However, the fundamental limit for phase noise is scarcely discussed in the literature. In a typically off-axis phase microscope system, the phase noise is limited to a few milliradians using a moderate camera. Common-path QPMs offer much reduced phase noise compared to typical Mach-Zehnder-based systems. However, further scaling down the phase noise becomes difficult. Here we propose a high-sensitivity common-path QPM that promises to reduce the phase noise by a factor of 10 (assuming the mechanical noise is negligible). This is achieved by a specifically designed signal filter, leaving only the subtle phase fluctuations coming from the dynamics sample scattering. By working at photon shot-noise limited detection, we can magnify the subtle phase contrast which is proportional to the camera well depth. We expect this system to have the height sensitivity similar to an atomic force microcopy, while measuring biological structures with a full field of view in a single-shot. We plan to use this system to study cell dynamics, particularly lamellipodial height fluctuations as well as stiffer cell membrane fluctuations.

Abstract:The strong need in materials and biological science has prompted the development of high-speed quantitative phase imaging. However, for phase retrieval applying digital micromirror devices (DMDs), the accuracy of the retrieved phase will be disturbed by the DMD-induced aberrations. Here, we propose a phase retrieval method based on measuring and correcting errors caused by phase non-uniformity of the device. Using only four binary amplitude masks and corresponding diffraction intensities, the proposed method achieves rapid convergence and high-quality reconstruction. The experiments prove the practical feasibility for general samples and the effective improvement of the retrieved phase accuracy.
Single-exposure cepstrum phase-shifting in-line digital holography based on dual-CCD

Abstract:Limited by the property of charge-coupled device (CCD), the recording optical path of in-line digital holography is commonly used to take full advantage of the CCD sampling space, which is in order to improve the performance of digital holographic measurement. But the reconstructed image of in-line digital holography is overlapped by the direct wave and the conjugate image, which is hard to be separated. To achieve the real-time high-resolution measurement of in-line digital holography, the dual-CCD single-exposure cepstrum phase-shifting in-line digital holography is put forward, which is based on the single phase-shifting cepstrum technology. Then two key technical schemes of this technology, including the method of adjusting dual-CCD's mirroring overlapping position and the optical structure of dual-CCD single-exposure π/2 phase-shifting, are given. Finally, the experimental results show that, the dual-CCD single-exposure cepstrum phase-shifting in-line digital holography can solve the overlapping problem well. Moreover, the real-time recording and reconstruction of in-line digital holography are initially realized.

Abstract:We propose a novel common-path quantitative phase imaging (QPI) method based on a digital micromirror device (DMD). The DMD is placed in a plane conjugate to the objective back-aperture plane for the purpose of generating two plane waves that illuminate the sample. A pinhole is used in the detection arm to filter one of the beams after sample to create a reference beam. Additionally, a transmission-type liquid crystal device, placed at the objective back-aperture plane, eliminates the specular reflection noise arising from all the “off” state DMD micromirrors, which is common in all DMD-based illuminations. We have demonstrated high sensitivity QPI, which has a measured spatial and temporal noise of 4.92 nm and 2.16 nm, respectively. Experiments with calibrated polystyrene beads illustrate the desired phase measurement accuracy. In addition, we have measured the dynamic height maps of red blood cell membrane fluctuations, showing the efficacy of the proposed system for live cell imaging. Most importantly, the DMD grants the system convenience in varying the interference fringe period on the camera to easily satisfy the pixel sampling conditions. This feature also alleviates the pinhole alignment complexity. We envision that the proposed DMD-based common-path QPI system will allow for system miniaturization and automation for a broader adaption.

Abstract:In 1948, Dennis Gabor proposed the concept of holography, providing a pioneering solution to a quantitative description of the optical wavefront. After 75 years of development, holographic imaging has become a powerful tool for optical wavefront measurement and quantitative phase imaging. The emergence of this technology has given fresh energy to physics, biology, and materials science. Digital holography (DH) possesses the quantitative advantages of wide-field, non-contact, precise, and dynamic measurement capability for complex-waves. DH has unique capabilities for the propagation of optical fields by measuring light scattering with phase information. It offers quantitative visualization of the refractive index and thickness distribution of weak absorption samples, which plays a vital role in the pathophysiology of various diseases and the characterization of various materials. It provides a possibility to bridge the gap between the imaging and scattering disciplines. The propagation of wavefront is described by the complex amplitude. The complex-value in the complex-domain is reconstructed from the intensity-value measurement by camera in the real-domain. Here, we regard the process of holographic recording and reconstruction as a transformation between complex-domain and real-domain, and discuss the mathematics and physical principles of reconstruction. We review the DH in underlying principles, technical approaches, and the breadth of applications. We conclude with emerging challenges and opportunities based on combining holographic imaging with other methodologies that expand the scope and utility of holographic imaging even further. The multidisciplinary nature brings technology and application experts together in label-free cell biology, analytical chemistry, clinical sciences, wavefront sensing, and semiconductor production.

Abstract:High-speed quantitative phase and amplitude imaging methods have led to numerous biological discoveries. For general samples, phase retrieval from a single-diffraction pattern has been an algorithmic and experimental challenge. Here we present a quantitative phase and amplitude imaging method applying an efficient support constraint to yield a rapid algorithmic convergence due to the removal of the twin image and spatial shift ambiguities. Compared to previous complex-valued imaging, our method is lenslet-free and relies neither on assumption based on sample sparsity nor interferometric measurements. Our method provides a robust method for imaging in materials and biological science, while its rapid imaging capability will benefit the investigation of dynamical processes.

Abstract:Quantitative phase imaging needs of a holographic setup in order to retrieve the sample phase distribution. Often this new phase information is obtained at the cost of reducing the useful range in other parameters of the imaging system such as field of view or resolution. We devised an experimental setup that allows complex amplitude evaluation and phase image quantification of three-dimensional (3-D) samples in wide-field digital holographic microscopy. The basis is a common-path interferometric configuration that can be implemented in two configurations depending on the position and the basic period of the grating: static grating and windowed Fourier filtering method or with phase-shifting moving grating. Both configurations allow complex amplitude recovery and, thus, quantitative phase imaging of the sample under test. Moreover, by combining the basic setup with tilted illumination and time multiplexing, it is possible to generate a synthetic aperture enlargement that improves the final image resolution. Experimental results validated our concepts.

Abstract:Digital holographic microscopy (DHM) suffers from a limited refocus depth range due to the employed microscope objective. The relationship between the refocus depth range of the DHM system and its optical parameters is studied theoretically and experimentally, and a method is proposed that combines wavefront coding with bicubic interpolation and extrapolation iteration to extend the refocus depth range. For obtaining a uniform point spread function on the refocus plane, a propagator kernel in numerical reconstruction is multiplied by a numerical logarithmic phase mask, and a deviation of the refocus distance is made. To improve the resolution of the refocused image, the size of the hologram is doubly enlarged. This approach can extend the refocus depth range from 0.9 up to 1.5 mm. This work provides important guidance for high-resolution imaging with a large refocus depth range in DHM.

Abstract:Quantitative phase imaging by digital holographic microscopy (DHM) is a nondestructive and label-free technique that has been playing an indispensable role in the fields of science, technology, and biomedical imaging. The technique is competent in imaging and analyzing label-free living cells and investigating reflective surfaces. Herein, we introduce a new configuration of a wide field-of-view single-shot common-path off-axis reflective DHM for the quantitative phase imaging of biological cells that leverages several advantages, including being less-vibration sensitive to external perturbations due to its common-path configuration, also being compact in size, simple in optical design, highly stable, and cost-effective. A detailed description of the proposed DHM system, including its optical design, working principle, and capability for phase imaging, is presented. The applications of the proposed system are demonstrated through quantitative phase imaging results obtained from the reflective surface (USAF resolution test target) as well as transparent samples (living plant cells). The proposed system could find its applications in the investigation of several biological specimens and the optical metrology of micro-surfaces.

Abstract:Digital holography is a versatile three-dimensional imaging technique that has the ability to record the complex wave-front of an imaged object in two-dimensions and retrieve it in three-dimensions. Several technical challenges of digital holographic systems have been overcome by proposing single-shot acquisition and common-path configurations. However, the limited fiel-of-view (FOV) of digital holography is the most fundamental and technically challenging aspect of this technology. With this in mind, we have developed a digital holographic microscope (DHM) with a doubled FOV together with it leverages single-shot acquisition, common-path, and off-axis configuration and operates in the reflection mode. The double FOV is achieved by spatial frequency multiplexing of two different areas of the object beam by the use of a cube beam splitter. The common-path and off-axis configuration are obtained by employing a plate beam splitter just before the microscope objective. Several experiments are carried out, and the results are presented to demonstrate the validity and effectiveness of the proposed DHM for quantitative phase imaging of (semi) transparent and reflective objects. Based on the experimental results, the proposed microscope shows advanced performance in biomedical imaging as well as inspection of engineered surfaces with its simplicity, higher stability (temporal and mechanical), compactness, low cost, and most importantly double FOV capabilities.

Abstract:Extracting quantitative phase information has received increased interest in many fields where either phase imaging or structure retrieval is an issue, such as optical testing, bio-medical imaging and materials science. In the past couple of decades, digital holography (DH) has emerged as a front-runner for phase imaging by providing quantitative phase measurements of the wave field with high accuracy and in near real-time [1]. However, DH systems need a highly coherent light source, suffer phase aberration, ambiguity and unwrapping problems, and cannot offer the highest spatial resolution. Recently, however, direct phase retrieval from intensity measurements using the Transport-of-Intensity Equation (TIE) [2, 3] has gained increasing attention. A minimum of two measurements of the spatial intensity of the optical wave in closely spaced planes perpendicular to the direction of propagation are needed to reconstruct the spatial phase of the wave by solving a second-order differential equation, i.e., with a non-iterative deterministic algorithm. In this paper, these two quantitative phase imaging methods: DH and TIE are introduced and compared. Two samples: a regular array of micro-bumps fabricated on Si substrate based on laser induced non-ablative texturing and a refractive quartz microlens array from SUSS MicroOptics were tested by DH and TIE. The results were compared and the merits and limitations of each method are discussed.

Abstract:Digital Holography Microscopy (DHM) is a powerful tool for three-dimensional imaging of micro-objects, since it yields an intensity image, called hologram. This is interference pattern, records the intensity information and allow reconstruction of the phase shift introduced by objects. With the development of computer and fast Fourier transformation technology, DHM has been widely used in biological and material tissues, etc.. In this paper, DHM is used to measure the abrupt edges and surface of reflect objects which are calibrated. Phase compensation uses the physical configuration to compensate the phase aberration, such as Microscope Objective (MO), spherical aberration and other curvature introduced by specimen or instruments in set-up. We apply this method to obtain the pure holograms of objects. The holograms produce more accurate phase-contrast imaging and three-dimensional imaging. Angular Spectrum Propagation (ASP) approach is a strict diffraction integration, and widely used for many applications, such as short-distance propagation, and the off-axis system and tilted plane. Here, we achieve numerical reconstruction of the hologram with ASP under phase compensation condition in an off-axis system.First, we use the fast Fourier transformation for the hologram and get the frequency image. Then, filter out the spectrum about the image which we need. Finally, inverse Fourier transform of the filtered spectrum and get the complex amplitude information. The phase image and intensity image of specimen is obtained after calculating the reconstruction distance.

Abstract:In various multiphase flows, the characterization of particle dynamics is important in the understanding of the interaction between particles and the surrounding flows. Digital holography (DH) is a versatile 3D imaging technique, which has shown great advantages in quantitative analysis and nonintrusive diagnosis of various particle fields. This review focuses on the advances and applications of DH in multiphase reactive/nonreactive flows in the last two decades. First, the basic principles of DH are introduced, including its mathematical background and representative experimental configurations. Then, the image processing algorithms for hologram reconstruction and automatic focusing are summarized, along with the methods for separating overlapping particles and tracking moving particles. As a prevailing and powerful tool, the recent applications of deep learning in processing holographic images is also included in this review. Furthermore, the applications of DH in the characterization of particle dynamics in multiphase reactive/nonreactive flows are surveyed in detail. Lastly, the review concludes with a discussion on the technical limits of DH and provides insights into its promising future research directions.

Abstract:Fourier ptychographic microscopy (FPM), as an emerging computational imaging method, has been applied to quantitative phase imaging with resolution bypassing the physical limit of the detection objective. Due to the weak illumination intensity and long image acquisition time, the achieved imaging speed in current FPM methods is still low, making them unsuitable for real-time imaging applications. We propose and demonstrate a high-speed FPM method based on using laser illumination and digital micro-mirror devices for illumination angle scanning. In this new, to the best of our knowledge, FPM method, we realized quantitative phase imaging and intensity imaging at over 42 frames per second (fps) with around 1 µm lateral resolution. The quantitative phase images have revealed membrane height fluctuations of red blood cells with nanometer-scale sensitivity, while the intensity images have resolved subcellular features in stained cancer tissue slices.

Abstract:Quantitative phase microscopy (QPM) is a powerful tool for label-free and noninvasive imaging of transparent specimens. In this paper, we propose a novel QPM approach that utilizes deep learning to reconstruct accurately the phase image of transparent specimens from a defocus bright-field image. A U-net based model is used to learn the mapping relation from the defocus intensity image to the phase distribution of a sample. Both the off-axis hologram and defocused bright-field image are recorded in pair for thousands of virtual samples generated by using a spatial light modulator. After the network is trained with the above data set, the network can fast and accurately reconstruct the phase information through a defocus bright-field intensity image. We envisage that this method will be widely applied in life science and industrial detection.

Abstract:Accurate depiction of waves in temporal and spatial is essential to the investigation of interactions between physical objects and waves. Digital holography (DH) can perform quantitative analysis of wave–matter interactions. Full detector-bandwidth reconstruction can be realized based on in-line DH. But the overlapping of twin images strongly prevents quantitative analysis. For off-axis DH, the object wave and the detector bandwidth need to satisfy certain conditions to perform reconstruction accurately. Here, we present a reliable approach involving a coupled configuration for combining two in-line holograms and one off-axis hologram, using a rapidly converging iterative procedure based on two-plane coupled phase retrieval (TwPCPR) method. It realizes a fast-convergence holographic calculation method. High-resolution and full-field reconstruction by exploiting the full bandwidth are demonstrated for complex-amplitude reconstruction. Off-axis optimization phase provides an effective initial guess to avoid stagnation and minimize the required measurements of multi-plane phase retrieval. The proposed strategy works well for more extended samples without any prior assumptions of the objects including support, non-negative, sparse constraints, etc. It helps to enhance and empower applications in wavefront sensing, computational microscopy and biological tissue analysis.
Coded aperture correlation holographic microscope for single-shot quantitative phase imaging with extended field of view

Abstract:Recently, a method of recording holograms of coherently illuminated three-dimensional scene without two-wave interference was demonstrated. The method is an extension of the coded aperture correlation holography from incoherent to coherent illumination. Although this method is practical for some tasks, it is not capable of imaging phase objects, a capability that is an important benefit of coherent holography. The present work addresses this limitation by using the same type of coded phase masks in a modified Mach-Zehnder interferometer. We show that by several comparative parameters, the coded aperture-based phase imaging is superior to the equivalent open aperture-based method. As an additional merit of the coded aperture approach, a framework for increasing the system's field of view is formulated and demonstrated for both amplitude and phase objects. The combination of high sensitivity quantitative phase microscope with increased field of view in a single camera shot holographic apparatus, has immense potential to serve as the preferred tool for examination of biological tissues and micro-organisms.
