Abstract:Memory is a way for people to acquire information through personal experiences and from media such as books, TV, and the internet. Relying on memory and the ability to gain new ones, the human brain is capable of adapting to survive in the ever-changing environment and can create marvels and build objects out of nature. Therefore, it is memory that creates modern civilization. Memory consists of synapses and neural ensembles in the brain which integrate multisensory information both spatially and temporally. Thus, memory bridges the link between cognitive domain information and physical domain neural connections, making them easier to track and study. Memory is the foundation of the brain's cognitive functions. Consequently, research into how the brain stores and retrieves memories reveals not only how the brain works to generate intelligence, but also suggests possible avenues for creating efficient machine learning algorithms that have evolved over millions of years. Recent breakthroughs in biotechnology have led to discoveries about the mechanisms underlying memory formation and retrieval, providing insight into neural memories at the cellular and synaptic levels as well as understanding neural coding for circuitry integration and operation. Particularly, the discovery of cellular memory engrams in the neocortex has revealed a highly parallel arrangement of memory circuits, which are located in layer ll of widely distributed cortical areas, including sensory cortices, primary visual cortices, and retrosplenial cortices. In this review, we summarize the most recent findings on memory engrams in the hippocampus and neocortex. We highlight the correlation between neural oscillations and cortical engram activities during memory formation. We also explore the evidence that long-range synchrony of theta-coupled gamma oscillations plays a key role in modulating cortical engram activity to create a brain-wide memory network. We explain how memory engrams in the neocortex are integrated and organized by indexing signals from the hippocampus, allowing memories to be registered allocated and retrieved in the brain. Lastly, we shed light on the molecular mechanisms especially epigenetic regulators involved in the physical regulation of memory persistency and flexibility. Deficiencies in the epigenetic regulation of these engrams may lead to memory deficits in various brain diseases. Additionally, the laminar-specific distribution of memory engrams in local cortical columns and the global organization of memory traces in distinct functional cortical regions may inspire the development of novel computing algorithms in machine learning to achieve brain-level intelligence. Taken together, the advances in methodology are paving the way for us to directly investigate the neural mechanisms of memory storage and retrieval in mammalian brain, giving insight into human cognition and inspiring new approaches in machine learning.

How the Brain Formulates Memory: A Spatio-Temporal Model Research Frontier.

Abstract:Memory is a complex process across different brain regions and a fundamental function for many cognitive behaviors. Emerging experimental results suggest that memories are represented by populations of neurons and organized in a categorical and hierarchical manner. However, it is still not clear how the neural mechanisms are emulated in computational models. In this paper, we present a spatio-temporal memory (STM) model using spiking neurons to explore the memory formulation and organization in the brain. Unlike previous approaches, this model employs temporal population codes as the neural representation of information and spike-timing-based learning methods to formulate the memory structure. It explicitly demonstrates that the complex spatio-temporal patterns are the internal neural representations of memory items. Two types of memory processes are analyzed and emulated: associative memory, i.e., spatio-temporal patterns driven by intra-assembly connections, and episodic memory, i.e., temporally separated spatio-temporal patterns linked by inter-assembly connections. Our model will provide a computational substrate based on low-level neural circuits for developing neuromorphic cognitive systems with wide applications.

Abstract:To comprehend the brain’s mental function and dysfunction, we focus on memory, its formation and loss. Our expanded tripartite mechanism of memory involves the following compartments: Stimulated brain cells (neurons + glial cells) which secrete dopants (>10 metals and >80 neuro-transmitters (NTs) and glio-transmitters (GTs)) into the extracellular matrix (nECM/PNN) enveloping all brain cells, that performs as a “memory material”. The metal-centered complexes formed within the nECM/PNN are molecular correlates of cognitive units of information (cuinfo) that encode experience. The transmitters (NTs+GTs) embody emotive quality that elicit physiologic reactions from organs and glands to encode emotive states. As for decoding the cuinfo, aggregates (mosaics) of sensors (GPCR- and K-channel family proteins) within the lipid-bilayer comprising the cell membranes are capable of “reading” and integrating the cuinfo, transliterating them into an experiential mental state. As to memory loss, aside from gross injuries or anatomical abnormalities, we look to molecular-level decay, which involve processes such as Toxicity or deficiency of trace metals, degradation of the nECM/PNN or it’s by enzymatic or redox (Fenton) reactions, uncontrolled lipid peroxidation (i.e. endocannabinoids) or autoimmune antibodies (i.e. anti- metallothionein) or lysozyme amyloid fibril formation. Interference with the optimal operation of the expanded tripartite mechanism of emotive memory is reflected by clinical features of memory loss in patients with degenerative disorders.

Abstract:The ability to store information about the past to dynamically predict and prepare for the future is among the most fundamental tasks the brain performs. To date, the problems of understanding how the brain stores and organizes information about the past (memory) and how the brain represents and processes temporal information for adaptive behavior have generally been studied as distinct cognitive functions. This Symposium explores the inherent link between memory and temporal cognition, as well as the potential shared neural mechanisms between them. We suggest that working memory and implicit timing are interconnected and may share overlapping neural mechanisms. Additionally, we explore how temporal structure is encoded in associative and episodic memory and, conversely, the influences of episodic memory on subsequent temporal anticipation and the perception of time. We suggest that neural sequences provide a general computational motif that contributes to timing and working memory, as well as the spatiotemporal coding and recall of episodes.

Abstract:We review recent work on three major lines of memory research: a) the possible role of the protein kinase M-zeta (PKMzeta) in memory persistence; b) the processes of "synaptic tagging and capture" in memory formation; c) the modulation of extinction learning, widely used in the psychotherapy of fear memories under the name of "exposure therapy". PKMzeta is a form of protein kinase C (PKC) that apparently remains stimulated for months after the consolidation of a given memory. Synaptic tagging is a mechanism whereby the weak activation of one synapse can tag it with a protein so other synapses in the same cell can reactivate it by producing other proteins that bind to the tag. Extinction, once mistakenly labeled as a form of forgetting, is by itself a form of learning; through it animals can learn to inhibit a response. We now know it can be modulated by neurotransmitters or by synaptic tagging, which should enable better control of its clinical use.

Abstract:Acquisition of language semantics is widely believed to be facilitated by biological mechanisms of associative learning [1]. However, current understanding of brain mechanisms underlying learning and memory is primarily based on simple models, including animal models, with mechanisms of learning in the cognitive realm largely unknown. Semantic language learning serves as a compelling example of these cognitive processes, whose brain mechanisms pose a challenge to explanation at the level of basic neural networks. Particularly, many processes underlying such learning still lack clear understanding, including the mechanisms that enable correlational coincidence of neuronal representation required for Hebbian plasticity, the reverberatory replay of the to-be associated representations in working memory, and the involvement of consolidation, among others. We conducted a series of studies modeling the acquisition of semantic knowledge of action-related vocabulary to investigate these questions. We implemented a unique trial-and-error-based procedure for semantic acquisition, in which participants obtained associations between newly introduced pseudowords and actions through active learning. The experimental procedure comprised the delivery of acoustically equivalent pseudowords that were previously unknown to the subjects. Participants were instructed to execute movements of their hands and feet in response to specific pseudowords. Following each trial, participants received either positive or negative feedback depending on their performance. During the learning phase, MEG was recorded, as well as during passive presentations of the same pseudowords before and after learning to control for attention and motor preparation effects. Our analysis involved event-related fields and beta oscillations. Our experiments showed that cortical plasticity related to word learning could be detected immediately after learning, without the need for prolonged consolidation time [2]. We observed two effects during learning that could contribute to Hebbian plasticity by linking auditory speech representations of pseudowords with associated movements. First, we observed the reactivation of auditory speech representations during the initialization of movement. This mechanism apparently creates the conditions for temporal coincidence of activations in two representations, specifically an auditory speech representation and a representation of a motor program. During the learning process, we observed a considerable rise in beta oscillations that occurred during and after action execution. This effect comprised a typical beta-rebound in the sensorimotor regions and a significant surge in beta activity in various associative cortical regions. We postulate that these beta oscillations constitute a reverberation mechanism that reinforces the associative connection and guards it against potential interference [3]. In further experiments, we administered the learning process over two consecutive days with an overnight interval between recordings. We evaluated beta power dynamics during the acquisition of novel pseudowords on day 1 and during task rehearsal following overnight sleep on day 2. Beta event-related synchronization in the frontal regions emerged upon the attainment of the pseudoword learning rules on day 1, and remained unchanged after sleep and subsequent rehearsal on day 2. The prefrontal beta rhythm’s dynamics mirrored the corresponding behavioral changes: subjects executed the task with minimal errors on day 2 upon meeting the learning criterion on day 1. Beta event-related synchronization augmented continuously in the posterior temporal and parietal cortices on day 2 and potentially contributed towards the assimilation of newly-acquired associations in long-term memory. In summary, our research offers fresh perspectives on the intricate neural mechanisms underlying plasticity in the human brain within the cognitive domain.

Abstract:Magnetoencephalography (MEG) was used to compare memory processes in two experiments, one involving recognition of word pairs and the other involving recall of newly learned arithmetic facts. A combination of hidden semi-Markov models and multivariate pattern analysis was used to locate brief “bumps” in the sensor data that marked the onset of different stages of cognitive processing. These bumps identified a separation between a retrieval stage that identified relevant information in memory and a decision stage that determined what response was implied by that information. The encoding, retrieval, decision, and response stages displayed striking similarities across the two experiments in their duration and brain activation patterns. Retrieval and decision processes involve distinct brain activation patterns. We conclude that memory processes for two different tasks, associative recognition versus arithmetic retrieval, follow a common spatiotemporal neural pattern and that both tasks have distinct retrieval and decision stages.

Abstract:Abstract Memory is a complex process that spans multiple time-scales and stages, and, as expected, involves multiple brain regions. Traditionally, computational models of memory are either too abstract (Shiffrin & Steyvers, 1997) to be meaningfully connected to a biological substrate, or, when explicitly connected, are narrowly focused on one specific region and process (Blum & Abbott, 1996; Weber et al., 2017). By contrast, a comprehensive model of memory with a plausible neural interpretation would be extremely valuable to drive further research in memory function and dysfunction. In this paper, we attempt to fill in this gap by providing a detailed biological analysis of ACT-R’s declarative memory system. This system, developed over four decades, has evolved into a consistent framework that describes how memories are formed, retrieved, forgotten, mistaken, and merged. Building on existing mappings between some components and their biological counterpart, as well as the existing literature, this paper provides a comprehensive view of how the framework’s various computations map onto different brain regions, their network dynamics and functional connectivity, and biological structure. We also show that these mappings provide further insights and explanations for puzzling findings in the memory disorders literature. Finally, we outline the remaining gaps (such as the transition from episodic to semantic memory) and how they could be addressed by future research and modeling efforts.

Abstract:Fear extinction is a phenomenon that involves a gradual reduction in conditioned fear responses through repeated exposure to fear-inducing cues. Functional brain connectivity assessments, such as functional magnetic resonance imaging (fMRI), provide valuable insights into how brain regions communicate during these processes. Stress, a ubiquitous aspect of life, influences fear learning and extinction by changing the activity of the amygdala, prefrontal cortex, and hippocampus, leading to enhanced fear responses and/or impaired extinction. Glucocorticoid receptors (GRs) are key to the stress response and show a dual function in fear regulation: while they enhance the consolidation of fear memories, they also facilitate extinction. Accordingly, GR dysregulation is associated with anxiety and mood disorders. Recent advancements in cognitive neuroscience underscore the need for a comprehensive understanding that integrates perspectives from the molecular, cellular, and systems levels. In particular, neuropharmacology provides valuable insights into neurotransmitter and receptor systems, aiding the investigation of mechanisms underlying fear regulation and potential therapeutic targets. A notable player in this context is cortisol, a key stress hormone, which significantly influences both fear memory reconsolidation and extinction processes. Gaining a thorough understanding of these intricate interactions has implications in terms of addressing psychiatric disorders related to stress. This review sheds light on the complex interactions between cognitive processes, emotions, and their neural bases. In this endeavor, our aim is to reshape the comprehension of fear, stress, and their implications for emotional well-being, ultimately aiding in the development of therapeutic interventions.

Abstract:Many mental disorders and neurodegenerative and neurodevelopmental diseases involve cognitive deficits. Remarkable advances and new technologies are providing a clearer picture of the molecular basis of cognition. In conjunction with an SFN2010 symposium, we provided here a brief overview of the molecular mechanisms of cognition, with emphasis on the development of treatments for cognitive disorders. Activity-dependent changes in gene expression and protein synthesis integrate with synapse selection to form memory circuits. A neuronal activity-dependent molecular tagging system that uses the gene expression program to record memory circuit formation represents one new tool to study cognition. Regulation of protein translation, protein degradation, cytoskeletal dynamics, extracellular matrix interactions, second messenger signaling, and neurotransmitter receptor trafficking and function are all components of synaptic remodeling essential for cognition. Selective targeting of specific effectors in these processes, such as NMDA receptors, may serve as an effective strategy to treat cognitive deficits.

Abstract:The acquisition of fear associative memory requires brain processes of coordinated neural activity within the amygdala, prefrontal cortex (PFC), hippocampus, thalamus and brainstem. After fear consolidation, a suppression of fear memory in the absence of danger is crucial to permit adaptive coping behavior. Acquisition and maintenance of fear extinction critically depend on amygdala-PFC projections. The robust correspondence between the brain networks encompassed cortical and subcortical hubs involved into fear processing in humans and in other species underscores the potential utility of comparing the modulation of brain circuitry in humans and animals, as a crucial step to inform the comprehension of fear mechanisms and the development of treatments for fear-related disorders. The present review is aimed at providing a comprehensive description of the literature on recent clinical and experimental researches regarding the noninvasive brain stimulation and optogenetics. These innovative manipulations applied over specific hubs of fear matrix during fear acquisition, consolidation, reconsolidation and extinction allow an accurate characterization of specific brain circuits and their peculiar interaction within the specific fear processing.

Abstract:The successful development of neural prostheses requires an understanding of the neurobiological bases of cognitive processes, i.e., how the collective activity of populations of neurons results in a higher level process not predictable based on knowledge of the individual neurons and/or synapses alone. We have been studying and applying novel methods for representing nonlinear transformations of multiple spike train inputs (multiple time series of pulse train inputs) produced by synaptic and field interactions among multiple subclasses of neurons arrayed in multiple layers of incompletely connected units. We have been applying our methods to study of the hippocampus, a cortical brain structure that has been demonstrated, in humans and in animals, to perform the cognitive function of encoding new long-term (declarative) memories. Without their hippocampi, animals and humans retain a short-term memory (memory lasting approximately 1 min), and long-term memory for information learned prior to loss of hippocampal function. Results of more than 20 years of studies have demonstrated that both individual hippocampal neurons, and populations of hippocampal cells, e.g., the neurons comprising one of the three principal subsystems of the hippocampus, induce strong, higher order, nonlinear transformations of hippocampal inputs into hippocampal outputs. For one synaptic input or for a population of synchronously active synaptic inputs, such a transformation is represented by a sequence of action potential inputs being changed into a different sequence of action potential outputs. In other words, an incoming temporal pattern is transformed into a different, outgoing temporal pattern. For multiple, asynchronous synaptic inputs, such a transformation is represented by a spatiotemporal pattern of action potential inputs being changed into a different spatiotemporal pattern of action potential outputs. Our primary thesis is that the encoding of short-term memories into new, long-term memories represents the collective set of nonlinearities induced by the three or four principal subsystems of the hippocampus, i.e., entorhinal cortex-to-dentate gyrus, dentate gyrus-to-CA3 pyramidal cell region, CA3-to-CA1 pyramidal cell region, and CA1-to-subicular cortex. This hypothesis will be supported by studies using in vivo hippocampal multineuron recordings from animals performing memory tasks that require hippocampal function. The implications for this hypothesis will be discussed in the context of "cognitive prostheses"-neural prostheses for cortical brain regions believed to support cognitive functions, and that often are subject to damage due to stroke, epilepsy, dementia, and closed head trauma.

Abstract:<p>Psychological theories posit that the hippocampus rapidly forms associations among ongoing events as they unfold. During a subsequent maturation process, so‐called systems memory consolidation, these associations are gradually stabilized within distributed neocortical circuits through close interactions between the hippocampus and neocortex. In the past fifty years, a major effort in neurobiological research has been directed towards translating these descriptive accounts into tangible, biological processes in the brain. Until the early 2000s, most studies exclusively focused on examining whether the hippocampus becomes unnecessary for memory retrieval once the memory has been consolidated. With recent methodological advances, however, the field shifted attention to several other theoretical accounts and began to uncover the genetic, physiological, and structural underpinnings of systems memory consolidation at an unprecedented level of precision. Here I review these neurobiological findings in the past fifteen years within a framework of six essential predictions extracted from the psychological theories. Genetic approaches have made it possible to tag neurons that were activated during memory encoding and investigate their physiological and genetic profiles as well as reactivation patterns during subsequent retrieval. In parallel, electrophysiological and imaging approaches detected signs of the gradual refinement of memory representations and its underlying hippocampal‐neocortical dialogue in millisecond‐resolved neural firing patterns, the inter‐region coupling of neural activity, across‐day stability of neural ensemble activity, and functional connectome. This summary represents substantial progress in our understanding of neurobiological mechanisms of systems memory consolidation whilst also identifying several essential remaining questions for future investigations.</p>
