Abstract:Highlights • Global Prevalence of HNSCC: HNSCC is the world's seventh most prevalent cancer, with roughly 660,000 new cases annually, representing 4.5% of all cancer diagnoses globally. • Role of HPV in HNSCC: HPV infection, notably HPV-16 and HPV-18, plays a crucial role in certain HNSCC cases, particularly in oropharyngeal cancer. • Impact of Oropharyngeal Cancer: Oropharyngeal cancer, a subtype of HNSCC, imposes a substantial global burden, with approximately 98,400 new cases and 48,100 deaths reported in 2020. • Distinct Molecular Profiles in HPV-Positive OPSCC: HPV-positive OPSCC exhibits unique molecular profiles, including p16 overexpression, and shows increased sensitivity to chemotherapy and radiation therapy compared to HPV-negative cases. • Future Directions in Research and Treatment: Future research aims to understand HPV-induced carcinogenesis, identify new therapeutic targets, and develop personalized treatments for HPV-positive tumors in HNSCC, promising improved outcomes.
