Abstract:In this review, we introduce some chip-based waveguide biosensing and imaging techniques, which significantly reduce the complexity of the entire system. These techniques use a well-confined evanescent field to interact with the surrounding materials and achieve high sensitivity sensing and high signal-to-noise ratio (SNR) super-resolution imaging. The fabrication process of these chips is simple and compatible with conventional semiconductor fabrication methods, allowing high-yield production. Combined with recently developed chip-based light sources, these techniques offer the possibility of biosensing and super-resolution imaging based on integrated circuits.

Abstract:Owing to the unique optophysical properties of nanomaterials and their self-calibration characteristics, nanomaterial-based (e.g., polymer dots (Pdots) quantum dots (QDs), silicon nanorods (SiNRs), and gold nanoparticle (AuNPs), etc.) ratiometric fluorescent sensors play an essential role in numerous biosensing and cell imaging applications. The dual-emission ratiometric fluorescence technique has the function of effective internal referencing, thereby avoiding the influence of various analyte-independent confounding factors. The sensitivity and precision of the detection can therefore be greatly improved. In this review, the recent progress in nanomaterial-based dual-emission ratiometric fluorescent biosensors is systematically summarized. First, we introduce two general design approaches for dual-emission ratiometric fluorescent sensors, involving ratiometric fluorescence with changes of one response signal and two reversible signals. Then, some recent typical examples of nanomaterial-based dual-emission ratiometric fluorescent biosensors are illustrated in detail. Finally, probable challenges and future outlooks for dual-emission ratiometric fluorescent nanosensors for biosensing and cell imaging are rationally discussed.

Abstract:Comprehensive Summary Cancers and chronic diseases have always been global health problems. The occurrence and development of such diseases are closely related to the abnormalities of proteins, nucleic acids, ions or small molecules in the body. Nowadays, nanopores/nanochannels have emerged as a powerful platform for detecting these biomolecules based on the electrical signal variation caused by biomolecules passing. However, detection relied on the electrical signal easily suffered from the clogging defects, low throughput, and strong background signals. Fortunately, the emergence of designing nanopores/nanochannels based on electrical and optical dual signal response has brought innovative impetus to biological detection, which can also identify the chemical compositions and conformations of the biomolecules. In this review, we summarize the reasonable preparation of nanopores/nanochannels with electrical and optical dual signal response and its application in biological detection. According to different biomolecules, we divide the targets into four types, including nucleic acids, small molecules, ions and proteins. In each section, the design of representative examples and the principle of dual signal generation are introduced and discussed. Abstract:The complexity of biological samples determines that the detection of a single biomolecule is unable to satisfy actual needs. Moreover, the "false positives" results caused by a single biomolecule detections easily leads to erroneous clinical diagnosis and treatment. Thus, it is important for the homogenous quantification of multiple biomolecules in not only basic research but also practical application. As a consequent, a large number of literatures have been exploited to monitor multiple biomolecules in homogenous solution, enabling facilitating the development of the disease diagnosis, treatment as well as drug discovery. One-dimensional nanomaterials and two-dimensional nanomaterials have special physical and chemical properties, such as good electrochemical properties, stable structure, large specific surface area, and biocompatibility, which are widely used in electrochemical and fluorescent detection of biomolecules. This tutorial review highlights the recent development for the detection of multiple biomolecules by using nanomaterials including one-dimensional materials（1DMs） as well as twodimensional materials（2DMs）.

Abstract:Biosensors have achieved significant progress in the past decades and are now widely used for diagnostic and therapeutic purposes. A biosensor is defined as a device for analyte detection or physiological signal measurements, which usually combines selective biological elements and transducers that convert signals resulting from the interaction between the analyte and biological elements to other easily measured outputs (eg, electrical, optical, and thermal signals)[1]. A broad range of biomaterials have been involved in building various biosensors. The major components usually include a probe that selectively responds to a certain biological element, such as deoxyribonucleic acid (DNA), aptamer, antibody, enzyme, cell, and so on, and a transducer that transforms the physiochemical information into optical, electrical, magnetic, electrochemical, and thermal signals. Due to the advantages of high surface area, miniaturized size, and associated new physical properties, nanomaterials have drawn a great deal of attention as components for biosensors. Abstract:Abstract Two-dimensional (2D) materials have attracted signifcant attention due to their unique chemical and physical characteristics. The specific structures and large surface area of 2D materials lead to great potentials in sensing applications with the advantages of high sensitivity, high efficiency, and environmental friendliness. As a result, a great variety of devices have been developed based on 2D materials and utilized as electronic, chemical, biological, and even multifunctional sensors. Importantly, the high performance of these sensors is largely attributed to the synthetic strategies of high-quality 2D materials, where the exfoliation in the liquid phase is one of the most efficient methods. In this review, we firstly summarize the recent progress on the solution methods for the synthesis of high-quality graphene as well as non-carbon 2D materials. Then the main focus of this review article is shifted to the transistor-type sensors, especially the biosensors and chemical sensors, on the basis of these solution-processed 2D materials. In addition, the remaining challenges in this research field are discussed, and possible future directions of development are also proposed from the aspects of materials, processing, and devices.

Abstract:The sensitive detection of analytes of different sizes is crucial significance for environmental protection, food safety and medical diagnostics. The confined space of nanochannels provides a location closest to the molecular reaction behaviors in real systems, thereby opening new opportunities for the precise detection of analytes. However, due to the susceptibility to external interference on the confined space of nanochannels, the high sensitivity nature of the current signals through the nanochannels is more troubling for the detection reliability. Combining highly sensitive optical signals with the sensitive current signals of solid-state nanochannels establishes a nanochannel detection platform based on electro-optical dual signals, potentially offering more sensitive, specific, and accuracy detection of analytes. This review summarizes the last five years of applications of solid-state nanochannels based on electro-optical dual signals in analytes detection. Firstly, the detection principles of solid-state nanochannels and the construction strategies of nanochannel electro-optical sensing platforms are discussed. Subsequently, the review comprehensively outlines the applications involving nanochannels with electrical signals combined with fluorescence signals, electrical signals combined with surface-enhanced Raman spectroscopy signals, and electrical signals combined with other optical signals in analyte detection. Additionally, the perspectives and difficulties of nanochannels are investigated on the basis of electro-optical dual signals.

Abstract:Nanomaterials show great potential for applications in biosensing due to their unique physical, chemical, and biological properties. However, the single-modal signal sensing mechanism greatly limits the development of single-modal nanoprobes and their related sensors. The multi-modal nanoprobes can realize the output of fluorescence, colorimetric, electrochemical, and magnetic signals through composite nanomaterials, which can effectively compensate for the defects of single-modal nanoprobes. Following the multi-modal nanoprobes, the multi-modal biosensors break through the performance limitation of the current single-modal signal and realize multi-modal signal reading. Herein, the current status and classification of multi-modal nanoprobes are provided. Moreover, the multi-modal signal sensing mechanisms and the working principle of multi-modal biosensing platforms are discussed in detail. We also focus on the applications in pharmaceutical detection, food and environmental fields. Finally, we highlight this field's challenges and development prospects to create potential enlightenment.

Abstract:The drive for a simultaneous analysis of multiple targets with excellent accuracy and efficiency, which is often required in both basic biomedical research and clinical applications, demands the development of multiplexed bioassays with desired throughput. With the development of nanotechnologies, innovative multiplex optical bioassays have been achieved. Nanomaterials exhibit unique physical and chemical properties such as easily tunable size, large surface-to-volume ratio, excellent catalysis and the desired signal transduction mechanism, which makes them excellent candidates for the fabrication of novel optical nanoprobes. This mini review summarizes nanomaterial-based optical multiplex sensors from the last 5 years. Specific optical techniques covered in this review are fluorescence, surface-enhanced Raman scattering (SERS), localized surface plasmon resonance (LSPR), chemiluminescence (CL), and the multimodality with fundamentals and examples.

Abstract:With the continuous advancement of science and technology, the application of nanomaterials in the field of biosensors is becoming increasingly widespread. Nanobiosensors are widely used due to their high sensitivity, selectivity, small size, and low cost, finding applications in medical diagnostics, environmental monitoring, biotechnology, and industrial automation, among others. This paper primarily discusses the research progress of biosensors based on nanomaterials and their applications in biological detection and medical diagnosis. It introduces the working principles of nanomaterial-based biosensors, provides practical application cases of nanomaterial biosensors in various fields, such as protein detection, DNA detection and pathogen detection. And this paper also looks forward to the prospects, as well as the advantages and disadvantages, of nanomaterial-based biosensors. With the deepening of nanomaterial research and the continuous innovation of technology, biosensors based on nanomaterials are expected to play an increasingly important role in biological detection, medical diagnosis, and biomedical research. Simultaneously, they also hold the potential for widespread application in clinical diagnosis, biosecurity, and environmental protection.

Abstract:DNA has excellent molecular recognition properties. At the same time, DNA has a programmable structure, high stability, and can be easily modified, making DNA attractive for biosensor design. To convert DNA hybridization or aptamer binding events to physically detectable signals, various nanomaterials have been extensively exploited to take advantage of their optical and surface properties. A popular sensing scheme is through the adsorption of a fluorescently-labeled DNA probe, where detection is achieved by target-induced probe desorption and fluorescence recovery. Another method is to use DNA to protect the colloidal stability of nanomaterials, where subsequent target binding can decrease the protection ability and induce aggregation; this method has mainly been used for gold nanoparticles. This Perspective summarizes some of our work in examining the sensing mechanisms, and we articulate the importance of the understanding of DNA/surface and target/surface interactions for the development of practical DNA-based biosensors.

Abstract:Surface plasmon resonance biosensors are drawing attention due to their real-time, label-free, and rapid characteristic. To detect trace biomarkers (ct-DNA, mi-RNA, PD-L1), plasmonic and metal oxide nanoparticles have been utilized for signal amplification and have shown exciting results. To achieve uniform, reproducible, simple, and sensitive sensor interface construction, two-dimensional materials such as graphene and molybdenum sulfide have opened a research upsurge and show a great possibility in the surface plasmon resonance biosensing field due to their unique large specific surface area, excellent in-plane electron transport effect, and singular optical properties. Here, we provide an overview of the recent research progress on two-dimensional materials in the field of surface plasmon resonance biosensors, including the optoelectronic properties of two-dimensional materials, sensitivity enhancement mechanism, and interface construction. Finally, the challenges and development directions of detection sensitivity enhancement are summarized and prospected. Our work will provide a useful platform to expand the further application of two-dimensional materials in surface plasmon resonance biosensors.

Abstract:The advancement of biosensing has led to increased demands for precise target recognition and highly sensitive detection. Nanomaterials have emerged as indispensable contributors to this development process. Concurrently, the CRISPR/Cas (Clustered Regularly Interspaced Short Palindromic Repeats/CRISPR-associated system) also has surfaced as a novel tool that brings new prospects to the field of biosensing. Its notable benefits include accurate recognition, rapid response, ease of operation, and cost effectiveness. Significantly, the integration of nanomaterials and CRISPR/Cas exhibits immense potential for diverse applications. To enhance comprehension of biosensing research utilizing nanomaterials and CRISPR/Cas, this review comprehensively summarizes and analyzes the distinct functional roles of both components in biosensing approaches. The focus lies on exploring the applications of various nanomaterials (such as metal nanoparticles, metal oxides, carbon nanomaterials, porous framework materials, and other nanomaterials) combined with CRISPR/Cas in the biosensing field. Abstract:As widely used commercial sensors, amperometric biosensors based on electrochemical (EC) reactions exhibit good linearity and reproducibility but suffer from a low dynamic range and poor limit of detection (LoD). In contrast, biosensors utilizing field-effect transistors (FETs) demonstrate a wide detection range and ultralow LoDs but are susceptible to interferences or ionic strength influences and have poor quantitative accuracy due to their nonlinear response mechanism. Here, EC and carbon nanotube (CNT) FET-based biosensors are integrated into one chip via a compatible fabrication process. The dual-mode biosensing chip (DMSC) provides the advantages from both the EC biosensors and FET biosensors; these advantages include high reliability and accuracy for quantitatively detecting glucose, a dynamic detection range spanning 6 orders of magnitude, and a detection limit of 1 fM. The introduction of a time-division multiplexing detection method enables the DMSC to carry out complementary detection of the same target, providing early warning of specific abnormal biomarkers and accurate determination of their concentration in real time. The DMSC approach is generally applicable to the detection of other biomarkers and provides exciting opportunities for more accurate self-monitoring diagnostics in real situations. Electrochemical (EC) and carbon nanotube (CNT) FET-based biosensors are integrated into one chip via a compatible fabrication process. Abstract:Energy transfer-based biosensing is one of the most effective methods for fast and accurate biodetection. By designing appropriate energy transfer pairs as optical probes, a large diversity of analytes, ranging from small molecules to viruses, can be detected with high sensitivity and selectivity. Particularly, the development of optical nanoprobes has greatly improved the capability of energy transfer-based biosensing systems. By controlling the composition and structure of nanomaterials, both their optical characteristics and energy transfer pathways can be rationally manipulated, thereby enabling novel sensing processes. In this article, we focus on various nanomaterials as components of energy transfer probes, and review recent advances in biodetection using optical nanoprobes in different biological settings.

Abstract:Biosensors contribute a lot to the reliable and sensitive detection in various fields, especially emerging trends of in-field and real-time detection for point-of-care diagnosis, food safety and environmental monitoring. The signal amplification that improves the analytical performance and the compact integration of various biosensing components with/in miniaturized and portable devices are essential but still challenging. Integrating the merits of bio-active proteins (enzyme, antibody, etc.) and nanomaterials (nanoparticles, nanotubes, nanosheets, nanoflowers etc.) with abundant physicochemical properties, numerous protein-nanomaterial hybrids (PN hybrids) have been designed and applied for biosensing in recent years. PN hybrids can serve as not only sensitive probes for analyte recognition and signal generation/amplification thereby enhancing analytical performance, but also miniaturized and full-functional sensing components that are easily combined with other devices, greatly simplifying the construction and assay procedures. In this review, the state-of-art strategies of PN hybrids for biosensing are summarized from the view of the role of nanomaterial components, i.e. immobilization matrix, catalyst, and label. Recent advances for the emerging in-field detection applications of PN hybrids with the incorporation of portable hand-held readers and miniaturized devices are then surveyed. The features of PN hybrids for the construction of these miniaturized biosensors are focused. The integration and synergy between proteins and nanomaterials for biosensing is emphasized and discussed.
