Abstract:In the traditional Nuclear Magnetic Resonance(NMR),the tested substance should be located on the center of homogeneous magnetic field of superconductive magnet,which limited the application of NMR to some extents.Recently a novel NMR spectrometer,the NMR-MOUSE (nuclear magnetic resonance mobile universal surface explorer),was developed and characterized by small volume,convenient transferring and surface analysis,which can be used to on-line analysis on the surface of tested substances and is sensitive to material changes,thus allowing non-destructive assessment and the life time predications of materials.The fundamental principle of this novel NMR-MOUSE was introduced in this paper,with its application in characterizing of heterogeneous rubber and plastic polymers discussed.

Abstract:Many polysaccharides are allowed for direct food use, where they serve a number of useful functions including dietary fiber, bulking agent, thickener, encapsulant, gelling agent, foam and emulsion stabilizer, protective colloid, emulsifier and suspending agent, adhesive and binder, flocculant, swelling agent, film/coat former, or syneresis inhibitor. Many of these polysaccharides have complex structures or are mixtures with different components. Over the years, NMR has been a premiere technique for characterizing these materials. NMR characterization can help identify the materials in question, quantify the different functional groups present, and detect minor components and impurities. Above all, the high resolution achieved in solution NMR can provide detailed structural information on composition, sequence distribution, substitution pattern, and molecular weights (in some cases) for individual polysaccharides. Concurrent application of other analytical techniques, such as methylation, esterification, fractionation, mass spectrometry, and chromatographic methods, has enabled structural information on even complex polysaccharides or mixtures to be obtained. In this article a review is given of the solution NMR of food polysaccharides, with emphasis on papers published in the past 20 years. Included in the review is a survey of 21 common food polysaccharides, the current understanding of their structures, and the techniques used for their determination.

In this paper, the basal principle of NMR and MRI was illuminated. Some introductions of application study on NRI in inner quality, mature, inner flaw and damage, storage of fruit were presented. The excellence and limitation in research and application were talked about and the prospect of NRI applied in fruit was offered.

Abstract:Polysaccharides are ubiquitous biomolecules found in nature that contain various biological and pharmacological activities that are employed in functional foods and therapeutic agents. Natural polysaccharides are obtained mainly by extraction and purification, which may serve as reliable procedures to enhance the quality and the yield of polysaccharide products. Moreover, structural analysis of polysaccharides proves to be promising and crucial for elucidating structure–activity relationships. Therefore, this report summarizes the recent developments and applications in extraction, separation, purification, and structural analysis of polysaccharides of plants and fungi.

Comprehensive Reviews in Food Science and Food Safety

Abstract:Polysaccharides are natural polymer compounds widely distributed in plants, animals, and microorganisms, most of which have a broad spectrum of biological activities to promote human health. They could also be used as texture modifiers in food industry due to their excellent rheological and mechanical properties. Many researchers have shown that nonthermal processing technologies have numerous advantages, such as high extraction efficiency, short extraction time, and environmental friendliness, in the extraction of polysaccharides compared with the traditional extraction methods. Moreover, nonthermal technologies could effectively change the physicochemical properties and structural characteristics of polysaccharides to improve their biological activities or processing properties. Therefore, a comprehensive summary about the extraction and modification of polysaccharides by nonthermal technologies, including ultrasound, high hydrostatic pressure, pulsed electric fields, and cold plasma, was provided in this review. In particular, the underlying mechanisms, processing operations, and current application status of these technologies were discussed. In addition, the applications of combining nonthermal techniques with other technological methods in polysaccharide extraction and modification were briefly introduced.

Abstract:INTRODUCTION:Natural products, the small organic molecules produced by plants, microbes and invertebrates, often present in the form of a mixture, this leads to the structural characterisation of natural extracts often requiring time-consuming multistep purification procedures. Nuclear magnetic resonance (NMR) technology is traditionally utilised as a tool for the structural elucidation of pure compounds. Contemporarily, an up-to-date trend in the application of NMR in natural product research is shifting to the direct NMR analysis of crude mixtures, to obtain molecular structure and biological activity information without performing cumbersome separation.OBJECTIVE:To review works of literature on the evolution, principle and progress of NMR technologies for analysing mixtures, we focus on the successful application of NMR technologies in direct analysis of natural product extracts.METHODOLOGY:Based on our research experience, academic tracking and extensive literature search, which involved, but not limited to, the use of various databases, like Web of Knowledge and PubMed. The keywords used, in various combinations, to retrieve recent literature on the successful application of NMR technologies to sheer natural product extracts, and excluded artificially natural product mixture and biofluid.RESULTS:NMR technologies for direct natural extracts analysis, including two-dimensional J-resolved spectroscopy (2D-JRES), pure shift NMR, diffusion-ordered NMR spectroscopy (DOSY), statistical correlation spectroscopy (STOCSY), concentration-ordered NMR spectroscopy (CORDY), saturation transfer difference (STD) and water-ligand observed via gradient spectroscopy (WaterLOGSY) were illustrated.CONCLUSIONS:By these methods, molecular structure and biological activity information will be directly obtained from NMR analysis of natural products extract, aiming to save experimental time and expenses.
A Novel Critical Control Point and Chemical Marker Identification Method for the Multi-Step Process Control of Herbal Medicines Via NMR Spectroscopy and Chemometrics.

Abstract:Herbal medicines have played a vital role in maintaining the health of the world population in the past thousands of years, and have proved to be an effective therapy. It is important to improve our understanding of the effects of the multi-step processing in herbal medicines on the chemical changes to ensure product quality. A proton nuclear paramagnetic resonance (1H NMR)-based evaluation strategy was developed for an efficient process variation exploration and diversified metabolite identification. In this study, 48 process intermediates from 6 commercial batches of the multi-step manufacturing chain of Danshen processing were obtained. Hierarchical classification analysis (HCA) tree based on 1H NMR spectra clustered the samples according to the processing steps, which indicates that 1H NMR has the potential capability for critical control point identification based on its adequate information of the organic compounds. Then, principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis (OPLS-DA) were applied to distinguish the major metabolite differences between the intermediates before and after the critical control point. In this case, the alkali-isolation and acid-dissolution method was recognized as the most critical process in the multi-step chain of Danshen extract manufacturing. Potential metabolites with the larger amplitude of variation and contributing the most to the discrimination were found to be potential quality markers by S-plot, including several previously undetected amino acids. The results in this study are consistent with previous research studies and reference experiments conducted with other analytical tools. Taken together, they prove that 1H NMR with chemometrics is a very effective process quality control tool to provide comprehensive information on the chemical changes during the processing of herbal medicines, and help with the identification of critical control points and potential critical quality markers.
Quantification of food polysaccharide mixtures by 1H NMR

Abstract:Polysaccharides are food ingredients that critically determine rheological properties and shelf life. A qualitative and quantitative assessment on food-specific polysaccharide mixtures by 1H NMR is presented. The method is based on the identification of intact polysaccharides, combined with a quantitative analysis of their monosaccharide constituents. Identification of the polysaccharides is achieved by 1H NMR line shape fitting with pure compound spectra. The monomeric composition was determined using the Saeman hydrolysis procedure, followed by direct monosaccharide quantification by 1H NMR. In the quantification, both the monosaccharide degradation during hydrolysis, as well as a correction for the non-instantaneous polysaccharide dissolution were taken into account. These factors were particularly important for the quantification of pectins. The method showed overall good repeatability (RSDr=4.1±0.9%) and within-laboratory reproducibility (RSDR=6.1±1.4%) for various food polysaccharides. Polysaccharide mixtures were quantitatively resolved by a non-negative least squares estimation, using identified polysaccharides and their molar monosaccharide stoichiometry as prior knowledge. The accuracy and precision of the presented method make it applicable to a wide range of food polysaccharide mixtures with complex and overlapping 1H NMR spectra.

Abstract:The application of NMR crystallography to organic molecules is exemplified by two case studies. For the tosylate salt of the active pharmaceutical ingredient, Ritlectinib, solid-state NMR spectra are presented at a 1H Larmor frequency of 1 GHz and a magic-angle spinning (MAS) frequency of 60 kHz. Specifically, 14N-1H heteronuclear multiple-quantum coherence (HMQC) and 1H-1H double-quantum (DQ) single-quantum (SQ) correlation experiments are powerful probes of hydrogen bonding interactions. A full assignment of the 1H, 13C and 14N/ 15N chemical shifts is achieved using also 1H-13C cross polarization (CP) HETCOR spectra together with gauge-including projector augmented wave (GIPAW) DFT calculation for the geometry-optimised X-ray diffraction crystal structure that is reported here (CCDC 2352028). In addition, GIPAW calculations are presented for the 13C chemical shifts in the two polymorphs of cellulose for which diffraction structures are available. For both case studies, a focus is on the discrepancy between experiment and GIPAW calculation.

Abstract:Polysaccharide characterization posts the most difficult challenge to available analytical technologies compared to other types of biomolecules. Plant polysaccharides are reported to have numerous medicinal values, but their effect can be different based on the types of plants, and even regions of productions and conditions of cultivation. However, the molecular basis of the differences of these polysaccharides is largely unknown. In this study, direct analysis in real time mass spectrometry (DART-MS) was used to generate polysaccharide fingerprints. Large saccharides can break down into characteristic small fragments in the DART source via pyrolysis, and the products are then detected by high resolution MS. Temperature was shown to be a crucial parameter for the decomposition of large polysaccharide. The general behavior of carbohydrates in DART-MS was also studied through the investigation of a number of mono- and oligosaccharide standards. The chemical formula and putative ionic forms of the fragments were proposed based on accurate mass with less than 10 ppm mass errors. Multivariate data analysis shows the clear differentiation of different plant species. Intensities of marker ions compared among samples also showed obvious differences. The combination of DART-MS analysis and mechanochemical extraction method used in this work demonstrates a simple, fast, and high throughput analytical protocol for the efficient evaluation of molecular features in plant polysaccharides.

Abstract:<p>Two-dimensional NMR spectroscopies are one of the most frequently used techniques for the structural determination of carbohydrates. However, the data analysis is challenging because of the signal overlap in the <sup>1</sup>H homonuclear correlation spectra. We attempted to explore a general strategy for the structural determination of carbohydrates by combined multi-dimensional spectroscopies. The strategy was applied to a human milk oligosaccharide lacto-N-difucohexaose I, that has been previously studied by conventional two-dimensional NMR spectroscopy. Assignment of the intra-residue resonances of the hexasaccharide using the three-dimensional spectrum was straightforward. Consequently, data analysis of the multi-dimensional spectra was significantly simplified, leading to a quicker determination of the intra- and inter-residue connections in the hexasaccharide. Application of the NMR strategy to chondroitin sulfate from bovine cartilage revealed two repeating disaccharide regions of the A and C units of chondroitin sulfate, indicating the high potential of this technique for the structural determination of complex polysaccharides.</p>

Abstract:The extraction methods of saccharides from plants were developed from solid-liquid extraction toultrasonic extraction and microwave assisted extraction, and achieved the results of sample and solvent con-sumption, high speed and environment friendly. The solid phase extraction was adopted into purification process, and its packings have being modified all the way on structures and functions, the materials variedfrom C18, NH2,ion exchange column to molecularly imprinting materials and optimized ionic liquid column. As to detec-tion, different methods were developed for monosaccharides to meet the requirements of chromatographic analysis, because most of them leak UV absorption and fluorescence properties. In this paper, the development of sample pretreatment methods and chromatographic analysis of monosaccharides in the plant samples was reviewed. The extraction, purification, separation and detection of monosaccharides from different samples were discussed.

Abstract:Polysaccharides are widely distributed in natural sources from monocytic microorganisms to higher animals, and are found in a variety of biological activities in recent decades. Natural polysaccharides have the characteristics of large molecular weight, diverse composition, and complex structure, so their purification and structural analysis are difficult issues in research. Chromatography as a powerful separation technique, plays an irreplaceable role in the separation and structural analysis of natural polysaccharides, especially in the purification of polysaccharides, the separation of hydrolysates, and the analysis of monosaccharide composition. The separation mechanisms and application of different chromatographic methods in the studies of polysaccharides were summarized in this review. Moreover, the advantages and drawbacks of various chromatography methods were discussed as well.

Abstract:In this paper,the extracting technique of sulfated polysaccharide is discussed and SPS is identified.The results show that the optimum extraction conditions as following: the time of extracting is(2 h),the temperature of extracting is 90℃,the quantity of water to kelp is 25.The yield of product is 1.99%.The content of sulfated radical and polysaccharide is 61.26%.

Abstract:Current practices for structural analysis of extremely large-molecular-weight polysaccharides via solution-state nuclear magnetic resonance (NMR) spectroscopy incorporate partial depolymerization protocols that enable polysaccharide solubilization in suitable solvents. Non-specific depolymerization techniques utilized for glycosidic bond cleavage, such as chemical degradation or ultrasonication, potentially generate structural fragments that can complicate complete and accurate characterization of polysaccharide structures. Utilization of appropriate enzymes for polysaccharide degradation, on the other hand, requires prior structural knowledge and optimal enzyme activity conditions that are not available to an analyst working with novel or unknown compounds. Herein, we describe an application of a permethylation strategy that allows the complete dissolution of intact polysaccharides for NMR structural characterization. This approach is utilized for NMR analysis of Xylella fastidiosa extracellular polysaccharide (EPS), which is essential for the virulence of the plant pathogen that affects multiple commercial crops and is responsible for multibillion dollar losses each year.

Abstract:NMR-based metabolic profiling techniques can simultaneously track changes in many plant metabolites and have found a number of applications in both systems biology and biosafety. Together with multivariate statistical analyses, NMR spectroscopy has been successfully applied to the characterisation of various herbs and plant products for quality control, authentication, determining geographical origin and for detecting adulteration of products. Additionally, the metabolic consequences of plant extracts have been demonstrated in experimental animals and in man using NMR-based metabonomics approaches to characterise the response. Here the application of NMR spectroscopy and chemometric tools for analysing plant-based products and their metabolic consequences are considered with particular emphasis on deconvolving biological complexity and minimising confounding biological variability with analytical 'noise'.

Abstract:Carboxymethyl xylan (CMX) was synthesized and characterized. A method based on quantitative 13C NMR was developed to obtain detailed structural information. The total degree of substitution (DS), partial DS at 2- and 3-O-positions, and the quantitation of each unit in CMX could be acquired simultaneously by this method without decomposing the polymer backbone, thus avoiding errors caused by polymer hydrolysis with traditional methods using HPLC and 1H NMR spectrometry. Moreover, different from methods of direct detection of carboxyl groups, such as acidometric and potentiometric titrations, this method was not affected by probable impurity introduced during carboxymethylation.
