Abstract:Cortical feedback is thought to mediate cognitive processes like attention, prediction, and awareness. Understanding its function requires identifying the organizational logic of feedback axons relaying different signals. We measured retinotopic specificity in inputs from the lateromedial visual area in mouse primary visual cortex (V1) by mapping receptive fields in feedback boutons and relating them to those of neurons in their vicinity. Lateromedial visual area inputs in layer 1 targeted, on average, retinotopically matched locations in V1, but many of them relayed distal visual information. Orientation-selective axons overspread around the retinotopically matched location perpendicularly to their preferred orientation. Direction-selective axons were biased to visual areas shifted from the retinotopically matched position along the angle of their antipreferred direction. Our results show that feedback inputs show tuning-dependent retinotopic specificity. By targeting locations that would be activated by stimuli orthogonal to or opposite to a cell’s own tuning, feedback could potentially enhance visual representations in time and space.

Abstract:A single visual stimulus activates neurons in many different cortical areas. A major challenge in cortical physiology is to understand how the neural activity in these numerous active zones leads to a unified percept of the visual scene. The anatomical basis for these interactions is the dense network of connections that link the visual areas. Within this network, feedforward connections transmit signals from lower-order areas such as V1 or V2 to higher-order areas. In addition, there is a dense web of feedback connections which, despite their anatomical prominence1,2,3,4, remain functionally mysterious5,6,7,8. Here we show, using reversible inactivation of a higher-order area (monkey area V5/MT), that feedback connections serve to amplify and focus activity of neurons in lower-order areas, and that they are important in the differentiation of figure from ground, particularly in the case of stimuli of low visibility. More specifically, we show that feedback connections facilitate responses to objects moving within the classical receptive field; enhance suppression evoked by background stimuli in the surrounding region; and have the strongest effects for stimuli of low salience.

Abstract:Sensory processing in the neocortex requires both feedforward and feedback information flow between cortical areas1. In feedback processing, higher-level representations provide contextual information to lower levels, and facilitate perceptual functions such as contour integration and figure–ground segmentation2,3. However, we have limited understanding of the circuit and cellular mechanisms that mediate feedback influence. Here we use long-range all-optical connectivity mapping in mice to show that feedback influence from the lateromedial higher visual area (LM) to the primary visual cortex (V1) is spatially organized. When the source and target of feedback represent the same area of visual space, feedback is relatively suppressive. By contrast, when the source is offset from the target in visual space, feedback is relatively facilitating. Two-photon calcium imaging data show that this facilitating feedback is nonlinearly integrated in the apical tuft dendrites of V1 pyramidal neurons: retinotopically offset (surround) visual stimuli drive local dendritic calcium signals indicative of regenerative events, and two-photon optogenetic activation of LM neurons projecting to identified feedback-recipient spines in V1 can drive similar branch-specific local calcium signals. Our results show how neocortical feedback connectivity and nonlinear dendritic integration can together form a substrate to support both predictive and cooperative contextual interactions.

Development of Retinotopic Feedback: Layer 6 Pyramids to Lateral Geniculate Principal Cells

Abstract:The development of many feedforward pathways in the brain, from sensory inputs to neocortex, have been studied and modeled extensively, but the development of feedback connections, which tend to occur after the development of feedforward pathways, have received less attention. The abundance of feedback connections within neocortex and between neocortex and thalamus suggests that understanding feedback connections is crucial to understanding connectivity and signal processing in the brain. It is well known that many neural layers are arranged topologically with respect to sensory input, and many neural models impose a symmetry of connections between layers, commonly referred to as reciprocal connectivity. Here, we are concerned with how such reciprocal, feedback connections develop so that the topology of the sensory input is preserved. We focus on feedback connections from layer 6 of visual area V1 to primary neurons in the Lateral Geniculate Nucleus (LGN). The proposed model is based on the hypothesis that feedback connections from V1-L6 to LGN use voltage-activated T-channels to appropriately establish and modify synapses in spite of unavoidable temporal delays. We also hypothesize that developmental spindling relates to synaptogenesis and memory consolidation.

Abstract:Neurons in primary visual cortex are driven by feedforward visual inputs and top-down contextual inputs. The nature of this contextual information is difficult to study, as responses to feedforward and top-down inputs overlap in time and are difficult to disentangle experimentally. To address this issue, we measured responses to natural images and partially occluded versions of these images in the visual cortex of mice. Assessing neuronal responses before and after familiarizing mice with the non-occluded images allowed us to study experience-dependent and stimulus-specific contextual responses in pyramidal cells (PyCs) in cortical layers 2/3 and 5 in the absence of feedforward input. Surprisingly, in the same retinotopic region of cortex, we found that separate populations of PyCs in layer 2/3 responded to occluded and non-occluded images. Responses of PyCs selective for occluded images were strengthened upon familiarization and decoding analysis revealed they contained image-specific information, suggesting that they signaled the absence of predicted visual stimuli. Responses of PyCs selective for non-occluded scenes were weaker for familiarized images but stronger for unfamiliar images, suggesting that these neurons signaled the presence of unpredicted visual stimuli. Layer 5 also contained PyCs preferring either feedforward or contextual inputs, but their responses were more complex and strengthening of responses to occluded images required task engagement. The results show that visual experience decreases the activity of neurons responding to known feedforward inputs but increases the activity of neurons responding to contextual inputs tied to expected stimuli.

Retinotopic biases in contextual feedback signals to V1 for object and scene processing

Abstract:Identifying the objects embedded in natural scenes relies on recurrent processing between lower and higher visual areas. How is cortical feedback information related to objects and scenes organised in lower visual areas? The spatial organisation of cortical feedback converging in early visual cortex during object and scene processing could be retinotopically specific as it is coded in V1, or object centred as coded in higher areas, or both. Here, we characterise object and scene-related feedback information to V1. Participants identified foreground objects or background scenes in images with occluded central and peripheral subsections, allowing us to isolate feedback activity to foveal and peripheral regions of V1. Using fMRI and multivoxel pattern classification, we found that feedback of object information is projected to foveal V1 cortex with increased detail during an object identification task. Background scene information is projected to both foveal and peripheral V1 but can be disrupted by a sufficiently demanding object discrimination task. We suggest that the feedback connections during scene perception project back to earlier visual areas an automatic sketch of occluded information to the predicted retinotopic location. In the case of a cognitive task however, feedback pathways project content to foveal retinotopic space, potentially for introspection, functioning as a cognitive active blackboard and not necessarily predicting the object’s location. This feedback architecture could reflect the internal mapping in V1 of the brain’s endogenous models of the visual environment that are used to predict perceptual inputs.

Abstract:The physiological response properties of neurons in the visual system are inherited mainly from feedforward inputs. Interestingly, feedback inputs often outnumber feedforward inputs. Although they are numerous, feedback connections are weaker, slower, and considered to be modulatory, in contrast to fast, high-efficacy feedforward connections. Accordingly, the functional role of feedback in visual processing has remained a fundamental mystery in vision science. At the core of this mystery are questions about whether feedback circuits regulate spatial receptive field properties versus temporal responses among target neurons, or whether feedback serves a more global role in arousal or attention. These proposed functions are not mutually exclusive, and there is compelling evidence to support multiple functional roles for feedback. In this review, the role of feedback in vision will be explored mainly from the perspective of corticothalamic feedback. Further generalized principles of feedback applicable to corticocortical connections will also be considered.

Abstract:Using whole cell recordings, we studied excitatory and inhibitory postsynaptic currents (EPSCs, IPSCs) in feedforward (FF) and feedback (FB) circuits between areas V1 and LM (lateromedial) in developing mouse visual cortex. We found that in mice reared with normal visual experience, FF and FB synapses onto layer 2/3 pyramidal neurons develop equal but submaximal strengths whose EPSCs are increased by monocular lid suture. In contrast, the development and experience-dependence of FF- and FB-IPSCs is pathway-specific. The difference develops during the critical period by strengthening FF-IPSCs, while keeping FB-IPSC amplitudes constant. Monocular lid suture increases FB-IPSCs but does not affect FF-IPSCs.

Abstract:Experience dependent plasticity in the visual cortex is a key paradigm for the study of mechanisms underpinning learning and memory. Despite this, studies involving manipulating visual experience have largely been limited to the primary visual cortex, V1, across various species. Here we investigated the effects of monocular deprivation (MD) on the ocular dominance (OD) and orientation selectivity of neurons in four visual cortical areas in the mouse: the binocular zone of V1 (V1b), the putative "ventral stream" area LM and the putative "dorsal stream" areas AL and PM. We employed two-photon calcium imaging to record neuronal responses in young adult mice before MD, immediately after MD, and following binocular recovery. OD shifts following MD were greatest in LM and smallest in AL and PM; in LM and AL, these shifts were mediated primarily through a reduction of deprived-eye responses, in V1b and LM through an increase in response through the non-deprived eye. The OD index recovered to pre-MD levels within 2 weeks in V1 only. MD caused a reduction in orientation selectivity of deprived-eye responses in V1b and LM only. Our results suggest that changes in OD in higher visual areas are not uniformly inherited from V1.

Abstract:Sensory cortical areas guide behavior by transforming stimulus-driven inputs into selective responses representing relevant features. A classic example is the representation of edge orientations in the visual cortex, where layer 4 (L4) neurons co-activated by an orientation provide feedforward inputs to specific functional modules in layer 2/3 (L2/3) that share strong recurrent connections. The aligned state of feedforward-recurrent interactions is critical for amplifying selective cortical responses, but how it develops remains unclear. Using simultaneous electrophysiology and calcium imaging in visually naive animals we find that coactivity of L4 neurons and L2/3 modular responses elicited by oriented gratings lacks the tight relationship to orientation preference found in experienced animals. One factor that could contribute to the lack of functionally specific coactivity is high variability in naive L4 neuron responses that decreases significantly following experience. But a computational model of feedforward-recurrent interaction suggests that high variability alone is insufficient to explain the naive state and provides a biological signature of feedforward recurrent misalignment that we confirm with whole-cell recordings: dynamic changes in orientation tuning of L2/3 subthreshold responses shortly after stimulus onset. In conclusion, we provide diverse evidence for a realignment of feedforward-recurrent interactions following experience that is critical for building reliable sensory representations with interlaminar temporal coherence.

Abstract:Although once regarded as a simple sensory relay on the way to the cortex, it is increasingly apparent that the thalamus has a role in the ongoing moment-by-moment processing of sensory input and in cognition. This involves extensive corticofugal feedback connections and the interplay of these with the local thalamic circuitry and the other converging inputs. Here, using the feline visual system as the primary model, some of the latest developments in this field are reviewed and placed in the perspective of an integrated view of system function. Cortical feedback mediated by ionotropic and metabotropic glutamate receptors, and effects mediated by the neuromodulator nitric oxide, all have a role in integrating the thalamic mechanism into the cortical circuit. The essential point is that the perspective of higher-level sensory mechanisms shifts and modulates the thalamic circuitry in ways that optimize abstraction of a meaningful representation of the external world. This review is part of the TINS special issue on The Neural Substrates of Cognition.

Visual experience-dependent auditory and visual plasticity in the mouse visual cortex

Abstract:A lack of early visual experience causes pronounced auditory and visual cross-modal changes. However, the visual cortical region-specific cross-modal organization down to the single neuron level remains unknown. Here we used two-photon calcium imaging in awake mice that were reared in darkness from birth to map auditory and visual responsiveness of single neurons. We targeted neurons in the primary visual cortex (V1) and higher visual cortical areas (HVAs) that resemble the ventral and dorsal stream regions. We found that lateral dorsal stream areas showed a pronounced increase in auditory response strength, even after accounting for tone-induced whisker movement. Strikingly, this was accompanied by a decreased visual drive, measured in number of recruited neurons and response strength, although these visual effects were more widespread across cortical regions. Together, these results provide a comprehensive functional map of auditory and visual cross-modal changes after a lack of early visual experience across the mouse visual cortex. Moreover, our results suggest that a lack of visual drive of dorsal stream regions might provide an opportunity for remaining senses to take over.

Differential destinations, dynamics, and functions of high- and low-order features in the feedback signal during object processing

Abstract:Brain is a hierarchical information processing system, in which the feedback signals from high-level to low-level regions are critical. The feedback signals may convey complex high-order features (e.g., category, identity) and simple low-order features (e.g., orientation, spatial frequency) to sensory cortex to interact with the feedforward information, but how these types of feedback information are represented and how they differ in facilitating visual processing is unclear. The current study used the peripheral object discrimination task, 7T fMRI, and MEG to isolate feedback from feedforward signals in human early visual cortex. The results showed that feedback signals conveyed both low-order features natively encoded in early visual cortex and high-order features generated in high-level regions, but with different spatial and temporal properties. The high-order feedback information targeted both superficial and deep layers, whereas the low-order feedback information reached only deep layers in V1. In addition, MEG results revealed that the feedback information from occipito-temporal to early visual cortex emerged around 200 ms after stimulus onset, and only the representational strength of high-order feedback information was significantly correlated with behavioral performance. These results indicate that the complex and simple components of feedback information play different roles in predictive processing mechanisms to facilitate sensory processing.

Recent visual experience reshapes V4 neuronal activity and improves perceptual performance

Abstract:Recent visual experience heavily influences our visual perception, but how this is mediated by the reshaping of neuronal activity to alter and improve perceptual discrimination remains unknown. We recorded from populations of neurons in visual cortical area V4 while monkeys performed a natural image change detection task under different experience conditions. We found that maximizing the recent experience with a particular image led to an improvement in the ability to detect a change in that image. This improvement was associated with decreased neural responses to the image, consistent with neuronal changes previously seen in studies of adaptation and expectation. We found that the magnitude of behavioral improvement was correlated with the magnitude of response suppression. Furthermore, this suppression of activity led to an increase in signal separation, providing evidence that a reduction in activity can improve stimulus encoding. Within populations of neurons, greater recent experience was associated with decreased trial-to-trial shared variability, indicating that a reduction in variability is a key means by which experience influences perception. Taken together, the results of our study contribute to an understanding of how recent visual experience can shape our perception and behavior through modulating activity patterns in mid-level visual cortex.

Abstract:The cortical visual system consists of many richly interconnected areas. Each area is characterized by more or less specific receptive field tuning properties. However, these tuning properties reflect only a subset of the interactions that occur within and between areas. Neuronal responses may be modulated by perceptual context or attention. These modulations reflect lateral interactions within areas and feedback from higher to lower areas. Recent work is beginning to unravel how horizontal and feedback connections each contribute to modulatory effects and what the role of these modulations is in vision. Whereas receptive field tuning properties reflect feedforward processing, modulations evoked by horizontal and feedback connections may reflect the integration of information that underlies perception.

Abstract:Representational drift – the gradual continuous change of neuronal representations – has been observed across many brain areas. It is unclear whether drift is caused by synaptic plasticity elicited by sensory experience, or by the intrinsic volatility of synapses. Here, using chronic two-photon calcium imaging in mouse primary visual cortex, we find that the preferred stimulus orientation of individual neurons slowly drifts over the course of weeks. By using cylinder lens goggles to limit visual experience to a narrow range of orientations, we show that the direction of drift, but not its magnitude, is biased by the statistics of visual input. A network model suggests that drift of preferred orientation largely results from synaptic volatility, which under normal visual conditions is counteracted by experience-driven Hebbian mechanisms, stabilizing preferred orientation. Under deprivation conditions these Hebbian mechanisms enable adaptation. Thus, Hebbian synaptic plasticity steers drift to match the statistics of the environment.

Abstract:Abstract The majority of synaptic inputs to the primary visual cortex (V1) are non-feedforward, instead originating from local and anatomical feedback connections. Animal electrophysiology experiments show that feedback signals originating from higher visual areas with larger receptive fields modulate the surround receptive fields of V1 neurons. Theories of cortical processing propose various roles for feedback and feedforward processing, but systematically investigating their independent contributions to cortical processing is challenging because feedback and feedforward processes coexist even in single neurons. Capitalising on the larger receptive fields of higher visual areas compared to primary visual cortex (V1), we used an occlusion paradigm that isolates top-down influences from feedforward processing. We utilised functional magnetic resonance imaging (fMRI) and multi-voxel pattern analysis methods in humans viewing natural scene images. We parametrically measured how the availability of contextual information determines the presence of detectable feedback information in non-stimulated V1, and how feedback information interacts with feedforward processing. We show that increasing the visibility of the contextual surround increases scene-specific feedback information, and that this contextual feedback enhances feedforward information. Our findings are in line with theories that cortical feedback signals transmit internal models of predicted inputs. Significance Statement The visual system has circuit mechanisms for processing scene context. These circuits involve lateral and feedback inputs to neurons. These inputs interact with feedforward inputs and modulate neuronal responses to visual stimuli presented outside their receptive fields. Systematically investigating independent contributions of feedback and feedforward processes is challenging because they coexist even in single neurons. Here we use an occlusion paradigm to isolate feedback and lateral signals in human participants viewing natural scene images in fMRI. We show that increasing the visibility of the contextual surround increases scene-specific feedback information, which also enhances feedforward signals. Our findings are in line with theories that cortical feedback signals carry abstract internal models that combine with more detailed representations in primary visual cortex.

Abstract:Neural circuits in sensory pathways develop through a general strategy of overproduction of synapses followed by activity-driven pruning to fine-tune connectivity for optimal function. The early visual pathway, consisting of the retina → visual thalamus → primary visual cortex, has served for decades as a powerful model system for probing the mechanisms and logic of this process. In addition to these feedforward projections, the early visual pathway also includes a substantial feedback component in the form of corticothalamic projections from the deepest layer of primary visual cortex. The role of this feedback in visual processing has been studied extensively in mature animals, yet historically, its role in development has received comparatively little attention. Recent technological advances allowing for selective manipulation of neural activity in development led to the uncovering of a role for feedback in guiding the refinement of the forward projection from retina to visual thalamus. Here we discuss the implications of feedback exerting influence on the development of sensory pathways. We propose several possible advantages to constructing neural circuits with top-down regulation, and discuss the potential significance of this finding for certain neurologic disorders.
The combination of feedforward and feedback processing accounts for contextual effects in visual cortex

Abstract:Sensory systems must combine local features with context to infer meaning. Accordingly, context profoundly influences neural responses. We developed a unified circuit model demonstrating how feedforward and feedback inputs are integrated to produce three forms of contextual effects in mouse primary visual cortex (V1). First, reanalyzing existing data, we discovered that increasing stimulus size only weakly increases the area of V1 neural response, conflicting with previous models of surround suppression (SS). Second, through modeling, we found that, in Layer 2/3, (1) SS and its contrast dependence are largely inherited from Layer 4; (2) Inverse responses (IR) – size-tuned responses to a gray “hole” in a full-field grating – are driven by feedback connections provided they are sufficiently wide; (3) Cross-orientation surround facilitation is induced by the summation of feedback input driving IR with the feedforward-driven classical center response. The model accounts for many previous findings and makes multiple testable predictions.
