Abstract:Formaldehyde, as one of the simplest reactive carbonyl species（RCS）, is regarded as a potential carcinogen and a sick house syndrome gas. Recent studies have shown that abnormally high levels of formaldehyde may result in cognitive decline and spatial memory deficits, asthmatic symptoms,Alzheimer’s disease, and cancer. Due to the harmfulness of high levels of formaldehyde in nature and humans, it is of great significance to further elucidate the roles and functions of formaldehyde by a noninvasive detection approach. Fluorescence imaging has become a powerful and popular tool in monitoring bio-species owing to their high sensitivity and selectivity, excellent spatiotemporal resolution and non-invasion nature. Therefore, fluorescent probes are widely applied to track and detect formaldehyde in vitro and in vivo which have attracted more and more interest recently. This review focuses on various strategies to design the fluorescent probes for detecting formaldehyde based on different recognition groups.

Abstract:A simple and stable fluorescent probe L based on a novel ortho-diaminorhodamine derivative for discrimination and detection of formaldehyde (FA), methylglyoxal (MGO) and oxalaldehyde (OA) from other aldehydes has been developed. This reaction-based probe, displayed distinct emission patterns for FA (turn-on) and MGO (turn-off) by single wavelength excitation, can be used for imaging of FA and MGO in living cells.

Abstract:We report a novel turn-on two-photon fluorescent probe (TPNF) for detecting formaldehyde based on aza-Cope rearrangement and intramolecular charge transfer (ICT). The probe, containing a 1,8-naphthalimide dye with a homoallylamino group, has was shown with high selectivity, sensitivity and low cytotoxicity and it was capable of detecting formaldehyde in live cells. Moreover, a time-dependent density functional theory (TD-DFT) calculation was performed to demonstrate the turn on mechanism of the probe via ICT effect. Finally, a zebrafish larvae imaging experiment reveals that TPNF displayed a significant signal change when treated with FA in vivo. These results indicated that TPNF is a promising tool in more biomedical researches. (C) 2016 Elsevier B.V. All rights reserved.

Abstract:Formaldehyde (FA) is one of the most important reactive carbonyl species. It is not only exogenously generated from the environment but also continuously produced in most organisms. Low concentrations of FA take part in several important physiological processes, while elevated levels of FA are associated with different kinds of diseases. Fluorescent probes have emerged as promising chemical tools for FA detection and real-time visualization in food, air, and living organisms, etc., enabling the discovery of the diverse functions of FA in physiology and pathology processes. This review will summarize recent progress in the development of FA probes, including both small-molecular organic FA probes and functional material-based probes. In addition, prospects for future design and research will be also discussed.

Abstract:Formaldehyde (FA) is not only used as a basic chemical for industrial products, but also a necessary metabolite for regulating human physiological activities. However, excessive intake from the external environment or unbalanced metabolism homeostasis of the internal environment would cause critical diseases to the human body such as organ cancers, Alzheimer?s disease and so on. The high sensitivity and selectivity with visualization and in-situ detection by means of small-molecular fluorescent probe, provides superiority for identification of FA and biological image in vitro and in vivo, and a new detection method for the trace detection of FA in real products as well. In the past five years, fluorescent probes of FA have developed rapidly. This article mainly reviews the research progress of fluorescent probes for FA recognition and applications, including the general reaction mechanisms and the applications in organisms and samples with commercial products. It is concluded that the small molecular fluorescent probes should be continuously improved in structures and optical characteristics to not only assist long-term research in biomedicine, but also lead to the achievable goals of fast (in situ) detecting FA in practice from actual products.

Abstract:Formaldehyde (FA) plays important roles in many physiological processes and closely relates to human health. However, the specific detection of FA remains a great challenge. Herein, we report a rational strategy for specific detection and imaging of FA through combining characteristic structure of a fluorophore merged with the modified ortho-phenylenediamine as a recognition moiety. After the reaction of FA and the propyl-modified ortho-phenylenediamine, the probe NP1 and NP2 based on naphthalimide fluorophore exhibited the turn-on fluorescence response for the detection of FA. In particular, the probe NP2 with the greater steric hindrance presented superior specificity for imaging the endogenous FA in living cells. This work may serve as a general strategy to design specific recognition moieties for FA probes.

Abstract:Formaldehyde (FA) is a harmful substance commonly found in the environment, so effective testing is essential. Consequently, we designed and synthesized a novel fluorescent probe, which is based on covalent post-synthetic modification (PSM) of UiO-66-NH2 by naphthylamide (NI). The combination of the metal-organic frameworks (MOFs) and the covalent PSM makes the probe with large amount of hydrazine group exposed on the surface, which results excellent FA sensing performance like an ultrafast response time (20 s) and a low detection limit of 0.167 μM. The presence of covalent bonds can lock the fluorophore and prevent strong π-π interactions to exhibit aggregation-induced emission (AIE). Further we confirmed that such probe can be applied to real water samples and portable test strip experiment. In addition, experimental evidence such as fluorescence lifetime decay curves suggest that the main mechanism of fluorescence quenching is Förster resonance energy transfer (FRET).

Abstract:An important challenge for reaction-based fluorescent probes is that they generally require analyte consumption for fluorescence signal generation, thus creating potential perturbation of native analyte homeostasis or change of local concentrations. Herein, we reported two formaldehyde (FA) regeneration fluorescent probes, NAP-FAP-1 and NAP-FAP-2. An unprecedented regiospecific FA-induced intramolecularity strategy is implemented in the probe design, which adopts 3-(benzylamino)-succinimide as the FA-selective reaction group. The probes are able to capture the analyte molecule, induce regiospecific imide bond cleavage, and then release the captured FA molecule with simultaneous fluorescence turn-on response via a unique dual PeT/ICT quenching mechanism. The probes have shown potentials in detection, comparison, and imaging of FA levels intracellularly and inside lysosomes. These features make them useful for the study of FA homeostasis and functions in biological systems with minimal perturbation.

Abstract:Energy transfer between fluorescent dyes and quenchers is widely used in the design of light-up probes. Although dual quenchers are more effective in offering lower background signals and higher turn-on ratios than one quencher, such probes are less explored in practice as they require both quenchers to be within the proximity of the fluorescent core. In this contribution, we utilized intramolecular motion and photoinduced electron transfer (PET) as quenching mechanisms to build super-quenched light-up probes based on fluorogens with aggregation-induced emission. The optimized light-up probe possesses negligible background and is able to detect not only free formaldehyde (FA) but also polymeric FA, with an unprecedented turn-on ratio of >4900. We envision that this novel dual quenching strategy will help to develop various light-up probes for analyte sensing.

Abstract:A novel and simple camphor-based fluorescent probe (CH) was synthesized for detecting formaldehyde (FA) with high specificity and efficiency. A significantly enhanced green fluorescence was clearly observed only with the addition of FA because of specific recognition site homoallylamino group of FA. After treating with FA, the homoallylamino group on probe CH was converted to aldehyde group via the 2-aza-Cope rearrangement mechanism, which was demonstrated with high-resolution mass spectrometer (HRMS) analysis and density functional theory (DFT). Moreover, the experiment results manifested that the detection of FA with probe CH can be performed in a nearly perfect aqueous solution with low detection limit, large Stokes shift and wide pH adaptation range. Meanwhile, a simple filter paper, loaded with probe CH, was available to detect gaseous FA. The probe CH was also successfully applied to the detection of FA in real food and water samples. Furthermore, the fluorescence imaging experiment verified that the probe CH can effectively monitor FA in living zebrafish.

Abstract:A two-photon nanoparticle probe was designed and developed based on the principle of intermolecular interaction of the aggregation-induced locally excited emission luminescence mechanism. The probe has the advantages of simple synthesis, convenient use, strong atomic economy, good biocompatibility, and photobleaching resistance. It can produce a specific and sensitive response to formaldehyde, help detect FA in normal cells and cancer cells, and is expected to become a specific detection probe for FA in vitro and in vivo.

Abstract:As an environmental pollutant and carcinogen, formaldehyde (FA) is widely used in many fields, such as building materials, biomedicine, chemical industry, and so on. In vivo, as an active carbonyl substance, FA also plays a very important role. However, excessive FA in living organisms can cause many diseases. Therefore, it is extremely important to ensure the balance of FA concentration in organisms. Based on this, a regenerated FA fluorescence probe NE-FA was studied for the detection of FA. The probe used hydroxynaphthalimide as a fluorophore and 2-methylaminobenzoate ester as a recognition receptor. After the probe reacts with FA, it can induce ester hydrolysis to regenerate FA, while simultaneously releasing enhanced fluorescent signal. In the selective experiments, the probe exhibits good selectivity to FA. In addition, the probe has been successfully applied to monitor FA in cells and zebrafish. A novel hydroxynaphthalimide-derived regenerative fluorescent probe was developed to monitor formaldehyde in cells and zebrafish.

Abstract:The fluorescence monitoring of formaldehyde in real environmental samples and live plant tissues is of great importance for physiological and pathological studies. However, there is a lack of suitable chemical tools to directly trace and measure the formaldehyde activity in bio-systems, and developing effective and, in particular, selective sensors for mapping formaldehyde in live tissues still remains a great challenge. Here, we demonstrate for the first time that the ratiometric fluorescence monitoring of formaldehyde in live plant tissues is achieved with a newly developed ratiometric fluorescent probe, FAP, which effectively eliminated interference from other comparative analytes. Live tissue analyses reveal that FAP can potentially detect exogenous and endogenous formaldehyde in live Arabidopsis thaliana tissues, exposing a potential application for biological and pathological studies of formaldehyde.

Abstract:A new fluorescent chemosensor DTH for distinguish detection formaldehyde (FA) and acetaldehyde (AA) has been synthesized based on 2, 5-dihydroxy-p-benzenedicarbonamide as fluorophore and hydrazine as reaction group. DTH is a ratiometric probe for formaldehyde with fluorescence quantum yield is 0.055 and a turn on probe for acetaldehyde with a higher fluorescence quantum yield (0.122) due to different electron donating property of -H (formaldehyde) and -CH3 group (acetaldehyde), FA-DTH and AA-DTH, which were products of formaldehyde and acetaldehyde with DTH, respectively, showed different fluorescence variation. Interestingly, the maximum fluorescence emission peak wavelength varies with different molar ratio of the mixture solution of FA/AA and it was found that the wavelength is linear with the molar ratio of FA/AA, demonstrating that DTH fluorescence probe can be used not only to distinguish detect formaldehyde and acetaldehyde, but also to analyze the approximate content of formaldehyde and acetaldehyde in the mixed solution. Therefore, DTH is an excellent probe, which is expected to be applied in cells and distinguish detection of formaldehyde and acetaldehyde in the environment.

Abstract:Formaldehyde (FA) is one of reactive carbonyl species (RCS), which is involved in various physiological and pathological processes of organisms. Herein, a novel fluorescent probe ( FAP1 ) containing lysosomal localization group for the detection of FA was designed and synthesized. The probe exhibited good fluorescence recognition performances for FA, such as large Stokes shift (130 nm), specific selectivity, good anti-interference ability, and low minimum detection limit (16 nM). It is worth mentioning that the probe has good biocompatibility and could be applied to the imaging detection of FA in lysosome.

Abstract:Formaldehyde (FA) has been detected via the Hantzsch reaction for many decades. However, the Hantzsch reaction has been rarely used to detect FA in biological systems due to the disadvantages of small-molecule probes (including toxicity and poor water solubility). In this study, polymeric fluorescent probes were developed to resolve these issues associated with small molecules, and FA in living systems was successfully detected via the Hantzsch reaction. These water-soluble polymers were easily scaled-up (∼25 g) by radical polymerization using commercial monomers. These polymers exhibited similar, albeit better, sensitivity to FA compared to water-soluble small molecules, primarily indicative of the advantages of polymers for the detection of FA via the Hantzsch reaction. The polymer structures were highly biocompatible with the probes; thus, these polymers can effectively detect endogenous FA in cells or zebrafish in a safe manner. This result confirmed the superiority of polymers in safety as biocompatible materials. This study highlights a straightforward method for exploring probes for the detection of FA in living systems. It offers functional polymers for bioimaging and extends the application scope of the Hantzsch reaction, reflecting the utility of a broad study of organic reactions in interdisciplinary fields as well as possible key implications in organic chemistry, analytical chemistry, and polymer chemistry.

Abstract:Amine moiety‐based fluorescence probes 2‐(phenylethynyl)aniline (BA‐1), 3‐(phenylethynyl)aniline (BA‐2), and 4‐(phenylethynyl)aniline (BA‐3) were synthesized from coupling reaction with diphenylacetylene as fluorophore and amino group as acceptor. The different formaldehyde detection performance of BA‐1, BA‐2 and BA‐3 revealed that the position of amino group on the benzene ring of diphenylacetylene had a significant impact on the recognition for formaldehyde. A novel amine moiety‐based fluorescent probe (BA) was prepared using diphenylacetylene as fluorophore and amino group as acceptor. The probe achieves detection of formaldehyde through fluorescence enhancement by inhibition the photoinduced electron transfer (PET) between diphenylacetylene and amino group. And the different formaldehyde detection performance of 2‐(phenylethynyl)aniline (BA‐1), 3‐(phenylethynyl)aniline (BA‐2) and 4‐(phenylethynyl)aniline (BA‐3) indicates that the position of amino recognition group on the benzene ring of diphenylacetylene has a significant impact on the detection ability of this type fluorescent probe. When the amino group is in the ortho‐position of the benzene ring, probe BA‐1 can rapidly enhance the fluorescence intensity within 20 min with limit of detection as low as 0.75 μM; When the amino group is located in meta‐position of benzene ring, the solution of BA‐2 also exhibited a certain degree of fluorescence enhancement, but it is weaker than BA‐1; When the amino group is at the para‐position of benzene ring, BA‐3 has almost no response to formaldehyde.

Abstract:Formaldehyde (FA), as the simplest endogenous carbonyl molecule, participates in many biosynthesis and metabolism in living organisms, such as nucleotides and adenosine triphosphate (ATP). FA concentrations are sub-millimolar in the normal healthy body, but can rise significantly in a number of disease pathologies. As a result, detecting endogenous FA is critical for illness diagnosis and rehabilitation therapy monitoring. Recent studies have focused on the FA-responsive turn-on fluorescence probe, which has huge promise in the detection and visualization of FA in living cells and organisms, as well as exceptional use in disease diagnosis and therapeutic monitoring. This review summarizes the fluorescence luminescence mechanism and design concepts of FA fluorescent probes, as well as their recent applications in bioimaging and illness diagnostics. Additionally, this article indicates the present dilemma of FA-responsive fluorescent probe, including selectivity, specificity, and detection mode, which may provide references for the development of FA-responsive fluorescent probes.
