Abstract:Background: Abnormalities of the mevalonate pathway, an important cellular metabolic pathway, are common in many diseases including cardiovascular disease. The mevalonate pathway related enzyme expressions in pressure overload-induced cardiac hypertrophy and associated diastolic dysfunction remains largely unknown. This study aims to investigate whether the expression of mevalonate pathway related enzyme is altered during the progression of cardiac hypertrophy and associated diastolic dysfunction induced by pressure overload. Methods: Male Sprague-Dawley (SD) rats were randomly divided into two groups: the suprarenal abdominal aortic coarctation (AAC) group and the sham group. Results: Histological and echocardiographic assessments showed that there was a significant cardiovascular remodeling in the AAC group compared with the sham group after 3 weeks post-operatively, and the left ventricular (LV) diastolic function was reduced at 8 and 14 weeks post-operatively in the AAC group, without any change in systolic function during the study. The tissue of the heart and the abdominal aorta proximal to the coarctation showed over-expression of several enzymes, including 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR), farnesyl diphosphate synthase (FDPS), farnesyltransferase-alpha (FNTA), farnesyltransferase-beta (FNTB), geranylgeranyltransferase type I (GGTase-I) and the activation of their downstream proteins was enhanced. Conclusions: AAC induced compensatory LV hypertrophy to decompensatory diastolic dysfunction, accompanied by altered expression of several key enzymes in the mevalonate pathway. Copyright (C) 2013 S. Role of Losartan and Carvedilol on in Extracellular Signal-Regulated Kinases in Myocardium of Spontaneously Hypertensive Rats

Abstract:OBJECTIVE To explore the role of losartan and carvedilol on extracellular signal-regulated kinase (ERK) and mitogen-actirated protein kinase phosphatase-1 (MKP-1) in the myocardium of spontaneously hypertensive rat (SHR). METHODS Twenty-one 14-week old SHRs were randomily divided into 3 groups, 7 rats each. One group was given with carvedilol 30 mg·kg~(-1)·d~(-1), one group was given with losartan 10 mg·kg~(-1)·d~(-1) for 8 weeks, while the SHR sham group and 7 Wistar Kyoto rats(WKY)as control group were routinely given with vehicle (0.5% carboxymethyl cellulose) only for equal time. Then the activity of ERK and the expression of MKP-1 mRNA in myocardium were studied by immuno-precipitation and RT-PCR, respectively. RESULTS ①After administration for 8 weeks, the blood pressure of the SHR group treated with carvedilol or losartan decreased significantly (P0.01). LVW/BW ratio in SHR with carvedilol or losatan treatment was lower than that in SHR sham group (carvedilol group, P0.01, losartan group, P0.05), and the reduction in hypertrophy with losartan treatment was more than with carvedilol (P0.05). ②After treatment in SHR, the activity of ERK in the group with carvedilol or losartan can treatment was lower than that in the SHR control (P0.01), while the expression of MKP-1 mRNA of them was higher (P0.05). CONCLUSION Both carvedilol and losartan can inhibit the activity of ERK, increase the expression of MKP-1 mRNA, and then relieve cardiacl hypertrophy.
Losartan Reduced Connexin43 Expression in Left Ventricular Myocardium of Spontaneously Hypertensive Rats

Abstract:Objective To assess the effect of angiotensin II type 1 (AT 1 ) receptor antagonist losartan on myocardium connexin43 (Cx43) gap junction (GJ) expression in spontaneously hypertensive rats (SHRs) and investigate possible mechanisms. Methods Sixteen 9-week-old male SHRs and 8 age-matched male Wistar-Kyoto (WKY) rats were included in this study. SHRs were randomly divided into two groups to receive losartan at 30 mg/(kg·d) by oral gavage once daily for 8 weeks (SHR-L) or vehicle (0.9% saline) to act as controls (SHR-V); WKY rats receiving vehicle for 8 weeks served as normotensive controls. At the end of the experiment, rats were sacrificed and the hearts were removed. Expressions of Cx43 and nuclear factor-kappaB p65 (NF-κB p65) proteins in all three groups were observed and further investigations on the effect of angiotensin II type 1 receptor antagonist losartan (30 mg/(kg·d), 8 weeks) on Cx43 expression were conducted with Western blot and immunohistochemistry. NF-κB p65 protein in nuclear extracts was determined by Western blot. Results Left ventricular (LV) hypertrophy was prominent in SHRs, Cx43 and NF-κB p65 protein expressions were obviously upregulated and Cx43 distribution was dispersed over the cell surface. Treatment with losarton reduced the over-expressions of Cx43 and NF-κB p65 in LV myocardium. The distribution of Cx43 gap junction also became much regular and confined to intercalated disk after losartan treatment. Conclusion Cx43 level was upregulated in LV myocardium of SHR during early stage of hypertrophy. Angiotensin II type 1 receptor antagonist losartan prevented Cx43 gap junction remodeling in hypertrophied left ventricles, possibly through the NF-κB pathway.
Regulation of Phospholamban and Sarcoplasmic Reticulum Ca2+-ATPase by Atorvastatin: Implication for Cardiac Hypertrophy

Abstract:Abnormal intracellular Ca2+ homeostasis in the myocardium has been suggested as the cause of cardiac hypertrophy, and this process can be prevented by the HMG-CoA reductase inhibitors, statins. In the present study, the effect of atorvastatin on left ventricular hypertrophy was investigated, and then whether the underlying mechanism was related to a defect in intracellular Ca2+ homeostasis explored. Twelve spontaneously hypertensive rats (SHR), at 8 weeks old, were used in this study, and received either distilled water or atorvastatin for ten weeks, with age-matched normotensive Wistar-Kyoto rats (WKY) used as controls. RT-PCR and western blotting were used to detect the mRNA and protein expressions of phospholamban (PLB) and sarcoplasmic reticulum Ca2+-ATPase (SERCA2a), respectively, and a colorimetric method used to examine the SERCA2a activity. Additionally, cardiac hypertrophic indices, such as the cardiosomatic ratio, left ventricular weight to body weight (LVW/BW) ratio and cardiomyocytes transverse diameter (TDM), together with the systolic blood pressure (SBP) and serum lipids levels were also examined. After ten weeks, significant decreases were observed in both the mRNA and protein expression levels of SERCA2a, as well as its activity, in the hypertrophied hearts of the SHR. The administration of atorvastatin to the same strains of rats effectively inhibited these decreases, and the above cardiac hypertrophie indices, as well as the SBP and serum lipids levels were significantly decreased. However, no significant changes in the expressions of PLB were observed in WKY, SHR and atorvastatin-treated SHR. These findings demonstrated that through regulation of the PLB and SERCA2a levels in the hearts of SHR, atorvastatin can prevent the cardiac hypertrophy caused due to pressure overload, which provides a relatively new insight into the mechanism of atorvastatin in the prevention of cardiac hypertrophy.
Effects of combination of irbesartan and perindopril on calcineurin expression and sarcoplasmic reticulum Ca 2+ -ATPase activity in rat cardiac pressure-overload hypertrophy

Abstract:Aim: To observe effects of angiotensin (Ang) II receptor antagonist (AT1) irbesartan and angiotensin-converting enzyme (ACE) inhibitor perindopril on rat myocardium calcineurin expression and sarcoplasmic reticulum Ca 2+ -ATPase activity in the model of pressure-overload cardiac hypertrophy. Methods: Forty male adult Sprague Dawley rats were divided into 5 groups. One group was treated by sham operation; four groups were myocardium hypertrophy cases caused by banding aortic above renal artery. Drugs were given one week after operation. Group 1: sham group, rats ( n =8) were gavaged with normal saline 2 ml/(kg·d) (ig); Group 2: control group, rats ( n =8) were treated with normal saline 2 ml/(kg·d) (ig); Group 3: rats ( n =8) were given perindopril 2 mg/(kg·d) (ig); Group 4: rats ( n =8) were treated with irbesartan 20 mg/(kg·d) (ig); Group 5: rats ( n =8) were given irbesartan 20 mg/(kg·d) plus perindopril 2 mg/(kg·d) (ig). Morphometric determination, calcineurin expression and sarcoplasmic reticulum Ca 2+ -ATPase activity were done at the end of 6 week of drug intervention. Expression of calcineurin in myocardium was detected by immunohistochemistry. Results: Left ventricular mass index (LVMI), transverse diameter of myocardial cell (TDM), calcineurin activity were remarkably decreased after drug intervention and this decrease was most remarkable in the combination drug therapy group. Sarcoplasmic reticulum Ca 2+ -ATPase activity was increased after drug intervention, especially in the combined drug therapy group. Calcineurin expression in myocardium was remarkably decreased after drug intervention. LVMI was positively correlated with TDM and calcineurin, negatively correlated with sarcoplasmic reticulum Ca 2+ -ATPase. Conclusion: These data suggest that irbesartan and perindopril inhibit cardiac hypertrophy through the increased activity of sarcoplasmic reticulum Ca 2+ -ATPase and decreased expression of calcineurin. Their combination had better effects on regressing of ventricular hypertrophy.

Abstract:Objective:To determine the action of tissue Ang Ⅱ levels on endothelin (ET) system.Method: Our present study observed the effect of ACEI-Captopril on plasma ET levels and ET concentractions,ET,ETA receptor mRNA gene expressions in left ventricular and aortic tissues of 14 week and 26 week SHR,which were given captopril in their drinking water at a dose of 100 mg/kg per day for 12 weeks and compared with age- matched WKY rats. Result:Captopril did not reduce plasma ET levels,but ET concentracons,ET,ETA receptor mRNA gene expressions in left ventricular and aortic tissues were significantly inhibited by Captopril. Conclusion:Through inhibiting ET system,captopril may play a role in reducing blood pressure and regressing cardiac hypertrophy.
Effects of irbesartan and perindopril on pressure overload-induced cardiac hypertrophy in rats

Abstract:AIM: To observe the effects of irbesartan and perindopril on pressure-overload cardiac hypertrophy in rats. METHODS: 40 male adult Sprague Dawley rats were divided into 5 groups. One was sham operation group, other four were aortic banding groups. One week after operation, all rats were gavaged with normal saline, perindopril, irbesartan or combination of perindopril and irbesartan. Morphometric determination, calcineurin (CaN) expression, CaN and sarcoplasmic reticulum (SR) Ca 2+-ATPase activity were performed at the end of 6 weeks of drug intervention. RESULTS: Left ventricular mass index (LVMI), transverse diameter of myocardical cell (TDM), CaN activity were remarkably decreased after drug intervention and this decrease was most remarkable in the combination group. SR Ca 2+-ATPase activity increased after drug intervention, especially in the combination group. CaN expression in myocardium were remarkably decreased after drug intervention. LVMI was positively correlated with TDM and CaN, negatively correlated with SR Ca 2+-ATPase. CONCLUSION: Both irbesartan and perindopril decrease CaN activity, increase SR Ca 2+-ATPase activity and combination of them has synergic effects on regressing of ventricular hypertrophy.
Inhibition of Farnesyl Pyrophosphate Synthase Improves Pressure Overload Induced Chronic Cardiac Remodeling

Abstract:Farnesyl pyrophosphate synthase (FPPS) is a key enzyme in the mevalonate pathway. In our previous studies, we find that inhibition of FPPS attenuates angiotensin II-induced cardiac hypertrophy and fibrosis by suppressing RhoA while FPPS and Ras are up-regulated in pressure overload rats. In this study, we evaluate the effects and mechanisms of FPPS inhibition in pressure overload mice. Male FPPS-small interfering RNA (SiRNA) transgenic (Tg) mice and non-transgenic littermate control (NLC) were randomly divided into suprarenal abdominal aortic constriction (AAC) group and sham operation group. 12 weeks following AAC, mice were sacrificed by cervical dislocation. Histological and echocardiographic assessments showed that inhibition of FPPS improved chronic cardiac remodeling which was induced by AAC. The reductions of Ras farnesylation and GTP-Ras, as well as their downstream extracellular signal-related kinases 1/2 (ERK1/2) expression were observed in the heart of Tg-AAC mice compared with NLC-AAC mice, along with the reduction of fetal gene expression. We provide here important experimental evidence that inhibition of FPPS improves AAC induced chronic cardiac remodeling and fibrosis by the reduction of farnesylated Ras and the downregulation of Ras-ERK1/2 pathway.
Aliskiren Ameliorates Pressure Overload-Induced Heart Hypertrophy and Fibrosis in Mice

Abstract:Aliskiren (ALK) is a renin inhibitor that has been used in the treatment of hypertension. The aim of this study was to determine whether ALK could ameliorate pressure overload-induced heart hypertrophy and fibrosis, and to elucidate the mechanisms of action. Transverse aortic constriction (TAC) was performed in mice to induce heart pressure overload. ALK (150 mg·kg−1·d−1, po), the autophagy inhibitor 3-methyladenine (10 mg·kg−1 per week, ip) or the PKCβI inhibitor LY333531 (1 mg·kg−1·d-1, po) was administered to the mice for 4 weeks. Heart hypertrophy, fibrosis and function were evaluated based on echocardiography, histological and biochemical measurements. Mechanically stretched cardiomyocytes of rats were used for in vitro experiments. The levels of signaling proteins were measured using Western blotting, while the expression of the relevant genes was analyzed using real-time QRT-PCR. TAC induced marked heart hypertrophy and fibrosis, accompanied by high levels of Ang II in plasma and heart, and by PKCβI/α and ERK1/2 phosphorylation in heart. Meanwhile, TAC induced autophagic responses in heart, i.e. increases in autophagic structures, expression of Atg5 and Atg16 L1 mRNAs and LC3-II and Beclin-1 proteins. These pathological alterations in TAC-mice were significantly ameliorated or blocked by ALK administration. In TAC-mice, 3-methyladenine administration also ameliorated heart hypertrophy, fibrosis and dysfunction, while LY333531 administration inhibited ERK phosphorylation and autophagy in heart. In mechanically stretched cardiomyocytes, CGP53353 (a PKCβI inhibitor) prevented ERK phosphorylation and autophagic responses, while U0126 (an ERK inhibitor) blocked autophagic responses. ALK ameliorates heart hypertrophy, fibrosis and dysfunction in the mouse model in setting of chronic pressure overload, via suppressing Ang II-PKCβI-ERK1/2-regulated autophagy.
Effects Of Losartan On Pressure Overload-Induced Cardiac Gene Expression Profiling In Rats

Abstract:1. In the present study, the effects of losartan on myocardial gene expression changes following cardiac hypertrophy were investigated.2. Male Wistar rats were randomized to receive 5 or 30 mg/kg per day losartan (i.p.) 1 day after suprarenal abdominal aortic constriction. Two weeks later, cardiac morphology and function were recorded with echocardiography and mean arterial central pressure was measured using carotid catheters. Myocardial gene expression was assessed with cDNA microarrays.3. The ratios of left ventricular weights to bodyweights, the posterior thickness of the left ventricle and mean arterial central pressure were significantly increased by aortic constriction and attenuated by losartan in a dose-related manner. Genes in different functional categories were regulated in pressure overload-induced cardiac hypertrophy and the majority of changes in gene expression were inhibited by losartan in a dose-dependent manner.4. However, there were still some genes that were unaffected by losartan, even at a higher dose. In contrast, losartan, especially at a lower dose, was able to induce changes in the expression of several additional genes that were unregulated in simple aortic constriction.5. In conclusion, losartan is able to inhibit pressure overload-induced cardiac hypertrophy, as well as the majority of pressure overload-related changes in gene expression. The genes that remained unaffected or those that were additionally induced by losartan are likely to be new targets for investigation or therapy.
Attenuation of changes in sarcoplasmic reticular gene expression in cardiac hypertrophy by propranolol and verapamil

Abstract:The effects of propranolol and verapamil on contractile dysfunction, subcellular remodeling and changes in gene expression in cardiac hypertrophy due to pressure overload were examined. Rats were subjected to banding of the abdominal aorta and then treated with either propranolol (10 mg/kg daily), verapamil (5 mg/kg daily) or vehicle for 8 weeks after the surgery. Depression of the left ventricular function in the hypertrophied heart was associated with decreases in myofibrillar and myosin Ca2+ ATPase activities as well as Ca2+-pump and Ca2+-release activities of the sarcoplasmic reticulum (SR). The level of alpha-myosin heavy chain (alpha-MHC) mRNA was decreased while that of beta-MHC mRNA was increased in the pressure-overloaded heart. The level of SR Ca2+-pump ATPase (SERCA2) mRNA and protein content for SERCA2 were decreased in the pressure overloaded heart. Treatment of the hypertrophied animals with propranolol or verapamil resulted in preservation of the left ventricular function and prevention of the subcellular alterations. Shift in the alpha- and beta-MHC mRNA levels and changes in the expression in SERCA2 mRNA level and protein content were also attenuated by these treatments. The results suggest that blockade of beta-adrenoceptors or voltage-dependent calcium channels normalizes the cardiac gene expression, prevents subcellular remodeling and thus attenuates heart dysfunction in rats with cardiac hypertrophy. Furthermore, both cardiac beta-adrenoceptors and L-type Ca2+-channels may be involved in the genesis of cardiac hypertrophy due to pressure overload.
An evaluation of the L5178Y TK+/- mouse lymphoma forward mutation assay using 42 chemicals.

Abstract:The L5178YTK+/- mouse lymphoma assay (MLA) has been utilized in several laboratories as a short-term test for chemical-induced forward mutation in cultured mammalian cells. In order to evaluate several technical modifications to the MLA, 42 chemicals representing 9 chemical classes were tested and the results were compared with those published elsewhere as well as with findings in a genetic toxicology test battery currently used in this laboratory. A positive response for the induction of TK-/- mutants was obtained for 26 chemicals. With the exception of p-aminophenol, all of these compounds were recognized mutagens or carcinogens and were representative of direct-acting and activation-dependent genotoxins. 16 compounds did not induce TK-/- mutants and among these were 5 compounds that were considered to be mutagens or carcinogens. A comparison of the results of this study with those published elsewhere revealed a strong agreement among findings for this test irrespective of minor technical variations. It was concluded that the MLA is a useful system for identifying chemical mutagens in mammalian cells and can serve as a valuable component in a genetic toxicology test battery.

Effect of Losartan and Captopril on Expression of Cardiac Angiotensin II AT1 Receptor Mrna in Rats Following Myocardial Infarction.

Abstract:AIM: To study the effects of losartan (Los) and captopril (Cap) treatment on expression of cardiac angiotensin II (Ang) AT(1) receptor mRNA in rats following myocardial infarction (MI). METHODS: Twenty-four rats with MI after coronary ligation for 7 d were randomly divided into 4 groups: A) Cap in drinking water, ad lib (2 g.L-1), B) ig Los 10 mg.kg(-1).d(-1), C) ig Los 30 mg.kg(-1).d(-1), and D) placebo for 6 wk. Sham-ligation rats (group E) served as controls. The levels of cardiac Ang AT(1) receptor mRNA expression in each group (n = 6) were examined by Dot blot using digoxigenin-labeled cDNA probes. RESULTS: Comparing with reflected peak areas of hybridization positive signals in group D (2640 +/- 201 mu m(2)), the expression of the cardiac Ang AT(1) receptor mRNA was much lower in the 3 treated groups (group A 1360 +/- 134 mu m(2), group B 1430 +/- 244 mu m(2), group C 1310 +/- 95 mu m(2)) (P < 0.01). But no difference was found between the 3 treated groups and sham-ligation group (1230 +/- 233 mu m(2)) (P > 0.05). CONCLUSION: Los and Cap attenuated the increase of cardiac Ang AT(1) receptor mRNA expression in rats following MI.
COMPARATIVE EFFECTS OF LOSARTAN AND CAPTOPRIL ON VENTRICULAR REMODELING AND FUNCTION AFTER MYOCARDIAL INFARCTION IN THE RAT

Abstract:OBJECTIVE:To determine the effects of losartan and captopril treatment on ventricular remodeling and function after myocardial infarction in rats.METHODS:Thirty-two rats with MI induced by coronary ligation after seven days were divided into four groups randomly and treated with captopril(2 g.liter-1, group A), losartan(10 mg.kg-1.d-1, group B), losartan(30 mg.kg-1.d-1, group C) and placebo (no drug, group D) for six weeks, respectively. Sham-operated rats(group E) served as controls. Echocardiography was performed at 1 and 7 weeks after MI, respectively.RESULTS:Compared with the results before treatment, both LV end-diastolic internal diameter and volume decreased significantly and the thickened posterior wall was reversed in group A, B and C; the peak early filling velocity decreased whereas the peak velocity was increased in these three groups. There are no significant difference among the three treated groups. However, LV end-diastolic internal diameter and the E/A were still increased, whereas the thickness of anterior wall and the peak velocity of LV outflow were decreased in group A, B, and C after treatment comparing with group E.CONCLUSION:Both angiotensin converting enzyme inhibitor and angiotensin II receptor antagonist can prevent the ventricular remodeling and improve the ventricular function.
Captopril Related Kidney Damage: Renal Afferent Arteriolar Responses to Angiotensin II and Inflammatory Signaling.

Abstract:Captopril can have nephrotoxic effects, which are largely attributed to accumulated renin and "escaped" angiotensin II (Ang II). Here we test whether angiotensin converting enzyme-1 (ACE1) inhibition damages kidneys via alteration of renal afferent arteriolar responses to Ang II and inflammatory signaling. C57Bl/6 mice were given vehicle or captopril (60 mg/kg per day) for four weeks. Hypertension was obtained by minipump supplying Ang II (400 ng/kg per min) during the second 2 weeks. We assessed kidney histology by periodic acid-Schiff (PAS) and Masson staining, glomerular filtration rate (GFR) by FITC-labeled inulin clearance, and responses to Ang II assessed in afferent arterioles in vitro. Moreover, arteriolar H2O2 and catalase, plasma renin were assayed by commercial kits, and mRNAs of renin receptor, transforming growth factor-β (TGF-β) and cyclooxygenase-2 (COX-2) in the renal cortex, mRNAs of angiotensin receptor-1 (AT1R) and AT2R in the preglomerular arterioles were detected by RT-qPCR. The results showed that, compared to vehicle, mice given captopril showed lowered blood pressure, reduced GFR, increased plasma renin, renal interstitial fibrosis and tubular epithelial vacuolar degeneration, increased expression of mRNAs of renal TGF-β and COX-2, decreased production of H2O2 and increased catalase activity in preglomerular arterioles and enhanced afferent arteriolar Ang II contractions. The latter were blunted by incubation with H2O2. The mRNAs of renal microvascular AT1R and AT2R remained unaffected by captopril. Ang II-infused mice showed increased blood pressure and reduced afferent arteriolar Ang II responses. Administration of captopril to the Ang II-infused mice normalized blood pressure, but not arteriolar Ang II responses. We conclude that inhibition of ACE1 enhances renal microvascular reactivity to Ang II and may enhance important inflammatory pathways.
Olmesartan attenuates cardiac remodeling through DLL4/Notch1 pathway activation in pressure overload mice.

Abstract:Background: Notch1 signaling controls the cardiac adaptation to stress. We therefore aimed to validate whether olmesartan, a widely used angiotensin II type 1 receptor blocker, ameliorates cardiac remodeling and dysfunction via delta-like ligand 4 (DLL4)/Notch1 pathway in mice with chronic pressure overload. Methods: Cardiac pressure overload was produced by transverse aortic constriction (TAC). A total of 35 wide-type C57BL/6J mice were randomly divided into sham group, TAC group, TAC + olmesartan group, and TAC + olmsartan + DAPT group (DAPT: gamma-secretase inhibitor, Notch signaling inhibitor). Saline (10 mg.kg(-1).d(-1)) or the same volume of olmesartan liquor (3 mg.kg(-1).d(-1)) was administered by gavage, and DAPT (10 mu mole.kg(-1).d(-1)) by peritoneal injection. After 28 days of treatment, cardiac hemodynamics, echocardiography, and histology were evaluated, followed by quantitative polymerase chain reaction of fetal gene (ANP and SAA) expression. Notch1-related proteins and ERK1/2 were examined by western blot, and the serum level of angiotensin II was determined by means of enzyme-linked immunosorbent assay kits. Results: Persistent pressure overload-induced left ventricular hypertrophy, dysfunction, fibrosis, and microcirculation dysfunction, together with the upregulation of angiotensin II, ERK1/2, and fetal gene expression. By the activation of DLL4/Notch1, olmesartan decreased left ventricular hypertrophy and fibrosis, preserved cardiac function, and improved capillary density and coronary perfusion. All these curative effects were suppressed by pharmacological blockade of Notch signaling with DAPT. Conclusions: Our findings identify a heretofore unknown pharmacological mechanism that olmesartan improves cardiac remodeling and function via DLL4/Notch1 pathway activation in mice with chronic pressure overload, which may present a new therapeutic target for hypertension.
Renal Denervation Significantly Attenuates Cardiorenal Fibrosis in Rats with Sustained Pressure Overload.

Abstract:To investigate the effects of renal denervation (RDN) on comprehensive cardiac and renal fibrosis in cardiomyopathy. Five weeks after successful transverse aortic constriction (TAC)-induced cardiomyopathy model building, Sprague-Dawley rats were randomly assigned to three groups: (1) RDN, (2) sham, and (3) losartan. Sham TAC rats served as control group. Compared with control, TAC groups showed a significant decrease in left ventricle ejection fraction and increase in ventricular septum thickness and left atrium diameter on echocardiography after 5 weeks. At 10 weeks post-TAC, venous blood samples were collected for fibrosis biochemical assay. Heart and kidney samples were also harvested for fibrosis pathophysiological detection. Cardiac and renal fibrosis quantity results showed that, compared with sham group, collagen volume fraction was significantly decreased in RDN group more than in losartan group. Biochemical parameters such as tumor necrosis factor alpha, aldosterone, and B-type natriuretic peptide levels as well as biomarkers for fibrosis such as procollagen type I N-terminal propeptide and procollagen type III N-terminal propeptide concentrations were significantly decreased in RDN group in comparison with sham. In addition, compared with sham group, left ventricle tissue protein expression of transforming growth factor-beta 1 and angiotensin II type I receptor was downregulated, and angiotensin-converting enzyme 2 was upregulated in RDN but not in losartan group. RDN significantly attenuates cardiac and renal fibrosis in cardiomyopathy. Differing from losartan, which only has angiotensin II type I receptor inhibition effects, RDN comprehensively suppresses cardiac and renal fibrogenesis through multiple pathways. The Effects of Different Angiotensin II Type 1 Receptor Blockers on the Regulation of the ACE-AngII-AT1 and ACE2-Ang(1–7)-Mas Axes in Pressure Overload-Induced Cardiac Remodeling in Male Mice

Abstract:Angiotensin II (AngII) type 1 receptor blockers (ARBs) have been effectively used in hypertension and cardiac remodeling. However, the differences among them are still unclear. We designed this study to examine and compare the effects of several ARBs widely used in clinics, including Olmesartan, Candesartan, Telmisartan, Losartan, Valsartan and Irbesartan, on the ACE-AngII-AT1 axis and the ACE2-Ang(1–7)-Mas axis during the development of cardiac remodeling after pressure overload. Although all of the six ARBs, attenuated the development of cardiac hypertrophy and heart failure induced by transverse aortic constriction (TAC) for 2 or 4weeks in the wild-type mice evaluated by echocardiography and hemodynamic measurements, the degree of attenuation by Olmesartan, Candesartan and Losartan tended to be larger than that of the other three drugs tested. Additionally, the degree of downregulation of the ACE-AngII-AT1 axis and upregulation of the ACE2-Ang(1–7)-Mas axis was higher in response to Olmesartan, Candesartan and Losartan administration in vivo and in vitro. Moreover, in angiotensinogen-knockdown mice, TAC-induced cardiac hypertrophy and heart failure were inhibited by Olmesartan, Candesartan and Losartan but not by Telmisartan, Valsartan and Irbesartan administration. Furthermore, only Olmesartan and Candesartan could downregulate the ACE-AngII-AT1 axis and upregulate the ACE2-Ang(1–7)-Mas axis in vitro. Our data suggest that Olmesartan, Candesartan and Losartan could effectively inhibit pressure overload-induced cardiac remodeling even when with knockdown of Ang II, possibly through upregulation of the expression of the ACE2-Ang(1–7)-Mas axis and downregulation of the expression of the ACE-AngII-AT1 axis. In contrast, Telmisartan, Valsartan and Irbesartan only played a role in the presence of AngII, and Losartan had no effect in the presence of AngII in vitro.

Abstract:Angiotensin II (AngII) type 1-receptor blockers (ARBs) have been effectively used not only in the treatment of hypertension but also in cardiac protection. However, whether and why there are differences in these effects still remain unclear. Here we compared the effects of five commonly used ARBs (Candesartan, Olmesartan, Losartan, Telmisartan and Valsartan) on pressure overload-induced cardiac hypertrophy in mice model. Pressure overload was produced by constriction of the transverse aorta (TAC) for 2 weeks, which induced a significant elevation of blood pressure; ARBs or saline was administered through a stomach tube; Cardiac hypertrophy was evaluated by transthoracic echocardiography, cardiac histology and specific gene expression analyses. Although all the five ARBs, which did not repress the elevation of left ventricular pressure after TAC, attenuated the development of cardiac hypertrophy in the wild-type mice, the degrees of regression by Candesartan, Olmesartan and Losartan tended to be larger than those by Telmisartan and Valsartan. Furthermore, in angiotensinogen-knockout mice lacking endogenous AngII, TAC-induced cardiac hypertrophy was regressed by Candesartan, Olmesartan and Losartan but not by Telmisartan and Valsartan administration. Our data suggest that Candesartan, Olmesartan and Losartan can effectively inhibit pressure overload-induced cardiac hypertrophy even in the absence of AngII, whereas Telmisartan and Valsartan could exert the inhibitory effects only in the presence of AngII.
