Abstract:Therapeutic use of electroconvulsive shock (ECS) is 75% effective for the remission of treatment-resistant depression. Like other more common forms of antidepressant treatment such as fluoxetine, ECS has been shown to increase neurogenesis in the hippocampal dentate gyrus of rodent models. Yet the question of how ECS-induced neurogenesis supports improvement of depressive symptoms remains unknown. Here, we show that ECS-induced neurogenesis is necessary to improve depressive-like behavior of mice exposed to chronic corticosterone (Cort). We then use slice electrophysiology to show that optogenetic stimulation of adult-born neurons produces a greater hyperpolarization in mature granule neurons after ECS vs Sham treatment. We identify that this hyperpolarization requires the activation of metabotropic glutamate receptor 2 (mGluR2). Consistent with this finding, we observe reduced expression of the immediate early gene cFos in the granule cell layer of ECS vs Sham subjects. We then show that mGluR2 knockdown specifically in ventral granule neurons blunts the antidepressant-like behavioral effects of ECS. Using single nucleus RNA sequencing, we reveal major transcriptomic shifts in granule neurons after treatment with ECS+Cort or fluoxetine+Cort vs Cort alone. We identify a population of immature cells which has greater representation in both ECS+Cort and fluoxetine+Cort treated samples vs Cort alone. We also find global differences in ECS-vs fluoxetine-induced transcriptomic shifts. Together, these findings highlight a critical role for immature granule cells and mGluR2 signaling in the antidepressant action of ECS.

Differential changes in the expression of cyclic nucleotide phosphodiesterase isoforms in rat brains by chronic treatment with electroconvulsive shock

Abstract:Electroconvulsive shock (ECS) has been suggested to affect cAMP signaling pathways to exert therapeutic effects. ECS was recently reported to increase the expression of PDE4 isoforms in rat brain, however, these studies were limited to PDE4 family in the cerebral cortex and hippocampus. Thus, for comprehensive understanding of how ECS regulates PDE activity, the present study was performed to determine whether chronic ECS treatment induces differential changes in the expression of all the PDE isoforms in rat brains. We analyzed the mRNA expression of PDE isoforms in the rat hippocampus and striatum using reverse transcription polymerase chain reaction. We found chronic ECS treatment induced differential changes in the expression of PDE isoform 1, 2, 3, 4, 5 and 7 at the rat hippocampus and striatum. In the hippocampus, the expression of PDE1A/B (694%), PDE4A (158%), PDE4B (323 %), and PDE4D (181%) isoforms was increased from the controls, but the expression of PDE2 (62.8%) and PDE7 (37.8%) decreased by chronic ECS treatment. In the striatum, the expression of PDE1A/B (179%), PDE4A (223%), PDE4B (171%), and PDE4D (327%) was increased by chronic ECS treatment with the concomitant decrease in the expression of PDE2 (78.4%) and PDE3A (67.1%). In conclusion, chronic ECS treatment induces differential changes in the expression of most PDE isoforms including PDE1, PDE2, PDE3, PDE4, PDE5, and PDE7 in the rat hippocampus and striatum in an isoform- and brain region-specific manner. Such differential change is suggested to play an important role in regulation of the activity of PDE and cAMP system by ECS.

Abstract:N-methyl-D-asparate-receptors (NMDAR) are implicated in several neuropathological conditions including epilepsy. As a model of epilepsy, genetically epilepsy-prone rat was chosen to invesitegate the changes in expression of NMDAR1 mRNA after seizure in different brain regions. The results showed that cerebral cortex, hippocampus, subcortex and inferior colliculus had a time-dependent modulation of the NMDAR1 mRNA expression and increased about 111%, 113%, 165%, 202% above control values, respectively in 24h after seizure. This phenomenon demonstrates that NMDAR1 mRNA was regulated by seizure and may contribute to epileptic susceptibility.

Abstract:Background: Volumetric changes and glial pathology have been reported in the central nervous system (CNS) of patients with depressive disorder, an illness often associated with elevated glucocorticoid levels. Glucocorticoids reduce gliogenesis in the adult rat CNS. Electroconvulsive seizure (ECS)-treatment, an animal model for the antidepressant treatment electroconvulsive therapy, can enhance proliferation of glial cells. This study examined glial cell proliferation in response to ECS in rats whose glucocorticoid levels were elevated to mimic the conditions seen in depression. Methods: Rats were injected daily for seven days with either corticosterone or vehicle. ECS- or sham- treatment was given once daily during the first five days. Proliferating cells in the hippocampus were labeled with bromodeoxyuridine and analyzed for co-labeling with the glial cell markers NG2, Ox42, S-100beta and Rip. Results: ECS counteracted the glucocorticoid-induced inhibition of NG2+, Ox42+ and Rip+ cell proliferation, and the gliogenesis rate was restored to baseline levels. Volumetric changes in rats treated with ECS were detected. Conclusions: Our results show that ECS-treatment affects the proliferation of glial cells even in the presence of elevated levels of glucocorticoids. This result adds to an increasing number of studies suggesting that antidepressant treatment can counteract degenerative processes associated with major depression.

Abstract:Zinc transporters (ZnTs) and plasticity-related genes (PRGs) both play the key roles in the formation of hippocampal mossy fiber sprouting, which is associated with cognitive deficits following developmental seizures. Here, for the first time, we report the timing of expression pattern of ZnT-1, ZnT-3 and PRG-1 in hippocampus and cerebral cortex following developmental seizures. A seizure was induced by inhalant flurothyl daily in neonatal Sprague–Dawley rats from postnatal day 6 (P6). Rats were assigned into the recurrent-seizure group (RS, seizures induced in 6 consecutive days) and the control group. At 1.5h, 3h, 6h, 12h, 24h, 48h, 7d and 14d after the last seizures, the mRNA level was detected using RT-PCR method; PRG-1 protein level was examined by Western blotting analysis. At an early period of 12h and 48h after the last seizures, both ZnT-1 and ZnT-3 showed significantly down-regulated mRNA level in the cerebral cortex of RS group than those at the corresponding time point in control group. In the long-term time point of 14d after the last seizure, ZnT-3 mRNA and PRG-1 protein level in hippocampus were up-regulated while the mRNA level of ZnT-1 down-regulated; in addition, there were up-regulated level of both the mRNA and protein level of PRG-1 and down-regulated mRNA level of ZnT-3 in the cerebral cortex of RS group when compared to the control. Taken together, these dates are consistent with an important role for ZnT-1, ZnT-3 and PRG-1 in the pathophysiology of the long-term adverse effects of recurrent neonatal seizure-induced hippocampal mossy fiber sprouting and cognitive deficit.

Abstract:Objectives To observe cannabinoid receptor 1 mRNA expression in rat hippocampus at different stages of sleep deprivation after epilepsy. Methods Fifty male Sprague-Dawley rats weighing 220-250 g were divided randomly into two groups, the non-epileptic group as control (n = 25) and the epileptic group (n = 25). Each of the groups was subsequently divided randomly into cage control (CC), tank control (TC) and the sleep deprivation (SD) groups. The modified multiple platform methods were used in SD groups for the rapid eye movement sleep deprivation for 1 day and 3 or 5 consecutive days. Pentetrazole (FTZ) was used to induce convulsion. CB1 receptor mRNA was measured by using reverse transcription-polymerase chain reaction. The structural changes of hippocampus at different stages were studied using electro-microscope. Results Neuron apoptosis were found in the SD groups. After PTZ injection the SD groups suffered serious convulsion while the CC and TC groups did not. The CC and TC groups displayed increased expression of CB1 receptor after PTZ injection (P < 0.01). The expression of CB1 receptor mRNA in SD had no significant changes on different days of sleep deprivation between non-epileptic groups and epileptic groups (P > 0.05). Conclusions Sleep deprivation can cause neuron apoptosis and modify the expression of CB1 receptor mRNA. The increased expression of CB1 receptor might be a protective response to the prolonged wakefulness for neurons.
Long-Term Effects Of Early Seizure-Like Discharge On Neuronal Function And Nmdar Subunit Mrna Expression In Developing Cortical Neurons Of Rat

Abstract:Objective: To explore the long-term effect of early seizure-like discharge on neuronal function and NMDAR subunit mRNA expression in developing cortical neurons of rat. Methods: The seizure model was recurrent epileptiform discharges induced by transient magnesium-free media treatment in the primary cultured embryo cortical neurons of rat. According to different treatment to cortical neurons at cultured day 6, three groups were divided: regular DMEM medium (NOT), regular extracellular solution media (CONT) and magnesium-free extracellular solution media (MGF). The neurons were exposed to extracellular solution for 3 h at cultured day 6 in either CONT or MGF group, and then cultured in regular DMEM medium again. The expression of NMDAR subunit NR1, NR2A and NR2B mRNA was measured using fluorescent real-time quantitative RTPCR and MTT metabolic rate were detected at different cultured day (7, 12, 17 d). Results: Compared with group NOT and CONT respectively, in group MGF MTT metabolic rate decreased at 7 d and 12 d, NR1 mRNA expression decreased at 7 d and increased at 12 d(P<0.05), NR2A mRNA expression decreased at 12 d and increased at 17 d (P<0.05), and NR2B mRNA expression increased at 7 d and 17 d (P<0.05). There were not statistic significant differences of MTT metabolic rat at 17 d and mRNA expression of NR1 at 17 d, NR2A at 7 d, and NR2B at 12 d among three groups (P>0.05). Conclusion: The change of neuronal function and NMDAR subunit mRNA expression may last for relatively long time after early seizure like discharge in developing cortical neurons of rat.

Abstract:Epilepsy in children is associated with a broad spectrum of cognitive deficits, which is associated with hippocampal mossy fiber sprouting. The underlying molecular mechanisms involved in mossy fiber sprouting in hippocampus following developmental seizures are not completely known. We studied the timing of cognitive dysfunction following neonatal seizures and the relation of this cognitive impairment to zinc transporter 1 (ZnT-1), 3 (ZnT-3), calcium/calmodulin-dependent protein kinase II (CaMK II), plasticity-related gene 1 (PRG-1) expression in hippocampus. A seizure was induced by inhalant flurothyl daily in neonatal Sprague-Dawley rats from postnatal day 6 (P6). Rats were assigned into the single-seizure group (SS), the recurrent-seizure group (RS, seizures induced in six consecutive days), and the control group. During P41–P46 and P85–P90, the rats were tested for spatial learning and memory abilities with automatic Morris water maze task. At P90, mossy fiber sprouting and gene expression in hippocampus were determined subsequently by Timm staining and RT-PCR methods. The escape latencies from the water maze were significantly longer in rats of RS group than those of the control and SS groups at d4 of the first maze test and at d3, d4 of the second maze test. As far as Spatial Probe Test was concerned, the frequency of passing through the platform quadrant was significantly decreased in RS group than that in control and SS groups in the entire two probe tests. In rats with recurrent seizures (RS group), there was an increased distribution of Timm granules in both the supragranular region of the dentate gyrus and the stratum pyramidale of CA3 subfield in RS group, while remaining barely visible in control and SS groups; the Timm scores in CA3 and dentate gyrus in the RS animals were significantly higher than that in the control and SS groups. RT-PCR densitometry analysis showed that the ratios of hippocampal ZnT-1 to β-actin of SS and RS group were decreased significantly compared with that of control group. Meanwhile, CaMK II to β-actin of RS group was markedly lower compared with those of SS and control groups. Our results suggest that the long-term adverse effects of recurrent neonatal seizures on cognition and mossy fiber sprouting may be associated with the down-regulated expression of ZnT-1 and CaMK II in hippocampus.
A Transient Upregulation Of Glutamine Synthetase In The Dentate Gyrus Is Involved In Epileptogenesis Induced By Amygdala Kindling In The Rat

Abstract:Reduction of glutamine synthetase (GS) function is closely related to established epilepsy, but little is known regarding its role in epileptogenesis. The present study aimed to elucidate the functional changes of GS in the brain and its involvement in epileptogenesis using the amygdala kindling model of epilepsy induced by daily electrical stimulation of basolateral amygdala in rats. Both expression and activity of GS in the ipsilateral dentate gyrus (DG) were upregulated when kindled seizures progressed to stage 4. A single dose of L-methionine sulfoximine (MSO, in 2 mu l), a selective GS inhibitor, was administered into the ipsilateral DG on the third day following the first stage 3 seizure (just before GS was upregulated). It was found that low doses of MSO (5 or 10 mu g) significantly and dose-dependently reduced the severity of and susceptibility to evoked seizures, whereas MSO at a high dose (20 mu g) aggravated kindled seizures. In animals that seizure acquisition had been successfully suppressed with 10 mu g MSO, GS upregulation reoccurred when seizures re-progressed to stage 4 and re-administration of 10 mu g MSO consistently reduced the seizures. GLN at a dose of 1.5 mu g abolished the alleviative effect of 10 mu g MSO and deleterious effect of 20 mu g MSO on kindled seizures. Moreover, appropriate artificial microRNA interference (1 and 1.5x10(6) TU/2 mu l) of GS expression in the ipsilateral DG also inhibited seizure progression. In addition, a transient increase of GS expression and activity in the cortex was also observed during epileptogenesis evoked by pentylenetetrazole kindling. These results strongly suggest that a transient and region-specific upregulation of GS function occurs when epilepsy develops into a certain stage and eventually promotes the process of epileptogenesis. Inhibition of GS to an adequate degree and at an appropriate timing may be a potential therapeutic approach to interrupting epileptogenesis.

Abstract:Industrial workers more and more frequently seek medical advice for nasal and sinus symptoms that they attribute to occupational exposure. The present study comprised 10 paint-sprayers exposed to solvents (primarily toluene and isobutylacetate) and dust. The working environment was checked by an industrial hygienist and a careful clinical examination including biopsies from the nasal mucosa was carried out. The results revealed prominent nasal symptoms in 3 patients and 4 suffered from cough. The histological examination showed in no case a normal nasal mucosa. The histological grading showed a significantly higher score in the exposed group compared to a matched control group. However, no conclusive connection between the duration of exposure and the clinical symptoms and morphological changes was possible to make, which will necessitate an extended study to evaluate the exact importance of the duration of exposure. The study indicates the possibilities of an early presymptomatic detection of nasal mucosal disturbances and arises the question whether the existing TLVs are adequate in preventing damage to the nasal mucosa and adherent clinical symptoms.
[Effects of Edaravone on the Expression of GRP78, Caspase-12, and Neuron Apoptosis in Juvenile Rat Hippocampus after Status Convulsive].

Abstract:OBJECTIVE:To observe the expression of GRP78 (glucose regulated protein, GRP78), Caspase-12 and the change of neuron apoptosis in the juvenile rat hippocampus after status convulsive (SC), and to explore the effect of edaravone on them.METHODS:One hundred and ninety-five juvenile male Sprague-Dawley (SD) rats were randomly divided into normal saline control group (NS group), status convulsive group (SC group) and edaravone treatment group (ED group). Each group was further divided into five subgroups in different executed time points after SC. The rats in status convulsive group were kindled into epilepsy by lithium-pilocarpine method. Expression of GRP78 mRNA and caspase-12 mRNA was detected with reverse transcription-polymerase chain reaction (RT-PCR) method. Expressions of GRP78 and caspase-12 protein were detected with immunohistochemical methods. The neuron apoptosis was observed by TdT-mediated dUTP nick end labeling (TUNEL).RESULTS:(1) Measured by immunohistochemistry the value of OD of GRP78 (0.1480 ± 0.0164, 0.1682 ± 0.0114, and 0.1540 ± 0.0102, respectively, 12 h - 48 h points) and caspase-12 (0.1325 ± 0.0165, 0.1794 ± 0.0213, 0.1525 ± 0.0423, and 0.1309 ± 0.0199, respectively, 12 h-72 h points) positive cells in the SC group increased, there was a significant difference compared with NS group (GRP78: 0.1214 ± 0.0147, 0.1272 ± 0.0177, and 0.1260 ± 0.0157, respectively, 12 h-72 h points. Caspase-12: 0.1050 ± 0.0121, 0.1041 ± 0.0151, 0.1058 ± 0.0222, and 0.1036 ± 0.0186, respectively, 12 h - 72 h points) (P < 0.01, or P < 0.05). By ED intervention GRP78 (0.1550 ± 0.0131, 0.1886 ± 0.0154, and 0.1721 ± 0.0151, respectively, 12 h - 48 h points) positive cells value of the OD increased as compared with SC group (P < 0.01, or P < 0.05). and caspase-12 (0.1211 ± 0.0184, 0.1545 ± 0.0205, and 0.1085 ± 0.0219, respectively, 12 h, 24 h and 72 h points) positive cells value of the A decreased as compared with SC group (P < 0.01, or P < 0.05). (2) Measured by RT-PCR, the expression of GRP78 mRNA and caspase-12 mRNA trend was similar to protein. (3) The TUNEL positive cells in hippocampus CA(1) of SC group (11.41 ± 2.37) were more than that of NS group after the SC 12 h (P < 0.01), reached its highest level at 48 h (28.78 ± 5.11), after the intervention with edaravone (8.98 ± 2.22, 13.09 ± 2.54 and 20.57 ± 4.89, respectively, 12 h-48 h points), TUNEL positive cells showed a significant drop in SC group at 12 h-48 h time points (P < 0.01, or P < 0.05), but still significantly higher than that of the NS group (6.22 ± 1.50, 6.57 ± 1.61 and 6.72 ± 1.14, respectively) (P < 0.01, or P < 0.05), at the 4 h time point (NS group 6.29 ± 1.49, SC group 6.61 ± 1.71, ED group 5.75 ± 1.41) among the three groups, no significant difference in TUNEL positive cells was found (P = 0.759).CONCLUSIONS:The expression of GRP78 and caspase-12 increased after SC. Edaravone increased expression of GRP78 and decreased expression of caspase-12 in hippocampus rat with pilocarpine-induced seizures, reduced the number of neuronal apoptosis. These results suggest that edaravone may have protective effect against the hippocampal damage caused by status convulsive.
[Effects of Recurrent Febrile Seizures on Gamma-Aminobutyric Acid B Receptor Subunit Expressions in the Hippocampus of Developing Rats].

Abstract:OBJECTIVE:To study gamma-aminobutyric acid (GABA) B receptor subunits GABABR1 and GABABR2 protein expression changes after recurrent febrile seizures in the hippocampus of developing rats.METHODS:The rats were randomly divided into hyperthermia-treated group (n = 68) and control group (n = 24). According to the times of treatment, and to whether seizures developed, hyperthermia-treated group were subdivided into 4 groups: F1 (febrile, one-time, n = 12), FS1 (febrile seizure, one-time, n = 16), F10 (febrile, ten-time, n = 13), FS10 (febrile seizure, ten-time, n = 15). Immunohistochemical staining was used to demonstrate GABABR1 and GABABR2 expressions in brain sections (12 microns each).RESULTS:GABABR1 and GABABR2 protein expressions significantly decreased in dentate gyrus and CA1-CA3 neurons in FS10 compared with those in F10, F1, FS1 and control groups. The expressions of GABABR1 and GABABR2 protein showed less intense in F10 than those in FS1, F1 and control groups. No significant differences were observed among F1, FS1 and control group. Throughout the hippocampus, the expressions of GABABR1 and GABABR2 protein largely overlaped. GABABR1 protein expressions decreased seriously as compared with the expressions of GABABR2 protein in dentate gyrus, whereas GABABR2 protein expressions decreased more apparently in hippocampus CA1 and CA3 than that of GABABR1 in FS10.CONCLUSION:Recurrent febrile seizures and recurrent hyperthermia all can make the GABABR receptor protein expression down-regulated in the immature rat brain, and GABABR protein expression decreased more apparently in recurrent febrile seizure groups than that in recurrent hyperthermia groups. These suggest that GABABR subunits may play an important role in the immature rat febrile seizures and the difference between GABABR1 and GABABR2 protein expression changes is possible as a result of subunit reorganization, which may lead to the changes of the inhibitory function.

Long-term Effects of Neonatal Seizures on Subsequent Activity-Dependent NR1 and GABAA Receptor Α1 Expressions in Hippocampus of the Rat

Abstract:Objective To evaluate the pathophysiological mechanism of reduced seizure threshold following neonatal seizures by measuring the expression changes in N-methyl-D-aspartate (NMDA)receptor 1(NR1)and gamma-aminobutyric acid receptor A α1(GABAARα1)immunoreactivity which are the basic functional subunit in NMDA receptor or GABAA receptor family. Methods Seizures were induced by the inhalant flurothyl in 24 neonatal rats,starting from postnatal day 6.The 24 rats were subdivided into single seizure group (seizure lasted 30 min) and recurrent seizure group (seizures were induced for 30 min/d, totally 6 d). At day 7 and day 75 after the last seizure, brain homogenates were made. The expressions of NR1 and GABAARα1 proteins in hippocampus were examined by Westem blotting analysis. Results NR1 expression did not change significantly in single seizure group and recurrent seizure group at day 75, but was enhanced significantly in recurrent seizure group at day 7(P＜0.05).Meanwhile,polypeptide levels of GABAARα1 receptor subullit in the rat hippocanlpus decreased significantly in single seizure-treated rats at day 75 and recurrent seizure-treated rats at day 7 and 75 (P＜0.05). Conclusions Recurrent or single prolonged status epilepticus in neonatal rats might cause long-term modification on NR1 and GABAARα1 expressions in hippocampus at post-seizure 7 d or in adult, with excessive activation of excitatory neuronal circuits involving both reduction of GABAARα1 receptor and/or enhancement of NR1. This phenomenon might play a key role in long-term reduction of seizure threshold induced by early life status seizures.

Abstract:MicroRNAs (miRNAs) are small and endogenously expressed non-coding RNAs that negatively regulate the expression of protein-coding genes at the translational level. Emerging evidence suggests that miRNAs play critical roles in central nervous system under physiological and pathological conditions. However, their expression and functions in status epilepticus (SE) have not been well characterized thus far. Here, by using high-throughput sequencing, we characterized miRNA expression profile in rat hippocampus at 24 hours following SE induced by amygdala stimulation. After confirmation by qRT-PCR, six miRNAs were found to be differentially expressed in brain after SE. Subsequent Kyoto Encyclopedia of Genes and Genomes pathway analysis indicated that most of the predicted target genes for these six miRNAs were related to neuronal apoptosis. We then investigated the dynamic changes of these six miRNAs at different time-point (4 hours, 24 hours, 1 week and 3 weeks) after SE. Meanwhile, neuronal survival and apoptosis in the hippocampus after SE were evaluated by Nissl staining and terminal deoxynucleotidyl transferase-mediated dUTP end-labeling assay. We found that the expression of miR-874-3p, miR-20a-5p, miR-345-3p, miR-365-5p, and miR-764-3p were significantly increased from 24 hours to 1 week, whereas miR-99b-3p level was markedly decreased from 24 hours to 3 weeks after SE. Further analysis revealed that the levels of miR-365-5p and miR-99b-3p were significantly correlated with neuronal apoptosis after SE. Taken together, our data suggest that miRNAs are important modulators of SE-induced neuronal apoptosis. These findings also open new avenues for future studies aimed at developing strategies against neuronal apoptosis after SE.

Abstract:Glycogen synthase kinase-3 (GSK-3) has been linked to prodepressant-like effects in rodents. However, the roles of GSK-3β and the hippocampal dentate gyrus in regulating these behavioral effects remain unclear.A lentiviral vector was utilized to site-specifically express GSK-3β constitutively in the hippocampal DG in a mouse model of CMS. We examine the forced swim, tail suspension and the sucrose intake test. Acute and chronic administrations were conducted by dissolving fluoxetine hydrochloride (10ml/kg). We examine behavior tests as before, cellular apoptosis, proliferation and differentiation in the hippocampus.GSK-3β expression levels persistently and significantly increased in the hippocampus following lenti-GSK-3β injections. In mice previously exposed to CMS, pre-injection of lentivirus-expressing GSK-3β into the hippocampal dentate gyrus significantly decreased sucrose preferences in the sucrose intake test and increased immobility times in both forced swim and tail suspension tests. In addition, fluoxetine resulted in similar antidepressant-like effects following chronic, but not acute, administrations under the same experimental conditions. Cellular apoptosis was observed in the hippocampal DG using TUNEL, revealing many TUNEL-positive cells in the lenti-GSK-3β mice. There were no significant changes in proliferation and differentiation.We did not measure more biomarkers which were regulated by GSK-3β.Results from this study demonstrated that site-specific injection of a lentivirus induced continuous GSK-3β expression in the hippocampal dentate gyrus of mice, resulting in prodepressant-like effects and increased sensitivity to chronic mild stress. Furthermore, chronic fluoxetine administration reversed these prodepressant-like effects and decreased neuronal apoptosis in the hippocampal DG in GSK-3β-overexpressing mice.

Abstract:To evaluate the long-term effects of single or recurrent prolonged neonatal seizures on seizure threshold and neuronal activity in the brain, a novel “twist” seizure was induced by coupling early-life flurothyl-induced seizures with later exposure to pentylenetetrazol. The authors assigned six neonatal rats for each group: the single-seizure group (SS), the recurrent-seizure group (RS) and the control group. At postnatal day 46, seizure threshold was examined using pentylenetetrazol, and then the brain slices were evaluated with thionine staining, in situ end labeling and immunohistochemical studies. The Results showed that the rats in SS and RS groups all had reduced latencies to develop generalized tonic seizures induced by PTZ compared with controls (P<0.01). Morphologic changes, cell loss and apoptotic cells were observed only in those of RS group. Significant fos and NR2C-immunoreactive positive cells were seen in hippocampus of rats in both SS and RS groups compared with controls (P<0.01). A significant decrease in the number of GABA-A-α1 immunoreactive positive neurons was detected in hippocampus in rats of SS and RS groups compared with the controls (P<0.01). We conclude that neonatal rats subjected to prolonged seizures have pronounced long-term effects on seizure threshold and neuronal neurophysiological activity in the brain. Obvious neuronal injury, however, was only seen in rat with recurrent-seizures. Subtle brain damage might occur in rats experiencing single prolonged neonatal seizures.
Activation of microglia in rat model of epilepsy during persistent state and analysis of temporal and spatial expression of different types of microglia

Abstract:Objective To investigate the activation of microglia in state of continuity (SE) of epileptic rat model and the temporal and spatial expression of different types of microglia.Methods 60 healthy male Wistar rats were randomly divided into control group,SE at the beginning (NC group),SE lasting for 30 minutes (SE30min group),SE lasting for 1 hour (SE1h group),SE holding.For 2 hours (SE2h),12 rats in each group.Behavioral changes and activation of microglia in hippocampus were observed.The expression levels of IL-6,IL-1 beta and IL-10 and IL-4 were detected.Results In the SE group,with the prolongation of time,the response of MG ceils increased,and the activation of MG cells was most obvious in SE2h group.The expression levels of IL-10,IL-4 protein and mRNA in SE1h group and SE2h group were significantly higher than those in NC group and SE30min group,while the expression levels of IL-6,IL-1beta protein and mRNA in M1 group were significantly lower than those in NC group and SE30min group (P ＜ 0.05).Conclusion Activation of microglia can be seen in pilocarpine-induced epilepsy rat model.With the prolongation of seizure time,polarization of microglia gradually transforms into neurotrophic M2 cells.The expression levels of anti-inflammatory factors IL-10 and IL-4 will increase,while the expression of pro-inflammatory factors IL-6 and IL-1 beta will decrease.
Effect of ceftriaxone on the glutamate-glutamine cycle and seizure susceptibility of Tg2576 mouse model of Alzheimer's disease

Abstract:Background: Individuals with Alzheimer's disease (AD) have a heightened risk of epilepsy. However, the underlying mechanisms are not well-understood. Objective: We aimed to elucidate the role of the glutamate-glutamine cycle in this mechanism and test the effect of ceftriaxone, a glutamate transporter-1 (GLT-1) enhancer, on seizure susceptibility in the Tg2576 mouse model of AD. Methods: First, we assessed expression levels of key proteins in the glutamate-glutamine cycle in Tg2576 (n = 7) and wild-type littermates (n = 7), and subsequently in the kindling model of epilepsy (n = 6) and sham (n = 6). Then, kindling susceptibility was assessed in three groups: 200 mg/kg ceftriaxone-treated Tg2576 (Tg-Ceft, n = 9); saline-treated Tg2576 (Tg-Sal, n = 9); and saline-treated wild-type (WT-Sal, n = 15). Mice were treated for seven days before kindling, and seizure susceptibility compared between groups. Results: Protein levels of GLT-1 (p = 0.0093) and glutamine synthetase (p = 0.0016) were reduced in cortex of Tg2576 mice, compared to WT. Kindling increased GLT-1 (cortex: p < 0.0001, hippocampus: p = 0.0075), and glutaminase (cortex: p = 0.0044) protein levels, compared to sham. Both Tg-Ceft and WT-Sal displayed Class IV seizures in response to the first stimulation (p > 0.99), while Tg-Sal displayed Class V seizure (p = 0.0212 versus WT-Sal). Seizure susceptibility of Tg-Ceft was not different from Tg-Sal (p > 0.05), and kindling rates did not differ between groups. Conclusions: Disruptions to key components of the glutamate-glutamine cycle are observed in models of AD and epilepsy. However, increasing GLT-1 through ceftriaxone treatment did not influence seizure susceptibility in Tg2576 mice, suggesting this is not an effective strategy to lower seizure susceptibility in AD, or a higher dosage is needed.
