Abstract:The manipulation of unactivated aliphatic C–H bonds remains one of the most challenging tasks in synthetic chemistry. Direct hydrogen atom transfer (HAT) photocatalysis is an appealing approach to this goal. However, many methods are constrained due to low catalytic efficiency. Here we report the use of a Brønsted acid to enhance the efficiency of an inexpensive organic HAT photocatalyst, eosin Y. This strategy enables valuable transformations, including alkylation, heteroarylation and fluorination, of a wide array of unactivated C( sp 3 )–H bonds, using the alkane substrate as the limiting reagent. The process has been applied to the late-stage functionalization of natural products and pharmaceuticals to selectively form C–H-functionalized analogues. Experimental and computational mechanistic studies show that the HAT reactivity is significantly enhanced when the sp 3 oxygen atoms on eosin Y are protonated. The method has been shown to be general across different types of direct HAT photocatalysts, demonstrating its potential in native C–H bond functionalization.

Abstract:A simple, efficient method has been developed for the Co^(III)-H-catalyzed aerobic C-C bond cleavage of tertiary allylic alcohols to access ketones. This novel approach presents excellent chemoselectivity, good functional group compatibility, and high yields. This reaction occurs through a HAT-initiated peroxide intermediate, and an adjacent glycol-type diradical fragmentation process is recommended.

Abstract:Developing methods to directly transform C(sp3)‐H bonds is crucial in synthetic chemistry due to their prevalence in various organic compounds. While conventional protocols have largely relied on transition metal catalysis, recent advancements in organocatalysis, particularly with radical NHC catalysis have sparked interest in the direct functionalization of "inert" C(sp3)‐H bonds for cross C‐C coupling with carbonyl moieties. This strategy involves selective cleavage of C(sp3)‐H bonds to generate key carbon radicals, often achieved via hydrogen atom transfer (HAT) processes. By leveraging the bond dissociation energy (BDE) and polarity effects, HAT enables the rapid functionalization of diverse C(sp3)‐H substrates, such as ethers, amines, and alkanes. This mini‐review summarizes the progress in carbene organocatalytic functionalization of inert C(sp3)‐H bonds enabled by HAT processes, categorizing them into two sections: 1) C‐H functionalization involving acyl azolium intermediates; and 2) functionalization of C‐H bonds via reductive Breslow intermediates.

Abstract:Abstract Hydrogen atom abstraction from C(sp3)−H substrates for the generation of alkyl radical represents a desirable yet underexplored strategy in alkylation reaction since involving noble metal catalysts, stoichiometric oxidants, and limited scope are common drawbacks. Here we describe the synergistic utilization of photoredox and hydrogen atom transfer (HAT) catalysis to accomplish a general and practical functionalization of unactived C(sp3)−H centers, which features broad reaction scope, high functional group compatibility, and operational simplicity. A combination of validation experiments and density functional theory reveals that the N-centred radicals, generated from free N−H bond via photoredox catalyzed single-electron oxidation followed by deprotonation in a stepwise electron/proton transfer event, are the key intermediates that enable an intramolecular 1,5-HAT or intermolecular HAT process for nucleophilic carbon-centred radicals formation to achieve heteroarylation, alkylation, amination, cyanation, azidation, trifluoromethylthiolation, halogenation and deuteration. The practical value of this protocol was further demonstrated by the gram-scale synthesis and the late-stage functionalization of natural products and drug derivatives.

Abstract:Abstract Hydrogen atom abstraction (HAT) from C( sp 3 )–H bonds of naturally abundant alkanes for alkyl radical generation represents a promising yet underexplored strategy in the alkylation reaction designs since involving stoichiometric oxidants, excessive alkane loading, and limited scope are common drawbacks. Here we report a photo-induced and chemical oxidant-free cross-dehydrogenative coupling (CDC) between alkanes and heteroarenes using catalytic chloride and cobalt catalyst. Couplings of strong C( sp 3 )–H bond-containing substrates and complex heteroarenes, have been achieved with satisfactory yields. This dual catalytic platform features the in situ engendered chlorine radical for alkyl radical generation and exploits the cobaloxime catalyst to enable the hydrogen evolution for catalytic turnover. The practical value of this protocol was demonstrated by the gram-scale synthesis of alkylated heteroarene with merely 3 equiv. alkane loading.

Abstract:Hydrogen atom transfer (HAT) from a metal-hydride is a reliable and powerful method for functionalizing unsaturated C–C bonds in organic synthesis. Cobalt hydrides (Co–H) have garnered significant attention in this field, where the weak Co–H bonds are most commonly generated in a catalytic fashion through a mixture of stoichiometric amounts of peroxide oxidant and silane reductant. Here we show that the reverse process of HAT to an alkene, i.e. hydrogen atom abstraction of a C–H adjacent to a radical, can be leveraged to generate catalytically active Co–H species in a new application of shuttle catalysis coined shuttle HAT. This method obviates the need for stoichiometric reductant/oxidant mixtures thereby greatly simplifying the generation of Co–H under exceedingly mild reaction conditions. This approach opens the door for the introduction of functional handles (e.g., iodides) that were previously inaccessible through other catalytic approaches, and paves the way for new reagent design which incorporates this shuttle HAT platform. To demonstrate the generality of this shuttle HAT platform, five different reaction manifolds are shown, including the late-stage C(sp3) iodination of structurally diverse FDA approved drugs.

Abstract:A copper catalyzed aerobic formal addition of aliphatic C(sp3)–H bonds to alkenes via a phenyl radical mediated intermolecular HAT process is reported herein, in which the phenylhydrazine not only acts as a good HAT reagent precursor but also as an excellent hydrogen atom donor. This reaction affords an alternative method for the direct formal addition of the aliphatic C(sp3)–H bonds to alkenes utilizing air as the radical initiator without additional reductants at the ambient temperature. A variety of alkanes and electron‐deficient alkenes are successfully incorporated, furnishing a series of aliphatic C(sp3)–H formal addition products in 20‐93% yields. The preliminary mechanistic studies uncover that the 2‐tetrahydrofuranyl and phenyl radical are formed in this reaction.

Abstract:Modern photoredox catalysis has traditionally relied upon metal-to-ligand charge-transfer (MLCT) excitation of metal polypyridyl complexes for the utilization of light energy for the activation of organic substrates. Here, we demonstrate the catalytic application of ligand-to-metal charge-transfer (LMCT) excitation of cerium alkoxide complexes for the facile activation of alkanes utilizing abundant and inexpensive cerium trichloride as the catalyst. As demonstrated by cerium-catalyzed C-H amination and the alkylation of hydrocarbons, this reaction manifold has enabled the facile use of abundant alcohols as practical and selective hydrogen atom transfer (HAT) agents via the direct access of energetically challenging alkoxy radicals. Furthermore, the LMCT excitation event has been investigated through a series of spectroscopic experiments, revealing a rapid bond homolysis process and an effective production of alkoxy radicals, collectively ruling out the LMCT/homolysis event as the rate-determining step of this C-H functionalization.

Abstract:Here, we reported for the first time a mechanistically distinctive cobalt-catalyzed Markovnikov-type sequential semihydrogenation/hydrohydrazidation of aliphatic terminal alkynes in one pot. A cobalt hydride species was employed as two roles for both a unique metal-catalyzed Markovnikov-type insertion of the aliphatic terminal alkynes and then metal-catalyzed hydrogen atom transfer of alkenes. This operationally simple protocol exhibits excellent functional group tolerance and step economy. The hydrazone products could be easily transferred to various valuable amine derivatives.

Abstract:A synergetic catalytic system of organophotocatalyst 4CzIPN, thianthrene, and methanol was developed for visible‐lightinduced Giese reactions of various alkanes and alkenes, upon using alkoxy radicals as hydrogen atom transfer reagents. The reaction can be performed under mild conditions to provide the alkylated products in moderate to excellent yields, avoiding the use of any strong oxidants and transition metal reagents. Different from the known LMCT and PCET processes, the key to success of this approach is the in situ thianthrenation of methanol by thianthrene radical cation to trigger the generation of methoxy radical.

Abstract:Direct functionalization of alkenes with C(sp 3 )–H substrates offers unique opportunities for the rapid construction of pharmaceuticals and natural products. Although significant progress has been made over the past decades, the development of green, high step-economy methods to achieve these transformations under mild conditions without the need for pre-functionalization of C(sp 3 )–H bonds remains a substantial challenge. Therefore, the pursuit of such methodologies is highly desirable. Recently, the direct activation of C(sp 3 )–H bonds via photocatalytic hydrogen atom transfer (HAT), especially from unactivated alkanes, has shown great promise. Given the potential of this approach to generate a wide range of pharmaceutically relevant compounds, this review highlights the recent advancements in the direct functionalization of alkenes through photocatalytic HAT from C(sp 3 )–H compounds, as well as their applications in the synthesis and diversification of drugs, natural products, and bioactive molecules, aiming to provide medicinal chemists with a practical set of tools.

Abstract:Herein, a direct transformation of aliphatic C-H bonds to oxime ethers has been developed via light-promoted hydrogen atom transfer (HAT) in the absence of a photocatalyst. Singlet oxygen and chlorine radical are complementary C(sp3)-H bond cleaving agents in this reaction, enabling the extraction of hydrogen atoms from a diverse range of compounds, like cycloalkanes, ethers, amines, amides, and cyclic sulfides. This method excels in transforming common aliphatic C-H bonds into valuable oxime ethers featuring abundant chemical feedstocks, good functional group tolerance, and catalyst free conditions.
Copper-Mediated Hydrogen Atom Transfer Enabling General Catalytic Hydrofunctionalization of Olefins

Abstract:The renaissance of catalytic hydrogen atom transfer from a metal hydride (MHAT) offers advanced tools for practicing sophisticated radical chemistry on simple olefins. While 3d transition metals like cobalt, iron, and manganese have been extensively studied in catalytic MHAT, the potential of copper remains unexplored. This is due to the polar reactivity exhibited by classical nucleophilic Cu(I) hydrides. Here we report the first examples of catalytic hydrofunctionalization reactions enabled by copper-mediated MHAT. The Cu-MHAT process produces alkyl radicals in high chemo- and regioselectivity, which are subsequently captured by Cu(II) species to undergo coupling reactions with an exceptionally broad scope of oxygen-, nitrogen-, halogen-, and carbon-based nucleophiles. The distinct reactivity and selectivity observed in Cu-MHAT system is attributed to the involvement of Cu(II)–H species, a hydrogen atom donor that possesses substantially altered charge distribution and weakened bonding in contrast to conventional Cu(I) hydrides. Preliminary results suggest potential extension to asymmetric catalysis and radical polymerization are viable. This work opens up new opportunities for MHAT chemistry by going beyond the common oxidation states.

Abstract:Electrochemically driven hydrogen atom transfer (HAT) catalysis provides a complementary approach for the transformation of redox-inactive substrates that would be inaccessible to conventional electron transfer (ET) catalysis. Moreover, electrochemically driven HAT catalysis could promote organic transformations with either hydrogen atom abstraction or donation as the key step. It provides a versatile and effective tool for the direct functionalization of C(sp3)-H/Si-H bonds and the hydrofunctionalization of alkenes. Despite these attractive properties, electrochemically driven HAT catalysis has been largely overlooked due to the lack of understanding of both the catalytic mechanism and how catalyst selection should occur. In this Review, we give an overview of the HAT catalysis applications in the direct C(sp3)-H/Si-H functionalization and hydrofunctionalization of alkenes. The mechanistic pathways, physical properties of the HAT mediators, and state-of-the-art examples are described and discussed.
