Kinetic Study of No Oxidation over Pt/Gamma-Al2o3 and La0.8sr0.2mno3

Abstract:Catalytic oxidation of NO with O-2 over Pt/gamma-Al2O3 and La0.8Sr0.2MnO3 was conducted with a tubular reactor in range of space time 0.090-0.720 second, inlet NO concentration 0.03-0.2 vol%, O-2 concentration 2-10 vol% and at 473 K. Under the conditions of 0.720 s space time, 0.05 vol% NO and 5 vol% O-2 concentrations, the steady-state conversion of NO oxidation over Pt/gamma-Al2O3 and La0.8Sr0.2MnO3 reached 22.4 and 90.5 %, respectively. A kinetic model consisting of 12 elementary reactions fits the experimental data adequately, indicated that the rate-limiting step of NO oxidation over Pt/gamma-Al2O3 was the reaction between gaseous NO with dissociately adsorbed O, and over La0.8Sr0.2MnO3 was the desorption of product NO2.
On The Chemical And Electrochemical One-Electron Reduction Of Peroxynitrous Acid

Abstract:Peroxynitrous acid was reduced by cathodic linear sweep voltammetry at a gold electrode and by iodide at pH 3.2 and 5.6. The cathodic reduction wave was identified by measuring its decay in time, which was the same as observed by optical spectroscopy. The iodide oxidation was followed by optical measurement of the triiodide formation. Both reductions show one-electron stoichiometry, with the product n(alpha)alpha = 0.23 +/- 0.04 from the electrochemical experiments, in which alpha is the transfer coefficient and n(alpha) the number of electrons transferred, and an diiodine yield of ca. 0.5 equiv per equivalent of peroxynitrous acid. The voltammetric reduction was irreversible up to scan rates of 80 V s(-1). Both reductions were pH independent in the range studied. The voltammetric reduction is most likely an irreversible elemental reaction followed by a chemical decay that cannot be observed directly. Because of the pH independence, we conclude that both reductions have a common short-lived intermediate, namely [HOONO](.-). We estimate the electrode potential of the likely ONOOH/ONOOH.- couple to be larger than 1 V. The commonly used electrode potential Edegrees(ONOOH, H+/NO2., H2O) does not describe the chemistry of peroxynitrous acid.

Abstract:Significance: Peroxynitrite is a short-lived intracellular redox signaling species that is closely related to a number of diseases. Current detection methods for this anion lack the selectivity over other redox-active species present in the cell. In this report the authors demonstrate the selective detection of peroxynitrite by using an amphiphilic block copolymer through an in situ dioxirane oxidation–fragmentation strategy. Comment: The reaction mechanism involves: 1) in situ dioxirane formation by reaction between a trifluoromethyl ketone and peroxynitrite; 2) intramolecular oxygen atom transfer by oxidizing the electron-rich arene; 3) the resulting peroxide undergoes fragmentation, releasing a hydrophilic carboxylic acid group that causes the self-assembled structures to dissolve. This chemistry enables selective detection of peroxynitrite over a number of other redox-active interferents such as hydrogen peroxide. Scavenging Effects of Natural Phenols on Oxidizing Intermediates of Peroxynitrite.

Abstract:Most of peroxynitrite (ONOO-/ONOOH) is formed via the diffusion-limited reaction between nitric oxide and superoxide. In biological systems, the decomposition of ONOO- yields 30-35% of carbonate radical (CO3*-) and nitrogen dioxide (NO2*), which are strongly oxidizing intermediates and are suggested to take a part of the responsibility for the toxicity of nitric oxide (NO*) or ONOO-. Therefore, the current study focuses on the scavenging activities of phenols toward CO3*- and NO2* to protect biomolecules from damage caused by NO* or ONOO- using the technique of pulse radiolysis. From the build-up kinetic of the phenoxyl radicals and the decay kinetic of CO3*- radical, the rate constants of scavenging reactions were determined to be 1.9-3.4 x 10(8) M(-1) x s(-1) and 0.11-1.9 x 10(8) M(-1) x s(-1) for CO3*- and NO2* respectively. The results indicated that the tested phenols are the efficient scavengers of CO3*- and NO2*.
Influence of the NO/NO 2 Ratio on Oxidation Product Distributions under High-NO Conditions

Abstract:Organic oxidation reactions in the atmosphere can be challenging to parse due to the large number of branching points within each molecule's reaction mechanism. This complexity can complicate the attribution of observed effects to a particular chemical pathway. In this study, we simplify the chemistry of atmospherically relevant systems, and particularly the role of NOx, by generating individual alkoxy radicals via alkyl nitrite photolysis (to limit the number of accessible reaction pathways) and measuring their product distributions under different NO/NO2 ratios. Known concentrations of NO in the classically "high-NO" range are maintained in the chamber, thereby constraining first-generation RO2 (peroxy radicals) to react nearly exclusively with NO. Products are measured in both the gas phase (with a proton-transfer reaction mass spectrometer) and the particle phase (with an aerosol mass spectrometer). We observe substantial differences in measured products under varying NO/NO2 ratios (from ∼0.1 to >1); along with modeling simulations using the Master Chemical Mechanism (MCM), these results suggest indirect effects of NOx chemistry beyond the commonly cited RO2 + NO reaction. Specifically, lower-NO/NO2 ratios foster higher concentrations of secondary OH, higher concentrations of peroxyacyl nitrates (PAN, an atmospheric reservoir species), and a more highly oxidized product distribution that results in more secondary organic aerosol (SOA). Abstract:Peroxynitrite (ONOO-), which is usually generated as a response of the immune system or in inflammatory processes, is a powerful oxidant species, while hemoglobin is an important peroxynitrite scavenger in vivo. In this work, we have studied the interaction between peroxynitrite and hemoglobin through an electrochemical method and UV-Vis spectroscopy. It is found that peroxynitrite, at a relatively high concentration level, may make the protein exhibit a concentration-dependent increase of its catalytic activity towards hydrogen peroxide, whereas peroxynitrite at low concentration will result in the slight decrease of the catalytic activity. Further studies reveal that the diversification of the enzymatic activity is ascribed to the different extent of tyrosine nitration and, accordingly, the spatial conformation of the protein.
Photocontrollable Peroxynitrite Generator Based on N-methyl-N-nitrosoaminophenol for Cellular Application.

Abstract:We designed and synthesized a photocontrollable peroxynitrite (ONOO(-)) generator, P-NAP, which has N-methyl-N-nitrosoaminophenol structure with four methyl groups introduced onto the benzene ring to block reaction of the photodecomposition product with ONOO(-) and to lower the semiquinoneimine's redox potential. The semiquinoneimine intermediate generated by photoinduced release of nitric oxide (NO) reduces dissolved molecular oxygen to generate superoxide radical anion (O(2)(•-)), which reacts with NO to afford ONOO(-) under diffusion control (k = 6.7 × 10(9) M(-1) s(-1)). NO release from P-NAP under UV-A (330-380 nm) irradiation was confirmed by ESR spin trapping. Tyrosine nitration, characteristic of ONOO(-), was demonstrated by HPLC analysis of a photoirradiated aqueous solution of P-NAP and N-acetyl-l-tyrosine ethyl ester. ONOO(-) formation was confirmed with a ONOO(-)-specific fluorogenic probe, HKGreen-3, and compared with that from 3-(4-morpholinyl)sydnonimine hydrochloride (SIN-1), which is the most widely used ONOO(-) generator at present. The photoreaction of P-NAP was influenced by superoxide dismutase, indicating that generation of O(2)(•-) occurs before ONOO(-) formation. The quantum yield for formation of duroquinone, the main P-NAP photodecomposition product, was measured as 0.86 ± 0.07 at 334 nm with a potassium ferrioxalate actinometer. Generation of ONOO(-) from P-NAP in HCT-116 cells upon photoirradiation was successfully imaged with HKGreen-3A. This is the first example of a photocontrollable ONOO(-) donor applicable to cultured cells.

Abstract:Oxidation flow reactors that use low-pressure mercury lamps to produce hydroxyl (OH) radicals are an emerging technique for studying the oxidative aging of organic aerosols. Here, ozone (O3) is photolyzed at 254 nm to produce O(1D) radicals, which react with water vapor to produce OH. However, the need to use parts-per-million levels of O3 hinders the ability of oxidation flow reactors to simulate NOx-dependent secondary organic aerosol (SOA) formation pathways. Simple addition of nitric oxide (NO) results in fast conversion of NOx (NO + NO2) to nitric acid (HNO3), making it impossible to sustain NOx at levels that are sufficient to compete with hydroperoxy (HO2) radicals as a sink for organic peroxy (RO2) radicals. We developed a new method that is well suited to the characterization of NOx-dependent SOA formation pathways in oxidation flow reactors. NO and NO2 are produced via the reaction O(1D) + N2O → 2NO, followed by the reaction NO + O3 → NO2 + O2. Laboratory measurements coupled with photochemical model simulations suggest that O(1D) + N2O reactions can be used to systematically vary the relative branching ratio of RO2 + NO reactions relative to RO2 + HO2 and/or RO2 + RO2 reactions over a range of conditions relevant to atmospheric SOA formation. We demonstrate proof of concept using high-resolution time-of-flight chemical ionization mass spectrometer (HR-ToF-CIMS) measurements with nitrate (NO3−) reagent ion to detect gas-phase oxidation products of isoprene and α-pinene previously observed in NOx-influenced environments and in laboratory chamber experiments.

Apparent Kinetics Study on the Oxidation of p-Nitroethylbenzene Catalyzed by Zinc Tetraphenylporphyrin

Abstract:In the aerobic oxidation of p-nitroethylbenzene,it is found that bis(α-methyl-4-nitrobenzyl) peroxide as the key intermediate and p-nitroacetophenone as the main products.So the apparent kinetics was investigated in depth though the establishment of reaction model and rate equation of the oxidation of p-nitroethylbenzene catalyzed by zinc tetraphenylporphyrin(TPPZn) that base on the above compounds.Meanwhile,the apparent rate constants of two reactions were calculated according to the experimental data in the temperature range(110—150)℃.The results showed that the first step of oxidation of p-nitroethylbenzene to bis(α-methyl-4-nitrobenzyl) peroxide was a zero-order reaction with the apparent activation energy of 39.5 kJ/mol,and the second step of decomposition of bis(α-methyl-4-nitrobenzyl) peroxide to p-nitroacetophenone was a half-order reaction with the apparent activation energy of 44.6 kJ/mol.Comparing with the apparent rate constants k1 and k2 under same temperature,it was found that the k2 approximate equal to k1 at low temperatures,and the k2 greater then k1 at high temperatures.So,increasing temperature was favorable for the formation of p-nitroacetophenone and with a faster reaction rate by comparing apparent activation energies and reaction rate constants of two step reactions.

Abstract:The reaction product of nitric oxide and superoxide, peroxynitrite, is a potent biological oxidant. The most important oxidative protein modifications described for peroxynitrite are cysteine-thiol oxidation and tyrosine nitration. We have previously demonstrated that intrinsic heme-thiolate (P450)-dependent enzymatic catalysis increases the nitration of tyrosine 430 in prostacyclin synthase and results in loss of activity which contributes to endothelial dysfunction. We here report the sensitive peroxynitrite-dependent nitration of an over-expressed and partially purified human prostacyclin synthase (3.3 μM) with an EC50 value of 5 μM. Microsomal thiols in these preparations effectively compete for peroxynitrite and block the nitration of other proteins up to 50 μM peroxynitrite. Purified, recombinant PGIS showed a half-maximal nitration by 10 μM 3-morpholino sydnonimine (Sin-1) which increased in the presence of bicarbonate, and was only marginally induced by freely diffusing NO2-radicals generated by a peroxidase/nitrite/hydrogen peroxide system. Based on these observations, we would like to emphasize that prostacyclin synthase is among the most efficiently and sensitively nitrated proteins investigated by us so far. In the second part of the study, we identified two classes of peroxynitrite scavengers, blocking either peroxynitrite anion-mediated thiol oxidations or phenol/tyrosine nitrations by free radical mechanisms. Dithiopurines and dithiopyrimidines were highly effective in inhibiting both reaction types which could make this class of compounds interesting therapeutic tools. In the present work, we highlighted the impact of experimental conditions on the outcome of peroxynitrite-mediated nitrations. The limitations identified in this work need to be considered in the assessment of experimental data involving peroxynitrite.

Abstract:Nitric oxide (NO) is a widespread signaling molecule involved in the regulation of an impressive spectrum of diverse cellular functions. Superoxide anions (O-2) not only contribute to the localization of NO action by rapid inactivation, but also give rise to the formation of the potentially toxic species peroxynitrite (ONOO-) and other reactive nitrogen oxide species. The chemistry and biological effect of ONOO- depend on the relative rates of formation of NO and O-2. However, the simultaneous quantification of NO and O-2 has not been achieved yet due to their high rate of interaction, which is almost diffusion-controlled. A sensitive spectrophotometric assay was developed for the simultaneous quantification of NO and O-2 in aqueous solution that is based on the NO-induced oxidation of oxyhemoglobin (oxyHb) to methemoglobin and the O-2-mediated reduction of ferricytochrome c. Using a photodiode array photometer, spectral changes of either reaction were analyzed, and appropriate wavelengths were identified for the simultaneous monitoring of absorbance changes of the individual reactions. oxyHb oxidation was followed at 541.2 nm (isosbestic wavelength for the conversion of ferri- to ferrocytochrome c), and ferricytochrome c reduction was followed at 465 nm (wavelength at which absorbance changes during oxyHb to methemoglobin conversion were negligible), using 525 nm as the isosbestic point for both reactions. At final concentrations of 20 microM ferricytochrome c and 5 microM oxyHb, the molar extinction coefficients were determined to be epsilon465-525 = 7.3 mM-1 cm-1 and epsilon541.2-525 = 6.6 mM-1 cm-1, respectively. The rates of formation of either NO or O-2 determined with the combined assay were virtually identical to those measured with the classical oxyhemoglobin and cytochrome c assays, respectively. The assay was successfully adapted to either kinetic or end point determination in a cuvette or continuous on-line measurement of both radicals in a flow-through system. Maximal assay sensitivity was approximately 25 nM for NO and O-2. Cross-reactivity with ONOO- was controlled for by the presence of L-methionine. Generation of NO from the NO donor spermine diazeniumdiolate could be reliably quantified in the presence and absence of low, equimolar, and high flux rates of O-2. Likewise, O-2 enzymatically generated from hypoxanthine/xanthine oxidase could be specifically quantified with no difference in absolute rates in the presence or absence of concomitant NO generation at different flux rates. Nonenzymatic decomposition of 3-morpholinosydnonimine hydrochloride (100 microM) in phosphate buffer, pH 7.4 (37 degrees C), was found to be associated with almost stoichiometric production of NO and O-2 (1.24 microM NO/min and 1.12 microM O-2/min). Assay selectivity and applicability to biological systems were demonstrated in cultured endothelial cells and isolated aortic tissue using calcium ionophore and NADH for stimulation of NO and O-2 formation, respectively. Based on these data, a computer model was elaborated that successfully predicts the reaction of NO and O-2 with hemoprotein and may thus help to further elucidate these reactions. In conclusion, the nitric oxide/superoxide assay allows the specific, sensitive, and simultaneous detection of NO and O-2. The simulation model developed also allows the reliable prediction of the reaction between NO and O-2 as well as their kinetic interaction with other biomolecules. These new analytical tools will help to gain further insight into the physiological and pathophysiological significance of the formation of these radicals in cell homeostasis.
Profiling proteins involved in peroxynitrite homeostasis using ROS/RNS conditional proteomics

Abstract:Peroxynitrite (ONOO−), the product of the diffusion-controlled reaction of superoxide (O2−) with nitric oxide (NO), plays a crucial role in oxidative and nitrative stress and modulates key physiological processes such as redox signaling. While biological ONOO− is conventionally analyzed using 3-nitrotyrosine antibodies and fluorescent sensors, such probes lack specificity and sensitivity, making high-throughput and comprehensive profiling of ONOO−-associated proteins challenging. In this study, we used a conditional proteomics approach to investigate ONOO− homeostasis by identifying its protein neighbors in cells. We developed Peroxynitrite-responsive protein Labeling reagents (Porp-L) and, for the first time, discovered 2,6-dichlorophenol as an ideal moiety that can be selectively and rapidly activated by ONOO− for labeling of proximal proteins. The reaction of Porp-L with ONOO− generated several short-lived reactive intermediates that can modify Tyr, His, and Lys residues on the protein surface. We have demonstrated the Porp-L-based conditional proteomics in immune-stimulated macrophages, which indeed identified proteins known to be involved in the generation and modification of ONOO− and revealed the endoplasmic reticulum (ER) as a ONOO− hot spot. Moreover, we discovered a previously unknown role for Ero1a, an ER-resident protein, in the formation of ONOO−. Overall, Porp-L represent a promising research tool for advancing our understanding of the biological roles of ONOO−.

Overcoming the Oxygen Dilemma in Photoredox Catalysis: Near‐Infrared (NIR) Light‐Triggered Peroxynitrite Generation for Antibacterial Applications

Abstract:The peroxynitrite anion (ONOO- ) is closely associated with many diseases and the creation of ONOO- donors is an essential means of understanding its pathophysiological functions. However, it is challenging to develop ONOO- donors due to the difficulties in simultaneously producing highly reactive and short-lived nitric oxide (NO) and superoxide anion (O2 ⋅- ). Here, we report a novel strategy for constructing ONOO- donors by combining near-infrared (NIR)-mediated type I photosensitization and photoredox catalysis. The key design using a Nile blue analogue that can serve as both a type I photosensitizer and a metal-free photocatalyst. Intriguingly, the formation of O2 ⋅- via type I photosensitization avoids oxygen interference and instead activates nitrobenzofurazan-based NO donors via oxygen-tolerant NIR photoredox catalysis. The simultaneous release of O2 ⋅- and NO leads to ONOO- release, showing both antibacterial and antibiofilm activities.

Abstract:The oxidation kinetics of -isophorone (-IP) using molecular oxygen catalyzed by iron(III) acetylacetonate was investigated in a lab-scale agitator bubbling reactor. -IP was found to give keto-isophorone (KIP) and 4-hydroxy-3,5,5-trimethyl-2-cyclohexen-1-one (HIP) along with little isomerization product -isophorone (-IP). The results show that the oxidation reaction took place in the pseudo-first-order fast reaction regime. The experiment was conducted under the mass transfer reaction regime as the mass transfer resistances could not be easily eliminated. The intrinsic kinetics was obtained through apparent kinetics. The activation energy of oxidation of -IP to KIP is 70.5 +/- 4.1 kJ mol(-1), and the value of ln A(KIP) is 33.53 +/- 1.22. Meanwhile, the activation energy of oxidation of -IP to HIP is 86.4 +/- 5.4 kJ mol(-1) and the value of ln A(HIP) is 36.23 +/- 1.52, which could provide theoretical basis for industrial design, amplification of reactor, and the optimization of reaction.
