Fibroblast activation is the main pathological basis of prostatic hyperplasia and an important factor in promoting the progression of BPH. Recent studies have shown that autophagy participates in BPH progression and inhibits lipid accumulation. Our previous study found that stromal cells of early-progressed BPH are activated and there exist a large number of autophagic bubbles. We investigated whether autophagy promotes loss of lipids in fibroblast cells to provide energy for their activation in BPH progression. METHODS: Immunohistochemistry, immunofluorescence, oleic acid staining experiments were performed to determine the number and distribution of autophagic vacuoles and lipid droplets in early-progressed BPH stromal. Autophagy model induced by fast/Rapamycin and blocked with CQ or siATG7 in fibrogenic cells; immunofluorescence, qPCR, Western Blot, oleic acid staining experiment, ATP measurement to explore the activation and lipid metabolism of prostate fibroblasts under different autophagy conditions RESULTS: The number of autophagic vacuoles increased but lipid droplets and triglycerides decreased in the early-progressed BPH stromal; when fast or Rapamycin induced autophagy, the expression of COL-1 and a-SMA increased while the formation of lipid droplets and decreased ATP levels in human prostate primary interstitial fibroblasts (PrSC) and prostate fibroblasts cell line (WPMY-1); Using fatty acid oxime oxidation inhibitor (etomoxir) could attenuate the effects by autophagy.It gived rise to opposite role that the expression of COL-1 and a-SMA decreased while the formation of lipid droplets increased prostate fibroblasts cells, when inhibition of autophagy by using small interfering RNAs against Atg7 and chemical antagonists(CQ), and these effects were partially overcome by providing oleic acid(OA) as an energy substrate. CONCLUSIONS: Autophagy may promote fibroblast activation by regulating lipid metabolism to accelerate the progression of BPH, providing a theoretical basis for clinical control of BPH progression.
