Abstract:<p>In order to improve the degradation rate of silica framework and the targeted release ability of tumor drugs, this paper used a one-pot synthesis method to synthesize a redox with a silsesquioxane framework disulfide bridge and a particle size of about 150 nm. The triggered biodegradable organic-inorganic hybrid nanocapsule is a new and unique hollow mesoporous nanocapsule that is sequentially coupled with bovine serum albumin (BSA) and folic acid (FA) for the targeted release of adriamycin. As a "control gate", BSA can prevent the premature release of drugs until the BSA coating "opens the door" in response to Glutathione(GSH) to achieve drug release. FA specifically recognizes the folate receptor on the tumor surface to achieve tumor targeting. Under acidic PH and GSH, it can achieve accelerated and complete release of drugs, and effectively consume intracellular GSH, help break the redox barrier in hypoxic cells, cause oxidative stress, and effectively inhibit tumor growth.</p>

Abstract:In situ forming redox responsive nanoparticles have been developed based on amphiphilic copolymers-phototriggered disulfide-cross-link macromolecules (PDCM). Upon 405 nm light irradiation, the macrocyclic thiol caged coumarin phototrigger in PDCM can release free thiols, and these free thiols subsequently realize in situ disulfide cross-link via reacting with a pyridyl disulfide group inside the PDCM assembled nanoparticles. The phototriggered disulfide-cross-link strategy can be conducted rapidly, conveniently, and cleanly without adding any cross-linkers or catalysts. Via changing irradiation condition, nanoparticles with different cross-link densities can be formed. These nanoparticles can encapsulate hydrophobic guest molecules with good stability and achieve redox-triggered release under GSH reduction. Intracellular experiments show that these nanoparticles can be used as promising drug carriers.

Abstract:A versatile one-pot strategy for the preparation of reversibly cross-linked polymer-coated mesoporous silica nanoparticles (MSNs) via surface reversible addition-fragmentation chain transfer (RAFT) polymerization is presented for the first time in this paper. The less reactive monomer oligo(ethylene glycol) acrylate (OEGA) and the more reactive cross-linker N,N'-cystaminebismethacrylamide (CBMA) are chosen to be copolymerized on the external surfaces of RAFT agent-functionalized MSNs to form the cross-linked polymer shells. Owing to the reversible cleavage and restoration of disulfide bonds via reduction/oxidation reactions, the polymer shells can control the on/off switching of the nanopores and regulate the drug loading and release. The redox-responsive release of doxorubicin (DOX) from this drug carrier is realized. The protein adsorption, in vitro cytotoxicity assays, and endocytosis studies demonstrate that this biocompatible vehicle is a potential candidate for delivering drugs. It is expected that this versatile grafting strategy may help fabricate satisfying MSN-based drug delivery systems for clinical application.
Facile synthesis and polymerization of 1,4,5-oxadithiepan-2-one for disulfide-based redox-responsive drug delivery

Abstract:Chemotherapeutic drugs have been shown to be promising for the treatment of tumors, however, they are associated with several disadvantages including low aqueous solubility, non-specificity, and poor plasma stability. Hence, they accumulate in the health organs and create several adverse effects. Drug encapsulated stimuli-responsive nanoparticles bearing redox-responsive disulfide linkages have emerged as promising delivery vehicle to address these limitations. A glutathione (GSH) responsive disulfide-based drug delivery system based on a polyethylene glycol methyl ether (PEGME) and a polydisulfide (PDS) diblock copolymer has been developed. The cyclic lactone containing a disulfide link, 1,4,5-oxadithiepan-2-one, was prepared using a thiol-free method utilizing the reaction of 2-bromoethyl bromoacetate with sodium disulfide. Ring-opening polymerization (ROP) of 1,4,5-oxadithiepan-2-one using 1-butanol in the presence of Sn(oct)2 generated the homopolymer PDS whereas the diblock copolymer PEGME-b-PDS was obtained when PEGME was used as a macroinitiator. In aqueous solution, PEGME-b-PDS self-assembled into spherical-shaped micelles with a hydrodynamic radius of 146 and 116 nm as measured by Field-emission Scanning Electron Microscopy (FESEM) and Dynamic Light Scattering (DLS), respectively. At pH = 7.4, a Doxorubicin (DOX) loaded PEGME-b-PDS micelle showed a cumulative release of ∼98.2, ∼9.9, and ∼1.9 % in presence of 2 mM, 2 μM DTT and in absence of DTT, respectively clearly demonstrating the redox responsiveness of the PDS block. The best fit of kinetic data using with the Korsmeyer-Peppas model suggests a Fickian diffusion as well as chain-relaxation controlled drug release process. DOX loaded micelles and free DOX displayed similar cytotoxicity towards both 4T1 and MCF-7 cells indicating that the encapsulation inside the micelle did not affect the anticancer efficacy of the drug. The drug loaded micelle showed higher cellular uptake than free DOX in both 4T1 and MCF-7 lines and lower cellular uptake than free DOX in 3T3-L1 cell lines. This polymeric micellar nanoparticle may be further exploited for encapsulation of other hydrophobic drugs.

Abstract:Hollow mesoporous silica spheres (HMSS) have a hierarchical mesoporous structure composed of a hollow cavity and mesoporous shell available for high capacity drug storage. The covalent attachment of ZnO quantum dots (QDs) to the HMSS outer surface as gatekeepers via disulfide-conjugated two amide linkages encapsulated the anticancer drugs doxorubicin (DOX) within the HMSS cavities and pores, and minimized premature release of the drug. The controlled release of the drug from the ZnO-gated HMSS delivery system was realized by the dissolution of ZnO QDs upon a decrease in pH and cleavage of the disulfide bonds, which indicates that the pH- and redox-responsive controlled release of the drugs could be synergically stimulated by tumor cells with weakly acidic environments and high-expressed glutathione. The constructed ZnO-gated HMSS delivery system has promising applications in site-specific drug release for tumor chemotherapy.

Abstract:<p>Self-assembled nanostructures are highly promising for controlled delivery of therapeutic and diagnostic agents. However, for conventional drug delivery nanosystems, there exist some intrinsic limitations such as low drug-loading contents, premature and burst release, nanocarrier matrix-associated toxicity and immunogenicity, and poor shelf stability. To address these issues, the covalent integration of active drug molecules into prodrug and polyprodrug amphiphiles and fabrication of self-delivery nanomedicines via controlled molecular self-assembly have emerged as a new paradigm. Moreover, it is crucial to achieve on-demand and selective activation of prodrugs and polyprodrugs in a spatiotemporally controlled manner, thus considerably reducing the occurrence of systemic toxicities. In this context, dynamic variations of reductive/oxidative (redox) milieu across normal and pathological tissues, cells, and cytoplasmic compartments provide accessible biochemical stimuli for triggered release of intact drugs. In this review, we highlight recent progresses on emerging applications of redox-activatable nanostructures self-assembled from prodrug and polyprodrug amphiphiles.</p>

One-Pot Synthesis of Redox-Labile Polymer Capsules Via Emulsion Droplet-Mediated Precipitation Polymerization

Abstract:Monodisperse poly(vinylcaprolactam) (PVCL)-based capsules are prepared by precipitation polymerization of vinylcaprolactam (VCL) onto dimethyldiethoxysilane (DMDES) emulsion droplets and removal of the DMDES templates by ethanol. Polymer chains in the shells can be cross-linked during the polymerization by disulfide-containing cross-linker N,N'-bis(acryloyl) cystamine, which endows the capsules with an excellent redox-labile property. Versatility of this technique to prepare capsules with diverse components is demonstrated by the copolymerization of methacrylic acid (MAA) and VCL in the shell to prepare poly(vinylcaprolactam-co-methacrylic acid) (P(VCL-co-MAA)) capsules. The disulfide-bonded capsules can degrade efficiently into low molecular weight species (ca. 1200 Da) when the capsules are incubated with 10 mM glutathione (GSH) as the reducing agent. Delivery of the anticancer drug (doxorubicin, DOX) was also investigated in the P(VCL-co-MAA) capsules. The cumulative in vitro release of DOX-loaded capsules allows a relatively low DOX release at pH 7.4. However, a burst release (ca. 90% in 6 h) of DOX was observed in the presence of 10 mM GSH. Cell viability assays show that the P(VCL-co-MAA) capsules have negligible cytotoxicity to HeLa cancer cells. In comparison, DOX-loaded P(VCL-co-MAA) capsules cause significant cell death following internalization. The reported capsules represent a novel and versatile class of stimuli-responsive carriers for controlled drug delivery.

Abstract:The applications of bioresponsive materials are limited by the low levels of physiological triggers and spatiotemporal barriers in vivo. To address these issues, a light‐controlled “Trojan horse” strategy is proposed by encapsulating a photo‐caged reducing agent within reduction‐responsive polymer vesicles. The polymersomes act as an enzyme‐inspired nanoreactor for efficient photo‐synthesis of dithiothreitol in situ, which attacks the disulfide bonds in polymer backbones and disintegrates the vehicles from the inside in a nanoconfined space. The assembly‐catalyzed photoreduction reaction overcomes the temporal and spatial barriers for hyper‐responsivity and enables ultrafast drug release even at 0.014 mm of dithiothreitol residues, a concentration 715 times lower than that required to cleave disulfide bonds. In addition, in both vitro and vivo, the equipment of upconverting nanoparticles and photothermal agents creates a near‐infrared light‐activated and self‐heating nanoreactor, which allows for efficient intracellular drug delivery and excellent photo‐chemo‐immunotherapy of tumors. This work presents a new approach to designing smart materials and a promising platform for on‐demand drug delivery applications. Abstract:Reduction-responsive biodegradable polymeric micelles based on functional 2-methylene-1,3-dioxepane (MDO) copolymers are developed and investigated for triggered doxorubicin (DOX) release. The MDO-based copolymers P(MDO-co-PEGMA-co-PDSMA) are synthesized via the simple one-step radical ring-opening copolymerization of MDO, poly(ethylene glycol) methyl ether methacrylate (PEGMA), and pyridyldisulfide ethylmethacrylate (PDSMA). The copolymers can self-assemble to form micelles in aqueous solution. DOX, a model anticancer drug, is loaded into the micelles with the drug loading content (DLC) of 11.3%. The micelles can be disassembled under a reductive environment (10 x 10(-3)m glutathione), which results in a triggered drug release behavior. The glutathione-mediated intracellular drug release of DOX-loaded micelles is investigated against A549 cells. Confocal laser scanning microscopy (CLSM) results demonstrated that DOX-loaded micelles exhibits faster drug release in glutathione monoester (GSH-OEt)-pretreated A549 cells, compared with untreated and buthionine sulfoximine (BSO)-pretreated A549 cells. Based on the facile synthetic strategy, the reduction-sensitive biodegradable micelles with triggered intracellular drug release are promising for anticancer drug delivery.

Abstract:Redox and pH double stimulus-responsive drug delivery platform based on chitosan (CS), carbon dots (CDs) and hyaluronic acid (HA) gated mesoporous silica nanoparticles (MSN) for drug delivery. The DDS showed excellent drug loading and release properties with good biocompatibility. In addition, the DDS exhibited fluorescence bioimaging properties. The prepared MSN-CS-CDs@HA is demonstrated to be a promising cancer treatment platform.

Abstract:Based on disulfide-enriched multiblock copolymer vesicles, we present a straightforward sequential drug delivery system with dual-redox response that releases hydrophilic doxorubicin hydrochloride (DOX·HCl) and hydrophobic paclitaxel (PTX) under oxidative and reductive conditions, respectively. When compared to concurrent therapeutic delivery, the spatiotemporal control of drug release allows for an improved combination antitumor effect. The simple and smart nanocarrier has promising applications in the field of cancer therapy.
A Dual-Responsive Nanocapsule Via Disulfide-Induced Self-Assembly for Therapeutic Agent Delivery.

Abstract:One-step synthesis of fluorescent molecules (SNBDP) containing one disulfide bond and two o-nitrobenzyl groups was demonstrated via multi-component Passerini reaction. This hydrophobic SNBDP could self-assemble into nanocapsules (SNBDP NCs) in aqueous solution via disulfide-induced assembly. The obtained nanocapsules were stable in aqueous solution for several weeks and exhibited enhanced fluorescence when nanocapsules were destroyed due to disaggregation-induced emission. The nanocapsules not only were reduction-sensitive and light-responsive, but also could be endocytosed by HeLa cells for cellular imaging. The enhanced fluorescence in the glutathione (GSH) pretreated HeLa cells showed that the compound was reduction-sensitive in living cells. In vitro WST-8 assays showed the nanocapsules were biocompatible and could further be used as drug delivery carriers. Indocyanine green (ICG), a clinically approved NIR dye, was loaded into the nanocapsules (ICG@SNBDP NCs). ICG@SNBDP NCs showed enhanced photothermal efficacy compared with same concentration of free ICG under 808-nm laser irradiation. Consequently, ICG@SNBDP NCs upon NIR irradiation can effectively kill cancer cells through local hyperthermia. These results highlight the potential of disulfide-induced nanocapsules as smart nanoparticles for cellular imaging and therapeutic agent delivery.

Abstract:Many medical and chemical applications require target molecules to be delivered in a controlled manner at precise locations. Metal-organic frameworks (MOFs) have high porosity, large surface area, and tunable functionality and are promising carriers for such purposes. Current approaches for incorporating target molecules are based on multistep postfunctionalization. Here, we report a novel approach that combines MOF synthesis and molecule encapsulation in a one-pot process. We demonstrate that large drug and dye molecules can be encapsulated in zeolitic imidazolate framework (ZIF) crystals. The molecules are homogeneously distributed within the crystals, and their loadings can be tuned. We show that ZIF-8 crystals loaded with the anticancer drug doxorubicin (DOX) are efficient drug delivery vehicles in cancer therapy using pH-responsive release. Their efficacy on breast cancer cell lines is higher than that of free DOX. Our one-pot process opens new possibilities to construct multifunctional delivery systems for a wide range of applications.

Abstract:Tumor redox stimulus-responsive nanoparticulate drug delivery systems (nano-DDSs) have attracted considerable attention due to their thermodynamically stable microstructures and well-controlled drug release properties. However, drug-loading nanoparticle conformation and redox-triggered drug release mechanisms at the molecular level remain unclear. Herein, doxorubicin-conjugated polymers were constructed using disulfide bonds as linkages (PEG-SS-DOX), which loaded photosensitizer chlorin e6 (Ce6). We integrated multiple scale dynamic simulations (density functional theory (DFT) calculation, atomistic molecular dynamics (MD) simulation and dissipative particle dynamics (DPD) simulations) to elucidate the assembly/drug release dynamic processing. First, it was revealed that the emergence of the calculated bond flexible angle of disulfide bonds facilitated the assembly behavior and improved the stability of conformation. Sorted by the binding model, hydrogen bonding accounted for the major interactions between polymers and photosensitive drugs. DPD simulations were further delved into to acquire knowledge regarding the drug-free self-aggregation and Ce6-loaded assembly mechanism. The results show that nano-assembly conformation not only depended on the concentration of polymers, but also were associated with the polymer-drug ratio. Different from dicarbon bond-bridging polymers, disulfide bonds would contribute to the breakage of the polymer and the rapid release of DOX and Ce6. Our findings provide deep insights into the influence of redox-responsive chemical linkages and offer theoretical guidance to the rational design of specific stimulus-responsive nano-DDSs for cancer therapy.
Reducing Agent Triggered Templated Synthesis of Dynamic Covalent Poly(disulfide)s Nanonetwork: Remarkable Tuning in Noncovalent Encapsulation Stabilities and Cargo Release

Abstract:Here, we demonstrated a new methodology for the templated synthesis of cross-linked poly(disulfide)s based dynamic covalent nanonetwork as highly stable potential delivery vehicle in the field of chemotherapeutic applications. The synthesis is carried out by the treatment of nanoaggregate of biomass-derived lipoic acid based amphiphilic monomer with reducing agent at room temperature in open air condition and aqueous medium. The hydrodynamic diameter of the naoaggregate is found to be ~130 nm as probed by dynamic light scattering experiment. The control over cross-linking density is achieved by varying the ratio of monomer to reducing agent. The crosslinking percentage varied from ~13% to ~100%. This gives the opportunity of fine-tuning the stability of the nanocarrier itself, noncovalent encapsulation stabilities and kinetics of cargo release, which are highly relevant in the field of drug delivery applications. For highly crosslinked nanonetwork, in simulated redox condition of cancer cell, ~80% guest release was noted from the nanonetwork in a sustained manner. The controlled depolymerization of the polymer is accomplished by the use of specific mol% of the same reducing agent. Finally, the reversibility and recyclability of the poly(disulfide)s to the monomeric form is achieved by the treatment of the polymer with the external thiol in presence of an organic base.

Reduction-Triggered Transformation of Disulfide-Containing Micelles at Chemically Tunable Rates.

Abstract:A dilemma exists between the circulation stability and cargo release/mass diffusion at desired sites when designing delivery nanocarriers and in vivo nanoreactors. Reported herein are disulfide-crosslinked (DCL) micelles exhibiting reduction-triggered switching of crosslinking modules and synchronized hydrophobic-to-hydrophilic transition. Tumor cell targeted DCL micelles undergo cytoplasmic milieu triggered disulfide cleavage and self-immolative decaging reactions at chemically adjustable rates, generating primary amine moieties. Extensive amidation reactions with neighboring ester moieties then occur because of the high local concentration and suppression of the apparent amine pKa value within the hydrophobic cores, thus leading to the transformation of crosslinking modules and formation of tracelessly crosslinked (TCL) micelles, with hydrophilic cores, inside live cells. We further integrate this design principle with theranostic nanocarriers for selective intracellular drug transport guided by enhanced magnetic resonance (MR) imaging performance.

Abstract:The development of efficient multiresponsive drug delivery systems (DDSs) to control drug release has been widely explored. Herein, a facile strategy is reported that enables the micelles of the selenium‐containing polymer with the drug to be encapsulated in metal‐organic frameworks (MOFs), which serves as multiresponsive drug release by employing the selenium‐containing polymers with redox‐triggered property and the MOFs with pH‐triggered property in DDS. In this case, the micelles of selenium‐containing polymers, as core easily disassembles in the presence of redox agents, can then release the drug in MOFs matrixes. The ZIF‐8 (one type of MOFs) crystal frameworks serving as shell can collapse only under low pH conditions, and the drug can be further released. In the presence of external redox agents as well as the pH stimuli, the prepared nanocomposite (P@ZIF‐8) drug system exhibits the capability of multiresponsive release of the doxorubicin (DOX) and possesses good selectivity in releasing the DOX under low pH conditions instead of normal pH conditions. In addition, the merits of P@ZIF‐8 such as good biocompatibility, multiresponsive release properties, and especially the selective release properties under different pH conditions make the materials highly promising candidates for the realization of controlled drug delivery in tumor tissue systems.
Redox- and Temperature-Controlled Drug Release from Hollow Mesoporous Silica Nanoparticles.

Abstract:A controlled drug-delivery system has been developed based on mesoporous silica nanoparticles that deliver anticancer drugs into cancer cells with minimized side effects. The copolymer of two oligo(ethylene glycol) macromonomers cross-linked by the disulfide linker N,N'-bis(acryloyl)cystamine is used to cap hollow mesoporous silica nanoparticles (HMSNs) to form a core/shell structure. The HMSN core is applied as a drug storage unit for its high drug loading capability, whereas the polymer shell is employed as a switch owing to its redox/temperature dual responses. The release behavior in vitro of doxorubicin demonstrated that the loaded drugs could be released rapidly at higher temperature or in the presence of glutathione (GSH). Thus, the dual-stimulus polymer shell exhibiting a volume phase transition temperature higher than 37 °C can effectively avoid drug leakage in the bloodstream owing to the swollen state of the shell. Once internalized into cells, the carriers shed the polymer shell because of cleavage of the disulfide bonds by GSH, which results in the release of the loaded drugs in cytosol. This work may prove to be a significant development in on-demand drug release systems for cancer therapy.
Disulfide-Linked/Peptide-Incorporated Macrocycles: Unique Redox-Responsiveness and Application for Intracellular Cargo-Delivery

Abstract:The use of disulfide-linked macrocycles for biomedicine applications was not explored, which is, at least partially, due to the lack of a straightforward strategy to incorporate a cell-surface binding ligand into the macrocycles for facilitating cellular uptake. Herein, we discover that twin disulfides consisting of a flexible peptide and a structurally rigid organic molecule can be transformed into stable and trimeric macrocycles upon triggering with thiols. The disulfide linked/peptide incorporated macrocycles are substantially more stable in weakly reducing environments compared to their non-cyclic analogues due to the cooperativity of the three interlinked disulfide bonds. We further demonstrated that the trimeric macrocycles can be a promising scaffold for the development of intracellular cargo-delivery systems by incorporating a cell-penetrating peptide into the macrocycles.

Abstract:Direct delivery of proteins to the cytosol of cells holds tremendous potential in biological and medical applications. Engineering vehicles for escorting proteins to the cytosol in a controlled release fashion has thus generated considerable interest. We report here the preparation of redox-responsive single-protein nanocapsules for intracellular protein delivery. Through in situ interfacial polymerization, the target protein is noncovalently encapsulated into a positively-charged polymeric shell interconnected by disulfide-containing crosslinkers. The dissociation of the polymeric shell under reducing conditions and the subsequent release of protein were confirmed using cell-free assays in the presence of glutathione (GSH). The nanocapsules were demonstrated to be efficiently internalized into the cells and to release the protein in the reducing cytosol. Using the nanocapsule as a vehicle, we showed that active caspase 3 (CP-3) can be delivered and can induce apoptosis in a variety of human cancer cell lines, including HeLa, MCF-7 and U-87 MG. Our approach therefore presents an effective intracellular protein delivery strategy for therapeutic, diagnostic and reprogramming applications.
