Abstract:Day-old chicks were trained on a one-trial passive avoidance task by pecking at a small, shiny bead coated with either a bitter-tasting substance (methylanthranilate) or water. The undifferentiated spontaneous multi-unit activity recorded bilaterally from anaesthetized chicks 1-13 h after training on the above task exhibited a significant increase in the methylanthranilate-trained over water-control chicks within three structures of the right hemisphere: the hyperstriatum accessorium (47%, P less than 0.05), the medial hyperstriatum ventrale (49.1%, P less than 0.02) and the medial portion of the paleostriatum augmentatum (47.5%, P less than 0.02). Within the multi-unit record obtained from both groups of chicks there were periods of short duration (15-20 ms) containing high-frequency (400-450 Hz) large-amplitude (greater than or equal to 200 microV; 450 microV max peak-to-peak) spikes. As a consequence of training, there was a massive increase in the occurrence of these bursting epochs in the medial hyperstriatum ventrale of both the left (320%, P less than 0.001) and right (350%, P less than 0.001) hemispheres in methylanthranilate-trained compared to water-trained chicks. In addition, the mean number of spikes per burst at this site increased by 66% (P less than 0.001) with no change observed in any other structure sampled. The statistically significant increase in activity within the right hyperstriatum accessorium and medial paleostriatum augmentatum and the non-significant increase in these structures in the left hemisphere was produced almost entirely by tonic spiking. Enhanced spontaneous multi-unit activity recorded under anaesthesia following passive avoidance training in the chick is shown here to be a feature common to several medial forebrain structures. The magnitude of the elevation in bursting frequency and the degree of localization of this effect to the medial hyperstriatum ventrale of methylanthranilate trained chicks would appear to offer strong support to previous biochemical and morphological evidence implicating this structure in the process of memory consolidation for this task. The data reported here represent the first evidence of electrophysiological changes occurring as a consequence of passive avoidance training in the domestic chick.
Passive avoidance training results in lasting changes in deoxyglucose metabolism in left hemisphere regions of chick brain

Abstract:Day-old chicks peck when offered a bright bead; if the bead is coated with the bitter-tasting methylanthranilate (M) they avoid it thereafter. 2-[14C] Deoxyglucose injected 1 min prior to training shows increased uptake into the hyperstriatum ventrale (HV) and lobus parolfactorius (LPO) 30 min later compared with control birds which have pecked a water-coated bead (W). To distinguish effects of training from those of consolidation, and to study lateralization of the increased uptake, 2-[14C]deoxyglucose (4 muCi) was injected ip either 5 min before, or 10 or 30 min after training. Thirty minutes after injection, bilateral samples of medial hyperstriatum ventrale (MHV), LPO and palaeostriatum augmentatum (PA)-enriched regions were dissected. Specific radioactivity (dmp/mg X prot) in left and right MHV and left and right LPO was standardized on the mean PA-specific radioactivity for each bird. When 2-DG was injected 5 min prior to training, standardized radioactivity in the left LPO was 26% greater, and in the left MHV 13% greater in M than W birds. There were no differences in the right hemisphere. With injection 10 min after training, there was an increase of 22% in the left LPO of M birds over W, of 29% in the left MHV and 22% in the right MHV. If injection was delayed to 30 min after training, there was no increase in the LPO, but a 13% increase persisted in the left MHV. Enhanced 2DG metabolism following passive avoidance training is thus persistent, lateralized, and, in the MHV at least may represent an aspect of cellular reorganization consequent on experience but independent of the immediate concomitants of training--perhaps part of the process of memory consolidation.
Cellular correlates of stages of memory formation in the chick following passive avoidance training

Abstract:The process of memory formation has been investigated using the model of one-trial passive avoidance training in the one-day old domestic chick. We have unraveled a biochemically coherent cascade of processes which, beginning with transient ion and neurotransmitter flux, and by way of a sequence of interacting pre- and post-synaptic intracellular signalling steps, results in gene activation and the synthesis of cell adhesion molecules which appear to be the effective agents in the structural processes involved in remodelling of synaptic and neuronal circuits. Further, in a related series of experiments we have shown that these biochemical and morphological changes are accompanied by significant changes in the neurophysiological status of the neurons on the IMHV and LPO, in particular in terms of their engagement in bouts of high-frequency firing. However, much remains to be clarified, particularly the meaning of the time-dependent shifts in the location of the trace, and the ways in which these molecular and cellular events translate into changes in behavior in the animal.
Observation learning in day-old chicks using a one-trial passive avoidance learning paradigm

Abstract:We tested the hypothesis that day-old chicks, Gallus gallus domesticus, can learn to avoid an aversive stimulus if they observe the responses of another chick. In experiment 1, one of a pair of chicks (the actor) was allowed to peck at a bead coated in the bitter-tasting substance methylanthranilate (MeA), while we prevented the other chick (the observer) from pecking the bitter-tasting bead by separating the chicks with a piece of wire mesh. Both chicks avoided pecking at a similar but dry bead 0.5, 3 and 24 h after the observer chick saw the actor chick peck at an MeA-coated bead. By contrast, when the actor chick had pecked at a water-coated bead, both chicks continued to peck at a dry bead at 0.5, 3 and 24 h after training. Experiment 2 investigated whether observer chicks showed avoidance if they were prevented (by the insertion of an opaque barrier) from observing their companion pecking at the MeA-coated bead during either training or testing. Observer chicks that could not see their companion during training but could observe the actor chicks at test showed no subsequent avoidance whereas chicks that observed the actor chick at training, but not during testing, showed high levels of avoidance. Although the sensory cues (visual, auditory or olfactory) or types of behaviour (i.e. levels of pecking or head shaking) that the observer chick used to maintain avoidance remain unclear, the results show that chicks can learn about an aversive object by observing the responses of a conspecific. Abstract:Indirect evidence has implicated glutamate and gamma-amino butyric acid in memory formation for one-trial passive avoidance learning. We have further examined this by following the time course of glutamate and gamma-amino butyric acid release from slices prepared from the intermediate medial hyperstriatum ventrale of day-old chicks (Ross 1 Chunky) trained to avoid a bead covered in the aversant methylanthranilate. At various times after training, slices of left and right intermediate medial hyperstriatum ventrale were incubated in medium containing 50 mM potassium chloride and amino acid release was determined. Thirty minutes after training there was a bilateral increase in calcium-dependent glutamate release in slices from methylanthranilate-trained chicks compared to those trained to peck water. This increase was sustained until 1 h in the left hyperstriatum when an increase in calcium-dependent gamma-amino butyric acid release was also apparent. Glutamate uptake was also enhanced in left hyperstriatum (30 and 60 min) and in the right at 30 min. In the right intermediate medial hyperstriatum ventrale of methylanthranilate birds glutamate release was increased from 3 to 6.5 h and gamma-amino butyric acid at 6.5 h: a time that corresponded to the mobilization of a late process required if long-term memory was to be formed. These results confirm that the amino acids glutamate and gamma-amino butyric acid are released from the intermediate hyperstriatum ventrale in a calcium-dependent, neurotransmitter-like manner. Furthermore, changes in the release of these two amino acids accompany memory formation for a one-trial learning task in the day-old chick.

Abstract:To observe the expression of c-fos gene in brain tissue following experimental cranial injury of rats. Methods The Feeney’s free falling-body model was used to induce the brain injury. Immunohistochemistry and computer image analysis technology were used to evaluate the expression of c-fos gene in brain tissue after trauma. Results Two hours after the injury, c-fos-positive cells were markedly increased in the traumatic cerebral cortex, surround cortex and dentate gyrus. The c-fos gene expression was significantly different, among various parts of the brain and among different injury levels at the same part. Conclusion The expression of c-fos gene in rat’s brain is significantly increased following brain injury and the expression level is related to the injury level.

Abstract:The present study investigated the behavioral training-induced c-Fos expression in the nucleus basalis of Meynert (nbM) in differently aged rats. This study demonstrated that the c-Fos expression in nbM was significantly increased and the peak occurred at 2 h after dark-avoidance training. Although the increase of c-Fos expression was also observed after pseudotraining, the number of Fos-like immunoreactive neurons in pseudotrained rats was significantly less than that in dark-avoidance trained rats at each time-point. This result suggested that c-Fos expression might be involved in learning and memory processes. In addition, all the pseudotraining-, training- and memory arousing-induced c-Fos expression was decreased with increasing age, and the decrease was more notable in trained and memory aroused rats. This suggested that the total number of nbM neurons and/or the sensitivity of nbM neurons to experimental manipulations, especially learning and memory performance, might reduce during aging. Abstract:The expression of mRNA encoding luteinizing hormone receptor (LHR) and follicle-stimulating hormone receptor (FSHR) was studied in chicken gonads from day 10 of incubation to day 14 of posthatch by Northern hybridization and expression of each mRNA was compared between sexes. LHR mRNA levels in the left ovary increased from day 12 of incubation, reached a peak two days later and remained high thereafter. In contrast, those in the testes remained extremely low until day 18 of incubation but started increasing sharply from day 20 of incubation and reached peak levels at the posthatch day 7. FSHR mRNA levels of the gonads displayed similar changes between both sexes. The changes resembled those of LHR mRNA levels of the testis during the examined period. These results suggest that the earlier expression of LHR mRNA in the ovary is involved in earlier estrogen production and the later expression of LHR mRNA in the testis is involved in androgen production at the late stage of development in the chick. The later expression of FSHR mRNA may be associated with some gonadal functions in common to both sexes.

Abstract:Developmental changes in brain activation after pain stimulation and after passive movement of the hind paw were assessed by functional magnetic resonance imaging (fMRI) in pigs of postnatal ages 2, 4 and 6 months. Response patterns were correlated with histological maturation parameters. At 2 months, fMRI failed to detect brain activation after pain stimulation and revealed weak, but widespread activation after passive movement. At 4 months, strong reaction of numerous cortical areas on the contralateral side was seen after pain stimulation. Following passive movement, activation was weaker but more widespread, and the brainstem was also involved. By 6 months, cortical activation became more restricted to the contralateral sensory cortex and brainstem after pain stimulation and to the contralateral sensory and ipsilateral premotor and motor cortices after passive movement. Neocortical synaptophysin immunoreaction increased significantly between 2 and 4 months and slightly decreased by 6 months. The density of GABA-immunoreactive neurons and fibers significantly increased, reaching a maximum at 6 months. Our studies indicate that remodeling of synapses and development of inhibitory GABA neurons last until 6 months postnatally, when the fMRI response of the pig's brain also attains its mature adult pattern.

Abstract:Visual imprinting is a learning process whereby young animals come to prefer a visual stimulus after exposure to it (training). The available evidence indicates that the intermediate medial mesopallium (IMM) in the domestic chick forebrain is a site of memory formation during visual imprinting. We have studied the role of Src, an important non-receptor tyrosine kinase, in memory formation. Amounts of total Src (Total-Src) and its two phosphorylated forms, tyrosine-416 (activated, 416P-Src) and tyrosine-527 (inhibited, 527P-Src), were measured 1 and 24 h after training in the IMM and in a control brain region, the posterior pole of nidopallium (PPN). One hour after training, in the left IMM, we observed a positive correlation between the amount of 527P-Src and learning strength that was attributable to learning, and there was also a positive correlation between 416P-Src and learning strength that was attributable to a predisposition to learn readily. Twenty-four hours after training, the amount of Total-Src increased with learning strength in both the left and right IMM, and amount of 527P-Src increased with learning strength only in the left IMM; both correlations were attributable to learning. A further, negative, correlation between learning strength and 416P-Src/Total-Src in the left IMM reflected a predisposition to learn. No learning-related changes were found in the PPN control region. We suggest that there are two pools of Src; one of them in an active state and reflecting a predisposition to learn, and the second one in an inhibited condition, which increases as a result of learning. These two pools may represent two or more signaling pathways, namely, one pathway downstream of Src activated by tyrosine-416 phosphorylation and another upstream of Src, keeping the enzyme in an inactivated state via phosphorylation of tyrosine-527.

Abstract:Abstract Mice display signs of fear when neurons that express cFos during fear conditioning are artificially reactivated. This finding gave rise to the notion that cFos marks neurons that encode specific memories. Here we show that cFos expression patterns in the mouse dentate gyrus (DG) change dramatically from day to day in a water maze spatial learning paradigm, regardless of training level. Optogenetic inhibition of neurons that expressed cFos on the first training day affected performance days later, suggesting that these neurons continue to be important for spatial memory recall. The mechanism preventing repeated cFos expression in DG granule cells involves accumulation of ΔFosB, a long-lived splice variant of FosB. CA1 neurons, in contrast, repeatedly expressed cFos. Thus, cFos-expressing granule cells may encode new features being added to the internal representation during the last training session. This form of timestamping is thought to be required for the formation of episodic memories.

Abstract:Both positive feedback learning and negative feedback learning are essential for adapting and optimizing behavioral performance. There is increasing evidence in humans and animals that the ability of negative feedback learning emerges postnatally. Our work in rats, using a two-way active avoidance task (TWA) as an experimental paradigm for negative feedback learning, revealed that medial and lateral prefrontal regions of infant rats undergo dramatic synaptic reorganization during avoidance training, resulting in improved avoidance learning in adulthood. The aim of this study was to identify changes of cellular activation patterns during the course of training and in relation to infant pretraining. We applied a quantitative cellular imaging technique using the immunocytochemical detection of the activity marker early growth response protein 1 (Egr1) as a candidate contributing to learning-induced synaptic plasticity. We found region-specific cellular activity patterns, which indicate that during the acquisition phase, Egr1 expression is specifically elevated in cellular ensembles of the orbitofrontal, dorsal anterior cingulate and hippocampal CA1 region. During memory retrieval Egr1 expression is elevated in cellular ensembles of the dentate gyrus. Moreover, we, for the first time, show here that TWA training during infancy alters adult learning- and memory-related patterns of Egr1 expression in these brain regions. It is tempting to speculate that during infant learning, specific Egr1-expressing cellular ensembles are “tagged” representing long-term memory formation, and that these cell ensembles may be reactivated during adult learning.

Investigate c-Fos changes in genetically identified amygdala neurons after mild footshock stress

Abstract:The amygdala is a key brain region that processes stress-related inputs to reshape future behaviors. The lateral amygdala (LA), basolateral amygdala (BLA), and central amygdala (CeA) are important subregions that mediate different aspects of stress experiences from receiving sensory input to memory formation and behavioral responding. The principal neurons in these regions are glutamatergic pyramidal neurons, which are genetically separable into two subpopulations, protein phosphatase 1 regulatory subunit 1B-positive (Ppp1r1b, also known as DARPP-32) parvocellular neurons and R-spondin2-positive (Rspo2) magnocellular neurons. Recent studies show that these two subpopulations of amygdala neurons differentially regulate appetitive versus aversive behaviors. The research goal of this study is to explore whether amygdala Ppp1r1b and Rspo2 neurons are transcriptionally activated by moderate stress experience, such that persistent cellular changes are made to influence future functional output of these two subtypes of neurons. To test transcriptional activation, we focused on c-Fos, one of the early genes that are transiently expressed in response to cellular stimulations to regulate downstream gene transcription. Moderate stress was introduced through brief footshocks, with mice without footshock as controls. Between shocked and control mice, we observed similar numbers of Ppp1r1b or Rspo2 neurons per unit area that expressed c-Fos, which was consistent across LA-BLA and CeA. Moreover, in LA-BLA, Ppp1r1b/c-Fos cells consistently outnumber Rspo2/c-Fos cells across treatment conditions, and the reverse is true in CeA. These results suggest that moderate stress experience is not sufficient to induce robust transcriptional alterations in the two key subpopulations of amygdala neurons, and Ppp1r1b versus Rspo2 neuron activities, as measured by c-Fos expression levels, show differential dominance in amygdala subregions.

Relativistic effects in three-body bound states.

Abstract:We formulate relativistic and nonrelativistic two-particle dynamics in such a manner that the two-body binding energies are the same for both. We then formulate and solve the relativistic Faddeev equations for a simple s-wave potential (Malfliet-Tjon V). The relativistic effects are small (about 3%) and reduce the three-body binding energy. The expectation value of the relativistic energy operator with the nonrelativistic wave function is a fairly good approximation, but approximate expressions involving expansions in powers of the momentum are shown to be quite unreliable.
Gene expression analysis of Arc mRNA as a neuronal cell activity marker in the hippocampus and amygdala in two-way active avoidance test in rats

Abstract:Introduction: Immediate early genes are widely used as neuronal cell activity markers in neuroscience. The present study investigated the relationship between their expression and abnormality in context fear conditioning. Methods: The learning test (two-way active avoidance test) was conducted in male rats administered with nonselective muscarinic antagonist scopolamine or selective dopamine D1-like receptor antagonist SCH 23390 at a dose level of 2.0 or 0.1mg/kg, respectively, for 4days. Expression levels of Arc and Fos mRNA in the hippocampus and amygdala were also evaluated on the second day of dosing by fluorescent in situ hybridization (FISH) and reverse transcriptase-quantitative polymerase chain reaction (RT-qPCR). Results: Scopolamine had no effect on avoidance rate, but decreased freezing in the two-way active avoidance test. SCH 23390 decreased avoidance rate and increased freezing. In FISH and RT-qPCR assays, scopolamine decreased Arc mRNA in the hippocampus and amygdala, whereas SCH 23390 increased Arc mRNA in the hippocampus. By contrast, scopolamine and SCH 23390 did not change Fos mRNA expression compared to Arc mRNA expression. Discussion: The results of the learning test indicated that scopolamine or SCH 23390 respectively inhibited fear or context conditioning in rats. Furthermore, alteration of the expression of Arc mRNA but not of Fos mRNA in the hippocampus and amygdala of the brain was suggested to be a sensitive neuronal cell activity marker to detect behavioral abnormality in the two-way active avoidance test.

Abstract:Positive and negative feedback learning is essential to optimize behavioral performance. We used the two-way active avoidance (TWA) task as an experimental paradigm for negative feedback learning with the aim to test the hypothesis that neuronal ensembles activate the activity-regulated cytoskeletal (Arc/Arg3.1) protein during different phases of avoidance learning and during retrieval. A variety of studies in humans and other animals revealed that the ability of aversive feedback learning emerges postnatally. Our previous findings demonstrated that rats, which as infants are not capable to learn an active avoidance strategy, show improved avoidance learning as adults. Based on these findings, we further tested the hypothesis that specific neuronal ensembles are “tagged” during infant TWA training and then reactivated during adult re-exposure to the same learning task. Using cellular imaging by immunocytochemical detection of Arc/Arg3.1, we observed that, compared to the untrained control group, (1) only in the dentate gyrus the density of Arc/Arg3.1-expressing neurons was elevated during the acquisition phase of TWA learning, and (2) this increase in Arc/Arg3.1-expressing neurons was not specific for the TWA learning task. With respect to the effects of infant TWA training we found that compared to the naïve non-pretrained group (a) the infant pretraining group displayed a higher density of Arc/Arg3.1-expressing neurons in the anterior cingulate cortex during acquisition on training day 1, and (b) the infant pretraining group displayed elevated density of Arc/Arg3.1-expressing neurons in the dentate gyrus during retrieval on test day 5. Correlation analysis for the acquisition phase revealed for the ACd that the animals which showed the highest number of avoidances and the fastest escape latencies displayed the highest density of Arc/Arg3.1-expressing neurons. Taken together, we are the first to use the synaptic plasticity protein Arc/Arg3.1 to label neuronal ensembles which are involved in different phases of active avoidance learning and whose activity patterns are changing in response to previous learning experience during infancy. Our results indicate (1) that, despite the inability to learn an active avoidance response in infancy, lasting memory traces are formed encoding the subtasks that are learned in infancy (e.g., the association of the CS and UCS, escape strategy), which are encoded in the infant brain by neuronal ensembles, which alter their synaptic connectivity via activation of specific synaptic plasticity proteins such as Arc/Arg3.1 and Egr1, and (2) that during adult training these memories can be retrieved by reactivating these neuronal ensembles and their synaptic circuits and thereby accelerate learning.

Abstract:An 8-year-old boy of Down’s syndrome was referred to the Endocrine clinic with weight loss of three kilograms, irritability, change in behavior and restlessness. At presentation the child was irritable and restless, unable to sit still. He had up slanting eyes, epicanthic folds, high arched palate and protruding tongue. His height was 128 cm (90th percentile by the Indian Council Medical Research charts)(3). The weight was 20 Kg [50th percentile by ICMR charts]. He had a visible goiter, a bruit over the thyroid, exophthalmos and mental retardation. The mental age was 3.4 years. The serum Total Thyronine (T3) was 320 ng/dL (normal 55-180 ng/dL), Serum Thyroxin (T4) was 16 (μg/dL (normal 5.5-11(μg/dL). The Serum Thyrotropin (Serum TSH) was undetectable by third generation Immuno Radiometric assay. The radioiodine I uptakes at two hours was 76% and at twenty-four hours was 91.6%. Thyroid scan using Tc99 showed a diffuse uptake. The thyroid antimicrosomal antibody titers were not raised. He was treated initially with carbimazole 15 mg/day and propranolol 20 mg/day. There was symptomatic improvement after six weeks of treatment. The antithyroid drugs were continued for a period of eighteen months with regular monitoring of the thyroid function. There was a gain in weight by 7 Kg from the pre treatment weight. The child responded with clinical and biochemical remission after 18 months of treatment with carbimazole. The child has maintained remission on subsequent follow up for six months.
Functionally refined encoding of threat memory by distinct populations of basal forebrain cholinergic projection neurons

Abstract:Neurons of the basal forebrain nucleus basalis and posterior substantia innominata (NBM/SI p ) comprise the major source of cholinergic input to the basolateral amygdala (BLA). Using a genetically-encoded acetylcholine (ACh) sensor in mice, we demonstrate that BLA-projecting cholinergic neurons can 'learn' the association between a naïve tone and a foot shock (training) and release ACh in the BLA in response to the conditioned tone 24h later (recall). In the NBM/SI p cholinergic neurons express the immediate early gene, Fos following both training and memory recall. Cholinergic neurons that express Fos following memory recall display increased intrinsic excitability. Chemogenetic silencing of these learning-activated cholinergic neurons prevents expression of the defensive behavior to the tone. In contrast, we show that NBM/SI p cholinergic neurons are not activated by an innately threatening stimulus (predator odor). Instead, VP/SI a cholinergic neurons are activated and contribute to defensive behaviors in response to predator odor, an innately threatening stimulus. Taken together, we find that distinct populations of cholinergic neurons are recruited to signal distinct aversive stimuli, demonstrating functionally refined organization of specific types of memory within the cholinergic basal forebrain of mice.
