Abstract:Human cerebrospinal fluid (CSF) is an ideal source for identifying biomarkers for neurodegenerative diseases such as Alzheimer's disease (AD), Parkinson's disease (PD), and dementia with Lewy bodies (DLB). Proteomics has been used to analyze CSF in order to discover disease-associated proteins and elucidate the basic molecular mechanisms that either cause, or result from, central nervous system disorders. However, before undertaking a rational approach to CSF protein biomarkers of neurodegenerative diseases, it is crucial to extensively characterize the profiles of normal human CSF proteins. In this study, to idetify as many CSF proteins in well-characterized healthy young subjects as possible, SDS-PAGE gel electrophoresis was used to prefractionate the CSF proteins before further separation by multidimensional liquid chromatography and analyzed with LCQ or LTQ-FT mass spectrometry (MS). While a total of 466 proteins were identified by LCQ-MS/MS, a total of 608 proteins were identified by LTQ-FT MS/MS, which is 30% over those identified by LCQ-MS/MS. Our results demonstrated that with improved sample preparation and better instrumentation, a much deeper analysis of the human CSF proteome was achieved. Furthermore, we searched our previous MS data obtained from the aging study with the identical database used for the current LCQ and LTQ analysis; it is surprising that a change in database had a very significant effect on proteomic identification of proteins with only 66% overall overlap between two search results. When all proteins were combined, a total of 915 proteins were identified in the CSF of these young healthy subjects. In the end, issues related to sample preparation, proteomic instrumentation, and database search are discussed further in the context of characterization of human CSF proteome.
Screening of pesticides in serum, urine and cerebrospinal fluid collected from an urban population in China

Abstract:Human exposure to pesticides is a topic of public health concern for decades. Pesticide exposures have been assessed through the analysis of urine or blood matrices, but little is known on the accumulation of these chemicals in cerebrospinal fluid (CSF). CSF plays an important role in maintaining physical and chemical balance of the brain and central nervous system and any perturbation can have adverse effects on health. In this study, we investigated the occurrence of 222 pesticides in CSF from 91 individuals using gas chromatography-tandem mass spectrometry (GC-MS/MS). Measured pesticide concentrations in CSF were compared with those in 100 serum and urine specimens from individuals living in the same urban location. Twenty pesticides were found in CSF, serum and urine, at levels above the limit of detection. Three most frequently detected pesticides in CSF were biphenyl (100%), diphenylamine (75%), and hexachlorobenzene (63%). Median concentrations of biphenyl in CSF, serum and urine were 1.11, 10.6, and 1.10 ng/mL, respectively. Six triazole fungicides were found only in CSF, but not in other matrices. To our knowledge, this is the first study to report pesticide concentrations in CSF in a general urban population.

Abstract:OBJECTIVE:To detect common environmental pollutants in human body.METHODS:Urine samples were collected from 80 healthy subjects. Chromatography mass spectrometry (GC-MS), HPLC and ELISA were applied to detect several common environmental pollutants in urine samples.RESULTS:DBP and methylbenzene were present in 75.3% and 41.2% of urine samples. The methanal and AFM1 were found in most of urine samples (approximately 91≊97%). By contrast, PCBs, CPZ, 4, 5-DCC were found in less than 5 samples, but there was no TMT detected.CONCLUSION:Some of the environmental pollutants including carcinogens are detected in urine samples in this study.

Abstract:Exposure to environmental pollutants or contaminants is correlated with detrimental effects on human health, such as neurodegenerative diseases. Adopting hair as a biological matrix for biomonitoring is a significant innovation, since it can reflect the long-term chemical exposome, spanning months to years. However, only a limited number of studies have developed analytical strategies for profiling the chemical exposome in this heterogeneous biological matrix. In this study, a systematic investigation of the chemical extraction procedure from human hair was conducted, using a design of experiments and a high-resolution mass spectrometry (HRMS)-based suspect screening approach. The PlackettBurman (PB) design was applied to identify the significant variables influencing the number of detected features. Then, a central composite design was implemented to optimize the levels of each identified significant variable. Under the optimal conditions-15-minute pulverization, 25 mg of hair weight, 40 min of sonication, and a sonication temperature of 35 °C-approximately 32,000 and 15,000 aligned features were detected in positive and negative ion modes, respectively. This optimized analytical procedure was applied to hair samples from patients with Alzheimer's disease (AD) and individuals with normal cognitive function. Overall, 307 chemicals were identified using the suspect screening approach, with 37 chemicals differentiating patients with AD from controls. This study not only optimized an analytical procedure for characterizing the long-term chemical exposome in human hair but also explored the associations between AD and environmental factors.

Abstract:Data remain scarce regarding the occurrence of per- and polyfluoroalkyl substances (PFASs) in the human brain for better understanding the cerebral disorders. In this study, we measured the concentrations and profiles of 26 traditional and emerging PFASs in cerebrospinal fluid (CSF), which is a preferred matrix to monitor pollutants in the human brain. Our results indicated perfluorooctanesulfonate (PFOS), perfluorooctanoic acid (PFOA), perfluorohexanesulfonate (PFHxS) and n-methylperfluorooctanesulfonamidoacetic acid were the most frequently detected congeners (detection frequency >90%). As the predominant congeners, PFOA and PFOS contributed 27.7% and 14.5% of the total amount of PFASs (ΣPFASs), with respective mean concentration of 221 and 115 pg mL-1. In addition, the concentrations of ΣPFASs in CSF of males were generally higher than those of females, which may be related to the different half-lives of PFASs in different sexes. Interestingly, the concentrations of ΣPFASs and several individual congeners (e.g., perfluorohexanoic acid, perfluorodecanoic acid, perfluorononanoic acid, PFHxS and PFOS) increased with age. The highest concentration of ΣPFASs was found in the elderly compared with other age groups, which may be due to the decreased CSF output as age increased. Our data are valuable for further studies regarding the toxic effects of PFASs on the human brain.

Abstract:Despite being protected by the blood–brain barrier and the blood–cerebrospinal fluid barrier, cerebrospinal fluids can contain contaminants that trigger neurotoxic effects, yet there are few effective methods to analyze metabolites of industrial additives. Here, we established a fast and robust analytical methodology, based on ultra-high performance liquid chromatography interfaced with triple quadrupole mass spectrometry, to quantitatively determine concentrations of metabolites of synthetic antioxidants, organophosphate esters, and phthalate esters in human cerebrospinal fluids. The developed method exhibited highly sensitive and stable instrumental performance, satisfactory recoveries, minimal matrix effects, and trace level detection limits. We identified and quantified thirty industrial additive metabolites in cerebrospinal fluid samples, at 0.58–13 ng/mL total concentrations.

Screening Novel Per- and Polyfluoroalkyl Substances in Human Blood Based on Nontarget Analysis and Underestimated Potential Health Risks

Abstract:Nontarget analysis has gained prominence in screening novel perfluoroalkyl and polyfluoroalkyl substances (PFASs) in the environment, yet remaining limited in human biological matrices. In this study, 155 whole blood samples were collected from the general population in Shijiazhuang City, China. By nontarget analysis, 31 legacy and novel PFASs were assigned with the confidence level of 3 or above. For the first time, 11 PFASs were identified in human blood, including C1 and C3 perfluoroalkyl sulfonic acids (PFSAs), C4 ether PFSA, C8 ether perfluoroalkyl carboxylic acid (ether PFCA), C4-5 unsaturated perfluoroalkyl alcohols, C9-10 carboxylic acid-perfluoroalkyl sulfonamides (CA-PFSMs), and C1 perfluoroalkyl sulfonamide. It is surprising that the targeted PFASs were the highest in the suburban population which was impacted by industrial emission, while the novel PFASs identified by nontarget analysis, such as C1 PFSA and C9-11 CA-PFSMs, were the highest in the rural population who often drank contaminated groundwater. Combining the toxicity prediction results of the bioaccumulation potential, lethality to rats, and binding affinity to target proteins, C3 PFSA, C4 and C7 ether PFSAs, and C9-11 CA-PFSMs exhibit great health risks. These findings emphasize the necessity of broadening nontarget analysis in assessing the PFAS exposure risks, particularly in rural populations.
Scannotation: A Suspect Screening Tool for the Rapid Pre-Annotation of the Human LC-HRMS-Based Chemical Exposome

Abstract:In an increasingly chemically polluted environment, rapidly characterizing the human chemical exposome (i.e., chemical mixtures accumulating in humans) at the population scale is critical to understand its impact on health. High-resolution mass spectrometry (HRMS) profiling of complex biological matrices can theoretically provide a comprehensive picture of chemical exposures. However, annotating the detected chemical features, particularly low-abundant ones, remains a significant obstacle to implementing such approaches at a large scale. We present Scannotation (https://github.com/scannotation/Scannotation_software), an automated and user-friendly suspect screening tool for the rapid pre-annotation of HRMS preprocessed data sets. This software tool combines several MS1 chemical predictors, i.e., m/z, experimental and predicted retention times, isotopic patterns, and neutral loss patterns, to score the proximity between features and suspects, thus efficiently prioritizing tentative annotations to verify. Scannotation and MS-DIAL4 were used to annotate blood serum samples of 75 Breton adolescents. Scannotation's combination of MS1-based chemical predictors allowed us to annotate 89 chemically diverse environmental compounds with high confidence (confirmed by MS2 when available). These compounds included 62% of emerging molecules, for which no toxicological or human biomonitoring data are reported in the literature. The complementarity observed with MS-DIAL4 results demonstrates the relevance of Scannotation for the efficient pre-annotation of large-scale exposomics data sets.
PFASs in Cerebrospinal Fluids and Blood-CSF Barrier Permeability in Patients with Cognitive Impairment

Abstract:Attention has been drawn to the associations between PFASs and human cognitive decline. However, knowledge on the occurrence and permeability of PFASs in the brains of patients with cognitive impairment has not been reported. Here, we determined 30 PFASs in paired sera and cerebrospinal fluids (CSFs) from patients with cognitive impairment (n = 41) and controls without cognitive decline (n = 18). We revealed similar serum PFAS levels but different CSF PFAS levels, with lower CSF PFOA (median: 0.125 vs 0.303 ng/mL, p < 0.05), yet higher CSF PFOS (0.100 vs 0.052 ng/mL, p < 0.05) in patients than in controls. Blood-brain transfer rates also showed lower R-CSF/Serum values for PFOA and higher R-CSF/Serum values for PFOS in patients, implying potential heterogeneous associations with cognitive function. The R (CSF/Serum) values for C4-C14 perfluoroalkyl carboxylates exhibited a U-shape trend with increasing chain length. Logistic regression analyses demonstrated that CSF PFOS levels were linked to the heightened risk of cognitive impairment [odds ratio: 3.22 (1.18-11.8)] but not for serum PFOS. Toxicity inference results based on the Comparative Toxicogenomics Database suggested that PFOS in CSF may have a greater potential to impair human cognition than other PFASs. Our results contribute to a better understanding of brain PFAS exposure and its potential impact on cognitive function. [GRAPHICS]

Abstract:A method for determination of concentrations of polychlorinated biphenyl congeners (PCB-28, 52, 101, 118, 138, 153, 156, and 187) and organochlorine pesticides (hexachlorobenzene, alpha-hexachlorocyclohexane, beta-hexachlorocyclohexane, gamma-hexachlorocyclohexane, delta-hexachlorocyclohexane, p,p'-dichlorodiphenyl dichloroethylene, o,p'-dichlorodiphenyl trichloroethane, p,p'-dichlorodiphenyl dichloroethane, p,p'-dichlorodiphenyl trichloroethane, alpha-chlordane, gamma-chlordane, heptachlor, heptachlor epoxide, and aldrin) in human serum is developed. Recovery is assessed with artificial serum, in which PCBs and OCPs could not be detected. The method is then confirmed with pooled human serum. Experiments are performed by adding two concentrations of analytes (0.5 µg/L and 1.0 µg/L) to both matrices. The sample pretreatment process involves denaturing with a mixture of water-1-propanol (v:v, 85:15), extraction with a C-18 cartridge, and cleanup with an Alumina B cartridge. This process required about 2 mL of serum. The limit of detection ranged from 0.05-0.35 µg/L for all the analytes. Recovery of analytes at low and high spiking concentrations varied from 63-122% and 61-124% for artificial serum and pooled human serum, respectively. Relative standard deviation was lower than 16% and 18% for artificial serum and pooled human serum, respectively. Stability of the method, expressed as relative standard deviation, was lower than 14%. The method has been applied in epidemiological research.
Penetration of PFASs Across the Blood Cerebrospinal Fluid Barrier and Its Determinants in Humans

Abstract:Laboratory studies indicate that exposure to perfluoroalkyl and polyfluoroalkyl substances (PFASs) can induce neuro-behavioral effects in animals. However, the penetration of PFASs across the brain barrier and its determining factors are yet to be clarified in humans. We studied PFAS levels in 223 matched-pair serum and cerebrospinal fluid (CSF) samples from hospital in-patients using UPLC/MS/MS. Among the 21 target analytes, PFOA, PFOS, and 6:2 Cl-PFESA were dominant in serum, with mean concentrations of 7.4, 6.8, and 6.2 ng/mL, respectively, contributing 79% to the total PFAS burden in serum. In CSF, PFOA, PFOS, and 6:2 Cl-PFESA were again the dominant PFASs, with mean concentrations of 0.078, 0.028, and 0.051 ng/mL contributing 36%, 13%, and 24%, respectively, to the total PFAS burden in CSF. Furthermore, PFAS penetration (R-PFAS, PFAS(CSF)/PFAS(serum)) was positively correlated with the barrier permeability index R-Alb (Albumin(CSF)/Albumin(serum)), indicating that barrier integrity was the main determinant of PFAS penetration across the blood-CSF barrier. Positive associations between the R-PFAS values of the main PFASs and serum C-reactive protein were observed, implying that inflammation facilitates the penetration of PFASs across the brain barrier.

Abstract:Individuals are exposed to a wide variety of chemicals over their lifetime, yet current understanding of mixture toxicology is still limited. We present a two-step analytical method using a gas chromatograph-triple quadrupole mass spectrometer that requires less than 1 mL of sample. The method is applied to 183 plasma samples from a study population of children with autism spectrum disorder, their parents, and unrelated neurotypical children. We selected 156 environmental chemical compounds and ruled out chemicals with detection rates less than 20% of our study cohort (n = 61), as well as ones not amenable to the selected extraction and analytical methods (n = 34). The targeted method then focused on remaining chemicals (n = 61) plus 8 additional polychlorinated biphenyls (PCBs). Persistent pollutants, such as p,p′-dichlorodiphenyldichloroethylene (p,p′-DDE) and PCB congeners 118 and 180, were detected at high frequencies and several previously unreported chemicals, including 2,4,6-trichlorophenol, isosafrole, and hexachlorobutadiene, were frequently detected in our study cohort. This work highlights the benefits of employing a multi-step analytical method in exposure studies and demonstrates the efficacy of such methods for reporting novel information on previously unstudied pollutant exposures.

Abstract:Neurodegenerative diseases pose increasing challenges to global aging populations. Cognitive decline in older adults is an initial indicator of neurodegenerative diseases, yet comprehensive research on environmental chemical exposures related to cognitive decline is limited. This study uses Environment-Wide Association Study (EWAS) framework to investigate associations of environmental chemicals with cognitive function in individuals aged ≥60 years. We used the Digit Symbol Substitution Test (DSST) scores (lower scores indicate cognitive decline) and chemical biomarker data of the U.S. National Health and Nutrition Examination Survey (NHANES) spanning four cycles (1999-2000, 2001-2002, 2011-2012, 2013-2014). We conducted multiple survey-weighted regression to identify biomarkers associated with DSST scores, penalized logit regression to estimate odds ratio (OR) of cognitive decline with identified biomarkers, and correlation network analyses to examine relationships among identified biomarkers and cognitive decline. After correction for multiple comparisons, 27 out of 173 biomarkers having a ≥10% detection rate were associated with DSST scores ( -value <0.05). Among them, increased odds of cognitive decline were associated with elevated levels of blood lead (Pb) (OR = 1.12, 95% CI: 1.01,1.42), blood 1,4-dichlorobenzene (1,4-DCB) (OR = 1.34, 95% CI: 1.17, 1.54), and urinary 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol (NNAL) (OR = 1.34, 95% CI: 1.10, 1.62). Correlation network showed biomarkers that potentially impact cognitive decline upon related health conditions, such as stroke. In conclusion, leveraging the EWAS framework enables us to identify chemical biomarkers that were not previously discovered from traditional approaches of examining a small number of chemicals at a time. While our findings provide foundation for further research, longitudinal studies are warranted to elucidate causal relationships.

Abstract:Elevated concentrations of polychlorinated biphenyls (PCBs) found in environmental media and biota from typical e-waste dismantling sites have raised concerns regarding their human body burden and potential negative health effects. In the present study, the enantiomeric compositions of three typical chiral congeners (PCB-95, PCB-132, and PCB-149) were measured in 24 serum samples from e-waste workers by using gas chromatography coupled to triple quadrupole tandem mass spectrometry. The mean enantiomer fractions (EFs) of chiral congeners in serum from the workers were 0.655 ± 0.103, 0.679 ± 0.164, and 0.548 ± 0.095 for PCB-95, PCB-132, and PCB-149, respectively. The (+) enantiomers of PCB-95, PCB-132, and PCB-149 were enantioselectively enriched in serum. Significant positive correlations were observed between the EF of the chiral congener PCB-95 and the total concentration of OH-PCBs, suggesting that EF values of chiral PCBs could be used to indicate the extent of biological metabolism. In addition, the EF of PCB-95 in serum samples increased with increasing work duration of the e-waste workers, thus demonstrating the usefulness of EF values of chiral PCBs as tracers of human exposure to PCBs. Because of the enantioselective enrichment of (+) enantiomers of PCB-95, PCB-132, and PCB-149, further studies are needed to explore the metabolism and toxicity of chiral contaminants in humans.

Non-Target and Suspect Screening of Per- and Polyfluoroalkyl Substances in Airborne Particulate Matter in China.

Abstract:Airborne particulate matter (APM) has an important role in inhalation exposure, especially in China. The environmental occurrence of conventional and unknown per- and polyfluoroalkyl substances (PFASs) in APM remains unclear. Therefore, in this study, a two-stage experiment was designed to identify potential PFASs and to investigate their distribution in APM. Indoor and outdoor APM samples were collected from five selected cities in China. Through PFAS homologue analysis and suspect screening, 50 peaks were identified with different confidence levels (levels 1-3). Among the identified PFASs, 34 emerging PFASs including p-perfluorous nonenoxybenzenesulfonate, 6:2 polyfluoroalkyl phosphate diester, n:2 fluorotelomer sulfonates, n:2 fluorinated telomer acids, n:2 chlorinated polyfluoroalkyl ether sulfonic acids, 1:n polyfluoroalkyl ether carboxylic acids (1:n PFECAs), perfluoroalkyl dioic acids (PFdiOAs), hydro-substituted perfluoroalkyl dioic acids (H-PFdiOAs), and unsaturated perfluorinated alcohols (UPFAs) were identified in APM. In particular, 1:n PFECAs, PFdiOAs, H-PFdiOAs, and UPFAs were first detected in APM. Although human exposure to perfluorooctanoic acid via inhaled APM was noted to not be a risk (hazard quotient <0.1) in this study, the expansion of the PFASs screened in APM implies that human exposure to PFASs might be much more serious and should be considered in future risk assessments in China.
Comparing Liquid-Liquid, Solid-Phase, and Supported-Liquid Extraction for the Determination of Polycyclic Aromatic Hydrocarbons in Serum Samples and Their Application for Human Biomonitoring

Abstract:Polycyclic aromatic hydrocarbons (PAHs) are a class of persistent organic pollutants of great health concern. Epidemiological and animal studies have revealed the carcinogenic and mutagenic risk of PAHs, and therefore avoiding PAH exposure is a high priority. In this work, a comparison of liquid-liquid, solid-phase, and supported -liquid extraction was conducted for detecting PAHs in serum samples. The key features of mechanism, simplicity, recoveries, and background interference were compared for the three procedures. Solid-phase and supported -liquid extraction showed chromatography background interference and required method optimization to obtain satisfactory recoveries that were comparable to that of liquid-liquid extraction. Liquid-liquid extraction with hexane and high-performance liquid chromatography coupled with a fluorescence detection was employed for detecting PAHs in serum samples. The limits of quantification of the individual PAHs ranged from 16.0 ng/L to 40.0 ng/L, and recoveries were obtained from 69.6 % to 87.7 %. The method was used for human bio-monitoring of the target PAHs in the serum of 120 volunteers from Guangzhou. The mean total PAH concen-tration in the serum of adults (2.60 mu g/L) was significantly higher than that of teenagers (1.69 mu g/L). The total estimated daily intake of PAHs was calculated for naphthalene, fluorene, and phenanthrene, and a low health risk was obtained by the evaluation of their non-carcinogenic and carcinogenic risks.

Abstract:Background: Polycyclic aromatic hydrocarbons (PAHs), as organic pollutants widely present in daily environments, have been shown by existing epidemiological studies to be significantly associated with deficits in learning and memory functions in children and adults. However, the association between exposure to PAHs and cognitive function in older adults remains unclear. Additionally, existing related studies have only assessed the association between individual PAH exposures and cognitive assessments, overlooking the risks posed by mixed exposures. This study aims to use three statistical models to investigate the individual and overall effects of mixed PAH exposures on the cognition of older adults in the United States. Methods: The study cohort was obtained from the NHANES database, which included individuals aged 60 and older from 2011 to 2014. Weighted generalized linear models (GLM), weighted quantile sum (WQS) models, and Bayesian kernel machine regression (BKMR) models were utilized to evaluate the connections between urinary PAH metabolites and the standardized Z-scores of four cognitive tests: Immediate Recall Test (IRT), Delayed Recall Test (DRT), Animal Fluency Test (AFT), and Digit Symbol Substitution Test (DSST). Results: Our analysis involved 899 individuals aged 60 and above. In the fully adjusted GLM, 2-hydroxynaphthalene (2-OHNa), 3-hydroxyfluorene (3-OHFlu), and 2-hydroxyfluorene (2-OHFlu) demonstrated negative associations with DSST Z-scores. In the WQS model, six urinary PAH metabolites were negatively linked to AFT Z-scores (β (95% confidence intervals [CI]): -0.120 (-0.208, -0.033), p = 0.007) and DSST Z-scores (β (95% CI): -0.182 (-0.262, -0.103), p < 0.001). In both assessments, 2-OHNa exerted the greatest influence among the urinary PAH metabolites. In the BKMR model, there was an overall negative correlation between urinary PAH metabolites and AFT and DSST Z-scores when the concentration was within the 25th to 75th percentile, where 2-OHNa dominated the main effect of the mixture. The WQS and BKMR models were adjusted for all covariates. Conclusion: Increased concentrations of urinary PAH metabolites are associated with cognitive decline in older adults, mainly on language ability, executive function, sustained attention, working memory, and information processing speed, with 2-OHNa playing a major effect.
