Evaluations of Marine Collagen Peptides from tilapia skin on experimental oral ulcer model of mice

Abstract:The present study aimed to evaluate the oral ulcer healing process treated with Marine Collagen Peptides (MCPs) from tilapia skin. Oral ulcer models were established on the tongue mucosa of C57/BL6 mice by phenol liquids. 60 mice were randomly divided into four groups: blank group, normal saline group (NS), low concentration MCPs group (LMCPs) and high concentration MCPs group (HMCPs). Each substance was respectively administered on the ulcer area once daily and continued until sacrificed (after 1, 3, 5, 7 and 9 days of ulcer establishment). In addition to clinical and macroscopic evaluation, we also performed histological, histomorphometric and immunohistochemistry analysis. The results indicated that MCPs could accelerate the oral ulcer healing process effectively as a result of the high content Pro-Hyp effects. Firstly it reduced the numbers of inflammatory cells infiltration. Secondly it promoted collagenesis, angiogenesis and re-epithelialization from D5. Thirdly, it down-regulate the expressions of IL-1β and TNF-α in the mid-late stage. In general, MCPs has great potential for developing clinical applications on oral ulcer treatments.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to evaluate the therapeutic effect of Marine Collagen Peptides (MCPs) extracted from tilapia skin on the experimental oral ulcer model in mice. Specifically, the study aims to explore whether MCPs can accelerate the healing process of oral ulcers and verify its effect through clinical, macroscopic, histological, histomorphometric and immunohistochemical analyses. The research background of the paper points out that oral ulcer is a common oral mucosal disease with a high incidence, which brings physical and mental discomfort to patients. Although there are currently some anti - inflammatory treatment methods, such as local use of steroids, antibiotics, traditional Chinese medicine or lasers, long - term use of these drugs may produce side effects, and so far there is no definite treatment plan. Therefore, it is very necessary to develop a new type of therapeutic agent that can combat oral ulcers and has almost no side effects. The study selected 60 C57/BL6 mice, established an oral ulcer model on the tongue mucosa with phenol liquid, and then randomly divided the mice into four groups: blank control group, normal saline group (NS), low - concentration MCPs group (LMCPs) and high - concentration MCPs group (HMCPs). Each group was given the corresponding treatment once a day until sacrifice (on the 1st, 3rd, 5th, 7th and 9th days after ulcer establishment respectively). The research results show that MCPs can effectively accelerate the healing process of oral ulcers, mainly through the following aspects: 1. Reducing the infiltration of inflammatory cells; 2. Promoting collagen production, angiogenesis and re - epithelialization; 3. Down - regulating the expression of interleukin - 1β (IL - 1β) and tumor necrosis factor - α (TNF - α) in the middle and late stages. In conclusion, MCPs have great application potential in the treatment of oral ulcers.
