Abstract:Sex pheromones are crucial for mate finding in many animals. Long-range attraction, mate recognition, and the elicitation of sexual receptiveness during courtship are typically mediated by different compounds. It is widely unknown, however, how the different components of a species’ pheromone system influence each other. Here, we demonstrated in the parasitoid wasp Nasonia vitripennis that females quickly cease to respond to the male sex attractant after they contact a male’s oral secretion during courtship. We used this behavioral switch to monitor the fractionation of head extracts from male wasps for identification of the bioactive compounds as a blend of ethyl oleate, ethyl linoleate, and ethyl α-linolenate. This is the first identification of a cephalic courtship pheromone in parasitic Hymenoptera. Plasticity in pheromone-mediated sexual behavior of female insects has hitherto been attributed to the transfer of bioactive proteinaceous molecules with the male ejaculate. The pheromone interaction reported here sheds new light on the sexual communication of insects by showing that the sex pheromone response of females can be terminated by males independent of sperm transfer.

Abstract:The use of chemical cues and signals is essential for communication in insects. Wasps of the genus Nasonia (Hymenoptera, Pteromalidae) are gregarious parasitoids that lay their eggs into puparia of cyclorrhaphous flies. During their life cycle, various kinds of semiochemicals are used: (1) a male abdominal sex pheromone that attracts females and induces site fidelity in males, (2) a female-derived contact sex pheromone eliciting courtship behavior in males, (3) an oral male aphrodisiac eliciting receptivity signaling in females and causing a switch in the females' olfactory preferences, (4) chemicals derived from host habitat and host puparia used in olfactory host finding by female wasps, and (5) chemicals used by females to assess the quality and parasitization status of potential hosts. We review the literature on the chemical ecology of Nasonia spp. following the wasps' life cycle from emergence to oviposition. We depict biosynthetic pathways where available, discuss ecological implications, highlight differences among Nasonia species, summarize insights into their olfactory perception and associative learning abilities, and point out gaps in our understanding of the chemical ecology of these parasitoids to be addressed in future studies.

Abstract:Sex pheromones are pivotal for insect reproduction. However, the mechanism of sex pheromone communication remains enigmatic in hymenopteran parasitoids. Here we have identified the sex pheromone and elucidated the olfactory basis of sex pheromone communication in Campoletis chlorideae (Ichneumonidae), a solitary larval endoparasitoid of over 30 lepidopteran pests. Using coupled gas chromatography-electroantennogram detection, we identified two female-derived pheromone components, tetradecanal (14:Ald) and 2-heptadecanone (2-Hep) (1:4.6), eliciting strong antennal responses from males but weak responses from females. We observed that males but not females were attracted to both single components and the blend. The hexane-washed female cadavers failed to arouse males, and replenishing 14:Ald and 2-Hep could partially restore the sexual attraction of males. We further expressed six C. chlorideae male-biased odorant receptors in Drosophila T1 neurons and found that CchlOR18 and CchlOR47 were selectively tuned to 14:Ald and 2-Hep, respectively. To verify the biological significance of this data, we knocked down CchlOR18 and CchlOR47 individually or together in vivo and show that the attraction of C. chlorideae to their respective ligands was abolished. Moreover, the parasitoids defective in either of the receptors were less likely to court and copulate. Finally, we show that the sex pheromone and ( Z )-jasmone, a potent female attractant, can synergistically affect behaviors of virgin males and virgin females and ultimately increase the parasitic efficiency of C. chlorideae . Our study provides new insights into the molecular mechanism of sex pheromone communication in C. chlorideae that may permit manipulation of parasitoid behavior for pest control.

Abstract:Sexual selection theory asserts that females are well adapted to sense signals indicating the quality of potential mates. One crucial male quality parameter is functional fertility (i.e. the success of ejaculates in fertilizing eggs). The phenotype-linked fertility hypothesis (PLFH) predicts that functional fertility of males is reflected by phenotypic traits that influence female mate choice. Here, we show for Nasonia vitripennis, a parasitic wasp with haplodiploid sex determination and female-biased sex ratios, that females use olfactory cues to discriminate against sperm-limited males. We found sperm limitation in newly emerged and multiply mated males (seven or more previous matings) as indicated by a higher proportion of sons in the offspring fathered by these males. Sperm limitation correlated with clearly reduced pheromone titres. In behavioural bioassays, females oriented towards higher doses of the synthetic pheromone and were attracted more often to scent marks of males with a full sperm load than to those of sperm-limited males. Our data support the PLFH and suggest that N. vitripennis females are able to decrease the risk of getting constrained to produce suboptimal offspring sex ratios by orienting towards gradients of the male sex pheromone.

Abstract:Pheromones are secreted molecules that mediate animal communications. These olfactory signals can have substantial effects on physiology and likely play important roles in organismal survival in natural habitats. Here we show that a blend of two ascaroside pheromones produced by C. elegans males primes the female reproductive system in part by improving sperm guidance toward oocytes. Worms have different physiological responses to different ratios of the same two molecules, revealing an efficient mechanism for increasing coding potential of a limited repertoire of molecular signals. The endogenous function of the male sex pheromones has an important side benefit. It substantially ameliorates the detrimental effects of prolonged heat stress on hermaphrodite reproduction because it increases the effectiveness with which surviving gametes are used following stress. Hermaphroditic species are expected to lose female-specific traits in the course of evolution. Our results suggest that some of these traits could have serendipitous utility due to their ability to counter the effects of stress. We propose that this is a general mechanism by which some mating-related functions could be retained in hermaphroditic species, despite their expected decay.

Abstract:The nuptial flight of ants usually occurs during certain periods of the year. Alate females and males fly out of their nests to mate simultaneously. In the genus Camponotus, sex-specific chemicals are deposited in the male head; however, their roles in nuptial flight have not yet been clarified. This study aimed to elucidate the functions of male-specific chemicals in the Japanese carpenter ant Camponotus japonicus. First, we identified three chemicals characteristic to the male - methyl salicylate (MS), methyl 6-methylsalicylate (MMS), and methyl anthranilate (MA) - all of which triggered electroantennogram (EAG) responses in both alate males and females. As the relative content of MS was insufficient for GC comparison, we investigated the quantitative changes of MMS and MA in the male head capsules before and after flight under laboratory conditions. The amounts of both compounds were significantly reduced after flight, which suggested that males secrete them during flight. Thereafter, a field trap experiment was conducted in three fields of the Kyoto Prefecture, Japan, during the nuptial flight season in 2021 using MMS and MA as baits. The number of captured alate males was significantly higher than that of the females, suggesting that these compounds primarily attracted males rather than females. Considering the field conditions, if the local concentration of these chemicals is increased by male aggregation, females may be attracted as they also showed EAG responses. Our findings represent a first step toward understanding chemically mediated male lek formation during the process of male aggregation syndrome in this species.

Abstract:Gregarious parasitoids usually clump their cocoons together and the adults emerge in a synchronized fashion. This makes it easy for them to find mating partners and most copulations indeed take place at the natal patch. Yet, males should leave such sites when females are no longer receptive. As yet, this decision-making process and the possible involvement of pheromones were poorly understood. Here we report on a remarkable use of attractive and repellent pheromones of the well-studied gregarious parasitoid species Cotesia glomerata (L.) (Hymenoptera: Braconidae). Virgin C. glomerata females were found to release an attractive as well as a repellent compound, which in combination arrest males on the natal patch, but after mating the females stop the production of the attractant and the males are repelled. The repellent compound was identified as heptanal, which was also released by males, probably reducing male-male competition on the natal patch. We also confirmed that the sex ratio of the emerging wasps can vary considerably among patches, depending on the relative quality of hosts and the number of females that parasitize a host. The newly revealed use of attractive and repellent pheromone compounds by C. glomerata possibly helps maximize mating success under these variable conditions.

Abstract:Mate recognition is paramount for sexually reproducing animals, and many insects rely on cuticular hydrocarbons (CHCs) for close-range sexual communication. To ensure reliable mate recognition, intraspecific sex pheromone variability should be low. However, CHCs can be influenced by several factors, with the resulting variability potentially impacting sexual communication. While intraspecific CHC variability is a common phenomenon, the consequences thereof for mate recognition remain largely unknown. We investigated the effect of CHC variability on male responses in a parasitoid wasp showing a clear-cut within-population CHC polymorphism (three distinct female chemotypes, one thereof similar to male profiles). Males clearly discriminated between female and male CHCs, but not between female chemotypes in no-choice assays. When given a choice, a preference hierarchy emerged. Interestingly, the most attractive chemotype was the one most similar to male profiles. Mixtures of female CHCs were as attractive as chemotype-pure ones, while a female–male mixture negatively impacted male responses, indicating assessment of the entire, complex CHC profile composition. Our study reveals that the evaluation of CHC profiles can be strict towards ‘undesirable' features, but simultaneously tolerant enough to cover a range of variants. This reconciles reliable mate recognition with naturally occurring variability.

Abstract:The cuticle of virtually any insect is covered by a thin layer of cuticular hydrocarbons (CHCs) typically consisting of a complex mixture of n-alkanes, methyl-branched alkanes, and unsaturated hydrocarbons. Apart from their putative primary function as a desiccation barrier, CHCs are used by insects for communication. In many species of parasitoid wasps, males use CHCs to recognize females, and thus the composition of CHC profiles is typically species- and sex-specific. Sometimes, the biological activity of CHCs is synergized by more polar lipids. In most species, however, the contribution of polar lipids and the role of individual CHCs or CHC classes is unclear. Here we report a CHC-based contact sex pheromone in the pteromalid wasp Urolepis rufipes. Males of U. rufipes were arrested on and showed courtship behavior (wing fanning) and copulation attempts toward cadavers of females, whereas male cadavers and solvent-washed cadavers of either sex elicited no responses. Whole body extracts from females and the non-polar CHC fraction thereof elicited arrestment, courtship, and copulation attempts by males, whereas the fractions containing polar lipids were behaviorally inactive. Analyses of male- and female-derived CHC profiles revealed that they consisted exclusively of n-alkanes and methyl-branched alkanes. Removal of n-alkanes from female-derived CHCs with molecular sieves rendered the remaining methyl-branched fraction inactive. Synthetic n-alkanes in female-specific ratios also did not elicit any responses by males. Two 7-methylalkanes (7-MeC31 and 7-MeC33) were the only two components with a higher relative abundance in females compared to males. However, supplementation of male-derived CHCs with synthetic racemic 7-MeC31 and 7-MeC33 to levels found in females was not sufficient to render male-derived CHCs attractive to males. Enantiopure synthetic 7-MeC31 and 7-MeC33 might have caused different effects but were not available. We conclude that female-derived CHCs function as a contact sex pheromone in U. rufipes. Our data suggest that methyl-branched and n-alkanes act synergistically and that the sex-specific relative composition of the whole profile, rather than the abundances of single components, underlies the elicitation of male courtship behavior.
