Abstract:As a powerful tool to advance drug discovery, molecular imaging may provide new insights into the process of drug effect and therapy at cellular and molecular levels. When compared with other detection methods, fluorescence-based strategies are highly attractive and can be used to illuminate pathways of drugs' transport, with multi-color capacity, high specificity and good sensitivity. The conjugates of fluorescent molecules and therapeutic agents create exciting avenues for real-time monitoring of drug delivery and distribution, both in vitro and in vivo. In this short review, we discuss recent developments of small molecule-based fluorophore-drug conjugates, including non-cleavable and cleavable ones, that are capable of visualizing drug delivery.

Abstract:ConspectusEnzymes are essential for life, especially in the development of disease and on drug effects, but as we cannot yet directly observe the inside interactions and only partially observe biochemical outcomes, tools “translating” these processes into readable information are essential for better understanding of enzymes as well as for developing effective tools to fight against diseases. Therefore, sensitive small molecule probes suitable for direct in vivo monitoring of enzyme activities are ultimately desirable. For fulfilling this desire, two-photon small molecule enzymatic probes (TSMEPs) producing amplified fluorescent signals based on enzymatic conversion with better photophysical properties and deeper penetration in intact tissues and whole animals have been developed and demonstrated to be powerful in addressing the issues described above. Abstract:Fluorescent probes have emerged as indispensable chemical tools to the field of chemical biology and medicine. The ability to detect intracellular species and monitor physiological processes has not only advanced our knowledge in biology but has provided new approaches towards disease diagnosis. In this review, we detail the design criteria and strategies for some recently reported fluorescent probes that can detect a wide range of biologically important species in cells and in vivo. In doing so, we highlight the importance of each biological species and their role in biological systems and for disease progression. We then discuss the current problems and challenges of existing technologies and provide our perspective on the future directions of the research area. Overall, we hope this review will provide inspiration for researchers and prove as useful guide for the development of the next generation of fluorescent probes.

Abstract:Over the last few years, the development of fluorescent probes has received considerable attention. Fluorescence signaling allows noninvasive and harmless real-time imaging with great spectral resolution in living objects, which is extremely useful for modern biomedical applications. This review presents the basic photophysical principles and strategies for the rational design of fluorescent probes as visualization agents in medical diagnosis and drug delivery systems. Common photophysical phenomena, such as Intramolecular Charge Transfer (ICT), Twisted Intramolecular Charge Transfer (TICT), Photoinduced Electron Transfer (PET), Excited-State Intramolecular Proton Transfer (ESIPT), Fluorescent Resonance Energy Transfer (FRET), and Aggregation-Induced Emission (AIE), are described as platforms for fluorescence sensing and imaging in vivo and in vitro. The presented examples are focused on the visualization of pH, biologically important cations and anions, reactive oxygen species (ROS), viscosity, biomolecules, and enzymes that find application for diagnostic purposes. The general strategies regarding fluorescence probes as molecular logic devices and fluorescence-drug conjugates for theranostic and drug delivery systems are discussed. This work could be of help for researchers working in the field of fluorescence sensing compounds, molecular logic gates, and drug delivery.

Abstract:In recent years, a variety of receptor-targeted fluorescent probes have been developed and widely used to realize the visualization of certain receptors, which facilitates the early diagnosis and treatment of diseases. In this Review, we focus on the recent achievements in design, chemical structure, imaging characterization, and potential applications of receptor-targeted fluorescent probes from the past 10 years. The development and application of receptor-targeted fluorescent probes will expand our knowledge of the distribution and function of disease-related receptors, shed light on the drug discovery for clinical diseases where receptors are implicated, and feed into the diagnosis and treatment of a plethora of diseases, including tumors.

Abstract:Some important biological species and microenvironments maintain a complex and delicate dynamic balance in life systems, participating in the regulation of various physiological processes and playing indispensable roles in maintaining the healthy development of living bodies. Disruption of their homeostasis in living organisms can cause various diseases and even death. Therefore, real time monitoring of these biological species and microenvironments during different physiological and pathological processes is of great significance. Fluorescent-probe-based techniques have been recognized as one of the most powerful tools for real time imaging in biological samples. In this Account, we introduce the representative works from our group in the field of fluorescent probes for biological imaging capable of detecting metal ions, small bioactive molecules, and the microenvironment. The design strategies of small molecule fluorescent probes and their applications in biological imaging will be discussed. By regulating the design strategy and mechanism (e.g., ICT, PeT, and FRET) of the electronic and spectral characteristics of the fluorescent platforms, these chemical probes show high selectivity and diverse functions, which can be used for imaging of various physiological and pathological processes. Through the exploration of the rational response mechanism and design strategy, combined with a variety of imaging techniques, such as super-resolution imaging, photoacoustic (PA) imaging, etc., we have realized multimode imaging of the important biological analytes from the subcellular level to the in vivo level, which provides powerful means to study the physiological and pathological functions of these species and microenvironments. This Account aims to offer insights and inspiration for the development of novel fluorescent probes for biological imaging, which could provide powerful tools for the study of chemical biology. Overall, we represent a series of turn-on/turn-off/ratiometric fluorescent/PA probes to visually and dynamically trace biological species and microenvironments in cells and even in vivo that seek higher resolution and depth molecular imaging to improve diagnostic methods and clarify new discoveries related to chemical biology. Our future efforts will be devoted to developing multiorganelle targeted fluorescent probes to study the mechanism of subcellular organelle interaction and employing various dual-mode probes of NIR II and PA imaging to investigate the development of related diseases and treat the related diseases at subcellular and in vivo levels.

Abstract:Site-specific fluorescent labeling of proteins in vivo remains one of the most powerful techniques for imaging complex processes in live cells. Although fluorescent proteins in a variety of colors are useful tools for tracking expression and localization of fusion proteins in cells, these relative large tag can perturb protein folding, trafficking and function. Site-specific small-molecule fluorescent probe with advantages of small size, membrane permeability, low background and simple preparation will be an ideal tool in various studies of protein functions. This article summarizes the advances in selective labeling of proteins by using small-molecule fluo-rescent probes in recent years.

Abstract:Fluorescent and colorimetric probes are gaining interest in analyte detection, imaging, and real-time applications. However, it faces several challenges in achieving high sensitivity and selectivity, maintaining a strong signal-to-noise ratio, detecting analytes at extremely low concentrations, ensuring stability under different conditions and addressing issues related to cost, accessibility and instrumentation. Additionally, ensuring biological compatibility and enabling real-time monitoring are also challenges that need to overcome. We have investigated the sensing application of probe molecules towards various analytes like metal ions, anions, biomolecules, reactive oxygen species (ROS), reactive sulphur species (RSS), toxic agents and nanoparticles performed by our group together with critically comparing with the existing reports. This featured article provides the impact of fluorescent and colorimetric probes in advancing analytical techniques, imaging and real-time applications that transcend traditional boundaries.

Abstract:Super-resolution fluorescence microscopy is a powerful tool to visualize biomolecules and cellular structures at the nanometer scale. Employing these techniques in living cells has opened up the possibility to study dynamic processes with unprecedented spatial and temporal resolution. Different physical approaches to super-resolution microscopy have been introduced over the last years. A bottleneck to apply these approaches for live-cell imaging has become the availability of appropriate fluorescent probes that can be specifically attached to biomolecules. In this Perspective, we discuss the role of small-molecule fluorescent probes for live-cell super resolution microscopy and the challenges that need to be overcome for their generation. Recent trends in the development of labeling strategies are reviewed together with the required chemical and spectroscopic properties of the probes. Finally, selected examples of the use of small-molecule fluorescent probes in live-cell super resolution microscopy are given.

Abstract:A key current challenge in biological research is the elucidation of the that roles chemicals and chemical reactions play in cellular function and dysfunction. Of the available cellular imaging techniques, fluorescence imaging offers a balance between sensitivity and resolution, enabling the cost‐effective and rapid visualisation of model biological systems. Importantly, the use of responsive fluorescent probes in conjunction with ever‐advancing microscopy and flow cytometry techniques enables the visualisation, with high spatiotemporal resolution, of both specific chemical species and chemical reactions in living cells. Ideal responsive fluorescent probes are those that contain a fluorophore tethered to both a sensing unit, to ensure selectivity of response, and a targeting group, to control the sub‐cellular localisation of the probe. To date, probes that are both targeted and selective are relatively rare and most localised probes are discovered serendipitously rather than by design. A challenge in this field is therefore the identification of suitable fluorophore scaffolds that can be readily attached to both sensing and targeting groups. Here we review current strategies for dual‐functionalisation of fluorophores, highlighting key examples of targeted, responsive probes.

Abstract:In recent years, more and more chemists and biologists pay attention to the research of fluorescent probes, and great progress has been made in the design and application. In this review, we first discuss the excellent photophysical properties of 4-hydroxy-1,8-naphthalimide (HNI) structure, including excellent photostability, high fluorescence quantum yield, and easy modification of the structure. Then, by discussing the fluorescence behavior of the probe in the corresponding recognition process, the contributions of fluorescent probes built on the platform of HNI in the fields of chemical sensing, biological imaging, pharmaceutical chemistry, environment and food safety in the past ten years are systematically introduced (173 references). At the same time, we hope to develop more powerful fluorescent chemical sensors, which can play a more extensive and exciting role in the future.

Abstract:The emergence of bioorthogonal reactions has provided numerous opportunities for the design and fabrication of innovative fluorescence systems, particularly in the analytical field. These bioorthogonal reactions, including copper-catalyzed azide-alkyne cycloaddition (CuAAC), Staudinger ligation reaction, strain-promoted alkyne-azide cycloaddition (SPAAC), photoclick reactions, strain-promoted alkyne-nitrone cycloaddition (SPANC), and inverse-electron-demand Diels-Alder (IEDDA) reaction, have been extensively employed in the development of fluorescent probes. By combining a wide range of bioorthogonal reaction pairs with diverse fluorophores, fluorescent probes have been created to enable specific and selective sensing of biomolecules, drugs, and other biologically related organic compounds. This review provides a comprehensive overview of the development process of fluorescent probes based on bioorthogonal reactions. It covers the construction strategies, systematic classification, analysis, and applications of these fluorescent probes, while also discussing future prospects in this field.

Abstract:Life are governed by cellular complex metabolic processes. One of the on-going endeavours in the scientific community is to study and understand these dynamic biochemical reactions and the roles of biomolecules in sustaining life. Fluorescent probes have been widely used in the visualization of analytes in physiological and pathological processes, due to their advantages such as easy operation, low cost, high sensitivity, capability of multi-channel and real-time visualization in vivo. However, most fluorescent probes can only respond to single analyte, which is not suitable to mornitor multiple analytes in complex biological system. In recent five years, significantly more multi-analyte fluorescent probes based on 1, 8-naphthimide fluorophore have been designed in biological or environmental field. Categorized by the fluorescent mechanisms, such as single fluorescent mechanism (e. g. photoinduced electron transfer (PET), intramolecular charge transfer (ICT) and fluorescence resonance energy transfer (FRET)), dual fluorescent mechanisms (e. g. PET-ICT, PET-FRET, and ICT-FRET) and so on, this review highlights the recent progress in designing strategies, corresponding recognition process, optical property and cell imaging of 1, 8-naphthalimide-based multi-analyte fluorescent probes. Finally, the development trend and possible challenges of such fluorescent probes are prospected.

Abstract:In recent years, with the rise of precision therapy and other frontier fields, the demand of the high quality fluorescent probes with extremely low toxicity, good biocompatibility, strong stability and high brightness is more urgent than ever. Proteins that with abundant active groups and special pockets can bind with different probes to alter the fluoresce properties, and some can even greatly enhance the fluorescence signals. Moreover, some proteins can simultaneously bind with two different molecules to fabricate fluorescence resonance energy transfer (FRET) probes. Recent progress witnessed that the protein-mediated strategies have provided a promising way for enhancing the performance of fluorescent probe. This article will review the up-to-date progress of protein-mediated fluorescent probes. We firstly provide a systemically on the principle of protein fluorescent probes fabrication and the interaction mechanisms between proteins and molecular dyes. Then the bioapplications of these fluorescent probes in bioimaging and biosensing were reviewed. And finally the current challenges and perspectives were discussed in-depth. We hope that this review can provide some new ideas and inspiration for relevant researchers to develop a new generation of fluorescent probes.
