Abstract:Background: Colletotrichum gloeosporioides is important plant pathogens on a wide range of plant hosts such as citrus causing pre- or post-harvest infections as anthracnose, post-bloom fruit drop, tearstain and stem-end rot on fruit, or wither-tip of twigs. Method: The optimization of growth conditions of this pathogen was performed (solid media, temperature, pH and water potential under laboratory experiments). Results: Our results revealed that the maximum radial growth of C. gloeosporioides was recorded on SDA medium. All isolates were able to grow on PDA at temperatures of 15 and 30°C (over 0.7cm/day). Optimal growth radial was recorded at pH 5, 6, 7 and 8. Similar responses were obtained with both salt types, but, in general, C. gloeosporioides was more tolerant to KCl than NaCl. Conclusion: Studies of cultural, morphological traits of the pathogen are prominent to understand the response of the pathogen in different environmental and nutritional conditions.

Abstract:There have been considerable advances in the understanding of species concepts in the genus Colletotrichum. This has lead to the need to carry out fresh surveys of Colletotrichum species associated with important hosts. Colletotrichum species are associated with Citrus plants as saprobes, important pre-harvest and post-harvest pathogens, as well as endophytes. In this study, a total of 312 Colletotrichum strains were isolated from leaves, shoots and fruits of cultivated Citrus and Fortunella species with or without disease symptoms across the main citrus production areas in China. The morphology of all strains were studied and multilocus (ACT, TUB2, CAL, GAPDH, GS, ITS) phylogeny established. Strains were from four important species complexes of Colletotrichum, namely C. gloeosporioides species complex, C. boninense species complex, C. acutatum species complex and a final group including C. truncatum, which was rare on Citrus species. The species belonging to the C. gloeosporioides species complex comprised C. gloeosporioides and C. fructicola, the C. boninense complex comprised C. karstii and a new species C. citricola and the C. acutatum complex included a new species, C. citri. The ability of strains to cause anthracnose on citrus fruits was tested by inoculation and strains of Colletotrichum gloeosporioides, C. fructicola and C. truncatum were pathogenic.
Effects of Environment Factors on Conidia Germination,appressoria Formation and Pathogenicity of the Persimmon Anthracnose Pathogens Colletotrichum Gloeosporioides

Abstract:The conidia germination and appressoria formation of Colletotrichum gloeosporioides, the pathogen of persimmon anthracnose, was tested on concave glass slides in different environment. As the concentration of glucose went up, rates of conidia germinating increased but the percentage of appressoria formation decreased, and the length of germ tubes augmented and the diameter of appressoria was steady; with time prolonging, the percentage of conidia germinating and appressoria formation all climbed. The conidia could germinate and form appressoria over wide pH range from 2. 0 to 9. 0; the optimal pH for conidia germination and appressoria formation was between pH 5. 0 and pH 6. 0. The pathogenicity test under different pH condition illuminated that the disease occurred on twigs at 23℃ within pH 4. 0 - 8. 0, also appeared at 17℃ and pH 6. 0 without spore masses on lesions of twigs surface, however, was absent at 17℃ and pH 5. 0 as well as at 15℃ and pH 5. 0 to pH 6. 0. The optimal temperature for the mycelium growth was about 25℃ , and higher temperature inhibited its growth. The scanning electron microscopy examination showed that the length of germ tube, growing longitudinally along the ridge and transversely cross the ridges and grooves, varied greatly, and the appressoria formation occurred at the bottom of groove or near.

Abstract:Colletotrichum horii causes serious anthracnose on persimmon (Diospyros kaki cv. Wuheshi). The taxon was previously identified as C. gloeosporioides and only recently revealed to be C. horii based on molecular data and comparisons to type specimens. This fungus provides an important new model for examining plant–fungus interactions in the perennial persimmon crop. In this paper, we review available information on C. horii, with special focus on symptoms, morphological characteristics, phylogenetic analysis, host-specificity and pathogenicity testing, infection processes, and the effects of environment factors on anthracnose development, including a discussion on future prospects.

Abstract:Colletotrichum gloeosporioides cause citrus anthracnose, is a major threat to citrus quality and fruit production worldwide. In the present study, different experiments were designed to check the potential of various chemicals and biocontrol agents (BCA) against C . gloeosporioides and characterize pathogen from core citrus growing regions of citrus in Punjab, Pakistan. Identification of different pathogens and phylogenetic relationship of Colletotrichum spp. was also performed to examine the relationship between isolates. The phylogenetic relationship determined across 16 C. gloeosporioides isolates showed five distinct clades, highlighting the importance of genetic variation among isolates. Pathogens were identified on the base of the internal transcribed spacers (ITS) region. During survey the disease incidence on fruits and leaves were identified and C . gloeosporioides found to be the most prevalent pathogens. Integrated management was done using BCA ( Trichoderma harzianum and T. viride ) and the synthetic combined fungicide (Cymoxanil+Mancozeb) under field conditions. Results revealed that, T. harzianum with T. viride and Cymoxanil+Mancozeb gave maximum inhibition against C . gloeosporioides and enhance plant performance under field conditions. In conclusion, the BCA ( T. harzianum and T. viride ) and the Cymoxanil+Mancozeb could be a better strategy to mitigate the heavy loss of citrus fruit due to anthracnose disease.

Abstract:Citrus are economically important fruit crops produced worldwide. Anthracnose, post bloom fruit drop, and stem-end rot of fruit caused by Colletotrichum species are diseases that seriously threatens citrus production in Turkey. Surveys conducted in 2016–2017 allowed the detection of typical symptoms of anthracnose of the visited fields (126 among 203) in the citrus production area of the Eastern Mediterranean region of Turkey. A total of 194 Colletotrichum strains were obtained from different citrus cultivars, including lemon (85), mandarin (73), orange (30), sour orange (1) and grapefruit (5) and different plant tissues (twigs (90), leaves (84) and fruits (20)). A multi-locus phylogeny was established based on four genomic loci (ITS, GAPDH, ACT, CHS1), and the morphological characters of the isolates were determined. Based on morphological characters and phylogenetic analysis, isolates were identified as belonging to C. gloeosporioides and C. karsti. Pathogenicity trials showed that, both species were able to colonise leaves, twigs, and fruits of the susceptible cultivar Interdonato of Citrus limon. Symptoms that developed on inoculated tissues were similar to those observed in the field on naturally infected citrus plants. Pathogenicity trials showed that C. gloeosporioides was more virulent on twigs and leaves, and C. karsti on fruits. To the best of our knowledge, this is the most comprehensive study on identification and characterization of C. gloeosporioides and C. karsti associated with citrus anthracnose in Turkey.

Abstract:SUMMARY Colletotrichum acutatum causes anthracnose on a wide range of hosts including woody and herbaceous crops, ornamentals, fruits and conifers. Almond, citrus, lupin, olive and strawberry are some of the crops in which C. acutatum diseases are economically important. With the application of molecular markers and diagnostic PCR over the last 10-15 years, C. acutatum was identified as a major pathogen on a number of hosts instead of or along with C. gloeosporioides. C. acutatum displays high levels of genotypic and phenotypic diversity. The global populations of this cosmopolitan pathogen fit into at least eight distinct molecular groups, A1-A8, which show some degree of correlation with the morphological characteristics and varying patterns of host association and geographical distribution. The pathogen has complex epidemiology, exhibiting pathogenic and non-pathogenic lifestyles on target hosts, non-target crops and weeds. C. acutatum populations also show pathogenic variability and cross-infection potential in relation to a number of hosts. Molecular genetic tools are being developed to investigate the pathogenicity mechanisms of this key pathogen. This article mainly focuses on the global population diversity in C. acutatum, pathogen epidemiology and diagnosis, host colonization processes, and the development of tools for the identification and analysis of genes associated with pathogenicity. Background information on the pathogen origin, host range, disease symptoms and disease management strategies is also provided.
Population Structure and Genetic Diversity of Colletotrichum Gloeosporioides on Citrus in China

Abstract:To analyze the genetic structure and genetic diversity of Colletotrichum gloeosporioides as the dominant Colletotrichum species on Citrus, the Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) genetic diversity, including 63 strains isolated and selected from 8 different sites and 5 different citrus species, was studied. A total of 19 GAPDH haplotypes were identified by genetic analysis, and the main haplotype (haplotype 5) was distributed in 28 isolates, mainly from Citrus unchiu Hort. ex Tanaka (WG) and Citrus reticulata Blanco cv. Succosa (BDZ) in Huangyan (HY), Linhai (LH), and Jiande (JD) of Zhejiang province, and Mashui tangerine (MSJ) in Mengshan of Guangxi province (GX). Using the genetic differentiation index, Fst revealed significant genetic differentiation in C. gloeosporioides populations between Jiangxi province (JXGZ) and GX, HY, LH, JD, and Chun’an (CA) of Zhejiang province, and also revealed slightly less genetic differentiation for C. gloeosporioides populations between HY, LH, JD, GX, Shaanxi province (SX), and Quzhou (QZ) of Zhejiang province. In addition, Fst revealed great genetic differentiation between the C. gloeosporioides populations obtained from MSJ and Citrus paradise Macf (PTY), and also revealed weak genetic differentiation between the C. gloeosporioides populations obtained from Citrus sinensis Osbeck (QC), WG, and BDZ. The AMOVA test showed that the levels of genetic differentiation for C. gloeosporioides were 19% and 81% among and within geographic populations, respectively. It also showed that C. gloeosporioides had levels of genetic differentiation among and within host populations of 12% and 88%, respectively. The Mantel test showed that the genetic distance was not linearly correlated with geographical distance and the haplotype phylogenetic analysis showed that C. gloeosporioides from different regions and hosts were scattered in the phylogenetic tree, implying that the genetic differentiation was independent of host variety and geographical origin. We speculated that genetic differentiation may be mainly due to gene mutation, gene recombination, or gene migration within native populations and has nothing to do with natural selection triggered by geography or host variety.
In vitro toxicity and field control effect of six fungicides to Colletotrichum gloeosporioides cau-sing citrus anthracnose

Abstract:In order to obtain fungicides with high efficiency , low toxicity , and less pollution in controlling citrus an-thracnose, which caused by Colletotrichum gloeosporioides Penz., the toxicity of pyraclostrobin , azoxystrobin, pro-chloraz, pyraclostrobin· me-tiram, thiophanate-methyl and iprovalicarb to a strain of C.gloeosporioides was respec-tively tested by mycelium growth rate and spore germination .The results showed that the effective dose to reduce my-celial growth of C.gloeosporioides by 50%(EC50 values) were 0.81, 2.58, 0.066, 4.97, 1555.15 and 237.98 μg · mL-1 , whereas the EC 50 values of spore germination to pyraclostrobin , azoxystrobin , prochloraz and pyraclostrobin· me-tiram were 0.93 , 14.34 , 4.47 and 5.29μg· mL-1 .Field experiments showed that 95%of above citrus anthrac-nose could be inhibited by azoxystrobin , prochloraz , iprovalicarb and pyraclostrobin· me-tiram with the recommended concentrations on citrus anthracnose .Conclusively , azoxystrobin , prochloraz , iprovalicarb and pyraclostrobin· me-ti-ram are effective in controlling citrus anthracnose .
Characterization of Colletotrichum Isolates Causing Colletotrichum Dieback of Citrus in California

Abstract:Dieback caused by Colletotrichum spp. is an emerging disease in California citrus groves. A large-scale survey with emphasis on seasonal variations of latent infections was conducted throughout citrus orchards in Fresno, Kern, and Tulare counties in 2019 and 2020. Latent infections on citrus leaves and twigs varied markedly between years. Isolates of Colletotrichum spp. were obtained from asymptomatic tissue, and two groups were formed based on colony and spore morphology. The morphological groups were further identified based on multigene sequence analysis using the DNA regions ITS1-5.8S-ITS2, TUB2, and GAPDH. Results revealed that isolates belong to two phylogenetic species, C. gloeosporioides and C. karstii, being C. karstii more frequently isolated. Representative isolates of each species were further selected and characterized based on the response of physiological variables to temperature. Both species had similar optimum growth temperatures but differed in maximum growth rates, with C. gloeosporioides exhibiting a greater growth rate than that of C. karstii on media. Pathogenicity tests on citrus trees demonstrated the ability of C. gloeosporioides and C. karstii to cause lesions on twigs and no differences in aggressiveness. A fungicide screening performed in this study determined that the DMI fungicides were the most effective in reducing the mycelial growth of C. gloeosporioides and C. karstii. The QoI fungicides showed a remarkably inhibitory impact on spore germination of both species. On average, C. karstii was more sensitive to the DMI fungicides than C. gloeosporioides. The findings of this study provide new information to understand the Colletotrichum dieback of citrus.
Genomic and Transcriptomic Characterization of Alternaria alternata during Infection

Abstract:Host-pathogen interactions are the result of the continuously evolving dynamics of the genomic interphases between pathogens and the host plants. Alternaria brown spot (ABS) caused by the pathogen Alternaria alternata is a serious threat to tangerine production. Although recent studies have made significant advances in the characterization of A. alternata virulence factors, a gap exists in the regulation of virulent genes throughout the course of A. alternata infection on host plants. To gain a better understanding of the dynamic defense transcriptome in Alternaria alternata during Infection, we performed a comparative transcriptome approach. After inoculation on citrus, we found that 2142, 1964, 2359 genes were up-regulated, and 1948, 1434, 1996 genes were down-regulated at 12 hours-post-inoculation (hpi), 24 hpi and 48 hpi, respectively. Among these genes, 1333 genes were up-regulated at three time points, and 1054 genes were down-regulated, indicating that most of the differentially expressed genes at the early stage of infection tended to remain differentially expressed at the later stage of infection. In addition to the genes that are known to be part of the infection network in plant-pathogen interactions, many novel genes related to plant-pathogen interaction were identified. Interestingly, our results indicate that A. alternata is able to rapidly alter its gene expression pattern during infection process, which is vital for the successful colonization of the pathogen. Moreover, this rapid alteration of gene expression is likely to be an adaptive mechanism, enabling the pathogen to quickly respond to any changes in the environment and adapt to the host's defense system. This ability to modify gene expression quickly in the face of environmental changes could play a critical role in the successful establishment of infection. RT-qPCR analysis confirmed that the expression pattern of nine randomly selected genes from the peroxisome pathway were consistent with the RNA-seq data. Our study provided a comprehensive study of the expression of genes during A. alternata infection of citrus, which may facilitate the understanding of host-plant interactions in A. alternata.
[Characterization of a bacterial biocontrol strain 1404 and its efficacy in controlling postharvest citrus anthracnose]

Abstract:Objective: Anthracnose caused by Colletotrichum gloeosporioides (Penz.) Sacc. is a main disease in citrus production. To develop an effective biocontrol measure against citrus postharvest anthracnose, we screened antagonistic microbes and obtained a bacterial strain 1404 from the rhizospheric soil of chili plants in Nanning city, Guangxi, China. The objectives of the present study were to: (1) identify and characterize the antagonistic bacterium; and (2) to evaluate the efficacy of the antagonistic strain in controlling citrus postharvest anthracnose disease. Methods: Strain 1404 was identified by comparing its 16S rDNA sequence with related bacteria from GenBank database, as well as analyzing its morphological, physiological and biochemical characters. The antagonistic stability of the strain 1404 was determined by continuously transferring it on artificial media. The effect of the strain on suppressing citrus anthracnose at postharvest stage was tested by stab inoculation method. Results: The 16S rDNA of strain 1404 was amplified with primers PF1 (5'-AGAGTTTGATCATGGCTCAG-3') and PR1 (5'-TACGGTTACCTTGTTACGACTT-3') and its sequence submitted to GenBank (accession number: GU361113). Strain 1404 clustered with the GenBank-derived Brevibacillus brevis strains in the 16S-rDNA-sequence-based phylogenetic tree at 100% bootstrap level. The morphological traits, physiological and biochemical characters of strain 1404 agreed with that of Brevibacillus brevis. Less change in the suppressive ability of antagonist against growth of Colletotrichum gloeosporioides was observed during four continuous transfers on artificial media. The average control efficacy of the strain was 64. 9 % against the disease 20 days after the antagonist application. Conclusion: Strain 1404 was identified as Brevibacillus brevis based on its morphological traits, phyiological and biochemical characters as well as 16S rDNA sequence analysis. The antagonist was approved to be a promising biocontrol agent. This is the first report of Brevibacillus brevis as an effective antagonist against citrus postharvest anthracnose disease.

Abstract:Colletotrichum spp. are important pathogens of citrus that cause dieback of branches and postharvest disease. Globally, several species of Colletotrichum have been identified as causing anthracnose of citrus. One hundred and sixty-eight Colletotrichum isolates were collected from anthracnose symptoms on citrus stems, leaves, and fruit from Victoria, New South Wales, and Queensland, and from State herbaria in Australia. Colletotrichum australianum sp. nov., C. fructicola, C. gloeosporioides, C. karstii, C. siamense, and C. theobromicola were identified using multi-gene phylogenetic analyses based on seven genomic loci (ITS, gapdh, act, tub2, ApMat, gs, and chs-1) in the gloeosporioides complex and five genomic loci (ITS, tub2, act, chs-1, and his3) in the boninense complex, as well as morphological characters. Several isolates pathogenic to chili (Capsicum annuum), previously identified as C. queenslandicum, formed a clade with the citrus isolates described here as C. australianum sp. nov. The spore shape and culture characteristics of the chili and citrus isolates of C. australianum were similar and differed from those of C. queenslandicum. This is the first report of C. theobromicola isolated from citrus and the first detection of C. karstii and C. siamense associated with citrus anthracnose in Australia.

Abstract:Information on the Colletotrichum species associated with Citrus anthracnose was lacking. Surveys were conducted from January 2012 to February 2013 on different citrus species and varieties orchards in the Cap-Bon region (Tunisia) to investigate the prevalence of Colletotrichum species. Typical anthracnose symptoms such as dieback and wither-tip of twigs were noticed. The fungi associated with anthracnose were isolated and studied. Morphological features (mycelial and sporulation characterization), molecular identification by using ITS1 and ITS2 primers were done along with pathogenicity test monosporic cultures. Eight Colletotrichum isolates were collected from infected fruits, leaves and twigs and were identified as Colletotrichum gloeosporioides (seven isolates) and Colletotrichum fructicola (one isolate). All eight Colletotrichum isolates were pathogenic on Thomson, Malti and Meski orange varieties, Lunari lemon variety and sweet lime Bergamot detached fruit and leaves. This investigation revealed the prevalence, for the first time of C. fructicola, on Citrus in Tunisia.

Abstract:The grapefruit (Citrus paradisi) is an important crop for citrus farming in Mexico. During June 2019, in a plantation in Iguala, Guerrero, Mexico, symptoms of anthracnose on grapefruit fruits were observed. Based on morphological and molecular characterisation, the fungus isolated from the anthracnose was identified as Colletotrichum gloeosporioides. Koch's postulates were verified and fulfilled. To our knowledge, this is the first report of C. gloeosporioides causing anthracnose on grapefruit fruits in Mexico.

Abstract:The effects of illumination,temperature,pH value,humidity, carbon source and nitrogen source on the mycelial growth,sporulation and conidial germination of Colletotrichum truncatum was primarily studied.The results showed that illumination have no much effect on the mycelial growth,sporulation and conidial germination of the fungi.Its mycelium grew between 15-35℃,and optimum temperature was at 30℃.It grew between pH 3-12,and optimum pH volue was between pH 5-7.The conidia germinated between 15-30℃,and optimum was at 25℃,It germinate between pH 3-10,and optimum was between 6-8.The lethal temperature of conidia and mycelium was at 56℃ and 60℃,respectively.The conidia could not germinate when the relative humidity below 97%.The pathogen could use a variety of carbohydrates and nitrogen sources,but for the mycelial growth,the optimum carbon source was sucrose,and the optimum nitrogen source was yeast.For the sporulation,the optimum carbon source was maltose,and the optimum nitrogen source was yeast.

Abstract:Chilli is one of the most significant spice crops cultivated with numerous culinary, medicinal, and industrial applications. Colletotrichum have a wide host range causing anthracnose disease in various crops whereas in chili anthracnose caused by Colletotrichum capsici is the major constraint in chili production. Under severe environmental conditions, the symptoms appear all over the plant including fruit which lead to complete crop damage. The particular disease has a huge economic impact on yield reduction and also debars the export criteria due to the less quality of chili crop. Management of the disease is difficult under field conditions and majority of the management strategies are highly dependent on the use of chemicals. Solely rely on chemicals is harmful to environmental and human health so integrated disease management approach can help to manage the disease at the initial stage. Detection and identification of pathogen play a major role in pathogen management. Early identification and detection of the pathogen based on conventional and molecular methods are very important to reduce yield loss under field conditions. The current review consists of a detailed understanding of the pathogen, identification, detection and diagnosis, and the most recent research updates.

Abstract:Anthracnose leaf spot and ripe rot disease, caused by Colletotrichum gloeosporioides in India, is of economic importance. The objective of the investigation was to isolate and characterise bacterial antagonists from the grape berry surface. Nine bacteria out of the 46 that were isolated significantly inhibited C. gloeosporioides in a dual culture assay. Six of these nine microorganisms demonstrated pathogen inhibition by producing extracellular volatile and non-volatile compounds. Bacteria SB5, SB13, and SB15 were chosen for the polyhouse experiment because they demonstrated the best disease control in a detached leaf assay. At 1 × 10 6 CFU/ml of bacterial concentration, both curative and preventive trials showed significant disease inhibition. Based on morphological and molecular characterisation the bacteria were identified as Bacillus species. All three strains could be effectively used for the biological control of C. gloeosporioides .

Abstract:This work aimed to evaluate the effect of the yeasts Candida albicans, Pichia guilliermondii, Rhodotorula glutinis, Saccharomyces cerevisiae and Zygoascus hellenicus in the control of anthracnose in cucumbers. The influence of volatile and non-volatile compounds on mycelial growth and the effect of cell suspension and culture filtrate on conidial germination were evaluated. In a greenhouse, yeasts were tested on cucumber cotyledons, one cotyledon being treated and the other not; afterwards both cotyledons were challenged with C. orbiculare. The severity of the disease in both cotyledons was assessed by determining biological control and resistance induction. The production of volatile compounds from the yeasts R. glutinis and C. albicans reduced the pathogen growth by 90.7 and 90.0%, respectively. The production of non-volatile compounds stimulated the pathogen development. Conidia germination was affected when exposed to cell suspension of all tested isolates, ranging from 43 to 75%. For the culture filtrate from Z. hellenicus, it reduced the conidia germination by 11.4%. In the in vivo test, on the treated cotyledon, the yeasts Z. hellenicus, R. glutinis and S. cerevisiae reduced the anthracnose severity by 52.5, 50.0 and 42.5%, respectively.
