Abstract:OBJECTIVE:To observe the effect of Puerarin on L-type calcium channel in isolated rat ventricular myocytes.METHOD:The cardiac ventricular myocytes were isolated enzymatically by Langendorff perfusion techniques at constant flow rate. Whole-cell recording of patch-clamp techniques was used to observe the current of L-type calcium channel.RESULT:Puerarin 2.4 mmol x L(-1) could inhibit the current of L-type calcium channel of rat ventricular myocytes and this inhibition was time-dependent. Purerarin elevated the current-voltage (I-V) curve of calcium current.CONCLUSION:Puerarin can inhibit L-type calcium current of rat ventricular myocytes. Which implies that puerarin takes part in anti-myocardial ischemia and anti-arrhythmics partly due to the inhibition of L-type calcium channel.

Abstract:Aim: To explore the effects of benactyzine (BEN) on the action potential and contractile force in guinea pig papillary muscles. Methods: Conventional microelectrode technique was used to record the fast action potentials (FAP) and slow action potentials (SAP) of guinea pig papillary muscles. Results: Benactyzine 5, 10, 50 μmol·L -1 suppressed the maximal upstroke velocity ( v max ) of FAP and contractile force (Fc) concentration dependently while prolonged the action potential duration at 50%, 90% repolarization (APD 50 , APD 90 ) and effective refractory period (ERP) of FAP. The suppression on the v max was frequency dependent. Benactyzine 5, 10,50 μmol·L -1 lengthened the APD50, APD90 of SAP induced by isoprenaline or histamine when perfused with KCl 22 mmol·L -1 Tyrode's solution. The v max of the SAP was not decreased by benactyzine 5, 10 μmol·L -1 but by 50 μmol·L -1 . The effects on the SAP were antagonized by elevation of the extracellular calcium from 2.0 to 5.6 mmol·L -1 . The effects of benactyzine on SAP elicited by tetrodotoxin resembled that by isoprenaline or histamine except the more pronounced suppression on v max and action potential amplitude (APA). The persistent rapid spontaneous activity and triggered tachyarrhythmia induced by ouabain were also abolished immediately by benactyzine 5 μmol·L -1 . Conclusion: Benactyzine can inhibit Na +, K +, Ca 2+ transmembrane movement and intracellular Ca 2+ mobilization in the myocardium, and this may be the electrophysiological basis of its effects against experimental arrhythmias.

Abstract:Both patch clamp and laser scanning confocal microscopy (LSCM) was applied to appraise the Anisodamine's cardioprotective effects quantitatively and its mechanism were studied. MTT measurement was observed cell viability and Fluo-3/AM was utilized for real-time free calcium with LSCM; ICa,L exposed to Anisodamine was measured by whole-cell patch clamp recording technique. Our study observed that KCL-triggered calcium elevation could be decreased by Anisodamine at dose of 0.04 mmol/L and 0.08 mmol/L with the decreased value of 46.6%, 54.3% correspondingly. Further study of Anisodamine at 0.08 mmol/L showed a marked inhibitory modulation of L-type Ca2+ current density in a time-dependent manner with decreased ratio of (34.8 plusmn 7.9) % (n = 6, P < 0.01), from the value of -4.474 pA/pF to -2.882 pA/pF and accelerated Vil/2 of current inactivation curve from -14.7 mV to -28.4 mV and delayed Vil/2 of current activation curve from -15.6 mV to -9.51 mV. Our result suggests that Anisodamine conferred the cardioprotection by decreasing calcium elevation quantitatively; the likely mechanism was suggested to be responsible for inhibition of L-calcium channel

Abstract:It has been shown on auricle fibres of the frog that neurotoxin apamin in extremely low concentrations (10(-10)-10(-9) M) suppresses the current entering in the cell through calcium channels of the membrane, but it increases tonic contraction related to Na/Ca exchange. No significant effects on fast sodium and potassium channels were observed. The results show that the mechanism of apamin effect on the heart is to a large extent conditioned by specific influence on Ca transport through the cell membranes.
Anisodamine inhibits cardiac contraction and intracellular Ca2+ transients in isolated adult rat ventricular myocytes

Abstract:Increased cardiac workload often leads to serious complications during cardiac surgery such as pericardiopulmonary bypass. Various agents have been applied to lower peripheral resistance and cardiac workload, one of which, anisodamine, is widely used in Asia. However, the direct action of anisodamine on cardiac contractile property is essentially unknown. This study was designed to examine the influence of anisodamine on ventricular contractile function at the single cardiac myocyte level. Ventricular myocytes from adult rat hearts were stimulated to contract at 0.5 Hz, and mechanical and intracellular Ca2+ properties were evaluated using an IonOptix Myocam system. Contractile properties analyzed included peak shortening (PS), time-to-PS (TPS), time-to-90% relengthening (TR90), maximal velocity of shortening/relengthening (±dL/dt), intracellular Ca2+ fluorescence intensity change (ΔFFI) and decay (τ). Anisodamine exhibited a concentration-dependent (10−12–10−6 M) inhibition in PS and ΔFFI, with maximal inhibitions of 44.7% and 47.2%, respectively. Anisodamine inhibited ±dL/dt, lowered resting FFI but elicited no effect on TPS/TR90 and τ. Pretreatment with the nitric oxide synthase (NOS) inhibitor Nω-nitro-l-arginine methyl ester (l-NAME, 100 μM) abolished the inhibitory effect of anisodamine in cell shortening. In addition, anisodamine prevented cholinoceptor agonist carbachol-induced positive cardiac contractile response. This study demonstrated a direct cardiac depressive action of anisodamine at the myocyte level, which may be related to, at least in part, NO production and cholinoceptor antagonism.
Effects of Pethidine on Cardiac Electrophysiological Properties

Abstract:OBJECTIVE:To investigate the effects of pethidine on electrophysiological properties of the isolated ventricular myocytes and the underlying mechanism.METHODS:Langendorff was applied to perfuse rat heart model and whole-cell current clamp and voltage clamp techniques were used.RESULTS:Pethidine decreased heart rate (HR) in a concentration dependent manner and caused severe atrioventricular block (AVB) at >or=250 micromol/L. Pethidine reduced action potential amplitude and maximal rate of depolarization, prolonged action potential duration. Pethidine at 100 micromol/L decreased sodium currents (I(Na)), transient outward potassium currents (I(to)), delayed rectifier potassium currents (I(k)) and L-type calcium currents (I(Ca.L)) to (60.7+/-6.9)%, (55.4+/-5.6)%, (65.1+/-8.0)% and (67.4+/-10.1)% of control levels,respectively. These effects could be recovered by washout. Naloxone, an opioid receptor antagonist, could not abolish the effects of pethidine on ionic currents.CONCLUSION:Pethidine decreased HR and induced AVB, which may be related to the inhibition of I(Na), I(to), I(k) and I(Ca-L) of heart. The depression of cardiac currents is not mediated by opioid receptor.
Fast deterministic processor allocation

Abstract:Interval allocation has been suggested as a possible formalization for the PRAM of the (vaguely defined) processor allocation problem, which is of fundamental importance in parallel computing. The interval allocation problem is, given n nonnegative integers x1, . . ., xn, to allocate n nonoverlapping subarrays of sizes x1, . . ., xn from within a base array of O(?nj=1xj) cells. We show that interval allocation problems of size n can be solved in O((log log n)3) time with optimal speedup on a deterministic CRCW PRAM. In addition to a general solution to the processor allocation problem, this implies an improved deterministic algorithm for the problem of approximate summation. For both interval allocation and approximate summation, the fastest previous deterministic algorithms have running times of ?(log n/log log n). We describe an application to the problem of computing the connected components of an undirected graph. Finally we show that there is a nonuniform deterministic algorithm that solves interval allocation problems of size n in O(log log n) time with optimal speedup.
Aminophylline at clinically relevant concentrations affects inward rectifier potassium current in a dual way

Abstract:Bronchodilator aminophylline may induce atrial or less often ventricular arrhythmias. The mechanism of this proarrhythmic side effect has not been fully explained. Modifications of inward rectifier potassium (Kir) currents including IK1 are known to play an important role in arrhythmogenesis; however, no data on the aminophylline effect on these currents have been published. Hence, we tested the effect of aminophylline (3-100 µM) on IK1 in enzymatically isolated rat ventricular myocytes using the whole-cell patch-clamp technique. A dual steady-state effect of aminophylline was observed; either inhibition or activation was apparent in individual cells during the application of aminophylline at a given concentration. The smaller the magnitude of the control IK1, the more likely the activation of the current by aminophylline and vice versa. The effect was reversible; the relative changes at -50 and -110 mV did not differ. Using IK1 channel population model, the dual effect was explained by the interaction of aminophylline with two different channel populations, the first one being inhibited and the second one being activated. Considering various fractions of these two channel populations in individual cells, varying effects observed in the measured cells could be simulated. We propose that the dual aminophylline effect may be related to the direct and indirect effect of the drug on various Kir2.x subunits forming the homo- and heterotetrameric IK1 channels in a single cell. The observed IK1 changes induced by clinically relevant concentrations of aminophylline might contribute to arrhythmogenesis related to the use of this bronchodilator in clinical medicine.
Contractile Effects of Amphetamine, Pseudoephedrine, Nor-pseudoephedrine (Cathine), and Cathinone on Atrial Preparations of Mice and Humans

Abstract:Amphetamine derivatives are used worldwide legally or illegally and intoxications may be accompanied by cardiac arrhythmias. Here, we tested contractile effects of cumulative applied (±)-amphetamine, pseudoephedrine, nor-pseudoephedrine (cathine), and cathinone in electrically stimulated (1 Hz) human right atrial preparations (HAP) and mouse left atrial preparations and in spontaneously beating mouse right atrial preparations. In mouse atrial preparations, amphetamine increased force of contraction and beating rate in a concentration- and time-dependent manner, starting at 1 μM in left atrial preparations to 157.1% ± 3.0% and right atrial preparations to 146.6% ± 9.8% at 10 μM, respectively [mean ± standard error of the mean (SEM); n = 5; P < 0.05]. Pseudoephedrine, cathine, or cathinone alone were ineffective in mouse atrial preparations but after pre-incubation with the phosphodiesterase IV inhibitor rolipram (0.1 μM), a positive inotropic effect was noted (mean ± SEM: pseudoephedrine: 112.3% ± 9.8%; cathine: 109.0% ± 4.3%; cathinone: 138.3% ± 21.2%). The effects of all drugs were greatly attenuated by 10 μM cocaine or 10 μM propranolol treatments. However, In HAP, not only amphetamine (to a mean ± SEM of 208% ± 32%) but also pseudoephedrine (to a mean ± SEM of 287% ± 60%), cathine (to a mean ± SEM of 234% ± 52%), and cathinone (to a mean ± SEM of 217% ± 65%) increased force of contraction without the need of phosphodiesterase inhibition. The contractile effects in HAP were attenuated by 10 μM cocaine and antagonized by 10 μM propranolol. We conclude that amphetamine, pseudoephedrine, cathine, and cathinone act probably via release of noradrenaline from cardiac stores as indirect sympathomimetic agents in mouse and more pronounced in human atrial preparations.

Abstract:A potent carcinogen, 4-hydroxyaminoquinoline 1-oxide, was found to produce a stable free radical oxidatively. The structure of the free radical produced in dioxane was studied by analyizng the hyperfine structure of isotope-substituted 4-hydroxyaminoquinoline 1-oxides, and by comparing the values of the spin densities obtained from the coupling constants with those calculated by the unrestricted SCF MO method. It was concluded that the free radical has the same molecular skeleton as the parent compound, but that it loses a hydrogen atom on the nitrogen atom of the NHOH group.
Effects of cyclopiazonic acid on membrane currents, contraction and intracellular calcium transients in frog heart

Abstract:The effects of cyclopiazonic acid, a specific inhibitor of the sarcoplasmic reticulum Ca(2+)-ATPase, on membrane currents and contraction were investigated under voltage clamped conditions on frog atrial trabeculae using the double mannitol-gap technique. In ringer solution, cyclopiazonic acid (10 microM) decreased the phasic contraction at all levels of depolarization without significant change in the time to peak of the contraction and time constant of relaxation. In a low-sodium the amplitude and the current/voltage relationship of L-type and T-type Ca2+ currents were not modified while their inactivation phase was markedly slowed by cyclopiazonic acid. At all levels of depolarization the tonic contraction was increased by cyclopiazonic acid. In the presence of cyclopiazonic acid, no significant variation in diastolic level of intracellular calcium ([Ca2+]i) was found while the maximum amplitude of the Ca2+ transient associated with an action potential was reduced and its time course slowed. All the reversible changes observed could be interpreted by assuming that, in frog atria, sarcoplasmic reticulum was present and cyclopiazonic acid inhibits the sarcoplasmic reticulum Ca(2+)-ATPase. It is proposed that under normal conditions relaxation is due mainly to Na+/Ca2+ exchange. The increase in [Ca2+]i during the contraction phase is due to ICa that provokes a fast release of Ca2+ from the sarcoplasmic reticulum and to an influx of Ca2+ provided by reverse Na+/Ca2+ exchange.
Analysis of the negative inotropic effect of acetylcholine on frog atrial fibres

Abstract:Voltage-clamp experiments have been performed on frog atrial preparations in order to study the mechanism of the inotropic effect of acetylcholine (ACh) at various concentrations. The amplitude of the slow inward current (Is) is reduced even at low ACh concentrations; such low concentrations have little or no effect on potassium permeability. Dose-effect relationships for Is inhibition (Is/Is max) by ACh show a half amplitude dose (K0.5 around 8 X 10(-8) M ACh. The reduction of Is is attributed largely to a decrease of the maximal conductance of the slow channel (gs). Steady-state activation and inactivation parameters are not affected by ACh. Experiments in a Na-free solution (Na replaced by Li ions) or in a Ca-free solution (with EGTA) indicate that the "slow sodium current" is more sensitive to ACh than the "slow Ca current", although these two currents both seem to flow through the slow channel. The decrease of the phasic component of contraction observed in the presence of ACh is very well correlated with the decrease of Is (K0.5 = 8 X 10(-8) M ACh), while the increase of the tonic tension may be related to the outward potassium current induced by high concentrations of ACh. The significant difference between the half amplitude dose (K0.5) observed in the dose effect curves with ACh for Is inhibition (K0.5 = 8 X 10(-8) M) and for ACh-induced extra-current (K0.5 - 10(-6) M) may indicate the presence of two muscarinic receptors.
Effect of Cl- channel blockers on aconitine-induced arrhythmias in rat heart

Abstract:The effects of Cl- channel blockers 5-nitro-2-(3-phenylpropylamino)benzoic acid (NPPB) and niflumic acid (NFA) on aconitine-induced arrhythmias were investigated. Left ventricular pressure and electrocardiogram were monitored in Langendorff-perfused rat hearts. Whole-cell patch-clamp and current-clamp techniques were used to measure sodium current (I(Na)) and action potential (AP), respectively, in single rat cardiac ventricular myocytes. Addition of the Na+ channel agonist aconitine (0.1 microM) to the perfusion solution produced polymorphic ventricular arrhythmias with a latent period of 25.5 +/- 6.3 s. NPPB could reverse aconitine-induced arrhythmias. A similar effect was observed by using NFA. NPPB and NFA reversibly depressed the upstroke of the AP in a dose-dependent manner with IC50 values of approximately 12.3 and approximately 73.1 microM, respectively, without significantly affecting the resting potential of rat ventricular myocytes. Both Cl- channel blockers inhibited I(Na) and induced a leftward shift of the steady-state inactivation of I(Na). In conclusion, the results of this study demonstrate that NPPB as well as NFA can suppress aconitine-induced arrhythmias in rat hearts mainly by inhibiting cardiac I(Na).
Cantharidin and sodium fluoride attenuate the negative inotropic effects of carbachol in the isolated human atrium

Abstract:Carbachol, an agonist at muscarinic receptors, exerts a negative inotropic effect in human atrium. Carbachol can activate protein phosphatases (PP1 or PP2A). We hypothesized that cantharidin or sodium fluoride, inhibitors of PP1 and PP2A, may attenuate a negative inotropic effect of carbachol. During bypass-surgery trabeculae carneae of human atrial preparations (HAP) were obtained. These trabeculae were mounted in organ baths and electrically stimulated (1 Hz). Force of contraction was measured under isometric conditions. For comparison, we studied isolated electrically stimulated left atrial preparations (LA) from mice. Cantharidin (100 μM) and sodium fluoride (3 mM) increased force of contraction in LA ( n = 5–8, p < 0.05) by 113% ± 24.5% and by 100% ± 38.2% and in HAP ( n = 13–15, p < 0.05) by 625% ± 169% and by 196% ± 23.5%, respectively. Carbachol (1 μM) alone exerted a rapid transient maximum negative inotropic effect in LA ( n = 6) and HAP ( n = 14) to 46.9% ± 3.63% and 19.4% ± 3.74%, respectively ( p < 0.05). These negative inotropic effects were smaller in LA ( n = 4–6) and HAP ( n = 9–12) pretreated with 100 μM cantharidin and amounted to 58.0% ± 2.27% and 59.2% ± 6.19% or 3 mM sodium fluoride to 63.7% ± 9.84% and 46.3% ± 5.69%, ( p < 0.05). We suggest that carbachol, at least in part, exerts a negative inotropic effect in the human atrium by stimulating the enzymatic activity of PP1 and/or PP2A.

Electrophysiological Effects of Protopine in Cardiac Myocytes: Inhibition of Multiple Cation Channel Currents

Abstract:Protopine (Pro) from Corydalis tubers has been shown to have multiple actions on cardiovascular system, including anti-arrhythmic, anti-hypertensive and negative inotropic effects. Although it was thought that Pro exerts its actions through blocking Ca(2+) currents, the electrophysiological profile of Pro is unclear. The aim of this study is to elucidate the ionic mechanisms of Pro effects in the heart. In single isolated ventricular myocytes from guinea-pig, extracellular application of Pro markedly and reversibly abbreviates action potential duration, and decreases the rate of upstroke (dV/dt)(max), amplitude and overshoot of action potential in a dose-dependent manner. Additionally, it produces a slight, but significant hyperpolarization of the resting membrane potential. Pro at 25, 50 and 100 microM reduces L-type Ca(2+) current (I(Ca,L)) amplitude to 89.1, 61.9 and 45.8% of control, respectively, and significantly slows the decay kinetics of I(Ca,L) at higher concentration. The steady state inactivation of I(Ca,L) is shifted negatively by 5.9 - 7.0 mV (at 50 - 100 microM Pro), whereas the voltage-dependent activation of I(Ca,L) remains unchanged. In contrast, Pro at 100 microM has no evident effects on T-type Ca(2+) current (I(Ca,T)). In the presence of Pro, both the inward rectifier (I(K1)) and delayed rectifier (I(K)) potassium currents are variably inhibited, depending on Pro concentrations. Sodium current (I(Na)), recorded in low [Na(+)](o) (40 mM) solution, is more potently suppressed by Pro. At 25 microM, Pro significantly attenuated I(Na) at most of the test voltages (-60 approximately +40 mV, with a 53% reduction at -30 mV. Thus, Pro is not a selective Ca(2+) channel antagonist. Rather, it acts as a promiscuous inhibitor of cation channel currents including I(Ca,L), I(K), I(K1) as well as I(Na). These findings may provide some mechanistic explanations for the therapeutic actions of Pro in the heart.

Main funding source(s): PPP allowance, Health holland ZonmW Background 2-Aminoethoxydiphenyl Borate (2-APB) has been shown to prevent atrial fibrillation in animal models, supposedly through its inhibitory action on the intracellular IP3 receptor and subsequent calcium regulation. However, 2-APB may modulate other ion channels or transporters and the cellular mechanisms associated with the prevention of atrial fibrillation are not well established. We aimed at unravelling the cellular effects of 2-APB on atrial and ventricular action potential and currents. Methods Rabbit (New Zealand White) left atrial and ventricular cardiomyocytes were isolated from Langendorff-perfused hearts by enzymatic dissociation. Adult human atrial cardiomyocytes were obtained by enzymatic dissociation from left atrial appendages (LAA) routinely removed from AF patients undergoing minimally invasive surgical pulmonary vein isolation. Action potentials (APs) and K+ currents were recorded at 36.5 °C with the amphotericin-B-perforated or whole cell patch clamp technique. RNA was isolated from left atrial and ventricular tissue and sequenced on an Illumina HiSeq4000 platform. Results In rabbit atrial cardiomyocytes, extracellular application of 5 uM 2-APB had no effect on action potential duration (APD), but extracellular application of 50 uM 2-APB prolonged the APD by 40% and inhibited an outward K+-current. Inversely, extracellular application of 5 uM 2-APB shortened the APD of rabbit left ventricular cardiomyocytes by 20% while extracellular application 50 uM 2-APB did not prolong the APD. Additionally, 2-APB did not modulate K+ currents in ventricular rabbit cardiomyocytes. Intracellular application (in the patch pipet) of 50 uM 2-APB did not have any effects on atrial APD, suggesting that IP3 receptor inhibition is not involved. However, subsequent application of 50 uM 2-APB in the bath prolonged the APD by 40% indicating that 2-APB prolongs atrial APD by inhibiting one or more K+ channel types in the sarcolemmal membrane from the extracellular site. RNA sequencing analysis revealed potential targets channel including TRP and Kv channels differentially expressed in atria versus ventricle. In human atrial cardiomyocytes from AF patients, application of 50 uM 2-APB prolonged APD by 25%. Conclusions 2-APB prolongs rabbit and human atrial APD and shortens rabbit ventricular APD in a concentration dependent manner, most likely due to modulation of sarcolemmal K+ channels, differentially expressed in the two cell types. Pharmacological studies with 2-APB may reveal novel cellular mechanisms identifying potential new targets for the treatment of atrial fibrillation.

Abstract:Recent improvements in MR technology have enabled the development of volumetric three-dimensional (3D) imaging methods for fat-suppressed T1-weighted images of the entire upper abdomen with pixel sizes of approximately 2 mm in all dimensions and with acquisition times of less than 25 seconds for breath-hold imaging. When performed with a timing scheme, dynamic contrast-enhanced volumetric imaging of the liver can be performed with selective imaging during the arterial phase and portal venous phase of enhancement. The volumetric data sets can be reconstructed in any oblique plane, enabling improved detection, localization, and characterization of small liver lesions. The combination of high-resolution isotropic pixels and accurate timing also permits angiographic reconstructions of the 3D images, producing MR angiography and venography that can be useful in therapeutic planning, such as for catheter-based interventions or surgical resections or transplantation. Additionally, with use of a hepatobiliary contrast agent such as mangafodipir, T1-weighted volumetric MR cholangiography can be performed in patients with nonobstructed systems for depiction of intrahepatic biliary anatomy.

Abstract:Objective To observe the frequency-and concentration-dependent effects of late sodium current inhibitor on the interatrial and intraventricular conduction time and acetylcholine-induced atrial fibrillation (AF).Methods New-Zealand White rabbit hearts were isolated and perfused in Langendorff mode.Hearts were paced at right atrium and para-His bundle area,and electrical signals were recorded from the left atrium and the apex to measure the interatrial and intraventricular conduction time.Results Ranolazine (1-15 μmol/L) prolonged interatrial conduction time in frequency-and concentration-dependent manners.Ranolazine prolonged the intraventricular conduction time at high concentration (＞15 μmol/L) but not at low and therapeutic concentrations (1-10 μmol/L).Ranolazine also exerted the ability of reducing AF burden and terminating AF induced by acetylcholine.In contrast,TTX(tetrodotoxin) and propafenone (1-3 μmol/L) prolonged both interatrial and intraventricular conduction time.Conclusion Ranolazine is relatively atrial selective to prolong interatrial conduction time in frequency-and concentration-dependent manners and can suppresses acetylcholine-induced AF.
