Abstract:Spermatogenesis is the process by which diploid male germ cells propagate and differentiate into haploid flagellated spermatozoa. This highly complicated process is dependent on testicular somatic cells maturation. While the role of these somatic cells in spermatogenesis is relatively well established, knowledge about their development and maturation, particularly at the molecular level is limited. In this study, we profiled the testicular single-cell transcriptomes of Guanzhong black pigs at the age of 7, 30, 60, 90, and 150 days. Five types of Sertoli cells, five types of Leydig cells, and four types of peritubular myoid cells were identified. Histological analysis revealed the changes in proliferation levels and marker gene expressions, and prion-like protein gene (PRND) was identified as a novel marker for Sertoli cells. Additionally, integrated analyses of porcine and human datasets revealed similarities between human and pig testicular somatic cells. Overall, the data obtained in this study contribute to the understanding of testicular development in pigs as a model species.

Changes in Serum LH Concentrations During Normal and Abnormal Sexual Development in the Pig

Abstract:Blood samples were collected from fetal and prepubertal pigs, starting at 49 days postcoitum and ending with pigs 25 weeks old. In addition, blood samples were collected from unilaterally cryptorchid, freemartin and castrated pigs of similar ages. Serum LH was quantified by radioimmunoassay. The LH concentration in the serum of male and female fetuses was undetectable in the second trimester of gestation but increased gradually thereafter, up to two weeks after birth. Frequent sampling of 2-week old pigs showed irregular LH peaks (>2 ng/ml) at this time. In the third and fourth week after birth average LU concentrations decreased. Average LH concentrations increased to about 2 ng/ml in castrated pigs. LH concentrations observed for freemartin pigs were 1.3 ± 0.7 ng/ml. The changes in LH concentrations appeared to be well correlated with the differentiation and functional activity of the testis. No correlation could be detected between the changes in LH concentration and normal and abnormal gubernacular development.

Seasonal Influence of the Developmental Competence of Porcine Oocytes Matured in Vitro

Abstract:Porcine embryology is one of the most demanding fields in reproductive research of domestic animals due to its environment-sensitive oocytes.Plenty of lipid droplets in cytoplasm of porcine oocytes are the main cause of fragility,but their specific biological functions are still unknown.It is well recognized that environmental influence,such as high/low temperature and humidity,are associated with the reproduction performance in pigs.The goal of this study is to describe a seasonal variation in the developmental ability of porcine oocytes matured in vitro.Prepubertal gilt ovaries were collected more than two years(from spring to winter every year) from a local shambles to collect cumulus-oocytes-complexes(COCs) to mature for 44h in BSA-free NCSU23 supplemented with 10 ng/ mL EGF,10 ng/mL leptin,0.57 mmol/L cysteine,10 IU/mL PMSG and 10 IU/mL hCG.After cumulus cells removal,and the others were subjected to parthenogenetic activation,and the others were used to conduct somatic cell nuclear transfer(SCNT).In porcine cloning,cell lines established from 35~40 d Landrace fetus were used as nuclear donors,and the oocytes matured in vitro as cytoplast recipients.Embryos were in vitro cultured in NCSU23 supplemented with 4 mg/mL BSA.Cleavage and blastocyst rates were assessed on D2 and D6(D0:the day fusion and activation conducted).All the experiments were replicated at least three times.All data were analyzed by using SPSS(13.0) with one-way ANOVA.In this study,we compared the number of COCs(Grade AB,Oocytes surrounded by at least 3 layers of compacted cumulus cells and with uniform cytoplasm were defined Grade A. Oocytes surrounded by 2~3 layers of cumulus cells and with uniform cytoplasm were defined Grade B.) every ovary pair collected in different seasons(COCs/ovary pair) and meiotic maturation ability of in vitro-matured oocytes(experiment 1) and developmental competence of the parthenogenetic activated oocytes(experiment 2) as well as somatic cell nuclear transfer embryos(experiment 3) using oocytes collected from four different seasons.In experiment 1,we found that COCs/ovary pairs was significantly increased in spring(March to May) in comparison with the other seasons [7.6±3.5 vs 6.4±3.3,6.7±2.9,6.7±3.4 in summer(June to August),autumn(September to November),winter(December to Feberary),respectively,P0.05].However,no significant differences were found among the rates of oocytes progressing to MⅡ in four seasons (75.3%±3.3%,73.5%±2.3%,73.0%±4.2%,76.5%±6.7% in spring,summer,autumn,winter, respectively).In experiment 2,we saw that the rate of blastocyst formation of parthenogenetic embryos in summer prominently decreased compared to the other seasons(10.2%±2.7% vs 27.2%±3.5%,24.2%±3.2%,22.3%±3.4% in spring,autumn,winter, respectively,P0.05).In experiment 3,increased blastocyst rate of cloned porcine embryos were found in spring when compared to autumn and winter(12.2%±1.3% vs 10.3%±1.1%,8.1%±1.4%,P0.05).The results suggest there is a seasonal influence on the developmental competence of porcine oocytes matured in vitro.
Sertoli cells in the boar testis: changes during development and compensatory hypertrophy after hemicastration at different ages

Abstract:Changes in Sertoli cell numbers and testicular structure during normal development and compensatory hypertrophy were assessed in crossbred Meishan x White Composite males. Boars were assigned at birth to unilateral castration at 1, 10, 56, or 112 days or to remain as intact controls through 220 days. The first testes removed were compared to assess testicular development. At 220 days, testicular structure was evaluated in boars representing the 25% with the largest (Lg) testis and the 25% with the smallest (Sm) testis in each treatment group. The number of Sertoli cells per testis reached a maximum by Day 56 in Sm testis but not until Day 112 in Lg testis boars, indicating a longer duration of Sertoli cell proliferation in Lg testis boars. Unilateral castration of Lg testis boars on Days 1, 10, 56, and 112 caused the weight of the remaining testis to hypertrophy by 149%, 135%, 119%, and 120%, respectively, and total sperm production to increase to 127%, 128%, 97%, and 106%, respectively. However, Sertoli cell numbers changed little in hemicastrate boars. In Lg testis boars, compensatory hypertrophy primarily involved proliferation of Leydig cells and expansion of existing Sertoli cells with little increase in Sertoli cell numbers, but in Sm testis boars, it involved expansion of existing Leydig and Sertoli cells without increase in cell numbers. These results indicate that Lg and Sm testis boars display intriguing differences during both development and compensatory hypertrophy, and they identify a unique animal model for further studies of factors that program and control Sertoli cell proliferation.
Histological Changes of the Testicular Interstitium during Postnatal Development in Microminipigs

Abstract:Microminipigs have become an attractive animal model for the toxicology- and pharmacology-related studies because of their manageable size. In this study, the development of the testicular interstitium and steroidogenesis in microminipigs, from 0 to 12 months of age, were investigated. Testicular interstitium was mostly composed of two types of Leydig cells (large and small Leydig cells) and a few macrophages and mast cells. Large Leydig cells were observed in the peritubular area throughout all the ages. Small Leydig cells were present in the interlobular and subcapsular areas at an early age and then gradually converted into large Leydig cells. Testicular composition of the Leydig cells began to increase after 3 months of age, when spermatogenesis was completed, and reached approximately 35% at 12 months. Steroidogenic enzymes in Leydig cells were detected by immunohistochemistry from 0 month of age. Serum testosterone levels increased substantially from 1.5 to 4.5 months of age, which coincided well with the age of sexual development previously reported in microminipigs. Because the interstitial space of the testis has dramatic variations between species, the basic information obtained in the present study will be a useful reference for all the future toxicity evaluations in microminipigs.

Abstract:A germ–Sertoli cell coculture model was established to study effects of follicle-stimulating hormone (FSH) and testosterone (T) on testicular germ cell proliferation of the embryonic chickens. Germ and somatic cells were dispersed from 18-day-old embryonic testes and cultured in 96-well plates. Germ cells were characterized by expression of stem cell factor receptor c-kit. Germ cell proliferation was assessed by an increase in cell number and expression of proliferating cell nuclear antigen (PCNA). Results showed that the germ and Sertoli cells kept alive in serum-free McCoy’s 5A medium supplemented with insulin, transferrin, and selenite (ITS medium). Germ cells adhered to the free surface of Sertoli cells that spread the filopodia and formed a monolayer in ITS medium. In the serum-containing medium, Sertoli cells displayed an increment with a flat squamous form and only a few very large germ cell masses were found in the free surface of Sertoli cells. Many germ cells showed apoptosis in the McCoy’s 5A medium without ITS or serum. Only germ cells showed positive staining for c-kit in the coculture. Ovine FSH (0.25–1.0IU/ml) significantly increased the number of germ cells, and PCNA-labeling index (P<0.05). FSH also induced stronger c-kit expression compared with the control. In the FSH-treated groups, germ cells were manifested distinct knob-like form. Similar stimulating effect was found in the germ cell number by T treatments (10−7–10−6M). Furthermore, FSH (0.5IU/ml) combined with T significantly promoted higher testicular germ cell proliferation (P<0.05) compared with either FSH or T alone, which indicated that interaction of FSH and T might be additive. The above results showed that the serum-free germ–Sertoli cell coculture model allowed evaluating hormonal regulation of testicular germ cell proliferation. FSH and T promoted testicular germ cell proliferation probably through indirect effects on Sertoli cells.

Abstract:The pig is an important model for studying human diseases and is also a significant livestock species, yet its testicular development remains underexplored. Here, we employ single-cell RNA sequencing to characterize the transcriptomic landscapes across multiple developmental stages of Bama pig testes from fetal stage through infancy, puberty to adulthood, and made comparisons with those of humans and mice. We reveal an exceptionally early onset of porcine meiosis shortly after birth, and identify a distinct subtype of porcine spermatogonia resembling transcriptome state 0 spermatogonial stem cells identified in humans, which were previously thought to be primate specific. We also discover the persistent presence of proliferating progenitors for myoid cells in postnatal testes. The regulatory roles of Leydig cell steroidogenesis and estrogen synthesis in supporting cell lineages are also explored, including the potential impact of estrogen on Sertoli cell maturation and spermatogenesis. Overall, this study offers valuable insights into porcine testicular development, paving the way for future research in reproductive biology, advancements in agricultural breeding, and potential applications in translational medicine.

New warped LPC-Based Feature for Fast and robust speech/Music Discrimination

Abstract:ABSTRACT Automatic discrimination of speech and music is an im-portant tool in many multimedia applications. The pa-per presents a low complexity but effective approach forspeech/music discrimination, which exploits only one sim-ple feature, called Warped LPC-based Spectral Centroid(WLPC-SC). A three-component Gaussian Mixture Model(GMM) classiﬁer is used because it showed a slightly bet-ter performance than other Statistical Pattern Recognition(SPR) classiﬁers. Comparison between WLPC-SC andthe timbral features proposed in [11] is performed, aimingto assess the good discriminatory power of the proposedfeature. Experimental results reveal that our speech/musicdiscriminator is robust and fast, making it suitable for real-time multimedia applications. 1. INTRODUCTION Automatic discrimination between speech and music hasbecome a research topic of interest in the last few years.Several approaches have been described in the recent lit-erature for different applications [1][2] [3][4][5]. Eachof these uses different features and pattern classiﬁcationtechniques and describes results on different material.Saunders [1] proposed a real-time speech/music dis-criminator, which was used to automatically monitor theaudio content of FM audio channels. Four statistical fea-tures on the zero-crossing rate and one energy-related fea-ture were extracted, a multivariate-Gaussian classiﬁer wasapplied, which resulted in an accuracy of 98%.In Automatic Speech Recognition (ASR) of broadcastnews, it’s desirable to disable the input to the speech rec-ognizer during the non-speech portion of the audio stream.Scheirer and Slaney [2] developed a speech/music dis-crimination system for ASR of audio sound tracks. Thir-teen features to characterize distinct properties of speechand music, and three classiﬁcation schemes (MAP Gaus-sian, GMM and k-NN classiﬁers) were exploited, result-ing in an accuracy of over 90%.Another application that can beneﬁt from distinguish-ing speech from music is low bit-rate audio coding. De-signing an universal coder to reproduce well both speechand music is the best approach. However, it is not a trivialproblem. An alternative approach is to design a multi-mode coder that can accommodate different signals. Theappropriate module is selected using the output of a speech-music classiﬁer [6] [7].Automatic discrimination of speech and music is animportant tool in many multimedia applications. KhaledEl-Maleh et al. [3] combined the line spectral frequen-cies and zero-crossings-based features for frame-level nar-rowband speech/music discrimination. The classiﬁcationsystem operates using only a frame delay of 20 ms, mak-ing it suitable for real-time multimedia applications. Anemerging multimedia application is content-based index-ing and retrieval of audiovisual data. Audio content analy-sis is an important task for such application [8]. Minami etal. [9] proposed an audio-based approach to video index-ing, where a speech/music detector is used to help users tobrowse a video database.Comparative view of the value of different types of fea-tures in speech music discrimination is provided in [10],where four types of features (amplitudes, cepstra, pitchand zero-crossings) are compared for discriminating speechand music signals. Experimental results showed cepstraand delta cepstra bring the best performance. Mel Fre-quencies Spectral or Cepstral Coefﬁcients (MFSC or MFCC)are very often used features for audio classiﬁcation tasks,providing quite good results. In [4], MFSC’s ﬁrst orderstatistics are combined with neural networks to form aspeech music classiﬁer that is able to generalize from alittle amount of learning data. MFCC are a compact rep-resentation of the spectrum of an audio signal taking intoaccount the nonlinear human perception of pitch, as de-scribed by the mel scale. They are one of the most usedfeatures in speech recognition and have recently proposedin musical genre classiﬁcation of audio signals [11][12].Unlike the previous works, speech/music discrimina-tion approaches based on only one type of features arepresented in [13] and [5], which result in fast and robustclassiﬁcation systems. The approach in [13] takes psy-choacoustic knowledge into account in that it uses the lowfrequency modulation amplitudes over 20 critical bands toform a good discriminator for the task, while the approachin [5] exploits a new energy-related feature, called mod-iﬁed low energy ratio, that improves the results obtainedwith the classical low energy ratio.In this paper, we present our contribution to the de-sign of a robust speech/music discrimination system. Sirtuin 1-mediated autophagy regulates testosterone synthesis in Leydig cells of piglets

Abstract:Testosterone is secreted by Leydig cells (LCs), which play an important physiological role in preserving male secondary sex characteristics, protecting male reproductive function, and establishing the blood-testis barrier. Studies have shown that autophagy is particularly active in LCs; however, its involvement in testosterone synthesis in porcine LCs has not been fully explored. Therefore, this experiment aimed to investigate the influence of autophagy on testosterone secretion in porcine LCs and its potential regulatory mechanism. Our results demonstrated that both testicular autophagy and serum testosterone levels increased in piglets during postnatal development from 4 to 18 weeks. In addition, autophagy was found to degrade the Na+/H+ exchange regulatory factor 2 (NHERF2), leading to the up-regulation of scavenger receptor class B type 1 (SRB1). This process resulted in increased cholesterol intake and enhanced testosterone production. The observable level of sirtuin 1 (SIRT1) was directly proportional to the level of autophagy. In vitro investigations have shown that SIRT1 can affect the level of autophagy, cholesterol uptake as well as testosterone release. In conclusion, testosterone synthesis during pig development is regulated by SIRT1. SIRT1 mediates the degradation of NHERF2 through autophagy, thereby weakening its negative regulatory effect on the high-density lipoprotein receptor SRB1 in Leydig cells. This process increases cholesterol uptake and enhances testosterone synthesis.

Abstract:The effect of chronic pulsatile low-dose GnRH treatment on the juvenile testis and associated structures was evaluated in relation to hormonal parameters in the peripheral blood in the pig. Starting at 8 weeks of age, male pigs (crossbreds of Dutch Landrace and Yorkshire breeds) were injected 6 times daily im with 0, 75 or 250 ng GnRH/kg body weight during 4 weeks. Immediately after the treatment period, a GnRH stimulation test with 750 ng GnRH/kg iv was carried out. Samples for plasma LH, FSH, testosterone and 5 alpha DHT measurement were obtained weekly (basal level) and after GnRH stimulation. The pigs were castrated at 12 weeks of age and the weights and lengths of the testis, epididymis and cremaster muscle were recorded. Intratesticular testosterone and 5 alpha DHT concentrations were determined, and the testis and epididymis were evaluated for histological changes. Basal plasma hormone concentrations, intratesticular androgen concentrations and the response of the pituitary gland to stimulation had not been affected by GnRH treatment. Pigs receiving the higher treatment dose of GnRH showed less increase in testosterone levels in response to the stimulation dose at 12 weeks of age than the other pigs. Morphologically, no changes were observed in the epididymis and cremaster muscle after GnRH treatment and no signs of reactivation of structures that can provoke testicular descent could be seen. The development of the seminiferous epithelium was more advanced in the GnRH-treated groups, apparently in a dose-dependent manner.

Abstract:Spermatogenesis is a complex biological process crucial for male reproduction and is characterized by intricate interactions between testicular somatic cells and germ cells. Due to the cellular heterogeneity of the testes, investigating different cell types across developmental stages has been challenging. Single-cell RNA sequencing (scRNA-seq) has emerged as a valuable approach for addressing this limitation. Here, we conducted an unbiased transcriptomic study of spermatogenesis in sexually mature 4-month-old Hezuo pigs using 10× Genomics-based scRNA-seq. A total of 16,082 cells were collected from Hezuo pig testes, including germ cells (spermatogonia (SPG), spermatocytes (SPCs), spermatids (SPTs), and sperm (SP)) and somatic cells (Sertoli cells (SCs), Leydig cells (LCs), myoid cells (MCs), endothelial cells (ECs), and natural killer (NK) cells/macrophages). Pseudo-time analysis revealed that LCs and MCs originated from common progenitors in the Hezuo pig. Functional enrichment analysis indicated that the differentially expressed genes (DEGs) in the different types of testicular germ cells were enriched in the PI3K–AKT, Wnt, HIF-1, and adherens junction signaling pathways, while the DEGs in testicular somatic cells were enriched in ECM–receptor interaction and antigen processing and presentation. Moreover, genes related to spermatogenesis, male gamete generation, sperm part, sperm flagellum, and peptide biosynthesis were expressed throughout spermatogenesis. Using immunohistochemistry, we verified several stage-specific marker genes (such as UCHL1, WT1, SOX9, and ACTA2) for SPG, SCs, and MCs. By exploring the changes in the transcription patterns of various cell types during spermatogenesis, our study provided novel insights into spermatogenesis and testicular cells in the Hezuo pig, thereby laying the foundation for the breeding and preservation of this breed.

Identification and Analysis of Genes Related to Testicular Size in 14-Day-Old Piglets

Abstract:The RNA-Seq technology was used to screen the key genes that affect the early development of the testes of Duroc × Landrace × Yorkshire piglets, to determine the regulatory pathway and provide reference for subsequent reproductive performance research, breeding, and other production practices. This study selected 14-day-old Duroc × Landrace × Yorkshire piglets as the trial animals. Testes from piglets with similar weights and no pathological changes were divided into small testis (ST) and large testis (LT) groups, and the RNA-Seq screening of differentially expressed genes (DEGs) was performed to find candidate genes and regulatory pathways related to early testicular development. The results show that 570 DEGs were found in the ST and LT groups, with 281 upregulated and 289 downregulated. The DEGs were mainly enriched on 47 gene ontology (GO) functional items. The Kyoto encyclopedia of genes and genotypes (KEGG) enrichment analysis found that there were 44 significantly enriched KEGG signal pathways, and the regulation of testicular development mainly focused on the arachidonic acid metabolism, Wnt signaling pathway and GnRH secretion pathways. The PTGES, SFRP1, SPP1, PLA2G4E, KCNJ5, PTGS2, and HCN1 genes were found to be as closely related to the testicular development of these Duroc × Landrace × Yorkshire piglets, and the differential gene expression was consistent with the real-time quantitative reverse transcription PCR (real-time qRT-PCR) validation results. This study was validated by high-throughput sequencing analysis and real-time qRT-PCR, and showed that the PTGES, SFRP1, SPP1, PLA2G4E, KCNJ5, PTGS2, and HCN1 genes may be involved in the regulation of germ cell development, spermatogenesis and semen traits. These should be further studied as candidate genes for early testicular development and reproductive trait regulation in boars.

Gene Expression in Porcine Bulbourethral Glands

Abstract:The porcine bulbourethral glands produce a gel-type secretion. Although the role of these contributions to reproductive success remains murky, the bulbourethral glands are major accessory sex glands in this species. Isometric growth in the early neonatal interval is followed by allometric growth in the late juvenile interval (6 to 11 weeks of age), while circulating endogenous steroids are low. The rate of allometric growth increases during the peripuberal interval (16 to 20 weeks of age) when systemic testosterone is relatively high. Gene expression for androgen receptor (AR) and for the steroid 5 alpha-reductase 2 (SRD5A2) enzyme that synthesizes the more potent androgen dihydrotestosterone from its precursor was evaluated by qPCR analyses of bulbourethral gland tissue. Tissues were collected from control boars (2 weeks to 40 weeks of age) and from littermates of these boars treated with letrozole to suppress endogenous estrogen synthesis. Gene expression for these two key proteins in androgen signaling was quite low during the initial allometric growth in the late juvenile and prepuberal intervals, suggesting that this initial growth was not primarily stimulated by androgens. These observations are consistent with a more direct estrogen-mediated inhibition of growth via GPER previously proposed, with the sensitivity extending into the late juvenile interval when estrogens as well as androgens are normally relatively low.

Abstract:From fetal life to adulthood, the testis evolves through maturational phases showing specific morphologic and functional features in its different compartments. The seminiferous cords contain Sertoli and germ cells, surrounded by peritubular cells, and the interstitial tissue contains Leydig cells and connective tissue. Sertoli cells secrete anti-Müllerian hormone (AMH), whereas Leydig cells secrete androgens. In the fetal and early postnatal testis, Leydig cells actively secrete androgens. Sertoli cells are morphologically and functionally immature--e.g., they secrete high levels of AMH--and germ cells proliferate by mitosis but do not enter meiosis. During infancy and childhood, Leydig cells regress and testosterone secretion declines dramatically. Sertoli cells remain immature and spermatogenesis is arrested at the premeiotic stage. At puberty, Leydig cells differentiate again, and testosterone concentration increases and provokes Sertoli cell maturation--e.g., down-regulation of AMH expression--and germ cells undergo meiosis, the hallmark of adult spermatogenesis driving to sperm production. An intriguing feature of testicular development is that, although testosterone production is as active in the fetal and early postnatal periods as in puberty, Sertoli cells and spermatogenesis remain immature until pubertal onset. Here, we review the ontogeny of the androgen receptor expression in the testis and its impact on Sertoli cell maturation and the onset of pubertal spermatogenesis. We show that the absence of androgen receptor expression in Sertoli cells underlies a physiological stage of androgen insensitivity within the male gonad in the fetal and early postnatal periods.

Semen Modulates Cell Proliferation and Differentiation-Related Transcripts in the Pig Peri-Ovulatory Endometrium

Abstract:Uterine homeostasis is maintained after mating by eliminating pathogens, foreign cells, and proteins by a transient inflammation of the uterus. Such inflammation does not occur in the oviductal sperm reservoir (utero-tubal junction, UTJ), colonized by a population of potentially fertile spermatozoa before the inflammatory changes are triggered. Semen entry (spermatozoa and/or seminal plasma) modifies the expression of regulatory genes, including cell proliferation and differentiation-related transcripts. Considering pigs display a fractionated ejaculation, this study aims to determine whether different ejaculate fractions differentially modulate cell proliferation and differentiation-related transcripts in the sow reproductive tract during the peri-ovulatory stage. Using species-specific microarray analyses, the differential expression of 144 cell proliferation and differentiation-related transcripts was studied in specific segments: cervix (Cvx), distal and proximal uterus (DistUt, ProxUt), UTJ, isthmus (Isth), ampulla (Amp), and infundibulum (Inf) of the peri-ovulatory sow reproductive tract in response to semen and/or seminal plasma cervical deposition. Most mRNA expression changes were induced by mating. In addition, while mating upregulates the fibroblast growth factor 1 (FGF1, p-value DistUt = 0.0007; ProxUt = 0.0253) transcript in the endometrium, both its receptor, the fibroblast growth factor receptor 1 (FGFR1, p-value DistUt = 2.14 e−06; ProxUt = 0.0027; UTJ = 0.0458) transcript, and a potentiator of its biological effect, the fibroblast growth factor binding protein 1 (FGFBP1), were downregulated in the endometrium (p-value DistUt = 0.0068; ProxUt = 0.0011) and the UTJ (p-value UTJ = 0.0191). The FGFBP1 was downregulated in the whole oviduct after seminal depositions (p-value Isth = 0.0007; Amp = 0.0007; Inf = 6.87 e−05) and, interestingly, FGFR1 was downregulated in the endometrium in the absence of semen (p-value DistUt = 0.0097; ProxUt = 0.0456). In conclusion, the findings suggest that spermatozoa, seminal components, and the act of mating trigger, besides inflammation, differential mechanisms in the peri-ovulatory female reproductive tract, relevant for tissue repair.

Changes in the concentration of follicle-stimulating hormone in plasma during development in the guinea-pig

Abstract:The concentration of FSH in the plasma of guinea-pigs from day 50 of gestation to day 45 of postnatal life was assayed by a radioimmunological procedure utilizing a cross-reaction with the NIAMDD S6 antiserum to rat FSH. At 68 days of gestation the mean plasma FSH concentration of female foetuses was greater than that of the males, although the concentrations in the two sexes were similar on day 50 of gestation. Maternal levels remained consistently low throughout gestation. Postnatally there were no marked changes in plasma FSH levels through to maturity, although a transient rise in the male occurred over the first 3 days after birth. Gonadectomy on days 0, 5, 10, 15, 25 or 35 of postnatal life, or when adult, resulted in a significant increase in plasma levels of FSH within 10 days. The rise in plasma FSH concentration was greater in males than in females at all ages, although, a larger increase was observed in females spayed at 5 or 10 days of age than at other times.

Abstract:The testis produces gametes through spermatogenesis and evolves rapidly at both the morphological and molecular level in mammals 1,2,3,4,5,6 , probably owing to the evolutionary pressure on males to be reproductively successful 7 . However, the molecular evolution of individual spermatogenic cell types across mammals remains largely uncharacterized. Here we report evolutionary analyses of single-nucleus transcriptome data for testes from 11 species that cover the three main mammalian lineages (eutherians, marsupials and monotremes) and birds (the evolutionary outgroup), and include seven primates. We find that the rapid evolution of the testis was driven by accelerated fixation rates of gene expression changes, amino acid substitutions and new genes in late spermatogenic stages, probably facilitated by reduced pleiotropic constraints, haploid selection and transcriptionally permissive chromatin. We identify temporal expression changes of individual genes across species and conserved expression programs controlling ancestral spermatogenic processes. Genes predominantly expressed in spermatogonia (germ cells fuelling spermatogenesis) and Sertoli (somatic support) cells accumulated on X chromosomes during evolution, presumably owing to male-beneficial selective forces. Further work identified transcriptomal differences between X- and Y-bearing spermatids and uncovered that meiotic sex-chromosome inactivation (MSCI) also occurs in monotremes and hence is common to mammalian sex-chromosome systems. Thus, the mechanism of meiotic silencing of unsynapsed chromatin, which underlies MSCI, is an ancestral mammalian feature. Our study illuminates the molecular evolution of spermatogenesis and associated selective forces, and provides a resource for investigating the biology of the testis across mammals.

Abstract:Sertoli cells are somatic cells present in seminiferous tubules which have essential roles in regulating spermatogenesis. Considering that each Sertoli cell is able to support a limited number of germ cells, the final number of Sertoli cells reached during the proliferative period determines sperm production capacity. Only immature Sertoli cells, which have not established the blood-testis barrier, proliferate. A number of hormonal cues regulate Sertoli cell proliferation. Among them, FSH, the insulin family of growth factors, activin, and cytokines action must be highlighted. It has been demonstrated that cAMP/PKA, ERK1/2, PI3K/Akt, and mTORC1/p70SK6 pathways are the main signal transduction pathways involved in Sertoli cell proliferation. Additionally, c-Myc and hypoxia inducible factor are transcription factors which participate in the induction by FSH of various genes of relevance in cell cycle progression. Cessation of proliferation is a pre-requisite to Sertoli cell maturation accompanied by the establishment of the blood-testis barrier. With respect to this barrier, the participation of androgens, estrogens, thyroid hormones, retinoic acid and opioids has been reported. Additionally, two central enzymes that are involved in sensing cell energy status have been associated with the suppression of Sertoli cell proliferation, namely AMPK and Sirtuin 1 (SIRT1). Among the molecular mechanisms involved in the cessation of proliferation and in the maturation of Sertoli cells, it is worth mentioning the up-regulation of the cell cycle inhibitors p21Cip1, p27Kip, and p19INK4, and of the gap junction protein connexin 43. A decrease in Sertoli cell proliferation due to administration of certain therapeutic drugs and exposure to xenobiotic agents before puberty has been experimentally demonstrated. This review focuses on the hormones, locally produced factors, signal transduction pathways, and molecular mechanisms controlling Sertoli cell proliferation and maturation. The comprehension of how the final number of Sertoli cells in adulthood is established constitutes a pre-requisite to understand the underlying causes responsible for the progressive decrease in sperm production that has been observed during the last 50 years in humans.

Abstract:Our understanding of calcific aortic stenosis has changed dramatically during the past 3 decades, with the concept of a "mechanical" disease of aging now replaced by the concept of an active disease process at the tissue level that may be amenable to medical therapy. The ability of echocardiography to provide early diagnosis and an accurate measurement of disease severity has increased our knowledge of the natural history of this disease process and allows us to follow individual patients over time, long before valve replacement is needed. We now recognize that even mild symptoms are an indication for valve replacement when severe obstruction is present. This review discusses the optimal approach to measurement of disease severity, the presymptomatic disease course, and the underlying causes of calcific valve disease, followed by a summary of clinical trials of medical therapy and the current indications and choices for valve replacement.
