Abstract:Several studies have shown that KAI1 inhibits tumor metastasis, but its mechanism is not clear. The present study aimed to determine the role of KAI1 in lymphatic metastasis, specifically in pancreatic cancer.The KAI1 gene was transfected into the pancreatic cancer cell line MIA PaCa-2 and PANC-1 by using liposomes and selected by G418, and the protein was measured by Western blotting. After successful infection, the cell growth curve was studied by MTT, vascular endothelial growth factor C (VEGF-C) secretion by pancreatic cancer cell were measured by ELISA. The KAI1 and pCMV transfected MIA PaCa-2 cells were renamed as MIA PaCa-2-K and MIA PaCa-2-p. These two kinds of cells were injected into the subcuticular layer of nude mice; both tumor growth and metastasis through the lymphatic nodes were assessed. Lymphangiogenesis in tumors was measured by immunohistochemistry.The VEGF-C secretion was significantly reduced in MIA PaCa-2 cells compared with PANC-1 cells after being transfected with the KAI1 gene. The growth rate of subcutaneous tumors was similar after the injection of MIA PaCa-2-K, MIA PaCa-2, and MIA PaCa-2-p. MIA PaCa-2-K tumors showed slower lymphangiogenesis and lymph node metastasis compared with MIA PaCa-2 and MIA PaCa-2-p tumors.The overexpression of KAI1 inhibits the lymphangiogenesis and lymph node metastasis of MIA PaCa-2 pancreatic tumors.

Abstract:KAI1 is a metastasis suppressor gene on human chromosome 11p11.2 that encodes a glycoprotein of the transmembrane four superfamily. Reduced KAI1 expression associates with malignant progression of human prostatic, lung and pancreatic cancers, but the role of KAI1 protein in the malignant progression of other human cancers remains to be elucidated. We analysed KAI1 protein in normal and cancer cells of the prostate, ovary, bladder, endometrium, lung and melanocytes by Western blot to determine if KAI1 may be involved in multiple cancers. We also investigated the relationship of KAI1 expression and two other transmembrane four superfamily proteins, CD81 and CD9, in the cells. We found that KAI1 protein was downregulated in 31/42 of the cancer cell lines analysed. Alternatively, some ovarian, bladder and endometrial cells had distinct, heterogeneous KAI1 protein band patterns in Western blots that were due primarily to N-linked glycosylation. Most of the cancer cells expressed two other transmembrane four superfamily proteins, CD81 and CD9. Downregulation of KAI1 protein may be an indicator of metastatic potential in cancers of urogenital, gynecological, and pulmonary origin and in melanomas. KAI1 may also have post-translational modifications specific to tissue type or malignant progression.

Abstract:The objective of this study is to investigate the expression of tumor metastasis suppressor gene KAI1 protein and matrix metalloproteinase 2 (MMP-2) in breast carcinoma. We retrospectively analyzed 60 cases of patients from June, 2005 to June, 2011 treated with radical mastectomy for invasive ductal breast cancer in our department. The carcinoma tissue was stained for KAI1 and MMP2 with immunohistochemistry. In breast cancer group, the positive expression rate of KAI1 was 41.7%, which was significantly lower than control (83.3%) (chi(2) = 17.328, P < 0.05); the positive expression rate of MMP2 was 80.0%, which was significantly higher than control group (16.7%) (chi(2) = 41.637, P < 0.05). Within the breast cancer patients, the group with lymph node metastasis showed higher positive rate of KAI1 and MMP2 expression than those without lymph node metastasis (P < 0.05). The patients with tumor diameter more than 5 cm showed lower positive rate of KAI1 but higher positive rate of MMP-2 expression than patient group with tumor diameter less than 5 cm. Additionally, tumor-nodes-metastasis III (TNM III) grade patients showed lower positive rate of KAI1 but higher positive rate of MMP-2 expression than patient group in I and II grades. KAI1 and MMP-2 were highly expressed in breast cancer tissues. The positive expression of KAI1 and MMP-2 were associated with tumor metastasis, tumor size, TNM grades etc.
Expression of KAI1 Gene in Non-Small Cell Lung Cancer and Its Relationship with P53

Abstract:BACKGROUND:Recent researches have manifested that down-regulation of KAI1 relates to metastasis in various tumors, but its association with non-small cell lung cancer (NSCLC) and the mechanism for its down-regulation are not clear. The aim of this study is to investigate the expression of KAI1 in NSCLC and its relationship with clinicopathological characteristics and mutant P53 protein. METHODS:The expression of KAI1/CD82 and mutant P53 protein was detected in 48 cases of NSCLC tissues by Western blot, and KAI1 mRNA was detected by RT-PCR method, with 20 cases of pulmonary benign disease tissues and normal lung tissues as control.. RESULTS:The positive rate of KAI1 mRNA was 52% in lung cancer group and 90% in control group, respectively (P < 0.01), KAI1/CD82 was 48% and 85% respectively (P < 0.01), and mutant P53 protein was 65% and 5% respectively (P < 0.01). The positive rate of KAI1 mRNA, KAI1/CD82 and mutant P53 protein closely related to the tumor stages, cell differentiation and lymph node metastasis status (P < 0.05 or P < 0.01). The expression of KAI1/CD82 highly related to KAI1 mRNA (P < 0.01) and mutant P53 protein (P < 0.05), while expression of KAI1 mRNA did not relate to mutant P53 protein expression (P > 0.05). CONCLUSIONS:The down-regulation of KAI1 may relate to carcinogenesis, development and metastasis of NSCLC. Its reduction may occur mainly at transcriptional level and correlate with p53 in NSCLC. KAI1 and p53 might be helpful to predict the potential metastasis of NSCLC.

Abstract:Objective: To investigate the expression of KAI-1 mRNA in renal cell carcinoma and the relationship with the metastasis. Method: RT-PCR was used to detect KAI-1 mRNA expression in specimens from 35 cases with renal cancer and adjacent normal kidney tissues. Results: KAI-1 expression rate in renal cancer was significantly lower than that in normal kidney tissues (P< 0.05); tumors of patients with extrarenal invasion or regional lymph node metastasis (stage Ⅲ orⅣ ) had significantly lower KAI-1 expression levels than tumors of patients with stage Ⅰ or Ⅱ disease (P< 0.05).Conclusion: Decreased expression of the KAI-1 gene may be related to disease progression in patients with renal cancer.

Abstract:OBJECTIVE:To investigate the role of KAI1 gene expression and loss of heterozygosity (LOH) of KAI1 in metastatic potential and prognosis of pancreatic cancer.METHODS:The expression of KAI1 gene was studied by immunohistochemistry for CD82 on paraffin-embedded tumor tissues. The LOH of KAI1 gene was detected by microdissection, polymerase chain reaction (PCR) and denaturing high performance liquid chromatography (DHPLC).RESULTS:The positivity rate of CD82 in primary pancreatic cancer was 76% (47/62). CD82 expression was significantly higher (P < 0.01) in earlier tumor stages (I and II), as compared to the advanced tumor stages ( III and IV) in which nodal or distant metastases were present. The expression rate of CD82 in patients who survived for more than one year was higher than that in patients who survived for less than one year (P < 0.05). The percentage of LOH at D11S1344 and D11S1326 loci was 17%.CONCLUSIONS:The abnormal expression of CD82 which participates in malignant progression of pancreatic cancer is probably associated with LOH of KAI1 gene. Detection of CD82 expression and LOH of KAI1 gene may carry potential clinical significance in evaluating the metastatic potential and prognosis of pancreatic cancer.
Loss of Tumor Suppressors KAI1 and P27 Identifies a Unique Subgroup of Primary Melanoma Patients with Poor Prognosis

Abstract:Primary melanoma, a highly aggressive malignancy, exhibits heterogeneity in biologic behaviors, clinical characteristics, metastasis potential and mortality. The present study sought to identify the molecular signatures that define a subgroup of primary melanomas with high risks of metastasis and mortality. First, we identified the markers that best differentiated metastatic melanomas from primary melanomas by examining the expression of seven previously reported biomarkers (BRAF, Dicer, Fbw7, KAI1, MMP2, p27 and Tip60) in a training cohort consisting of 145 primary melanomas and 105 metastatic melanomas. KAI1 and p27, both tumor suppressors, emerged as best candidates. Loss of both tumor suppressors occurred in the majority (74.29%) of metastatic melanomas. Further, a subset (metastatic like, or "ML", 33.10%) of primary melanomas also lost these two tumor suppressors. Kaplan-Meier analysis indicated that ML subgroup of primary melanoma patients had much worse 5 year survival compared with other primary melanoma patients (P = 0.002). The result was confirmed in an independent validation cohort with 92 primary melanomas (P = 0.030) and in the combined cohort with 237 melanoma patients (P = 3.00E-4). Additionally, compared to KAI1 and p27 as an individual prognostic marker, the combined signature is more closely associated with melanoma patient survival (P = 0.025, 0.264 and 0.009, respectively). In conclusion, loss of both KAI1 and p27 defines a subgroup of primary melanoma patients with poor prognosis. This molecular signature may help in metastatic melanoma diagnosis and may provide information useful in identifying high-risk primary melanoma patients for more intensive clinical surveillance in the future.

Abstract:KAI1 gene is a novel metastasis suppressor gene. Its transcription level has been reported to be related with metastases in several kinds of tumors. The Authors used RT-PCR method to investigate the expression levels of KAI1 gene in hepatocellular carcinoma (HCC) tissues from 42 patients. The results demonstrated that 57.1% of tumor tissues were positive in KAI1 expression. There was no statistically significant difference on KAI1 gene expression rate between large (>5cm) and small (<=5cm) tumors, encapsulated and non-capsulated tumors, as well as alpha-fetoprotein (AFP) positive (>20ug/ml) and negative (<=20ug/ml) tumors; however, the expression rate was higher in tumors without intrahepatic metastasis than in those with intrahepatic metastasis (63.9% vs. 16.7%, P<0.05). It is suggested that KAI1 gene expression may be related to the metastases of hepatocellular car cinema.

Abstract:Abstract Objective: Melanoma is a highly heterogeneous disease. A certain subgroup of patients with primary melanomas exhibits a greater potential to develop metastatic disease and thus has worse survival. The present study attempts to identify the molecular features shared by metastatic melanomas and the subgroup of primary melanomas, and use the information to assist clinicians to design personalized therapy for primary melanoma patients. Patients and methods: Seven previously reported biomarkers, including BRAF, Dicer, Fbw7, KAI1, MMP2, p27 and Tip60, were investigated. A training cohort with 250 melanoma patients (145 primary melanomas and 105 metastatic melanomas) and an independent cohort with 92 primary melanoma patients were used for the study. Logistic regression analysis was used to identify discriminate biomarkers of metastatic melanoma from primary melanoma. Kaplan-Meier survival and multivariate Cox proportional hazard regression analysis were performed to assess the significance of the molecular signature in the prognosis of melanoma. Results: In the training cohort, we found the loss expression of KAI1 and p27 to be most significant between metastatic and primary melanoma. The primary melanoma patients with loss expression of both KAI1 and p27 had poor 5-year survival in both training cohort (P = 0.002) and independent cohort (P = 0.03). Multivariate Cox regression analysis showed that the KAI1-/p27- signature was an independent factor for disease- specific survival (P = 0.004). More important, compared to KAI1 and p27 as an individual prognostic marker, the KAI1-/p27- signature is more closely associated with melanoma patient survival: the P value is 0.004 for KAI1-/p27-, and 0.044, 0.181 for KAI1 and p27, respectively. Conclusion: Loss of both KAI1 and p27 defines a subgroup of primary melanoma patients with poor prognosis, and the combination signature is better than individual biomarker. The clinical significance of KAI1 and p27 warrants further assessment in prospective clinical trials. The Significance of KAI-1 Tumor Metastasis Suppression Gene Expression for the Metastasis of Renal Carcinoma

Abstract:Objective To test the statistical relationship between the loss of expression of KAI-1 on T1-3aN0M0 stage renal carcinoma specimens and the postoperative metastases. It implied that the loss of KAI-1 expression took part in the postoperative metastases of renal carcinoma. The study is to test testify the effect of KAI-1 to inhibit of renal carcinoma metastases by means of naked mice experiment. Methods A piece of KAI-1 negative specimen of renal carcinoma was chosen and was made to renal carcinoma cell solution and then these cell were incubated and stored. The KAI-1 negative renal carcinoma cells were transinfected into KAI-1 positive cells with KAI-1 gene plasmid. Every forty naked mice were inoculated with KAI-1 negative or positive renal carcinoma cell. The growth of subcutaneous tumor and the number of lung metastatic tumors in naked mice were counted and the results were compared by χ~2 testing. KAI-1 expression was examined with PCR and immunohistochemistry. Results Renal carcinoma cells were inoculated successfully in every naked mouse and tumor growth was observed and the tumors grew rapid dramatically from the second week to the third. There were 40 naked mice on which surface of lungs the grey and red nodules of 1～2mm in diameter were distinguished and the average number was 6 on surface of lung in each mouse with KAI-1 positive renal carcinoma(0～11). Otherwise,the average number was 31 in KAI-1 negative mice(0～57).There was statistical significant difference between the two groups(χ~2=5.01,P<0.05). Conclusion The loss of KAI-1 expression is able to promote the metastases of renal carcinoma. This kind of experimental evidence may help to prognosticate the clinical metastases of renal carcinoma by way of testing KAI-1 expression.
The expression levels of KAI1/CD82 mRNA in intestinal tumor and investigation the regulative mechanism

Abstract:Objective To investigate the expression levels of KAI1/CD82 mRNA in intestinal tumour and normal intestinal tissues and the regulative mechanism of KAI1/CD82 expression. Methods KAI1 mRNA of 18 cases with intestinal tumour as well as, normal intestinal tissues were detected using duplex reverse transcription polymerase chair reaction (duplex RT-PCR)method.Bel-7402 and SMMC-7721 HCC cell lines were cultured after being demethylated by 5-aza-deoxycytidine.Results The average expression levels of KAI1/CD82 mRNA in intestinal tumour tissues(0.78±0.22)were significantly by lower than that in normal intestinal tissues(1.53±0.12)P0.05 .KAI1/CD82 expression of intestinal tumour metastasic group(0.70±0.24)weren't significantly lower than that of non-metastasic group.After being demethylated by 5-aza-deoxycytidine,the expression level of KAI1/CD82 mRNA in Bel-7402 cell increased,but could't detect significant change in SMMC-7721 cell.Conclusion The indicated KAI1/CD82 in intestinal tumor might be a marker for intestinal tumour cell migration and that several regulatory mechanisms including gene demethylation might be involved in KAI1/CD82 expression.
The correlation of KAI1, Slug and vasculogenic mimicry in the prediction of metastasis and prognosis in colorectal carcinoma.

Abstract:KAI1 and the epithelial-mesenchymal transition regulator Slug, as well as vasculogenic mimicry (VM), play a significant role in the process of metastasis and prognosis in many human cancers. This study investigated the relationship of KAI1, Slug and VM and their connection with clinicopathological features, metastasis and their prognostic value in colorectal carcinoma (CRC). Immunohistochemistry was performed to analyze the expression of KAI, Slug, VM, E-cadherin (E-cad) and vimentin in 134 human CRC and corresponding normal mucosal tissues. The results showed that the protein expressions of KAI1, Slug and VM in tumor tissues were significantly different from adjacent normal mucosal tissues. The expression of KAI1 was negatively related, while the expressions of Slug and VM are positively related with differentiation, lymph node metastasis, distant metastasis and TNM stage. KAI1 was negatively related, while VM was positively related with invasion. The expression of KAI1 showed a negative correlation with Slug and VM. Moreover, the expression of Slug was positively associated with VM. A Kaplan-Meier analysis showed that the expression of Slug and VM was negatively and KAI1 expression was positively correlated with overall survival. Low KAI1 expression and high Slug and VM expression showed a poorer prognosis in CRC. Multivariate Cox regression analysis showed that the expression of KAI1, Slug, VM and TNM stage were independent predictors of OS in CRC. The expression of KAI1, Slug and VM was correlated with metastasis and prognosis. These findings suggest that KAI1, Slug and VM can be novel therapeutic targets to predict metastasis and prognosis in CRC patients.
Src/STAT3 Signaling Pathways Are Involved in KAI1-induced Downregulation of VEGF-C Expression in Pancreatic Cancer.

Abstract:A previous study by our group demonstrated that overexpression of KAI1 was associated with lymphatic metastasis in pancreatic cancer. The present study further investigated the signaling pathways involved in KAI1-induced downregulation of vascular endothelial growth factor C (VEGF-C) and lymphatic metastasis in pancreatic cancer. Immunohistochemistry was performed to examine KAI1 and VEGF-C expression in 28 surgically resected pancreatic cancer tissues. MIA PaCa-2 and PCAN1 pancreatic cancer cell lines were transfected with KAI1 overexpression vector. VEGF-C expression as well as phosphorylation of Src and signal transducer and activator of transcription (STAT)3 were assessed by western blot analysis. Furthermore, the signal transduction inhibitors PP2 and AG490 were used to block the Src and STAT3 signaling pathways, respectively. KAI1 was negatively correlated with VEGF-C expression in pancreatic tumor samples. In MIA PaCa-2 cells, VEGF-C expression was more significantly inhibited by restoration of KAI1 than that in PCAN1 cells. In addition, Src and STAT3 phosphorylation was decreased by KAI1 in MIA PaCa-2 cells. Of note, pre-treatment with PP2 efficiently reversed the KAI1-induced enhancement of Src and STAT3 phosphorylation as well as VEGF-C expression. Pre-treatment with AG490 efficiently reversed the KAI1-induced enhancement of STAT3 phosphorylation and VEGF-C expression, but had no effect on the upregulation of Src phosphorylation. The present study identified the involvement of Src/STAT3 signaling pathways in KAI1-induced downregulation of VEGF-C expression and suggested the implication of these pathways in lymphatic metastasis of pancreatic cancer.
Evaluation of the correlation of KAI1/CD82, CD44, MMP7 and β-catenin in the prediction of prognosis and metastasis in colorectal carcinoma

Abstract:Background To investigate the relationship of KAI1/CD82, CD44, matrix metalloproteinase 7 (MMP7) and β-catenin, and examine its association with clinicopathological features, metastasis and prognosis in colorectal carcinoma (CRC). Methods Immunohistochemical (IHC) analysis was used to detect the expression of KAI1/CD82, CD44, MMP7 and β-catenin in 174 archival surgical specimens of human CRC. Furthermore, clinicopathological features such as age, sex and so on were also collected retrospectively. Results CD44, MMP7 and β-catenin expression was positively associated with distant metastasis, lymph node metastasis and tumor-node-metastasis (TNM) stage. However, decreased KAI1/CD82 expression correlated significantly with distant metastasis, lymph node metastasis and TNM stage. KAI1/CD82 expression showed a negative correlation with CD44, MMP7 and β-catenin. Furthermore, β-catenin expression showed a positive correlation with CD44 and MMP7. Multivariate logistic regression analysis showed that KAI1/CD82 and β-catenin expression were significantly associated with lymph node metastasis and KAI1/CD82 was significantly associated with distant metastasis. Kaplan-Meier analysis revealed that CD44, MMP7 and β-catenin expression was negatively correlated with overall survival (OS), while KAI1/CD82 expression was positively correlated with OS. Low KAI1/CD82 expression and high expression of CD44, MMP7 and β-catenin was associated with a poor prognosis in CRC. Multivariate Cox regression analysis indicated that the expression of KAI1/CD82, MMP7 and β-catenin were independent predictors of OS in CRC. Conclusion The expression of KAI1/CD82, CD44, MMP7 and β-catenin is related to tumor metastasis and prognosis in CRC. Combined detection of these factors may be of significant value in predicting the prognosis and metastasis in CRC patients.

Abstract:To investigate the expressions of casein kinase II β(CK2β) and X-Linked inhibitor of apoptosis protein (XIAP) in cholangiocarcinoma (CCA) and evaluated their correlations with major clinicopathologic features and patients’ survival. Fifty CCA specimens and 20 normal liver tissues were included in the study. Immunohistochemical staining was used to determine the expression levels of CK2β, XIAP in normal and CCA tissues. The relationships of CK2β and XIAP expressions with clinicopathologic parameters and clinical outcome were evaluated. High immunostaining of CK2β and XIAP were observed in 66 % (33/50) and 68 % (34/50) of CCA tissues, which were significantly higher than that of normal liver tissues 0 % (0/20) and 25 % (5/20). The high expression of CK2β was significantly associated with TNM stage (P = 0.036), histological grade (P = 0.020) and high serum CEA level(P = 0.010), while high expression of XIAP was only associated with TNM stage(P = 0.014) and high serum CEA level(P = 0.001). By univariant analysis, patients with high expression of CK2β and XIAP demonstrate significantly poorer overall survival (P = 0.003 vs P = 0.018). Cox regression model showed that positive expression of CK2βis an independent factor of prognosis (P = 0.004). The expressions of CK2β and XIAP in CCA tissues showed strong correlations with the tumor progression, CK2β may be applied as a potential prognostic marker for CCA.
Effects of Altered Expression and Activity Levels of CK1δ and Ɛ on Tumor Growth and Survival of Colorectal Cancer Patients

Abstract:Colorectal cancer (CRC) is the fourth leading cause of cancer related death worldwide due to high apoptotic resistance and metastatic potential. Because mutations as well as deregulation of CK1 isoforms contribute to tumor development and tumor progression, CK1 has become an interesting drug target. In this study we show that CK1 isoforms are differently expressed in colon tumor cell lines and that growth of these cell lines can be inhibited by CK1‐specific inhibitors. Furthermore, expression of CK1δ and ɛ is changed in colorectal tumors compared to normal bowel epithelium, and high CK1ɛ expression levels significantly correlate with prolonged patients' survival. In addition to changes in CK1δ and ɛ expression, mutations within exon 3 of CK1δ were detected in colorectal tumors. These mutations influence ATP binding resulting in changes in kinetic parameters of CK1δ. Overexpression of these mutants in HT29 cells alters their ability to grow anchorage independently. Consistent with these results, these CK1δ mutants lead to differences in proliferation rate and tumor size in xenografts due to changes in gene expression, especially in genes involved in regulation of cell proliferation, cell cycle, and apoptosis. In summary, our results provide evidence that changes in the expression levels of CK1 isoforms in colorectal tumors correlate with patients' survival. Furthermore, CK1 mutants affect growth and proliferation of tumor cells and induce tumor growth in xenografts, leading to the assumption that CK1 isoforms provide interesting targets for the development of novel effective therapeutic concepts to treat colorectal cancer.

Abstract:Objective KAI 1/CD82, a member o TM4SF(Transmembrane 4 Superfamily),has important biological roles such as regulatiog cell adhesion and cell migration. The ratios of KAI 1/CD82mRNA to β actin cDNA in the same tissue was used as the expression level of KAI 1/CD82 mRNA in hepatocellular carcinoma and normal hepatic tissues,studies on the regulative mechanism.Methods KAI 1 mRNA of 8 cases with hepatocellular carcinoma and normal hepatic tissues were detected using duplex reverse transcription polymerase chain reaction (duplex RT PCR)method as compared with product of β actin.KAI 1/CD82mRNA and β actin mRNA were reversed transcription in one tube, so that the condition of reverse transcription polymerase chain reaction between KAI 1 and β-actin genes were same. Bel 7402 and SMMC 7721 of human hepatocellular carcinoma celllines were cultured after being demethylated by 5 aza deoxycytidine. The condition is 37℃ ,5%CO 2 saturation humidity,nonbacterium.The expression levels rations of KAI 1/β actin,mRNA by duplex reverse transcription polymerase chain rection were detected,used the instrument of light density scanning.Results The product of KAI 1 was 484 bp and the one of β actin was 370 bp.Under the condition of being sure that the extracted total RNA was pure, the results as follows ,the average expression levels of KAI 1/CD82 mRNA in depatocellular carcinoma (0.70±0.13)were significant lower than that in normal hepatic tissues (1.53±0.03) and that in paracancer tissues(1.47±0.18)P0.05.After being demethylated by 5 aza deoxycytidine, the expression level of KAI 1/CD82 mRNA in Bel-7402 cell increased,but couldn't detect significant change in SMMC 7721cell.Conclusion These results indicated that decreased KAI 1/CD82 in hepatocellular carcinoma might be a marker for hepatocellular carcinoma cell migration and that several regulatory mechanisms including gene demethylation might be involved in KAI 1/CD82 expression. The decreasing of the expression levels of KAI 1/CD82 mRNA affected by many factors and maybe demethylate gene.The mechanisms are different on various type cells.
Low Expression Of 2 '-5 ' Oligoadenylate Synthetase 1 Predicted Poor Prognosis In Colorectal Cancer

Abstract:2'-5' oligoadenylate synthetase 1 (OAS1) is an IFN inducible enzyme and can activate RNaseL, degrade cellular RNA and inhibit protein synthesis. Except anti-virus infection, OAS1 is involved in cell growth, differentiation, apoptosis and migration in several cancers. The purpose of this study was to clarify the role of OAS1 in progression of colorectal cancer (CRC) and to examine whether OAS1 can be used as a prognostic marker for CRC. 135 CRC patients who underwent surgical resection were investigated. The expression of OAS1 was evaluated by immunohistochemical staining to analyze the relationship between OAS1 protein expression and clinicopathological parameters. OAS1 was mainly localized in the nucleus of normal colorectal mucosa and tumor cells, scarcely in the cytoplasm. High expression of OAS1 was only detected in 31.1% of cases. Low expression of OAS1 was significantly correlated with poor pathologic differentiation, lymph node metastasis, distant metastasis, and advanced TNM stages (P < 0.05 for each). Patients with high OAS1 expression had a significantly better 5-year overall survival rate (P=0.001). The same results were found in different groups of patients with G2/G3, lymph node metastasis, TNM stage III/IV, and without distant metastasis (P < 0.05 for each). Moreover, multivariate analysis showed that OAS1 expression proved to be an independent prognostic factor (P=0.039). Patients with high expression of OAS1 exhibited better prognosis than those with low expression of OAS1. Thus, OAS1 could be a useful prognostic marker and potential therapeutic target for CRC patients.

Abstract:BACKGROUND: Transketolase-like protein 1 (TKTL1) is an isoform of tranketolase, a key protein in a cancer cell's glucose metabolism that causes rapid cell growth and controls the non-oxidative part of the pentose phosphate pathway (PPP). Its overexpression occurs in several human cancer types. Our purpose was to study whether TKTL1 expression in colorectal cancer tissue associates with these patients' prognosis.METHODS: We collected retrospectively patient data and tissue samples from 840 colorectal cancer patients treated at Helsinki University Hospital, then stained tumor tissue microarrays for TKTL1 by immunohistochemistry, and compared immunohistochemical tissue expression with clinico-pathological parameters and survival.RESULTS: High expression of TKTL1 associated with high Dukes stage, non-mucinous adenocarcinoma, and left-sided disease. Patients with high TKTL1 expression had poorer prognosis than those with low expression, with a 5-year disease-specific survival of 55.7% vs. 62.7%.CONCLUSION: We show that high TKTL1 in tumor tissue can lead to poor survival in colorectal cancer. TKTL1 thus can serve as a candidate marker for identifying patients at risk of recurrent disease.
