Abstract:NOTCH receptor 3 (NOTCH3) is known to regulate the transcription of oncogenes or tumor suppressor genes, thereby playing a crucial role in tumor development, invasion, maintenance, and chemotherapy resistance. However, the specific mechanism of how NOTCH3 drives immune infiltration in gastrointestinal cancer remains uncertain. The expression of NOTCH3 was analyzed through Western blot, PCR, Oncomine database, and the Tumor Immune Estimation Resource (TIMER) site. Kaplan–Meier plotter, PrognoScan database, and gene expression profile interactive analysis (GEPIA) were used to assess the impact of NOTCH3 on clinical prognosis. The correlation between NOTCH3 expression and immune infiltration gene markers was investigated using TIMER and GEPIA. NOTCH3 was found to be commonly overexpressed in various types of gastrointestinal tumors and was significantly associated with poor prognosis. Furthermore, the expression level of NOTCH3 showed a significant correlation with the tumor purity of gastrointestinal tumors and the extent of immune infiltration by different immune cells. Our findings suggest that NOTCH3 may act as a crucial regulator of tumor immune cell infiltration and can serve as a valuable prognostic biomarker in gastrointestinal cancers.

What problem does this paper attempt to address?
The problem this paper attempts to address is to explore the prognostic value of NOTCH3 in gastrointestinal cancers and its correlation with immune infiltration. Specifically, the researchers aim to evaluate the impact of NOTCH3 expression levels on the clinical prognosis of patients by analyzing its expression in different types of gastrointestinal tumors, and further investigate the relationship between NOTCH3 expression and tumor immune cell infiltration. ### Main Research Questions: 1. **Expression of NOTCH3 in Gastrointestinal Cancers**: The researchers explored the expression levels of NOTCH3 in gastrointestinal cancers through various experimental methods (such as Western blot, PCR, etc.) and database analyses (such as Oncomine, TIMER, etc.). 2. **Correlation between NOTCH3 and Patient Prognosis**: Using databases like Kaplan–Meier plotter and PrognoScan, they assessed the impact of NOTCH3 expression levels on patient overall survival (OS), progression-free survival (PFS), and other prognostic indicators. 3. **Relationship between NOTCH3 and Immune Infiltration**: Through TIMER and GEPIA databases, they analyzed the correlation between NOTCH3 expression and the infiltration levels of different types of immune cells (such as B cells, CD4+ T cells, CD8+ T cells, macrophages, etc.). ### Research Background: - Gastrointestinal cancers are a major global health threat, with high incidence and mortality rates for colorectal cancer, gastric cancer, liver cancer, and pancreatic cancer. - Immunotherapy has achieved significant therapeutic effects in certain types of cancers, but its application in gastrointestinal cancers still requires further research. - The NOTCH signaling pathway plays an important role in cell fate determination, proliferation, and differentiation. As a potential oncogene, NOTCH3 may play a key role in tumorigenesis, invasion, maintenance, and chemotherapy resistance. ### Research Significance: - By revealing the prognostic value of NOTCH3 in gastrointestinal cancers and its relationship with immune infiltration, this study can provide a theoretical basis for developing new immunotherapy targets. - It helps to better understand the immune microenvironment of gastrointestinal cancers, providing a reference for the design of personalized treatment plans. In summary, this paper aims to comprehensively evaluate the biological function of NOTCH3 in gastrointestinal cancers and its potential role in tumor immunology through systematic experiments and data analysis.
