Abstract:Abscisic acid (ABA) is the most important phytohormone involved in the response to drought stress. Subclass II of SNF1-related kinase 2 (SnRK2) is an important signaling kinase related to ABA signal transduction. It regulates the phosphorylation of the target transcription factors controlling the transcription of a wide range of ABA-responsive genes in Arabidopsis thaliana. The transgenic poplars (Populus tremula × P. tremuloides, clone T89) ectopically overexpressing AtSnRK2.8, encoding a subclass II SnRK2 kinase of A. thaliana, have been engineered but almost no change in its transcriptome was observed. In this study, we evaluated osmotic stress tolerance and stomatal behavior of the transgenic poplars maintained in the netted greenhouse. The transgenic poplars, line S22, showed a significantly higher tolerance to 20% PEG treatment than non-transgenic controls. The stomatal conductance of the transgenic poplars tended to be lower than the non-transgenic control. Microscopic observations of leaf imprints revealed that the transgenic poplars had significantly higher stomatal closures under the stress treatment than the non-transgenic control. In addition, the stomatal index was lower in the transgenic poplars than in the non-transgenic controls regardless of the stress treatment. These results suggested that AtSnRK2.8 is involved in the regulation of stomatal behavior. Furthermore, the transgenic poplars overexpressing AtSnRK2.8 might have improved abiotic stress tolerance through this stomatal regulation.

What problem does this paper attempt to address?
The main problem this paper attempts to address is improving the drought tolerance of poplar (*Populus tremula × P. tremuloides*) by heterologously expressing the SNF1-related kinase 2 (SnRK2) subclass II gene (AtSnRK2.8) from Arabidopsis (*Arabidopsis thaliana*). Specifically, the researchers aim to enhance the survival ability of poplar under drought conditions by regulating stomatal behavior. ### Main Issues 1. **Improving Poplar's Drought Tolerance**: By heterologously expressing the AtSnRK2.8 gene, researchers hope to enhance the survival ability of poplar under drought conditions. 2. **Stomatal Regulation Mechanism**: Investigate whether the AtSnRK2.8 gene improves poplar's drought tolerance by regulating stomatal behavior. ### Research Background - **ABA Signal Transduction**: Abscisic acid (ABA) is an important plant hormone in response to drought stress, and SnRK2 kinases play a key role in ABA signal transduction. - **Function of SnRK2 Genes**: SnRK2 genes in Arabidopsis are divided into three subclasses, with subclass II SnRK2 genes (such as AtSnRK2.8) playing a role in ABA-dependent stress responses. - **Stress Response in Poplar**: There is limited research on the abiotic stress response pathways in woody plants (such as poplar), necessitating further exploration of the function of SnRK2 genes in poplar. ### Research Methods - **Construction of Transgenic Poplar**: Using transgenic technology, the AtSnRK2.8 gene was introduced into poplar, resulting in the construction of two independent transgenic lines (S01 and S22). - **Stress Treatment**: Osmotic stress was simulated using polyethylene glycol (PEG) solution to evaluate the tolerance of transgenic poplar and non-transgenic control groups. - **Stomatal Behavior Analysis**: Stomatal conductance, stomatal index, and stomatal aperture were measured to assess changes in stomatal behavior in transgenic poplar. ### Main Findings - **Increased Tolerance**: Transgenic poplar showed significantly higher tolerance under 20% PEG treatment, maintaining higher levels of photosynthetic quantum yield (QY) and chlorophyll content (SPAD values). - **Reduced Stomatal Conductance**: Transgenic poplar exhibited lower stomatal conductance under both stress and non-stress conditions, suggesting that AtSnRK2.8 may reduce water evaporation by decreasing stomatal opening. - **Changes in Stomatal Index and Aperture**: The stomatal index of transgenic poplar was significantly lower than that of the non-transgenic control group, and the degree of stomatal closure was higher under stress conditions. ### Conclusion This study demonstrates that heterologous expression of the AtSnRK2.8 gene can significantly improve the drought tolerance of poplar, primarily through the regulation of stomatal behavior. This finding provides new insights for developing drought-resistant poplar varieties.
