Abstract:Objective To observe the effects of oxidative low density lipoprotein (ox LDL) on number and function of endothelial progenitor cells (EPCs) from peripheral blood. Methods Total mononuclear cells (MNC) were isolated from peripheral blood by Ficoll density gradient centrifugation, and then the cells were plated on fibronectin coated culture dishes. After incubation for 7 days, attached cells were collected and stimulated with different cencentrations of ox LDL (25, 50, 100 and 200 μg/ml), native LDL (100 μg/ml) or vehicle control for different time (6, 12, 24 and 48 h). EPC were characterized and adherent cells which were double positive for DiLDL uptake and lectin binding by direct fluorescent staining were observed under a laser scanning confocal microscope. EPC were further identified by demonstrating the expression of CD34, VEGFR 2 and AC133 with flow cytometry. Proliferation, migration and in vitro vasculogenesis activity of EPC were assayed by MTT assay, modified Boyden chamber assay and in vitro vasculogenesis kit, respectively. EPC adhesion assay was performed by replating those on fibronectin coated dishes, then adherent cells were counted. Results Incubation of isolated human MNC with ox LDL decreased the number of EPC in a concentration dependent manner, being maximum at 200 μg/ml (approximately 70% reduction, P0.01). In time course experiments performed with a concentration of 100 μg/ml ox LDL, the decrease of EPC number became apparent at 12 h and the maximal decrease was at 24 h (approximately 50% reduction, P0.01). In addition, ox LDL impaired proliferative, migratory, adhesive and in vitro vasculogenesis capacities of EPC in dose and time dependent manners. Conclusion Incubation of EPC with ox LDL reduces the number of EPC and impaires their function.

Abstract:Aim To ascertain whether Dimethylsulfoxide (DMSO)-soluble particles from cigarette smoke (DSP) stimulate the proliferation of vascular smooth muscle cells (VSMC). Methods Rabbit aortic vascular smooth muscle cells were used as target cell. The proliferation of VSMC was assayed by the measurement of MTT and flow cytometry. Cellular protein assay (ELISA) was used to observe the change of protein content. The amount of endothelin-1 (ET-1) secreted by VSMC was measured by radioimmunoassay (RIA). Results In contrast to the control group (without DSP), protein absorbance of VAMC added 1∶1000, 2∶1000 and 3∶1000 DSP did not make significant difference (P0.05). VSMC added 1∶1000 and 2∶1000 DSP showed proliferation significantly (P0.01), and yet 3∶1000 DSP group did not. The amount of ET-1 in the medium of DSP groups were significantly increased, especially in 2∶1000 DSP group (P0.01) and the result of flow cytometry showed that in 2∶1000 DSP group, the cell numbers of G 1 phase were decreased and cell numbers of S phase were increased markedly (P0.01). Conclusion 1∶1000 and 2∶1000 DSP can stimulate significantly the proliferation of VSMC, and ET-1 produced by VSMC may play an important role in the proliferation of VSMC.

Abstract:OBJECTIVE:To study the effect of parthenolide on the proliferation of vascular smooth muscle cell(VSMC) and its mechanism.METHOD:Vascular smooth muscle cell was cultured, the protein levels of c-fos, c-myc, p15, p16, p18, p19 were measured by Western blot method, cell cycle were examined with flow cytometry, and the DNA synthesis was determined by [3H]-TdR incorporation.RESULT:Parthenolide inhibited protein levels of c-fos, c-myc in a time-dependent manner but didn't affect the protein levels of p15, p16, p18, p19. Flow cytometric DNA analysis revealed that parthenolide increased significantly G0/G1 phase of VSMC and decreased S phase of VSMC in a dose-dependent manner. Parthenolide inhibited [3H]-TdR incorporation in a dose dependent manner.CONCLUSION:Parthenolide may inhibit proliferation of VSMC by inhibiting the expressions of c-fos, c-myc, but not the expressions of p15, p16, p18, p19.
Yangxueqingnao Particles Inhibit Rat Vascular Smooth Muscle Cell Proliferation Induced by Lysophosphatidic Acid

Abstract:Objective: To observe the effect of Yangxueqingnao particles on rat vascular smooth muscle cell (VSMC) proliferation induced by lysophosphatidic acid (LPA). Methods: The amount of 3 H-TdR ( 3 H-thymidine) admixed in cultured rat VSMC was measured and mitogen-activated protein kinase (MAPK) activity and lipid peroxidation end product malondialdehyde (MDA) content of the VSMC were assayed. Results: 1×10 −9 , 1×10 −8 , 1×10 −7 mol/L LPA in a concentration dependent manner, induced the amount of 3 H-TdR admixed, MAP kinase activity, and MDA content of the cultured rat VSMC to increase. However, 5%, 10%, and 15% Yangxueqingnao serum preincubation resulted in a decrease of 23.0%, 42.0%, and 52.0% ( P <0.01) respectively in the amount of 3 H-TdR admixed, a decline in VSMC MAP kinase activity of 13.9% ( P <0.05), 29.6% ( P <0.01), and 48.9% ( P <0.01) respectively, and also, a decrease in MDA content of VSMC of 19.4%, 24.7%, and 43.2% ( P <0.01) respectively, in the 1×10 −7 mol/L LPA-treated VSMC. Conclusions: LPA activates the proliferation and lipid peroxidation of VSMC in a concentration dependent manner. The LPA-induced VSMC proliferation is related to the activity of MAP kinases, enzymes involved in an intracellular signalling pathway. The results of the present study showed that Yangxueqingnao particles can effectively inhibit LPA-induced VSMC proliferation, MAP kinase activation, and reduce lipid peroxidative lesion.
[Effect of oxidative low-density lipoprotein on the proliferation of bone marrow stem cell- derived smooth muscle cells]

Abstract:Objective: To establish the model of bone mesenchymal stem cell-derived smooth muscle cells (BMSC-SMCs) and investigate the role of BMSC-SMCs in the development and progression of artherosclerosis. Methods: BMSCs were isolated from the femoral bone of SD rats by adherent tissue culture method, and vascular smooth muscle cells (VSMCs) were obtained from the thoracic aorta. The differentiation of BMSCs into BMSC-SMCs was induced in the conditioned medium. The specific markers of BMSCs and BMSC-SMCs were identified by immunofluorescence (IF) staining. After treatment with 80 mg/L oxidative low-density lipoprotein (ox-LDL) for 72 h, the growth characteristics of BMSC-SMCs and VSMCs were observed. Flow cytometry was applied to analyze the cell cycle of BMSC-SMCs and VSMCs. Results: BMCS-SMCs transformed into foam cells after treatment with ox-LDL, which was more obvious in comparison with VSMCs. The growth curve of BMSC-SMCs and VSMCs presented with an S-shape pattern with the cell doubling time of 20 and 32 h, which was reduced to 15 and 28 h after treatment with 80 mg/L ox-LDL, respectively. Flow cytometry showed that exposure to 80 mg/L ox-LDL significantly increased G(0)/G(1) and decreased S and G(2)/M phase cells in both BMSC-SMCs (P<0.01, n=3) and VSMCs (P<0.05, n=3) in comparison with the control cells. Conclusion: BMSC-SMC might be involved in the formation of fatty core and accelerate the development of atherosclerosis.

Abstract:The arterial smooth muscle cells (SMC) are the predominant type of cells within atherosclerotic (As) lesions, and their proliferation plays an important role in the process of As genesis. On the basis of the establishment of primary culture and sub-culture method for human arterial SMC, the effects of LDL, VLDL, HDL, OX-LDL, OX-VLDL, OX-HDL on DNA synthesis of cultured human arterial SMC were. observed. Results are as follows: (1) HDL had no stimulating effect on H-3-TdR incorporation into DNA of SMC (P>0.05); (2) LDL and VLDL showed the obvious stimulating effects (P<0.05); (3) OX LDL, OX-VLDL and OX-HDL increased significantly 3H-TdR incorporation into SMC DNA, respectively (P<0.01). These results suggest that the atherogenic roles of LDL, VLDL, OX-LDL, OX-VLDL, OX-HDL are closely related to their stimulating effects on DNA synthesis and the proliferation of the arterial SMCs.

Abstract:BACKGROUND: Endothelial dysfunction is thought to play a crucial role in the pathogenesis of atherosclerosis induced by ox-LDL. Recently, a variety of evidence suggested that endothelial progenitor cells (EPCs) participated in neovascularization and reendothelialization. However, effects of ox-LDL on EPCs number and activity are ill understood.METHODS: Total mononuclear cells (MNCs) were isolated from peripheral blood by Ficoll density gradient centrifugation, and then the cells were plated on fibronectin-coated culture dishes. After 7 days culture, attached cells were stimulated with ox-LDL (to make a series of final concentrations: 25 microg/mL, 50 microg/mL, 100 microg/mL, 200 microg/mL), native LDL (100 microg/mL) or vehicle control for the respective time points (6 h, 12 h, 24 h and 48 h). EPCs were characterized as adherent cells double positive for DiLDL-uptake and lectin binding by direct fluorescent staining under a laser scanning confocal microscope. EPCs were further documented by demonstrating the expression of KDR, VEGFR-2 and AC133 with flow cytometry. Proliferation, migration and in vitro vasculogenesis activity of EPCs were assayed by MTT assay, modified Boyden chamber assay and in vitro vasculogenesis kit, respectively. EPCs adhesion assay was performed by replating those on fibronectin-coated dishes, and then counting adherent cells.RESULTS: Incubation of isolated human EPCs with ox-LDL decreased the number of EPCs in concentration-dependent manner, maximum at 200 microg/mL (approximately 70% reduction, P < 0.001). In time-course experiments performed with an ox-LDL concentration of 100 microg/mL, decrease of EPCs number became apparent at 12 hours and reached the maximum at 24 hours (approximately 50% reduction, P < 0.01). In addition, ox-LDL dose and time dependently impaired EPC proliferative, migratory, adhesive and in vitro vasculogenesis capacity.CONCLUSION: The results of the present study defined a novel mechanism of action of ox-LDL: the reduction of EPCs with decreased functional activity.

Pgc-1 Alpha Inhibits Oleic Acid Induced Proliferation And Migration Of Rat Vascular Smooth Muscle Cells

Abstract:Background. Oleic acid (OA) stimulates vascular smooth muscle cell (VSMC) proliferation and migration. The precise mechanism is still unclear. We sought to investigate the effects of peroxisome proliferator-activated receptor gamma (PPAR gamma) coactivator-1 alpha (PGC-1 alpha) on OA-induced VSMC proliferation and migration. Principal Findings. Oleate and palmitate, the most abundant monounsaturated fatty acid and saturated fatty acid in plasma, respectively, differently affect the mRNA and protein levels of PGC-1 alpha in VSMCs. OA treatment resulted in a reduction of PGC-1 alpha expression, which may be responsible for the increase in VSMC proliferation and migration caused by this fatty acid. In fact, overexpression of PGC-1 alpha prevented OA-induced VSMC proliferation and migration while suppression of PGC-1 alpha by siRNA enhanced the effects of OA. In contrast, palmitic acid (PA) treatment led to opposite effects. This saturated fatty acid induced PGC-1 alpha expression and prevented OA-induced VSMC proliferation and migration. Mechanistic study demonstrated that the effects of PGC-1 alpha on VSMC proliferation and migration result from its capacity to prevent ERK phosphorylation. Conclusions. OA and PA regulate PGC-1 alpha expression in VSMCs differentially. OA stimulates VSMC proliferation and migration via suppression of PGC-1 alpha expression while PA reverses the effects of OA by inducing PGC-1 alpha expression. Upregulation of PGC-1 alpha in VSMCs provides a potential novel strategy in preventing atherosclerosis.
Effect of Oxidated Low-Density Lipoprotein on Early Apoptosis of Vascular Smooth Muscle Cell

Abstract:OBJECTIVE:To investigate the apoptosis in vascular smooth muscle cell (VSMC) induced by oxidated low-density lipoprotein cholesterol (ox-LDL), and to discuss the relative mechanism of the apoptosis of VSMC.METHODS:The growth index of VSMC of Wistar rat was determined. By using trypan blue to count sustaining cells, the proliferative effect of ox-LDL on culturing VSMC was observed. With flow cytometer, the apoptosis of VSMC induced by ox-LDL and the inhibitory effect of cyclooxygenase-2 (COX-2) inhibitor NS-398 on apoptosis were determined.RESULTS:After VSMC grew normally in culture, ox-LDL in concentrations of 35 mg/L and 50 mg/L could advance the growth period of VSMC to between 12 and 24 hours, and the proliferation of VSMC were obvious. The proliferation of VSMC induced by ox-LDL were inhibited by NS-398. The rate of apoptosis of VSMC in ox-LDL and NS-398 groups were higher than that of normal control.CONCLUSION:ox-LDL could induce an obvious proliferation of VSMC in culture. Although NS-398 can decrease the proliferation of VSMC, it could enhance ox-LDL to induce apoptosis.

Abstract:Aim: As the major atheroprotective particle in plasma, high-density lipoprotein (HDL) is oxidized during atherosclerotic processes. Oxidized HDL (ox-HDL) may lose its cardioprotective properties and develop a proinflammatory and proatherogenic phenotype. The proliferation and migration of vascular smooth muscle cells (VSMCs) play a crucial role in atherogenesis. However, the influence of ox-HDL on VSMC proliferation and migration remains poorly understood. Methods: VSMCs were treated with native HDL (N-HDL) or ox-HDL at varying concentrations for different time intervals and used in several analyses. The degree of cell proliferation was assayed using CCK-8 kits. The level of cell migration was determined using a Transwell chamber and scratch-wound assay. The presence of intracellular reactive oxygen species (ROS) was detected based on ROS-mediated 2',7'-dichlorofluorescein fluorescence. The activation of NADPH oxidase was measured in terms of the Rac1 activity and NADP(+)/NADPH ratio. Results: Compared to N-HDL, ox-HDL significantly promoted VSMC proliferation and migration in a dose-dependent manner. In addition, ox-HDL remarkably activated NADPH oxidase and enhanced ROS generation in the VSMCs. Diphenyleneiodonium chloride, an inhibitor of NADPH oxidase, and N-acetylcysteine, a ROS scavenger, efficiently inhibited the ROS production triggered by ox-HDL and subsequently blocked the proliferating and migrating effects of ox-HDL in the VSMCs. Conclusions: Ox-HDL significantly induces VSMC proliferation and migration by promoting NADPH oxidase activation and ROS production. Furthermore, the inhibition of NADPH oxidase and ROS generation blocks the proliferation and migration of VSMCs induced by ox-HDL. These proliferating and migrating effects of ox-HDL are closely related to its proinflammatory and proatherogenic roles.
[Effects of Oxidized-Low Density Lipoprotein on Expression of Urotensin II Receptor GPR14 in Rat Aortic Smooth Muscle Cells].

Abstract:OBJECTIVE:To investigate the effects of oxidized-low density lipoprotein (ox-LDL) and urotensin II (U II) on the proliferation of rat aortic smooth muscle cells (VSMCs) and the effect of ox-LDL on the mRNA expression of U II receptor GPR14 in. METHODS:Rat VSMCs were incubated with UII and ox-LDL at different concentrations. The viability of rat VSMCs was detected by MTT assay. Rat VSMCs were incubated with ox-LDL at the concentrations of 0, 0.1, 1, 10, and 50 microg/ml. Twelve hours later competitive RT-PCR was used to detect the effects of concentration of ox-LDP on the mRNA expression of GPR14 in the VSMCs. Another VSMCs were incubated with ox-LDL at the final concentration of 50 microg/ml, and 0, 2, 6, 12, and 24 hours later competitive RT-PCR was used to detect the mRNA expression of GPR14 in the VSMCs. (125)I-labelled U II was added into the culture fluid of VSMCs, and radioligand binding assay was used to calculate the maximum binding (B(max)). RESULTS:UII of the concentration of 50 nmol/L significantly increased the proliferation of VSMCs. When the concentration of ox-LDL was 10 microg/ml the proliferation of VSMCs (P < 0.01) was 0.678 +/- 0.061, increased by 121% compared with the control group (0.325 +/- 0.052, P < 0.01). 0.01 microg/ml of ox-LDL and 10 nmol/L of UII showed a synergistic effect on the proliferation of VSMCs with the MTT value increased to 162% +/- 29% that of the control group (P < 0.01). When the concentration of ox-LDL increase to 0.1 microg/ml the synergistic effect was attenuated with the MTT value of 153% +/- 22% that of the control group (P < 0.05) and when the concentration of ox-LDL increased to 1 microg/ml the synergistic effect diminished, with a MTT value of 123% +/- 13% that of the control group (P > 0.05). When the concentration of ox-LDL was 50 microg/ml the proliferation of VSMCs the MTT value decreased to 59% that of the of control group, however, ox-LDL of the concentration of 50 microg/ml combined with UII of the concentration of 10 nmol/L the MTT value increased to 68% that of the control group, showing that UII of that concentration significantly inhibited the cytotoxic effect of ox-LDL. Incubation of VSMCs with the ox-LDL at the concentration of 0.1 microg/ml the GPR14 mRNA expression of the VSMCs was 125.1% that of the control group (P < 0.01) as assessed by competitive RT-PCR, however, when the concentration of ox-LDL increased over 1 microg/ml the GPR14 mRNA expression does-dependently decreased as much as 72.6% that of the control group (all P < 0.01), with the maximal effect when the concentration of ox-LDL reached 50 microg/ml. ox-LDL at the concentration of 50 microg/ml time-dependently down-regulated the GPR14 mRNA expression with the maximal effect 12 hours after addition of ox-LDL and this effect was sustained for up to 24 hours. After incubation for 12 hours the binding of (125)I-UII to VSMCs was 117.5% that of the control group (P < 0.01) with the ox-LDL at the concentration of 0.1 microg/ml, and was 68.8% that of the control group (P < 0.01) with the ox-LDL at the concentration of 50 microg/ml. CONCLUSION:ox-LDL and with U-II. GPR14 at certain concentrations show synergetic effects on the proliferation of VSMCs. The GPR14 mRNA expression in VSMCs can be upregulated by low concentration of ox-LDL, while downregulated by high concentration of ox-LDL.
Ox-LDL induces apoptosis in human coronary artery endothelial cells: role of PKC, PTK, bcl-2, and Fas

Abstract:Oxidized low-density lipoprotein (ox-LDL) plays a critical role in the development of atherosclerosis. Recent studies show that ox-LDL may induce apoptosis of cultured rabbit smooth muscle cells and human macrophages. This study was designed to determine the modulation by ox-LDL of apoptosis in cultured human coronary arterial endothelial cells (HCAEC) during hypoxia-reoxygenation and to determine underlying mechanisms. When HCAEC were ∼85% confluent, the cells were exposed to hypoxia (24 h)-reoxygenation (3 h), native LDL, or ox-LDL. Fragmented DNA end-labeling, DNA laddering, and light and electron microscopy were used to determine changes characteristic of apoptosis. Ox-LDL (20 μg/ml) increased apoptosis during hypoxia-reoxygenation compared with hypoxia-reoxygenation alone ( P < 0.05). Low concentrations of ox-LDL (5 μg/ml) and native LDL (20 μg/ml) under identical conditions had no effect on the degree of apoptosis. Ox-LDL markedly decreased endogenous superoxide dismutase activity and increased lipid peroxidation in HCAEC. The presence of ox-LDL, but not native LDL, in cultured HCAEC resulted in the activation of protein kinase C (PKC) and protein tyrosine kinase (PTK). The specific PKC and PTK inhibitors significantly reduced ox-LDL-mediated apoptosis of HCAEC ( P < 0.05). Hypoxia-reoxygenation significantly increased Fas expression and decreased bcl-2 expression in HCAEC lysate as determined by Western analysis. Ox-LDL further increased Fas expression and decreased bcl-2 expression. These data indicate that ox-LDL enhances hypoxia-reoxygenation-mediated apoptosis in HCAEC. Ox-LDL-mediated apoptosis of HCAEC appears to involve activation of PKC and PTK. In addition, ox-LDL modulates Fas and bcl-2 protein expression in HCAEC. This study also suggests that ox-LDL is more important than native LDL in hypoxia-reoxygenation-induced apoptosis.

Abstract:AIM: Cross talk between oxidized low-density lipoprotein (ox-LDL) and angiotensin II(Ang-Ⅱ) may play an important role in the pathogenesis of atherosclerosis. In this study, we tested the hypothesis that AngⅡ can induce expression of lectin-like oxidized low-density lipoprotein receptor-1 (LOX-1), a new receptor for ox-LDL, in cultured THP-1 cells. METHODS: Cultured THP-1 cells were treated with 0.1 μmol/L PMA for 48 hours at first .Then the cells were incubated with 10 -9 -10 -5 mol/L Ang II for 24 hours, or with 10 -6 mol/L Ang II for various time up to 48 hours. LOX-1 protein expression was examined by cell enzyme linked immunosorbent assay (ELISA), and LOX-1 mRNA was detected by quantitative competitive reverse transcription-polymerase chain reaction (RT-PCR) . RESULTS: PMA-untreated THP-1 cells didn't express LOX-1 mRNA. After 48 h of PMA treatment, THP-1 cells differentiated and LOX-1 mRNA was detected. Incubation of AngII with THP-1 cells for 24 h significantly increased LOX-1 mRNA and protein expression in a concentration-dependent manner (10 -9 -10 -5 mol/L, P0.05 ). Otherwise , after treatment with 1×10 -6 mol/L Ang II for 3 hours, LOX-1 protein and mRNA expression were elevated ( P 0.05), reaching the maximum at 24 hours, then it declined. CONCLUSIONS: PMA induces differentiation and LOX-1 mRNA expression in THP-1 cells. Ang II upregulates LOX-1 expression in a concentration-dependent and time-dependent manner, and this effect of Ang II may contribute to the pathogenesis of atherosclerosis.
ox-LDL-Induced Endothelial Progenitor Cell Oxidative Stress via p38/Keap1/Nrf2 Pathway

Abstract:Background. Nrf2 which was recently reported to regulate the antioxidant genes and cellular redox regulators was highly expressed in EPCs. However, its role in ox-LDL-induced EPC oxidative stress and apoptosis has not been fully illustrated. Methods. EPCs isolated from human peripheral blood mononuclear cells were treated with different concentrations of ox-LDL, Keap1 siRNA, and a specific p38 MAPK inhibitor SB203580 and then used to assay the cytoplasmic Nrf2, nuclear Nrf2, NAD(P) H:quinone oxidoreductase 1 (NQO1) and Bax/Bcl-2 levels with Western blot, NQO1 mRNA levels with RT-PCR, ROS levels with H2DCF-DA, loss/disruption of mitochondrial membrane potential with JC-1, apoptosis with Annexin V and PI, migration with transwell chambers, and tube formation with Matrigel. Results. ox-LDL decreased the nuclear Nrf2/Histone H3 to cytoplasmic Nrf2/GAPDH ratio, NQO1 mRNA, and protein levels. ox-LDL enhanced ROS production, induced the loss of membrane potential, and increased the cell shrinkage, pyknotic nuclei, and apoptosis of EPCs. Keap1 siRNA increased Nrf2 nuclear translocation, NQO1 mRNA transcription, and protein expression and prevented ROS generation and formation of JC-1 monomers. ox-LDL increased the activation of p38. SB203580 significantly eliminated ox-LDL induced inhibition of Nrf2 nuclear translocation, depression of NQO1 mRNA transcription, generation of ROS, and formation of JC-1 monomers in EPCs. Keap1 siRNA decreased the Bax/Bcl-2 ratio which was increased by ox-LDL in EPCs. ox-LDL decreased EPC migration and tube formation. Keap1 siRNA preserved the migration and tube formation of EPCs. Conclusion. ox-LDL activated EPCs p38/Keap1/Nrf2 pathway and induced oxidative stress, dysfunction, and apoptosis of EPCs.

Abstract:Objectives Increasing evidences shown the important role of bone marrow-derived mesenchymal stem cells (BM-MSCs) in wound healing and vascular remodelling in vivo. However, their mechanism in the development of atherosclerosis and vessel calcify remain unclear. Objective To investigate the effects of Oxidatively Modified Low Density Lipoprotein (ox-LDL ) on proliferation and osteodiferenliation of BM-MSCs that Co-cultured with Smooth Muscle Cells with or without osteogenic inductor being then we try to explain the mechanism of how MSCs participate in atherosclerosis. Methods BM-MSCs and VSMCs were prepared from SD rats and co-cultured in a Transwell system, which allowed the diffusion of secreted factors but prevented cell contact between the two cell types. Four groups were designed according as factorial design, which were Combine group (both osteogenic inductor and ox-LDL), ox-LDL group, Osteogenic inductor group and control group. The proliferation of BM-MSCs was determined by MTT assay. The their effects on the osteogenic potential of BM-MSCs by cell morphology, real-time PCR, immunofluorescent staining, alkaline phosphatase (AKP) activity and Osteopontin (OPN) synthesis. Results 1. The growth curve showed that cells number of all four groups increased along with time. The ability of promoting cellular proliferation in ox-LDL (5 mg/L) group were showed strongest in four groups, but that of combine group were opposite at 7days, 10 days and 14 days. 2. At the cultured 7 days, 10 days, all groups express OPN mRNA and AKP and but the combine group was highest. There was interaction between the two factors (P < 0.01). 3. The result of Immunity histochemistry showed that the rate of OPN and AKP positive cells in combine group were the highest at 14 days. Conclusions Increasing evidences indicated a link between bone marrow cells and the pathogenesis of atherosclerosis. It was recognized that a higher percentage of circulating endothelial progenitor cells expressed osteocalcin in patients with coronary atherosclerosis. Widespread increased myeloid calcifying cells contribute to ectopic vascular calcification in type 2 diabetes. Furthermore, mouse model and human studies have suggested that bone marrow-derived progenitor cells may contribute to a variety of vascular pathologies including calcification, atherosclerosis and intimal hyperplasia. In view of the capacity of differentiation into osteoblast from BM-MSCs, we investigated whether BM-MSCs was involved in the process of angiosteosis in atherosclerosis when exposed to vascular injure and hyperlipemia. Thus, we mimicked the phenotype transformation of VSMC exposed to hyperlipemia to investigate the differentiation of BM-MSCs. Data reported in this study demonstrated that ox-LDL significantly promoted proliferation of BM-MSCs co-cultured with VSMC, but not osteoblast differentiation. The proproliferative role of ox-LDL was neutralised by Osteogenic inductor, but apoptosis was induced instead. We results showed that ox-LDL can promote the proliferation of BM-MSCs, and Osteogenic Inductor mediated Osteodifferentiation marker expression in BM-MSCs in vitro.

Abstract:Background/Objectives: Oxidized low-density lipoprotein (ox-LDL) is a proinflammatory particle associated with various diseases and affects cell proliferation and viability in multiple cell types. However, its impact on intestinal epithelial cells remains underexplored. This study investigates the effect of ox-LDL on colonic epithelial cell proliferation and viability, as well as the underlying mechanisms involved. Methods: The expression levels of ox-LDL receptors in human colonoids were analyzed at baseline and in response to proinflammatory signals by qRT-PCR. The effect of ox-LDL on organoid proliferation was analyzed using morphometric measurements, viability assays, and the incorporation of a thymidine analog into DNA. The generation of reactive oxygen species (ROS) was determined by Amplex Red assays. Additionally, ox-LDL-induced ROS-dependent organoid proliferation was studied by exposing colonoids to an antioxidant or ROS inhibitors. Results: Colonic epithelial cells express ox-LDL receptors. Ox-LDL significantly induces the proliferation of colonic epithelial cells, which are dependent on ROS generation. Notably, ROS scavengers and NADPH inhibitors reduced ox-LDL-induced proliferation, highlighting the crucial role of oxidative stress in this process. Conclusions: This study demonstrates for the first time that ox-LDL stimulates CEC proliferation mediated by ROS production and validates that the colonic organoid model enables the analysis of potential pharmacological strategies for intestinal diseases characterized by oxidative stress and inflammation.

Abstract:Aim To investigate whether the expression of groupⅣcytosolic phospholipase A2(cytosolic phospholipase A2,cPLA2),groupⅤsecretory phospholipase A2(sPLA2-Ⅴ) and groupⅩsecretory phospholipase A2 (sPLA2-Ⅹ) could be regulated by oxidized low density lipoprotein(ox-LDL) of different oxidation levels in the mouse macrophage cell line RAW264.7 cells,and to explore the inflammatory mechanisms of the interactions between ox-LDL and macrophages in the process of promoting atherosclerosis.Methods 3 types of ox-LDL(minimally modified low density lipoprotein or MM-LDL,lightly ox-LDL or ox_(4h)-LDL,heavily ox-LDL or ox_(48h)-LDL) were prepared by different oxidation ways.3 types of ox-LDL were used to incubate with RAW264.7 cells for 4 h and 8 h,then the cell lysate and cell culture supernatant were collected.The mRNA expression and protein expression of cPLA2,sPLA2-Ⅴand sPLA2-Ⅹwere determined respectively by reverse transcription polymerase chain reaction(RT-PCR) and Western blotting analysis. Results The mRNA expression and protein expression of sPLA2-Ⅴand sPLA2-Ⅹwere undetectable in untreated RAW264.7 cell.Compared with the PBS control group and the LDL group,the mRNA expression of cPLA2 was not affected by all types of ox-LDL(P>0.05).But the mRNA expression and protein expression of sPLA2-Ⅴand sPLA2-Ⅹwere significantly enhanced by the stimulation of MM-LDL and ox_(4h)-LDL in RAW264.7 cell(P<0.05),while ox_(48h)-LDL had a slight ability to induce the expression of sPLA2-Ⅴand sPLA2-Ⅹ(P<0.05).However,PBS control group and native LDL failed to induce the expression of sPLA2-Ⅴand sPLA2-Ⅹ.Conclusion The exposure of ox-LDL to RAW264.7 cells did not affect the mRNA expression of cPLA2,but could significantly induce RAW264.7 cells to express sPLA2-Ⅴand sPLA2-Ⅹ.Two types of mildly oxidized LDL(MM-LDL and ox_(4h)-LDL)-induced expression of sPLA2-Ⅴand sPLA2-Ⅹwas stronger than that of heavily oxidized LDL(ox_(48h)-LDL),indicating that mildly oxidized LDL was capable of inducing a strong inflammatory response on macrophages.

Abstract:Despite many alternatives to point tracking problem, iterative least squares solution solving the optical flow constraint has been the most popular approach used by many in the field. This paper attempts to leverage the former efforts to enhance such tracking methods by introducing geometric constraint to the tracking problem. In contrast to alternative geometry based methods, the proposed approach provides an online solution to optical flow estimation by exploiting Horn and Schunck flow estimation regularized by view geometric constraints. We particularly use the geometric invariants in the projective coordinates and conjecture that the traditional appearance solution needs to be geometrically regularized. Geometric regularization is achieved by estimating the posterior distribution of possible locations given the samples drawn from other tracked points which specify the scene geometry. Our experiments demonstrate that tracking can still be performed with great accuracy even when the features undergo appearance changes due to projective deformation of the template. Hence the resulting algorithm judges the quality of the tracked points based on not only appearance similarity but also geometric consistency.

Abstract:It is to observe the effects of oxLDL on endothelial cell (EC) proliferation and apoptosis and to test the protective effects of Vit E, C and β carotene. Methods: Bovine aortic EC were incubated in culture media with vit E, C, and β carotene for 12h, and then exposed to oxLDL for 24h respectively. Results:(1) bovine aortic EC were deformed and shed as observed under phase contrast microscope in oxLDL group. The phenomena in antioxdant vitamin groups were slighter.(2) Inhibitory rate of bovine aortic EC proliferation in vit E, vit C and β carotene groups was significantly lower than that of oxLDL group.(3) Number of G 1 phase cells in oxLDL group was increased compared with vit E, vit C and β carotene groups respectively.(4) The apoptotic peak in oxLDL group was higher than that in antioxidant vitamin groups. Conclusion: Antioxidant vitamins have definite protective effects on injury induced by oxLDL on morphology, proliferation and apoptosis in EC possibly through promoting the demage repair and proliferation.
