Abstract:Panax ginseng and Panax quinquefolius, which are commonly called Chinese ginseng and American ginseng respectively, have different medicinal properties and market values; however, these samples can be difficult to differentiate from one another based on physical appearances of the samples especially when they are in powdery or granular forms. A molecular technique is thus needed to overcome this difficulty; this technique is based on the nucleic acid test (NAT) conducted on the microfluidic chip surface. Three single nucleotide polymorphism (SNP) sites (i.e. N1, N2, N3) on the Panax genome that differ between P. ginseng (G) and P. quinquefolius (Q) have been selected to design probes for the NAT. Primers were designed to amplify the antisense strands by asymmetric PCR. We have developed three different NAT methodologies involving surface immobilization and subsequent (stop flow or dynamic) hybridization of probes (i.e. N1G, N1Q, N2G, N2Q, N3Q) to the antisense strands. These NAT methods consist of two steps, namely immobilization and hybridization, and each method is distinguished by what is immobilized on the microfluidic chip surface in the first step (i.e. probe, target or capture strand). These three NATs developed are called probe-target method 1, target-probe method 2 and three-strand complex method 3. Out of the three methods, it was found that the capture strand-target-probe method 3 provided the best differentiation of the ginseng species, in which a 3' NH2 capture strand is first immobilized and the antisense PCR strand is then bound, while N2G and N3Q probes are used for detection of P. ginseng (G) and P. quinquefolius (Q) respectively.Graphical

Abstract:Abstract When testing botanical ingredients of herbal medicines and dietary supplements, the complexity of botanical matrixes often requires the use of orthogonal methods to establish identification procedures suitable for quality control purposes. Genomic-based botanical identification methods are evolving and emerging as useful quality control tools to complement traditional morphological and chemical identification methods. Species-specific polymerase chain reaction methods are being evaluated for botanical quality control and as a cost-effective approach to identify and discriminate between closely related botanical species. This paper describes orthogonal identification of Panax ginseng, P. quinquefolius, and P. notoginseng materials in commerce as an example of the development and validation of a set of species-specific polymerase chain reaction methods to establish botanical identity in ginseng roots. This work also explored the possibility of extending the application of species-specific polymerase chain reaction methods to provide species identity information for processed materials, such as steamed roots and hydroalcoholic extracts, and showed success with this approach. Finally, the paper provides recommendations for an out-of-specification investigation of samples that may pass some of the orthogonal tests and fail others.

Application of a modified tetra-primer ARMS–PCR assay for rapid Panax species identity authentication in ginseng products

Abstract:Panax ginseng products can be adulterated with materials from other Panax species. The purpose of this study is to provide a rapid P. ginseng authentication method for simultaneous identification of P. ginseng and detection of adulteration in ginseng products at different processing stages. First, a tetra-primer ARMS–PCR assay was designed based on a single-nucleotide polymorphism (SNP) within the trnL-trnF region and was tested at 28 PCR cycles with DNA extracted from Botanical Reference Materials (BRMs). Next, 5′ end random nucleotide and 3′ terminus phosphorothioates linkage modifications were incorporated into the inner primers to improve sensitivity and specificity at 40 PCR cycles. Finally, the modified assay was validated using characterized market ginseng materials and the detection limit was determined. The modified tetra-primer ARMS–PCR assay can achieve the desired sensitivity and specificity using one set of reaction conditions in ginseng materials at different stages. In validation, it was able to correctly identify target species P. ginseng and differentiate it from closely related species. This study suggests that the modified tetra-primer ARMS–PCR assay can be used for the rapid, species identity authentication of P. ginseng material in ginseng products. This assay can be used to complement chemical analytical methods in quality control, so both species identity and processing attributes of ginseng products can be efficiently addressed.

Abstract:Forty minutes of abdominal aorta ligature (partial ischemia in the lumbosacral segments of the spinal cord) did not change significantly levels of norepinephrine (NE) and 5-hydroxytryptamine (5-HT) in the spinal cord of the dog in comparison with sham-operated controls. After a 40-minute interval of recirculation NE levels dropped nonsignificantly below levels of the control group and 5-HT significantly in lumbar segments. It thus appears that even though in the stage of recirculation in the spinal cord energy metabolism is restored, the functional deficit of monoamines may persist.

Abstract:Background: American ginseng (Panax quinquefolius L.) and Asian ginseng (Panax ginseng Meyer) products, such as slices, have a similar appearance, but they have significantly different prices, leading to widespread adulteration in the commercial market. Their aroma characteristics are attracting increasing attention and are supposed to be effective and nondestructive markers to determine adulteration. Methods: The aroma characteristics of American and Asian ginseng were investigated using gas chromatography-mass spectrometry(GC-MS) and an electronic nose (E-nose). Their volatile organic compounds were separated, classified, compared, and analyzed with different pattern recognition. Results: The E-nose showed a good performance in grouping with a principle component analysis explaining 94.45% of variance. A total of 69 aroma components were identified by GC-MS, with 35.6% common components and 64.6% special ingredients between the two ginsengs. It was observed that the components and the number of terpenes and alcohols were markedly different, indicating possible reasons for their difference. The results of pattern recognition confirmed that the E-nose processing result is similar to that of GC-MS. The interrelation between aroma constituents and sensors indicated that special sensors were highly related to some terpenes and alcohols. Accordingly, the contents of selected constituents were accurately predicted by corresponding sensors with most R-2 reaching 90%. Conclusion: Combined with advanced chemometrics, the E-nose is capable of discriminating between American and Asian ginseng in both qualitative and quantitative angles, presenting an accurate, rapid, and nondestructive reference approach. Abstract:The American ginsengs (Radix panacis Quinquefolii) of different geographical regions from Beijing (China), Toronto (Canada), Vancouver (Canada), Wisconsin (USA), and the wild American ginseng can be discriminated and identified by using Fourier transform-infrared spectroscopy (FT-IR) and two dimensional correlation infrared spectroscopy (2D-IR). The FT-IR spectra of the five kinds of American ginseng have small differences, and their secondary derivative spectra have obvious finger-print feature, especially in the range of 1750–400cm−1. Using the difference of the position and intensity of the special (peculiar) bands, which include both the peak at 1317cm−1 assigned to oxalate group, and other peaks near 1050, 1020, and 866cm−1 assigned to the starch, we can identify and discriminate them from each others. Furthermore, using the pattern of 2D-IR correlation spectra under thermal perturbation, we can very directly identify and discriminate them. As a result, the macroscopic IR finger-print method, which combines three-steps: general IR spectra, secondary derivative spectra as well as 2D-IR correlation spectra, can discriminate different American ginsengs effectively, fast, and nondestructively.

Abstract:A total of 44 root samples of American ginseng (Panax quinquefolius) originating from five different geographical areas (America = 6, Canada = 10, Beijing = 8, Jilin = 8, and Shandong = 12) were collected from specialty wholesalers in China. The C/N/H/O stable isotope ratios were determined using an elemental analyzer coupled with a stable isotope ratio mass spectrometer. Our results revealed that a single variable was unable to clearly discriminate between the various geographical origins of American ginseng, whereas the combination of δ18O and δ15N and the combination of δ13C, δ15N, and δ18O provided useful information for the identification of American ginseng from the five regions. Moreover, the application of different multivariate statistical approaches, including the one-way analysis of variance and linear discriminant analysis (LDA), to further explore the ability of C/N/H/O stable isotopes for determining the geographical origins of American ginseng identified a significant difference (P < 0.05) in the stable isotope ratios from different regions, and high recognition (90.9%) and satisfactory predictive ability (86.4%) were achieved with the LDA. The present findings suggest that the C/N/H/O stable isotopes can be potentially applied in the authentication of different geographical origins of American ginseng.

Abstract:AIM:To develop a method of identifying the existing of Panax notoginseng in products of traditional Chinese medicine compound Danshen.METHODS:Total ion chromatograms (TIC) of Panax notoginseng, P. ginseng, and P. quinquefolius were obtained by means of LC/MS. Extracted ion chromatograms (EIC) posses m/z of 770, 800, 932, 946 and 1,108 of above-mentioned three herbs was compared between species. EIC 800 and 946 were selected as differentiation marks to distinguish P. notoginseng from the other two species. The EIC 800 and 946 of P. notoginseng were also compared to the EICs obtained by the same method from Chinese patent medicines of compound Danshen pellet, compound Danshen tablet, and compound Danshen injection. EIC 800 and 946 of P. notoginseng and its products possess similar peaks, relative retention time, and relative integral areas. Main chemical constitutes of P. notoginseng were also identified by using LC/MS/MS.RESULTS:EIC 800 and 946 were obviously different between P. notoginseng, P. ginseng, and P. quinquefolius. Patent medicines of compound Danshen pellet, compound Danshen tablet, which consist of extractions from P. notoginseng, possess the characteristic EICs. The selected EICs were stable and reproductive.CONCLUSION:EIC 800 and 946, which correspond to ginsenoside Rg1, Re, and their isomers, can be used as identifying mark of P. notoginseng to differentiate it from other herbs, and also can be used to tell apart P. notoginseng from other herb extractions in Chinese patent medicines of compound Danshen.

Abstract:Black ginseng (BG) is processed ginseng traditionally made in Korea via the steaming and drying of ginseng root through three or more cycles, leading to changes in its appearance due to the Maillard reaction on its surface, resulting in a dark coloration. In this study, we explored markers for differentiating processed ginseng by analyzing the chemical characteristics of BG. We elucidated a new method for the structural identification of ginsenoside metabolites and described the features of processed ginseng using UPLC-QTOF-MS in the positive ion mode. We confirmed that maltose, glucose, and fructose, along with L-arginine, L-histidine, and L-lysine, were the key compounds responsible for the changes in the external quality of BG. These compounds can serve as important metabolic markers for distinguishing BG from conventionally processed ginseng. The major characteristics of white ginseng, red ginseng, and BG can be distinguished based on their high-polarity and low-polarity ginsenosides, and a precise method for the structural elucidation of ginsenosides in the positive ion mode is presented.

Abstract:Ginseng, Panax ginseng C. A. Meyer, is an industrial crop in China and Korea. The functional components in ginseng roots and rhizomes are characteristic ginsenosides. This work developed a new high-performance liquid chromatography coupled with electrospray ionization ion trap time-of-flight multistage mass spectrometry (LC-ESI-IT-TOF-MSn) method to identify the triterpenoids. Sixty compounds (1-60) including 58 triterpenoids were identified from the ginseng cultivated in China. Substances 1, 2, 7, 15-20, 35, 39, 45-47, 49, 55-57, 59, and 60 were identified for the first time. To evaluate the quality of ginseng cultivated in Northeast China, this paper developed a practical liquid chromatography-diode array detection (LC-DAD) method to simultaneously quantify 14 interesting ginsenosides in ginseng collected from 66 different producing areas for the first time. The results showed the quality of ginseng roots and rhizomes from different sources was different due to growing environment, cultivation technology, and so on. The developed LC-ESI-IT-TOF-MSn method can be used to identify many more ginsenosides and the LC-DAD method can be used not only to assess the quality of ginseng, but also to optimize the cultivation conditions for the production of ginsenosides.

Abstract:To distinguish wild Panax ginseng from cultivated Panax ginseng, high quality DNA were extracted from ginseng material of 5 mg or so. Based on improved SDS method, genomic fingerprints of ten individuals of wild ginseng and five of cultivated ginseng were obtained using direct amplification of length polymorphism (DALP)analysis. The DALP fingerprints showed that the genetic variation in wild ginseng group was much higher than that in cultivated ginseng group. The characteristic DALP fragments, which could be used to distinguish wild ginseng from cultivated ginseng were found. DALP has proved to be an effective technique to study DNA difference between wild ginseng and cultivated ginseng.
Differentiation of Panax quinquefolius grown in the USA and China using LC/MS-based chromatographic fingerprinting and chemometric approaches

Abstract:American ginseng (Panax quinquefolius) is one of the most commonly used herbal medicines in the world. Discriminating between P. quinquefolius grown in different countries is difficult using traditional quantitation methods. In this study, a liquid chromatographic mass spectrometry fingerprint combined with chemometric analysis was established to discriminate between American ginseng grown in the USA and China. Fifteen American ginseng samples grown in Wisconsin and 25 samples grown in China were used. The chromatographic fingerprints, representing the chemical compositions of the samples, made it possible to distinguish samples from the two locations. In addition, it was found that some ginsenosides varied widely from P. quinquefolius cultivated in these two countries. P. quinquefolius grown in the USA is higher in ginsenoside R(c), ginsenoside R(d), quinquenoside III/pseudo-ginsenoside RC₁, malonyl ginsenoside R(b1), and ginsenoside R(b2), but lower in ginsenoside R(b1) compared with P. quinquefolius grown in China. These ginsenosides may be responsible for the class separation seen using fingerprinting and chemometric approaches.

Rapid Discrimination of Panax quinquefolium and Panax ginseng Using the Proofman-Duplex-LMTIA Technique

Abstract:This study aims to establish a rapid identification method based on the Proofman-LMTIA technique for distinguishing between Panax quinquefolium and Panax ginseng. By targeting specific 18S rDNA sequences, suitable primers and Proofman probes labeled FAM or JOE were designed for LMTIA. Initially, single-species-primer Proofman-LMTIA assays were performed separately for each ginseng type to optimize reaction temperature, assess sensitivity and specificity, and determine the detection limit. Subsequently, both sets of primers and their corresponding probes were combined in the same reaction system to further optimize reaction conditions, evaluate sensitivity, and assess stability. Finally, the developed Proofman-duplex-LMTIA technique was employed to detect P. quinquefolium and P. ginseng slices available in the market. Single-plex Proofman-LMTIA assays revealed that the optimal reaction temperature for both P. quinquefolium and P. ginseng was 62 °C. The sensitivity was as low as 1 pg/μL, with a detection limit of 0.1%, and both showed excellent specificity. The optimal temperature for Proofman-duplex-LMTIA assays was 58 °C. This method could simultaneously identify P. quinquefolium and P. ginseng. Testing 6 samples of P. ginseng and 11 samples of P. quinquefolium from the market resulted in a 100% positive rate for all samples. This study successfully established a rapid, simple, sensitive, and specific Proofman-duplex-LMTIA identification method for P. quinquefolium and P. ginseng. It provides an effective means for quality control of P. quinquefolium, P. ginseng, and related products.

Abstract:Yunpoong is an important Korean ginseng (Panax ginseng C. A. Meyer) cultivar, but no molecular marker has been available to identify Yunpoong from other cultivars. In this study, we developed a single nucleotide polymorphism (SNP) marker for Yunpoong based on analysis of expressed sequence tags (ESTs) in an exon region of the glyceraldehyde 3-phosphate dehydrogenase (GAPDH) gene. This SNP marker had high specificity to authenticate Yunpoong in twelve different main ginseng cultivars. For application of the molecular marker, a rapid identification method was established based on the NaOH-Tris method and real-time polymerase chain reaction (PCR) in order to ensure more efficiency in the cultivar selection. The biggest feature of the NaOH-Tris method was that it made the extraction of DNA very simple and rapid in young leaf tissues. We only spent 1 min to extract DNA and directly used it to do PCR. In this report, the conventional DNA extraction method was used to develop molecular marker process, and the NaOH-Tris method was applied in screening large numbers of cultivars. Moreover, the greatest advantage of the real-time PCR compared with traditional PCR, is time saving and high efficiency. Thus, this strategy provides a rapid and reliable method for the specific identification of Yunpoong in a large number of samples.
Simultaneous Quantitative Assays of 15 Ginsenosides from 119 Batches of Ginseng Samples Representing 12 Traditional Chinese Medicines by Ultra-High Performance Liquid Chromatography Coupled with Charged Aerosol Detector

Abstract:Despite the extensive consumption of ginseng, precise quality control of different ginseng products is highly challenging due to the containing of ginsenosides in common for different Panax species or different parts (e.g. root, leaf, and flower) of a same species. Herein we performed a comparative investigation of diverse ginseng products by simultaneously assaying 15 saponins (notoginsenoside R1, ginsenosides Rg1, -Re, -Rf, -Ra2, -Rb1, -Rc, -Ro, -Rb2, -Rb3, -Rd, 20(R)-ginsenoside Rg3, 24(R)-pseudoginsenoside F11, chikusetsusaponins IV, and -IVa) using an ultra-high-performance liquid chromatography/charged aerosol detector (UHPLC-CAD) approach. Twelve Panax-derived ginseng products (involving P. ginseng root, P. quinquefolius root, P. notoginseng root, Red ginseng, P. ginseng leaf, P. quinquefolius leaf, P. notoginseng leaf, P. ginseng flower, P. quinquefolius flower, P. notoginseng flower, P. japonicus root, and P. japonicus var. major root) were considered. Benefiting from the condition optimization, the baseline resolution of 15 ginsenosides was achieved on a CORTECS UPLC Shield RP18 column. This method was validated as specific, precise (0.81–1.94% intra-day variation; 0.86–2.35% inter-day variation), and accurate (recovery: 90.73–107.5%), with good linearity (R2 > 0.999), high sensitivity (limit of detection: 0.02–0.21 μg; limit of quantitation: 0.04–0.42 μg) and sample stability (1.49–4.74%). Its application to 119 batches of ginseng samples unveiled vital information enabling the authentication of these different ginseng products. Detection of ginsenosides by CAD exhibited superiority over UV in sensitivity and the ability to monitor chromophore-free structures. Large-scale comparative studies by quantifying multiple markers provide methodological reference to the precise quality control of herbal medicine.
A method for the analysis of ginsenosides, malonyl ginsenosides, and hydrolyzed ginsenosides using high-performance liquid chromatography with ultraviolet and positive mode electrospray ionization mass spectrometric detection

Abstract:A high-performance liquid chromatographic separation coupled to diode array absorbance and positive mode electrospray mass spectrometric detection has been developed for the analysis of ginsenosides, malonyl ginsenosides, and hydrolyzed ginsenosides in extracts of Asian ginseng (Panax ginseng) and American ginseng (P. quinquefolius). The method is capable of separating, identifying, and quantifying the predominant ginsenosides found in heated alcoholic extracts of Asian and American ginseng roots routinely sold as nutraceuticals. It also separates and identifies the malonyl ginsenosides often found in cold alcoholic extracts of ginseng root and has the potential to quantify these compounds if pure standards are available. Furthermore, it can separate and identify ginsenoside hydrolysis products such as those readily produced in situations mimicking gastric situations, including those used for dissolution studies (i.e., 0.1 N HCl, 37 degrees C).
Enhanced Profiling and Quantification of Ginsenosides from Mountain-Cultivated Ginseng and Comparison with Garden-Cultivated Ginseng

Abstract:Panax ginseng can be generally divided into mountain-cultivated ginseng (MCG) and garden-cultivated ginseng (GCG). The market price of MCG is significantly higher than that of GCG. However, the chemical compositions of MCG and the differences from GCG remained unclear. In this study, an integrated strategy combing an offline two-dimensional liquid chromatography separation, LTQ-orbitrap dual mode acquisition, and Q-trap full quantification/quasi-quantification was proposed to explore and compare the chemical compositions of MCG. Consequently, 559 ginsenosides were characterized, among which 437 ginsenosides were in-depth characterized with α-chain and β-chain annotated. Subsequently, enhanced quantification of 213 ginsenosides was conducted in 57 batches of MCG and GCG. Ginsenosides were found more abundant in MCG than GCG. In addition, 25-year-old MCG could be distinctly differentiated from 15/20-year-old MCG. This strategy facilitated the enhanced profiling and comparison of ginsenosides, improved the quality control tactics of MCG and provided a reference approach for other ginseng related products.

Abstract:Ginseng has been used for prevention and treatment of disease for thousands of years in China and many other Asian countries. Phytochemical studies have indicated that ginsenosides, polysaccharides, alkaloids, and phenolic acids are the active constituents of ginseng. Main and branch roots of ginseng exhibit distinct bioactive behavior. Furthermore, the bioactive behavior of ginseng depends on its age. Traditional analysis is complex preparation and provides inadequate of chemical information of the original distribution of analytes. Therefore, in this study, ultraperformance liquid chromatography quadrupole/time of flight-mass spectrometry (UPLC-QTOF MS) and desorption electrospray ionization mass spectrometry imaging (DESI-MSI) combined with orthogonal partial least squares discriminant analysis were used to discriminate ginseng in different age and parts of ginseng, and profiled distribution of selected markers. The results indicated that UPLC-QTOF-MS and DESI-MSI could be used to determine the parts and age of ginseng. Fifteen variables including five of protopanaxatriol (PPT), four of protopanaxadiol (PPD), and six of other types were assumed as markers for different parts of ginseng. Moreover, four variables of PPT, four of PPD, and ten of other types were used to determine the age of ginseng samples. An analysis of localization of markers indicated that malonyl ginsenoside, including malonyl-ginsenoside Rb1, Rb2, Rc, and Rd was mainly distributed in the corks. Neutral ginsenoside Rg1, yesanchinoisde D, and chikusetsusaponin Iva were mainly distributed in the cork and phloem. Non-ginsenoside castanoside H, 20(S)-protopanaxatriol, unknown 2, saponin III and cistanoside C were distributed in all tissues. Ethyloleate, unknown 1 and monolinolein were distributed in the cork.

Abstract:Aim To establish a method for differentiaing commercial samples of Panax species including notoginseng,cultivated ginseng(Chinese ginseng and Korean ginseng),wild ginseng,red ginseng,three types of American ginsengs,and one American ginseng preparation with their HPLC fingerprints for assnrning the quality of different commercial samples of Panax species.Methods HPLC-UV method was used to establish their fingerprints,Zorbax Extend C18(250 mm×4.6 mm,5μm)was used as the analytical column,and acetonitrile/KH2PO4 aqueous solution Was used as the mobile phase with gradient elution.Resuits The fingerprints of different commercial samples of Panax species varied in their holistic chromatograms and some specific constituents.Conclusion This method is reliable, reproducible and simple.It could be applied in the routine authentication of different commercial samples of Panax species
