Abstract:In this research, a new DSA (Distyryl-anthracene) derivative with four carboxyl groups was designed and synthesized. This molecule exhibits aggregation-induced emission property (AIE). With the AIE character, a convenient and sensitive fluorescent probe for the detection of protamine, heparin and heparinase has been developed. The protamine can directly induce the aggregation of probe, which caused by electrostatic attraction. In this way, the turn-on detection of protamine is achieved, and the detection limit is as low as 30 ng mL-1. When heparin appears, the probes will be redisperse in solution, which causes a decrease in fluorescence intensity. Besides, this method also shows good selectivity and sensitivity, and the linear range of heparin is from 0.08 to 8 μg mL-1 with detection limit of 37 ng mL-1. After hydrolyzing heparin by heparinase, the probes rebind with protamine and the fluorescence enhance. The fluorescence enhancement was linearly related to the concentration of heparinase in the range of 0.02-2.0 μg mL-1 and detection limit as low as 143.7 ng mL-1. In addition, the results exhibited that the recovery percentage of heparinase in bovine samples reached to 96-101%.

Abstract:Heparin is a common blood anticoagulant, and is widely used in biomedical fields. Herein, we design a readily effective fluorescent probe for the detection and quantitation of Heparin. A nonemissive quinoline-malononitrile derivative QM-CO is affected to emit by Heparin, showing a near-infrared aggregation-induced emission (AIE), which produces the AIE effect more 4.62-fold fluorescence emission intensity than free QM-CO. The AIE probe enjoys a good linear relationship with the Heparin concentration is in the range of 0–1.75 μg/mL (R 2 = 0.9937), a low detection limit (down to 0.139 μg/mL), and a superior selectivity to correlative substances. Utilizing the AIE feature of QM-CO and the electrostatic adsorption from Heparin to QM-CO, the AIE-based near infrared fluorescent probe QM-CO achieved a long wavelength (665 nm) emission and good selectivity for Heparin. Scanning electron microscopy (SEM) suggests that the loose aggregate QM-CO ensembles are disassembled and respectively condensed to form many tightly packed QM-CO/Heparin ensembles, offering mechanistic insight into the microenvironment of QM-CO/Heparin ensembles with the aid of the zeta potential.

Abstract:In this strategy, the fluorescence sensor Nap-Co-T1 employing the fluorescence resonance energy transfer (FRET) mechanism was designed and synthesized to have an efficient response to Heparin, and the FRET mechanism was explored for different excitation-emission wavelengths with different distances between the energy acceptor and the energy donor (comparing with fluorescence sensor Nap-TPA-T2). Upon the addition of Heparin, the fluorescence emission of Nap-Co-T1 was turned on at 565 nm, and the fluorescence color changed of the solution from colorless to bright yellow. The limit of detection (LOD) was as low as 0.04 μg/mL. With the addition of antagonistic protamine (PRTM) to the sensor complex with Heparin, the fluorescence emission was turned off to a certain extent, and the reversibility of the "off-on-off" system was maintained for five cycles or more. In addition, Nap-Co-T1 provides rapid and sensitive detection of Heparin in human serum albumin solution and artificial urine and is highly sensitive to environmental viscosity.

Abstract:A novel fluorescence turn-on method for sensitive and selective detection of protamine was developed based on aggregation-induced emission enhancement (AIEE) characteristics of 4-(6'-carboxyl)hexyloxysalicylaldehyde azine (CHSA) induced by electrostatic interaction between protamine and CHSA(-). Under optimal conditions described, a large Stokes shift of approximately 198 nm could be observed, and the fluorescence enhancement at 538 nm was linearly related to the concentration of protamine in the range of 0.6-18 microg mL(-1) with the relative correlation coefficient of R(2) = 0.9996 (n = 11) and a detection limit as low as 43 ng mL(-1). The relative standard deviation (R.S.D.) was 2.0% (n = 5). The proposed method was successfully utilized in quantifying protamine in diluted horse serum. In addition, due to the special electrostatic association between protamine and heparin, CHSA could also be employed as a probe to study the interaction between them.

Abstract:A positively charged tetraphenylethene (TPE) derivative, TPE-4MN, was synthesized as a probe for heparin based on aggregation induced emission. On the addition of 5.0 mu g/mL of heparin, TPE-4MN showed an enhanced emission of about 10-fold. The change in fluorescence at 475 nm was linear over a range of heparin concentrations of 0-1.0 mu g/mL with an R = 0.99988 and the limit of detection (LOD) was calculated to be 0.75 mu g/mL. The mechanism of the detection was proven to be through an ion pairing interaction. TPE-4MN showed good selectivity for heparin over other types of polysaccharides and could easily distinguish heparin from heparan sulfate, a glycosaminoglycan having a similar structure to that of heparin. Abstract:A facile, rapidly responsive fluorescence turn-on probe for heparin with high selectivity and sensitivity was reported in this paper. The probe could aggregate on the negatively charged heparin template through electrostatic interactions and then display intense fluorescence due to its aggregation-induced emission (AIE) characteristics. Under optimal condition, the probe showed high selectivity to heparin over chondroitin sulfate(ChS), hyaluronic acid (HA), dextran (DeX) and other substances, with a linear range of 0.2–14μg/mL, and a detection limit of 57.6ng/mL. In diluted serum, it also showed good performance.

Abstract:A novel ratiometric fluorescent strategy was proposed for sensitive detection of protamine and heparin based on N,P‐doped CDs and Eosin Y. A novel ratiometric fluorescent assay was proposed for protamine and heparin based on blue‐emitting N,P‐doped carbon dots (N,P‐CDs) and green‐emitting Eosin Y. The N,P‐CDs were prepared using arginine as carbon and nitrogen sources according to hydrothermal method with favorable luminescence intensity, photostability, and good reproducibility. And the N,P‐CDs were used as internal reference fluorescence molecules. The anionic Eosin Y was used as fluorescence signal molecule. Protamine with positive group could interact with Eosin Y through electrostatic effect, leading to the fluorescence quenching of Eosin Y. Upon the addition of heparin, heparin was combined with protamine with stronger affinity through electrostatic interactions and hydrogen bonds, which preventing the bonding of protamine with Eosin Y and obvious fluorescence emissions of Eosin Y at 538 nm could be observed. Meanwhile, the fluorescence signal of N,P‐CDs at 420 nm remained stable during the whole process. A good linear range between the fluorescence emission intensity ratio of I538/I420 was obtained for protamine from 0.01 to 40 μg/mL with a low detection limit of 5.6 ng/mL, and heparin from 0 to 35 μg/mL with a low detection limit of 2.5 ng/mL. Moreover, the method was further applied to detection of heparin with satisfactory results in fetal bovine serum samples.

Abstract:Cu（Ⅱ） detection is important because it plays crucial role in several biological processes and ecological systems.Fluorescent techniques have attracted more and more attention in Cu（Ⅱ） detection.In this report,we contribute a novel strategy to use fluorescence spectroscopy for Cu（Ⅱ） specific detection.The specificity relies on the fact that,of the many metal cations,only Cu（Ⅱ） can catalyze the hydrolyzation of a-amino acid ester.The novelty originates from the unique aggregation-induced emission（AIE） property of the fluorescent label.We designed a model a-amino acid ester（TPE-Ala） constructed with alanine and tetraphenylethene-functionalized methanol（TPE-methanol）.In comparison with the precursor TPE-Ala, TPE-methanol has lower solubility and is easy to form aggregates in water,thereby displaying a higher fluorescent response.Thus,the Cu（Ⅱ） catalyzed hydrolyzation can be monitored by recording the fluorescence enhancement and fluorescent detection Cu（Ⅱ） is rationally achieved.

Abstract:Heparin is a widely used anticoagulant and the quantification of heparin concentration is pivotal for clinical use. However, previous fluorescent probes for heparin detection are usually based on fluorophores with aggregation-caused quenching (ACQ) properties, which severely restrict their applications for quantitative measurement of heparin in a wide range. Herein, we develop an aggregation-induced emission (AIE) probe HPQ-TBP-I for light up detection of heparin based on the electrostatic interaction-triggered formation of the HPQ-TBP/heparin complex and simultaneous displacement of the fluorescence quencher iodide ion. A linear relationship from 0 to 14 mu M of heparin accompanied with a low detection limit of 22 nM was achieved, which fully covers the whole clinical dose range (1.7-10 mu M). It is anticipated that this easily accessible and sensitive AIE probe is promising for clinical applications.
AIE-active TPA modified Schiff base for successive sensing of Cu 2+ and His via on-off-on method and its application in bioimaging

Abstract:In this article, a novel triphenylamine-modified salicylaldehyde Schiff base 2-(((4-(diphenylamino)phenyl)imino)methyl)-4-(pyridine-4-yl)phenol (HL) was synthesized and structurally characterized. HL possessed D-π-A structure and exhibited typical AIE property in THF/H 2 O. It was applied to selectively recognize Cu 2+ through on-off mode in THF/H 2 O (1/9, v/v), and the fluorescence attenuation was ascribed to paramagnetic quenching effect of Cu 2+ together with the abatement of HL aggregates. Hence the detection limit achieved as low as 1.32×10 -7 M. The spectroscopic and ESI-HRMS results revealed the 1:2 complexation ratio of Cu 2+ with HL. The mechanism for sensing Cu 2+ was further confirmed by DFT calculations. In virtue of the large affinity between Cu 2+ and His, the resultant CuL 2 system was further used to detect His via off-on method based on displacement of ligands. The detection limit for His reached 5.14×10 -8 M. Furthermore, HL was available to prepare handy indicator papers for on-site recognition of Cu 2+ and His. Confocal fluorescent imaging demonstrated that HL could sequentially respond to intracellular Cu 2+ and His.

Abstract:A ratiometric fluorescence sensor based on pyrene was designed for selective detection of heparin in HEPES (N-(2-hydroxyethyl)piperazine-N'-ethanesulfonic acid) buffer and serum sample. Pyrene and long-chain alkanes were linked through bisquaternary functionality in the sensor which could interact with heparin via supramolecular assembly. A ratiometric fluorescent signal change of the sensor can be observed because of the specific monomer-excimer conversion of pyrene which is modulated by the supramolecular self-assembly of sensor and heparin. Upon addition of heparin, the excimer emission of the sensor at 489 nm is observed and the monomer emission intensity at 395 nm decreases concomitantly. Addition of heparin derivatives with very similar structure such as chondroitin 4-sulfate or hyaluronic add to the same sensor solution only leads to very smaller changes in intensity ratios probably because of lower charge density and more distant spatial distribution of anions (or disadvantageous spatial orientation of anions) as compared to those of heparin. The novel sensor can effectively differentiate heparin from its derivatives with relatively low background interference and wide linear response in HEPES and serum. A linear calibration curve is obtained from 0 to 3.4 mu M for heparin quantification in serum.
Aggregation-Induced Emission Enhancement of CdSe QDs by Protamine and its Application to Sensitively and Selectively Detect Heparin

Abstract:Background: Heparin, it is commercially used as an anticoagulant in surgical procedures for the prevention of blood clotting. However, overdose and prolonged use of heparin often induce potentially fatal bleeding complication. So, it is of crucial importance to monitor closely heparin levels for the sake of health. In this work, a sensitive fluorescence sensing platform to detect heparin was set up based on MPA-CdSe QDs (quantum dots) and protamine enhanced fluorescent system. Methods: The image of CdSe QDs was taken on a JEM-2100 transmission electron microscope (JEOL Ltd.). The fluorescence spectrum was recorded on a FluoroMax-4 fluorescence spectrophotometer (Horiba, USA). UV–vis absorption spectrum was recorded using a Shimadzu UV-2450 Spectrophotometer (Tokyo, Japan). A vortex mixer IKA MS3 digital was selected to mix the solution. Results: Under optimized conditions, the linear response to detect heparin ranges from 0.06 to 14 μg mL-1 with a detection limit of 8 ng mL-1. The approach showed a highly selective response to heparin in the presence of 16 interfered substances. Conclusion: A simple method for the detection of heparin was developed based on MPA-CdSe QDs and protamine enhanced fluorescent system. The electrostatic effect between MPA-CdSe QDs and protamine resulted in strong fluorescence enhancement from the MPA-CdSe QDs. Moreover, the addition of heparin could cause a significant fluorescence decrease due to the strong affinity of protamine and heparin. Under optimal conditions, this method displayed a low detection limit and good selectivity over other substances.

Abstract:In this paper a label-free fluorescent sensor for probing the interaction between heparin and protein was reported. Heparin, the bioactive polyanions, formed supramolecular assemblies with cationic surfactant cetyltrimethyl ammonium bromide (CTAB). The environment-dependent dye pyrene, encapsulated in hydrophobic interiors of the supramolecular assemblies worked as the fluorescence probe. Once the heparin-binding protein was added, competing interactions of protein with heparin would weaken the interaction between CTAB and heparin. As a result, the noncovalently sequestered pyrene would be released upon disassembly and the fluorescence of the released pyrene was subsequently decreased. The binding events were exemplified by protamine and Tat peptide, these processes were also verified by DLS and TEM. Such a strategy is appealing as organic synthesis was traded off against supramolecular assembly. This label-free fluorescent system is simple, selective, convenient, and can serve as a good complement to other existing methods, also this method has the potential for preprimary drug screening.

Abstract:Heparin is a highly negatively charged sulfated linear polymer glycosaminoglycan that has been widely used as an anticoagulant in medicine. Protamine is a cationic protein rich in arginine that is used to treat the blood-brain barrier during excess heparin surgery. Trypsin is the most important digestive enzyme-encoding generated by the pancreas and can specifically cleave the carboxyl ends of arginine and lysine residues. Heparin, protamine, and trypsin interact and constrain each other, and their fluctuations reflect the body's dysfunction. Therefore, it is necessary to develop a fast, sensitive, and highly selective assay for regularly monitoring the levels of heparin, protamine, and trypsin in serum. Herein, a fluorescent and colorimetric dual-mode upconversion nanoparticle (UCNP) biosensor was used for the determination of heparin, protamine, and trypsin based on the oxidase-mimicking activity of Ce 4+ and electrostatic control. The biosensor exhibited sensitive detection of heparin, protamine, and trypsin with low limits of detection (LODs) of 16 ng/mL, 87 ng/mL and 31 ng/mL, respectively. Furthermore, the designed biosensor could eliminate autofluorescence, which not only effectively increased the accuracy of the sensor but also provided a new sensing pathway for the detection of differently charged biotargets.

Polycation-Induced Benzoperylene Probe Excimer Formation and the Ratiometric Detection of Heparin and Heparinase

Abstract:A benzoperylene probe excimer emission in an aqueous buffer solution is observed for the first time, and a novel ratiometric fluorescence method based on the probe excimer emission for the sensitive detection of heparin and heparinase is demonstrated. A negatively charged benzoperylene derivative, 6-(benzo[ghi]perylene-1,2-dicarboxylic imide-yl)hexanoic acid (BPDI), was employed. A polycation, poly(diallyldimethylammonium) chloride (poly-DDA), could induce aggregation of BPDI through noncovalent interactions. A decrease of BPDI monomer emission and a simultaneous increase of BPDI excimer emission were observed. Upon the addition of heparin, the strong binding between heparin and poly-DDA caused release of BPDI monomer molecules, and an excimer-monomer emission signal transition was detected. However, after the enzymatic hydrolysis of heparin by heparinase, heparin was hydrolyzed into small fragments, which weakened the competitive binding of heparin to poly-DDA. Poly-DDA induced aggregation of BPDI, and a monomer-excimer emission signal transition was detected. Our assay is simple, rapid, inexpensive, sensitive and selective, which could facilitate the heparin and heparinase related biochemical and biomedical research.

Abstract:In this work, a novel fluorescence “turn off–on” nanosensor for the determination of heparin and heparinase based on CuInS2 quantum dots (QDs) was established. CuInS2 QDs (modified by l-cysteine) featuring amino groups were directly prepared in aqueous solution via a hydrothermal synthesis method. The amino groups on the surface of CuInS2 QDs can interact with sulfate and carboxylate groups in heparin via electrostatic interactions and hydrogen bonding, which led the fluorescence of CuInS2 QDs to “turn-off”. However, the heparin could be hydrolyzed into small fragments in the presence of heparinase, which resulted in the fluorescence of CuInS2 QDs being recovered. Therefore, the addition of heparinase to the heparin/CuInS2 QDs system activated the fluorescence of CuInS2 QDs to “turn-on” state. Thus, the determination of heparin and heparinase could be achieved by monitoring the fluorescence “turn off–on”. Under the optimum conditions, there was a good linear relationship between I/I0 (I and I0 were the fluorescence intensity of CuInS2 QDs in the presence and absence of heparin, respectively) and heparin concentration in the range of 0.05–15μmolL−1 with the detection limit of 12.46nmolL−1. The linear detection for heparinase was in the range of 0.2–5μgmL−1 with the detection limit of 0.07μgmL−1. The proposed nanosensor was employed for the detection of heparin in fetal bovine serum samples with satisfactory results.

Abstract:We have reported an optical indicator displacement assay (IDA) for heparin with a UV-vis absorbance and fluorescence dual-readout based on pyranine/methyl viologen (MV2+). Upon introducing heparin, pyranine/MV2+ shows a clearly observable increase in UV-vis absorbance and a turn-on of the fluorescence signal. We have demonstrated that the ionic nature of buffers significantly affects the pyranine displacement and the zwitterionic HEPES was most suitable for heparin sensing. After careful screening of experimental conditions, the pyranine/MV2+-based optical chemosensor exhibits a fast, sensitive, and selective response toward heparin. It shows dynamic linear concentration of heparin in the ranges of 0.1-40 U·mL-1 and 0.01-20 U·mL-1 for the absorptive and fluorescent measurements, respectively, which both cover the clinically relevant levels of heparin. As with the animal experiments, the optical chemosensor has been demonstrated to be selective and effective for heparin level qualification in rat plasma. The chemosensor is readily accessible, cost-effective, and reliable, which holds a great promise for potential application on clinical and biological studies. Furthermore, this IDA system can serve as an IMPLICATION logic gate with a reversible and switchable logical manner.
Nanozyme catalysis-assisted ratiometric multicolor sensing of heparin based on target-specific electrostatic-induced aggregation

Abstract:Monitoring the level of heparin in clinical matrices is significant because of its pivotal role in preventing thrombosis. Compared to traditional single-signal sensors, multi-signal ratiometric detection can provide anti-interference results especially in complicated environments. However, fabricating an easy-to-operation, low-cost and robust sensor for the ratiometric detection of heparin still remains challenging. Here we propose a novel nanosensor for the ratiometric multicolor sensing of heparin with high performance. The sensor is based on the specific electrostatic interaction between the target and a positively charged species generated from nanozyme catalysis. FeMoO4 nanorods are explored as an oxidase mimic for the first time, showing a high activity at neutral pH to catalyze the colorless 3,3',5,5'-tetramethylbenzidine (TMB) oxidation to blue TMBox with an absorbance at 652 nm. Heparin can induce the rapid aggregation of the produced TMBox intermediate with rich positive charges due to their strong electrostatic interaction, leading to the formation of a purple Heparin-TMBox complex providing a signal at 565 nm. With the increase of heparin, the color changes from blue to indigo and further purple, enabling the multicolor sensing of the target. As a result, ultrasensitive determination of heparin was obtained with a very low detection limit. The fabricated nanosensor could differentiate heparin from complex species with no interferences, and it provided reliable analytical results for heparin in both serum and plasma. With robust performance, low cost and facile fabrication, the sensor holds great potential in monitoring heparin for clinical applications.

Abstract:Multi-functional fluorescent probes have attracted more interest recently. Herein, two simple fluorescent probes (FAS and DPAS) from AIEgens with the ESIPT process of salicylaldehyde Schiff-base structures are developed. The aggregates of FAS and DPAS exhibit good selectivity and sensitivity towards Cu2+ in aqueous media without any water-miscible organic solutions as buffers. Their larger association constants and appropriate hydrophilic and hydrophobic properties play key roles in achieving lower detection limits. Based on their fluorescence on/off processes in the presence or absence of Hg2+, the composite probes are further prepared.

Abstract:Two novel conjugated polymers containing 2,3-di(pyridin-2-yl)quinoxaline (DPQ) unit (P1) and 2,3-di(thiophen-2-yl)quinoxaline (DTQ) unit (P2) were designed and synthesized through Sonogashira coupling reaction. P1 provided high sensitivity and selectivity to silver ions: color change from green to brown red and fluorescent emission quenching, which can be observed by naked eye, even in the presence of interference cations. However, P2 with the similar polymer backbone to P1 showed no absorption and fluorescence changes toward any cations. Therefore, the DPQ units in P1 exhibited a key role for the selective recognition of Ag+, which was further confirmed by the 1H NMR titration, density functional theory (DFT) calculations and electrochemical studies. These results not only imply that DPQ based conjugated polymer is a promising probe of Ag+ ions but also afford a good guidance to design effective polymer sensors in future.
