Genetic diversity and construction of a core collection in Chinese Gastrodia elata germplasm resources

Abstract:Gastrodia elata BL (subtribe: Gastrodiinae Lindl; family: Orchidance) is a perennial herb with high medicinal value. In China, a substantial number of wild and cultivated G. elata germplasm resources have been preserved; however, many of them exhibit genetic mixing necessitating genetic differentiation for effective germplasm management. In this study, we developed 15 novel simple sequence repeats (SSRs) and applied them in combination with 7 existing SSRs to evaluate the genetic diversity and population structure of 743 G. elata accessions excluding 24 potentially redundant genotypes, from China. Through STRUCTURE analysis, 158 pure G. elata f. elata , 60 pure G. elata f. glauca , and 38 pure G. elata f. viridis accessions were identified, whereas, the remaining accessions were idenified as potential hybrids derived from diverse crossing parents. The G. elata hybrids exhibited the highest genetic diversity, followed by G. elata f. viridis and G. elata f. elata , whereas G. elata f. glauca exhibited the lowest diversity. A major genetic differentiation existed between G. elata f. glauca and the other G. elata types. The results of Mantel test revealed a positive correlation between genetic and geographic distances. However, substantial genetic exchanges or mixing were observed in the G. elata germplasm from varying geographical regions. Based on the genetic variation, a core collection comprising 297 G. elata individuals was constructed, which is a vital resource for conserving and breeding of this medicinal plant.
