between single particles and the related physical properties. The results illustrate that long-range anisotropic forces drive the formation of chains, which then clump and fold to maximize close range van der Waals interactions.
Revealing dynamic processes of materials in liquids using liquid cell transmission electron microscopy

Abstract:The recent development for in situ transmission electron microscopy, which allows imaging through liquids with high spatial resolution, has attracted significant interests across the research fields of materials science, physics, chemistry and biology. The key enabling technology is a liquid cell. We fabricate liquid cells with thin viewing windows through a sequential microfabrication process, including silicon nitride membrane deposition, photolithographic patterning, wafer etching, cell bonding, etc. A liquid cell with the dimensions of a regular TEM grid can fit in any standard TEM sample holder. About 100 nanoliters reaction solution is loaded into the reservoirs and about 30 picoliters liquid is drawn into the viewing windows by capillary force. Subsequently, the cell is sealed and loaded into a microscope for in situ imaging. Inside the TEM, the electron beam goes through the thin liquid layer sandwiched between two silicon nitride membranes. Dynamic processes of nanoparticles in liquids, such as nucleation and growth of nanocrystals, diffusion and assembly of nanoparticles, etc., have been imaged in real time with sub-nanometer resolution. We have also applied this method to other research areas, e.g., imaging proteins in water. Liquid cell TEM is poised to play a major role in revealing dynamic processes of materials in their working environments. It may also bring high impact in the study of biological processes in their native environment.
Real-time Observation of Dynamic Heterogeneity of Gold Nanorods on Plasma Membrane with Darkfield Microscopy

Abstract:The plasma membrane possesses a complicated structure, on which the protein clusters are randomly but orderly distributed to maintain the regular morphology and function of cells. Investigating the detailed dynamic behaviors of nanoparticles (NPs) on cytomembrane is of great importance to understand cellular mechanisms and advance the bio-nano technologies for drug delivery, photothermal therapy, immunotherapy, etc. In this work, to study the dynamic heterogeneous interactions between NPs and cell membrane with high resolution, we established a simple method to efficiently track the translational and rotational diffusion of individual gold nanorods (AuNRs) on cell membranes. This method is based on that an anisotropic AuNR appears as a colored spot under a darkfield microscope (DFM) equipped with a color camera. While obtaining its lateral position, the polar angle of the AuNR can be calculated simultaneously from intensity difference between the R and G channels. Careful analysis shows that the lateral motion of single AuNRs do not follow normal Brownian diffusion, which could be attributed to their hop diffusion in the dynamically varying picket-fence structure of the live cell membrane. Furthermore, 4 different rotation-translation patterns of the AuNR are observed due to spatiotemporal heterogeneity of the cytomembrane. This simple but robust method for simultaneously obtaining the location and orientation of anisotropic plasmonic nanoparticles could be further applied to the analysis of complicated biological and biomedical processes.
Visualizing Diffusional Dynamics of Gold Nanorods on Cell Membrane Using Single Nanoparticle Darkfield Microscopy

Abstract:Analyzing the diffusional dynamics of nanoparticles on cell membrane plays a significant role in better understanding the cellular uptake process and provides a theoretical basis for the rational design of nano-medicine delivery. Single particle tracking (SPT) analysis could probe the position and orientation of individual nanoparticles on cell membrane, and reveal their translational and rotational states. Here, we show how to use traditional dark-field microscopy to monitor the dynamics of gold nanorods (AuNRs) on live cell membrane. We also show how to extract the location and orientation of AuNRs using ImageJ and MATLAB, and how to characterize the diffusive states of AuNRs. Statistical analysis of hundreds of particles show that single AuNRs perform Brownian motion on the surface of U87 MG cell membrane. However, individual long trajectory analysis shows that AuNRs have two distinctly different types of motion states on the membrane, namely long-range transport and limited-area confinement. Our SPT methods can be potentially used to study the surface or intracellular particle diffusion in different biological cells and can become a powerful tool for investigations of complex cellular mechanisms.

Abstract:Scanning transmission electron microscopy of various fluid and hydrated nanomaterial samples has revealed multiple imaging artifacts and electron beam-fluid interactions. These phenomena include growth of crystals on the fluid stage windows, repulsion of particles from the irradiated area, bubble formation, and the loss of atomic information during prolonged imaging of individual nanoparticles. Here we provide a comprehensive review of these fluid stage artifacts, and we present new experimental evidence that sheds light on their origins in terms of experimental apparatus issues and indirect electron beam sample interactions with the fluid layer. A key finding is that many artifacts are a result of indirect electron beam interactions, such as production of reactive radicals in the water by radiolysis, and the associated crystal growth. The results presented here will provide a methodology for minimizing fluid stage imaging artifacts and acquiring quantitative in situ observations of nanomaterial behavior in a liquid environment.

Abstract:Liquid-phase transmission electron microscopy (TEM) has been recently applied to materials chemistry to gain fundamental understanding of various reaction and phase transition dynamics at nanometer resolution. However, quantitative extraction of physical and chemical parameters from the liquid-phase TEM videos remains bottlenecked by the lack of automated analysis methods compatible with the videos' high noisiness and spatial heterogeneity. Here, we integrate, for the first time, liquid-phase TEM imaging with our customized analysis framework based on a machine learning model called U-Net neural network. This combination is made possible by our workflow to generate simulated TEM images as the training data with well-defined ground truth. We apply this framework to three typical systems of colloidal nanoparticles, concerning their diffusion and interaction, reaction kinetics, and assembly dynamics, all resolved in real-time and real-space by liquid-phase TEM. A diversity of properties for differently shaped anisotropic nanoparticles are mapped, including the anisotropic interaction landscape of nanoprisms, curvature-dependent and staged etching profiles of nanorods, and an unexpected kinetic law of first-order chaining assembly of concave nanocubes. These systems representing properties at the nanoscale are otherwise experimentally inaccessible. Compared to the prevalent image segmentation methods, U-Net shows a superior capability to predict the position and shape boundary of nanoparticles from highly noisy and fluctuating background—a challenge common and sometimes inevitable in liquid-phase TEM videos. We expect our framework to push the potency of liquid-phase TEM to its full quantitative level and to shed insights, in high-throughput and statistically significant fashion, on the nanoscale dynamics of synthetic and biological nanomaterials.Machine-learning-assisted extracting of nanoscale dynamics from liquid-phase TEM videos.This article has not yet been cited by other publications.

Direct Visualization of Photomorphic Reaction Dynamics of Plasmonic Nanoparticles in Liquid by 4D Electron Microscopy.

Abstract:Liquid-cell electron microscopy (LC-EM) provides a unique approach for in situ imaging of morphology changes of nanocrystals in liquids under electron beam irradiation. However, nanoscale real-time imaging of chemical and physical reaction processes in liquids under optical stimulus is still challenging. Here, we report direct observation of photomorphic reaction dynamics of gold nanoparticles (AuNPs) in water by liquid-cell four-dimensional electron microscopy (4D-EM) with high spatiotemporal resolution. The photoinduced agglomeration, coalescence, and fusion dynamics of AuNPs at different temperatures are studied. At low laser fluences, the AuNPs show a continuous aggregation in several seconds, and the aggregate size decreases with increasing fluence. At higher fluences close to the melting threshold of AuNPs, the aggregates further coalesced into nanoplates. While at fluences far above the melting threshold, the aggregates fully fuse into bigger NPs, which is completed within tens of nanoseconds. This liquid-cell 4D-EM would also permit study of other numerical physical and chemical reaction processes in their native environments.

Abstract:Objective-type total internal reflection fluorescence technique and Gaussian fit single particle tracking algorithm have been used to study the constrained Brownian motion characteristics of single fluorescent microspheres of 40 nm diameter in real-time. Three-dimensional tracking has been realized from the two-dimensional images by utilizing the exponential decay of the fluorescence intensity excited by the evanescent field. Trajectory analyses have shown that single fluorescent beads diffused in a constrained Brownian mode with a diffusion coefficient a little smaller than theoretical estimates in the plane-parallel direction.
