Abstract:Characterizing the complex and diverse interactions between cells and nanomaterials (NMs) is essential to advance biomedical applications such as drug delivery. Given the diverse physiochemical properties of NMs, the complex microenvironment and barriers they encounter in vivo, as well as the inherent heterogeneity of cells, it is no surprise that cells can respond very differently to NMs, resulting in disparate cell-nanomaterial interactions (CNI) that affect the choice and reproducibility of downstream applications. Acquiring a deeper and more accurate understanding of the interactions between cells and NMs would unlock the full potential and optimize the performance of nanotherapeutics such as targeted drug delivery and diagnostic. This review provides a comprehensive overview of traditional and emerging methods for characterizing CNI. Here, we discuss traditional techniques including fluorescence-based technologies, electron microscopy and mass spectrometry, and emerging methods like DNA barcoding, with their underlying principles, applications, and properties in the context of CNI studies, and evaluate their strengths and limitations. Additionally, we discuss existing challenges and future prospects in the field of CNI characterization. We hope that through this review, we can inspire the development of next-generation tools for characterizing CNI and optimize the design of NMs to further enhance their biomedical applications.
